Chitin, also known as chitin, is a structurally homologous polysaccharide formed by the polymerization of N-acetylglucosamine through beta linkage. It is widely found in the shells of crustaceans, the crustaceans of insects, and the cell walls of fungi, and also in some green algae, mainly to serve as a skeleton for supporting the body and to protect the body. The polygonatum polysaccharide in the present application may be commercially available, for example, as a product sold under the product number BL-2020102804 by the biotechnology limited of baysu, shanxi.
Ganoderan is a secondary metabolite of mycelium of fungus belonging to genus Ganoderma of family Polyporaceae, and is present in mycelium and fruiting body of fungus belonging to genus Ganoderma. The ganoderan is a light brown to tan powder, and is soluble in hot water. The ganoderan has the effects of reducing blood sugar and blood fat, resisting thrombus and oxidation, scavenging free radicals, resisting aging, radiation and tumor, promoting blood circulation, regulating immunity, regulating nucleic acid and protein metabolism, promoting DNA synthesis, and promoting proliferation of LAK cell in cord blood. The ganoderma lucidum polysaccharide has special physiological activity and clinical effect, is safe and nontoxic, and can be widely applied to the industries of medicine, food and cosmetics.
The ganoderan of the present application may be commercially available, for example, as SQLZ from St.John, Biotech, Inc.
In some preferred embodiments, the composition comprising the rehmannia glutinosa oligosaccharide comprises the following components in parts by weight: 2-3 parts of polygonatum polysaccharide and 5-7 parts of ganoderma polysaccharide.
The second aspect of the invention provides application of the composition containing the rehmannia glutinosa oligosaccharide in skin care products, which are characterized in that the skin care products comprise essence water, facial masks and emulsion.
The rehmannia glutinosa oligosaccharide in the application can be prepared from rehmannia glutinosa powder or can be directly purchased.
In one embodiment, the method for preparing the rehmannia glutinosa oligosaccharide comprises: crushing a rehmannia root raw material to 10-30 meshes; dynamically extracting the rehmannia powder by taking subcritical water as an extracting solution; filtering to obtain supernatant, purifying the supernatant by anion exchange resin, and collecting effluent liquid; adding deionized water for elution, collecting eluent, and mixing the effluent and the eluent; desalting the obtained feed liquid by using cation exchange resin; adding deionized water for elution, collecting eluent, and mixing the effluent and the eluent; filtering the obtained feed liquid with microporous membrane, concentrating under reduced pressure, and drying.
The specific preparation parameters of the rehmanniae radix oligosaccharides in the present application are not particularly limited, and those skilled in the art can routinely select the parameters according to the actually required properties of the rehmanniae radix oligosaccharide species.
The preparation method of stachyose comprises the following steps: crushing a rehmannia root raw material to 10-30 meshes; and dynamically extracting the rehmannia powder by taking subcritical water as an extracting solution, wherein the material-to-liquid ratio of the rehmannia powder to the water is 1 g: 6mL, the extraction time is 30min, the extraction pressure is 5MPa, and the extraction temperature is 120 ℃; filtering to obtain supernatant, purifying the supernatant with anion exchange resin at flow rate of 3m3Resin/h, collecting effluent liquid; adding deionized water for elution, collecting eluent, and mixing the effluent and the eluent; desalting the obtained feed solution with cation exchange resin at flow rate of 10m3Resin/h, collecting effluent liquid; adding deionized water for elution, collecting eluent, and mixing effluent and eluent; filtering the obtained feed liquid with 0.6-0.8 μm microporous membrane, concentrating at 70 deg.C under reduced pressure, and drying to obtain stachyose product.
Examples
The technical solution of the present invention is described in detail by the following examples, but the scope of the present invention is not limited to the examples. The starting materials in the present invention are commercially available, unless otherwise specified, and can be prepared from rehmannia glutinosa, if conditions permit.
The preparation method of stachyose comprises the following steps: crushing a rehmannia root raw material to 10-30 meshes; and dynamically extracting the rehmannia powder by taking subcritical water as an extracting solution, wherein the material-to-liquid ratio of the rehmannia powder to the water is 1 g: 6mL, the extraction time is 30min, the extraction pressure is 5MPa, and the extraction temperature is 120 ℃; filtering to obtain supernatant, purifying the supernatant with anion exchange resin at flow rate of 3m3Resin/h, yieldCollecting and discharging liquid; adding deionized water for elution, collecting eluent, and mixing effluent and eluent; desalting the obtained feed solution with cation exchange resin at flow rate of 10m3Resin/h, collecting effluent liquid; adding deionized water for elution, collecting eluent, and mixing the effluent and the eluent; filtering the obtained feed liquid with 0.6-0.8 μm microporous membrane, concentrating at 70 deg.C under reduced pressure, and drying to obtain stachyose product.
The raffinose in this application is purchased from Shanghai Michelin Biochemical technology, Inc. under the product designation D832616-1 g.
Mannotriose in this application is available from Shanghai Michelin Biotechnology Limited under the trade designation M860556-5 mg.
The verbascose is obtained from the product sold under the trade name B27137-5mg from Shanghai-derived leaf Biotechnology Ltd.
The tremella polysaccharide in the application is purchased from a product sold by Shanghai Michelin Biotechnology Limited with a product number of T861210-20 mg.
Aloe polysaccharide in this application is purchased from Shanghai-derived leaf Biotech Co., Ltd under a product number of B24395-20 mg.
Fucoidan in this application is purchased from Shanghai Michelin Biotechnology Ltd under the trade designation F885876-1 g.
The cordyceps polysaccharide in the application is purchased from a product sold by Jiantong biological products Limited in Shaanxi with the product number of 5495553145.
Polyporus polysaccharide in this application is purchased from the product sold under the trade name REL-ZL10 by Sierra bioengineering, Inc.
The achyranthes bidentata polysaccharides are purchased from Shanghai Van ecology Biotech Co., Ltd under the trade name FT-R8514.
Chitin in this application was purchased from a product sold by Shenzhen topology Biotechnology Limited.
The lycium barbarum polysaccharides in the application are purchased from the product with the product number of 003 sold by saint biotechnology limited of Shaannan province.
The polygonatum polysaccharide in the application is purchased from a product with a product number of BL-2020102804 sold by Shaanxi Bolin biotechnology limited company.
The ganoderan in this application is purchased from the product sold by sienna holy biotechnology limited under the product number SQLZ.
The elm bark polysaccharide in this application is purchased from ningxia vanilla biotechnology limited with a specification of 20: 1.
Example 1
Example 1 provides a composition comprising a rehmannia glutinosa oligosaccharide, comprising the following components in parts by weight: 4 parts of tremella polysaccharide, 6 parts of aloe polysaccharide, 7 parts of fucoidin, 6 parts of raffinose, 6 parts of stachyose, 6 parts of mannotriose, 6 parts of verbascose, 2.5 parts of cordyceps polysaccharide, 5 parts of polyporus polysaccharide, 6 parts of achyranthes polysaccharide, 5 parts of chitin, 8 parts of lycium polysaccharide, 2.5 parts of polygonatum polysaccharide, 6 parts of ganoderma polysaccharide and 5 parts of elm bark polysaccharide.
Example 2
Embodiment 2 provides a composition containing a rehmannia glutinosa oligosaccharide, which comprises the following components in parts by weight: 1 part of tremella polysaccharide, 2 parts of aloe polysaccharide, 4 parts of fucoidin, 2.5 parts of raffinose, 2.5 parts of stachyose, 2.5 parts of mannotriose, 2.5 parts of verbascose, 1 part of cordyceps polysaccharide, 1 part of polyporus polysaccharide, 2 parts of achyranthes polysaccharide, 3 parts of chitin, 4 parts of lycium barbarum polysaccharide, 1 part of polygonatum polysaccharide, 3 parts of ganoderma polysaccharide and 1 part of elm bark polysaccharide.

Evaluation of Performance
1. Efficacy measurement of different examples for moisturizing the stratum corneum of the skin
Healthy skin should be smooth, moist, and elastic, which is largely attributable to the moisture in the skin, and these appearance signs are related to the function of the skin's own moisturizing system. The moisturizing system has the functions of maintaining a certain moisture content of the skin and resisting the damage of evaporative dryness, even embrittlement and the like to the skin caused by internal and external factors. When the water content of the skin is sufficient, the skin becomes full of water, and particularly, the water content of the stratum corneum directly influences the apparent state of the skin.
Whether the skin looks dry on the surface mainly depends on the water content of the horny layer of the epidermis, and the skin is moist, compact and elastic, and is the most ideal skin state; when the water content of the skin is too low, the skin becomes dry, rough and even cracked. Meanwhile, when the skin is in a dry and dry state, the skin is very fragile and is easily influenced by the environment, so that a series of problems of dull skin, easy sensitization, aging and the like occur.
80 volunteers were selected for the test and divided into 8 groups of 10 persons each, one group being a control group and not treated, and the remaining 7 groups, and the test was performed using the skin care products prepared using the compositions of examples 1-3 and comparative examples 1-4 as additives, which were essence water prepared from the composition of the present application in 10% by weight and deionized water in 90% by weight. The 80 volunteers respectively wash the face with clear water and wipe clean, at the moment, the digital skin moisture detector is used for measuring the moisture content of the skin, then the skin care product is uniformly smeared on the face, then the moisture content of the skin is respectively measured after 0.5h, 1h, 2h, 4h and 8h, the average value of each group of measurement results is calculated, and the average value of the moisture content is obtained, which is shown in the following table.

From the above table, it can be seen that the use of the composition of the present application is effective in increasing skin moisture content, with example 1 being the most preferred example.
Finally, it should be noted that the above-mentioned embodiments are only preferred embodiments of the present invention, but the embodiments of the present invention are not limited by the above-mentioned embodiments, and any other changes, modifications, substitutions, combinations, and simplifications which do not depart from the spirit and principle of the present invention should be regarded as equivalent replacements within the protection scope of the present invention.

Claims (10)

1. A composition comprising a rehmannia glutinosa oligosaccharide, comprising at least one of the following: the polysaccharide is one or more of raffinose, stachyose, mannotriose and verbascose.
2. A composition comprising a rehmannia glutinosa oligosaccharide according to claim 1, comprising the following components in parts by weight: 1-8 parts of tremella polysaccharide, 2-10 parts of aloe polysaccharide, 4-10 parts of fucoidin, 10-40 parts of rehmannia oligosaccharide and 1-10 parts of elm bark polysaccharide.
3. A composition comprising a rehmanniae oligosaccharide according to claim 2, comprising the following components in parts by weight: 3-5 parts of tremella polysaccharide, 4-8 parts of aloe polysaccharide, 6-8 parts of fucoidin, 20-32 parts of rehmannia root oligosaccharide and 4-6 parts of elm bark polysaccharide.
4. A composition comprising rehmania oligosaccharide according to claim 3, further comprising the following components in parts by weight: 1-4 parts of cordyceps polysaccharide, 1-8 parts of polyporus polysaccharide and 2-12 parts of achyranthes polysaccharide.
5. A composition comprising rehmania oligosaccharide according to claim 4, wherein the composition comprises the following components in parts by weight: 2-3 parts of cordyceps polysaccharide, 3-6 parts of polyporus polysaccharide and 4-8 parts of achyranthes polysaccharide.
6. A composition comprising rehmania oligosaccharide according to claim 5, further comprising the following components in parts by weight: 3-9 parts of chitin and 4-15 parts of lycium barbarum polysaccharide.
7. A composition comprising rehmania oligosaccharide according to claim 6, wherein the composition comprises the following components in parts by weight: 4-7 parts of chitin and 6-10 parts of lycium barbarum polysaccharide.
8. A composition comprising rehmania oligosaccharide according to claim 7, further comprising the following components in parts by weight: 1-5 parts of polygonatum polysaccharide and 3-10 parts of ganoderma polysaccharide.
9. A composition comprising rehmania oligosaccharide according to claim 8, wherein the composition comprises the following components in parts by weight: 2-3 parts of polygonatum polysaccharide and 5-7 parts of ganoderma polysaccharide.
10. Use of a composition comprising a rehmanniae oligosaccharide according to any of claims 1-9 in a skin care product comprising serum, mask, lotion.
