Abstract
本发明涉及一种防治库尔勒香梨枝枯病的方法,将发酵得到的土壤改良剂撒施梨树旁边开的沟槽内,覆土浇水,在梨树发病关键期配合喷洒生物复配杀菌剂防控。其中土壤改良剂发酵原料有效利用废弃物梨渣与秸秆、高分子聚合物协同发酵,土壤改良剂中的有益菌可以与枝枯病的病原菌发生拮抗作用,有效抑制土壤中病原菌的生长和传播。香梨梨渣富含多糖、多酚,可以促进发酵菌剂中有益菌的生长,从而增强对土壤中致病菌的抑菌性。通过香梨渣的生物强化腐殖化获得的土壤改良剂,获得健康的微生物生态系统,同时补充土壤有机质含量,实现土壤中蚯蚓数量显著增加,解决土壤碱性化、肥力下降的问题,提高香梨植株的抗病能力,提升香梨产量和果品品质的目的。

技术领域
本申请涉及农业技术领域,具体涉及一种防治库尔勒香梨枝枯病的方法。
背景技术
库尔勒香梨是新疆维吾尔自治区特产,中国国家地理标志产品。因具有色泽悦目、味甜爽滑、香气浓郁、皮薄肉细、酥脆爽口、汁多渣少、落地即碎、入口即化、耐久贮藏、营养丰富等特点,被誉为“梨中珍品”、“果中王子”。从2016年开始,枝枯病开始在库尔勒香梨种植园爆发并蔓延扩散,出现整园死树现象;其次,当地传统的香梨种植和管理技术落后,长期以施用化肥为主,大水漫灌,植保用药不规范等粗放式的肥、水、植保管理,导致土壤生态退化,次生盐渍化、土壤碱化逐年加重,最终体现在香梨品质的退化和产量下降。香梨枝枯病的发病特征是:侵染梨树的多种组织和器官。初期侵染嫩梢部位和花序,然后病原菌顺着韧皮内里往近根部位侵染,具有从嫩枝浸染到侧枝,从侧枝侵染主干,严重者枯死的特征。症状在花梗上表现为水渍状、灰绿色病变,随之花瓣由红变褐或黑色。花序之间相互传染,发病的花序不脱落。在果实上,早期侵染的果实不膨大,色泽黑暗;呈现红褐色或黑色病斑。在新梢枝条上首先表现为灰绿色病变,随之整个新梢萎蔫下垂,最后枯死。在叶上,在树皮上,发病后,略凹陷,着色也略深,皮下组织呈水渍状。一旦主茎皮下组织色变深,呈水渍状,表明该树病重,一般半季就死树,而且传染性极强。在根部,被侵染的根根腐。
香梨枝枯病和香梨品质的退化严重威胁到库尔勒香梨产业的可持续发展,针对香梨枝枯病,当地政府从2016年枝枯病爆发开始,诚邀全国的专家学者对香梨枝枯病的防治做了大量的调研和试验,至今还没有找到有效的香梨枝枯病防治和香梨品质提升技术措施。
土壤改良剂又称土壤调理剂,是指可以改善土壤物理性,促进作物养分吸收,而本身不提供植物养分的一种物料。随着现代农业的发展,土壤改良剂作为一种改善土壤性状和肥力的有效手段被大力采用,而减少化学药品的使用,采用土壤修复与生物技术彻底改善果园、农田病害的防治手段正逐渐受到重视。另一方面,随着可持续发展模式的深入人心,对日益增加的固体废弃物的资源化处置显得非常重要和必要。循环利用农林废弃物,因地制宜,就近取材,不仅可以节约农业成本,同时对促进农业生态可持续发展起到推进作用。
因此,研究一种防治库尔勒香梨枝枯病的方法,就近利用农业废弃原料发酵得到的土壤改良剂与生物复配杀菌剂配合作用,调理土壤微生物环境,提高梨树枝枯病的免疫能力,提高库尔勒香梨产量具有重要意义。
发明内容
本申请的发明目的是:有效利用废弃物梨渣与秸秆、高分子聚合物协同发酵获得土壤改良剂,通过香梨渣的生物强化腐殖化,补充土壤有机质含量的同时,增加土壤生物活性;土壤改良剂中的有益菌与枝枯病的病原菌发生拮抗作用,有效抑制病原菌的生长。香梨梨渣富含多糖、多酚类物质,如奎尼酸、绿原酸等,利用多糖和多酚物质作为益生元能够促进发酵菌剂中有益菌的生长的特性,从而增强土壤的抑菌性,获得健康的微生物生态系统;同时实现土壤中蚯蚓数量显著增加,解决土壤碱性化、肥力下降的问题,从而实现提高香梨的抗病能力,提升香梨产量和果品品质。
为实现上述发明目的,解决上述技术问题,本发明提供一种防治库尔勒香梨枝枯病的方法,其特征在于:所述方法包括以下步骤:
将发酵得到的土壤改良剂撒施梨树旁边开的沟槽内,覆土浇水,施用量为每亩300-500kg,在施用土壤改良剂后,在梨树发病关键期配合喷洒生物复配杀菌剂防控;
所述土壤改良剂的制备方法包括以下步骤:将梨渣、农林废弃秸秆分别粉碎,投入混料机混合搅拌10-20分钟,加入一定量高分子聚合物,继续搅拌5-10分钟,梨渣、农林废弃秸秆与高分子聚合物的质量比为4:(13-15):(1-2),混匀后加入占混料总量1%-3%的复合发酵剂,复合发酵剂有效活菌数≥2.00×108cfu·g-1,加水搅拌至半干半湿状态,含水量35%-45%,得到发酵原料,将发酵原料堆放在发酵容器内微氧发酵,每隔4-6天翻动一次,保持含水量35-45%,发酵30-40天,完成发酵,加入5%-10%天然矿物类原料混合搅拌均匀,得到土壤改良剂;
所述生物复配杀菌剂由5%中生菌素可湿性粉剂、8%多抗霉素可湿性粉剂和N-(5-三氟甲基-3,4-噻二唑-2-基)-2-氟苯甲酰胺组成,质量比为1:1:1,生物复配杀菌剂稀释为600倍液,每年施药两次,第一次在开花前,第二次在7月或8月高温干旱期,第一年每亩喷洒60kg,第二年及以后的用药量减半。

本发明的有益效果:
1、本发明土壤改良剂原料加入梨渣进行发酵,解决了库尔勒香梨产地大量梨渣浪费的问题,同时与农林废弃物和高分子聚合物配合发酵,不但实现了农林废弃物的再利用,还能有效增强土壤的抑菌性,改善土壤理化性质,增加土壤有机质含量,解决土壤碱性化、肥力下降的问题。
2、土壤改良剂中的菌种选择可以与枝枯病的病原菌发生拮抗作用,有效抑制病原菌的生长,阻断病原菌在土壤中传播。
3、香梨梨渣富含多糖、多酚类物质,如奎尼酸、绿原酸等,利用多糖和多酚物质作为益生元能够促进发酵菌剂中有益菌的生长的特性,选择香梨梨渣作为益生元显著提高枯草芽孢杆菌及植物乳杆菌的数量,调理改善土壤微生物群系,从而增强对土壤中致病菌的抑菌性,获得健康的微生物生态系统,可达到更好的土壤改良效果及防治枝枯病效果,提高香梨的抗病能力。
4、通过香梨渣的生物强化腐殖化,培育土壤健康的生态系统,增加土壤生物活性,土壤中蚯蚓数量显著增加,从而有效的改善了土壤团粒结构,减少土壤板结,增加土壤孔隙度,增加土壤自净化功能,提高抗病免疫能力。
5、本发明土壤改良剂与生物复配杀菌剂结合使用,能够减少杀菌剂的使用,减少农药残留,同时达到更好地防治香梨枝枯病的效果,提高香梨的亩产量及一级果的比例,减少梨树枝枯病的发病率。
6、本发明土壤改良剂促进香梨总酸含量降低,增加了总糖的含量,糖酸比得到提高,改善了香梨的口感。
7、土壤改良剂中加入高分子聚合物可以提高土壤结构中矿物质的稳定性,提高土壤的通气性、透水性,提高土壤分散颗粒的团聚作用。

实验一:土壤改良前后有机质、pH、氮磷钾、蚯蚓数量的变化对比
实验例1:采用实施例1的方法,土壤改良剂施用量为每亩400kg;
实验例2:采用实施例2的方法,土壤改良剂施用量为每亩400kg;
实验例3:采用实施例1的方法,土壤改良剂施用量为替换为每亩1000kg;
空白例:不添加土壤改良剂,仅向梨树喷洒生物复配杀菌剂;
对比例1:采用实施例1的方法,将梨渣替换为等量花大豆秸秆;
对比例2:采用实施例1的方法,将复合菌剂中地衣芽孢杆菌替换为等量枯草芽孢杆菌,解淀粉芽孢杆菌替换为等量枯草芽孢杆菌。
试验方法:
在库尔勒香梨成品园,种植密度4m x 5m,树龄7-12年,划分不同片区施用不同的土壤改良剂,每月取一次土壤样品,深度为0-30cm,每个样品取三个平行样品,每月保持在同一取样点取样。采用土壤有机质用重铬酸钾氧化外加热法测定,总氮用碱性过硫酸钾法测定,总磷用硫酸-过氧化氢消煮钼锑抗比色法测定,速效钾用乙酸铵浸提火焰光度计(WFX-200)法测定,土壤pH用pH计测定(土水比1:2.5)。
实验结果1:

结果显示,空白例有机质含量最少,对比例1缺少梨渣原料的土壤改良剂施用土壤中的有机质高于空白例,少于实验例1-3。对比例2菌种的变化导致有机质的减少速度加快。实验例1-3有机质平均含量提高,且能够在随后的几个月中保持稳定。实验例2梨渣相对含量减少,有机质的含量也相对于实验例1有所降低。实验例3相对于实验例1大幅提高了土壤调理剂的用量,但是土壤中有机质的含量与实验例1相接近,说明本发明限定的土壤改良剂使用量能在最节约成本的情况下,达到最好的土壤有机质含量提高效果。

结果显示,空白例pH呈现碱性,对比例1提高了土壤pH,少于实验例。对比例2菌种的变化并未影响土壤pH。实验例pH显著下降,改善了土壤的碱性环境。
实验结果3:

结果显示,空白例氮磷钾含量最少,对比例1缺少梨渣的土壤改良剂处理后,土壤中的氮磷钾高于空白例,明显少于实验例1-3。对比例2菌种的变化导致氮磷钾的减少速度加快。实验例氮磷钾平均含量提高,且能够在随后的几个月中保持稳定。实验例2梨渣相对含量减少,氮磷钾的含量也相对于实验例1有所降低。实验例3提高了土壤改良剂用量,很好地保持土壤中氮磷钾的含量但相对与实验例1增幅不显著。

使用土壤改良剂一个月后,对实验例1-3、空白例、对比例1-2根据单位体积每1m3田间地表蚯蚓粪堆数量和可见蚯蚓数量进行抽样统计,。统计结果可知:空白例土壤中蚯蚓粪堆数量和可见蚯蚓数量含量最少,对比例1缺少梨渣原料的土壤改良剂施用土壤中的数量高于空白例,少于实验例1-3。实验例1-3土壤中,改土1个月即出现小蚯蚓,在随后的几个月中蚯蚓粪堆数量和可见蚯蚓数量明显增多。实验例3相对于实验例1结果相接近,说明本发明限定的土壤改良剂使用量的过分提高,不能使土壤中蚯蚓粪堆数量和可见蚯蚓数量含量更多。
实验二 土壤改良剂对香梨枝枯病防治及香梨品质改善试验
实验例1-3:分别将实施例1-3发酵得到的土壤改良剂撒于梨树旁边开的沟槽内,覆土施压后浇水,施用量为每亩400kg。在施用土壤改良剂后,向梨树喷洒生物复配杀菌剂,所述生物复配杀菌剂由5%中生菌素可湿性粉剂、8%多抗霉素可湿性粉剂和N-(5-三氟甲基-3,4-噻二唑-2-基)-2-氟苯甲酰胺组成。5%中生菌素可湿性粉剂、8%多抗霉素可湿性粉剂和N-(5-三氟甲基-3,4-噻二唑-2-基)-2-氟苯甲酰胺的质量比为1:1:1。生物复配杀菌剂稀释为600倍液,每亩喷洒60kg。
空白例:不添加土壤改良剂和杀菌剂。
对比例1:土壤改良剂采用实施例1方法,将复合菌剂中地衣芽孢杆菌替换为等量枯草芽孢杆菌,解淀粉芽孢杆菌替换为等量枯草芽孢杆菌。
对比例2:省略土壤改良剂,杀菌剂喷洒量翻倍。
对比例3:省略杀菌剂,土壤改良剂采用实施例1方法制备,施用量为每亩400kg。
试验方法:
在库尔勒香梨成品园,种植密度4m x 5m,树龄7-12年,划分不同片区施用不同的土壤改良剂,施用量每亩300kg,深度为0-30cm,生物复配杀菌剂稀释为600倍液,每亩喷洒60kg。每月至少一次记录相关病害、虫害和草害的基本信息,以拍照/摄影为主。
测产方案:在每个试验园区选择10株树,准确数出每株树所结的果实总数,在每株香梨树上随机取鲜果5-10个,分别称重,取平均值即为其单果质量,以每株树的结果总数与这株树的平均单果质量之积计算单株产量,测得单株产量后折合每亩的产量。
对香梨果实品质研究的试验方法如下:总糖参照GB5009.7食品中还原糖的测定,总酸参照GB/T12456食品中总酸的测定方法,可溶性固形物含量采用阿贝折光仪法进行测定。
表4枝枯病树统计
实验结果:砍树量指因枝枯病重而砍掉的香梨树的数量,从表4可知,未添加土壤改良剂和生物复配杀菌剂的空白对照组有57棵香梨树有枝枯病症,发病率30%。仅加入生物复配杀菌剂或者土壤改良剂的对比例2和3枝枯病的发病率有所降低,分别为22.3%和21.2%。实验例1-3同时加入土壤改良剂和生物复配杀菌剂后产生了抑菌的协同效果,且从实验例1和2的对比看,梨渣含量的增加有助于减少枝枯病的发病率,从实验例1和3的对比看,复合菌剂加入量的增加也同样有助于减少枝枯病的发病率,说明土壤改良剂对于枝枯病菌的抑制作用是复合菌剂与梨渣共同作用产生的。从对比例1也可以看出,省略了两种菌后,相比于实验例发病率和砍树率都有所上升。
表5测定每亩产量和分级
实验结果:从果品的分级亩产量可以看出,土壤改良对香梨产量和品质的促进作用是明显的。和空白对照组比较,对比例2和3的亩产量分别增加79%和114%。实验例1和实验例2的亩产量分别增加245%和200%,相比于仅添加生物复配杀菌剂的对比例2,实验例1和实验例2的亩产量分别增加93%和68%,可以看出土壤改良剂和生物复配杀菌剂共同使用产生了协同增效的效果,增加了亩产量,而复合菌剂的增加对亩产量的增加影响并不大。
表6香梨果实品质变化
实验结果:从香梨品质的变化可以看出,除了空白对照组以外,香梨可溶性固形物的含量几乎没有变化,土壤改良对可溶性固形物含量的影响不明显。实验例1-3的总糖含量相比于空白例和对比例有所提高,土壤改良促进香梨总酸含量降低,总糖含量升高。一般来讲,糖酸比越大,水果味道以甜为主;糖酸比越小,水果味道以酸为主。好吃的水果一般糖酸比都比较高。土壤改良使香梨更可口。经过一个80人的盲试品尝测试,85%的测试者认为实验例1的香梨比空白对照的皮薄,味甜,比对比例的香梨甜酸度更适中。盲试品尝测试的结果和香梨糖酸比的变化趋势相吻合。
实验三库尔勒香梨梨渣原料对复配菌种生长的促进对比试验
以实施例1中的土壤改良剂获得方法进行发酵,对比省略香梨梨渣原料的发酵对比例。发酵过程中,每隔一定时间对发酵产物取样,进行一定的梯度稀释,选择合适稀释度的样品涂布于相应的培养基并对其中的菌群计数,结果如表7所示(菌群单位为lg(cfu/mL)):
结果显示,土壤改良剂的发酵制备过程中,采用香梨梨渣原料与省略香梨梨渣原料相对比,含有香梨梨渣原料的发酵产物中枯草芽孢杆菌及植物乳杆菌的数量显著高于对比例,但对酿酒酵母的数量增加不明显。由此结果可知,将香梨梨渣作为发酵原料,可以有效提高复合菌剂中枯草芽孢杆菌和植物乳杆菌的扩繁数量。本领域技术人员知晓,香梨梨渣中含有大量多酚、多糖类物质,比如奎尼酸、绿原酸等,这些活性成分可作为益生元促进益生菌的生长和繁殖。由本实验可知,采用香梨梨渣原料作为土壤改良剂的发酵原料,可有效促进复合菌剂以及土壤中有益菌枯草芽孢杆菌和植物乳杆菌的扩繁速度,从而促进香梨枝枯病感染土壤中的微生物群系的生态恢复。
以上描述了本发明优选实施方式,然其并非用以限定本发明。本领域技术人员对在此公开的实施方案可进行并不偏离本发明范畴和精神的改进和变化。
Claims (1)
1.一种防治库尔勒香梨枝枯病的方法,其特征在于:所述方法包括以下步骤:
将发酵得到的土壤改良剂撒施梨树旁边开的沟槽内,覆土浇水,施用量为每亩300-500kg,在施用土壤改良剂后,在梨树发病关键期配合喷洒生物复配杀菌剂防控;
所述土壤改良剂的制备方法包括以下步骤:将梨渣、农林废弃秸秆分别粉碎,投入混料机混合搅拌10-20分钟,加入一定量高分子聚合物,继续搅拌5-10分钟,梨渣、农林废弃秸秆与高分子聚合物的质量比为4:(13-15):(1-2),混匀后加入占混料总量1%-3%的复合发酵剂,复合发酵剂有效活菌数≥2.00×108cfu·g-1,加水搅拌至半干半湿状态,含水量35%-45%,得到发酵原料,将发酵原料堆放在发酵容器内微氧发酵,每隔4-6天翻动一次,保持含水量35-45%,发酵30-40天,完成发酵,加入5%-10%天然矿物类原料混合搅拌均匀,得到土壤改良剂;
所述生物复配杀菌剂由5%中生菌素可湿性粉剂、8%多抗霉素可湿性粉剂和N-(5-三氟甲基-3,4-噻二唑-2-基)-2-氟苯甲酰胺组成,质量比为1:1:1,生物复配杀菌剂稀释为600倍液,每年施药两次,第一次在开花前,第二次在7月或8月高温干旱期,第一年每亩喷洒60kg,第二年及以后的用药量减半;
所述梨渣为库尔勒香梨梨渣;所述农林废弃秸秆由玉米秸秆、大豆秸秆、花生茎叶中的一种或多种组成;
所述高分子聚合物由聚乙二醇、聚乙烯醇、聚丙烯酰胺中的一种或多种组成;
所述复合发酵剂由枯草芽孢杆菌、地衣芽孢杆菌、长柄木霉、解淀粉芽孢杆菌、白腐菌、植物乳杆菌、假单胞菌、酿酒酵母菌,按质量比:1:(1-3):(1-3):(1-3):(1-3):(1-3):(1-3):(1-3)复配所得;
所述天然矿物类原料由磷矿粉、白云石粉、钾矿粉中的一种或两种组成。
