Abstract
本发明提供了一种用蔗糖、电气石粉等组成的培养基,利用该培养基培养由面包酵母、嗜酸乳杆菌、植物乳杆菌、光合细菌、放线菌、地衣芽孢杆菌等组合而成的EM菌剂,获得原菌液,可应用于垃圾除臭中,使分解恶臭的有益微生物迅速生长繁殖,同时在各种酶的作用下,迅速分解产生恶臭的物质一氨、硫化氢、不饱和脂肪酸等,从而达到迅速、有效除臭的目的。

本发明涉及微生物发酵培养技术领域,尤其是涉及一种EM菌培养基及其发酵产品在垃圾除臭中的应用。
背景技术:
自然界中存在着许多有用的微生物,被广泛的应用于农业、工业和环保等方面。在众多微生物中,我们将其中一部分对于环境保护、土壤改良、保持宿主健康等具有重要的作用的微生物称为EM(effective microorganism)。
EM菌包括光合细菌、乳酸菌、酵母菌、放线菌等,采用适当的比例和独特的发酵工艺,把经过仔细筛选出来的好气性和嫌气性有益微生物混合培养,形成多种多样的微生物群落,这些菌群在生长过程中产生的有益物质及其分泌物质成为各自或相互生长的基质(食物),正是通过这样一种共生增殖关系,组成了复杂而稳定的微生态系统,形成功能多样的强大而又独特的优势。向垃圾喷洒EM菌剂后,可以分解恶臭的有益微生物迅速生长繁殖,同时在各种酶的作用下,迅速分解产生恶臭的物质-氨、硫化氢、不饱和脂肪酸等,从而达到迅速、有效除臭的目的。EM菌剂可以应用在居民小区的垃圾间、垃圾中转站等场所,也可用于垃圾处理,清除异味不产生二次污染,减少蚊蝇的大量滋生,防止传染性疾病的传播,有利于环境保护,而且处理费用低廉,操作简便。
鉴于EM菌剂在应用上的巨大优势,目前关于EM菌剂的各种技术产品应运而生,液态与固态菌剂的培养基与培养方法也多种多样。
电气石(tourmaline)是一种含硼的带有附加阴离子的环状硅酸盐类矿物,目前已在环保、医疗保健、纺织、涂料等方面开发应用。
目前,尚未有将电气石粉应用于EM菌剂培养促进EM菌剂生长相关报道。
发明内容:
本发明要解决的技术问题是:如何利用电气石粉促进EM菌剂生长并提高EM菌剂在垃圾除臭中的作用。
电气石的主要化学成分为SiO2、FeO、Fe2O3、B2O3、Al2O3、Na2O、MgO、Li2O、MnO2等。电气石晶体结构决定其具有永久带电和永久保持着正极和负极的特性,具有形成电场的特性,电气石存在永久性电极,与水分子作用,将水分子电离形成H+和OH-。H+会形成水合氢离子(H3O+)或形成H2,OH-形成水合羟基离子H3O2 -,H3O2 -散发到空气中即为空气负离子;电气石具有放射4~18μm波长的远红外线的特性,电气石还具有表面活性和吸附性能。由于电气石的这些特性,在EM菌剂培养基中添加一定量的电气石粉可以增加培养基中的溶解氧,促进EM菌剂的生长。而电气石的电场特性和远红外线特性也可以刺激EM菌剂的生长。
本发明使用一种EM菌剂落地发酵罐(授权公告号:CN 214781861 U)进行培养。该落地发酵罐是将多个不同发酵流程的罐体和功能集成到一个罐上,得到可全自动升温和恒温控制、好氧和厌氧交替发酵的发酵罐,实现混料、搅拌加热、好氧发酵、厌氧发酵、物料循环、出料等发酵的整个流程。该落地发酵系统将EM菌剂发酵环节前移到田间地头、养殖场旁,省却了大量工厂生产成本,同时还省去包装、运输、营销等环节的费用,实现生产和应用零距离,应用成本降低80%以上。
本发明的技术方案如下:
EM菌培养基,由以下质量百分数的原料组成:
蔗糖5%-12.5%,蛋白胨0.5%-2%,氯化钠0.1%-0.2%,磷酸氢二钾0.1%-0.2%,硫酸镁0.05%-0.2%,电气石粉0.01%-0.05%,加水补足至100%;所述原料中的粉末皆粉碎至≤200目。
所述EM菌培养方法,按照以下步骤进行:
(1)在落地发酵系统发酵罐中配好用蔗糖、电气石粉等组成的EM菌培养基;
(2)将由面包酵母、嗜酸乳杆菌、植物乳杆菌、光合细菌、放线菌组成的复合EM菌种倒入培养基中,添加完全部原料后,充分搅拌均匀,并用盖子将其盖好密封;
(3)盖上盖子恒温38℃发酵12小时到48小时,再常温发酵3-4天后密闭发酵15-25天,待pH达到3.8以下即可终止发酵,加入已另行发酵好的地衣芽孢杆菌发酵液,此液体即为原菌液产品。
所述EM菌种由面包酵母、嗜酸乳杆菌、植物乳杆菌、光合细菌、放线菌和地衣芽孢杆菌组成。
上述制备得到的原菌液产品用于垃圾除臭的一般方法为:第一天向原始积存垃圾喷洒50倍的原菌液稀释液,菌液与垃圾的比例为1∶1000,如果垃圾场内有排沼气管道,可用原菌液稀释液灌入垃圾底层以增加处理效果;以后按处理进度而定,每天按垃圾数量向新到垃圾喷洒50倍原菌液稀释液。
本发明有益的技术效果在于:
EM菌剂与电气石的协同作用可以促进有益微生物生长,加速垃圾的发酵分解,在短时间内减少堆积垃圾的体积;抑制腐败细菌的生长,改善有机物的分解途径,减少氨和硫化氢的释放量和氨类物质的产生;抑制蚊蝇的蛹、蛆孵化,达到减少蚊蝇滋生的长远效果,减少疾病。

蔗糖5%,蛋白胨2%,氯化钠0.1%,磷酸氢二钾0.2%,硫酸镁0.05%,电气石粉0.05%,加水补足至100%;所述原料中的粉末皆粉碎至≤200目。
上述培养基进行EM菌培养方法,按照以下步骤进行:
(1)在落地发酵系统发酵罐中配好用蔗糖、电气石粉等组成的EM菌培养基;
(2)将由面包酵母、嗜酸乳杆菌、植物乳杆菌、光合细菌、放线菌组成的复合EM菌种倒入培养基中,添加完全部原料后,充分搅拌均匀,并用盖子将其盖好密封;
(3)盖上盖子恒温38℃发酵12小时到48小时,再常温发酵3-4天后密闭发酵15-25天,待pH达到3.8以下即可终止发酵,加入已另行发酵好的地衣芽孢杆菌发酵液,此液体即为原菌液产品。
所述EM菌种由面包酵母、嗜酸乳杆菌、植物乳杆菌、光合细菌、放线菌和地衣芽孢杆菌组成。
实施例2
本实施例提供的EM菌培养基,由以下质量百分数的原料组成:
蔗糖8%,蛋白胨1%,氯化钠0.2%,磷酸氢二钾0.1%,硫酸镁0.05%,电气石粉0.03%,加水补足至100%;
所述原料中的粉末皆粉碎至≤200目。
上述培养基进行EM菌培养方法,按照以下步骤进行:
(1)在落地发酵系统发酵罐中配好用蔗糖、电气石粉等组成的EM菌培养基;
(2)将由面包酵母、嗜酸乳杆菌、植物乳杆菌、光合细菌、放线菌组成的复合EM菌种倒入培养基中,添加完全部原料后,充分搅拌均匀,并用盖子将其盖好密封;
(3)盖上盖子恒温38℃发酵12小时到48小时,再常温发酵3-4天后密闭发酵15-25天,待pH达到3.8以下即可终止发酵,加入已另行发酵好的地衣芽孢杆菌发酵液,此液体即为原菌液产品。

所述EM菌种由面包酵母、嗜酸乳杆菌、植物乳杆菌、光合细菌、放线菌和地衣芽孢杆菌组成。
应用数据:
将制备得到的原菌液,第一天向原始积存垃圾喷洒50倍的原菌液稀释液,菌液与垃圾的比例为1∶1000,以后按处理进度而定,每天按垃圾数量向新到垃圾喷洒50倍原菌液稀释液。经处理后,氨气、硫化氢、臭味减少80%以上。

Claims (5)
1.一种EM菌培养基,其特征在于,由以下质量百分数的原料组成:
蔗糖5%-12.5%,蛋白胨0.5%-2%,氯化钠0.1%-0.2%,磷酸氢二钾0.1%-0.2%,硫酸镁0.05%-0.2%,电气石粉0.01%-0.05%,加水补足至100%;所述原料中的粉末皆粉碎至≤200目;
2.根据权利要求1所述的EM菌培养基,其特征在于,所述EM菌种由面包酵母、嗜酸乳杆菌、植物乳杆菌、光合细菌、放线菌、地衣芽孢杆菌组成;
3.一种EM菌培养基进行培养EM菌种的方法,其特征在于,按照以下步骤进行:
(1)在落地发酵系统发酵罐中配好用蔗糖、电气石粉等组成的EM菌培养基;
(2)将由面包酵母、嗜酸乳杆菌、植物乳杆菌、光合细菌、放线菌组成的复合EM菌种倒入培养基中,添加完全部原料后,充分搅拌均匀,并用盖子将其盖好密封;
(3)盖上盖子恒温38℃发酵12小时到48小时,再常温发酵3-4天后密闭发酵15-25天,待pH达到3.8以下即可终止发酵,加入已另行发酵好的地衣芽孢杆菌发酵液,此液体即为原菌液产品;
4.根据权利要求3所述的一种培养EM菌培养方法,其特征在于:使用一种EM菌剂落地发酵罐进行培养。
5.根据权利要求3所述的一种培养EM菌培养方法,其特征在于:首先分别制备由面包酵母、嗜酸乳杆菌、植物乳杆菌、光合细菌、放线菌组成的复合EM菌种和地衣芽孢杆菌的发酵液,然后将上述发酵液按下述重量配比混合:复合EM菌种发酵液7份,地衣芽孢杆菌发酵液3份。
