Abstract
A ready-to-drink burdock bird's nest peptide liquid compound beverage easy to digest comprises the following raw materials in parts by weight: 20-40 parts of burdock enzymolysis liquid, 20-30 parts of cubilose peptide liquid, 15-25 parts of tremella extract, 10-25 parts of pear juice, 10-20 parts of rose extract and 0.1-0.2 part of carrageenan. The preparation method comprises the following steps: preparing burdock enzymolysis liquid and bird's nest peptide liquid; proportionally compounding the burdock enzymolysis liquid, the bird's nest peptide liquid, the tremella extract, the pear juice, the rose extract and the carrageenan, homogenizing, uniformly mixing, filling and sterilizing to obtain the compound beverage. The method can simplify the treatment steps, reduce the probability of microbial contamination and the requirements on equipment, reduce energy consumption, improve the use amount of the enzyme, reduce the variety of the used enzyme and effectively degrade the protein; the prepared beverage retains the flavor of fresh burdock, and has low carbon water content, rich nutrient components and easy absorption by human body.

Technical Field
The invention relates to the technical field of beverages, in particular to a ready-to-drink burdock bird's nest peptide liquid composite beverage easy to digest and a preparation method thereof, and belongs to the technical field of food processing.
Background
The burdock is a feverfew of Campanulales, has rich nutritive value, contains rich protein, amino acid, cellulose, vitamin and various mineral elements, and also contains special components such as inulin and chlorogenic acid; in addition, the burdock contains rich soluble fiber, which is the main component of burdock pharmacological activity, and has the effects of promoting development, promoting intestinal flora balance, inhibiting tumor growth, resisting bacteria, and the like. The burdock has high fiber content, and is easy to age if not preserved properly after being harvested and unearthed, so that the original taste and flavor are lost. Therefore, the quality loss caused by the sale process can be well avoided by developing the burdock product. The developed products of the burdock beverage are relatively few, the burdock tea is the most common burdock tea, and the dried burdock sliced tea has the problems of strong earthy taste, poor taste, inconvenience for drinking after being soaked in boiling water and the like. The other types of burdock beverages and burdock fermented beverages are basically prepared by simply boiling and extracting burdock with water and mixing the burdock with red dates, longan, milk, Chinese herbal medicine raw materials and the like, and the burdock beverages have the problems that the flavor of fresh burdock is basically covered, the carbohydrate content is too high, the digestion rate is low, the nutrient content is single and the burdock fermented beverage is difficult to absorb and the like.
The edible bird's nest mainly comprises water, ash, fat, protein and saccharide, and comprises five kinds of semi-essential amino acids and six kinds of essential amino acids. The carbohydrate and protein in the cubilose usually exist in a form of combined glycoprotein, the cubilose mainly contains five monosaccharides or monosaccharide derivatives, wherein N-acetyl neuraminic acid (sialic acid) is a monosaccharide conversion product, the sialic acid can promote and improve the nervous system and the immune system of a human body after being absorbed by the human body, and the content of the sialic acid accounts for about 10 percent of the cubilose, which is difficult to achieve in other purely natural foods. The protein in the cubilose is subjected to soaking and stewing, and a large amount of insoluble protein exists. After simulating gastrointestinal tract digestion, protein fragment electrophoresis analysis shows that the digested supernatant has over 50% of high molecular protein incapable of being degraded. This indicates that although the protein content in the edible bird's nest is high, some protein can not be absorbed by human body, which causes great waste to the edible bird's nest which is a precious food raw material. Although the literature has already carried out research reports in this respect, most of the existing research aims at extracting and purifying certain specific components of the cubilose and then applying the components, but does not reserve the full-spectrum nutritional components in the cubilose; in addition, the preparation method of the bird's nest related products also has the problems of excessive treatment steps, high probability of microbial contamination, higher equipment requirement, high energy consumption, obviously lower enzyme dosage, too wide enzyme variety, incapability of effectively degrading proteins and the like.
At present, a plurality of relevant documents about burdock beverages or cubilose products are reported, but no beverage prepared by compounding burdock and cubilose is reported.
Disclosure of Invention
The invention aims to provide a ready-to-drink burdock bird's nest peptide liquid composite beverage easy to digest and a preparation method thereof, the method can simplify the treatment steps, reduce the probability of microbial contamination and the requirements on equipment, reduce energy consumption, improve the use amount of enzyme and reduce the variety of the used enzyme, and can effectively degrade protein; the prepared beverage retains the flavor of fresh burdock, and has low carbon water content, rich nutrient components and easy absorption by human body.
In order to achieve the purpose, the invention provides a ready-to-drink burdock bird's nest peptide liquid compound drink which is easy to digest and comprises the following raw materials in parts by weight: 20-40 parts of burdock enzymolysis liquid, 20-30 parts of cubilose peptide liquid, 15-25 parts of tremella extract, 10-25 parts of pear juice, 10-20 parts of rose extract and 0.1-0.2 part of carrageenan.
The invention also provides a preparation method of the ready-to-drink burdock bird's nest peptide liquid composite beverage easy to digest, which comprises the following steps:
(1) preparing burdock enzymolysis liquid;
(1-1) cleaning and peeling fresh burdock roots, and removing squeezed damaged and rotten tissues;
(1-2) cutting the burdock roots into slices with the thickness of 2-3mm, and soaking the slices in a color fixative;
(1-3) fishing out the burdock slices from the color fixative, draining, soaking in boiling water for blanching for 80-120s, fishing out, cooling and draining;
(1-4) adding pure water into the burdock slices to obtain a mixture, wherein the mass ratio of the burdock slices to the pure water is 1 (15-25), adding a complex enzyme according to the proportion of 1-3% of the mass of the mixture for stirring and enzymolysis, the complex enzyme comprises cellulase and papain, and the mass ratio of the cellulase to the papain is (3-1): 1, carrying out enzymolysis at the temperature of 40-50 ℃ for 2-5h, filtering and collecting the filtrate I through 300-mesh and 400-mesh filter cloth after the enzymolysis is finished, and boiling the filtrate I for 20min to inactivate enzyme to obtain an enzymolysis liquid;
(1-5) adding pure water into the filter residue obtained by filtering in the step (1-4), wherein the mass ratio of the filter residue to the pure water is 1: (5-10), extracting in a boiling water bath for 2-3h, filtering and collecting through 300-mesh filter cloth with 400 meshes to obtain a filtrate II, and combining the filtrate II with the enzymatic hydrolysate obtained in the step (1-4) to obtain the burdock enzymatic hydrolysate;
(2) preparing bird's nest peptide liquid;
(2-1) drying and grinding dry swallow marigold, sieving with a 80-100 mesh sieve to obtain dry swallow marigold powder, adding pure water into the dry swallow marigold powder to obtain a mixed solution, wherein the mass ratio of the dry swallow marigold powder to the pure water is 1: (40-60), extracting the mixed solution in bath water at 70-90 ℃ for 3-6h, and cooling to obtain a cubilose colloid;
(2-2) adding flavourzyme into the cubilose colloid according to the mass fraction of 2-3% for enzymolysis, wherein the pH value is 6-8, the temperature is 35-55 ℃, the time is 3-4h, then boiling for 20min for enzyme deactivation to obtain an enzymolysis liquid, and ultrafiltering the enzymolysis liquid by an ultrafiltration membrane to collect a micromolecular protein and polypeptide solution with the KD or less;
(2-3) adding neutral protease into the macromolecular protein intercepted by ultrafiltration of an ultrafiltration membrane in the step (2-2) according to the mass fraction of 2-3%, adjusting the pH to 6.5-8, adjusting the enzymolysis temperature to 40-55 ℃, carrying out enzymolysis for 3-4h, boiling for 20min to inactivate enzyme to obtain an enzymolysis liquid, and mixing the enzymolysis liquid with the micromolecular protein and polypeptide solution collected in the step (2-2) to obtain an enzymolysis bird's nest peptide liquid;
(3) compounding the burdock enzymolysis liquid, the bird's nest peptide liquid, the tremella extract, the pear juice, the rose extract and the carrageenan according to the proportion, homogenizing, mixing uniformly, filling and sterilizing to obtain the burdock bird's nest peptide liquid compound beverage.
Further, the preparation method of the tremella extract comprises the following steps: removing roots and impurities from tremella, crushing to obtain tremella powder particles, adding pure water into the tremella powder particles to obtain mixed liquid, wherein the mass ratio of the tremella powder particles to the pure water is 1: (80-100), extracting the mixed solution in boiling water for 1.5-2h, and finally filtering and collecting the tremella extract through 400-mesh filter cloth.
Further, after pure water is added into the tremella powder particles to obtain a mixed solution, sodium bicarbonate is added into the mixed solution, and the mass of the sodium bicarbonate is 1-2% of that of the tremella powder particles.
Further, the preparation method of the pear juice comprises the following steps: the method comprises the following steps of sequentially cleaning, peeling, removing bad spots and insect eyes, removing stones and cutting pears into blocks, and adding pure water into the pear blocks to obtain a mixture, wherein the mass ratio of the pear blocks to the pure water is 1: (2-4), boiling the mixture in boiling water for 10min to inactivate enzyme, pulping, filtering through filter cloth with 300 meshes and 400 meshes, and collecting to obtain the pear juice.
Preferably, the pear is one of Dangshan pear, duck pear and Korla pear.
Further, the preparation method of the rose leaching solution comprises the following steps: removing sepals, flower stems and leaves from the dried rose flowers, and adding pure water into the dried rose flowers to obtain a mixture, wherein the mass ratio of the dried rose flowers to the pure water is 1: (8-15), extracting the mixture at 70-85 deg.C for 30-50min, and filtering with 100 mesh filter cloth to obtain rose leachate.
Preferably, in the step (1-2), the color fixative is a citric acid solution with a mass fraction of 0.7-1.1% or a mixed solution of a citric acid solution and a vitamin C solution with a mass fraction of 0.4-0.75%.
Compared with the prior art, the invention has the following advantages:
(1) in the burdock extracting solution provided by the invention, the content of the inulin can reach 11.45%, the content of the chlorogenic acid can reach 0.25mg/mL, the bioactive components in the burdock are well reserved, and the water-soluble fiber content of the product is improved;
(2) the cubilose used by the invention can be a complete cubilose, or a cubilose strip or cubilose fragment, and no quality and sensory influence is caused on the product no matter which cubilose is used, so that the utilization rate of the cubilose is better improved;
(3) compared with the common stewing method, part of macromolecular protein of the cubilose peptide liquid prepared by the preparation method is degraded into small-fragment peptide or amino acid, so that the cubilose peptide liquid is more favorable for digestion and absorption; after the provided bird's nest enzymatic hydrolysate is detected, the protein is obviously degraded, the polypeptide content is increased from 0.85mg/mL to 1.5mg/mL, the amino acid content is increased from 8.3 mu g/mL to 65.4 mu g/mL, the sialic acid content is not obviously changed, and the most important component, namely sialic acid, in the bird's nest is better reserved;
(4) the compound beverage prepared by the invention is rich in various substances beneficial to human bodies, such as polysaccharides, proteins, polypeptides, amino acids, dietary fibers, chlorogenic acid and the like, is easy to digest and absorb, has a hydroxyl radical clearance rate of over 41.6 percent, a DPPH radical clearance rate of over 80.6 percent and an ABTS radical clearance rate of over 59.3 percent, has an obvious antioxidant effect and has a good health-care function.
The invention selects enzymes which can efficiently hydrolyze protein and fiber components to prepare burdock enzymolysis liquid and cubilose peptide liquid, hydrolyzes burdock and cubilose into components which are easy to absorb, and then prepares the compound health-care beverage by matching with other raw materials. The invention simplifies the treatment steps, reduces the probability of microbial contamination and the requirements on equipment, reduces energy consumption, improves the use amount of enzyme, reduces the variety of the used enzyme, and can effectively degrade protein; the prepared beverage keeps the flavor of fresh burdock, has lower carbohydrate content, low sugar content, fresh and sweet taste, more easily accepted flavor and rich nutrient components; in addition, the comparison of the change before and after the digestion simulation of the drink shows that the sialic acid in the drink is obviously reduced and the content of the amino acid is obviously increased after the drink is digested, and the amino acid serving as the minimum amino-containing component unit in a living body can be directly absorbed by the human body. All components except the polysaccharide are degraded to different degrees, so that the content of small molecular components such as amino acid and the like is increased, the components are easier to digest and absorb, and the polysaccharide has good market application value.

Example one
A preparation method of a ready-to-drink burdock bird's nest peptide liquid compound beverage easy to digest comprises the following steps:
(1) preparing a burdock extracting solution;
(1-1) selecting fresh burdock, wherein the skin is relatively complete, the damage is not more than two parts, and the damaged area is less than the area of 1 yuan of coin; cleaning fresh radix Arctii, peeling, and removing squeezed damaged and rotten tissue;
(1-2) cutting burdock roots into slices with the thickness of 3mm, immediately soaking the slices in citric acid with the mass fraction of 0.8% for color protection treatment to prevent oxidation discoloration;
(1-3) fishing out the burdock slices from the color fixative, draining, immediately immersing into boiling water for blanching for 120s after draining, fishing out, cooling and draining, wherein polyphenol oxidase in the burdock is inactivated and the burdock has good color and luster;
(1-4) adding pure water into the burdock slices to obtain a mixture, wherein the mass ratio of the burdock slices to the pure water is 1: 25, adding a complex enzyme according to the proportion of 3% of the mass of the mixture, stirring and performing enzymolysis, wherein the complex enzyme is cellulase and papain, and the mass ratio of the cellulase to the papain is 1:1, carrying out enzymolysis at 40 ℃ for 5h, continuously and slowly stirring, filtering and collecting filtrate I through 400-mesh filter cloth after enzymolysis is finished, boiling the filtrate I for 20min, and inactivating enzyme to obtain an enzymolysis solution;
(1-5) adding pure water into the filter residue obtained by filtering in the step (1-4), wherein the mass ratio of the filter residue to the pure water is 1: 5, extracting in a boiling water bath for 2 hours, filtering and collecting through 400-mesh filter cloth to obtain a filtrate II, and combining the filtrate II with the enzymatic hydrolysate obtained in the step (1-4) to obtain the burdock enzymatic hydrolysate;
(2) preparing bird's nest peptide liquid;
(2-1) drying and grinding dry swallow marigold, sieving with a 80-mesh sieve to obtain dry swallow marigold powder, and adding pure water into the dry swallow marigold powder to obtain a mixed solution, wherein the mass ratio of the dry swallow marigold powder to the pure water is 1: 60, extracting the mixed solution in bath water at 70 ℃ for 6 hours, and cooling to obtain a cubilose colloid;
(2-2) adding flavourzyme into the cubilose colloid according to the mass fraction of 2% for enzymolysis, adjusting the pH to 6, controlling the temperature to 55 ℃, keeping the time for 3 hours, boiling for 20min to inactivate enzyme to obtain an enzymolysis liquid, and ultrafiltering the enzymolysis liquid by an ultrafiltration membrane to collect micromolecular protein and polypeptide solution with the KD or less;
(2-3) adding neutral protease into the macromolecular protein intercepted by ultrafiltration of an ultrafiltration membrane in the step (2-2) according to the mass fraction of 2%, adjusting the pH to 6.5, adjusting the enzymolysis temperature to 55 ℃, carrying out enzymolysis for 3 hours, boiling for 20min to inactivate enzyme to obtain an enzymolysis liquid, and mixing the enzymolysis liquid with the micromolecular protein and polypeptide solution collected in the step (2-2) to obtain an enzymolysis bird's nest peptide liquid;
(3) preparing a composite beverage;
taking 40 parts of burdock extract, 20 parts of bird's nest peptide liquid, 10 parts of pear juice, 20 parts of rose extract and 15 parts of tremella extract as raw materials, mixing and compounding the raw materials in proportion to obtain mixed liquid, adding 0.2 part of carrageenan into the mixed liquid, homogenizing the mixed liquid at the pressure of 20MPa and the temperature of 40 ℃, keeping the mixed liquid at the temperature of 121 ℃ for 10min, sterilizing and filling the mixed liquid, and packaging the mixed liquid to obtain the burdock bird's nest peptide liquid composite beverage. The beverage is in a light yellow semitransparent state, has no obvious precipitate after standing within 14 days, has fresh and slightly sweet taste, slightly has the special protein flavor of cubilose, has no obvious bitter and earthy taste of burdock, has certain fruit and rose fragrance, and has smooth mouthfeel and no sticky feeling.

(1-3) fishing out the burdock slices from the color fixative, draining, immediately immersing into boiling water for blanching for 80s after draining, fishing out, cooling and draining, wherein polyphenol oxidase in the burdock is inactivated and the burdock has good color and luster;

The beverage has palatable sweetness, obvious fruit fragrance, slight specific protein flavor of edible bird's nest, no bitter and astringent taste and earthy taste of burdock, and refreshing taste.
The preparation method of the pear juice comprises the following steps: selecting fresh pear with complete surface and rot and mildew condition not exceeding 1 yuan coin area, sequentially cleaning pear, peeling, removing necrotic spots and insect eyes, removing core, and cutting into 1cm3Adding pure water into the pear blocks to obtain a mixture, wherein the mass ratio of the pear blocks to the pure water is 1: and 4, boiling the mixture in boiling water for 10min to inactivate enzyme, cooling and pulping, and filtering and collecting through filter cloth with 300 meshes and 400 meshes to obtain the pear juice.

The preparation method of the rose leaching solution comprises the following steps: removing sepals, flower stems and leaves from the dried rose flowers, and adding pure water into the dried rose flowers to obtain a mixture, wherein the mass ratio of the dried rose flowers to the pure water is 1: and 8, extracting the mixture at 85 ℃ for 30min, and finally filtering and collecting the rose leachate through a filter cloth of 100 meshes.
And (3) carrying out a digestion simulation experiment on the composite beverage, and detecting the change condition of 7 indexes, namely the total sugar content, the sialic acid content, the protein content, the polypeptide content, the amino acid content, the chlorogenic acid content and the soluble solid content in the sample before and after digestion by using the composite beverage prepared in the first embodiment.
1. Test method
(1) Simulated gastric fluid digestion experiment steps: taking 250mL of beverage in a conical flask, adjusting the pH value of the sample to 1.3 by using 1mol/L HCl after the beverage is put in a water bath at 37 ℃, adding 250mL of simulated gastric juice, digesting the sample in a constant-temperature water bath at 37 ℃ for 1h in a dark condition, and putting the sample in a water bath at 70 ℃ for 5min to inactivate after the digestion is finished.
(2) Simulating intestinal juice digestion experiment steps: 250mL of a sample after simulated gastric digestion was taken, adjusted to pH 6.8 with 1mol/L sodium hydroxide, and 250mL of intestinal fluid was added. Continuously placing in a 37 deg.C water bath for 3 hr under dark condition, placing the sample in 70 deg.C water bath for 5min, cooling, centrifuging at 10000r/min for 6min, and collecting supernatant for determination.
2. Test results
The simulated changes of total sugar, sialic acid and soluble solid before and after digestion are respectively shown in figure 1, figure 2 and figure 3, and the simulated changes of protein, polypeptide, amino acid and chlorogenic acid before and after digestion are shown in figure 4. The results in the figure show a significant decrease in sialic acid and a significant increase in amino acid content. Amino acids, which are the smallest amino group-containing unit in the living body, are directly absorbed by the human body. All components except the polysaccharide are degraded to different degrees, so that the content of small molecular components such as amino acid and the like is increased, and the components are easier to digest and absorb. Although the polysaccharide is not obviously degraded, the research results show that the macromolecular structure of the components has obvious correlation with the immunity improvement of human bodies and has special effects except nutrition.
Claims (8)
1. The ready-to-drink burdock bird nest peptide liquid compound beverage easy to digest is characterized by comprising the following raw materials in parts by weight: 20-40 parts of burdock enzymolysis liquid, 20-30 parts of cubilose peptide liquid, 15-25 parts of tremella extract, 10-25 parts of pear juice, 10-20 parts of rose extract and 0.1-0.2 part of carrageenan.
2. The preparation method of the ready-to-drink digestible burdock bird's nest peptide liquid compound drink according to claim 1, is characterized by comprising the following steps:
(1) preparing burdock enzymolysis liquid;
(1-1) cleaning and peeling fresh burdock roots, and removing squeezed damaged and rotten tissues;
(1-2) cutting the burdock roots into slices with the thickness of 2-3mm, and soaking the slices in a color fixative;
(1-3) fishing out the burdock slices from the color fixative, draining, soaking in boiling water for blanching for 80-120s, fishing out, cooling and draining;
(1-4) adding pure water into the burdock slices to obtain a mixture, wherein the mass ratio of the burdock slices to the pure water is 1 (15-25), adding a complex enzyme according to the proportion of 1-3% of the mass of the mixture for stirring and enzymolysis, the complex enzyme comprises cellulase and papain, and the mass ratio of the cellulase to the papain is (3-1): 1, carrying out enzymolysis at the temperature of 40-50 ℃ for 2-5h, filtering and collecting the filtrate I through 300-mesh and 400-mesh filter cloth after the enzymolysis is finished, and boiling the filtrate I for 20min to inactivate enzyme to obtain an enzymolysis liquid;
(1-5) adding pure water into the filter residue obtained by filtering in the step (1-4), wherein the mass ratio of the filter residue to the pure water is 1: (5-10), extracting in a boiling water bath for 2-3h, filtering and collecting through 300-mesh filter cloth with 400 meshes to obtain a filtrate II, and combining the filtrate II with the enzymatic hydrolysate obtained in the step (1-4) to obtain the burdock enzymatic hydrolysate;
(2) preparing bird's nest peptide liquid;
(2-1) drying and grinding dry swallow marigold, sieving with a 80-100 mesh sieve to obtain dry swallow marigold powder, adding pure water into the dry swallow marigold powder to obtain a mixed solution, wherein the mass ratio of the dry swallow marigold powder to the pure water is 1: (40-60), extracting the mixed solution in bath water at 70-90 ℃ for 3-6h, and cooling to obtain a cubilose colloid;
(2-2) adding flavourzyme into the cubilose colloid according to the mass fraction of 2-3% for enzymolysis, wherein the pH value is 6-8, the temperature is 35-55 ℃, the time is 3-4h, then boiling for 20min for enzyme deactivation to obtain an enzymolysis liquid, and ultrafiltering the enzymolysis liquid by an ultrafiltration membrane to collect a micromolecular protein and polypeptide solution with the KD or less;
(2-3) adding neutral protease into the macromolecular protein intercepted by ultrafiltration of an ultrafiltration membrane in the step (2-2) according to the mass fraction of 2-3%, adjusting the pH to 6.5-8, adjusting the enzymolysis temperature to 40-55 ℃, carrying out enzymolysis for 3-4h, boiling for 20min to inactivate enzyme to obtain an enzymolysis liquid, and mixing the enzymolysis liquid with the micromolecular protein and polypeptide solution collected in the step (2-2) to obtain an enzymolysis bird's nest peptide liquid;
(3) compounding the burdock enzymolysis liquid, the bird's nest peptide liquid, the tremella extract, the pear juice, the rose extract and the carrageenan according to the proportion, homogenizing, mixing uniformly, filling and sterilizing to obtain the burdock bird's nest peptide liquid compound beverage.
3. The preparation method of the ready-to-drink burdock edible bird's nest peptide liquid composite drink easy to digest according to claim 2 is characterized in that the preparation method of the white fungus extract liquid is as follows: removing roots and impurities from tremella, crushing to obtain tremella powder particles, adding pure water into the tremella powder particles to obtain mixed liquid, wherein the mass ratio of the tremella powder particles to the pure water is 1: (80-100), extracting the mixed solution in boiling water for 1.5-2h, and finally filtering and collecting the tremella extract through 400-mesh filter cloth.
