Abstract
本发明提供了IRAK1抑制剂联合PARP抑制剂在制备抗肿瘤的药剂中的应用,涉及生物医药技术领域。本发明发现在IRAK1高表达的肿瘤中,IRAK1能促进NBS1磷酸化的发生,进而提高了DNA同源重组修复效率,导致PARP抑制剂治疗不敏感;如果抑制IRAK1酶活,会使NBS1磷酸化减低,同源重组修复效率下降,此时联合PRAP抑制剂的使用,具有明显的合成致死效应。本发明为IRAK1高表达的肿瘤找到了一种新的治疗方案。
Description
技术领域
本发明属于生物医药技术领域,具体涉及IRAK1抑制剂联合PARP抑制剂在制备抗肿瘤的药剂中的应用。
背景技术
胃癌(Gastric cancer,GC)是一种起源于胃黏膜上皮细胞的恶性肿瘤,主要在胃窦小弯侧及幽门区发病。胃癌的发病率和死亡率极高。胃癌高发病的主要原因是胃癌的致病因素多样化,比如幽门螺旋杆菌感染、高硝酸盐和亚硝酸盐饮食、家族遗传性因素、化学暴露、吸烟、饮酒等因素。胃癌高死亡率的主要原因是胃癌的治疗手段及有效的方法有限导致的,目前对于胃癌的治疗方法主要有手术、化疗、免疫疗法、分子靶向治疗和放疗五种方法。但是这些治疗方法及手段对提高患者的总体生存率来说并不太理想。因此,迫切需要找到一些有效的手段和方法来治疗胃癌。
在二十世纪,科学家提出了“合成致死”这个概念,所谓的“合成致死”是指在细胞中,当一个基因发生突变导致功能缺失时,细胞不会死亡,如果两个基因同时发生突变导致功能缺失时,细胞就会死亡。“合成致死”的概念使药物特异性杀伤肿瘤细胞成为了可能,因为肿瘤的发生,通常是由于一些功能比较重要的基因发生突变导致的,对于非肿瘤细胞,该基因则是正常的。因此,可以专门设计和开发一些药物特异性地杀伤肿瘤细胞,而对正常细胞没有明显的杀伤作用。PARP抑制剂就是根据“合成致死”概念开发出来最具有代表性的药物,在一些肿瘤中,比如乳腺癌或者卵巢癌,通常会有BRCA1/2基因突变,BRCA1/2基因突变将会导致同源重组修复受损。对于这类肿瘤,如果使用PARP抑制剂阻止碱基切除修复,肿瘤细胞内自发的DNA单链断裂会变成DNA双链断裂,最终由于同源重组修复缺陷而死亡,这就是PARP抑制剂合成致死的原理。目前,已有四种PARP抑制剂被FDA批准用来治疗乳腺癌、卵巢癌、胰腺癌和前列腺癌等肿瘤。
由于在DNA同源重组修复这条信号通路中,如果其中一个基因发生突变或者功能缺失,同源重组修复就会有缺陷。因此,在后来的研究中,科学家们发现不仅是在BRCA1/2突变或其他同源重组相关基因突变的肿瘤中,PARP抑制剂具有合成致死效应,而且在没有同源重组修复基因突变的肿瘤中,只要抑制与同源重组修复相关基因的功能,PARP抑制剂也能对肿瘤细胞产生明显的合成致死效应。这让PARP抑制剂的运用范围得到了极大的扩展,也让很多之前难以治疗的肿瘤有了新的治疗方法,于是寻找新的药物联合PARP抑制剂来治疗肿瘤成为了一个研究热点。
蛋白激酶抑制剂是一类抑制蛋白质发生磷酸化修饰的药物,目前被FDA批准上市的激酶抑制剂共有52种,其中有46种是用来治疗肿瘤的,例如作用于 RAF激酶的索拉芬妮,主要用于治疗肾细胞癌;作用于EGFR-TK激酶的吉非替尼,主要用于治疗非小细胞肺癌等。激酶抑制剂靶向药物与传统的化疗药物相比,其优势是安全性更高、毒副作用小,治疗效果更明显。因此,激酶抑制剂被认为是肿瘤治疗过程中最有潜力的靶向药物之一。
PARP抑制剂和激酶抑制剂虽然在肿瘤治疗过程中取得了不错的疗效,但是单一的激酶抑制剂或者PARP抑制剂对胃癌的治疗均没有明显的疗效。
发明内容
本发明的目的提供IRAK1抑制剂联合PARP抑制剂在制备抗肿瘤的药剂中的应用,IRAK1抑制剂联合PARP抑制剂使用,可产生明显的协同致死效应,有助于肿瘤的治疗。
本发明提供了IRAK1抑制剂联合PARP抑制剂在制备抗肿瘤的药剂中的应用。
优选的,所述IRAK1抑制剂包括IRAK1/4抑制剂。
优选的,所述PARP抑制剂包括Olaparib或Niparib。
优选的,所述抗肿瘤包括抗IRAK1高表达的肿瘤细胞。

优选的,所述药物组合物中,IRAK1抑制剂和Olaparib的质量比为5:1~5:6,IRAK1抑制剂和Niparib的质量比为1:10。
本发明还提供了一种抗IRAK1高表达的肿瘤细胞的药物,包括IRAK1抑制剂和Olaparib及Niparib。

具体实施方式
本发明提供了一种IRAK1抑制剂联合PARP抑制剂在制备抗肿瘤的药剂中的应用。
本发明所述IRAK1抑制剂,优选包括IRAK1/4抑制剂。在本发明中,由于目前没有IRAK1的选择性抑制剂,只有IRAK4的选择性抑制剂以及能够同时抑制IRAK1和IRAK4酶活的IRAK1/4抑制剂,因此本发明实施例中使用IRAK1/4 抑制剂用来与PARP抑制剂进行联合使用,从而产生合成致死效应。本发明实施例中,使用IRAK4特有的抑制剂与Olaparib进行联用,但发现IRAK4抑制剂与 Olaparib不存在合成致死效应,说明IRAK1/4抑制剂与PARP抑制剂具有合成致死效应主要是由于抑制IRAK1酶活所导致,因此可将IRAK1可以作为胃癌治疗的一个潜在靶点。
本发明所述PARP抑制剂包括Olaparib(奥拉帕尼)或Niraparib。本发明对所述IRAK1抑制剂和PARP抑制剂的来源并没有特殊限定,利用本领域的常规市售试剂即可。
本发明所述抗肿瘤优选包括抗IRAK1高表达的肿瘤细胞,更优选包括胃癌细胞和宫颈癌细胞,如在本发明实施例中利用多种胃癌细胞系,如SGC7901、 SNU1和MKN45,证实了IRAK1/4抑制剂与Olaparib均具有合成致死效应;同时还利用宫颈癌细胞系HeLa细胞进行了实验验证,发现无论是敲除IRAK1还是抑制IRAK1酶活,Olaparib的敏感性均有所增强,证实了在IRAK1高表达的非胃癌细胞系中,IRAK1/4抑制剂与Olaparib也具有合成致死效应;同时 IRAK1/4抑制剂与Niraparib也具有类似的合成致死效应。
本发明还提供了一种抗IRAK1高表达的肿瘤细胞的药物组合物,包括 IRAK1抑制剂和Olaparib,或IRAK1抑制剂和Niraparib。
本发明中,由于没有IRAK1的选择性抑制剂,因此所称IRAK1抑制剂实际上指IRAK1/4抑制剂,且在所述药物组合物中,IRAK1/4抑制剂和Olaparib的质量比,优选为5:1~5:6,IRAK1抑制剂和Niparib的质量比优选为1:10。
本发明还提供了一种抗IRAK1高表达的肿瘤细胞的药物,包括IRAK1抑制剂和Olaparib,或IRAK1抑制剂和Niraparib。
本发明所述药物中优选还包括药学上可接受的辅料。
在IRAK1高表达的肿瘤中,IRAK1能促进NBS1磷酸化的发生,进而提高了DNA同源重组修复效率,导致PARP抑制剂治疗不敏感;如果抑制IRAK1 酶活,会使NBS1磷酸化减低,同源重组修复效率下降,此时联合PRAP抑制剂的使用,具有明显的合成致死效应。
为了进一步说明本发明,下面结合附图和实施例对本发明提供的IRAK1抑制剂联合PARP抑制剂在制备抗肿瘤的药剂中的应用进行详细地描述,但不能将它们理解为对本发明保护范围的限定。
本发明实施例中,所用的试剂和耗材,如非特别说明均为常规得到:
1、细胞系、质粒、菌株、实验动物
细胞系:HEK293T细胞和HeLa细胞为本室传代培养,胃癌细胞系SGC7901 由北京大学免疫学系韩文玲教授馈赠,胃癌细胞系MGC803、SNU1、MKN45 及胃非癌变的上皮细胞系GES-1由北京大学肿瘤医院实验室馈赠。
质粒:本研究使用的SFB/MYC/GST标签的质粒均由Gateway系统构建。
菌株:本研究使用的大肠杆菌TOP10、BL21和DH5α感受态均由本室制备和保存。
实验动物:本研究使用的实验小鼠购买自北京维通利华实验动物技术有限公司。实验动物均饲养于清洁级或SPF级环境内,所有动物实验均符合北京大学医学部伦理委员会的规定。

在本发明实施例中,每个实验都重复三次或三次以上,实验数据采用t检验进行分析,当p<0.05具有统计学意义,*表示p<0.05,**表示p<0.01,***表示p<0.001,****表示p<0.0001。
实施例1
一、实验与方法
1、质粒构建
本发明使用的SFB标签、Myc标签及GST标签的质粒均通过Gateway克隆系统构建,按GatewayTM BP ClonaseTM酶混合物(货号:11789013)和Gateway TM LR ClonaseTM酶混合物(货号:11791043)试剂盒说明书进行操作。
2、质粒的提取
质粒小量和大量提取,按天根生物公司的试剂盒说明书进行。
3、细胞培养、复苏及冻存
1)细胞复苏与培养
从液氮或-80℃冰箱中取出细胞,37℃水浴锅中融化,离心,加入培养基将细胞重悬,然后转移到培养皿中,37℃恒温培养箱中培养,每36~48h传代一次。 HEK293T细胞及HeLa细胞使用DMEM培养基,胃癌细胞系使用1640培养基。
2)细胞冻存
Verse或无菌PBS洗涤细胞,胰酶消化,加入培养基中合胰酶,吹打并将细胞转移至离心管中,离心,弃尽上清,加入细胞冻存液重悬细胞并转移至冻存管中。4℃放置30min,-20℃放置2h,-80℃放置24~48h,液氮中长期冻存。
4、质粒转染
接种细胞,当细胞融合度达到60%~80%时进行转染;根据培养皿的大小配制转染体系,例如,以6cm细胞培养皿为例,向EP管中加入500μL的OPTI-MEM 培养基,再加入5μg质粒,轻轻混匀,然后加入15μL转染试剂,充分混匀后静置15min,将质粒DNA和转染试剂的混合液逐滴加入到细胞培养皿中,轻轻混匀,放入培养箱中继续培养。转染后4~6h更换新的培养基,转染后36~48h 后收集细胞,进行后续实验。
5、SFB标签稳定表达细胞株的制备
HEK293T细胞铺板,当细胞生长融合度达到60%~80%时转染带SFB标签的质粒,转染后4~6h换液,36~48h后加入终浓度为2μg/ml嘌呤霉素培养基,培养6~8天后进行密度梯度稀释,将细胞稀释至终浓度为1个细胞/100μL,稀释后的细胞转移到96孔板中培养,当细胞生长到一定密度后,依次转移到24 孔板、12孔板、6孔板和10cm培养皿中生长,通过Western blot和免疫荧光鉴定出稳定表达的细胞株。
6、CRISPR-Cas9载体的构建及基因敲除细胞的制备
1)通过http://tools.genome-engineering.org网站对IRAK1基因的gRNA(表 2)进行评分,筛选出评分最高、脱靶率最小的两条gRNA。
2)合成引物,构建质粒。
3)测序正确后进行质粒大提、转染,嘌呤霉素筛选,梯度稀释获得单克隆细胞株、通过Western blot和T载体链接测序确认得到基因敲除的细胞株。
7、蛋白质免疫印迹实验(Western blot)
收集细胞,4℃,400g离心5min,弃尽上清,预冷PBS洗涤细胞一遍;弃尽上清,向细胞沉淀中加入细胞裂解液RIPA或NETN和蛋白酶抑制剂,充分混匀,4℃,转盘上旋转裂解20min;4℃,13400g离心10min,吸取上清,采用 Broadford法对细胞总蛋白进行定量,向蛋白样品中加入SDS上样缓冲液,100℃,加热10min;电泳、转膜,5%脱脂牛奶或5%BSA室温封闭1h;加入一抗,4℃孵育过夜;洗膜,加入二抗,室温孵育1h,洗膜,曝光,分析实验结果。
8、免疫共沉淀
1)HEK293T细胞铺板,当细胞生长融合度达到60%~80%时转染两个不同标签的质粒,转染后4~6h换液,36~48h后收集细胞;
2)收集细胞,4℃,400g离心5min,弃尽上清,PBS洗涤细胞一遍;
3)加入600~800μL NETN裂解液,4℃转盘上裂解20min;
4)4℃,13400g,离心10min;
5)将上清转移到新的EP管中,混匀,取40~50μL作为input,加入SDS 蛋白上样缓冲液,100℃加热5~10min;剩下的样品中加入20~30μL S-protein Agarose,4℃转盘上孵育2~3h;
6)4℃,400g离心1min,弃尽上清,加入预冷的NETN裂解液洗涤S-beads 3~5遍,弃尽上清;
8)加入20μL 2×SDS蛋白上样缓冲液,100℃加热10min;
9)电泳、转模、5%脱脂牛奶室温封闭1h,一抗4℃孵育过夜;TBST洗膜三次,每次10min,HRP标记的二抗室温孵育1h,TBST洗膜三次,曝光,结果分析。
9、串联亲和纯化偶联质谱分析
1)选择稳定表达状态最佳的细胞,扩增至20~30盘,IR照射或者加入10 μmol/LCPT处理4~6h,收集细胞,离心,PBS洗涤细胞一遍,弃尽上清;
2)加入12-15ml NETN裂解细胞,4℃裂解20min,离心,将上清(soluble) 转移到离心管中,加入100μL Strepavidin-sepharose,4℃转盘上孵育2~4h;
3)将沉淀(Chromatin,染色质)转移到EP管中,PBS洗涤2~3次;
4)加入Nuclease,超声裂解,37℃水浴锅中孵育5min,离心,收集上清于EP管中,加入50μL Strepavidin-sepharose,4℃转盘上孵育2~4h;
5)离心,弃尽上清,NETN洗涤四次,加入终浓度为5mg/ml Biotin竞争性结合Strepavidin-sepharose上的蛋白,4℃转盘过夜;
6)离心,向上清中加入60μL S-beads,结合3~4h;
7)离心,NETN洗涤S-beads 4~6次,弃尽上清;加入25μL 2×loading buffer,100℃加热10min;
8)电泳,考马斯亮蓝染色,脱色,拍照,切下目的条带,进行质谱检测。
10、免疫荧光实验
1)细胞培养皿中加入盖玻片,铺板,待细胞贴壁后进行相应的处理;
2)弃尽培养基,PBS洗涤细胞,多聚甲醛固定15min;
3)PBS洗涤细胞,0.25%Triton-X100打孔液处理5min;
4)PBS洗涤细胞,2%BSA室温封闭5min;
5)加入一抗,室温孵育1h;PBS洗涤细胞,加入二抗,室温避光孵育30min;
7)PBS洗涤细胞,DAPI染核,封片,共聚焦显微镜下分析实验结果。
11、彗星实验
本研究运用碱性彗星实验,实验步骤如下:
1)称取低熔点琼脂糖溶于PBS中(W/V为1%),65℃水浴锅中溶解;
2)称取正常熔点的琼脂糖溶于PBS中(W/V为0.5%),微波炉加热溶解,取180μL溶解后琼脂糖液均匀浇注到玻片上,加盖玻片,待琼脂糖胶凝固;
3)消化细胞,将细胞制成单细胞悬液,取下步骤2中的盖玻片,将单细胞悬液与1%LMA(Low Melting Agarose)等体积混合;吹打均匀后迅速吸取180 μL混合液滴加到第1层琼脂糖上,加上盖玻片,待琼脂糖胶凝固;
4)将凝胶玻片放置于平皿中,去掉盖玻片,加入预冷的细胞裂解液(2.5mol/LNaCl,0.1mol/L EDTA-2Na,10mmol/L Tris-HCl,pH 10.0,10%DMSO,1%Triton -X100),4℃裂解2h或过夜;
5)取出载玻片,双蒸水洗涤载玻片;加入电泳缓冲液(300mmol/L NaOH, 1mmol/LEDTA-2Na,pH 13.0)解旋0.5~1h;
6)20V,200mA,冰浴中电泳30min;中和液(0.4mol/L Tris-HCl,pH 7.50) 中和;滴加100μL PI避光染色10min;PBS洗涤,荧光显微镜下进行结果分析。
12、流式细胞术检测细胞周期
1)胰酶消化细胞,离心,PBS洗涤;加入250μL PBS重悬细胞,逐滴加入750μL预冷的无水乙醇,轻轻吹打混匀,置于4℃固定过夜;
3)离心,弃尽上清,加入1ml预冷的PBS重悬细胞,4℃,500g离心5min;
4)弃尽上清,加入120μL预冷的PBS,80μL 0.25%Triton-X100打孔液及终浓度为100μg/ml RNase,重悬细胞,37℃水浴锅中孵育30min;
5)加入终浓度为50μg/ml PI,室温避光孵育30min;
6)加入预冷的PBS重悬细胞,300目筛网过滤,流式上机分析结果。
13、克隆形成实验
1)胰酶消化细胞,室温,离心;
2)加入新鲜培养基,重悬细胞,计数,梯度稀释细胞密度为1000个/ml;
3)铺板,每个6cm培养皿加入1ml梯度稀释好的细胞,再补加3ml新鲜培养基,充分振荡,使细胞分布均匀,培养8~12h;
4)进行相应的处理,例如IR照射、CPT及PARP抑制剂等药物处理,每隔3-4天更换一次新鲜的培养基,培养9~15天;如果是联合用药处理,则提前 24h加入激酶抑制剂,然后再加入PARP抑制剂处理24h,PBS洗涤细胞三遍,再加入含有激酶抑制剂的培养基处理48h,PBS洗涤细胞三遍,加入新鲜培养基继续培养7~11天;
5)弃尽培养基,考马斯亮蓝染色30min,清水脱色、晾干、拍照及统计克隆数,相对存活率=处理组实验细胞克隆数/非处理组实验细胞克隆数×100%。
14、电离辐射实验
提前8~12h进行细胞铺板,将细胞放入X射线辐射仪中,设置辐射剂量,进行照射,照射完成后根据实验目的收集细胞或继续培养所需时间。
15、CCK8(Cell Counting Kit-8)筛选激酶抑制剂与PARP抑制剂合成致死实验
1)消化细胞,离心;弃尽上清,加入培养基重悬细胞,计数;
2)将细胞稀释至2×104/ml;分为四组,分别加入5μmol/L Olaparib、10μmol/L 激酶抑制剂、5μmol/L Olaparib+10μmol/L激酶抑制剂和对照组,对照组加入等体积的DMSO;
3)将96孔板的边缘孔加入200μL的PBS,中心孔加入稀释后的细胞100μL,每组细胞铺六个复孔,三个复孔作为0h时间点的OD测量值,三个复孔作为 96h时间点OD测量值;
4)加入10μL CCK8;培养2h,测定OD450吸光度值,作为0h时间点测量值;
5)剩下的细胞继续培养96h,然后加入10μL CCK8,孵育2h,测定OD450吸光度值,分析合成致死的结果。
16、全蛋白磷酸化组学质谱分析
将HeLa细胞和HeLa-IRAK1-KO细胞通过射线/Olaparib/CPT处理后,收集细胞于冻存管中,液氮速冻,然后转移到干冰中,送往杭州景杰生物公司进行全蛋白磷酸化组学质谱分析。
17、邻位连接技术(Proximity ligation assay,PLA)
按Duolink In Situ PLA(货号DUO92001)试剂盒说明书进行操作。
二、结果与分析
1.通过激酶抑制剂库和生物数据库筛选出IRAK1抑制剂与PARP抑制剂具在胃癌细胞中具有合成致死效应
在胃癌细胞系MGC803中使用PARP抑制剂Olaparib与240种激酶抑制剂进行联合用药筛选(图1中A)。筛选结果发现多种激酶抑制剂与Olaparib具有合成致死效应(图1中B),在合成致死效应排名前十的激酶抑制剂分别是Src 抑制剂、ULK1抑制剂、IGF-1R抑制剂、ATM抑制剂、ERK抑制剂、PKD1抑制剂、PAK4抑制剂、ROCK1抑制剂、IRAK1/4抑制剂和MTOR抑制剂。
为区分是IRAK1酶活还是IRAK4酶活或是二者共同发挥作用,进而与PARP 抑制剂具有合成致死效应,使用IRAK4特有的抑制剂与Olaparib进行联用,发现IRAK4抑制剂与Olaparib不存在合成致死效应(图1中C),这说明IRAK1/4 抑制剂与PARP抑制剂具有合成致死效应主要是由于抑制IRAK1酶活所导致的。
通过生物数据库对五个候选基因进行进一步筛选,以便确认最终的候选者。首先通过人类蛋白质组学数据库对五个候选基因进行分析,发现在免疫组织化学染色中,IRAK1蛋白在正常胃组织和胃癌组织中的表达差异最大,表达量也最高;而PKD1和MTOR在正常胃组织中的表达高于胃癌组织;PAK4、ROCK1 在正常胃组织和胃癌中的表达差异不大(图1中D、E)。综合激酶抑制剂库和生物数据库的筛选结果,本发明认为IRAK1可以作为胃癌治疗的一个潜在靶点。
2、在多种胃癌细胞中IRAK1抑制剂与PARP抑制剂具有合成致死效应
在胃癌细胞系SGC7901、SNU1和MKN45中进行实验探索。实验结果表明, IRAK1/4抑制剂与Olaparib在胃癌细胞系SGC7901、SNU1和MKN45中均具有合成致死效应(图2中A)。这些实验结果排除了细胞特异性影响,说明在多种胃癌细胞中,IRAK1/4抑制剂与Olaparib均具有合成致死效应。
由于CCK8实验主要是在短时间内观察细胞的增殖状态,为了进一步探索 IRAK1/4抑制剂与PARP抑制剂在体外的长期协同致死效应,本发明在细胞 SGC7901和MGC803中进行了单克隆形成实验。实验结果提示在胃癌细胞 SGC7901中,IRAK1/4抑制剂与Olaparib具有非常明显的合成致死效应(图2 中B、C)。
为了排除IRAK1/4抑制剂脱靶效应导致的协同致死效应,通过CRISPR-Cas9 技术制备IRAK1的KO细胞(图3中C),在IRAK1的KO细胞中,加入Olaparib 也具有明显的致死效应(图2中D-F)。证明敲除IRAK1或者抑制其酶活都能使 Olaparib的敏感性增强,产生良好的合成致死效应。在胃癌细胞MGC803中也存在类似的实验结果,即IRAK1/4抑制剂与Olaparib具有良好的协同致死效应 (图2中G、H)。
3、在其他IRAK1高表达的肿瘤细胞中IRAK1抑制剂与PARP抑制剂也具有合成致死效应
在HeLa细胞中也进行与上述相同的实验,结果显示无论是敲除IRAK1还是抑制IRAK1酶活,Olaparib的敏感性均有所增加(图3中A、B)。
使用碱性彗星实验来观察二者联用后对细胞产生的损伤情况。结果发现在 HeLa细胞中,单独使用IRAK1/4抑制剂时,细胞并没有产生明显的拖尾;单独使用Olaparib时细胞产生了一定的拖尾;当二者联用时,细胞产生了明显的拖尾(图3中C),证实IRAK1/4抑制剂与PARP抑制剂联用时会导致细胞产生更严重的DNA损伤。
另外,敲除IRAK1也会导致细胞产生一定的拖尾,这说明敲除IRAK1可能会导致一定的自发性DNA损伤产生,在IRAK1 KO细胞中加入Olaparib处理后,彗星实验拖尾现象呈现明显增加。
4、IRAK1/4抑制剂与PARP抑制剂在非癌变的胃上皮细胞中不具有合成致死效应
选择非癌变的胃上皮细胞系GES-1进行单克隆形成实验。实验结果提示在 GES-1细胞中,IRAK1/4抑制剂与Olaparib联用后并没有产生明显的协同致死效应(图4中A、B),这说明IRAK1/4抑制剂与PARP抑制剂联用时只对肿瘤细胞产生选择性的杀伤,而对非肿瘤细胞的杀伤作用并没有因为联用而增强。为了进一步探索IRAK1/4抑制剂与Olaparib联用时对细胞是否在短期内会造成 DNA损伤,进行碱性彗星电泳实验探索,实验结果显示IRAK1/4抑制剂与 Olaparib联用时,细胞产生的拖尾长度没有比使用单一药物产生所产生的拖尾长度强(图4中C),这说明IRAK1/4抑制剂与Olaparib联用时对非肿瘤细胞并不产生明显的毒副作用,两种抑制剂联用后的毒副作用并没有比单一药物的毒副作用强,这提示IRAK1/4抑制剂与Olaparib联用可以作为IRAK1高表达肿瘤治疗的候选方案之一。
5、裸鼠成瘤实验证实IRAK1/4抑制剂与PARP抑制剂在体内具有协同致死效应
Niraparib是被FDA批准上市的另一种PARP抑制剂,其对PARP家族酶活抑制具有更强的效力,我们在体内动物实验中使用了Niraparib抑制剂。我们首先将胃癌细胞MGC803接种于Balb/c裸鼠皮下,当肿瘤生长到一定大小时,根据肿瘤大小和小鼠体重随机将小鼠分为4组:分别为对照组、IRAK1/4抑制剂组(5mg/kg)、Niraparib(50mg/kg)组和IRAK1/4抑制剂(5mg/kg)+Niraparib(50mg/kg) 组,对照组给予DMSO,实验组分别给予IRAK1抑制剂、Niraparib和两种药物联用治疗,每隔两天腹腔给药一次,同时测量肿瘤的大小和小鼠体重的变化,在肿瘤长到一定大小后将小鼠处死,取出肿瘤,进行数据分析。
实验结果提示当进行药物治疗一定时间后,相比于对照组,Niraparib组的肿瘤体积有所减小,IRAK1/4抑制剂组的肿瘤也有所减小,而IRAK1抑制剂与 Niraparib联用组的肿瘤则明显减小(图5中A、B);另外,药物联用组肿瘤的重量也明显小于对照组、IRAK1/4抑制剂组和Niraparib组(图5中C)。在全程的治疗中,对照组、IRAK1/4抑制剂组、Niraparib组和药物联用组的小鼠体重变化并没有明显的区别(图5中D),这说明不论是IRAK1/4抑制剂、Niraparib 还是两药联合运用后对小鼠均没有明显的毒副作用,这提示IRAK1抑制剂与PARP抑制剂可以作为胃癌治疗的候选药物。
6、敲除IRAK1会导致轻微的基因组不稳定
通过碱性彗星电泳实验观察药物处理后细胞的DNA损伤情况时,发现相比于对照组,在HeLa细胞和SGC7901细胞中敲除IRAK1,细胞均能产生明显的拖尾(图6中A、B),这说明敲除IRAK1后细胞会有一定的自发性DNA损伤产生。
在蛋白水平上检查γ-H2AX的变化,发现敲除IRAK1后,γ-H2AX在蛋白水平上有一定程度的升高(图6中C),这进一步证实敲除IRAK1会导致细胞有轻微的自发性基因组不稳定。在实验中,敲除IRAK1后,相比于对照组细胞, KO组细胞的微核有所增加,极少部分细胞还存在有丝分裂障碍,细胞核型大小不一致或者产生多核(图6中D),这些实验结果说明IRAK1在维持基因组的稳定性方面可能发挥着一些以前没有被报道过的功能。
7、敲除IRAK1或者抑制其酶活会导致细胞DNA修复受损
当DNA受到损伤后,损伤修复蛋白就会被招募到DNA断裂处,如果用相应蛋白的抗体进行免疫荧光染色,可以观察到细胞核中有荧光亮点的聚集,这些荧光亮点被称为Foci,因此Foci代表了损伤修复蛋白在损伤位点的募集。
综合前面的实验结果可知敲除IRAK1会导致基因组不稳定,γ-H2AX在蛋白水平上升高,这提示IRAK1在DNA损伤修复中可能发挥着重要的作用,接下来,本发明还通过实验对其进行进一步的探究。首先,将HeLa细胞分为三组: WT组、IRAK1/4i组和IRAK1-KO组,分别给予3Gy射线(IR)处理,然后让细胞恢复24h,再通过免疫荧光检测γ-H2AX foci的变化,探索敲除IRAK1或者抑制其酶活是否影响DNA修复。在实验结果中发现敲除IRAK1或者抑制其酶活,γ-H2AX foci会明显增多(图7中A、B)。同时也通过流式细胞术检测细胞周期变化,实验结果证明敲除IRAK1后细胞周期没有发生明显的改变(图7 中C),这说明γ-H2AX foci的变化不是由于细胞周期导致的,是敲除IRAK1或者抑制其酶活后DNA修复受损所导致的。
8、敲除IRAK1会导致RAD51招募受损,RPA2(S4-S8)磷酸化降低
检测更多的DNA损伤修复相关蛋白的foci变化。将WT组细胞和IRAK1-KO 组细胞给予10Gy的射线(IR)处理,让细胞恢复3小时,然后通过免疫荧光检测DNA损伤修复相关蛋白foci的变化,发现敲除IRAK1后RAD51的招募明显受损(图8中A、B),同时也检测了细胞周期的变化,发现敲除IRAK1对细胞周期并没有产生明显的影响(图8中C),这说明RAD51 foci的招募受损不是由于细胞周期导致的,而是敲除IRAK1所导致的。另外,通过免疫荧光和Western blot发现敲除IRAK1后RPA2(S4-S8)的磷酸化明显减弱(图8中D-F)。这些实验结果证明,敲除IRAK1或者抑制其酶活会导致DNA修复受损,这种受损方式主要是通过RAD51介导的同源重组修复方式来发挥作用的。
9、敲除IRAK1不影响53BP1的招募
在射线处理的条件下检测53BP1 foci的招募变化。通过免疫荧光实验,发现敲除IRAK1对53BP1的招募没有明显影响(图9中A、B),这说明IRAK1 不参与非同源末端连接的调节。
综上可知,IRAK1参与了DNA损伤修复调节,敲除IRAK1或者抑制其酶活会导致DNA修复受损。
10、在DNA损伤后IRAK1会发生磷酸化
由于IRAK1属于激酶,本发明首先探索在PARP抑制剂、CPT和电离辐射处理后IRAK1能否被激活。实验结果显示,当PARP抑制剂、CPT和电离辐射造成DNA损伤后,IRAK1会发生磷酸化(图10中A、B)。因此,将HeLa细胞和HeLa-IRAK1 KO细胞进行DNA损伤处理后,送到杭州景杰生物公司进行全蛋白磷酸化组学质谱分析(图10中C、D)。
11、全蛋白磷酸化组学质谱揭示IRAK1调节了NBS1的磷酸化
在对质谱结果进行基于GO数据库分类和KEGG通路分析之后,发现敲除 IRAK1会导致DNA同源重组修复信号通路下调(图11中A),这与前面观察到敲除IRAK1或者抑制其酶活会导致γ-H2AX foci增多、RAD51招募受损、 RPA2磷酸化降低和53BP1的招募不受影响的结果相符合,这说明IRAK1确实参与了DNA损伤修复调节,而且主要是调节了同源重组修复相关的信号通路。
在全蛋白磷酸化组学质谱结果中,发现了很多与DNA损伤修复和DNA复制相关的分子,尤其是当敲除IRAK1后,NBS1(NBN)S432和S343位点的磷酸化明显降低(图11中B)。NBS1属于MRN复合物(MRE11-RAD50-NBS1) 的组成成员之一,MRN复合物参与了DNA损伤的感知、反应、DNA双链断裂 (DSB)修复、DNA重组、端粒完整性的维持、细胞周期检查点的控制和减数分裂等功能。这说明IRAK1很可能是通过调节NBS1的磷酸化,进而参与了DNA 损伤修复调节。
12、在DNA损伤后IRAK1会向细胞核中聚集
全蛋白磷酸化组学质谱结果提示IRAK1可能调节了NBS1的磷酸化,由于 NBS1主要定位于细胞核,而IRAK1主要定位于细胞质,也有文献报道IRAK1 可以定位于细胞核,因此通过免疫荧光对IRAK1的定位进行分析,发现IRAK1 大部分定位于细胞质,少部分定位于细胞核(图12)。在射线处理后,IRAK1 定位于细胞核的部分明显增加(图12),这说明在DNA损伤发生后,可能是细胞质中的IRAK1入核调节了NBS1的磷酸化。
13、实验结果证实IRAK1调节着NBS1和ATM的磷酸化
在HeLa细胞中通过Western blot验证NBS1的磷酸化变化,发现在IR处理 1h后,敲除IRAK1或者抑制其酶活会导致NBS1 S343位点的磷酸化明显降低 (图13中A);为了排除细胞系的特异性,也在胃癌细胞MGC803中进行了同样的实验,实验结果进一步证实了抑制IRAK1酶活会导致NBS1 S343和S432 位点的磷酸化明显降低(图13中B)。
同时抑制IRAK1酶活还会导致ATM S1981位点的磷酸化降低(图14中B)。为了进一步证明IRAK1调节着NBS1的磷酸化,在HeLa细胞和胃癌细胞 SGC7901中进行免疫荧光实验探索,得到了同样的结论,敲除IRAK1或者抑制其酶活会导致NBS1 S343位点的磷酸化明显降低(图13中C、D)。在Olaparib 处理的条件下,也得到了同样的结果,当敲除IRAK1或者抑制其酶活会导致 NBS1 S343和S432位点的磷酸化明显降低(图13中E)。
最后,在IRAK1过表达的细胞中,NBS1 S343、S432及ATM S1981位点的磷酸化有所增强,而在IRAK1酶活缺失突变体过表达的细胞中没有检测到这一现象(图13中F)。这些实验结果充分证实了IRAK1调节着NBS1 S343、S432 和ATM S1981位点的磷酸化,进而调节了DNA同源重组修复。
14、在蛋白水平上IRAK1不影响NBS1的表达及定位分布
对IRAK1影响NBS1磷酸化的机制进行了进一步探索。由于蛋白的表达水平和定位分布都会影响到磷酸化的变化,在全蛋白磷酸化组学质谱结果中,磷酸化位点降低最明显的是SRRM1的S549和S551位点,而SRRM1是一个与转录调节相关的分子。因此,本发明首先在mRNA水平上对ATM及MRN复合物进行分析,发现抑制IRAK1酶活会导致ATM的mRNA水平略有增高,但是MRN复合物的mRNA水平则没有明显的改变;在IRAK1的KO细胞中,ATM 及RAD50的mRNA水平没有明显的改变,而NBS1和MRE11的mRNA水平有所降低(图14中A)。接下来,在蛋白水平上检测MRN复合物的表达情况及定位变化,发现敲除IRAK1或者抑制其酶活对MRN复合物的蛋白表达水平没有明显影响,细胞质和细胞核内MRN复合物的分布也没有明显的区别(图 14中B)。另外,对MRN复合物可溶部分和染色质部分进行分离,结果显示不管有无射线处理,敲除IRAK1或者抑制其酶活均不影响MRN复合物的定位分布(图14中C)。这些实验结果说明IRAK1并不是通过影响MRN复合物的表达和定位来调节NBS1的磷酸化。
15、IRAK1与NBS1存在相互作用,在DNA损伤后IRAK1与NBS1的相互作用更强
首先,对NBS1进行亲和串联质谱分析,质谱结果显示其存在8个IRAK1 的肽段(图15中A),这提示NBS1与IRAK1可能存在相互作用。接下来,在 SFB-IRAK1的稳定表达细胞中进行pull down实验,发现IRAK1与NBS1存在相互作用,并且在DNA损伤发生后的相互作用更强(图15中B)。
邻近连接技术(Proximity ligation assay,PLA)是一种可以研究蛋白质定位、定量、相互作用、修饰及功能的体外蛋白质分析技术。其原理是通过一对亲和探针对目的分子双识别,继而产生可扩增的检测信号,将蛋白质的检测转变成为对DNA的检测,实现痕量蛋白质的分析,具有极高的检测灵敏度和特异性。为了近一步确认NBS1与IRAK1是否真的存在相互作用,在SFB-IRAK1稳定表达的细胞株进行了PLA实验。实验结果显示NBS1与IRAK1存在共定位,并且在DNA损伤发生后二者的共定位增加(图15中C)。以上的实验结果说明 IRAK1与NBS1存在相互作用,在DNA损伤后,二者的相互作用更强,在IRAK1 高表达的肿瘤中,IRAK1能促进NBS1的磷酸化发生,进而会招募更多的修复蛋白RPA及RAD51到损伤的DNA处,提高了同源重组修复效率,导致肿瘤细胞对PARP抑制剂不敏感。如果使用IRAK1/4的抑制剂抑制IRAK1酶活,则 NBS1磷酸化降低,修复蛋白RPA及RAD51的招募受损,同源重组修复效率降低,进而增加了肿瘤细胞对PARP抑制剂的敏感性(图16)。
尽管上述实施例对本发明做出了详尽的描述,但它仅仅是本发明一部分实施例,而不是全部实施例,人们还可以根据本实施例在不经创造性前提下获得其他实施例,这些实施例都属于本发明保护范围。

1.IRAK1抑制剂联合PARP抑制剂在制备抗肿瘤的药剂中的应用。
2.根据权利要求1所述应用,其特征在于,所述IRAK1抑制剂包括IRAK1/4抑制剂。
3.根据权利要求1所述应用,其特征在于,所述PARP抑制剂包括Olaparib或Niparib。
4.根据权利要求1所述应用,其特征在于,所述抗肿瘤包括抗IRAK1高表达的肿瘤细胞。
5.根据权利要求1或4所述应用,其特征在于,所述IRAK1高表达的肿瘤细胞包括胃癌细胞和宫颈癌细胞。
6.一种抗IRAK1高表达的肿瘤细胞的药物组合物,其特征在于,包括IRAK1抑制剂和Olaparib,或IRAK1/4抑制剂和Niparib。
7.根据权利要求6所述药物组合物,其特征在于,所述药物组合物中,IRAK1抑制剂和Olaparib的质量比为5:1~5:6,IRAK1抑制剂和Niparib的质量比为1:10。
8.一种抗IRAK1高表达的肿瘤细胞的药物,其特征在于,包括IRAK1抑制剂和Olaparib,或IRAK1/4抑制剂和Niparib。
9.根据权利要求8所述药物,其特征在于,还包括药学上可接受的辅料。
