Abstract
本发明涉及了纳米二氧化钛在制备预防和/或治疗重金属包括镉造成的肾损伤的制剂中的应用。本发明发现了纳米二氧化钛对镉诱导的大鼠肾脏毒性有明显的拮抗作用,表现为纳米二氧化钛可显著降低镉经口暴露引起的尿液中肾损伤标志物的增加,包括显著降低N‑乙酰‑β‑D氨基葡萄糖苷酶B型同工酶、尿素和肌酐等;减轻镉暴露引起的急性肾小管损伤;机制可能与联合暴露减少镉的吸收和分布、增加镉的排泄,降低镉的生物利用度有关。因此,可将纳米二氧化钛应用于药物或者食品或者保健品,用于预防和/或治疗肾损伤,特别是用于预防和/或治疗重金属包括镉造成的肾损伤。

技术领域
本发明属于生物医药领域,具体涉及纳米二氧化钛在防治肾损伤中的应用。
背景技术
纳米二氧化钛(Titanium dioxide nanoparticles,nTiO2)是各种工业中生产最广泛的纳米材料之一,据估算全球每年的nTiO2消费量达1000万吨,主要应用于化妆品和涂料。nTiO2根据晶型可分为锐钛矿型,金红石型及板钛矿型,其中锐钛矿型及金红石型较为常见;又可根据疏水性分为亲水型和疏水型。nTiO2良好的抗菌效果和阻隔能力使其复合包装材料得以广泛的应用;因其对紫外线具有吸收作用,nTiO2用于防晒霜及喷雾的生产;而nTiO2良好的吸附特性及光催化特性在环境污染治理中为废气的吸收及废水的处理提供了有效的方法。二氧化钛可作为着色剂(E171)添加到糖果涂层、口香糖、冰淇淋、牙膏等食品和日用品中,而E171中的二氧化钛颗粒一部分为纳米级,按数量计占5%~50%,按质量计最多占30%,因而nTiO2也可出现在食品中。关于nTiO2的毒性研究很多,例如有研究认为nTiO2可通过血脑屏障,造成脑损伤及神经毒性,但也有部分研究表明nTiO2无显著毒性,总之,目前尚未得到毒性的明确结论。此外,nTiO2在体内吸收、分布研究也仍未得到一致的结果。各种研究结果的差异可能与nTiO2颗粒在晶型、粒径、疏水性、介质及前处理方式上的不同可能有关。
镉(Cd)是银白色有光泽的金属,广泛用于合金制造、电镀等工业领域,采矿活动及工业废水中的Cd造成环境水及土壤污染后,由于大部分Cd化合物有较高的溶解度,极易被植物吸收,通过环境及食物链在无脊椎动物和脊椎动物体内蓄积。Cd污染在我国乃至全世界一直是一个突出的环境问题,Cd暴露及其毒性也一直深受人们关注。肾脏是长期Cd暴露的关键靶器官。长期经口暴露于Cd,可引起肾脏的组织病理学损伤,首先受到损伤的是肾皮质的近端小管,表现为近端小管上皮细胞损伤、间质纤维化、肾小管细胞再生受限。肾脏的主要功能是代谢生成尿液,尿液中的尿蛋白、尿素、肌酐等是临床上检测肾病的常用指标。此外,存在于肾小管上皮细胞刷状缘的N-乙酰-β-D氨基葡萄糖苷酶(NAG)等小分子蛋白酶是Cd早期肾损伤的敏感标志物。同时,Cd被国际癌症研究机构(IARC)列为I型致癌物,吸入致肺癌。也有研究表明Cd还可致肾癌、肝癌、前列腺癌,但证据尚不充分。PAN等人报道,随年龄增长引发的前列腺癌和乳腺癌的发病率与慢性低剂量Cd接触相关。现如今,尽管Cd排放问题在法规监管后有了很大的改善,但由于Cd无法被生物降解,Cd污染仍是一个长期的威胁。
目前文献报道的nTiO2和Cd联合短期毒性研究主要集中在体外试验和以水生生物为模型的环境毒性试验。这些研究的结论存在争议,其中既有协同作用,也有拮抗作用。
针对目前食品中的nTiO2和仍然突出的重金属Cd高风险污染问题,尽管目前关于nTiO2与重金属(包括Cd)的联合毒性研究表明,nTiO2可以通过吸附作用改变重金属在机体中的有效浓度,从而改变重金属毒性,然而,关于nTiO2对Cd诱导的大鼠肾毒性的影响仍是未知的。
发明内容
本发明的目的是提供一种nTiO2在医药或食品中的新应用。
包括以下技术方案。
nTiO2在制备预防和/或治疗肾损伤的制剂中的应用。
nTiO2在制备预防和/或治疗重金属造成的肾损伤的制剂中的应用。
一种预防和/或治疗重金属造成的肾损伤的制剂,其活性成分包括有nTiO2。
在其中一些实施例中,所述重金属为Cd。
在其中一些实施例中,所述nTiO2为食品级的亲水型nTiO2。
在其中一些实施例中,所述nTiO2的晶型为锐钛矿型。
在其中一些实施例中,所述nTiO2的平均粒径为40.9±9.6nm。
在其中一些实施例中,所述预防和/或治疗肾损伤包括显著降低N-乙酰-β-D氨基葡萄糖苷酶B型同工酶、尿素和肌酐等。

本发明的发明人通过大量实验,发现了nTiO2对Cd的肾脏毒性有拮抗作用:Cd组大鼠从第一个月开始尿中N-乙酰-β-D氨基葡萄糖苷酶等肾损伤标志物显著升高,并持续到实验结束。与Cd组相比,nTiO2+Cd联合组大鼠肾损伤标志物N-乙酰-β-D氨基葡萄糖苷酶B型同工酶、尿素和肌酐等明显下降,第一个月析因分析显示nTiO2与Cd联合暴露对尿素和肌酐有显著的拮抗作用。在对大鼠肾脏组织病理学及超微结构的影响的实验中:Cd组大鼠出现急性肾小管损伤,光镜下表现为上皮细胞肿胀、胞浆脱落、管型及炎症细胞浸润。透射电镜(TEM)超微结构观察显示,肾脏近曲小管上皮细胞线粒体空泡变性、微绒毛消失;而nTiO2+Cd联合组大鼠肾脏的病理改变较Cd组减轻;机制可能与联合暴露减少大鼠体内Cd的吸收和分布,增加Cd的排泄,降低Cd的生物利用度有关。
因此,可将nTiO2应用于药物或者食品或者保健品中,用于防治肾损伤,特别是用于防治重金属包括镉造成的肾损伤。

具体实施方式
本发明下列实施例未注明具体条件的实验方法,通常按照常规条件,或按照制造厂商所建议的条件。实施例中所用到的各种常用化学试剂,均为市售产品。
除非另有定义,本发明所使用的所有的技术和科学术语与属于本发明的技术领域的技术人员通常理解的含义相同。本发明的说明书中所使用的术语只是为了描述具体的实施例的目的,不用于限制本发明。
以下实验中,所述的CdCl2和nTiO2来源如下:
CdCl2:CAS号:10108-64-2;美国Sigma;白色粉末。
nTiO2:CAS号:13463-67-7;上海阿拉丁;晶型:锐钛矿;粒径:40nm;性状:白色粉末。所涉及到的实验试剂如下表:
为了便于理解本发明,下面将对本发明进行更全面的描述。
实施例1
1纳米二氧化钛表征
1.1外貌形态及粒径表征
采用扫描电子显微镜(SEM)和透射电子显微镜(TEM)对nTiO2的表面形态及粒径进行检测。
①蘸取少量nTiO2粉末平铺一层至碳导电胶布,置于电镜台上,进行喷金处理后,用扫描电子显微镜(SEM)对纳米颗粒的表面形态进行扫描拍照,选取图中150个粒子,用Image J(1.52v)软件对其粒径进行测量。
②将少量nTiO2粉末溶于无水乙醇,涡旋30s,利用超声仪超声20min并涡旋3min使其成为澄清透明的悬浮液,将悬浮液滴在铜网上,用透射电子显微镜(TEM)对样品的微观形貌进行观察。
参见图1:nTiO2的SEM(图1a)及TEM(图1b)图像如图所示。由SEM图像可看出,nTiO2的形状近似球形。经Image J软件测量得到的平均粒径为40.9±9.6nm。由TEM图像可看出,nTiO2在超纯水中较易聚集成团,难以分散成单个粒子。
1.2流体动力学直径及zeta电位分析
将少量nTiO2粉末溶于超纯水中制成澄清透明悬浮液,涡旋30s,超声20min后涡旋3min,取适量至上样杯后,室温下静置15min,用动态光散射(DLS)检测流体动力学直径及进行zeta电位分析。
参见图2:nTiO2在超纯水中的流体动力学直径及zeta电位检测结果如图2所示。流体粒径三次检测的平均值为326.6nm,多分散系数(Polymer dispersity index,PDI)为0.595,提示nTiO2颗粒在超纯水中可能存在有聚集成团的情况。检测的Zeta电位为-14.7mV,证明室温下超纯水的pH值大于nTiO2的等电点,当其分散在超纯水中时带负电,利于对阳离子的吸附。
1.3元素组成分析
取少量nTiO2粉末粘在碳导电胶布上,用能量色散X射线光谱仪(EDX)进行元素组成检测。结果见表1.3。

1.4晶体结构表征
取少量nTiO2粉末,研磨过300目筛,装入铝制样品盘,压平,放入X射线衍射仪(XRD)检测晶体结构。测试条件:电压40kv电流40mA扫描角度:3-90°扫描速度:6°/min,入射狭缝:1/2°,一维探测器D/teX Ultra,步长0.02°,扫描模式:连续扫描。
图3中可见样品呈现出良好结晶性,各衍射峰尖锐分明。图谱在25.2858°,36.931°,37.782°,38.527°,48.017°,53.905°,55.025°,62.809°处的特征峰分别对应锐钛矿的晶面,而在27.42°对应金红石的晶面,除这两种TiO2相,无其他杂相出现。
1.5比表面积测定
称取一定量的nTiO2置于U型测试管中,在单点模式下,以300℃的温度预处理15分钟后,以N2为吸附质,He为载气,氦氮比7:3,测试程序下样品管放入液氮杯中保温,进行N2吸附测试,吸附温度为77.30K,气体浓度为30.00%。
通过BET法测得样品的比表面积为49.2059m2/g。

40只SPF级5周龄雌性SD种大鼠,体重80~100g,购于广东省医学实验动物中心,实验动物使用许可证号:SYXK(粤)2018-0002。涉及动物的程序已经动物实验伦理委员会批准。实验室动物的管理参照《良好实验室规范》(Good Laboratory Practice,GLP)的要求。大鼠饲养在温度为20-26℃和相对湿度为40-70%的标准SPF环境中,动物2只/笼,合笼饲养,采用12h:12h昼夜间断照明。动物分组及暴露:大鼠检疫5天,期间每日检查动物1次,记录动物的体形、被毛、皮肤、精神、粪便、眼睛、口鼻、呼吸等体征,如发现不健康的动物立即剔除,选用健康大鼠进行实验。检疫最后一天称重,根据动物体重分层,随机分为4组:对照组、nTiO2组(灌胃给予100mg/kg BW nTiO2混悬液)、Cd组(饮水给予50mg Cd/L CdCl2)和nTiO2+Cd组(饮水给予50mg Cd/L CdCl2,灌胃给予100mg/kg BW nTiO2混悬液),每组10只动物,每周给样7次,连续90天。在灌胃前将nTiO2混悬液涡旋30s,利用超声仪超声20min并涡旋3min。基于2016年EFSA的评估,通过饮食暴露于nTiO2的最大暴露量为1.04mg/kg BW,我们以100为安全系数设定了100mg/kg BW nTiO2的剂量;根据文献报道,我们选择了暴露12周引起明显的肾毒性50mg Cd/L CdCl2的剂量。动物自由摄食和饮水。经测定,纯净饮用水以及nTiO2混悬液中使用的超纯水均不含Cd,所用的灭菌饲料中Cd的浓度为0.095μg/g。
表2.1动物分组
为进行实验的质量控制,防止交叉污染,本实验对实验区域及实验器材进行了染Cd与非染Cd的严谨区分。为保护环境,本实验对实验过程中产生的Cd废水进行无害化处理。
3标本采集与指标
3.1一般临床观察:每天观察并记录1次,观察大鼠精神状态、外观体征、行为活动、腺体分泌、呼吸、粪便及死亡等情况及其他毒性症状。
各试验组大鼠一般生理体征、行为表现、外观、皮毛、黏膜、分泌物、大小便等均未见异常,试验期间动物无死亡。
3.2体重:前6周每周记录2次,之后每周测定1次;第90天记录所有动物体重,解剖当日记录空腹体重。
各剂量组大鼠体重与对照组比较,差异均无统计学意义(P>0.05);各剂量组总增重与对照组比较,差异均无统计学意义(P>0.05),见图4。
3.3摄食量:每周测定1次,根据增重及摄食量计算食物利用率。
各组摄食量如图5所示:
(1)与对照组相比,nTiO2组摄食量未见统计学差异;
(2)与对照组相比,Cd组第一周、第八周摄食量显著降低;
(3)与对照组相比,nTiO2+Cd组第一周、第七周、第十周摄食量显著降低;与Cd组相比第七周摄食量显著降低。
(4)各剂量组试验期间总摄食量与对照组比较,差异均无统计学意义。
3.4食物利用率
各组食物利用率如图6所示(其中第五周、第九周的食物利用率因尿液采集24h禁食而下降):
(1)与对照组相比,nTiO2组食物利用率仅在第九周显著升高;
(2)与对照组相比,Cd组食物利用率在第二周、第六周、第十三周显著增加,第八周显著降低;
(3)nTiO2+Cd组与对照组相比,第十三周食物利用率显著增加;与Cd组相比,第二周食物利用率显著降低,而第八周显著增加。
(4)各剂量组试验期间总食物利用率与对照组比较,差异均无统计学意义。

综上,剂量组在个别时间点与对照组相比摄食量与食物利用率有显著差异,但这些差异未见时间连续性,同时各组总摄食量及总食物利用率未见统计学差异,因此nTiO2与Cd单独或联合给样对大鼠的摄食量和食物利用率无明显影响。
3.5饮水量:每周测定2次。根据试验期间饮水量及Cd浓度计算Cd实际摄入量。各组饮水量及Cd实际摄入量如图7所示:
(1)nTiO2组的饮水量与对照组相比仅在第三、第八周有显著差异(P<0.05);在整个实验过程的总饮水量无显著差异。
(2)Cd组与对照组比较,在整个实验过程的饮水量均显著降低,差异有统计学意义(P<0.01);总饮水量与对照组相比显著降低(P<0.01)。
(3)nTiO2+Cd组与对照组比较,在整个实验过程的饮水量显著降低,差异有统计学意义(P<0.01);与Cd组相比,部分时间点有统计学差异,但总饮水量无显著差异。
(4)根据饮水量计算Cd实际摄入量,Cd组和nTiO2+Cd组在实验期间的平均每日Cd实际摄入量分别为0.996±0.043mg/rat(3.75-4.45mg/kg BW)和0.941±0.097mg/rat(3.17-4.73mg/kg BW)。
4实验方法
4.1尿液采集与检测
(1)尿液采集
尿液采集瓶提前用硝酸(优纯级,五倍稀释)浸泡24h后,用去离子水冲洗3次,高压灭菌,晾干后使用,以去除原有的Cd离子。在给予受试物第一个月、第二个月、第三个月(终期)动物进代谢笼采集尿液。在上午完成灌胃后,将动物装入代谢笼,禁食不禁水,仍给予受试物(含Cd饮水),收集24h尿液。收集瓶置于冰袋上,并在下午置换一轮冰袋,以低温保存尿液。第一、第二个月完成尿液采集后动物回笼继续进行实验,第三个月完成尿液采集后动物进行麻醉解剖。
(2)尿指标检测
尿液采集完成后,立即检测尿常规与尿生化。
尿常规指标(干化学法):相对密度(SG)、pH、潜血(BLD)、尿胆原(UBG)、胆红素(BIL)、酮体(KET)、尿蛋白(PRO)、微量白蛋白(MAlb)、尿糖(GLU)、白细胞(LEU)、亚硝酸盐(NIT)。
尿生化指标:总蛋白(TP)、MAlb、尿碱性磷酸酶(ALP)、尿肌酐(CRE)、尿素(Urea)、尿酸(UA)、总N-乙酰-β-D氨基葡萄糖苷酶(NAG-T)及其B型同工酶NAG-B含量。NAG-B的检测采用加热法,该法的原理为NAG-B有较好的热稳定性,利用加热使NAG的A型同工酶失活后,检测剩余的NAG活性,即为NAG-B的活性。取150μL尿液于1.5ml EP管中,用恒温金属浴55℃加热15min后,再进行NAG-B含量检测。
4.2血液采集与检测
(1)麻醉和采血
待解剖大鼠在解剖前禁食不禁水,解剖当日称量空腹体重。使用2.25%的戊巴比妥钠溶液麻醉,剂量为45mg/kg BW,注射体积为0.2ml/100g BW。以分离胶非抗凝真空采血管从腹主动脉采血,采血完毕切断腹主动脉放血安乐死。
(2)血液指标检查
血液生化指标:分离胶促凝采血管以3000r/min离心10min分离血清样本,以全自动生化仪检测血生化指标。检测指标包括:Urea、CRE、TP、白蛋白(Alb)、UA、ALP。
4.3病理学观察
(1)大体解剖:实验第90天动物解剖并进行大体观察,取双侧肾脏并称量肾重,精确至三位小数。左肾用于组织病理学检查。
(2)组织病理学检查:左侧肾脏组织标本固定于10%中性福尔马林液中后,将其石蜡包埋,切成5μm切片,常规苏木精和伊红(H&E)染色。使用光学显微镜进行观察和拍照。
(3)TEM超微结构观察:将肾皮质组织样品切成1mm3块,固定在2.5%戊二醛(GA)电镜固定液中,并在0.1M磷酸盐缓冲盐水(PBS)中漂洗三次,每次10min,然后在室温下将样品置于1%四氧化锇溶液中1h。如上所述再次冲洗后,组织在梯度浓度乙醇和丙酮中脱水,然后用环氧酸盐12(Ted Pella)包埋。超薄切片用超薄切片机切割。样品用醋酸铀酰染色30分钟,然后与柠檬酸铅对比15分钟,并在120kV模式下用TEM进行检查。观察肾脏近曲小管及肾小球两个部位(主要观察近曲小管),寻找包括细胞膜、核膜、线粒体膜断裂,染色质凝聚不规则,线粒体内膜肿胀,粗面内质网扩张等改变;肾小球内皮细胞、蒂细胞、足细胞和系膜细胞的超微结构。
4.4大鼠血液、肾脏、粪便和尿液中Cd含量的测定
(1)血液
依4.2(1),用分离胶促凝管采集血液后,用肝素抗凝管从大鼠腹主动脉采集全血,于-20℃保存血样。检测前解冻摇匀,取200μL加1ml浓硝酸过夜后90℃恒温水浴4h(盖子旋松),再加超纯水定容,ICP-MS检测Cd含量。
(2)肾皮质
取右肾皮质切分(0.1-0.4g)后置于冻存管内,液氮急冻后于-80℃保存。检测前将样本从-80℃取出,加至消化管中,加5ml浓硝酸过夜;微波消解后170℃赶酸至溶液澄清透明;将溶液移至离心管,用超纯水冲洗管壁,定容后ICP-MS检测Cd含量。
(3)尿液
采集24h尿液后,取3ml于EP管中,-80℃保存。检测前尿液从-80℃取出解冻摇匀,ICP-MS检测Cd含量。
(4)粪便
第三个月采集尿液同时,用离心管采集大鼠24h粪便,于-80℃保存,离心管提前称重并记录空管重量,烘干后称量总重,记录粪便重量;粪便在85℃烘至恒重后,研磨粉碎成粉末状,称取0.02-0.07g至硝化管中,加入5ml浓硝酸过夜;经微波消解后170℃赶酸至溶液澄清透明,将溶液移至离心管,定容后ICP-MS检测Cd含量。
微波消解程序设定为:0-10min升到130℃保持5min,然后10min升到160℃,保持5min,20min升到170℃再保持10min。
5实验数据统计方法
使用SPSS 25.0(SPSS Inc.,Chicago,IL,USA)对实验数据进行分析。采用均数±标准差或频数和率对数据进行统计描述。对照组和试验组以及Cd组和nTiO2+Cd组之间的组间比较采用参数检验中的Student T检验(双尾)或非参数检验中的Mann-WhitneyU检验。P<0.05被认为是差异具有统计学意义。Cd和nTiO2的交互作用采用双因素方差分析(Two-way ANOVA,即析因分析),同时轮廓图(交互图)用于比较边际均值。
6实验结果与分析
6.1尿常规检查结果
第一个月尿常规检测结果:
(1)nTiO2组与对照组相比无显著异常;
(2)Cd组与对照组比较,KET、PRO、SG、MAlb、VC均显著升高;
(3)nTiO2+Cd组与对照组相比,PRO、SG、MAlb显著升高;与Cd组相比,PRO、KET降低,其中KET有显著意义(P<0.05)。
第二个月尿常规检查结果:
(1)nTiO2组与对照组相比无显著异常;
(2)Cd组与对照组相比,PRO、MAlb显著升高;其余指标与对照组相比无显著异常;
(3)nTiO2+Cd组与对照组相比PRO、MAlb显著升高;与Cd组相比无显著异常。
第三个月尿常规检查结果:
(1)nTiO2组与对照组相比无显著异常;
(2)Cd组与对照组比较,SG显著升高、pH显著降低;
(3)nTiO2+Cd组与对照组相比,pH显著降低;与Cd组相比SG显著下降(P<0.05)。
综上,尿常规结果表明,nTiO2对大鼠尿常规指标无显著影响;Cd暴露一个月后导致大鼠PRO和MAlb的显著升高,提示Cd导致大鼠肾脏受到损伤;nTiO2+Cd组与Cd组相比,KET、PRO、SG、MAlb均降低,且KET有统计学意义(P<0.05),体现肾毒性的减轻。

6.2尿生化分析结果
第一个月尿生化分析结果(图8,图9,图10):
(1)nTiO2组与对照组相比TP有显著升高,其余指标无显著差异;
(2)Cd组与对照组相比,NAG-T、NAG-B、MAlb、ALP、Urea、TP、CRE、UA显著升高;
(3)nTiO2+Cd组与对照组相比,NAG-T、MAlb、ALP、Urea、TP、CRE、UA显著升高;与Cd组相比,NAG-B、Urea与CRE显著降低;
(4)析因分析表明在NAG-T、MAlb、ALP、Urea、TP、CRE、UA指标中,Cd的主效应显著(P<0.01);Cd与nTiO2联合对Urea、CRE有显著拮抗效应(F=12.328,P=0.012;F=14.672,P<0.01)。
第二个月尿生化检测(图11):
(1)nTiO2组与对照组相比,各指标无显著差异;
(2)Cd组与对照组相比,Urea、TP、CRE、UA显著升高;
(3)nTiO2+Cd组与对照组相比,NAG-T、Urea、TP、CRE、UA显著升高;与Cd组相比,各项指标无显著差异;
(4)析因分析表明在NAG-T、MAlb、ALP、Urea、TP、CRE、UA指标中,Cd的主效应显著(P<0.05);但二者未见显著交互效应。
第三个月尿生化检测(图12):
(1)nTiO2组与对照组相比,各指标无显著差异;
(2)Cd组与对照组相比,NAG-T、NAG-B、ALP、Urea、TP、CRE、UA显著升高;
(3)nTiO2+Cd组与对照组相比,ALP、Urea、TP、CRE、UA显著升高;与Cd组相比,各项指标无显著差异;
(4)析因分析表明在NAG-T、NAG-B、ALP、Urea、TP、CRE、UA指标中,Cd的主效应显著(P<0.05);但二者未见显著交互效应。
NAG是存在于肾小管上皮细胞中的一种相对高分子量溶酶体酶,肾小管上皮细胞损伤时NAG释放到尿中,尿NAG增加。因此尿NAG水平在检测急性肾损伤时高度敏感,先于血清肌酐的增加,其中NAG的B型同工酶水平与肾损伤直接相关。本发明中第一个月Cd组大鼠的NAG-T及NAG-B显著升高,而联合组中两指标较Cd组下降,其中NAG-B有显著意义(P<0.05)(图8a,图8c);轮廓图显示nTiO2与Cd有交互效应(图8中b,d)。尿Urea及CRE是在临床上检测肾功能的常用指标。本发明中第一个月Cd组尿Urea及CRE显著上升(P<0.01),而联合组较Cd组显著下降(P<0.01)(图9a,图9c);轮廓图(图9b,图9d)结果与析因分析一致,表明Cd和nTiO2的联合暴露对Urea及CRE有显著的拮抗作用(F=12.328,P=0.012;F=14.672,P<0.01)。
尿常规及尿生化结果显示,每日灌胃100mg/kg BW nTiO2混悬液对大鼠肾脏无显著影响;每日自由饮用50mg Cd/L饮水从第一个月起可对大鼠产生显著的肾毒性,直至实验结束;Cd与nTiO2联合暴露,在第一个月表现出对Cd肾毒性的拮抗作用,但第二个月及终期未见显著交互作用。
6.3血清生化分析
从表6.3可见,试验终期各剂量组各项生化指标与对照组比较,差异均无统计学意义(P>0.05)。
对于Cd导致肾毒性而言,肾小管是早期主要受损的靶点。血清肌酐、尿素等指标的改变主要是由肾小球滤过功能障碍导致,不如尿液肾小管损伤标志物敏感。
6.4病理学观察结果
6.4.1大体观察、脏器重及脏器系数分析
大体观察未发现各组大鼠有明显病变。与对照组相比,终期各剂量组大鼠肾脏绝对重量与脏器系数差异均无统计学意义(P>0.05)。
6.4.2组织病理学观察结果
对肾脏进行组织病理学检查,结果如下:
(1)对照组:如图13a,镜下可见正常的肾皮质及髓质结构。该组大鼠肾小球无扩张,肾小球毛细血管无充血,间质偶见扩张充血的静脉;少数近曲小管上皮细胞近基质膜侧可见空泡形成,基本无间质炎症,血管正常。
(2)nTiO2组:如图13b,镜下可见基本正常的肾皮质及髓质结构。该组大鼠肾小球无扩张,肾小球毛细血管无充血,间质基本正常;肾小管病变较轻,仅表现为轻微的水肿,未见小管上皮细胞胞浆脱落;髓质区个别小管腔可见蓝染,脱落的上皮成分。
(3)Cd组:如图13c,该组大鼠肾脏可以见到不多于25%的急性肾小管损伤,主要表现为近端肾小管上皮细胞肿胀以及胞浆脱落,形成类似蛋白管型样的成分,少数(3/6)肾间质可见小簇单个核细胞浸润,主要位于小动脉周围。肾小球未见扩张、充血等改变,未见远曲小管的异常,未见修复性改变。
(4)nTiO2+Cd组:如图13d,该组大鼠肾脏肾小管病变轻,仅表现为轻微上皮细胞水肿,个别肾组织(2/6)可见到小管管腔扩张,但尚不足以诊断为急性肾小管损伤。髓质区个别小管腔可见蓝染,脱落的上皮成分(1/6)。肾小球未见扩张、充血或缺血等改变,未见修复性改变。
组织病理学结果显示,Cd导致急性肾小管损伤,通常是以近端肾小管上皮细胞变性、蛋白管型、炎症细胞浸润为特征;而与nTiO2共暴露使损伤减轻。
6.4.3 TEM超微结构观察结果
我们重点研究了与Cd诱导的肾损伤相关的超微结构变化,尤其是近端肾小管上皮细胞。超微结构观察显示了对照组的正常亚细胞结构,如正常微绒毛、细胞核和线粒体(图14a)。nTiO2组大鼠肾脏没有明显的超微结构变化(图14b)。Cd组微绒毛消失,细胞质和细胞器脱落至管腔(图14c)。线粒体肿胀,部分线粒体内嵴完全消失,形成透明的电子密度区(图14d)。在nTiO2+Cd组大鼠中,肾小管管腔中偶尔可见脱落的细胞器,但肾小管上皮细胞的微绒毛基本正常(图14e)。该组线粒体的部分外膜和内嵴溶解,但未发现线粒体内嵴完全消失(图14f)。
超微结构的变化证实了Cd组和nTiO2+Cd组出现肾损伤,而nTiO2+Cd组的损伤较Cd组减轻。这些发现与组织病理学检查结果一致。
6.4.4血液、肾脏、粪便和尿液Cd含量
通过ICP-MS测得血液、肾脏中的Cd浓度;通过24h粪便总重及总尿量计算得24h粪便或尿液Cd排泄总量。在Cd组和nTiO2+Cd组中,接触Cd显著增加血液中的Cd浓度(图15a)以及肾皮质中的Cd浓度(图15b)。与Cd组相比,nTiO2+Cd组血液和肾皮质的Cd含量降低19.2%和8.6%。同时,两组大鼠粪便和尿液中的Cd排泄量显著增加(图15c,d)。与Cd组相比,nTiO2+Cd组24h粪便中Cd含量升高3.7%,而24h尿Cd含量降低28.6%。
Cd组和联合组暴露90天后,24h尿Cd含量显著升高。尿Cd含量是慢性Cd暴露的一个指标。正常情况下,每天通过尿液排出的Cd是有限的;而当近端肾小管损伤时,尿Cd排泄量明显增加。研究表明,尿Cd含量与肾脏损伤呈正相关关系。在本发明中,联合组24h尿Cd排泄量减少了28.6%,与前面的结果一致,证实了联合暴露后对Cd引起肾功能损伤的改善。
在本发明中,联合组粪便中的Cd含量升高3.7%,而血液和肾脏中Cd含量分别下降19.2%和8.6%。本发明中观察到的拮抗作用可能归因于nTiO2导致Cd的生物利用度降低。nTiO2可通过吸附形成nTiO2-Cd络合物,使Cd的排泄量增加,Cd在体内的吸收和分布下降,导致Cd的生物利用度降低,引起肾毒性降低。
研究表明,nTiO2与Cd联合暴露可降低介质中游离Cd2+的浓度,而产生毒作用的是游离Cd2+。研究发现,nTiO2能降低培养液和生物体中Cu、Pb、Zn等重金属的游离离子浓度。Wang等人通过EDX光谱证实,Cd在介质中分散后与nTiO2聚集体中的Ti和O元素发生重叠,表明nTiO2粒子能够有效地吸附和富集Cd。对大肠杆菌的研究表明,当培养基中Cd2+浓度随nTiO2浓度的增加而降低时,nTiO2与Cd共暴露后,其LC50和生长速率均增加。在另一项对紫贻贝(Mytilus gallospeccialis Lam.)的研究中,Cd和nTiO2共暴露后,Cd的遗传毒性有所改善,贻贝鳃中Cd含量降低14.8%。Yang等人计算了nTiO2在细胞表面的最大扩散和实际吸收速率,提出如果纳米颗粒的尺寸大于10nm,nTiO2的吸收将受到细胞表面扩散的限制。因此,当纳米颗粒尺寸大于10nm时,复合物很难进入细胞,更可能产生拮抗作用,这与本发明一致。
再者,nTiO2的强吸附能力及与Cd的络合物结构可阻止Cd的解吸。Ahamed等人发现nTiO2显著抑制了Pb在A549细胞中的生物利用度和毒性,由于nTiO2吸附能力强,即使在细胞内也不存在游离Pb2+。Rocco等人根据定量构效关系(Quantitative Structure-ActivityRelationship,QSAR)计算方法计算发现,nTiO2-Cd络合物呈“三明治”结构,其中中心Cd被结构表面的nTiO2完全遮蔽。即使络合物被吸收并在细胞内转运,毒性也可能会降低,因为nTiO2对Cd的高结合亲和力限制了Cd的解吸,降低了游离Cd2+浓度。
以上所述实施例仅表达了本发明的几种实施方式,其描述较为具体和详细但并不能因此而理解为对发明专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进,这些都属于本发明的保护范围。因此,本发明专利的保护范围应以所附权利要求为准。
