Abstract
本发明公开CREM/ICER基因或其转录体作为靶点在制备抗HIV药物中的应用。本发明是基于发明人发现CREM/ICER可以作为一个调控HIV感染和复制的关键蛋白,通过将CREM/ICER当作靶基因制备药物,能用于调控HIV病毒对CD4 T的感染和复制,所述药物可以是根据CREM/ICER为靶基因制备的抑制剂或小干扰RNA等药物。本发明还提供了根据CREM/ICER制备的siRNA,对CREM/ICER转录后特定剪切体的特异性好,可以特异性的抑制目的CREM/ICER剪切体的表达,从而达到抑制HIV感染和复制的效果,其可用于开发HIV治疗性疫苗,将会取得巨大的社会效益和经济价值。

技术领域
本发明属于生物技术与医学领域,特别涉及CREM/ICER基因或其转录体作为靶点在制备抗HIV药物中的应用。
背景技术
艾滋病,即获得性免疫缺陷综合征(acquired immunodeficiency syndrome,AIDS),由人免疫缺陷病毒(human immunodeficiency virus,HIV)引起。目前,艾滋病临床治疗方法主要是高效抗逆传录病毒疗法(Highly active antiretroviral therapy,HAART)。HAART能有效减少病毒载量,保护CD4 T细胞免受HIV病毒的攻击,进而保护免疫系统,延长了HIV感染者的存活时间。然而,目前抗病毒疗法只能控制病毒复制,无法清除HIV感染者体内的病毒。此外,抗HIV药物还有严重的副作用,高额的治疗费用,以及容易产生针对药物的耐药病毒株等一系列缺点。因此,研发有效的抗HIV的药物和针对HIV的疫苗是目前全球的研究热点。
现在的研究发现一些宿主基因可以控制病毒的感染,或者加速病毒感染后的恢复。通过调控这些宿主基因,可以阻断病毒感染和复制中的特定步骤,从而起到控制感染的作用。在HIV病毒整合到宿主基因组后,HIV的Tat蛋白控制正向转录延伸因子P-TEFb,辅助RNA聚合酶II(RNA polymerase II)对整合进宿主的HIV基因转录,从而完成病毒的感染周期。在HIV感染后,RNA聚合酶II对HIV感染周期的完成起到关键作用。之前的研究发现,CREM/ICER(cAMP responsive element modulator/inducible cAMP early repressor)可特异性的调节RNA聚合酶II的转录。同时,细胞外的HIV-1Tat蛋白可以诱导Jurkat CD4 T细胞系和外周淋巴细胞中的CREM/ICER的表达。这些结果说明CREM与HIV的感染和复制可能有着密切的关系,但到目前为止,还没有通过CREM/ICER基因来控制HIV感染的报道。
CREM属于cAMP反应结合蛋白家族,含有20个外显子并高度保守。通过对不同启动子的选择性识别,转录元件的使用,和外显子的可变剪切,最终翻译形成大量异构体(>30种)。CREM的每种异构体都有不同的蛋白结构域和功能位点,从而对下游基因起到抑制或激活的的作用。CREM的表达受细胞,组织和发育特异性机制的严格调控。ICER为CREM的一种,ICER使用CREM近3’端的启动子(P2启动子)转录翻译大于10种异构体。由于ICER缺少由P1启动子转录的CREM的5’端,在功能上和由P1启动子转录的CREM异构体相反。P1启动子转录的CREM异构体对下游基因起激活作用,而由P2启动子转录的ICER异构体对下游基因的功能起抑制作用。CREM/ICER基因编码的不同异构体都含有富含碱性亮氨酸的拉链结构域bZIP(Basic Leucine Zipper Domain),bZIP结构域可识别被称为cAMP响应元件上(cAMPresponsive element,CRE)的一段DNA回文结构(5’-TGACGTCA-3’)。多种细胞外信号都可以让CREM/ICER产生信号级联反应,比如生长因子或激素与跨膜受体结合后可驱动腺苷酸环化酶产生高水平的cAMP。然后,cAMP可以促进蛋白激酶(例如PKA,PKC以及酪蛋白激酶I和II)的酶促活性,进而磷酸化并激活CREM/ICER蛋白。

本发明的目的通过下述技术方案实现:CREM/ICER基因或其转录体作为靶点在制备抗HIV药物中的应用,是基于本发明发明人发现CREM/ICER可以作为一个调控HIV感染和复制的蛋白,通过将CREM/ICER当作靶基因制备药物,可以用于调控HIV病毒对CD4 T的感染和CD4 T细胞中的复制,所述药物可以是根据CREM/ICER为靶基因制备的抑制剂或小干扰RNA等药物,本领域技术人员可以根据CREM/ICER为靶基因进行设计。所述的转录体是由CREM/ICER不同外显子转录后通过可变剪切得到的转录体。
所述的CREM/ICER基因或其转录体作为靶点在制备抗HIV药物中的应用,优选以CREM/ICER基因的外显子15、16、17、18、19、20中的至少一个作为靶点,通过抑制外显子的转录和表达,得到抗HIV药物。
所述的转录体优选为含有外显子15和19拼接体的转录体、含有外显子16和19拼接体的转录体或含有外显子15和18拼接体的转录体。
所述的抗HIV药物优选为干扰RNA;优选为siRNA。
一种抗HIV药物,是依据上述应用筛选得到,为siRNA 1、siRNA2和siRNA3中的至少一种;

一种治疗HIV药物,包含编码上述抗HIV药物的DNA序列;优选包含启动子和编码上述抗HIV药物的DNA序列;更优选包含载体和编码上述抗HIV药物的DNA序列,载体上含有启动子。
所述的启动子优选为U6启动子或T7启动子。
所述的载体优选为慢病毒载体;更优选为能转录干扰RNA的慢病毒载体;最优选为pSIH1-H1-Puro载体。

本发明相对于现有技术具有如下的优点及效果:
1)本发明发明人发现CREM/ICER可以作为一个调控HIV感染和复制的关键蛋白,通过将CREM/ICER当作靶基因制备药物,可以用于调控HIV病毒对CD4 T的感染和CD4 T细胞中的复制,所述药物可以是根据CREM/ICER为靶基因制备的抑制剂或小干扰RNA等药物,本领域技术人员可以根据CREM/ICER为靶基因进行设计得到抗HIV药物。
2)本发明还提供了根据CREM/ICER制备的siRNA,易合成,成本低,对CREM/ICER特异性好,可以特异性的抑制CREM/ICER的表达,从而达到抑制HIV感染和复制的效果;
3)本发明提供的siRNA药物能够有效抑制HIV,可用于开发HIV治疗性疫苗,将会取得巨大的社会效益和经济价值。

具体实施方式
下面结合实施例及附图对本发明作进一步详细的描述,但本发明的实施方式不限于此。
实施例中未注明具体技术或条件者,按照本领域内的文献所描述的技术或条件,或者按照产品说明书进行。所用试剂或仪器未注明生产厂商者,均为可通过正规渠道商购获得的常规产品。
实施例1:针对CREM的shRNA的设计
根据人类基因的外显子使用频率,不同组织的表达,剪切体的构成模式的网站所提供的CREM基因信息(网址为:https://gtexportal.org/home/gene/CREM),筛选CREM产生抑制性信号的变体所在的外显子,CREM基因的外显子和变体信息如图1所示,筛选出外显子15、16、17、18、19、20。针对这些外显子设计特异性靶向CREM基因的siRNA。在公共的siRNA设计网站(网址为:https://portals.broadinstitute.org/gpp/public/seq/search)上输入含有筛选出的外显子的mRNA序列,按照网站指示预测可以沉默CREM mRNA表达的siRNA序列。
针对CREM基因共选择了5个待干扰的靶序列,信息见表1。

在合成的双链DNA片段一端引入BamHI酶切位点,另一端引入EcoRI酶切位点。shRNA的设计原则如图2所示。
其中,正义链代表插入的干扰片段,反义链代表和正义链的反向互补序列。
以针对编号1的DNA oligo设计为例(针对于外显子15和19):

将设计得到的DNA oligo序列交由上海生工生物有限公司合成。
实施例2:慢病毒的构建和包装
为了得到稳定表达针对HIV病毒基因组的siRNA,本实施例通过构建siRNA表达载体,将其与慢病毒包装系统共同转染进入人胚肾HEK 293细胞得到慢病毒颗粒。具体步骤如下:
1)shRNA的双链化:
将实施例1人工合成的每种shRNA对应的的正义链和反义链序列分别用双蒸水溶解,浓度为100μM。取相应的正义链和反义链寡聚物溶液各22.5μl,加入5μl的10×退火缓冲液(100mM Tris,pH 7.5~8.0、500mM NaCl、10mM EDTA),在PCR仪上进行退火反应。程序如下:95℃2分钟;每分钟温度降低1℃,梯度缓慢降温至25℃;4℃保存。退火处理后得到浓度为100μM的shRNA模板。将所得模板溶液使用双蒸水稀释1000倍,终浓度为100nM,用于连接反应。
2)pSIH1-H1-Puro载体的线性化:
将10×CutSmart buffer 5μl,BamHI和EcoRI各1μl,pSIH1-H1-Puro载体(产品号:SI500A-1,System Biosciences,美国)5μg,ddH2O补充至50μl进行酶切处理。在37℃酶切24小时,使用1%的琼脂糖凝胶电泳后用DNA回收试剂盒回收。
3)pSIH1-H1-Puro重组载体的构建:
按照10×T4 Ligation Buffer(T4连接酶缓冲液)2μl,线性化的pSIH1-H1-Puro(BamHI+EcoRI双酶切)4μl,shDNA模板(100nM)1μl,T4 DNA连接酶(5U/μl)1μl,双蒸水12μl的体系进行载体的连接反应。在16℃连接过夜(16-18小时)后,转化到DH5α感受态细胞。细菌培养16小时后,每个连接反应挑取4个菌落,接种到4mL含100μg/mL氨苄霉素的LB液体培养基中,37℃培养16小时,使用质粒提取试剂盒(天根生化科技有限公司)提取质粒。所提取的质粒送到苏州金维智生物科技有限公司进行测序鉴定。
4)慢病毒包装:
将状态良好,处于对数期的人胚肾HEK 293细胞(ATCC)用0.25%(w/v)胰酶消化,用含10%(v/v)胎牛血清DMEM完全培养基悬浮成单细胞悬液,细胞计数后,按照每孔6×105个细胞接种于6孔板中,培养18-24h细胞融合度达到70%左右时,换成不含抗生素的DMEM完全培养基。培养2小时后,使用Lipofectamine 2000按照说明书进行转染。转染步骤为(下面步骤中的物质的量为每孔的量):使用200μL Opti-MEM无血清MEM培养液用稀释1μg重组干扰质粒,再加入1μg慢病毒包装系统(Lentiviral Packaging Mix,Thermo Fisher,美国),加入6μl Lipofectamine 2000转染试剂,充分混匀,室温放置15分钟;然后将混合物逐滴加入对应的细胞上;6-8小时后换成含10%(v/v)胎牛血清的DMEM完全培养基。培养48小时后收集细胞培养上清(病毒液)。使用1500rpm离心10分钟去细胞及碎片,离心所得的上清液使用0.45μm滤膜获得病毒原液,分装冻存在-80℃。
5)慢病毒浓度测定:
将转染得到的6种慢病毒,分别为shRNA1-5和对照(对照为实施例1中包装shControl的慢病毒),使用p24 ELISA定量检测试剂盒(PerkinElmer,美国)检测病毒液中Gag(p24)的浓度,病毒浓度根据标准品换算为ng/ml。
实施例3:建立能够永久表达siRNA的CD4 T细胞株
本实施例选取对HIV敏感的CD4 T细胞株,使用慢病毒感染后筛选载体插入到宿主细胞基因组能稳定表达shRNA的细胞系。具体步骤如下:
1)分别培养如下CD4+细胞系:
Jurkat T细胞系(人T淋巴细胞白血病细胞,ATCC,美国),来源于人的CD4+T细胞,表达CD4、CXCR4和CCR5,可被CXCR4和CCR5嗜性的HIV病毒感染;
THP-1细胞系(ATCC,美国),来源于人的单核细胞,表达CD4和CXCR4,可被CXCR4嗜性的HIV病毒感染;
HuT78细胞系(ATCC,美国),来源于人的CD4+T细胞,表达CD4、CXCR4和低水平的CCR5,可被CXCR4嗜性的HIV病毒感染;
A3R5细胞系(Creative Biogene,美国),来源于人的CD4+T细胞,转入GFP蛋白,高表达CD4、CXCR4和CCR5,可被CXCR4和CCR5嗜性的HIV病毒感染;
CEM Luciferase细胞系(Creative Biogene,美国),来源于人的CD4+T细胞,转入Luciferase指示基因,HIV感染后可激活Luciferase活性,高表达CD4和CXCR4,可被CXCR4嗜性的HIV病毒感染;
以上细胞系的培养条件均为含10%胎牛血清的RPMI-1640完全培养基。
2)在48孔板中分别加入上述CD4+细胞系(细胞浓度均为1×105个细胞/孔/500μl),用实施例2步骤4)中制备的慢病毒20μl分别感染上述CD4+细胞,再加入8μg/ml(终浓度)的聚凝胺(polybrene)以促进感染;
3)感染18~24小时后去掉上清,换上新的完全培养基;
4)感染48小时后,加入2μg/mL(终浓度)的嘌呤霉素(puromycin)进行抗性筛选,每三天换一次新的含嘌呤霉素的培养基;
5)连续培养14~21天后,稳定表达siRNA的细胞株建立成功,可进行后续的HIV病毒感染实验。
实施例4:细胞内HIV病毒p24实验检测稳定表达CREM siRNA对HIV-1的抑制作用
1)HIV NL4-3病毒制备,具体包括以下步骤:
将人胚肾HEK 293细胞按照每孔6×105个细胞接种于6孔板中,培养18-24h细胞融合度达到70%左右时,换成不含抗生素的DMEM完全培养基。培养2小时后,使用Lipofectamine 2000按照说明书进行转染。转染步骤为:使用200μL Opti-MEM无血清MEM培养液用稀释2μg的HIV NL4-3质粒(BioVector NTCC保藏中心,北京),加入6μl的Lipofectamine 2000转染试剂,充分混匀,室温放置15分钟;然后将混合物逐滴加入对应的细胞上;6-8小时后换成含10%(v/v)胎牛血清的DMEM完全培养基。培养48小时后收集细胞培养上清(病毒液)。使用1500rpm离心10分钟去细胞及碎片,离心所得的上清液使用0.45μm滤膜获得病毒原液,分装冻存在-80℃。使用p24 ELISA定量检测试剂盒(PerkinElmer,美国)检测病毒液中Gag(p24)的浓度,病毒浓度根据标准品换算为ng/ml。
2)HIV病毒感染,具体包括以下步骤:
将实施例3中得到的稳定表达siRNA的Jurkat T细胞按2×104个细胞/孔接种到96孔U型细胞培养板,然后每孔加入20ng p24Gag的HIV NL4-3病毒,细胞置于5%CO2、37℃培养;培养18~24小时后,去掉含HIV病毒的培养基,使用含有10%(v/v)胎牛血清的RPMI-1640完全培养基洗一次;使用300g离心7分钟,去掉上清后加入200μl的含有10%(v/v)胎牛血清的RPMI-1640完全培养基,细胞置于5%CO2、37℃培养;分别在病毒感染细胞3天和7天的时候对细胞内病毒染色。
3)细胞内病毒染色,具体包括以下步骤:
将实施例4步骤2)中的细胞使用1×PBS洗一遍,加入分辨死细胞的染色剂(Live/Dead Fixable Near-IR Dead Cell Stain Kit,Thermo Fisher,美国),在4℃染色20分钟;300g离心7分钟弃上清,加入细胞固定剂和破膜剂(Fixation/Permeabilization SolutionKit,BD,美国),在4℃静置30分钟;400g离心7分钟弃上清,加入PE荧光标记的anti-p24抗体(KC57-RD1,Beckman Coulter,美国)1μl,在4℃静置30分钟;使用破膜剂洗液在400g离心7分钟条件下洗两次,用2%(v/v)多聚甲醛固定细胞;使用流式细胞仪检测细胞中的HIV病毒水平。
细胞内病毒检测结果如图3所示:实施例1中CREM基因的siRNA(siRNA 1~3,特别是siRNA 1和siRNA 2)明显抑制HIV-1NL4-3病毒在Jurkat T细胞中的复制。
实施例5:荧光素酶(Luciferase)实验检测稳定表达CREM siRNA对HIV-1的抑制作用
将实施例3中得到的稳定表达siRNA的CEM Luciferase细胞按2.5×104个细胞/孔/100μl接种到96孔平底细胞培养板,然后每孔加入100μl含5ng p24Gag的HIV NL4-3病毒的含10%(v/v)胎牛血清的RPMI-1640完全培养基;细胞置于5%CO2,37℃培养;培养48小时后,去掉100μl培养基,加入1%(v/v)的Triton X-100裂解细胞;室温放置10分钟让细胞充分裂解后,转移100μl上清至新的96孔板,加入萤火虫萤光素酶(Luciferase)检测试剂(Promega,美国),使用化学发光检测仪器(Luminometer)检测发光值。
荧光素酶检测结果如图4所示:实施例1中CREM基因的siRNA(siRNA 1和siRNA 2)明显抑制HIV-1 NL4-3病毒在CEM Luciferase T中复制。
实施例6:CREM siRNA在不同的CD4细胞系中对HIV-1的抑制作用
本实施例的大部分具体实施内容与实施例4相同,不同的是使用多种CD4细胞系检测CREM siRNA对HIV-1的抑制作用,具体包括以下:
在HIV病毒感染环节,将实施例3中得到的稳定表达siRNA的THP-1细胞、HuT78细胞、A3R5细胞和CEM Luciferase细胞分别按2.5×104个细胞/孔接种到96孔U型细胞培养板。对THP-1细胞和HuT78细胞每孔分别加50ng p24Gag的HIV NL4-3病毒;对A3R5细胞和CEMLuciferase细胞每孔分别加5ng p24Gag的HIV NL4-3病毒。
细胞置于5%CO2,37℃培养。其余步骤与实施例4相同。
细胞内病毒检测结果如图5所示:实施例1中CREM基因的siRNA(siRNA 1和siRNA2)明显抑制HIV-1NL4-3病毒在不同的CD4细胞中的复制。
实施例7:CREM siRNA对多种HIV病毒的抑制作用
本实施例的大部分具体实施内容与实施例4相同,不同的是使用CREM siRNA检测对多种HIV病毒的抑制作用,具体包括以下:
在HIV病毒感染环节,将实施例3中得到的稳定表达siRNA的Jurkat T细胞按2.5×104/孔接种到96孔U型细胞培养板,然后每孔或加入25ng p24Gag的HIV-1Ba-L病毒(Advanced Biotechnologies,美国),或加入25ng p24Gag的HIV-1IIIB病毒(AdvancedBiotechnologies,美国),或加入25ng p24Gag的HIV-1 MN病毒(AdvancedBiotechnologies,美国)。细胞置于5%CO2、37℃培养。其余步骤与实施例4相同。
细胞内病毒检测结果如图6所示:实施例1中CREM基因的siRNA明显抑制多种HIV病毒的复制。
实施例8:CREM siRNA长时间抑制HIV病毒
本实施例的大部分具体实施内容与实施例4相同,不同的是检测CREM siRNA能否长时间抑制HIV病毒的感染和复制,具体包括以下:
在HIV NL4-3病毒感染稳定表达CREM siRNA的Jurkat T细胞后,分别在病毒感染第3天、第5天、第7天、第10天和第14天取细胞样品并进行细胞内病毒染色。
细胞内病毒检测结果如图7所示:实施例1中CREM基因的siRNA(siRNA 1和siRNA2)能长时间控制HIV病毒在非常低的水平。
上述实施例为本发明较佳的实施方式,但本发明的实施方式并不受上述实施例的限制,其他的任何未背离本发明的精神实质与原理下所作的改变、修饰、替代、组合、简化,均应为等效的置换方式,都包含在本发明的保护范围之内。

1.CREM/ICER基因或其转录体的抑制剂在制备抗HIV药物中的应用,其特征在于:
所述的抑制剂为siRNA 1和siRNA2中的至少一种;其中,
siRNA 1正义链:5’-UAACUGGAGAUGACACAGCUG-3’;
siRNA 1反义链:5’-CAGCUGUGUCAUCUCCAGUUA-3’;
siRNA 2正义链:5’-UAGUGCUGCCACUGGUGACAU-3’;
siRNA 2反义链:5’-AUGUCACCAGUGGCAGCACUA-3’。
