Abstract
本发明公开了一种基于聚乙烯亚胺和普鲁士蓝的肿瘤纳米药物及其制备方法和应用。所述纳米药物在血液循环中,由于PEG化诱导的隐形效应而延长其血液循环时间,并且PEI的细胞毒性也被屏蔽,具有良好的生物相容性;其在肿瘤弱酸性微环境中,可脱落PEG外壳,暴露PEI的正电荷,促进肿瘤细胞对纳米粒子的摄取,并引发PEI的细胞毒性,有助于提高光动力治疗。同时,其具有普鲁士蓝纳米颗粒类过氧化氢酶活性,可以将肿瘤细胞中内源性过氧化氢转化为氧气来减轻肿瘤的乏氧,进一步增强肿瘤光动力治疗。本发明的纳米药物通过聚乙烯亚胺细胞毒性和普鲁士蓝乏氧缓解双途径增强肿瘤光动力治疗效率,对乳腺癌细胞具有显著的杀伤作用。

技术领域
本发明涉及纳米药物技术领域,具体地,涉及一种基于聚乙烯亚胺和普鲁士蓝的肿瘤纳米药物及其制备方法和应用。
背景技术
聚乙烯亚胺(PEI)是一种阳离子聚合物。由于PEI具有较高的阳离子电荷,可以与正常细胞和肿瘤细胞中的阴离子组分相互作用,可导致细胞皱缩、细胞质空泡或细胞死亡。虽然PEI具有细胞毒性,但仍作为最典型的阳离子聚合物之一,目前已经被广泛用于基因和药物递送系统。但是目前仅利用PEI作为载体,并尚无合理利用PEI的细胞毒性,避免对正常细胞或组织的破坏,来进行肿瘤的治疗的报道。
光动力疗法(Photodynamic therapy,PDT)是一种联合利用光敏剂、光(特定波长和强度)和氧气,通过光动力反应产生活性氧,通常是具有细胞毒性的单线态氧(1O2)来杀伤肿瘤细胞,达到治疗肿瘤的目的。PDT已经在临床治疗肿瘤中得到广泛应用。PDT的疗效依赖于光敏剂、氧气以及光源三要素的协同配合。然而许多恶性实体瘤固有的缺氧微环境及PDT过程会进一步加剧缺氧情况,这严重限制了PDT治疗效果。
为了克服肿瘤缺氧以提高PDT的疗效,有研究利用阻断缺氧诱导因子1上调来缓解肿瘤缺氧、阻碍肿瘤呼吸以减轻细胞内氧气消耗、运输外源性氧气或在肿瘤细胞原位产生氧气。肿瘤微环境(缺氧、H2O2升高和pH值降低)促进了癌症增殖和转移,因此调控肿瘤微环境的治疗对于消融肿瘤细胞具有重要意义。特别是具有类过氧化氢酶活性的普鲁士蓝纳米颗粒(Prussian blue,PB)已用于将肿瘤细胞中内源性H2O2催化成O2以改善PDT疗效。普鲁士蓝纳米酶的广泛使用归因于普鲁士蓝具有良好的生物安全性,并且普鲁士蓝在临床上作为一种解毒剂,已被用于治疗重金属铯和铊中毒。
近年来随着科学家对肿瘤的研究不断深入以及纳米医药的发展,光动力治疗效率已有所提高。但由于肿瘤的复杂性和患者的差异性,严重影响了PDT疗效的发挥。在肿瘤光动力治疗研究方面,仍然有很大的探索空间。如何提高光动力治疗效率仍是本领域亟需解决的技术问题。可见,高效低毒光敏剂纳米药物的研发具有重要的现实意义和长远的科学意义。
发明内容
本发明的目的是为了克服现有技术的上述不足,提供一种基于聚乙烯亚胺和普鲁士蓝的肿瘤纳米药物及其制备方法和应用。其工作原理如图1所示,本发明利用聚乙烯亚胺PEI既作为载体也作为发挥抑制肿瘤作用的药物,利用PEI的毒性增强光动力治疗的效果,并通过普鲁士蓝PB纳米颗粒作为纳米酶催化肿瘤内源性H2O2产生O2以缓解肿瘤细胞乏氧,实现了双途径增强肿瘤光动力治疗。本发明制备的高效低毒纳米药物PEG-Ce6-PEI@PB,通过聚乙二醇PEG来调节PEI的细胞毒性,且避免对正常细胞或组织的破坏(PEG修饰到PEI上可通过稳定PEI空间构象和减少PEI的正电荷来有效降低PEI细胞毒性)。具体过程:PEI细胞毒性通过醛基化的聚乙二醇来调节,PEI-PEG席夫碱交联响应于血流(pH 7.4)和肿瘤弱酸性条件(pH 6.8)之间的pH变化。在血流(pH 7.4)环境中,席夫碱比较稳定,PEI的细胞毒性被屏蔽,同时PEG化诱导的隐形效应,提高了PEG-Ce6-PEI@PB纳米药物血液循环时间。随后,PEG-Ce6-PEI@PB纳米药物通过增强的渗透和滞留效应(EPR)在肿瘤部位有效积累,利用肿瘤弱酸环境分解席夫碱触发PEG分离,暴露PEI的正电荷,促进肿瘤细胞对纳米药物的摄取,并引发PEI的细胞毒性。因此,PEG-Ce6-PEI@PB纳米药物具有可调的PEI细胞毒性以及改善细胞内吞作用,以增强光动力治疗效应。在肿瘤细胞内部,PEG-Ce6-PEI@PB纳米药物表现出类过氧化氢酶的活性,可催化H2O2生成O2以缓解肿瘤细胞乏氧,进一步增强光动力治疗效应。最后,在激光照射条件下,PEG-Ce6-PEI@PB协载的光敏剂Ce6将胞内O2转化为细胞毒性分子1O2,杀伤细胞,实现了双途径增强肿瘤光动力治疗。结果表明本发明制备得到的响应肿瘤微环境的纳米药物PEG-Ce6-PEI@PB在乳腺癌治疗中具有良好的生物安全性和显著的抑制肿瘤生长特性,并具有纳米药物的高效低毒优势,发挥了光动力治疗的优势。
本发明的可用于双途径增强肿瘤光动力治疗的纳米药物PEG-Ce6-PEI@PB利用普鲁士蓝纳米颗粒PB、聚乙烯亚胺PEI、二氢卟吩e6(光敏剂Ce6)和聚乙二醇PEG制备得到,如图2所示,其具体的制备方法如下:首先利用单前体(K3[Fe(CN)6])合成法制备了普鲁士蓝PB纳米颗粒,然后利用自组装技术将聚乙烯亚胺修饰在普鲁士蓝纳米颗粒表面形成PEI@PB,接着分别利用酰胺化和醛胺缩合反应在PEI@PB表面修饰光敏剂和聚乙二醇,得到一种基于聚乙烯亚胺和普鲁士蓝双途径增强光动力治疗的肿瘤纳米药物PEG-Ce6-PEI@PB。
本发明的第一个目的是提供一种基于聚乙烯亚胺和普鲁士蓝双途径增强肿瘤光动力治疗的纳米药物。
本发明的第二个目的是提供一种基于聚乙烯亚胺和普鲁士蓝的纳米药物的制备方法。
本发明的第三个目的是提供所述的制备方法制备得到的基于聚乙烯亚胺和普鲁士蓝的纳米药物。
本发明的第四个目的是提供所述的基于聚乙烯亚胺和普鲁士蓝的纳米药物在肿瘤治疗中的应用。
为了实现上述目的,本发明是通过以下方案予以实现的:
本发明要求保护一种基于聚乙烯亚胺和普鲁士蓝的纳米药物,所述纳米药物为普鲁士蓝纳米颗粒表面修饰有聚乙烯亚胺,并在表面修饰有二氢卟吩e6和聚乙二醇。
本发明还要求保护一种基于聚乙烯亚胺和普鲁士蓝的纳米药物的制备方法,包括以下步骤:
S1.制备普鲁士蓝PB纳米颗粒;
S2.S1得到的普鲁士蓝PB纳米颗粒和聚乙烯亚胺通过自组装得到PEI@PB纳米颗粒;
S3.S2得到的PEI@PB纳米颗粒表面修饰二氢卟吩e6得到Ce6-PEI@PB纳米颗粒;
S4.S3得到的Ce6-PEI@PB纳米颗粒表面修饰聚乙二醇得到PEG-Ce6-PEI@PB纳米药物。
优选地,步骤S1中,K3[Fe(CN)6]、PVP、HCl和水,充分混合至澄清后,在75~90℃充分反应,固液分离收集沉淀即为普鲁士蓝PB纳米颗粒。
更优选地,步骤S1中,K3[Fe(CN)6]、PVP、HCl和水,充分混合至澄清,再在80℃充分反应18~24小时,固液分离收集沉淀即为普鲁士蓝PB纳米颗粒。
更优选地,反应时间为20小时。
更优选地,步骤S1中,K3[Fe(CN)6]、PVP和质量分数36%~38%HCl的用量为0.3~0.4g:7.5~9g:80~95μL。

与现有技术相比,本发明具有以下有益效果:
本发明的纳米药物PEG-Ce6-PEI@PB可用于双途径增强肿瘤光动力治疗。纳米药物PEG-Ce6-PEI@PB在血流环境中(pH~7.4),由于PEG化诱导的隐形效应而提高其血液循环时间,并且PEI的细胞毒性也被屏蔽,具有良好的生物相容性。PEG-Ce6-PEI@PB纳米药物在肿瘤弱酸性微环境中(pH~6.8),可脱落PEG外壳,暴露PEI的正电荷,促进肿瘤细胞对纳米粒子的摄取,并引发PEI的细胞毒性,有助于提高光动力治疗。同时,PEG-Ce6-PEI@PB纳米药物具有普鲁士蓝纳米颗粒类过氧化氢酶活性,可以将肿瘤细胞中内源性过氧化氢转化为氧气来减轻肿瘤的乏氧,进一步增强肿瘤光动力治疗。本发明制备得到的PEG-Ce6-PEI@PB纳米药物对乳腺癌细胞具有显著的杀伤作用,表明通过调节PEI细胞毒性和普鲁士蓝减轻肿瘤乏氧双途径,显著提高了肿瘤光动力治疗效率,为增强光动力治疗开辟了一条新途径,为高效低毒纳米药物的研发提供了新思路。

具体实施方式
下面结合说明书附图及具体实施例对本发明作出进一步地详细阐述,所述实施例只用于解释本发明,并非用于限定本发明的范围。下述实施例中所使用的试验方法如无特殊说明,均为常规方法;所使用的材料、试剂等,如无特殊说明,为可从商业途径得到的试剂和材料。
1、主要试剂
铁氰化钾(K3[Fe(CN)6],纯度>99.5%),聚乙烯吡咯烷酮(PVP-K30,AR)和光敏剂二氢卟吩e6(Chlorin e6,Ce6)购自阿拉丁试剂有限公司(中国)。聚乙烯亚胺(20000Da)购自西亚化学科技(山东)有限公司。聚乙二醇(2000Da)购自上海甄准生物科技有限公司。CellCounting Kit 8(CCK8)试剂盒购自东仁化学科技(上海)有限公司。
2、仪器
Eppendorf离心机、仪科数显恒温磁力搅拌器、岛津分光光度计、马尔文粒度仪、瑞士帝肯酶标仪、蔡司荧光显微镜、贝克曼库尔特流式细胞仪等。
3、细胞培养
人乳腺癌细胞系(MCF7)用含有10%FBS、链霉素(100μg mL-1)和青霉素(100U mL-1)的DMEM培养基培养,培养环境为37℃的5%CO2细胞培养箱。
实施例1 PEG-Ce6-PEI@PB纳米药物的合成
PEG-Ce6-PEI@PB纳米药物的合成路线如图2所示。
1、普鲁士蓝PB纳米颗粒的制备
将0.367g K3[Fe(CN)6]、8g PVP和90μL HCl(质量分数为36%~38%)混合在100mL去离子水中,在室温下搅拌30分钟得到澄清溶液;接下来,使反应在80℃下进行20小时;离心(11,000转/分,15分钟)后收集PB,然后在去离子水中洗涤,制备得到普鲁士蓝PB纳米颗粒。
2、PEI@PB纳米颗粒的制备
PEI@PB纳米颗粒是由阳离子PEI和带负电荷的PB的静电相互作用形成的:在磁力搅拌作用下将PEI溶液(2mL,20mg mL-1,pH 5)添加到5mL PB(16mg mL-1)分散液中;反应24小时后,通过去离子水透析获得产物PEI@PB纳米颗粒。
3、Ce6-PEI@PB纳米颗粒的制备
光敏剂二氢卟吩e6(Ce6)通过酰胺反应共价连接到PEI@PB:首先将Ce6(11.9mg)溶解在5mL二甲基亚砜中,依次引入EDC(5.75mg)和NHS(3.45mg)活化Ce6的羧基1小时;然后将制备的PEI@PB纳米颗粒(CPB=3.89mg mL-1,12.25mL)添加到混合物中,在室温下搅拌24小时;然后通过磷酸盐缓冲溶液(Phosphate buffer solution,PBS)pH 7.4进行透析以去除游离的EDC、NHS和游离的Ce6,获得Ce6-PEI@PB纳米颗粒。
4、PEG-Ce6-PEI@PB纳米药物的制备
将1mL甲氧基-聚乙二醇-醛基MPEG-CHO(4mg mL-1)加入到5mL Ce6-PEI@PB(CPB=1.6mg mL-1)纳米颗粒溶液中,在室温下搅拌24小时。通过用PBS透析的方式提纯得到PEG-Ce6-PEI@PB纳米药物。
实施例2透射电镜检测纳米颗粒
一、实验方法
将实施例1制备的PB纳米颗粒和PEG-Ce6-PEI@PB纳米药物的溶液悬液滴在覆有碳膜的铜网上,待其晾干。使用JEOL JEM-1400plus透射电子显微镜,在电子加速电压为120kV条件下,观察粒子形态。
二、实验结果
检测结果如图3a所示,结果显示实施例1制备的PB纳米颗粒及PEG-Ce6-PEI@PB纳米药物表现出均匀的立方形态。
实施例3纳米颗粒的粒径检测
一、实验方法
分别取1mL实施例1制备的PB纳米颗粒和PEG-Ce6-PEI@PB纳米药物的分散液进行水合粒径检测。纳米颗粒样本分散于PBS中,使用马尔文纳米分析仪测量检测粒径。
二、实验结果
检测结果如图3b所示,结果显示实施例1制备的PB纳米颗粒及PEG-Ce6-PEI@PB纳米药物平均尺寸均为100nm左右。
实施例4纳米颗粒的Zeta电势检测
一、实验方法
分别取1mL的实施例1制备得到的PB纳米颗粒、PEI@PB纳米颗粒、Ce6-PEI@PB纳米颗粒和PEG-Ce6-PEI@PB纳米药物的分散液进行zeta电势检测。电势测定中,纳米颗粒样本分散于PBS pH 7.4中,使用马尔文纳米分析仪测量。
二、实验结果
实施例1制备的PB纳米颗粒、PEI@PB纳米颗粒、Ce6-PEI@PB纳米颗粒和PEG-Ce6-PEI@PB纳米药物的zeta电位依次为-22.8±3.6、-3.4±1.2、-15.7±1.0、-22.4±2.1mV(图4)。上述纳米材料之间的电荷差异验证了在普鲁十蓝纳米颗粒PB表面成功修饰了阳离子聚乙烯亚胺PEI,光敏剂二氢卟吩e6和聚乙二醇PEG。
实施例5紫外可见吸收光谱检测
一、实验方法
将实施例1制备的纳米药物PEG-Ce6-PEI@PB、普鲁士蓝纳米颗粒PB及光敏剂Ce6充分溶解于PBS中,用紫外可见分光光度计测样品吸收前,将待测样品超声2分钟,以保证溶质充分溶解。紫外可见分光光度计测量的区间为300~900nm。
二、实验结果
实施例1制备的PEG-Ce6-PEI@PB纳米药物的紫外-可见吸收光谱在404和655nm处显示典型的Ce6吸收峰,PB吸收峰在720nm附近(图5)。该结果进一步证明了实施例1制备的PEG-Ce6-PEI@PB纳米药物的成功合成。
实施例6 PEG-Ce6-PEI@PB的类过氧化氢酶活性
一、实验方法
为了验证实施例1制备的PEG-Ce6-PEI@PB纳米药物具有类似过氧化氢酶的特性,能够催化H2O2产生O2。将实施例1制备的PEG-Ce6-PEI@PB纳米药物分散在5mL PBS中,并在混合液中加入H2O2(最终浓度1wt%),以PBS、H2O2或不加H2O2的实施例1制备的PEG-Ce6-PEI@PB纳米药物分散液为对照,用便携式溶解氧仪测量15分钟内各溶液内的氧气含量变化。并拍照记录15分钟时各溶液内的氧气气泡变化。
二、实验结果
利用便携式溶解氧仪评估了实施例1制备的PEG-Ce6-PEI@PB纳米药物的类过氧化氢酶活性,结果如图6所示。在监测混合溶液15分钟时,含有H2O2组的实施例1制备的PEG-Ce6-PEI@PB纳米药物的溶液中的O2浓度显着增加,并且可以清楚地观察到明显的O2气泡,但在其他组中气泡可以忽略不计(图6中的插图),说明实施例1制备的PEG-Ce6-PEI@PB具有普鲁士蓝纳米颗粒PB的类过氧化氢酶活性。
实施例7溶液中PEG-Ce6-PEI@PB的单态线氧1O2产生检测
一、实验方法
荧光探针Singlet oxygen sensor green(SOSG)是单态线氧1O2的荧光探针。SOSG探针在其初始状态发弱蓝色荧光,但在与1O2反应激活时会发出绿色荧光信号(λex=488nm,λem=530nm)。SOSG遇到1O2时其绿色荧光会增强。实施例1制备的PEG-Ce6-PEI@PB纳米药物(Ce6:1μM)与SOSG(5μM)混匀后,在660nm激光(34mW cm-2)辐照时,检测不同时间点SOSG的荧光强度。
二、实验结果
结果如图7所示,实施例1制备的PEG-Ce6-PEI@PB纳米药物在激光照射下会将基态的O2转化为具有细胞毒性的单态线氧1O2,随着激光照射时间的延长,累积的SOSG荧光强度逐渐升高。PEG-Ce6-PEI@PB纳米药物与H2O2的1O2产率高于没有H2O2的产率,这是由于PB催化H2O2提供了额外的O2。结果表明,具有类过氧化氢酶活性的实施例1制备的PEG-Ce6-PEI@PB纳米药物可以催化H2O2产生O2,不仅可以改善肿瘤缺氧,还可以增强1O2的产生,从而提高PDT效率。
实施例8肿瘤弱酸微环境引发PEG脱落以增强细胞对PEG-Ce6-PEI@PB药物的摄取
一、实验方法
根据细胞内Ce6荧光荧光强度来研究肿瘤弱酸微环境引发的实施例1制备的PEG-Ce6-PEI@PB纳米药物表面PEG脱离特性。将MCF7细胞传代到共聚焦培养皿中,过夜后将细胞与实施例1制备的PEG-Ce6-PEI@PB纳米药物或Ce6-PEI@PB纳米颗粒(具有等效浓度的Ce6)在不同pH孵育6小时。最后使用荧光显微镜及流式细胞仪分析细胞内的Ce6荧光强度。
二、实验结果
实施例1制备的PEG-Ce6-PEI@PB纳米药物的工作原理及制备过程如图1和图2所示,简述如下:首先利用单前体合成法制备了普鲁士蓝纳米颗粒PB,然后利用自组装技术将聚乙烯亚胺修饰在普鲁士蓝纳米颗粒表面形成PEI@PB,接着分别利用酰胺化和醛胺缩合反应在PEI@PB表面修饰光敏剂和聚乙二醇。其中,醛基化聚乙二醇是通过醛胺缩合反应(席夫碱)修饰在PEI@PB表面。通常情况下,实体瘤微环境的酸性(pH 6.8)比血流和正常组织(pH7.4)的酸性更强。PEI-PEG席夫碱交联响应于血流(pH 7.4)和肿瘤弱酸性条件(pH 6.8)之间的pH变化。在血流(pH 7.4)环境中,席夫碱比较稳定,PEG-Ce6-PEI@PB处于稳定状态。同时PEG化诱导的隐形效应,提高了实施例1制备的PEG-Ce6-PEI@PB血液循环时间。随后,实施例1制备的PEG-Ce6-PEI@PB通过增强的渗透和滞留效应(EPR)在肿瘤部位有效积累,利用肿瘤弱酸环境分解席夫碱触发PEG分离,暴露PEI的正电荷,促进肿瘤细胞对纳米粒子的摄取。
为了验证这一点,将实施例1制备的PEG-Ce6-PEI@PB纳米药物在pH 7.4和pH 6.8下分别施用于MCF7细胞以模拟生理条件和肿瘤弱酸微环境。通过荧光显微镜监测来自实施例1制备的PEG-Ce6-PEI@PB纳米药物和Ce6-PEI@PB纳米颗粒的Ce6的红色荧光强度(图8a)。进入细胞的PEG-Ce6-PEI@PB(pH 7.4)、PEG-Ce6-PEI@PB(pH 6.8)和Ce6-PEI@PB(pH 7.4)的量依次增加。进一步,通过流式细胞仪的定量分析(图8b),表明当pH值从7.4降低到6.8时,实施例1制备的PEG-Ce6-PEI@PB纳米药物的荧光强度增加了约20%。此外,与实施例1制备的PEG-Ce6-PEI@PB纳米药物在pH 7.4时相比,Ce6-PEI@PB纳米颗粒的细胞内荧光强度增加了约30%,结果显示肿瘤弱酸微环境可以引发实施例1制备的PEG-Ce6-PEI@PB纳米颗粒表面PEG脱落,并且有助于肿瘤细胞对纳米药物实施例1制备的PEG-Ce6-PEI@PB纳米药物的摄取。
实施例9 PEG-Ce6-PEI@PB催化细胞内内源性H2O2产生O2
一、实验方法
MCF7细胞与400μL[(Ru(dpp)3)]Cl2(浓度:10μg mL-1)一起孵育12小时,用PBS洗涤细胞3次,再将细胞与不同浓度的实施例1制备的PEG-Ce6-PEI@PB纳米药物再培养6小时。PBS清洗细胞后补加新鲜的培养基,通过荧光显微镜观察细胞。此外,还进行了流式细胞术分析以评估细胞内氧浓度。
二、实验结果
为了检测实施例1制备的PEG-Ce6-PEI@PB纳米药物作为纳米酶催化细胞内内源性H2O2产生O2,利用荧光探针[Ru(dpp)3]Cl2作为O2指示剂来识别细胞内O2含量,其荧光在富氧条件下被猝灭。荧光显微镜(图9a)和流式细胞仪(图9b)显示,[Ru(dpp)3]Cl2荧光强度的降低与实施例1制备的PEG-Ce6-PEI@PB纳米药物的浓度相关,细胞孵育的实施例1制备的PEG-Ce6-PEI@PB纳米药物浓度越高,氧气荧光探针[Ru(dpp)3]Cl2的荧光越弱。这些结果进一步证实了实施例1制备的PEG-Ce6-PEI@PB纳米药物的类过氧化氢酶活性,可以催化MCF7细胞内内源性H2O2产生O2。
实施例10肿瘤细胞内单线态氧1O2检测
一、实验方法
为了分析细胞内1O2含量,将MCF7细胞接种于35mm细胞培养皿中。MCF7细胞分别与pH 7.4、pH 6.8条件下的实施例1制备的PEG-Ce6-PEI@PB纳米药物(其中以Ce6计浓度为3μgmL-1)一起孵育6小时。随后用无血清培养基冲洗细胞,并在37℃下用SOSG(5μM)或NAC(5mM)预培养1小时。经660nm激光(34mW cm-2,5min)照射后,使用荧光显微镜拍照记录。
二、实验结果
利用1O2荧光探针SOSG评估激光照射后实施例1制备的PEG-Ce6-PEI@PB纳米药物处理的MCF7细胞中单线态氧1O2的产生,如图10所示。在没有660nm激光照射的组中,SOSG没有明显的绿色荧光。实施例1制备的PEG-Ce6-PEI@PB纳米药物的SOSG的绿色荧光强度在激光照射后增加,说明PB和PEI可以作为Ce6的合适纳米载体,而不会对其产生的1O2造成不利影响。在pH 6.8时,1O2的产生高于中性条件(pH 7.4),这归因于在弱酸性条件下细胞对实施例1制备的PEG-Ce6-PEI@PB的摄取增加。并且PEG-Ce6-PEI@PB纳米药物在pH 6.8下产生1O2可以被活性氧清除剂NAC阻断,进一步证实PEG-Ce6-PEI@PB纳米药物在MCF7细胞中产生了1O2。
实施例11 PEG-Ce6-PEI@PB纳米药物的血液相容性测定
一、实验方法
将含有2%新鲜小鼠红细胞的实施例1制备的PEG-Ce6-PEI@PB纳米药物溶液置于1.5mL的ep管中。水和PBS分别作为阳性和阴性对照。3小时后,将混合物离心(3000转/分,5分钟)。然后对样品拍照并通过酶标仪读取上清液在540nm处的吸光度。使用以下公式评估血液相容性:溶血率(%)=(OD样品–OD阴性对照)/(OD阳性对照–OD阴性对照)×100。
二、实验结果
实施例1制备的纳米药物PEG-Ce6-PEI@PB的溶血性实验结果,如图11所示。实施例1制备的PEG-Ce6-PEI@PB纳米药物即使浓度高达200μg mL-1其溶血率仍低于5%,表明实施例1制备的PEG-Ce6-PEI@PB纳米药物具有良好的血液相容性。
实施例12 PEG-Ce6-PEI@PB纳米药物的细胞毒性
一、实验方法
将MCF7细胞种植在96孔板中24小时。每孔加入100μL具有不同浓度实施例1制备的PEI@PB纳米颗粒、Ce6@PB纳米颗粒和Ce6-PEI@PB纳米颗粒的新鲜培养基。12小时后用新鲜的DMEM替换细胞后,用34mW cm-2的660nm激光照射12分钟。再孵育24小时后,将细胞转移到含有CCK8的新鲜培养基中1小时,然后在450nm处记录吸光度以确定相对肿瘤细胞活力。
二、实验结果
利用CCK8法研究实施例1制备的PEG-Ce6-PEI@PB纳米药物的抗肿瘤效率。用(1)实施例1制备的PEI@PB纳米颗粒(仅PEI细胞毒性)、(2)实施例1制备的Ce6@PB纳米颗粒加激光(仅乏氧缓解单途径增强PDT)和(3)实施例1制备的Ce6-PEI@PB纳米颗粒加激光(PEI细胞毒性和乏氧缓解双途径增强PDT)处理MCF7细胞。三个实验组的细胞毒性随着药物浓度的增加而增加(图12a)。与单独的PEI细胞毒性或乏氧缓解单途径增强PDT相比,Ce6-PEI@PB纳米药物在660nm激光照射下表现出较低的细胞活力,表明PEI细胞毒性和乏氧缓解双途径显著提高了肿瘤光动力治疗效率。
实施例13 PEG-Ce6-PEI@PB纳米药物的细胞凋亡和坏死试验
一、实验方法
将MCF7细胞接种在六孔板中,细胞密度为每孔1.5×105个细胞。细胞接种过夜后,用实施例1制备的PEI@PB、Ce6@PB、和Ce6-PEI@PB纳米粒子处理细胞12小时。随后,将细胞用PBS洗涤并暴露于激光组的660nm激光(34mW cm-2,12分钟)。24小时后,收集所有细胞,重悬于膜联蛋白结合缓冲液中,并根据说明书利用Annexin V-FITC/PI染色。最后,通过流式细胞术测定细胞凋亡和坏死的百分比。
二、实验结果
利用Annexin V-FITC/PI凋亡和坏死试剂盒,并借助流式细胞术来定量细胞凋亡和坏死的比例(图12b)。其与CCK8检测结果一致,与其他处理组相比,激光照射的实施例1制备的Ce6-PEI@PB纳米粒子处理的细胞活性最低,早期凋亡和晚期凋亡/坏死率最高。实验表明PEG-Ce6-PEI@PB纳米粒子对乳腺癌细胞具有显著的杀伤作用,进一步验证了PEI细胞毒性和普鲁士蓝乏氧缓解双途径显著提高了肿瘤光动力治疗效率。
综上,本研究构建了一种新型基于聚乙烯亚胺和普鲁士蓝双途径增强肿瘤光动力治疗的纳米药物PEG-Ce6-PEI@PB。该纳米药物PEG-Ce6-PEI@PB不仅可以在弱酸性条件下脱落PEG,有助于肿瘤细胞内吞,提高了肿瘤细胞内纳米药物的含量,并引发PEI细胞毒性,还可以催化肿瘤细胞中内源性过氧化氢转化为氧气缓解肿瘤乏氧,提高了肿瘤光动力学治疗效率。同时,PEG-Ce6-PEI@PB纳米药物表现出良好的血液相容性。更重要的是,PEG-Ce6-PEI@PB对乳腺癌细胞具有显著的杀伤作用,验证了PEI细胞毒性和普鲁士蓝乏氧缓解双途径显著提高了肿瘤光动力治疗效率,发挥了光动力治疗的优势,为增强光动力治疗开辟了一条新途径。
最后所应当说明的是,以上实施例仅用以说明本发明的技术方案而非对本发明保护范围的限制,对于本领域的普通技术人员来说,在上述说明及思路的基础上还可以做出其它不同形式的变化或变动,这里无需也无法对所有的实施方式予以穷举。凡在本发明的精神和原则之内所作的任何修改、等同替换和改进等,均应包含在本发明权利要求的保护范围之内。
Claims (3)
1.一种基于聚乙烯亚胺和普鲁士蓝的纳米药物的制备方法,其特征在于,包括以下步骤:
S1 . 将K3[Fe(CN)6]、PVP、HCl和水充分混合至澄清,在75~90℃充分反应,固液分离收集沉淀制得普鲁士蓝PB纳米颗粒;
S2 . S1得到的普鲁士蓝PB纳米颗粒与聚乙烯亚胺按照质量比2:1在溶液中混合,充分反应后纯化即得PEI@PB纳米颗粒;
S3 .在溶液中二氢卟吩e6活化羧基之后与S2得到的PEI@PB纳米颗粒按照质量比11.9:47.7充分混合,充分反应进行共价连接后,纯化即得Ce6-PEI@PB纳米颗粒;
S4 .在溶液中甲氧基-聚乙二醇-醛基与S3得到的Ce6-PEI@PB纳米颗粒按照质量比1:2充分混合,充分反应进行醛胺缩合反应后,纯化即得PEG-Ce6-PEI@PB纳米药物。
2.权利要求1所述的制备方法制备得到的基于聚乙烯亚胺和普鲁士蓝的纳米药物。
3.权利要求2所述的基于聚乙烯亚胺和普鲁士蓝的纳米药物在制备治疗肿瘤药物中的应用,其特征在于,肿瘤药物为基于聚乙烯亚胺和普鲁士蓝双途径增强光动力治疗的肿瘤纳米药物,所述肿瘤为乳腺癌。
