本发明公开了一种基于高频gamma神经振荡的自适应ASSR增强装置,装置包括:提取模块,用于通过重复高频刺激诱发gamma频段ASSR响应,提取中央脑区的EEG信号事件相关谱扰动特征进行预处理,计算事件相关谱扰动能量大小,并进行在线反馈;调制模块,用于根据在线反馈能量值,联合经颅交流电刺激非侵入式神经调控,自适应输出刺激电流强度,实现对gamma频段的ASSR个体化针对性闭环调制,改善或增强特定的神经功能,推广至神经工程学、临床医学等领域,获得可观的社会效益和经济效益。
Description
技术领域
本发明涉及神经工程领域,尤其涉及一种基于高频gamma神经振荡的自适应ASSR增强装置。
背景技术
大脑皮质网络中,gamma神经振荡(30-100Hz)被揭示源于小清蛋白(Parvalbumin,PV)抑制性γ-氨基丁酸能(GABAergic)中间神经元和兴奋性锥体神经元之间的突触相互作用,被认为是协调信息处理的基本机制,与认知功能密切相关[1]。近年来,gamma振荡表征异常在抑郁症、双相情感障碍以及精神分裂症等各种精神疾病中被相继报道。听觉稳态响应(Auditory Steady-state Response,ASSR)是在重复且间隔固定的声音刺激下诱发的脑电反应,由整个听觉神经系统产生,在头皮处采集得到,具有良好的频率特异性。ASSR被认为可反映PV中间神经元维持兴奋/抑制平衡的功能,是可被用来评估gamma神经网络同步振荡和时间信息传递状态的有效手段。多项研究揭示精神分裂症、双相情感障碍、抑郁症等诸类临床精神疾病中均表现出40Hz ASSR响应不足[2-4]。
近两年,调制神经振荡的调控模式越来越受关注,以期通过非侵入式施加特定频率的外源振荡去调制相应频段的大脑内源性神经振荡,从而改善相应的认知功能和临床症状。其中,经颅交流电刺激(transcranial alternating current stimulation,tACS)通过放置在头皮的刺激电极输出特定频率波形调制特定频率的内源性神经振荡,取得一定成效。德国慕尼黑大学团队发现,作用于额区的gamma-tACS能有效缓解抑郁患者的临床症状与认知功能[5]。此外,多项研究证实了tACS可以引起听觉诱发活动的变化[6,7]。但由于个体间的生理差异,个体间gamma ASSR响应强度各不相同。根据个体gamma ASSR响应强度差异,施加不同强度的刺激,更有助于发展基于神经振荡解码的闭环自适应ASSR调制策略。

发明内容
本发明提供了一种基于高频gamma神经振荡的自适应ASSR增强装置,本发明通过重复高频刺激诱发gamma频段ASSR响应,提取中央脑区的EEG信号事件相关谱扰动特征进行在线反馈,联合tACS非侵入式神经调控技术,实现对gamma ASSR个体化闭环调制,在该过程中,tACS电流幅度根据给定时间的大脑状态进行动态调整,以提高tACS的有效性,进而改善或增强某些特定的神经功能,推广至神经工程学、临床医学等领域,获得可观的社会效益和经济效益,详见下文描述:

每部分包含n组刺激任务,每组刺激任务包括:3s的刺激呈现时间和1s的静息时间,静息时间设置为1s,用于保证各组刺激之间的信息不产生交叉影响;
刺激呈现期间为40Hz clicks听觉刺激范式,在线提取相应的EEG信号进行分析,获取刺激电流强度,自适应控制电刺激器输出对应刺激,刺激位点是两侧颞叶T3,T4。

应用64导脑电采集系统及其专用采集软件,数据采集参数设置为采样率1000Hz,0.5~100Hz硬件带通滤波,50Hz工频陷波;
诱发范式控制通过Matlab专用工具箱编写实现,同时向脑电放大器发送同步事件代码以保证数据同步性;
在施加tACS刺激前,记录40Hz click刺激下用户的脑电信号,提取Cz导联下的EEG数据进行0.5-100Hz滤波,根据刺激诱发的时间标签对数据进行分段截取。

本发明提供的技术方案的有益效果是:
1、本发明设计可稳定诱发gamma振荡的听觉稳态诱发范式;对采集到的中央区域的脑电(electroencephalography,EEG)数据进行预处理;计算事件相关谱扰动能量大小;根据在线反馈能量值,联合经颅交流电刺激(transcranial Alternating CurrentStimulation,tACS)技术自适应输出刺激电流强度,实现对gamma频段的ASSR个体化针对性调制,进而改善或增强某些特定的神经功能,为临床精神疾病的脑功能基础研究和神经调控技术发展提供有力帮助;
2、本发明联合高频听觉稳态诱发范式,通过采集中央区域的脑电ASSR响应数据,提取事件相关谱扰动能值,通过在线反馈能量值的大小,自适应输出tACS的刺激电流强度,进而实现对gamma ASSR个体化针对性调制,从而改善相应的认知功能和临床症状,为发展临床精神疾病的神经调控方案提供有力帮助;
3、本发明通过重复高频刺激可稳定有效在线诱发用户gamma频段ASSR响应,通过提取中央脑区Cz单导联下的事件相关谱扰动特征,在线优化tACS刺激电流幅度,实现对gamma ASSR个体化闭环调制;在该过程中,tACS电流幅度根据给定时间的大脑状态进行动态调整,克服了个体间生理差异,更好地提高tACS的有效性,更加个性化的调制gammaASSR,有望为临床各种精神疾病表现出的40Hz ASSR响应不足提供治疗新思路,为安全、非侵入性神经干预提供关键技术支撑。

具体实施方式
为使本发明的目的、技术方案和优点更加清楚,下面对本发明实施方式作进一步地详细描述。
本发明实施例的设计如图1所示,其技术流程包括:通过40Hz click听觉刺激范式对个体ASSR进行诱发,提取中央脑区Cz导联下的gamma ASSR能量值;通过在线反馈能量值的大小,自适应调整tACS的刺激电流强度(1mA,1.5mA,2mA),进而实现对个体的gamma ASSR个性化调制。
听觉刺激范式设计如图2所示,每部分(Session)包含n组刺激任务(可根据用户具体情况合理选择),每组刺激任务包括:3s的刺激呈现时间和1s的静息时间。静息时间设置为1s,是为保证各组刺激之间的信息不产生交叉影响。其中,刺激呈现期间为40Hz clicks听觉刺激范式。随后,在线提取相应的EEG信号进行分析,获取刺激电流强度,自适应控制电刺激器输出对应刺激,刺激位点是两侧颞叶T3,T4,刺激时长可根据用户具体情况进行刺激设置。
EEG采集部分应用64导脑电采集系统及其专用采集软件,数据采集参数设置为采样率1000Hz,0.5~100Hz硬件带通滤波,50Hz工频陷波。系统诱发范式控制通过Matlab专用工具箱(Psychtoolbox)编写实现,同时向脑电放大器发送同步事件代码以保证数据同步性。
EEG数据预处理:在施加tACS刺激前,记录40Hz click刺激下用户的脑电信号,提取Cz导联下的EEG数据进行0.5-100Hz滤波(尽量保证ASSR特征基频和谐波信息不丢失),根据刺激诱发的时间标签对数据进行分段截取。
基于短时傅里叶变换(STFT)的事件相关谱扰动(Event-related SpectralPerturbation,ERSP)提取分析click刺激诱发的ASSR信号,对每个组的能量值进行计算:
其中,f表示频率,t表示时间,k是对应的组刺激任务的标号,Fk(f,t)2表示的是第k组的t时刻下对应的f频率的能量值。根据用户需求,选择特定时间段的窄带频率下的能量值(例如:选择刺激后1–3s的38–42Hz的ASSR响应能量),应用叠加平均方法得到个体平均的40Hz ASSR能量值,如下式所示:
其中,fmin与fmax分别是窄带频率的下限频率与上限频率,tmin与tmax分别是截取时间的下限时刻与上限时刻。根据在线得到的ERSPoutput的大小判定输出神经调控刺激参数A电流强度大小:
其中,T阈表示的是能量阈值,可根据用户人群进行自行设置。通过对ERSPoutput的大小判定,得到A的具体数值,进而控制刺激器的输出电流强度。
例如,当A=1mA时,刺激器会在两侧听觉皮层,即颞叶T3和T4施加1mA的40Hz tACS来调节个体的ASSR响应。tACS调控结束后,可再次通过声音刺激下EEG谱特征分析,评估个体gamma ASSR能量变化,进一步优化下次tACS刺激的神经调控参数,提高tACS调控的有效性,个性化增强用户ASSR响应。
本发明设计了一种基于高频gamma神经振荡的自适应ASSR增强装置。该项发明可在抑郁症、精神分裂症、双相障碍患者以及听力障碍者等神经精神障碍中ASSR受损后的改善与治疗方面产生巨大应用潜力和深远影响,有力推动新一代神经调控技术的发展与应用,有望获得可观的社会效益和经济效益。
本发明实施例对各器件的型号除做特殊说明的以外,其他器件的型号不做限制,只要能完成上述功能的器件均可。

提取模块,用于通过重复高频刺激诱发gamma频段ASSR响应,提取中央脑区的EEG信号事件相关谱扰动特征进行预处理,计算事件相关谱扰动能量大小,并进行在线反馈;
调制模块,用于根据在线反馈能量值,联合经颅交流电刺激非侵入式神经调控,自适应输出刺激电流强度,实现对gamma频段的ASSR个体化针对性闭环调制,改善或增强特定的神经功能;
