Description

Probiotic composition for improving hair health condition and application thereof
Technical Field
The invention relates to the technical field of probiotics, in particular to a probiotic composition for improving the health condition of hair and application thereof.
Background
Alopecia is a physiological phenomenon caused by the fact that the number of new hairs on the scalp is less than the number of fallen hairs, hair is thinned for a long time, and then partial or all areas of the scalp have no hairs, namely complete alopecia is caused. Although the hair loss does not affect the health of the body, the hair loss greatly affects the beauty of the body, and particularly causes great trouble to the body and the heart of people in the current 'exterior-emphasized' age.
Factors that lead to hair loss are: seasonal alopecia, neurogenic alopecia, chemical alopecia, physical alopecia, lipoid alopecia (androgenetic alopecia), and the like, and among these factors, lipoid alopecia (androgenetic alopecia) and neurogenic alopecia are the most common.
The neurogenic alopecia is related to stress, and the excessive stress can cause physical and mental fatigue, liver fire and liver depression. According to the traditional Chinese medicine, the liver depression hurts the spleen, namely the liver takes the spleen (liver wood is used for tonifying spleen earth), and the hair loss is further caused after the spleen and the stomach are weak.
Seborrheic alopecia (seborrhoeic alopecia, SA) is also called androgenetic alopecia, and is called as 'carious alopecia' and 'decayed hair tinea' in traditional Chinese medicine. The common skin damage of the disease is the top of the head or M-type progressive alopecia of the forehead, and the plain scalp is itchy, oily and dandruff. Seborrheic alopecia is divided into acute and chronic. In the acute stage, the scalp is oiled, or dandruff or pimple, or itch, and the hair is scooped up and falls off in a short time and even completely fades. The scalp in the chronic period is greasy and shiny, is oiled, has a large amount of grey bran-like dandruff, is dry, lacks luster, has severe pruritus, and mostly slowly falls off from the two sides of the forehead and the top of the head, and baldness is formed after years or decades, but the alopecia totalis is not easy to form.
The western medicine considers that the causes of seborrheic alopecia are complex and related to genetic factors, endocrine dyscrasia, emotion, autoimmunity and the like, but the main related factors are androgen metabolism disorder and are closely related to the levels of testosterone and a metabolite Dihydrotestosterone (DHT) thereof, 5 alpha-reductase of follicular unit and Androgen Receptor (AR). Only two drugs, finasteride and minoxidil, have received acceptance by the united states Food and Drug Administration (FDA) (u.s.a) as alopecia therapeutics to date. However, because of unpredictable effect and side effect, and long-term administration is needed, the alopecia is more serious after the medicine is stopped, and the use is limited.
The traditional Chinese medicine recognizes alopecia earlier, and considers that hair growth and metabolism are originated from viscera, originated from essence and blood, and are honored in meridians and collaterals, and are longer than skin striae. The record of Huangdi's classic on medicine: the excessive blood-qi can lead to the strong kidney-qi, so it turns black; deficiency of blood-qi can lead to kidney-qi deficiency, leading to white hair and loss, suggesting that hair loss is related to kidney-qi and blood-qi. Wang Qingren, yi Lin correction records that the hair falls off and all medical books are concerned with blood damage. Lingshu-Yin twenty five people cloud: "blood and qi are less, without hair, and rare and withered if there is blood and qi. The theory of the etiology of various diseases and the hair loss state are recorded in the book: if blood is abundant, the hair will be lustered, so the beard and hair will be beautiful; if the blood and qi are weakened, the meridians are exhausted and unable to nourish, so the beard and hair fall off. Plain questions and five zang organs generation cloud: the kidney also binds to the bone, it also nourishes hair, and it also governs spleen. Therefore, it is \8230, and if it is eaten more often, it will cause bone pain and fall. According to the above classic, the hair is rich and moist and closely related to qi and blood. The spleen is the source of the generation and transformation of qi and blood, and is the acquired root, and the function of the spleen in transportation and transformation can make qi and blood up to the vertex, thereby nourishing hair, and making hair glossy and tough. The students think that spleen earth is healthy and healthy for ever, one of the spleen earth can generate qi and blood and cultivate the origin of hair growth, and roots of all things are planted in the earth, which is the 'earth grows all things'; secondly, the lung governs skin and hair adheres to the skin, and grows by means of lung qi diffusing and moistening, so that the lung can be fumigated and hair can be moisturized by virtue of hilling; thirdly, the hair can tonify the innate essence in the acquired, the spleen and earth are healthy and strong, the acquired essence is strong, and the innate essence can tonify the innate essence, so that the kidney essence is full and the hair is luxuriant.
Therefore, no matter what the liver and kidney are responsible for, it is the essential method for promoting hair growth, strengthening and consolidating hair, preventing alopecia by the way of spleen-earth, transporting and transforming qi and blood.
The probiotics has the powerful effect of conditioning the gastrointestinal tract, promotes the nutrient absorption and keeps the gastrointestinal tract healthy by regulating the host mucous membrane and system immune function or regulating the flora balance in the intestinal tract, has the probiotic function, has the functions of promoting the digestion and absorption of nutrient substances, improving the immunity of the organism, maintaining the intestinal tract flora structure balance, inhibiting intestinal inflammation, protecting the intestinal tract mucous membrane barrier and the like, is related to the functions of the spleen and the stomach in the traditional Chinese medicine, and promotes the functions and the effects, namely, the functions are equivalent to the functions of strengthening and transporting the spleen and the stomach in the traditional Chinese medicine. Therefore, from the perspective of traditional Chinese medicine, the probiotics has the function of strengthening and transporting the spleen and stomach.
Therefore, the method has the advantages of screening and optimizing probiotics and the combination thereof, strengthening the spleen and stomach, and transporting and transforming qi and blood to achieve the effects of promoting hair growth, fixing hair and preventing alopecia, and is a feasible and innovative direction.
Patent CN110325194A discloses a composition containing fermentation product of aquatic animals for preventing alopecia and promoting hair growth, which contains lactobacillus and yeast fermentation product of aquatic animals or aquatic animal processing by-products as effective ingredients, wherein the lactobacillus includes lactobacillus acidophilus, pediococcus pentosaceus, pediococcus cerevisiae, etc. The composition has effects of inhibiting and preventing alopecia and promoting hair growth, but the disclosed strain is many and complicated, and is not clear and optimized. And the samples used in the implementation examples are fermented fish oil. The preparation process of the fermented fish oil comprises the following steps: adding Lactobacillus plantarum and Saccharomyces cerevisiae, fermenting, extracting fish oil with hexane, and recovering hexane to obtain fermented fish oil. Thus, the product is free of active probiotics and is also substantially free of probiotic or metabolite components thereof (little fat soluble components will be incorporated into the fish oil). Therefore, it is not confirmed that the effective ingredients for preventing hair loss and promoting hair growth include probiotics.
The market lacks a safe and healthy probiotic product which can be orally taken or/and externally applied and has better effects of promoting hair growth, fixing hair and preventing alopecia.

Detailed Description
The present invention is further described below with reference to specific embodiments, and it should be noted that, without conflict, any combination between the embodiments or technical features described below may form a new embodiment. The raw materials, equipments and the like used in the following examples are commercially available unless otherwise specified.
In the examples of the present invention, the probiotic strains used were all from Guangdong Yivikov technologies, inc.
Test system: the mouse assay system was set up as follows.
In the following experiments of the embodiments of the present invention, the experiments of mice were used to verify the hair growth promotion, hair fixing and hair loss prevention, and the animal model was a sodium sulfide depilation mouse model.
Animal sources: selecting Kunming white mice, which are from southern medical university animal experiment center, have the body weight of 18-22g and are male, and the age is 6-8 weeks.
Establishing a model: taking a mouse, breeding the mouse for 1 week, adapting to the environment, marking a depilation region on the back of the mouse by picric acid, shaving the hair of the mouse marking region by a shaver, and then removing the hair by 6% Na 2 And (S) carrying out depilation by using the solution. The depilatory area is 10% of the surface area of the mouse body (the depilatory area is about 6 cm calculated according to the Meeh-Rubner formula) 2 Thus, it was determined that each mouse was 6 cm 2 Unhairing), selecting smooth and non-traumatic mouse as model success model.
Grouping and trial method: the mice successfully modeled are randomly grouped into 8 mice each group, and the mice are divided into a blank control group, a positive control group and a probiotic group. All groups of animals were dosed 1/day for 30 consecutive days.
Blank control group (no drug, jar stomach distilled water).
Positive control group (finasteride, gavage 0.15 mg/kg). When the finasteride is used for treating alopecia, the application method comprises the following steps: orally taking for 1 mg/time and 1 time/day, wherein the dosage of the mouse is 10 times of that of the human body according to the average weight of 70 kilograms of the human body, the gastric lavage amount of the mouse is 0.143 mg/kilogram, and the whole is taken according to 0.15 mg/kilogram.
Probiotic group: mice were gavaged with probiotic powder at 22 hundred million CFU/kg.
Index measurement: the measurement indexes include the length of the new hair, the weight of the new hair and the number of hair follicles.
(1) Measurement of the Length of newborn Hair in mice
At the end of the experiment, 10 new hairs were randomly picked up in the experimental area of each mouse using tweezers and a magnifying glass, the length of the 10 new hairs was measured with a vernier caliper, the average value of the new hair length of each mouse was calculated, and finally the average value of each group was calculated and counted as X.
(2) Mouse new hair weight determination
All new hairs were carefully shaved with a shaver on the last day of dosing, weighed and the mean new hair weight (including the 10 hair weight measured above) was calculated for each group of mice and calculated as Y.
(3) Determination of number of hair follicles in mice
At the end of the experiment, all mice were sacrificed, the skin of the back experimental area where hairs had been shaved was taken, the skin was fixed with 10% formalin, the hair follicle cross section was taken, the number of hair follicles was observed under a microscope, 3 fields were randomly selected for each under x 100 conditions, the number of hair follicles was counted, the average of the 3 fields was taken as a sample hair follicle count, and was counted as Z.
When scoring the test sample, the above-mentioned 3 criteria were considered in combination. The embodiment of the invention adopts a standard score calculation method, and the specific calculation process is as follows: average hair length group (X) i ) Each group of data was compared to a blank control group (X) 0 ) By a difference of (X) i -X 0 ) The maximum difference is 100 points, the blank control group is 0 points (the blank group is also counted as an index group, namely the blank control group-blank control group, and 0 is obtained, the counting standard is 0 point, if the data of the sample group is smaller than that of the blank group, the sample has the reverse phase effect and is directly shaved off). The other groups were scored as: (X) i -X 0 )/(X i -X 0Max *100. The average hair weight Y and the average hair follicle number Z are calculated according to the same method.
Among the factors, hair follicles are a key factor for hair growth, and healthy hair can grow only in mature hair follicles, so that the weight is given to 40%, and the hair length index and the hair weight index are given to 30% respectively. The final scores for each group were calculated according to the following formula: each set of scores = (X) i -X 0 )/(X i -X 0Max *100*30%+(Y i -Y 0 )/(Y i -Y 0Max *100*30%+(Z i -Z 0 )/(Z i -Z 0Max *100*40%。
Verification system one: screening test for probiotics to promote hair growth, fix hair and prevent alopecia
1. Preliminary screening test for probiotics
A series of probiotic strains are subjected to a mouse hair-fixing and hair-loss-preventing test, and comprise bifidobacterium adolescentis Y54, bifidobacterium bifidum Y22, lactobacillus acidophilus K43, lactobacillus paracasei K9, bifidobacterium animalis subsp lactis Y6, bifidobacterium longum W68, lactobacillus reuteri K07, pediococcus pentosaceus LN-PT16, lactobacillus rhamnosus YGG and lactobacillus fermentum FM-12. And (3) taking freeze-dried powder of the strain, performing intragastric administration on the mouse according to the test system, testing, measuring the 3 groups of indexes after the test is finished, and calculating the standard score. The specific results are shown in Table 1 below.
Mouse dose conversion method: for a probiotic experiment group, the probiotic is eaten per person in an amount of 150 hundred million CFU (circulating fluid units) per day, the weight of a human body is 70 kilograms on average, the dosage per kilogram of the human body is 2.14 hundred million/kg, the dosage of a white mouse is 10 times that of the human body, namely 21.4 hundred million CFU/kg, and the whole is taken in an amount of 22 hundred million CFU/kg (counted upwards) to be irrigated to the stomach of the mouse (in order to facilitate the operation of irrigating the stomach, 1ml of purified water is added for suspension during the irrigation of the stomach).
The positive control group (finasteride, gavage 0.15 mg/kg) has a gavage volume of 1ml.
The blank group was subjected to intragastric administration with distilled water, and the intragastric administration volume was 1ml.
TABLE 1 test results of different strains
Figure 409634DEST_PATH_IMAGE001
From the results of the standard components in table 1 above, the standard component values of a single strain are, from large to small: bifidobacterium bifidum Y22, lactobacillus rhamnosus YGG, lactobacillus reuteri K07, lactobacillus fermentum FM-12, lactobacillus acidophilus K43 group, bifidobacterium animalis subspecies lactis Y6 group, pediococcus pentosaceus LN-PT16 group, bifidobacterium longum W68 group, lactobacillus paracasei K9 group, and Bifidobacterium adolescentis Y54 group.
Wherein the standard score of Bifidobacterium bifidum Y22, lactobacillus rhamnosus YGG, lactobacillus reuteri K07 and Lactobacillus fermentum FM-12 is higher and is separated from other groups, which indicates that the above 4 strains have better hair growth promoting effect.
The function of each single strain is analyzed, wherein bifidobacterium bifidum Y22 and lactobacillus fermentum FM-12 have better oil control effect. Lactobacillus rhamnosus YGG and Lactobacillus reuteri K07 have good anti-inflammatory effect. While both oil control and anti-inflammatory have positive effects on hair growth. Of course, the 4 strains also have better gastrointestinal tract conditioning function.
2. Probiotic combination screening assay
4 strains screened out are compounded to verify whether better effects can be obtained. The results are shown in table 2 below.
TABLE 2 recording table of the results of the combined tests of strains
Figure 530037DEST_PATH_IMAGE002
Remarking: the above proportions are counted by the total number of colonies, and are proportioned in equal proportion. The standard score calculation method is the same as that in Table 1.
The results in table 2 above show that the use of oil-controlling bacteria in combination with anti-inflammatory bacteria results in better hair growth than a combination of strains with only a single effect. The 2 strains in the 3 rd group are oil control bacteria, the 2 strains in the 4 th group are anti-inflammatory bacteria, the scores of the bacteria are not much different from those of single strains and are all lower than those of the groups 5-9, and basically no synergistic effect exists.
And the results of groups 5-9 show that the average hair length X (mm), the average hair weight Y (mg) and the average hair follicle number Z (number) are better than the single-strain groups of bifidobacterium bifidum Y22, lactobacillus rhamnosus YGG, lactobacillus reuteri K07 and lactobacillus fermentum FM-12 in the table 1, which indicates that the efficacy is better than that of the single strain by adopting oil-controlling bacteria and anti-inflammatory bacteria for matching. Particularly, in the group 7, the standard score obtained by compounding 4 strains of bacteria is the highest, which shows that the 4 strains of bacteria have the best matching effect.
3. Probiotic combination ratio screening test
4 strains of the screened bacteria are compounded according to different proportions, and the effects of promoting hair growth, fixing hair and preventing alopecia of the strain combination under different proportions are verified. The ratios of the 4 strains and the test results are shown in Table 3 below.
TABLE 3 Strain proportioning Table
Figure 903249DEST_PATH_IMAGE003
Remarking: the above proportions are counted and proportioned according to the total number of colonies.
The composite strain freeze-dried powder of the combinations 1 to 9 is taken, and the gavage test is carried out on the mice according to the test system, and the test results are shown in the following table 4.
TABLE 4 test results of different ratios of strains
Figure 564038DEST_PATH_IMAGE004
From the test data results in table 4 above, the test results of hair-fixing and anti-hair loss are as follows: the compound strain combination is 9, the compound strain combination is 7, the compound strain combination is 8, the compound strain combination is 1, the compound strain combination is 2, the compound strain combination is 3, the compound strain combination is 4, the compound strain combination is 5, and the compound strain combination is 6.
By combining the results of Table 1, the compound strain can better promote hair growth, fix hair and prevent alopecia. And when the 4 strains are uniformly matched, the effect is better. When the proportion of one strain in the formula is smaller, the score value is reduced. Particularly, when the proportion of strains with the same functions is small, the score value is reduced more obviously.
The results in table 4 show that different strains, especially different functional strains, can exert a synergistic effect. In the combination 1-9, the weight ratio of the bifidobacterium bifidum Y22 to the lactobacillus fermentum FM-12 to the lactobacillus rhamnosus YGG to the lactobacillus reuteri K07 is 1 (0.125-8) to (0.125-8). Particularly, the effect is better when the weight ratio of the bifidobacterium bifidum Y22 to the lactobacillus fermentum FM-12 to the lactobacillus rhamnosus YGG to the lactobacillus reuteri K07 is 1 (0.125-1.5) to (0.5-2) to (0.125-1.75) (the combination is 1-2 and 7-9). And bifidobacterium bifidum Y22: lactobacillus rhamnosus YGG: lactobacillus reuteri K07: lactobacillus fermentum FM-12=2.5:2.5:2.5:2.5, namely 4 strains, the effect is best.
And (2) verifying a system II: application extension
According to the functions and research of the traditional Chinese medicines, part of the traditional Chinese medicines also have good effects of promoting hair growth, fixing hair, preventing alopecia and blackening hair. Selecting a traditional Chinese medicine composition with better nutrients required by fermentation, adding glucose as a nutrient substrate, performing combined fermentation by using the above 4 strains of bacteria, filtering fermentation liquor (rough filtering, retaining viable bacteria), freeze-drying to obtain the probiotic fermented traditional Chinese medicine composition, and verifying the effect of promoting hair growth.
Meanwhile, in order to verify the effect of the 4 strains of bacteria in external application, two probiotic samples are prepared for verification, and the sample 1 is probiotic freeze-dried powder added with purified water to obtain probiotic suspension. Sample 2 is a lysate obtained by lysing sample 1.

The specific results are shown in Table 5 below.
TABLE 5 mouse test results of applied samples are recorded in Table
Figure 581672DEST_PATH_IMAGE006
Remarking: the standard score calculation method is the same as that in table 1.
As can be seen from table 5 above:
(1) Group 6, example 4 (suspension) and group 7, example 5 (clear), the composite probiotic lysate had a hair growth promoting effect, and the clear form of group 7 was slightly superior to the suspension form of group 6.
(2) Group 3 the group 1 of the example had a comprehensive score higher than that of the group 4 of the example 2 and also higher than that of the group 8 of the composite probiotics group, which indicates that the effect of promoting hair growth by the combined action of the probiotic fermented traditional Chinese medicines is better than that of the single traditional Chinese medicine or the single composite probiotics.
(3) The combined score of the group 9 composite probiotic + example 5 (clarified) group was higher than that of the group 8 composite probiotic group and that of the group 7 example 5 (clarified) group, indicating that the oral composite probiotic and the topical composite probiotic lysate had a synergistic effect on hair growth. The group 5 composite probiotic suspension is externally applied, the score value is very low and is less than 10, the result difference with a blank control group is very small, and the result shows that the hair growth promotion effect is almost absent when the pure probiotics are externally applied.
The research results show that the probiotics oral administration, the probiotics fermented traditional Chinese medicine product oral administration and the probiotics lysate external coating have the hair growth promoting effect, and compared with the oral administration of distilled water, the hair growth speed is higher, the weight of newly grown hair is heavier, and the number of newly grown hair follicles is more.
The above embodiments are only preferred embodiments of the present invention, and the scope of the present invention should not be limited thereby, and any insubstantial changes and substitutions made by those skilled in the art based on the present invention are intended to be covered by the claims.
