技术领域
本发明涉及生物医药技术领域,具体涉及一种复合纳米颗粒、制备方法、应用。
背景技术
免疫调节物,通过协调人体免疫系统来消灭癌细胞,癌症免疫治疗领域取得了重大进展。其中一种重要的免疫调节物,如模式识别受体(PRRs)是一种基因编码的宿主传感器,它可以启动先天免疫信号通路,导致促炎细胞因子的产生,从而指导适应性免疫。然而,此类免疫刺激药物都具有高毒性,它们都对正常的细胞具有杀伤力,此外,还具有水溶性差及产生不良的药代动力学,这些缺点限制了此类免疫刺激药物在癌症免疫治疗中的直接应用。
纳米技术,在癌症免疫治疗方面,具有许多优势,如向免疫细胞靶向传递,可增强临床效果,减少不良事件的发生,这将有助于癌症疫苗和免疫调节剂的递送。迄今为止,各种纳米结构已被用来运载各种分子货物的载体,它能够稳定运载货物的“生物活性”,增加货物在体液中的溶解度,同时,减少系统性副作用的产生。因此,基于纳米颗粒的靶向给予模式识别受体激动剂无疑是抗肿瘤治疗的重要途径。
发明内容
有鉴于此,本发明提供一种复合纳米颗粒,以期解决上述技术问题。
为实现上述技术目的,本发明提供一种复合纳米颗粒,包括:脂质体囊泡包裹的介孔硅纳米颗粒;
其中,上述介孔硅纳米颗粒的表面修饰有第一模式识别受体激动剂和第二模式识别受体激动剂;
上述脂质体囊泡由二油酰磷脂酰胆碱、(2,3-二油酰基-丙基)三甲胺和式(I)所示的化合物A组成;
根据本发明实施例,其中,上述第一模式识别受体激动剂包括以下任意一种:

将上述负电性的纳米颗粒与脂质体囊泡混合,得到复合纳米颗粒。
根据本发明实施例,其中,上述将介孔硅纳米颗粒与第一模式识别受体激动剂反应,得到修饰正电性的纳米颗粒,包括:
将第一模式识别受体激动剂、氯硅烷和有机碱,在无水无氧及的条件下混合反应10~60min后,得到第一混合溶液;

根据本发明实施例,脂质体囊泡包裹介孔硅纳米颗粒形成的复合纳米颗粒,由于介孔硅纳米颗粒表面修饰有两种模式识别受体激动剂,可以对肿瘤部位微酸环境响应,从而将两种模式识别受体激动剂同时靶向递送至肿瘤部位,在静脉给药后,由于实体瘤增强渗透滞留效应以及配体驱动的主动靶向效应,会优先在肿瘤部位积聚,并通过受体介导的内吞作用进入肿瘤微环境的抗原呈递细胞内,脂质体囊泡在酸性条件下的不稳定促进了pH触发的模式识别受体激动剂从内体释放,两种模式识别受体激动剂一同发挥作用,协同激活免疫系统;利用纳米技术和免疫调节物的结合,纳米结构作为运载分子的载体,利用纳米结构具有靶向传递的优点,可稳定运载货物,保证生物活性,减少副作用的发生,增强临床效果,减少不良事件的发生,具有重要的应用价值。

相关技术中的免疫调节物,如模式识别受体,它们都具有高毒性,杀伤正常细胞,水溶性差,产生不良药代动力力学等副作用,限制了免疫刺激药物在癌症免疫治疗中的应用。
因此,本发明提供了一种复合纳米颗粒,包括:脂质体囊泡包裹的介孔硅纳米颗粒;其中,上述介孔硅纳米颗粒的表面修饰有第一模式识别受体激动剂和第二模式识别受体激动剂;上述脂质体囊泡由二油酰磷脂酰胆碱、(2,3-二油酰基-丙基)三甲胺和式(I)所示的化合物A组成;

本发明实施例中,复合纳米颗粒可以对肿瘤部位微酸环境响应,从而将两种模式识别受体激动剂同时靶向递送至肿瘤部位。其中,介孔硅纳米颗粒能够抑制脂质体不稳定的双层波动,而脂质体囊泡的包覆能够减少介孔硅负载的药物在运输过程中的泄露;通过在介孔硅纳米颗粒表面修饰硅烷醇基团与氯硅烷反应键接第一模式识别受体激动剂,可以实现其酸响应释放,同时第一模式识别受体激动剂将介孔硅的表面修饰为正电荷,可以通过静电作用吸附另一种负电性的第二模式识别受体激动剂。此外,脂质体囊泡用聚乙二醇长链和甘露糖肿瘤靶向配体进行修饰,可以增加纳米颗粒在体内的循环时间以及实现肿瘤部位抗原呈递细胞的靶向和内吞。
具体地,通过以上方法得到的复合纳米颗粒在静脉给药后,由于实体瘤增强渗透滞留效应以及配体驱动的主动靶向效应,会优先在肿瘤部位积聚,并通过受体介导的内吞作用进入肿瘤微环境的抗原呈递细胞内,脂质体囊泡在酸性条件下的不稳定促进了pH触发的从内体释放,第一模式识别受体激动剂与第二模式识别受体激动剂一同发挥作用,协同激活免疫系统。

作为本发明的另一个方面,本发明还提供了复合纳米颗粒的制备方法,包括:将介孔硅纳米颗粒与第一模式识别受体激动剂反应,得到修饰正电性的纳米颗粒;
将上述正电性的纳米颗粒与第二模式识别受体激动剂混合,得到负电性的纳米颗粒;
将上述负电性的纳米颗粒与脂质体囊泡混合,得到复合纳米颗粒。
本发明实施例中,上述第一模式识别受体激动剂将介孔硅的表面修饰为正电荷,得到正电性的纳米颗粒;正电性的纳米颗粒通过静电作用吸附第二模式识别受体激动剂,得到负电性的纳米颗粒;
第一模式识别受体激动剂和第二模式识别受体激动剂,通常在抗原呈递细胞上表达,如树突状细胞和巨噬细胞,可以识别病原体相关分子模式和危险相关分子模式,启动先天免疫信号通路,导致促炎细胞因子的产生,从而指导适应性免疫。
根据本发明实施例,其中,上述将介孔硅纳米颗粒与第一模式识别受体激动剂反应,得到修饰正电性的纳米颗粒,包括:上述第一模式识别受体激动剂、氯硅烷和有机碱,在无水无氧的条件下,混合反应10~60min后得到第一混合溶液;再将介孔硅纳米颗粒加入上述第一混合溶液中,在无水无氧的条件下,反应1~4h,得到正电性的纳米颗粒;根据本发明的实施例,其中,有机碱包括N,N-二异丙基乙胺。
本发明实施例中,在无水无氧条件下,将氯硅烷分散在无水N,N-二甲基甲酰胺溶剂中,加入1%~2%的有机碱,保持反应条件为碱性,再加入第一模式识别受体激动剂,其与氯硅烷摩尔比例为1~3:1,例如,1~1:1、1~2:1、1~3:1,通过硅烷醇基团混合反应10~60min,例如,10min、30min、40min、60min;再加入介孔硅,通过其硅烷醇基团与氯硅烷的另一端的硅氯键反应1~4h,例如,1h、2h、3h、4h;反应结束后,用乙醚重复沉降、洗涤多次最终得到修饰了第一模式识别受体激动剂的正电性介孔硅纳米颗粒。本发明实施例中,上述氯硅烷包括:二氯二甲基硅烷、二氯二乙基硅烷、二氯二异丙基硅烷,其中:甲基、乙基、异丙基基团水解速率不同,会影响第一模式识别受体激动剂酸响应释放的速率。
本发明实施例中,上述无氧气氛可以为氮气气氛或者惰性气氛。
根据本发明实施例,其中,将上述正电性的纳米颗粒与第二模式识别受体激动剂混合,得到负电性的纳米颗粒,包括:将上述第二模式识别受体激动剂加入上述正电性的纳米颗粒中,混合搅拌0.5~4h,例如,0.5h、1h、2h、4h,得到负电性的纳米颗粒。

本发明实施例中,其中,上述预设时长包括24h。本发明实施例中,上述化合物A是由二硬脂酰基磷脂酰乙醇胺-聚乙二醇2000-N-羟基琥珀酰亚胺与4-氨基苯基-α-d-吡喃甘露糖苷通过交联反应制得的。具体为将氨基苯基-α-d-吡喃甘露糖苷溶于无水二甲基亚砜(DMSO)中20~60分钟,加入等比例二硬脂酰基磷脂酰乙醇胺-聚乙二醇2000-N-羟基琥珀酰亚胺反应24~48h,。通过使用截留分子量为1000Da的透析膜对反应混合物进行透析提纯48~72h,冻干后得到产物。
本发明实施例中,将二油酰磷脂酰胆碱、(2,3-二油酰基-丙基)-三甲胺、化合物A按照配比溶解在氯仿中,旋转蒸发去掉氯仿后形成一层薄膜;在薄膜中加入超纯水,超声5~30min,将得到的混合液通过200nm挤出器挤出得到脂质体囊泡;将脂质体囊泡与负电性纳米颗粒在500~1200rpm下,混合搅拌预设时长;反应结束后将混合物在1000~3000g,4℃条件下,用30k超滤管洗涤3-4次,得到脂质体囊泡。
作为本发明的另一个方面,本发明还提供了复合纳米颗粒在治疗癌症药物中的应用。
本发明提供的复合纳米颗粒在静脉给药后,由于实体瘤增强渗透滞留效应以及配体驱动的主动靶向效应,会优先在肿瘤部位积聚,并通过受体介导的内吞作用进入肿瘤微环境的抗原呈递细胞内,脂质体囊泡在酸性条件下的不稳定促进了pH触发的模式识别受体激动剂从内体释放,两种模式识别受体激动剂一同发挥作用,协同激活免疫系统。
下面结合具体实施例对本发明的技术方案作进一步说明,但需要注意的是,下述的实施例仅用于说明本发明的技术方案,但本发明并不限于此。
实施例1
介孔硅(MSNs)的制备
将1.1mmol十六烷基三甲基溴化铵(CTAB)加入到含200ml水和1.2ml 2M NaOH水溶液中,并在搅拌下将混合物加热至70℃,搅拌1h。然后,向反应溶液中滴入提前混合的2ml原硅酸四乙酯(TEOS),再加入12mL乙酸乙酯,搅拌12小时。用等体积丙酮沉淀,10000rpm下离心分离,将得到的固体用无水乙醇洗涤3次以除去未反应的物质并分散在80ml乙醇中。向其中加入4ml HCl并在60℃回流下搅拌12小时。最后用无水乙醇和去离子水依次洗涤几次后,将介孔硅再分散于超纯水中冻干,得到介孔硅。
通过透射电子显微镜对实施例1制备的介孔硅进行表征,表征结果如图2所示。
图2示意性示出了本发明实施例制备的介孔硅纳米颗粒的透射电子显微镜表征图。
实施例2
对介孔硅表面接雷西莫特(MSNs-R)
在手套箱的无水无氧条件下,将0.028mmol二氯二甲基硅烷分散在150ul无水N,N-二甲基甲酰胺溶剂中,加入5ul的N,N-二异丙基乙胺,再加入0.019mmol的雷西莫特(R848),与二氯二甲基硅烷反应30min;再向反应体系中加入2mg介孔硅反应2h;反应结束后,将反应溶液滴入40ml乙醚中沉降、再用40ml乙醚洗涤多次得到白色固体介孔硅表面接雷西莫特(MSNs-R),将介孔硅表面接雷西莫特分散在1ml超纯水中,得到介孔硅表面接雷西莫特(MSNs-R)。
实施例3
化合物A的制备
将3.7nmol 4-氨基苯基-α-d-吡喃甘露糖苷溶于500ul的无水二甲基亚砜(DMSO)中30min,加入3.7nmol二硬脂酰基磷脂酰乙醇胺-聚乙二醇2000-N-羟基琥珀酰亚胺,30℃搅拌48h。通过使用截留分子量为1000Da的膜对反应混合物进行透析提纯48h。产物经冻干,得到化合物A产物。用核磁共振氢谱(H-NMR)对其化学结构进行表征。
图4示意性示出了本发明实施例制备的化合物A核磁共振氢谱图。
如图4所示,1H NMR(400MHz,DMSO)δ9.62(s,1H),7.35(d,J=8.6Hz,1H),7.00(d,J=8.9Hz,1H),5.24(s,1H),4.98(s,1H),4.81(d,J=5.2Hz,1H),4.72(d,J=6.0Hz,1H),4.44(s,1H),4.17(s,1H),3.85-3.12(m,80H),3.38(d,J=30.8Hz,38H),3.38(d,J=30.8Hz,38H),3.30-3.12(m,2H),2.50(s,10H),1.23(s,5H)。核磁结果得出,产物在化学位移值为7.35和7.00处多了两个峰,为产物4-氨基苯基-α-d-吡喃甘露糖苷中苯环的峰,可证明反应生成了化合物A。
实施例4
脂质体囊泡的制备
将2.4nmol二油酰磷脂酰胆碱、0.4nmol(2,3-二油酰基-丙基)-三甲胺、0.2nmol化合物A溶解在氯仿中,旋转蒸发去掉氯仿后形成一层薄膜,在薄膜中加入1ml超纯水,超声5分钟,将得到的混合液通过200nm挤出器挤出得到粒径为150nm左右的脂质体囊泡(Lipo-M)。
实施例5
将150ug CpG寡脱氧核苷酸激动剂加入到400ul浓度约为3.5mg/ml的介孔硅表面接雷西莫特(MSNs-R)分散液中,在1200rpm剧烈搅拌1h后,加入400ul浓度为2.46mg/ml的脂质体囊泡,在室温下800rpm搅拌过夜,用30k超滤管1500g,4℃,15min多次洗涤纳米颗粒,浓缩至1ml,得到复合纳米颗粒(CpG@MSNs-R@Lipo-M)。
图1示意性示出了本发明实施例中复合纳米颗粒的合成路线。
如图1所示,实施例1-5的制备步骤详细描述了复合纳米颗粒的合成路线。
通过透射电子显微镜对实施例5制备的复合纳米颗粒进行表征,结果如图3所示。

通过对实施例5制备的复合纳米颗粒进行动态光散射粒径测试,结果如图5所示。
图5示意性示出了本发明实施例制备的复合纳米颗粒的动态光散射粒径测试结果。
如图5所示,复合纳米颗粒的动态光散射粒径SIZE在150nm左右。
实施例6
对实施例1-5制备得到的介孔硅(MSNs)、介孔硅表面接雷西莫特(MSNs-R)、脂质体囊泡(Lipo-M)、复合纳米颗粒(CpG@MSNs-R@Lipo-M)进行动态光散射粒径、电势、PDI表征,结果如表1所示。

从表1可知,介孔硅的动态光散射粒径为122.3nm、电势为-21.1mV、PDI为0.203;介孔硅表面接雷西莫特的动态光散射粒径为234.3nm、电势为33.6mV、PDI为1.000;脂质体囊泡的动态光散射粒径为165.3nm、电势为29.1mV、PDI为0.106;复合纳米颗粒的动态光散射粒径为211.9nm、电势为-10.2mV、PDI为0.250,可见,介孔硅是负电性的;负电性的介孔硅通过表面接雷西莫特,变成正电性;脂质体囊泡是正电性的;复合纳米颗粒通过脂质体囊泡包裹的介孔硅表面接雷西莫特,最终带有负电性。PDI越大,分散性越差,MSNs-R的PDI最大,复合纳米颗粒降低了MSNs-R的PDI。因此,复合纳米颗粒在包裹了脂质体囊泡后,粒径减小,分散性明显提高。
实施例7
本实施例采用实施例1中的制备方法,其不同之处在于探究了不同雷西莫特(R848)和介孔硅(MSNs)的比例对雷西莫特(R848)的包封率以及介孔硅表面接雷西莫特(MSNs-R)电势和粒径的影响。

从表2可知,随着R848:MSNs质量比、R848投量、二氯二甲基硅烷(DCDMS)投量的增加,MSNs投量固定,R848包封率从0.12,增加到0.44,动态光散射粒径SIZE和PDI具有先增后降,又增的趋势,电势ZETA逐渐增大。
实施例8
本实施例采用实施例1中的制备方法,其不同之处在于改变了CpG寡脱氧核苷酸(CpG ODN)的投入,探究对雷西莫特(R848)和CpG ODN包封量、复合纳米颗粒(CpG@MSNs-R@Lipo-M)的电势和粒径的影响。

从表3可知,随着CpG寡脱氧核苷酸的投入量的增加,R848包封量成逐渐增加的趋势,CpG包封量从2.8ug、5.1ug、28.1ug、变化到到47.5ug。动态光散射粒径SIZE从240.3nm、215.6nm、211.9nm、变化到225.4nm,PDI分别为0.21、0.26、0.25及0.22,电势ZETA从5.2mV、-9.6mV、-10.2mV、变化到-23.6mV。
实施例9
应用实例1酸性pH触发纳米粒子中雷西莫特的释放
肿瘤组织处于微酸环境(pH为6.0~6.5),而且在肿瘤微环境中的抗原呈递细胞的溶酶体内部pH可达5.0。基于此,将实施例1中制备的复合纳米颗粒(CpG@MSNs-R@Lipo-M纳米颗粒)各取150ul加入到12个20k透析杯中,6个/组放置到配制好的pH分别为5.0和7.4的4L缓冲溶液中。取样点为0.5h、1h、2h、6h、12h、36h,到达取样时间点时取出样品,并对体积定量后,取50ul样品用4倍体积的乙腈破乳,用酶标仪对每份样品进行紫外测试,从而计算得雷西莫特的释放率。剩余样品旋蒸去除水后,用氯仿沉淀洗涤3次,将沉淀用500ul水溶解,在nanodrop上测CpG的紫外吸收,从而计算得CpG的释放率。
图6示意性示出了本发明实施例制备的复合纳米颗粒的药物体外释放曲线。
如图6所示,从图6可以看出,相比较于pH 7.4的中性环境,雷西莫特在pH 5.0的酸性环境下释放更快,整体释放速率较慢,另外,CpG在pH 5时在前30min内迅速释放完全,而在pH 7.4时则释放缓慢,这均表明了该纳米颗粒两种药物的酸响应性能较好。
实施例10
应用实例2采用本发明制备方法得到的纳米颗粒的药效试验
将小鼠B16F10细胞(1*105)皮下接种于6-8周龄雌性C57BL/6小鼠下侧翼。当肿瘤体积达到50~100mm3时,每隔两天分别静脉注射PBS、Free R848 and CpG、CpG@MSNs-R@Lipo-M、CpG@MSNs@Lipo-M and MSNs-R@Lipo-M纳米颗粒,共给4次,给药计划详见图7。
图7示意性示出了本发明实施例制备的复合纳米颗粒的给药计划示意图。
其中雷西莫特和CpG的剂量分别为2mg/kg和5ug/鼠。
设定小鼠生存终点为肿瘤体积达到3000mm3,通过肿瘤体积大小变化。
图8示意性示出了本发明实施例制备的复合纳米颗粒的药效试验,包括肿瘤生长曲线。
从图8可以看出,根据Two way ANOVA统计分析,各组肿瘤大小差异为:PBS与CpG@MSNs-R@Lipo-M组****p<0.0001,Free R848 and CpG与CpG@MSNs-R@Lipo-M组*p=0.0273,而CpG@MSNs-R@Lipo-M组与CpG@MSNs@Lipo-M and MSNs-R@Lipo-M组未见显著性差异。
因此,可以得出结论,相比对照组,复合纳米颗粒递送双激动剂表现出更强的抑瘤效果。

Claims (8)
1.一种复合纳米颗粒,包括:
脂质体囊泡包裹的介孔硅纳米颗粒;
其中,所述介孔硅纳米颗粒的表面修饰有第一模式识别受体激动剂和第二模式识别受体激动剂;其中,所述第一模式识别受体激动剂为雷西莫特或嘎德莫特,所述第二模式识别受体激动剂为CpG寡脱氧核苷酸、Poly(dT)、聚肌苷酸:聚胞苷酸、聚腺尿苷酸、dsDNA-EC、G3-YSD、HSV-60、Poly(dA:dT)、Poly(dG:dC)、3p-hpRNA、5'ppp-dsRNA、VACV-70、ssPolyU、ORN Sa19、环鸟苷单磷酸腺苷、环磷酸腺苷、环磷酸鸟苷中的任意一种,
通过在所述介孔硅纳米颗粒表面修饰硅烷醇基团与氯硅烷反应键接所述第一模式识别受体激动剂,使得所述介孔硅纳米颗粒的表面修饰为正电荷,且以静电作用吸附负电性的所述第二模式识别受体激动剂;
所述脂质体囊泡由二油酰磷脂酰胆碱、(2,3-二油酰基-丙基)三甲胺和式(I)所示的化合物A组成;
2.根据权利要求1所述的复合纳米颗粒,其中,所述脂质体囊泡中二油酰磷脂酰胆碱、(2,3-二油酰基-丙基)-三甲胺、所述化合物A的摩尔比为2~10:0.5~4:0.5~10。
3.一种权利要求1~2任一项所述的复合纳米颗粒的制备方法,包括:
将介孔硅纳米颗粒与第一模式识别受体激动剂反应,得到修饰正电性的纳米颗粒;
将所述正电性的纳米颗粒与第二模式识别受体激动剂混合,得到负电性的纳米颗粒;
将所述负电性的纳米颗粒与脂质体囊泡混合,得到所述复合纳米颗粒。
4.根据权利要求3所述的制备方法,其中,所述将介孔硅纳米颗粒与第一模式识别受体激动剂反应,得到修饰正电性的纳米颗粒,包括:
将所述第一模式识别受体激动剂、氯硅烷和有机碱在无水无氧气的条件下,混合反应10~60min后,得到第一混合溶液;
再将介孔硅纳米颗粒在无水无氧气的条件下,加入所述第一混合溶液中,反应1~4h,得到正电性的纳米颗粒。
5.根据权利要求4所述的制备方法,其中,所述有机碱包括N,N-二异丙基乙胺。
6.根据权利要求3所述的制备方法,其中,将所述正电性的纳米颗粒与第二模式识别受体激动剂混合,得到负电性的纳米颗粒,包括:
将所述第二模式识别受体激动剂加入所述正电性的纳米颗粒中,混合搅拌0.5~4h,得到所述负电性的纳米颗粒。
7.根据权利要求3所述的制备方法,其中,将所述负电性的纳米颗粒与脂质体囊泡混合,得到复合纳米颗粒,包括:
将二硬脂酰基磷脂酰乙醇胺-聚乙二醇2000-N-羟基琥珀酰亚胺与4-氨基苯基-α-d-吡喃甘露糖苷通过交联反应,制得化合物A;
再将二油酰磷脂酰胆碱、(2,3-二油酰基-丙基)-三甲胺和所述化合物A溶解在氯仿中,通过旋转蒸发法得到脂质体囊泡;
将所述脂质体囊泡与所述负电性的纳米颗粒混合搅拌预设时长,得到所述复合纳米颗粒。
8.一种权利要求1~2任一项所述的复合纳米颗粒在制备用于治疗癌症的药物中的应用。
