技术领域
本发明涉及重金属检测技术领域,具体涉及一种同位素示踪沉积物重金属生物可利用性的ICP-MS法。
背景技术
重金属的生物可利用性是指重金属被生物吸收的性质,最开始被认为是在水体环境中污染物在生物传输和生物反应中被利用的程度,后来扩展到了土壤、底泥等环境中(Benson etal.,1994)。在环境生态毒理学领域学者看来,生物可利用性是指沉积物中重金属等污染物直接被生物所利用及在生物体内产生毒性效应的难易程度,即污染物则越容易为生物所吸收利用代表其生物可利用性越高,可应用于沉积物污染物生态风险评估(锺寰和王文雄,2011)。不同学科对生物可利用性的理解和定义不同,生物可利用性所包涵的内容非常广泛,而基于化学概念上的生物可利用性则是指污染物质如重金属元素是否能被生物吸收以及其潜在的毒性,其实质在于研究污染物与生物之间的相互关系。
美国国家研究委员会(National Research Council,2003)认为生物可利用性可分为固相(沉积物或土壤)与污染物结合的三个主要过程:首先与沉积物的结合并从沉积物释放的污染物;其次相关污染物迁移到达生物膜的过程;再次污染物从生物膜外部环境穿过生物的生理屏障(如细胞膜等体表屏障)进入到生物体内的过程。
单纯依靠研究重金属元素的总量往往很难表征其污染特性和危害,不同形态的重金属的生物可利用性和对生物的毒性不同。金属在沉积物中与各化学相的结合以及在各种条件下的释放,直接影响着金属在沉积物中的赋存形态,从而决定着金属的生物可利用性。
发明内容
重金属等污染物对生物的危害取决于生物对重金属的吸收利用效率,传统的沉积物测定重金属总量与实际上对生物的危害不一定成正相关性,生物可利用性能够更客观评估沉积重金属中对生物的危害。
一般用室内培养胁迫实验来评估重金属的生物可利用性,相关实验培养方法耗时长,针对特定模式生物,对于不同区域不同生物没有可比性,因此,需要开发一种可靠的方法来评估重金属的生物可利用性。
牛血清白蛋白用于模拟生物肠道消化液对重金属的吸收,经同位素示踪技术验证其可行性,可用于环境中重金属的危害评估,该方法相比传统的其他方法,操作简便,耗时短,可以更准确的评估环境中重金属的危害,从而为相关环境保护和管理提供科学支撑。
本发明的技术方案如下:
一种同位素示踪沉积物重金属生物可利用性的ICP-MS法,其包括如下步骤:
(3)沉积物重金属总量的测定
(4)同位素标记示踪:通过合成不同铅同位素标记的沉积物中离子交换态铅(206Pb,以硝酸铅形式存在),将其添加到沉积物中
(3)生物可利用性提取:将沉积物样品用加入牛血清白蛋白溶液,震荡、萃取;
(4)用ICP-MS测定牛血清白蛋白中重金属含量和同位素含量;
以百分比含量表示的生物可利用态重金属(S,%)计算如下:
CB——以mg/kg表示的生物可利用态重金属;
CT——沉积物中重金属总量(mg/kg);
Cdissolved——提取后牛血清白蛋白提取液中溶解重金属的浓度(μg/L);
Cbackground——牛血清白蛋白提取液中的溶解重金属本底浓度(μg/L);
L——加入沉积物中进行培养的消化液体积(L);
m——提取试验中所用的沉积物质量(g)。
将牛血清白蛋白粉末用纯水配制成5g/L(0.045mol/L)BSA溶液,牛血清白蛋白溶液 (5g/L)
样品载入:样品通过进样器经蠕动泵(Pump 1,提升转速0.5rps,稳定转速0.3rps)进入雾化器(Nebulizer)雾化成气溶胶进入矩管(Torch),在高温等离子体中原子化,再通过样品锥与截取锥,在离子透镜中带正电荷离子进行聚焦,通过四级杆电磁场根据质量/电荷比值进行偏转,进入检测器监测。
i)四极杆质谱优化:利用标准调谐液优化仪器性能,控制89Y,70/140Ce++/Ce+双电荷指标,156/140CeO/Ce氧化物指标,等离子体稳定后,优化四级杆质谱,从而保证测定结果的精密度。
ii)质量歧视效应:主要是由于离子传输过程中的空间电荷效应,对于质量歧视效应可通过外标或内标法进行校正。外标法校正:是利用已知同位素比值的Pb标准来进行校正。
内标法校正:选择与待测元素质量数相近的已知同位素比值的元素来进行校正。
一般选择Tl元素(203Tl/205Tl)按照下式来进行计算:
其中:Z为已知标准的同位素比值,I为离子的检测强度。
iii)基体效应以及离子干扰
ICP-MS分析过程中,样品中存在的204Hg会对丰度较低的204Pb带来离子干扰,可按下面的干扰校正方程来进行消除:

iv)死时间
在高计数率下,ICP-MS仪器所用的通道式电子倍增管和脉冲计数系统采集的计数比实际的少,这是因为检测器及计数系统存在死时间,死时间将导致仪器计数率在超过某个额定值时成为非线性关系。采用质量浓度分别为0、5、10、20、40、60ng/mL 的GBW04424~27标准溶液对死时间进行测定,选择最为合适的死时间使仪器达到最佳信噪比。
用ICP-MS实际样品测定前,应进行空白试验,按照样品测量的步骤,对空白样品进行测量,空白值应控制在0.1μg/L以内。
实验空白实验进行如下:随机抽取按流程进行清洗干净的容器,加入实验步骤所用的溶液,分析这份空白溶液中各个待测金属的含量,得到容器空白;测试每个步骤所用试剂溶液中各种待测金属的含量,得到试剂空白;在进行分级提取实验时,一份空白样品容器中不加入沉积物样品也将进行与样品相同的操作流程,最后按照实验要求测试,得到系统空白。
内标校正
使用45Sc,72Ge,89Y,115In,209Bi作内标物,所有的空白、标准溶液及样品均在线添加内标,混合后内标浓度为15μg/L,校正方式为虚拟内标校正(VIS)
干扰及消除
1)通过控制氧化物指标及双电荷指标,以及89Y计数灵敏度的方法,来优化仪器条件。
2)通过He碰撞模式消除大部分多原子离子干扰,如测定砷时,75As在75质量数处存在40Ar35Cl+,通过碰撞模式,He可以将大部分40Ar35Cl+打碎为单原子,从而消除40Ar35Cl+对As测定的干扰。
3)通过选择适当同位素避开干扰,如测定锌时,66Zn在66质量数存在31P35Cl+干扰,通过选择68Zn测定来避开干扰。
本发明的优点:
通过不同同位素标记,从而确定重金属的生物可利用部分的明确来源,传统的分析检测方法不能判断重金属的来源。从而为重金属污染迁移,如何通过食物链累积富集提供明确的科学依据,为环境管理部门提供技术支撑。
利用牛血清白蛋白仿生萃取剂模拟底栖无脊椎中肠胃消化液,利用铅同位素进行示踪标记,该萃取剂具有易获得,成分与底栖生物肠胃消化液蛋白成分类似的优点,从而建立沉积物中的重金属对海洋生物特别是底栖生物的生物可利用性定量分析方法。

具体实施方式
为了使本领域的技术人员更好地理解本发明的技术方案,下面进一步披露一些非限制实施例对本发明作进一步的详细说明。
2.1生物可利用性提取
用蚌式挖泥斗采集表层沉积物样品,采集上来的样品用塑料勺取表层0~5cm沉积物密封于预先冲入氮气的洁净聚乙烯塑料封口袋内,冷冻保存带回实验室。在实验室中,湿沉积物样品在冷冻干燥器中冻干,剔除砾石、贝壳等杂物,用玛瑙研钵研磨后用160 目(近似孔径为0.01mm)尼龙筛进行筛分,四分法缩分分取10~20g制备好的样品,装袋放入冰箱冷冻保存,作为待测样品。
本专利主要利用牛血清白蛋白(Bovine Serum Albumin,BSA)作为仿生萃取剂研究沉积物重金属的生物可利用性,将牛血清白蛋白粉末用纯水配制成5g/L(0.045mol/L)BSA溶液,将冷冻的新鲜沉积物样品用冷冻干燥仪冻干后,低温研磨过筛,称取2.5g 冻干新鲜样品放入250mL具塞三角烧瓶,加入187ml牛血清白蛋白溶液(5g/L)(Kalman andTurner,2007),旋紧盖子放在摇床振荡器以100rpm室温避光培育萃取20h(Zhong andWang,2008&2009),后将液体倒入50mL具塞聚丙烯离心塑料管,放入旋转离心仪以3000rpm离心半小时。每组均进行3个平行样品分析,另外取空白三角烧瓶,不放置样品,同样按照样品步骤进行实验,做5组样品空白实验(空白实验等质量控制要求见3.1.3)。取上清液20mL加入0.2mL默克农残级别硝酸酸化,用ICP-MS直接分析牛血清白蛋白提取液重金属含量(见2.2)。

CB——以mg/kg表示的生物可利用态重金属;
CT——沉积物中重金属总量(mg/kg);
Cdissolved——提取后牛血清白蛋白提取液中溶解重金属的浓度(μg/L);
Cbackground——牛血清白蛋白提取液中的溶解重金属本底浓度(μg/L);
L——加入沉积物中进行培养的消化液体积(L);
m——提取试验中所用的沉积物质量(g)。
2.2同位素标记示踪
利用同位素示踪技术,通过合成不同铅同位素标记的沉积物中离子交换态铅(206Pb,以硝酸铅形式存在),将其添加到沉积物中。分别称取两份,2.5g冻干新鲜样品放入250 mL具塞三角烧瓶,其中一份再加入准确量1.87ml 250ppb离子交换态铅206Pb进行同位素示踪,混合均匀后,再按照2.1加入牛血清白蛋白进行生物可利用性萃取,根据 Pb206的加标回收率,验证牛血清白蛋白仿生萃取生物可利用性的可行性。
根据同位素铅206标记示踪试验,铅206的加标回收率达到了95.6%,证明了牛血清白蛋白可以基本完全萃取离子交换态铅,牛血清白蛋白可以用于仿生萃取。
2.3沉积物重金属总量测定
准确称取0.5000g样品,置于密闭微波消解仪聚四氟乙烯消解罐中,加入8ml高纯浓硝酸和2ml高纯氢氟酸消解,使用Milestone微波消解系统180℃梯度升温,高压消解2h,消解完成后于电热板上赶酸,冷却卸压后,冷却取出将消解液转移至50ml离心管,用MilliQ超纯水定容至50ml后定容至100.0ml。样品消解时每批样品(40个/批) 加入2个超纯水样品用以扣除空白,以及一个近海海洋沉积物成分标准物质(GBW07314) 和南海海洋沉积物成分分析标准物质(GBW07334)用作质量控制。
2.4仪器试剂
仪器:Agilent 7900型带有碰撞/反应模式(ORS)的电感耦合等离子体质谱 (DFISICP MS)。
试剂:硝酸(Suprapur级,美国默克公司),Agilent Environmental calibrationstandard(P/N 51834688,美国安捷伦公司),超纯水(电阻率≥18.2MΩ,Millipore 纯水制备系统)。
2.5仪器方法
样品载入[图1]:样品通过进样器经蠕动泵(Pump 1,提升转速0.5rps,稳定转速0.3rps)进入雾化器(Nebulizer)雾化成气溶胶进入矩管(Torch),在高温等离子体中原子化,再通过样品锥与截取锥,在离子透镜中带正电荷离子进行聚焦,通过四级杆电磁场根据质量/电荷比值进行偏转,进入检测器监测。而大液滴的气溶胶在雾化室(Spray Chamber)冷凝,经蠕动泵2(Pump 2,转速0.3rps)进入废液桶(Drain)。
2.4实验条件
RF功率:1450W,等离子体气流速:15.0L/min,载气流速:0.60L/min,补偿气流速:0.60L/min,雾化室温度:2℃,氧化物指标:156CeO+/140Ce+<0.5%,双电荷指标:70Ce++/140Ce+<1.5%,灵敏度89Y(1μg/L)≥1200M cps。
使用Agilent Environmental calibration standard(P/N 51834688,美国安捷伦公司)先配制成中间液,再逐级稀释配制0、1.0、2.0、5.0、10.0μg/L(铜、铅、锌、镉、铬)工作曲线。
2.5工作曲线
由于海水等高盐样品在测试时,会在锥口结盐导致基线漂移,影响结果准确性,采用标准加入法来消除海水基体干扰。使用离岸距离大于200海里的外海干净海水作为基体,在基体海水中加入已稀释的混合标准溶液,配制0、1.0、2.0、5.0、10.0μg/L工作曲线。
2.6试验空白控制
用ICP-MS实际样品测定前,应进行空白试验,按照样品测量的步骤,对空白样品进行测量,空白值应控制在0.1μg/L以内。
实验空白实验进行如下:随机抽取按流程进行清洗干净的容器,加入实验步骤所用的溶液,分析这份空白溶液中各个待测金属的含量,得到容器空白;测试每个步骤所用试剂溶液中各种待测金属的含量,得到试剂空白;在进行分级提取实验时,一份空白样品容器中不加入沉积物样品也将进行与样品相同的操作流程,最后按照实验要求测试,得到系统空白。
2.7内标校正
使用45Sc,72Ge,89Y,115In,209Bi作内标物,所有的空白、标准溶液及样品均在线添加内标,混合后内标浓度为15μg/L,校正方式为虚拟内标校正(VIS).
2.8干扰及消除
1)通过控制氧化物指标及双电荷指标,以及89Y计数灵敏度的方法,来优化仪器条件。
2)通过He碰撞模式消除大部分多原子离子干扰,如测定砷时,75As在75质量数处存在40Ar35Cl+,通过碰撞模式,He可以将大部分40Ar35Cl+打碎为单原子,从而消除40Ar35Cl+对As测定的干扰。
3)通过选择适当同位素避开干扰,如测定锌时,66Zn在66质量数存在31P35Cl+干扰,通过选择68Zn测定来避开干扰。
由于同位素间的质量差很小,因而分析精密度和准确度对于测定结果具有较大的影响。影响ICP-MS测定铅同位素精密度的因素主要来源于由于等离子体和四极杆质谱过滤器造成的仪器噪音和不稳定性;而影响样品分析准确性的因素主要包括:质量歧视效应、离子干扰以及四极杆质谱的死时间,因而本申请课题拟从以下几个方面对测定准确性进行研究:
i)四极杆质谱优化:利用标准调谐液优化仪器性能,控制89Y,70/140Ce++/Ce+双电荷指标,156/140CeO/Ce氧化物指标,等离子体稳定后,优化四级杆质谱,从而保证测定结果的精密度。
ii)质量歧视效应:主要是由于离子传输过程中的空间电荷效应,对于质量歧视效应可通过外标或内标法进行校正。外标法校正:是利用已知同位素比值的Pb标准来进行校正。
内标法校正:选择与待测元素质量数相近的已知同位素比值的元素来进行校正。
一般选择Tl元素(203Tl/205Tl)按照下式来进行计算:
其中:Z为已知标准的同位素比值,I为离子的检测强度。
iii)基体效应以及离子干扰
ICP-MS分析过程中,样品中存在的204Hg会对丰度较低的204Pb带来离子干扰,可按下面的干扰校正方程来进行消除:
其中Abundance(204Hg)/Abundance(202Hg)=0.23
iv)死时间
在高计数率下,ICP-MS仪器所用的通道式电子倍增管和脉冲计数系统采集的计数比实际的少,这是因为检测器及计数系统存在死时间,死时间将导致仪器计数率在超过某个额定值时成为非线性关系。本申请课题拟采用质量浓度分别为0、5、10、20、40、60ng/mL的GBW04424~27标准溶液对死时间进行测定,选择最为合适的死时间使仪器达到最佳信噪比。
实施例1珠江口沉积物
采集了珠江口沉积物样品,珠江口沉积物中生物可利用态重金属含量见表3.1-1与表3.1-2,空间分布见表3.1-1。
珠江口沉积物中生物可利用态砷含量的分布范围在3.20~26.7mg/kg,占沉积物砷总量的提取百分比为0.78%~6.52%,平均值为8.71mg/kg(平均3.21%),沉积物中生物可利用态砷高值出现在珠江口中部海域。沉积物中生物可利用态镉含量的分布范围在0.05~2.04mg/kg,占沉积物镉总量的提取百分比为0.81%~53.01%,平均值为0.61mg/kg(平均15.90%),镉生物可利用态分布与总量分布类似,西岸海域沉积物中生物可利用态镉较高。沉积物中生物可利用态铬含量的分布范围在1.88~77.7mg/kg,占沉积物铬总量的提取百分比为0.27%~5.94%,平均值为16.4mg/kg(平均2.53%),中部海域沉积物中生物可利用态铬较高。沉积物中生物可利用态铜含量的分布范围在28.0~663.7 mg/kg,占沉积物铜总量的提取百分比为7.98%~50.24%,平均值为93.7mg/kg(平均 24.19%)。铜生物可利用态分布与总量分布类似,西岸东四大口门附近海域和深圳宝安海域沉积物中生物可利用态铜较高。
珠江口沉积物中生物可利用态铅含量的分布范围在1.27~28.8mg/kg,占沉积物铜总量的提取百分比为0.03%~5.03%,平均值为6.54mg/kg(平均1.57%),中部靠近香港海域沉积物中生物可利用态铅较高。沉积物生物可利用性锌含量的分布范围在 1.20~166.6mg/kg,占沉积物锌总量的提取百分比为2.04%~14.39%,平均值为60.3mg/kg (平均6.75%),其中珠江口西岸的附近海域的浓度较高,并且呈现珠江口内向珠江口外逐级降低趋势。
本专利方法用于研究珠江口沉积物铜的生物可利用性百分比含量在24.19%(7.98%~50.24%),高于美国缅因湾沉积物海洋底栖无脊椎动物肠消化液提取中Cu的生物可利用态百分比含量(0~14%)(Mayer et al.,1996),可能主要由于不同生物的消化液具有不同的消化活性及有机质,以及沉积物本身的地球化学性质也是影响两个不同区域沉积物重金属生物可利用性的重要因子(Mayer et al.,2001)。本专利方法用于研究珠江口铜生物可利用性百分比含量也高于Wang等(2012a)利用光裸方格星虫肠消化液提取珠江口的铜生物可利用性百分比含量(2.8~27.5%),采用同样区域的沉积物作为研究对象排除了不同沉积物含有的有机质控制着沉积物中Cu等重金属的生物可利用性可能 (Mcaloonand Mason,2003),这可能与牛血清白蛋白本身的分子基团上含有较多的巯基 -C-SH和二硫键-C-S-S-C-等含硫键有关,较容易与沉积物的铜离子形成金属硫键(蛋白 -S-Cu)有关。因此,采用不同提取能力的生物提取液所得到的生物可利用性重金属含量也有所差异。

本专利方法用于研究珠江口沉积物锌的生物可利用性百分比含量在6.75%(2.04%~14.39%),与Wang等(2012a)利用光裸方格星虫肠消化液提取珠江口的锌生物可利用性百分比含量7.93%(2.55~21.34%)相当,略低于同样用光裸方格星虫肠消化液提取辽宁湾海域沉积物中Zn的生物可利用态百分比含量8%(5~15%)(Yan and Wang,2002)。说明对于提取沉积物生物可利用态Zn的能力,牛血清白蛋白的提取能力与光裸方格星虫肠消化液相当。
本专利方法用于研究珠江口沉积物铅的生物可利用性百分比含量在1.57%(0.03%~5.03%),明显低于珠江口沉积物其它重金属生物可利用性百分比含量,导致铅元素相对其它重金属较难提取的原因,Mayer等(1996)认为可能由于沉积物具有较小的粒度(如粘土、粉砂)导致Pb更容易被沉积物吸附从而更难被释放到提取液,Chen等 (2000)认为可能是Pb形成配合物、氢氧化物和硫化物等导致被沉积物吸附。而 Wang(2012a)发现利用光裸方格星虫肠消化液提取珠江口沉积物中生物可利用态Pb出现负值,认为可能是含有相对较高比例不稳定态Pb(可交换态和碳酸盐结合态)的沉积物中的Pb在消化液培养过程中能够溶解出来,但已经溶解出来的这些Pb随着培养时间的增加重新被沉积物中其他更强的结合态吸附,从而造成Pb的损失。本专利方法应用于珠江口沉积物实际案例说明牛血清白蛋白相对其它天然底栖生物肠液提取液来说更适用于沉积物中Pb的生物可利用性研究。
以上所述仅为本发明的优选实施例而已,并不用于限制本发明,对于本领域的技术人员来说,本发明可以有各种更改和变化。凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。
Claims (8)
1.一种同位素示踪沉积物重金属生物可利用性的ICP-MS法,其包括如下步骤:
(1)沉积物重金属总量的测定
(2)同位素标记示踪:通过合成不同铅同位素标记的沉积物中离子交换态铅(206Pb,以硝酸铅形式存在),将其添加到沉积物中
(3)生物可利用性提取:将沉积物样品用加入牛血清白蛋白溶液,震荡、萃取;
(4)用ICP-MS测定牛血清白蛋白中重金属含量和同位素含量;
以百分比含量表示的生物可利用态重金属(S,%)计算如下:
CB——以mg/kg表示的生物可利用态重金属;
CT——沉积物中重金属总量(mg/kg);
Cdissolved——提取后牛血清白蛋白提取液中溶解重金属的浓度(μg/L);
Cbackground——牛血清白蛋白提取液中的溶解重金属本底浓度(μg/L);
L——加入沉积物中进行培养的消化液体积(L);
m——提取试验中所用的沉积物质量(g)。
2.如权利要求1所述的ICP-MS法,所述牛血清白蛋白溶液的为BSA溶液,牛血清白蛋白溶液的浓度约为5g/L。
3.如权利要求1所述的ICP-MS法,所述离子交换态铅为206Pb,以硝酸铅形式存在。
4.如权利要求1所述的ICP-MS法,所述ICP-MS测定测定包括:将样品通过进样器经蠕动泵(Pump 1,提升转速0.5fps,稳定转速0.3rps)进入雾化器(Nebulizer)雾化成气溶胶进入矩管(Torch),在高温等离子体中原子化,再通过样品锥与截取锥,在离子透镜中带正电荷离子进行聚焦,通过四级杆电磁场根据质量/电荷比值进行偏转,进入检测器监测。
5.如权利要求1所述的ICP-MS法,所述四极杆质谱优化包括:利用标准调谐液优化仪器性能,控制89Y,70/140Ce++/Ce+双电荷指标,156/140CeO/Ce氧化物指标,等离子体稳定后,优化四级杆质谱,从而保证测定结果的精密度。
8.如权利要求1所述的ICP-MS法,所述ICP-MS测定还包括确定仪器的死时间:采用质量浓度分别为0、5、10、20、40、60ng/mL的GBW04424~27标准溶液对死时间进行测定,选择最为合适的死时间使仪器达到最佳信噪比。
