技术领域
本发明属于生物医药领域,涉及用于药物筛选的试验动物模型领域,具体涉及视网膜新生血管疾病动物模型、构建方法及其应用。
背景技术
视网膜新生血管属于异常血管增生,其主要发生在视网膜中央静脉阻塞、年龄相关性黄斑变性、糖尿病视网膜疾病等,是临床病例中致盲的主要原因之一。迄今为止,视网膜新生血管发生的分子机制并不完全清楚,因此,构建与之相关的动物模型是进行该疾病机制研究、针对该疾病的预防及药物开发的重要基础。
视网膜新生血管模型主要有三类:早产儿视网膜病变模型、糖尿病视网膜病变模型、视网膜静脉阻塞动物模型。
1)氧诱导视网膜病变模型(OIR模型):该模型通过高氧再相对缺氧的饲养环境来产生新生小鼠视网膜新生血管,即将出生第7天的小鼠与哺乳母鼠一起放置于密闭氧箱中,氧浓度维持在75%±2%,第12天小鼠与哺乳母鼠放回室内标准环境下饲养(氧浓度21%),此时小鼠视网膜新生血管发生,5天后达高峰,随后新生血管开始消退,一周后几乎完全消退。这一造模方法的操作十分简单,并且造模成功率高,重复性良好,近几年在视网膜新生血管研究中被广泛应用。但是,该模型存在以下缺陷:
①.OIR模型在极短的几天时间内通过短期高氧和低氧诱导产生急性视网膜新生血管,这种新生血管的发生仅类似于人类早产儿视网膜病变,与其它慢性视网膜新生血管病程并不一致,甚至其发病的分子机制也不相同。
②.该模型在实际造模过程中存在动物死亡率高、成膜率低且不稳定等缺点,同时也受到特定设备的制约。
③.OIR模型更多地反映的是新生鼠视网膜新生血管的状态,对于成年小鼠视网膜新生血管的代表性不佳。
④.氧诱导的视网膜新生血管模型在1~2周时间即可自然消退,这一特性不利于部分长时间干预的研究。
2)糖尿病视网膜病变新生血管模型:糖尿病视网膜病变中,视网膜新生血管是主要表现。现阶段用于研究糖尿病视网膜病变以及并发症的动物模型较多,主要包括以下3种:药物或饮食诱发性、转基因或基因敲除以及自发性遗传性。
诱发性模型的优缺点:优点是耗时短、成本低、方法简便、重复性较好、短期内可批量造模,是目前最常用的模型。缺点是药物对其他组织有一定的毒性损害;动物间有个体差异;动物死亡风险较大;不能出现完全的视网膜病变新生血管病变。
转基因或基因敲除模型的优缺点:优点是病变由转入的外源性基因引起,在疾病的基因水平上,病因清楚,模型动物间表现的差异小。缺点是该类模型主要是针对某些特定基因突变导致的病变,从而不能成为一种广谱的动物模型;而且,该模型操作较复杂,成本昂贵,不适合大批量造模从而限制了它的广泛应用。
自发性糖尿病模型的优缺点:该模型动物未经过任何有意识的人工处置,多数采用有自发性糖尿病倾向的近交系纯种动物,按照饲养条件喂养,自发成模,最接近人类疾病的发病过程。这类模型的优点是具有同质的遗传背景,能控制环境因素,可对这种多因素疾病进行基因分析。缺点是频繁的同系繁殖和单基因遗传,使其发生糖尿病的遗传同质性与人类有差异。同时,对饲养和繁殖条件的要求高,价格昂贵。
总的来说,糖尿病动物模型由于是全身性疾病,因此其诱导产生的视网膜新生血管不能简单视为单纯性视网膜新生血管,需考虑多种混杂因素,对特定病理过程的代表性不佳。

发明内容
针对现有视网膜新生血管疾病动物模型存在的问题,本发明基于小胶质细胞活化这一视网膜新生血管共同病理特征,旨在提供一种造模稳定性强、诱导周期短且具有良好代表性的视网膜新生血管疾病动物模型及其构建方法与应用。
基于上述目的,本发明采用的技术方案如下:
第一方面,本发明提供一种视网膜新生血管疾病动物模型的构建方法,将源于M1型活化小胶质细胞的外泌体注射于动物的眼球组织,诱导产生视网膜新生血管。
发明人所在研究团队发现在现有的多种视网膜新生血管动物模型中,视网膜小胶质细胞出现了异常的聚集和活化,并首次证实M1型活化小胶质细胞来源的外泌体可直接促进新生血管的产生,也可以通过促进原位静息小胶质细胞活化、富集促进血管信号而间接促进视网膜新生血管的产生,这种炎症-新生血管的病理模式符合大多数视网膜新生血管产生的病理过程,因此,本发明利用源于M1型活化小胶质细胞的外泌体诱导产生视网膜新生血管构建的动物模型,对视网膜病理过程具有良好的代表性。
另外,相对于经典的OIR模型在构建时受到高氧箱设备的限制,致使许多研究团队无法建立该模型,OIR模型的应用受限;而本发明基于外泌体诱导产生视网膜新生血管,采用常规外泌体的分离方式即可从M1型活化小胶质细胞中分离出外泌体,外泌体的获取方式简单,且整个模型构建过程中不依赖于特定设备,因而具有较好的推广应用前景。
进一步地,将源于M1型活化小胶质细胞的外泌体注射于动物眼球组织的玻璃体腔,诱导产生视网膜新生血管。
采用玻璃体腔注射,注射针头只需穿过外侧的巩膜,色素上皮层和局部视网膜,便可进入玻璃体腔,同时,玻璃体腔注射仅在进针部位穿过视网膜,不会造成视网膜脱离,对视网膜细胞的损伤小,炎症反应较轻,玻璃体腔注射具有操作简单、损伤小、作用范围广的优势。
进一步地,外泌体的单次注射量为:0.5~1ug/uL;注射频次为:每2~3天注射1次,注射持续时间为:5~6周。
本发明在6周左右的时间内,通过向动物眼球组织(如玻璃体腔)定期注射外泌体诱导动物产生视网膜新生血管,相对于现有视网膜新生血管疾病动物模型如链脲佐菌素诱导的糖尿病视网膜新生血管疾病动物模型的诱导周期通常超过6个月,本发明构建动物模型的方法显著缩短了新生血管的诱导时间,亦即显著缩短了视网膜新生血管疾病动物模型的构建周期。
进一步地,M1型活化小胶质细胞由小胶质细胞经LPS诱导生成。
进一步地,诱导小胶质细胞生成M1型活化小胶质细胞的LPS的量为50~100ng/mL,诱导时间为24h~48h。
LPS激活小胶质细胞膜上的TLR4受体,下游激活NF-κB信号通路,诱导小胶质细胞发生M1型活化。
进一步地,M1型活化小胶质细胞的外泌体由差速超速离心分离得到或由外泌体提取试剂盒提取得到。
本发明诱导产生视网膜新生血管的外泌体,可以通过外泌体提取试剂盒提取得到或者利用超高速离心机进行差速超速离心分离得到,将分离得到的外泌体进行动物眼球组织如玻璃体注射即可构建视网膜新生血管疾病动物模型,操作简单,且不依赖于特定设备,便于推广应用。
进一步地,动物为6~8周龄的成年鼠或8~12周龄的成年兔,或者为动物实验中其他常用的成年动物。
本发明以成年鼠或兔作为诱导对象构建视网膜新生血管疾病动物模型,由于成年动物在诱导过程中不会涉及到发育因素,使得构建的成年动物模型相对于幼年动物模型(如OIR模型),具有更好的稳定性和代表性,具有较高的研究价值。
第二方面,本发明提供一种由上述方法构建得到的视网膜新生血管疾病动物模型。
本发明通过向动物的眼球组织注射源于M1型活化小胶质细胞的外泌体,诱导其产生视网膜新生血管,由该方法构建的视网膜新生血管疾病动物模型具有造模稳定,成功率在80%以上,且连续观测1~2个月,视网膜新生血管能够稳定存在,具有较好的稳定性和病理状态持续性。
第三方面,本发明提供上述视网膜新生血管疾病动物模型在视网膜新生血管疾病研究中的应用。
针对目前视网膜新生血管发生的分子机制并不完全清楚,本发明构建的视网膜新生血管疾病动物模型具有良好的稳定性和病理状态的持续性,能够为视网膜新生血管疾病的发病机制的研究提供重要基础。
第四方面,本发明提供上述视网膜新生血管疾病动物模型在预防或治疗视网膜新生血管疾病药物开发中的应用。
与现有技术相比,本发明的有益效果如下:
(1)本发明首次通过活化小胶质细胞的外泌体诱导产生视网膜新生血管以构建动物模型,外泌体通过直接促进新生血管产生,同时又促进原位静息小胶质细胞活化及富集,促进血管信号而间接促进视网膜新生血管的产生,这种炎症-新生血管的病理模式符合大多数视网膜新生血管产生的病理过程,使得由本发明方法构建的视网膜新生血管疾病动物模型具有良好的代表性。
(2)本发明构建视网膜新生血管疾病动物模型的方法,不依赖于特定设备,外泌体的分离采用外泌体提取试剂盒或超高速离心机进行超速离心分离获得,在实验室中依据本发明方法能够自行构建该视网膜新生血管疾病动物模型,具有良好的推广应用价值。
(3)本发明动物模型以成年动物为构建对象,相对于幼年动物疾病模型,降低了发育因素的影响,具有更高的稳定性、持续性和良好的代表性,具有较高的研究价值和应用价值。
(4)本发明利用外泌体诱导构建视网膜新生血管疾病动物模型方法,诱导时间为5~6周,即本发明在6周的时间内能够构建具有良好稳定性、代表性的动物模型,显著缩短了模型构建过程中的诱导时间。
综上所述,本发明构建视网膜新生血管疾病动物模型的周期短、成功率高并具有良好的代表性,且构建的动物模型具有良好的稳定性和病理状态的持续性,在视网膜新生血管疾病的发病机制研究、预防或治疗视网膜新生血管疾病药物开发等方面具有广阔的应用前景。

具体实施方式
为更好地说明本发明的目的、技术方案和优点,下面将结合具体实施例对本发明作进一步说明。本领域技术人员应当理解,此处所描述的具体实施例仅用以解释本发明,并不用于限定本发明。
实施例中所用的试验方法如无特殊说明,均为常规方法;所用的材料、试剂等,如无特殊说明,均可从商业途径得到。
实施例1视网膜新生血管疾病动物模型的构建
本实施例以小胶质细胞BV2为例进行说明视网膜新生血管疾病动物模型的构建方法,具体包括如下步骤:
1、小胶质细胞BV2活化
将BV2细胞于培养液中培养至呈对数生长期时,更换培养液为含100ng/mL LPS的培养液,培养24~48h后即可获得M1型活化的BV2细胞悬浮液,其中,BV2细胞购买于国家实验细胞资源共享平台,LPS脂多糖货号为sigma L4391。
2、外泌体的提取、鉴定
(1)外泌体的提取
利用外泌体提取试剂盒进行分离M1型活化的BV2细胞的外泌体,或者采用如下差速高速离心分离的方法,从M1型活化的BV2细胞悬浮液中分离出外泌体。
将M1型活化的BV2细胞悬浮液,以300×g离心10min,2000×g离心10min,10000×g、4℃离心30min,经0.22μm滤网过滤除去残留细胞,然后将悬浮液以100000×g、4℃超离70min(BecKman Coulter,California,USA),收集沉淀,PBS重悬沉淀物后再以100000×g、4℃超离70min;最后将沉淀重悬于100μL PBS中,即得源于M1型活化小胶质细胞BV2的外泌体悬液,记为M1-EXO。
以未经LPS活化处理的小胶质细胞BV2作为对照,由该小胶质细胞参照上述方法分离得到的外泌体记为M0-EXO。
(2)外泌体的鉴定
向外泌体悬液M1-EXO和M0-EXO中分别加入3%(v/v)戊二醛,滴入碳涂覆的铜网格中,5min后用蒸馏水反复洗涤后,用4%乙酸铀酰染色10min,再用1%甲基纤维素溶液处理5min,待网格干燥后使用TEM 1011透射电子显微镜在80KV(JEOL-1200EX)下成像,结果如图1A所示,M1-EXO和M0-EXO均为双凹“茶托样”圆形囊泡。
使用配备有快速视频捕获和粒子追踪软件的Nanosight LM10系统(NanosightLtd,Navato,CA)分析外泌体的大小,通过测量布朗运动的速率来计算纳米粒子浓度和粒度分布,结果如图1B所示,M0-EXO的直径为132.2±40.5nm,M1-EXO的直径为136.4±43.1nm。
进一步利用蛋白质印迹法(Western Blot)对外泌体蛋白CD63、HSP70、TSG101进行检测,结果如图1C所示,可以看到外泌体蛋白CD63、HSP70、TSG101在M0-EXO和M1-EXO中均有表达。
以上结果证明了M0-EXO和M1-EXO均具有外泌体性质。
3、小鼠玻璃体腔注射外泌体构建视网膜新生血管疾病动物模型
(1)向6~8周龄的小鼠玻璃体腔内注射源于M1型活化小胶质细胞BV2的外泌体M1-EXO,单次注射量为1ug/uL;注射频次为每3天注射1次,注射持续时间为6周,以构建视网膜新生血管疾病动物模型。向小鼠玻璃体腔注射外泌体的具体过程如下:
在解剖显微镜下固定好小鼠头位,充分暴露眼球,于角膜缘后1mm处,避开血管组织,利用破囊针头做巩膜穿刺。使用33G Hamilton显微注射器以约45度角从穿刺口避开晶状体斜穿入玻璃体腔内。保持针头不移动的情况下缓慢推注注射器,使外泌体缓慢释放,推注完成后停留30秒后缓慢拔出针头,立即用消毒棉签封闭针孔,覆盖结膜。术后利用羟甲基纤维素凝胶涂眼,以确保在小鼠苏醒前眼球表面的湿润。待到小鼠完全复苏后放回鼠笼。在手术中及复苏过程中使用电热毯保温,以确保实验动物的顺利苏醒、避免术后患病和死亡。
所有动物实验程序均严格遵循中山大学中山眼科中心《关于动物应用于伦理学规定》,伦理编号为SYXK(YUE)2017-093。所有动物饲养、实验操作及处死原则均遵循视觉与眼科学研究学会(ARVO)《关于动物实验处理规定相关要求》进行。
(2)视网膜新生血管疾病动物模型的鉴定
为明确M1型活化BV2细胞来源的外泌体是否具有诱导视网膜新生血管的作用,同时进一步鉴定上述构建的视网膜新生血管疾病动物模型是否产生了视网膜新生血管,参照上述步骤(1)向小鼠玻璃体腔内注射源于M1型活化小胶质细胞BV2的外泌体M1-EXO的方法,向小鼠玻璃体腔内分别注射PBS、M0-EX0作为对照,同时设置假手术组消除操作影响,其中,假手术组为进行上述相同的注射操作过程,但不注射任何物质。
上述四组试验分别为PBS组、M0-EX0组、M1-EXO组、假手术组(SHAM),每组实验中实验动物数量不小于5只;在试验持续注射6周后,处死小鼠,对小鼠眼球组织进行冰冻切片和免疫荧光染色进行检测,具体方法如下:
修剪新鲜眼球组织表面肌肉、筋膜等组织,4%新鲜配制的PFA固定眼球,4℃过夜。次日于10%蔗糖溶液中脱水2h后,以30%蔗糖溶液过夜脱水。将过夜脱水后的眼球组织浸入OCT包埋剂,待OCT完全浸入眼球组织后,将包埋模具放入-80℃冰箱,待组织完全冰冻后行组织切片。使用冰冻切片机,将眼球组织切成10μm厚度的组织切片,做好标记,储存于-80℃冰箱备用。
切片洗脱OCT后,用5%BSA封闭1小时,使用以下一抗:Iba-1(Abcam,ab178847),SDF-1(Abcam,ab25117),CXCR4(Abcam,ab124824),PECAM-1(Santa Cruz,sc-376764),Ki67(Cell Signaling Technology,9129),与第二抗体和DAPI分别避光孵育后,使用防淬灭封片剂封片。
试验结果分析方法为:两组间比较采用双尾Student T-test检测;三组及三组以上结果组间统计学差异采用单因素方差分析(One-way ANOVA)和多重比较Tukey's HSD检测。应用R(version 4.0.0)进行数据分析,应用GraphPad Prism 8(California)软件进行可视化。结果记录为均数±标准误(SEM)形式。在所有实验数据统计结果中,当p<0.05认为具有统计学意义。
免疫荧光检测结果如图2A和2B所示,M1-EXO玻璃体腔注射后,出现突破内界膜且具有增殖活性的内皮细胞,表明M1-EXO诱导产生了视网膜新生血管。此外,由图2C和图2D可以看出,M1-EXO组的血管新生相关蛋白SDF-1和CXCR4表达量也明显增多,进一步证实源于M1型活化小胶质细胞BV2的外泌体诱导小鼠眼球组织产生了视网膜新生血管。表明,本发明采用将源于M1型活化小胶质细胞的外泌体注射于小鼠眼球组织的玻璃体腔方法,成功构建了视网膜新生血管疾病动物模型。
采用如表1中分四批次对77只小鼠,按照上述方法进行玻璃体腔注射M1-EXO构建视网膜新生血管疾病动物模型,造模的成功率在80%以上。

随后对上述成功构建的67只视网膜新生血管疾病动物模型进行连续观测1~2个月,对小鼠组织进行切片染色,结果如图3所示,可以看到由本发明构建的模型,在成功建模后的1~2个月,视网膜新生血管依然能够稳定存在,表明由本发明方法构建的视网膜新生血管疾病动物模型具有良好的病理持续性和稳定性。
最后所应当说明的是,以上实施例仅用以说明本发明的技术方案而非对本发明保护范围的限制,尽管参照较佳实施例对本发明作了详细说明,本领域的普通技术人员应当理解,可以对本发明的技术方案进行修改或者等同替换,而不脱离本发明技术方案的实质和范围。
Claims (10)
1.一种视网膜新生血管疾病动物模型的构建方法,其特征在于,将源于M1型活化小胶质细胞的外泌体注射于动物的眼球组织,诱导产生视网膜新生血管。
2.根据权利要求1所述视网膜新生血管疾病动物模型的构建方法,其特征在于,所述源于M1型活化小胶质细胞的外泌体注射于动物眼球组织的玻璃体腔,诱导产生视网膜新生血管。
3.根据权利要求1或2所述视网膜新生血管疾病动物模型的构建方法,其特征在于,所述外泌体的单次注射量为:0.5~1ug/uL;注射频次为:每2~3天注射1次,注射持续时间为:5~6周。
4.根据权利要求1或2所述视网膜新生血管疾病动物模型的构建方法,其特征在于,所述M1型活化小胶质细胞由小胶质细胞经LPS诱导生成。
5.根据权利要求4所述视网膜新生血管疾病动物模型的构建方法,其特征在于,所述诱导小胶质细胞生成M1型活化小胶质细胞的LPS的量为50~100ng/mL,诱导时间为24h~48h。
6.根据权利要求1或2所述视网膜新生血管疾病动物模型的构建方法,其特征在于,所述M1型活化小胶质细胞的外泌体由差速超速离心分离得到或由外泌体提取试剂盒提取得到。
7.根据权利要求1或2所述视网膜新生血管疾病动物模型的构建方法,其特征在于,所述动物为6~8周龄的成年鼠或8~12周龄的成年兔。
