Abstract

背景技术
病毒感染可以引发人体的多种疾病。流感病毒感染是引起流行性感冒的一个主要因素,它传染性强,传播速度快,主要通过空气中的飞沫、人与人之间的接触或与被污染物品的接触传播。典型的临床症状是:急起高热、全身疼痛、显著乏力和轻度呼吸道症状。每年的秋冬季节是其高发期。肠道病毒71型主要引起手足口病,还可引起无菌性脑膜炎、脑干脑炎和脊髓灰质炎样的麻痹等多种神经系统疾病。患者表现为低热、流涕、食欲下降、口痛、呕吐、腹泻等。口腔黏膜出现小疱疹,常分布于舌、颊黏膜、硬腭,也可以出现在扁桃体、牙龈及咽部等,疱疹破溃后形成溃疡。手足口病和中枢神经系统感染是肠道病毒71 型感染而引起的两大常见临床症状。水疱性口炎病毒主要感染牛马猪等禽类,具有兽医上和畜牧业经济上的重要性,被感染的动物的症状多为发热,嗜睡,食欲下降,口腔、乳头、趾间及蹄冠上出现水疱性病灶和腿部的罐状环带,可造成局部的继发性的细菌和真菌感染,带来重大的经济损失。I型单纯疱疹病毒在人群中的感染率高达90%,常呈隐形感染并持续存在,当机体抵抗力下降时,体内潜伏的HSV被激活而发病,临床特征为皮肤黏膜成簇出现的单房性小水疱,常发生于面部,易复发。
白细胞介素(interleukin,IL)最早是指由白细胞产生且只在白细胞间发挥调节作用的细胞因子。后来发现,除白细胞外,其他细胞也能产生白细胞介素,而且白细胞介素也能作用于其他靶细胞,具有广泛的生物学功能和意义。IL-6最初由于实验体系和功能的不同,曾分别被命名为杂交瘤/浆细胞瘤生长因子,干细胞刺激因子,T细胞分化因子,B细胞分化因子和B细胞刺激因子-2等。于1986年正式命名白细胞介素-6(interleukin 6,IL-6)。IL6基因位于第7号染色体,长约5kb,分子量26kDa,由5个外显子和4个内含子组成。成熟的IL6分子由184个氨基酸组成,其蛋白质二级结构由4个α螺旋和C端受体结合点所组成,与受体结合的重要位点是179位的精氨酸。淋巴细胞和某些非淋巴样细胞可产生 IL6,如T细胞、B细胞、成纤维细胞、内皮细胞、单核细胞、肾小球系膜细胞和角朊细胞等。IL6能作用于多种靶细胞,具有促进细胞分化、刺激细胞生长等生物学效应,并能参与机体的多种免疫调节反应。IL-27是IL-6/IL-12家族的一个异源二聚体成员,是Ⅰ型细胞因子。IL-27是由螺旋蛋白p28和可溶性细胞因子受体样成分EBI3(EB病毒诱导基因3, Epstein-Barr virus-inducedgene 3)组成。EBI3自身没有表现出直接的活性,必须跟其它蛋白一起才能形成有活性的细胞因子。EBI3的二聚体搭档是IL-27A,一个螺旋状的细胞因子,是通过与其同源的IL-12/p35和IL-6而被鉴定的。
现有技术中能够抗多种病毒感染的药物较少,因此有必要开发一种新的能够抗多种病毒感染的药物。
发明内容
本发明目的是提供IL-6-IL-27复合物及其在制备抗病毒药物中的应用,IL-6-IL-27复合物为白介素6和白介素27形成的复合物,可以阻断IAV、VSV、EV71、HSV-I、SeV病毒感染,从而发挥抗病毒作用。
在本发明的第一方面,提供了一种IL-6-IL-27复合物,所述IL-6-IL-27复合物为IL-6 和IL-27相互作用形成的复合物。
在本发明的第二方面,提供了一种IL-6-IL-27复合物的表达载体,所述表达载体能够表达IL-6、IL-27A、EBi3和IL-27。

从SeV感染的人THP1细胞中提取细胞总RNA,反转录获得cDNA,以所述cDNA为模板扩增获得人IL-6和IL-27的cDNA;
将所述IL-6和IL-27的cDNA克隆到pcDNA3.1(+)载体上,获得真核表达质粒pcDNA3.1-IL-6-IL-27。
进一步地,所述IL-6和IL-27的cDNA克隆到pcDNA3.1(+)载体的NheI/XhoI酶切位点上。
在本发明的第四方面,提供了一种所述的IL-6-IL-27复合物和所述的IL-6-IL-27复合物的表达载体在制备抗病毒药物中的应用。

本发明证实使用IL-6-IL-27复合物处理可以抑制多种病毒的感染,包括流感病毒(IAV)、水泡型口炎病毒(VSV)、肠道病毒71(EV71)、I型单纯疱疹病毒(HSV-I)和仙台病毒(SeV)。病毒感染IL-6-IL-27复合物预处理的细胞,实验组与对照组相比,病毒的复制水平得到了很大程度的抑制。因此IL-6-IL-27复合物具有抗病毒感染的活性。这一新的生物学功能的发现可以作为研究IL-6-IL-27复合物在抑制病毒感染方面的潜在功能。
此外,L-6和IL-27是多功能细胞因子,可调节免疫反应、造血、组织损伤的急性期反应和炎症反应。IL-6在COVID-19感染期间发挥基础性作用,与细胞因子风暴的启动和进展有关,对感染者可能造成致命后果。IL-6的过表达可引起炎症因子风暴和免疫细胞毒。IL-6 和IL-27相互作用形成的复合物可缓解IL-6过表达所造成的问题,也可抑制COVID-19的复制和表达。未来的研究应该阐明IL-6-IL-27复合物在先天免疫和抗病毒活性中的作用,特别是对于COVID-19。

本申请实施例的技术方案为解决上述技术问题,总体思路如下:
细胞因子白介素(IL)是一类参与免疫反应表达和调节的细胞因子,它们可以由多种细胞产生,并能作用于多种细胞发挥功能,目前已得到承认的白介素成员至少达38个。在我们前期的研究工作中发现可溶性白细胞介素6受体(sIL-6R)和IL-27的亚基IL-27A在宿主抗病毒天然免疫中紧密联系并发挥重要作用。IL-6、IL-27作为IL-6/IL-12家族的重要成员,它们之间是否存在某种相互作用形成IL-6-Il-27复合物参与天然免疫反应和炎症反应是我们本课题要研究的主要内容。
在本发明实施例中,我们首先从多个实验角度验证IL-6、IL-27A、EBi3之间的相互作用确认IL-6-Il-27复合物的存在。然后从细胞水平探究IL-6-IL-27复合物在抗病毒天然免疫反应和炎症反应中的作用,我们从感染了甲型流感病毒的患者的血清和咽拭子样本中检测到IL-6-IL-27复合物(如图1和图2所示)。
随后,构建表达IL-6-IL-27复合物的质粒以探索其生物学功能。
结果表明,IL-6-IL-27复合物比IL-6,IL-27A和EBi3的各个亚基具有更强的抗病毒作用。
此外,IL-6-IL-27复合物的活性主要由IL-27A亚基和受体IL-27Rα介导。IL-6-IL-27复合物可通过与衔接蛋白MAVS相互作用,增强TRAF3和TRAF6的泛素化以及NF-κB向核易位,积极调节病毒触发的干扰素(IFN)和IFN刺激基因的表达。分泌的IL-6-IL-27复合物可诱导STAT1和STAT3磷酸化,并显示抗病毒活性。
总之,我们通过不同的实验方法鉴定了一个之前未被认识的复合物,并将其命名为 IL-6-IL-27复合物,这种复合物对多种病毒感染表现出强大的抗病毒活性。
我们的研究还表明,IL-6-IL-27复合物作为新型的激活因子,通过与MAVS相互作用,泛素化TRAF6和TRAF3,正向调控NF-κB和JAK-STAT信号通路。我们的结果证明了IL-6,IL-27A和EBi3在细胞内和细胞外形成先前未知的复合物,揭示了IL-6-IL-27复合物在抗病毒天然免疫通路中的分子机制,论证了该复合物在宿主抗病毒反应中所起的重要作用。本课题大胆提出IL-6-IL-27复合物的新观念,对传统的关于白细胞介素的观念提出新的挑战,并研究其在抗病毒天然免疫中的重要作用,有望为流行性疾病的预防和治疗提供新的方法。
IL-6和IL-27是白细胞介素(IL)-6家族细胞因子的成员,这些细胞因子有一个共同的受体gp130。我们检测了IL-6-IL-27流感病毒感染患者的血清和咽拭子样本。我们用流感病毒感染患者的样本、过表达IL-6-IL-27复合物质粒、人源重组蛋白IL-6和IL-27和细胞共定位实验等证实了IL-6-IL-27复合物亚基之间的相互作用。
我们之前证明了可溶性IL-6R/IL-27A蛋白在介导病毒诱导的III型IFN的产生中的作用。此外,我们还发现sIL-6R在抗病毒免疫应答期间病毒触发的I型IFN诱导和IL-27A介导的抗病毒功能中的作用。有趣的是,在本研究中,我们观察到IL-6-IL-27复合物比 IL-27A或EBi3的单个亚基成分具有更强的抗病毒作用,这有助于描述这种反应的分子机制。作为IL-27的一个亚基,Il-27A是一种四螺旋细胞因子,需要与EBi3结合才能有效分泌和功能激活。IL-27最初被描述为促炎细胞因子,但这种细胞因子也有抗病毒作用,包括抑制 HIV和丙型肝炎病毒的复制。我们论证了IL-6-IL-27复合物通过不同的机制介导抗病毒反应。在这项研究中,我们发现该复合物可以抑制多种病毒的复制,包括H1N1病毒、HSV-1 病毒、EV71病毒、VSV病毒和SeV病毒。根据之前的研究结果和我们目前的研究结果显示,IL-6-IL-27复合物是一种新的抗病毒因子,对多种病毒感染具有抗病毒活性。
由于IL6-IL27复合物由三个亚基组成,我们需要确认哪个亚基是抗病毒活性最重要的决定因素。为此,我们干扰了IL-6-IL-27复合物的单个亚基的表达。当IL-27A在A549细胞中被沉默时,H1N1复制被促进,IFNs和ISGs的表达降低(如图3-6,3-7所示)。为了进一步探索IL-6-IL-27复合物介导的抗病毒应答的机制,我们将重点关注IFNs和炎症因子。 IL-6-IL-27复合物增强病毒触发的IFN-α、IFN-β、IFN-λ1和ISGs的表达,包括OAS1、PKR 和MxA。最近的几篇报道也表明,IL-6、IL-27A和EBi3在炎症性疾病中发挥重要作用,因此我们接下来检测过表达IL-6-IL-27复合物的细胞中促炎细胞因子的产生,如IL-1β、IL-8 和TNF-α。我们的结论是,该复合物显示出对IL-8调控的强烈影响,这与Masaaki Murakami 最近的一篇报告描述的一致。综上所述,IL-6-IL-27复合物在病毒触发免疫应答中强烈促进了IFNs和ISGs的表达,IL-27Rα和IL-27A在这一过程中发挥了重要作用。
病毒触发IFN的产生受到各种分子和机制的精确调控。IL-6R和IL-27A可激活MAVS和TRAF6,表明IL-6-IL-27复合物与MAVS/TRAF6或MAVS/TRAF3复合物存在一定关系。一些证据表明IL-6-IL-27复合体可以靶向病毒触发信号通路中的MAVS/TRAF3/6。我们的 CoIP实验表明IL-6-IL-27复合物与MAVS相互作用,泛素化实验表明细胞内IL-6-IL-27复合物可与TRAF6或TRAF3相互作用并促进其泛素化。由于在抗病毒免疫应答中, MAVS/TRAF6信号复合物在IRF3和NF-κB激活中发挥关键作用,我们的结果也证明了 IL-6-IL-27复合物正向调控宿主的抗病毒活性,促进NF-κB向细胞核的转移。然后与IFN 基因的特定启动子元素结合激活基因表达。考虑到受体和IL-6-IL-27亚基在病毒触发的先天免疫应答中的重要作用,我们接下来试图探索IL-6-IL-27复合体的哪个受体介导其抗病毒应答。为此,我们使用了pLKO.1-TRC克隆载体系统构建IL-6R-KD细胞系、IL-27Rα-KD细胞系和gp130-KD细胞系,并探讨其在KD细胞中的抗病毒作用和免疫应答。在A549细胞中敲除IL-6R、IL-27Rα和gp130后,在IL-27Rα-KD细胞中H1N1复制增强最明显,而在 IL-27Rα-KD细胞中恢复IL-27Rα表达后,H1N1复制受到明显的抑制。
许多报道和实验表明IL-6和IL-27与JAK-STAT信号通路有关。虽然我们已经发现了一种新的IFNs和ISGs的激活剂,但我们接下来想知道的是IL-6-IL-27复合物是否参与了JAK-STAT信号通路的调控。STAT磷酸化分析表明,IL-6-IL-27复合物可增强病毒感染后STAT1和STAT3的磷酸化,从而调控下游基因的表达。

基于我们的研究结果,我们提出了IL-6-IL-27复合物诱导宿主抗病毒天然免疫的工作模型(如图16所示)。在该模型中,病毒感染诱导IL-6-IL-27复合物的表达。该复合物可与MAVS、 TRAF6、TRAF3相互作用,通过MAVS/TRAF3/6通路诱导IFN表达。IL-6-IL-27和MAVS/TRAF3/6复合物通过调节p50和p65与IFN启动子的结合诱导IFN的表达。IL-6-IL-27 复合物分泌到细胞外环境中,可与其他细胞相互作用,诱导STAT1和STAT3磷酸化,从而激活下游基因发挥其抗病毒功能。总之,本专利发现了一种此前未知的IL-6-IL-27复合物,并展示了其调节病毒复制和抗病毒反应的功能,促进了我们对宿主免疫应答病毒感染的认识,为研制抗病毒药物提供理论基础。
下面将结合实施例及实验数据对本申请的IL-6-IL-27复合物及其在制备抗病毒药物中的应用进行详细说明。

本专利中所有表达质粒均构建于pcDNA3.1(+)载体上,采用青岛生物科技有限公司的同源重组克隆试剂盒进行克隆。从SeV感染的人THP1细胞中提取细胞总RNA,扩增人IL-6、 EBi3和IL-27A的cDNA,并直接克隆到pcDNA3.1(+)载体上。IL-6用C端3×HA标记, EBi3用C端Myc标记,IL-27A用C端V5标记。在IL-27表达质粒中,将EBi3和IL27A 的编码序列与编码肽链的序列GSGSGGSGGSGSGKL连接,并添加C端V5标签。IL-6荧光蛋白由IL6、GFP和C端ER保留信号KDEL组成,命名为IL6-GFP-KDEL。IL-27荧光蛋白由IL27、DsRed和C端KDEL信号组成,命名为IL27-DsRed-KDEL。
此外,构建了过表达MyD88、MAVS、TRAF6、TRAF3、TBK1、IRF3、p50和p65的质粒。

1、IL-6-HA质粒的构建方法为通过PCR获得,退火温度为60℃,引物序列如SEQ IDNO.1-2所示;再通过同源重组的方法将目的片段构建于pcDNA3.1(+)空载上。

6、IL27-DsRed-KDEL的构建方法为通过PCR获得,退火温度为60℃,引物序列如SEQID NO.11-12所示;再通过同源重组的方法将目的片段构建于pcDNA3.1(+)空载上。
7、MAVS-Flag的构建方法为通过PCR获得,退火温度为60℃,引物序列如SEQ IDNO.13-14所示;再通过同源重组的方法将目的片段构建于pcDNA3.1(+)空载上。

14、IL-6-IL-27复合表达质粒的构建
从SeV感染的人THP1细胞中提取细胞总RNA,反转录获得cDNA,以所述cDNA 为模板扩增获得人IL-6和IL-27的cDNA;扩增人IL-6的引物对序列分别如SEQ ID NO.1-2 所示和SEQ ID NO.3-4所示;
将所述IL-6和IL-27的cDNA克隆到pcDNA3.1(+)载体的NheI/XhoI酶切位点上,通过两个自剪切的Linker E2A和P2A将其连接,获得真核表达质粒pcDNA3.1-IL-6-IL-27。

各质粒都通过测序确定序列正确,且瞬时转染入细胞后皆能正常表达,相关表达结果可见图3和图4。
实施例2、内源相互作用
一、血清样本
本专利中用到的10例流感病毒感染患者血清标本和45例流感病毒感染患者的咽拭子样本均来自湖北省疾病预防控制中心。临床样本的收集是根据《赫尔辛基宣言》的原则进行的,并根据《人类受试者保护指南》得到武汉大学机构审查委员会的批准。所有研究参与者都为样本收集和后续分析提供了书面知情同意。
二、血清样本的处理
10例IAV感染者血清标本来自湖北省疾病预防控制中心。将血清样品混合后分为三组,每组取1ml样品用于CoIP实验。其余样品用PBS稀释至蛋白浓度5.00g/l,以5μl的负荷量进行western blot检测。
我们还采集了湖北省疾病预防控制中心45例IAV感染患者的咽拭子样本。咽拭子样本混合并浓缩于10kd超滤管中,将浓缩样品分为3组,每组500μl用于CoIP实验,其余样品进行免疫印迹。
三、内源的相互作用
向肝癌细胞加TGF-β刺激,刺激1小时时间后收取细胞,加入适量细胞裂解缓冲液(含蛋白酶抑制剂),冰上裂解30min,细胞裂解液于4℃,最大转速离心30min后取上清得到细胞总蛋白。取少量裂解液以备Western blot分析(input分析),剩余裂解液加5μg相应的抗体加入到细胞裂解液4℃缓慢摇晃孵育过夜;取10μl protein G琼脂糖珠,用适量裂解缓冲液洗3次,每次3,000rpm离心3min;将预处理过的琼脂糖珠加入到和抗体孵育过夜的细胞裂解液中4℃缓慢摇晃孵育2-4h,使抗体与琼脂糖珠偶连;免疫沉淀反应后,在4℃以 3,000rpm速度离心3min,将琼脂糖珠离心至管底;将上清小心吸去,琼脂糖珠用1ml裂解缓冲液洗3-4次;最后加入30μl的2×SDS上样缓冲液,沸水煮5分钟;Western blotting 分析。
由图1可知,血清样本的内源IP确认IL-6、IL-27A、EBi3之间的相互作用,证明了IL-6-Il-27复合物的存在;
由图2可知,咽拭子样本的内源IP确认IL-6、IL-27A、EBi3之间的相互作用,证明了IL-6-Il-27复合物的存在。
实施例3通过免疫共沉淀确定相互作用
将实施例1构建的IL-6-HA、IL-27-V5、IL-27A-V5和EBi3-Myc表达质粒进行不同的组合进行共转,共转后30小时,收取细胞,加入适量细胞裂解缓冲液(含蛋白酶抑制剂),冰上裂解30min,细胞裂解液于4℃,最大转速离心15min后取上清得到细胞总蛋白。其中, IL-6-HA、IL-27-V5、IL-27A-V5和EBi3-Myc蛋白的KD分别为25KD、28KD、54KD和34KD;
取少量裂解液以备Western blot分析(input分析);
剩余裂解液加1μg相应的抗体(剩余裂解液分为4份,其中一份加入1ug抗HA抗体共同孵育,其中一份加入1ug IgG共同孵育,其中一份加入1ug V5共同孵育,其中一份加入1ugMyc共同孵育),后加入到细胞裂解液,4℃缓慢摇晃孵育过夜;取10μl protein G琼脂糖珠,用适量裂解缓冲液洗3次,每次3,000rpm离心3min;将预处理过的protein G琼脂糖珠加入到和抗体孵育过夜的细胞裂解液中4℃缓慢摇晃孵育2-4h,使抗体与protein G 琼脂糖珠偶连;免疫沉淀反应后,在4℃以3,000rpm速度离心3min,将琼脂糖珠离心至管底;将上清小心吸去,琼脂糖珠用1ml裂解缓冲液洗3-4次;最后加入30μl的2×SDS上样缓冲液,沸水煮5分钟;Western blotting分析。
由图3可知,IL-6-HA、IL-27-V5、IL-27A-V5和EBi3-Myc发生相互作用。具体地:
图3A为IL-6-HA和IL-27A-V5共转后的正拉和反拉的结果,表明IL-6-HA和IL-27A-V5 存在相互作用;
图3B为IL-6-HA和EBi3-Myc共转后的正拉和反拉的结果;表明IL-6-HA和EBi3-Myc存在相互作用;
图3C为IL-27A-V5和EBi3-Myc共转后的正拉和反拉的结果;表明IL-27A-V5和EBi3-Myc存在相互作用;
图3D为IL-6-HA和IL-27A-V5共转后的正拉和反拉的结果;表明IL-6-HA和IL-27A-V5 存在相互作用。
实施例4共定位应荧光检测
共聚焦免疫荧光
(1)4%多聚甲醛固定15min,PBS洗涤3次;
(2)然后用含4%BSA的PBS在室温下封闭1h;
(3)固定细胞与原代Ab在4℃孵育过夜;
(4)第二天,用含有0.01%Tween 20和1%BSA的PBS洗涤3次;
(5)与第二抗体(ProteinTech组)孵育1h后,用含有0.01%Tween 20和1%BSA的PBS 洗涤5次;
(6)用DAPI(H1200)在室温下对细胞核进行染色5分钟,然后用激光共聚焦显微镜(fluview FV1000;奥林巴斯)进行观察。
由图3E可知,IL6和IL27都主要存在于细胞质内,且它们之间有很好的共定位,呈现出明显的点状分布。
实施例5细胞水平检测IL-6-IL-27复合物对多种病毒的影响
一、RNA的提取、RT-PCR和real time PCR
1.用Trizol处理的细胞中,以每管加入1ml Trizol裂解细胞为例;
2.在收集了细胞裂解液的EP管中加入200ul的氯仿,振荡涡旋15s,静置3min;
3.4℃离心机12000rpm 15min;
5.取最上层上清(约200ul)至新的无RNA酶的EP管中,每管中加入500ul异丙醇,静置10分钟;
6.4℃离心机12000rpm 10min;
7.小心吸除上清,留下管底少量沉淀,每管中加入1000ul的无RNA酶的75%的无水乙醇;
8.轻柔的上下颠倒之后会清洗掉沉淀表面的残留杂质,4℃离心机12000rpm5min;
9.除去上清之后,室温下静置直至管内水分完全蒸发(约5-10min),此过程注意观察干燥程度,待水分挥发之后沉淀变成无色;
10.加入适量无RNA酶的水溶解沉淀,并55℃水浴锅水浴十分钟帮助沉淀充分溶解;
11.测量RNA的浓度用于后续的实验。
12.待检测完RNA的浓度之后,根据实验需求,取一定量RNA;
13.加入引物、逆转录酶、dNTPs、RNAase inhibiter,平行和对照样本都配制成相同体系;
14.然后在事先准备的42℃水浴锅水浴60min,再95℃水浴锅水浴5min,得到的就是相对的稳定的cDNA,可以在-20℃冰箱保存,或者直接进行后续实验。
real time PCR是采用SYBR Green染料和双链DNA镶嵌发光为原理。在实验室购买的 Mix中包含了反应所需的原料,只需加入模板、引物和ddH2O即可。每个样设置三个复孔,待点完样之后,贴上封闭用的膜,采用机器默认的程序对样本进行实时定量的扩增。本课题所采取的的qPCR引物如表3所示。

二、流式细胞计数
VSV-eGFP感染A549细胞为例
VSV-eGFP感染细胞之后用胰酶消化细胞;
离心之后收集细胞,然后用PBS重悬细胞;
轻柔的多次吹打细胞利于形成单个细胞,采用轴流式专业管,在流式仪器中上样;
设定流式的激发光和接收光之后,开始读数,检测10000个细胞之后即可停止,每个样本重复三次;
采用流式仪器自带软件和对照组对比之后即可知道靶标样本被VSV-eGFP感染的数量和比例。
三、病毒感染
A549,RD和VERO细胞分别转染p-IL6-IL27,pIL27,pIL6,pIL27A和pEBI3质粒,转染12小时后将细胞换为无血清培养基,再过12小时后,利用H1N1、EV71、HSV-1-GFP 和VSV-eGFP分别感染细胞。12小时后收取H1N1感染的A549和EV71感染的RD,并提取细胞总RNA,反转录后进行real time-PCR检测;24小时后收取HSV-1-GFP感染的VERO 细胞,和VSV-eGFP感染的A549细胞,利用荧光倒置显微镜和流式细胞仪检测细胞感染情况。荧光实验中采用12孔板,转染2-3ug,不同组别转染量相同。
结果如图5所示,转染了p-IL6-IL-27的细胞对四种病毒包括流感病毒(IAV)、水泡型口炎病毒(VSV)、肠道病毒71(EV71)、I型单纯疱疹病毒(HSV-I)和仙台病毒(SeV) 都有抑制效果,且相对于其它亚基组,p-IL6-IL-27的抑制效果最强,且具有统计学差异。病毒感染IL-6-IL-27复合物预处理的细胞,实验组与对照组相比,病毒的复制水平得到了很大程度的抑制。通过图5C和5D我们可以看到,相比空载和其他亚基,显微镜下观察到 IL6-IL27复合物存在的组细胞病变最少,说明IL-6-IL-27复合物可以很好地保护细胞抵抗由病毒感染造成的病变。为了更加直观的体现IL-6-IL-27复合物的抗病毒效果,我们还单独定量比较了IL-6-IL-27复合物和IL-27的抗病毒效果,结果显示IL-6-IL-27复合物比IL-27有更强的抗病毒作用。
实施例6构建稳定敲减的细胞系探索IL-6-IL-27复合物主要通过哪个亚基发挥抗病毒作用
一、敲减细胞系的构建
1、将靶向IL-6、IL-27A、EBi3、IL-6R、IL-27Rα和gp130序列的寡核苷酸克隆到pLKO.1-TRC克隆载体的AgeI和BamHI位点。
2、实验用稳定敲减(KD)细胞系都使用pLKO.1-TRC克隆载体系统构建shRNA来完成。设计表3所示的shRNA靶序列交由上海生工生物公司合成含有shRNA序列的引物构建pLKO.1-shRNA质粒。所有用于shRNA质粒的插入DNA序列均列于表4。

3、重组慢病毒表达载体进行病毒包装:
(1)将构建的pLKO.1-IL-6;pLKO.1-IL-27A;pLKO.1-EBi3;pLKO.1-IL-6R;pLKO.1-IL-27RαshRNA和pLKO.1-gp130 shRNA,与慢病毒包装质粒pMD2.G(Addgene #12259),psPAX2(Addgene#12260)共转染,用PEI MAX按4:3:1的比例转染进HEK293T 细胞;按照病毒包装辅助试剂盒操作说明书进行操作;
(2)24h后,用新鲜培养基替换原来的培养基;
(3)连续两天收集病毒,用低蛋白结合过滤器(Millipore,SLHV013SL)过滤,直接使用或保存在-80℃;
(4)将细胞放入35mm的细胞培养皿中,当细胞融合度达到70%时,加入含有8mg/ml嘌呤霉素的新鲜培养基进行孵育,并加入0.1ml的慢病毒;
(5)24h后,用嘌呤霉素(A549细胞为1.0mg/ml)将病毒感染的细胞至少压力筛选3天以上,然后进行检测,获得敲减细胞系。
二、确定IL-6-IL-27复合物中发挥关键作用的亚基
将p-IL6-IL27质粒与三种不同干扰质粒shIL-6,shEBI3和shIL27A分别共转染进Huh7 或A549细胞中,转染12小时后将细胞换为无血清培养基,再过12小时后,利用SeV感染Huh7细胞,利用H1N1感染A549细胞,感染12小时后,收取细胞,提取总RNA,反转录,并用realtime PCR检测细胞内IFNα、IFNβ和IFNλ1,及其下游OAS1、PKR和MxA 表达变化情况。
图6(A-C)和图7(A-C)的结果表明干扰质粒shIL-6,shEBI3和shIL27A在Huh7 细胞和A549细胞中都能发挥作用,能显著降低胞内IL-6,EBI3和IL27A的表达。而图6 (D-E)和图7(D-E)的结果则说明在干扰IL-27A表达后,IL-6-IL-27复合物对干扰素和下游ISG的激活能力下降最为明显,说明IL-27A是该复合物中起作用的主要分子。
实施例7 IL-6-IL-27复合物激活细胞内的炎症因子
一、双荧光素酶报告基因测定
将A549细胞接种于24孔板中,将荧光素酶报告质粒按适当比例与Renilla荧光素酶质粒共同转染,pRL-TK作为内参,添加空载使每个孔中转染的DNA总量相等。转染24h后,用被动裂解缓冲液(Promega,E1941)破坏细胞,根据试剂的使用说明书,用双荧光素酶报告基因测定法(Promega,E1910)测定荧光素酶的活性。
二、鉴定IL-6-IL-27激活的炎症因子
用空载、IL-6、IL-27、IL-6-IL-27复合物的表达质粒和含有IFN-β、IFN-λ1、NF-κB或ISRE启动子的荧光素酶报告质粒共转染A549细胞,12小时后换为无血清培养基,再过12小时感染SeV,感染24小时后进行报告基因检测,pRL-TK作为对照。
此外,用空载、IL-6、IL-27或IL-6-IL-27复合表达质粒转染A549细胞,12小时后换为无血清培养基,再过12小时感染SeV或H1N1,SeV感染24小时,或H1N1感染12小时后,用qRT-PCR检测IFN-α、IFN-β、IFN-λ1、IL-1β、TNF-α、IL-32、IL-8、OAS1、PKR 和MxA的表达水平差异。
图8A的荧光素酶活性报告基因结果显示,过表达IL-6-IL-27复合物可刺激IFN-β、IFN-λ、NF-κB和ISRE启动子表达,且效果最为显著。而图8(B-C)结果显示,在SeV感染的A549细胞中,当IL-6-IL-27复合物过表达时,IFN-α、IFN-β、IFN-λ1和IL-8的mRNA 水平也随之升高;在过表达IL6-IL27复合物的A549细胞中,感染H1N1后,IFN和ISGs 的表达水平也明显增强,且IL-6-IL-27复合物的效果都是最为显著的。
实施例8 IL-6-IL-27复合物通过与MAVS相互作用调控干扰素通路
报告基因检测方法见实施例7,免疫共沉淀检测方法见实施例3。
一、鉴定IL-6-IL-27影响的通路蛋白
为了探索IL-6-IL-27复合物与IFN-β、NF-κB和ISRE信号通路上的蛋白之间的相互作用,将IFN-β、NF-κB和ISRE荧光素酶报告质粒与IL-6-IL-27复合物表达质粒与信号转导分子共转染。图9的结果表明,IL-6-IL-27复合物以剂量依赖的方式增强RIG-I、MAVS-和TRAF3触发的IFN-β、NF-κb和ISRE激活,但对TBK1-、IRF3-、p50-或p65触发的IFN-β、 NF-κb和ISRE激活影响较小。这些数据表明IL-6-IL-27复合物参与了MAVS相关TRAF3 信号转导。
二、检测IL-6-IL-27与MAVS的相互作用
接下来通过瞬时转染和CoIP实验来确定了MAVS与病毒触发信号通路中IL-6-IL-27复合物的相互作用。图10(A-C)结果显示MAVS与IL-6-IL-27复合物的三个亚基IL-6、IL-27A 和EBI3都存在相互作用。此外,我们选择了MAVS下游的TBK1作为负对照,图10(D-E) 中CoIP的结果也证实IL-6-IL-27的三个亚基与TBK1之间不存在相互作用。
实施例9 IL-6-IL-27复合物增强TRAF3和TRF6的泛素化并促进NF-κB入核
免疫共沉淀检测方法见实施例3。
一、检测IL-6-IL-27复合物对TRAF3和TRAF6泛素化的影响
在转染不同泛素和TRAF3/6的情况下共转染IL-6-IL-27复合物,随后检测IL-6-IL-27 复合物对TRAF3/6与泛素化免疫共沉淀结果的影响,图11的结果显示IL-6-IL-27复合物明显加强了TRAF3或TRAF6的泛素化,且IL-6-IL-27复合物主要增强了K63-linked的泛素化修饰,而K48-linked的泛素化效果没有变化。该结果说明IL-6-IL-27复合物能够增强TRAF3和TRAF6的K63-linked泛素化。
二、核质分离检测NF-κB入核情况
由于TRAF3和TRAF6的泛素化直接决定了下游NF-κB的入核情况,因此,进一步检测了IL-6-IL-27对NF-κB入核的影响,方法如下:
1.配制裂解液A,每1ml裂解液A中含有10ul的蛋白酶抑制剂、10ul的1M Tris-HCl、5ul的1M Mgcl2、10ul的1M NaCl、1ul的1M DTT和964ul的ddH2O。
2.配制裂解液C,每1毫升裂解液C中有20ul的1M HEPES-KOH(pH7.9)、1.5ul的 1MMgcl2、500ul的1M NaCl、1ul的1M DTT、0.4ul的0.5M EDTA、100ul的10%NP-40、 10ul的蛋白酶抑制剂和367.1ul的ddH2O。
3.收取靶标细胞置于EP管后,用PBS清洗两次,4℃离心机5000离心5分钟,吸除上清之后加入十倍沉淀体积的裂解液A,放置冰上裂解30分钟,期间15min用移液抢吹打一次。
4.加入1/20倍体积的10%的NP40,涡旋5秒,然后在冰上静置5分钟再涡旋5秒,4℃离心机3000rpm离心20分钟,此时的上清就是细胞质蛋白。
5.用5倍裂解液A重悬沉淀,冰上静置一刻钟,3000转5分钟后除去上清,加入10 倍体积裂解液C,进行超声,一刻钟后再次超声,4℃12000rpm离心30分钟,此时的上清就是细胞核蛋白溶液。
图12显示的就是IL-6-IL-27影响下细胞质和细胞核中NF-κB的两个亚基p50和p65的分布情况,可以发现IL-6-IL-27能够显著增强p50和p65的入核,从而使NF-κB入核发挥转录激活功能,促进干扰素下游基因表达。
实施例10 IL-6-IL-27复合物通过IL-27Rα和STAT1/3发挥胞外功能
一、确定IL-27Rα是IL-6-IL-27复合物的主要受体
敲减细胞系的构建详见实施例6。
由于IL-6和IL-27已知的受体包括三个,分别是IL-6R、gp130和IL-27Rα,因此首先构建了这三个受体敲减的A549细胞系,随后将IL-6-IL-27复合物转染进这三种细胞系内,并用H1N1病毒感染,随后检测病毒在胞内的复制情况,同时我们在每组细胞中也进行了回复实验。图13的结果显示敲减IL-27Rα后,IL-6-IL-27的抗病毒效果几乎消失,而当同时回复IL-27Rα表达后,该复合物的抗病毒效果也得到恢复,敲除另两种受体IL-6-IL-27的抗病毒效果都得到保留,这说明IL-27Rα是IL-6-IL-27的主要受体。
二、外源的IL-6和IL-27能直接形成IL-6-IL-27复合物并发挥抗病毒功能
免疫共沉淀检测详见实施例3.
为进一步检测IL-6-IL-27在胞外的功能,我们首先利用人工合成的hIL-6和hIL-27进行了免疫共沉淀检测,图14A的结果显示,IL-6和IL-27能直接在胞外形成IL-6-IL-27复合物。此外,图14B中,将hIL-6和hIL-27共同加入A549的无血清培养基上清中,发现其有很强的抗病毒效果,大致与IFNα相当,比单体的IL-27和IL-6显著更强。
三、IL-6-IL-27主要通过激活STAT1/3信号通路发挥胞外的抗病毒功能
由于IL-6和IL-27都是通过STAT家族成员来发挥激活功能的,因此进一步检测了IL-6-IL-27复合物对STAT1/2/3磷酸化修饰的影响,从图15的结果可以看出,IL-6-IL-27复合物主要激活了STAT1和STAT3的磷酸化,而STAT2的磷酸化几乎不受影响。
综上,通过以上一系列实验,我们提出了IL-6-IL-27复合物在病毒感染先天免疫中的作用的工作模型(如图16所示)。在该模型中,病毒感染诱导IL-6-IL-27复合物的表达。该复合物可与MAVS、TRAF6、TRAF3相互作用,通过MAVS/TRAF3/6通路诱导IFN表达。 IL-6-IL-27和MAVS/TRAF3/6复合物通过调节p50和p65与IFN启动子的结合诱导IFN的表达。IL-6-IL-27复合物分泌到细胞外环境中,可与其他细胞相互作用,诱导STAT1和STAT3 磷酸化,从而激活下游基因发挥其抗病毒功能。总之,本研究发现了一种此前未知的 IL-6-IL-27复合物,并展示了其调节病毒复制和抗病毒反应的功能,促进了我们对宿主免疫应答病毒感染的认识。
最后,还需要说明的是,术语“包括”、“包含”或者其任何其他变体意在涵盖非排他性的包含,从而使得包括一系列要素的过程、方法、物品或者设备不仅包括那些要素,而且还包括没有明确列出的其他要素,或者是还包括为这种过程、方法、物品或者设备所固有的要素。
尽管已描述了本发明的优选实施例,但本领域内的技术人员一旦得知了基本创造性概念,则可对这些实施例作出另外的变更和修改。所以,所附权利要求意欲解释为包括优选实施例以及落入本发明范围的所有变更和修改。
显然,本领域的技术人员可以对本发明进行各种改动和变型而不脱离本发明的精神和范围。这样,倘若本发明的这些修改和变型属于本发明权利要求及其等同技术的范围之内,则本发明也意图包含这些改动和变型在内。

Claims (1)
1.IL-6-IL-27复合物的表达载体在制备抗病毒药物中的应用,其特征在于,所述抗病毒药物包括抗IAV、VSV、EV71、HSV-I、SeV中的至少一种病毒的药物;所述IL-6-IL-27复合物的表达载体的构建方法包括:
从SeV感染的人THP1细胞中提取细胞总RNA,反转录获得cDNA,以所述cDNA为模板扩增获得人IL-6和IL-27的cDNA;其中,扩增人IL-6的引物对序列如SEQ ID NO.1-2所示,扩增人IL-27的引物对序列如SEQ ID NO.3-4所示;
将所述IL-6和IL-27的cDNA克隆到pcDNA3.1(+)载体的NheI/XhoI酶切位点上,获得真核表达质粒pcDNA3.1-IL-6-IL-27。
