本发明公开了一种野葛嫩枝扦插的方法,属于矿山生态环境保护与恢复治理苗木类用材繁育技术领域;所述方法包括如下步骤:选材、浸泡、基质处理、扦插及扦插管理。本发明扦插种苗于两周后开始生根,生根速度快,根系数量众多,并着生根瘤,成活率较高,在较短时间内达到增产的目的。

本发明属于矿山生态环境保护与恢复治理苗木类用材繁育技术领域;,具体涉及一种野葛嫩枝扦插的方法。
背景技术
野葛属于豆科葛属的木质落叶藤本植物,在矿山生态环境保护与恢复治理中起着重要作用。其生长量大,攀援高度可达30米,匍匐地面甚至可达百米;其蔓叶重叠,郁闭度可达到100%;其根系发达而强大,深固在土壤中,可有效减轻雨水对地表的冲刷,防止水土流失。其耐旱、耐瘠薄性强;这些特性使其成为矿山生态修复优选的先锋植物之一,
野葛属于多年生豆科类藤本植物,葛根是葛属植物野葛的根茎,是《中华人民共和国药典》规定的正品药材,野葛根中的葛根总黄酮具有降血压、血糖、血脂、抗氧化、清除自由基、增强免疫力,抗癌、抗肿瘤等作用。以往葛根的商品供应主要依靠采集野生植株,过度采挖已导致野生资源日趋减少。而野葛作为一种适用于“白茬山”治理的优良攀援植物品种,矿山生态环境保护与恢复治理领域对其种苗的需求越来种苗越大,为避免采挖野生种苗对环境的破坏,批量快速地获得野葛种苗,服务于矿山生态环境保护与恢复治理领域,寻找一种高效便捷的野葛育苗技术方法就显得十分必要。
发明内容
为了解决上述背景技术中的问题,本发明提供了一种野葛嫩枝扦插的方法,扦插后两周即可生根,生根粗壮均一,根系数量众多,提高了后期葛根的产量和品种。
为实现上述目的,本发明提供如下技术方案:
一种野葛嫩枝扦插的方法,包括如下步骤:
S1、选材:选取2年生以上的野葛母株,取其当年生新梢直径5 mm半木质化枝条作为作为扦插条,插条长度控制在25-30 cm,并保留2个茎节以上,1片小叶,基部节下部分保留2-3 cm长,并剪成45-60度角斜面;
S2、浸泡:将剪取的插条在浸泡液A里面浸泡0.5-1h,取出,洗净沥干,然后将插条的形态学下端在浸泡液B中浸泡1 h,取出;
S3、基质处理:在自动喷灌棚内将混合基质整理成20-40cm厚的扦插床,用0.05-0.2%的高锰酸钾均匀喷灌基质,罩上塑料膜,密闭12-48h;
S4、扦插:扦插前,用清水喷淋基质后,使用木棍在基质上插孔,再将处理好的插条的1/3插入已准备好扦插床内。
S5、扦插管理:扦插完成后,保持扦插床湿度75-85%,温度28℃以下,每隔1周用800-1000倍多菌灵和0.05-0.2%高锰酸钾进行交替灭菌,每隔两周除1次杂草。
进一步地,所述浸泡液A为900-1200倍多菌灵溶液,优选地,所述浸泡液A为1000倍多菌灵溶液。
进一步地,所述浸泡液B由以下原料制备而成:萘乙酸 80-120 mg/L、青霉素80-120 mg/L、硼0.2-0.5 mg/L、维生素C15-30 mg/L、余量为水。
优选地,所述浸泡液B以下原料制备而成:萘乙酸100 mg/L、青霉素100 mg/L、硼0.3 mg/L、维生素C20 mg/L、余量为水。
进一步地,所述基质由河沙、蛭石、草炭土组成,各组分体积比为:V河沙:V蛭石:V草炭土=1:1:1。
本发明具有以下有益效果:
1、本发明提供的野葛嫩枝扦插的方法具有操作简单,处理时间短,在市场应用上更具优势。
2、本发明提供的野葛嫩枝扦插的方法在扦插前先将基质浇透水,用木棍在基质上插孔,减少插条在插入基质时对生根部位进行的机械损伤,提高和保证了扦插种苗的生根率和成活率。
3、本发明提供的野葛嫩枝扦插的方法在选材时选择当年生新梢直径5 mm的半木质化枝条,生长健壮,组织充实,腋芽饱满,更能快速收获成熟大苗。
4、浸泡液A对扦插种苗具有消毒杀菌的能力,浸泡液B不仅可以为切口进行消毒杀菌,还能促进插条快速生根,进一步提高和保证了扦插种苗的生根率和成活率。
5、本发明提供的野葛嫩枝扦插的方法,扦插种苗生根速度快,生根粗壮均一,根系数量众多,并着生根瘤,在根瘤内,根瘤菌从豆科植物根的皮层细胞中吸取碳水化合物、矿质盐类及水分,以进行生长和繁殖,同时他们又把游离的氮通过固氮作用固定下来,转变为植物所能利用的含氮化合物,供植物所需,这样,根瘤菌与根就形成了互相依赖的共生关系,提高了后期葛根的产量和质量。

以下通过具体实施例进一步说明本发明,但本领域技术人员应当知晓本发明的具体实施例并不以任何方式限制本发明,且在本发明基础上所作出的任何等同替换均落入本发明的保护范围。
实施例1:
一种野葛嫩枝扦插的方法,包括以下步骤:
S1、选材:选取2年生以上的野葛母株,取其当年生新梢直径5 mm半木质化枝条作为作为扦插条,插条长度控制在25 cm,并保留2个茎节以上,1片小叶,基部节下部分保留3cm长,剪成45度角斜面;
S2、浸泡:将剪取的插条在浸泡液A里面浸泡0.5 h,取出,洗净沥干,然后将插条的形态学下端在浸泡液B中浸泡1h,取出;其中,浸泡液A由为1000倍多菌灵溶液,浸泡液B由以下原料制备而成:萘乙酸 100 mg/L,青霉素100 mg/L,硼0.3 mg/L,维生素C20 mg/L。
S3、基质处理:在自动喷灌棚内将混合基质整理成30cm厚的扦插床,用0.1%的高锰酸钾均匀喷灌基质,罩上塑料膜,密闭24h后,对基质进行彻底消毒;所述基质由河沙、蛭石、草炭土组成,各组分体积比为:V河沙:V蛭石:V草炭土=1:1:1。
S4、扦插:扦插前,用清水喷淋基质后,使用木棍在基质上插孔,再处理好的插条的1/3插入已准备好苗床内。
S5、扦插管理:扦插完成后,保持扦插床湿度75-85%,温度28℃以下,每隔1周用800-1000倍多菌灵和0.05-0.2%高锰酸钾进行交替灭菌,每隔两周除1次杂草。
实施例2:
一种野葛嫩枝扦插的方法,包括以下步骤:
S1、选材:选取2年生以上的野葛母株,取其当年生新梢直径5 mm半木质化枝条作为作为扦插条,插条长度控制在30 cm,并保留2个茎节以上,1片小叶,基部节下部分保留2cm长,剪成60度角斜面;
S2、浸泡:将剪取的插条在浸泡液A里面浸泡1 h,取出,洗净沥干,然后将插条的形态学下端在浸泡液B中浸泡1.5 h,取出;其中,浸泡液A由为1200倍多菌灵溶液,浸泡液B由以下原料制备而成:萘乙酸120 mg/L、青霉素120 mg/L、硼0.4 mg/L、维生素C30mg/L。
S3、基质处理:在自动喷灌棚内将混合基质整理成30cm厚的扦插床,用0.15%的高锰酸钾均匀喷灌基质,罩上塑料膜,密闭36h;对基质进行彻底消毒;所述基质由河沙、蛭石、草炭土组成,各组分体积比为:V河沙:V蛭石:V草炭土=1:1:1。
S4、扦插:扦插前,用清水喷淋基质后,使用木棍在基质上插孔,再处理好的插条的1/3插入已准备好苗床内。
S5、扦插管理:扦插完成后,保持扦插床湿度75-85%,温度28℃以下,每隔1周用800-1000倍多菌灵和0.05-0.2%高锰酸钾进行交替灭菌,每隔两周除1次杂草。
验证试验:
试验组1 :
浸泡液A换成纯净水,其余步骤与实施例1完全相同。
试验组2 :
浸泡液B换成纯净水,其余步骤与实施例1完全相同。
试验组3 :
浸泡液A和浸泡液B同时换成纯净水,其余步骤与实施例1完全相同:
试验组4:
扦插前,不进行处理,直接将处理好的野葛种苗扦插,其余步骤与实施例1完全相同。

Claims (6)

1.一种野葛嫩枝扦插的方法,其特征在于,包括如下步骤:
S1、选材:
选取2年生以上的野葛母株,取其当年生新梢直径5mm半木质化枝条作为扦插条,插条长度控制在25-30 cm,并保留2个以上茎节,1片小叶,基部节下部分保留2-3cm长,并剪成45-60度角斜面;
S2、浸泡:
将剪取的插条在浸泡液A里面浸泡0.5-1h,取出,洗净沥干,然后将插条的形态学下端在浸泡液B中浸泡1h,取出;
S3、基质处理:
在自动喷灌棚内将混合基质整理成20-40cm厚的扦插床,用0.05-0.2%的高锰酸钾均匀喷灌基质,罩上塑料膜,密闭12-48h;
S4、扦插:
扦插前,用清水喷淋基质后,使用木棍在基质上插孔,再将处理好的插条的1/3插入已准备好的扦插床内;
S5、扦插后管理:
扦插完成后,保持扦插床湿度75-85%,温度28℃以下,每隔1周用800-1000倍多菌灵和0.05-0.2%高锰酸钾进行交替灭菌,每隔两周除1次杂草。
2.根据权利要求1所述的野葛嫩枝扦插的方法,其特征在于,所述浸泡液A为900-1200倍多菌灵溶液。
3.根据权利要求2所述的野葛嫩枝扦插的方法,其特征在于,所述浸泡液A为1000倍多菌灵溶液。
4.根据权利要求1所述的野葛嫩枝扦插的方法,其特征在于,所述浸泡液B由以下原料制备而成:萘乙酸80-120mg/L、青霉素80-120mg/L、硼0.2-0.5mg/L、维生素C15-30mg/L,余量为水。
