Abstract
本发明公开了一种锌离子在抗肠道病毒EV‑D68中的应用。锌离子通过抑制病毒和感染靶细胞的表面吸附以及抑制病毒颗粒从感染细胞中释放的双重机制实现抗病毒作用。锌离子在体外对EV‑D68感染具有显著的抗病毒活性,锌盐处理可有效抑制EV‑D68病毒RNA的复制、蛋白质合成、感染性病毒颗粒的释放,并抑制细胞病变效应;此外,锌离子载体吡咯烷二硫代氨基甲酸盐(pyrrolidine dithiocarbamate,PDTC)可将锌离子输送到细胞内,增强了锌离子抗EV‑D68病毒的活性。因此,补充锌离子能够作为抗EV‑D68病毒感染的强有力的治疗策略。
Description
技术领域
本发明涉及一种锌离子在抗肠道病毒EV-D68中的应用。
背景技术
肠道病毒D68型 (EV-D68)是一种呼吸道病毒病原体,可引起严重的呼吸道疾病和神经系统症状。2014年,EV-D68病毒在美国引发了大规模呼吸道传染病的流行,引起不同程度的呼吸系统疾病,严重者出现了哮喘、肺炎等严重的呼吸道感染甚至死亡。此外,EV-D68感染还能引发无菌性脑膜炎、脑炎等神经系统并发症,导致患者出现小儿麻痹症类似的急性弛缓性脊髓炎(AFM),引起了国际公共卫生领域的广泛关注,目前还没有有效的EV-D68抗病毒药物或疫苗。
EV-D68病毒的生命周期包括病毒的粘附和侵入、基因组脱衣壳、RNA复制、蛋白质翻译、病毒颗粒组装及释放几个阶段。病毒颗粒首先与细胞表面受体结合,通过受体介导的内吞作用进入细胞后进行脱衣壳,在细胞质中释放病毒基因组RNA,病毒RNA作为模板进行蛋白翻译,产生一个多聚蛋白,再由病毒蛋白酶2A、3C和3CD水解成结构蛋白(VP0,VP1和VP3)以及非结构蛋白(2A,2B,2C,3A,3B,3C,3D)。病毒RNA的合成在病毒聚合酶3D的作用下,以正链RNA为模板生成负链RNA,再以负链RNA为模板生成新的正链RNA,新合成的病毒RNA继续作为模板进行蛋白质翻译和病毒RNA合成,最后,病毒颗粒组装及成熟后释放到细胞外。
锌离子是人体第二丰富的过渡金属离子,它在细胞命运和发育,基因转录和病毒感染等多个生物过程中起着重要的作用,锌离子对病毒感染的影响机制包括:1.锌离子能够通过破坏多聚蛋白的加工和中断病毒蛋白的合成来抑制病毒复制;2.阻断病毒蛋白酶的活性,抑制转录;3.干扰病毒和细胞膜的融合以阻断病毒的粘附和入侵;4.抑制病毒基因组复制。此外,锌通过维持免疫稳态,在增强宿主对病毒感染的抵抗力方面发挥着至关重要的作用。相反,锌的缺乏会损害细胞免疫反应,削弱细胞免疫效果。
发明内容
本发明的第一目的在于提供一种锌离子在抗肠道病毒EV-D68中的应用。
本发明的第二目的在于提供一种锌离子在抗肠道病毒EV-D68感染治疗中的医药用途。
锌离子在体外对EV-D68感染具有显著的抗病毒活性,锌盐处理可有效抑制EV-D68病毒RNA的复制、蛋白质合成、感染性病毒颗粒的释放,并抑制细胞病变效应;此外,锌离子载体吡咯烷二硫代氨基甲酸盐(pyrrolidine dithiocarbamate,PDTC)可将锌离子输送到细胞内,增强了锌离子抗EV-D68病毒的活性。因此,补充锌离子能够作为抗EV-D68病毒感染的强有力的治疗策略。
本发明首次提出,锌离子在体外能够抑制EV-D68原始株病毒的复制。
通过病毒的吸附和入侵实验证明锌离子通过抑制EV-D68病毒的吸附和入侵感染靶细胞抑制病毒复制。
通过实验证明锌离子通过抑制EV-D68病毒颗粒从感染靶细胞中释放进一步抑制病毒复制。
本发明首次提出,锌离子同样能够抑制EV-D68流行株病毒的复制。
通过在细胞中加入锌离子载体吡咯烷二硫代氨基甲酸盐,促进锌离子的细胞内转运,结果显示锌离子载体能够通过促进锌离子细胞内流从而增强锌离子的抗EV-D68病毒活性。
通过测试不同锌盐拮抗EV-D68病毒复制的能力,结果显示氯化锌、硫酸锌和醋酸锌都能够抑制EV-D68病毒复制。
本发明的有益效果在于:锌离子能够抑制EV-D68病毒吸附于靶细胞表面的能力,并且在病毒释放阶段抑制子代病毒颗粒从感染细胞中释放,通过双重抑制作用有效地阻断了EV-D68病毒感染;同时,锌离子能够广谱性抑制EV-D68流行病毒株的复制,人体血浆中锌的生理水平为10-20μM,而摄入的锌只有约10-20%被吸收,本发明中所用锌离子有效浓度EC50远低于细胞毒性浓度IC50,因此,本发明提供了一种锌补充剂可作为潜在的预防和治疗EV-D68病毒感染的用途。

参见图1至图6所示,本发明公开了一种锌离子在抗肠道病毒EV-D68中的应用。锌离子能够广谱性抑制肠道病毒EV-D68的复制,锌离子通过抑制病毒和感染靶细胞的表面吸附以及抑制病毒颗粒从感染细胞中释放的双重机制实现抗病毒作用,进一步实验证明锌离子载体能够通过促进锌离子细胞内流增强锌离子的抗病毒能力,以及多种形式的锌盐对EV-D68病毒的抑制能力相似,为EV-D68病毒的防治提供了新的思路,锌离子的补充可以应用于肠道病毒EV-D68感染的预防和治疗。
实施例1:
EV-D68病毒靶细胞感染能力体外检测:
本实施例中,EV-D68病毒感染能力检测细胞系选用人横纹肌肉瘤细胞RD作为病毒复制靶细胞,RD细胞分成2组,每组包括阴性对照细胞和EV-D68病毒感染细胞,阴性对照细胞加入无菌水,病毒感染细胞加入EV-D68病毒原始株Fermon(MOI=0.1),感染2小时后,弃上清,使用DMEM清洗2次,补入细胞培养液,第1组加入0.01mM氯化锌,第2组加入0.1mM氯化锌,继续感染48小时后,观察病毒导致的细胞病变效应(cytopathic effect,CPE),使用RD细胞测定病毒上清滴度,并通过免疫印迹实验检测细胞上清中EV-D68病毒衣壳蛋白VP1的表达情况,结果显示,氯化锌能够显著抑制EV-D68感染导致的细胞病变情况,并且呈剂量依赖性(参见图1中A部分);病毒上清滴度和病毒结构蛋白VP1的表达也呈现氯化锌剂量依赖性降低(参见图1中B、C部分),说明氯化锌能够有效抑制EV-D68病毒的复制。
实施例2:
EV-D68病毒吸附和入侵靶细胞能力检测:
本实施例中,在4度条件下检测病毒吸附能力,在37度条件下检测病毒的入侵能力(见图2中A部分),RD细胞分为2组,每组包括阴性对照细胞和EV-D68病毒感染细胞,预先使用0.1mM氯化锌处理细胞24小时,第1组在4度条件下,第2组在37度条件下,加入EV-D68病毒原始株Fermon(MOI=0.1),阴性对照组加入无菌水,孵育2小时,使用PBS清洗两次,收集细胞,提取RNA,反转录成cDNA,使用EV-D68病毒特异性引物进行RT-PCR检测细胞中病毒RNA含量,结果显示,氯化锌能够抑制EV-D68病毒的吸附和入侵(见图2中B、C部分),说明锌离子通过抑制EV-D68病毒吸附和入侵抑制病毒复制。
实施例3:
EV-D68病毒颗粒释放能力检测:
本实施例中,RD细胞分成2组,每组包括阴性对照细胞和EV-D68病毒感染细胞,阴性对照细胞加入无菌水,病毒感染细胞加入EV-D68病毒原始株Fermon(MOI=0.1),感染2小时后,弃上清,使用DMEM清洗2次,补入细胞培养液,第1组加入0.01mM氯化锌,第2组加入0.1mM氯化锌,继续感染48小时后,免疫印迹实验检测细胞上清和细胞内病毒衣壳蛋白VP1的表达情况,结果显示,氯化锌能够显著减弱细胞上清中病毒衣壳蛋白VP1的含量(见图3),说明锌离子通过抑制EV-D68病毒颗粒从靶细胞中释放抑制病毒复制。
实施例4:
锌离子抑制EV-D68流行株病毒能力检测:
为了评估氯化锌抑制EV-D68病毒的广谱性,本实施例检测了氯化锌对EV-D68流行株病毒MO(US/MO/14–18947)和KY(US/KY/14–18953)的抑制能力,RD细胞分成2组,每组包括阴性对照细胞、EV-D68流行株病毒MO(US/MO/14–18947)和KY(US/KY/14–18953)感染细胞,阴性对照细胞加入无菌水,感染2小时后,弃上清,使用DMEM清洗2次,补入细胞培养液,第1组加入0.01mM氯化锌,第2组加入0.1mM氯化锌,继续感染48小时后,观察病毒导致的细胞病变效应,使用RD细胞测定病毒上清滴度,并通过免疫印迹实验检测细胞上清中EV-D68病毒衣壳蛋白VP1的表达情况,结果显示,氯化锌能够显著抑制EV-D68流行株病毒感染导致的细胞病变情况,并且呈剂量依赖性(见图4中A部分);病毒上清滴度和病毒结构蛋白VP1的表达也呈现氯化锌剂量依赖性降低(见图4中B、C、D、E部分),说明氯化锌能够广谱性抑制EV-D68流行株病毒的复制。
实施例5:
锌离子载体促进锌离子抑制EV-D68病毒复制能力检测:
吡咯烷二硫代氨基甲酸盐(PDTC)是一种锌离子载体,能够促进锌离子进入细胞,本实施例中,RD细胞分成3组,每组包括阴性对照细胞和EV-D68病毒感染细胞,阴性对照细胞加入无菌水,病毒感染细胞加入EV-D68病毒原始株Fermon(MOI=0.1),感染2小时后,弃上清,使用DMEM清洗2次,补入细胞培养液,第1组加入0.0005mM氯化锌,第2组加入65uM PDTC,第3组联合加入0.0005mM氯化锌和65uM PDTC,继续感染48小时后,观察病毒导致的细胞病变效应,使用RD细胞测定病毒上清滴度,并通过免疫印迹实验检测细胞中EV-D68病毒衣壳蛋白VP1的表达情况,结果显示,对比氯化锌和PDTC单独处理组,联合使用氯化锌和PDTC能够显著增强氯化锌对细胞病变效应的抑制(见图5中A部分),增强氯化锌对细胞上清滴度以及细胞内病毒蛋白VP1表达的抑制(见图5中B、C部分),说明锌离子载体通过促进锌离子细胞内流增强氯化锌的抗病毒能力。
实施例6:
不同锌盐拮抗EV-D68病毒复制的能力检测:
RD细胞分成6组,每组包括阴性对照细胞和EV-D68病毒感染细胞,阴性对照细胞加入无菌水,病毒感染细胞加入EV-D68病毒原始株Fermon(MOI=0.1),感染2小时后,弃上清,使用DMEM清洗2次,补入细胞培养液,第1组加入0.01mM氯化锌,第2组加入0.1mM氯化锌,第3组加入0.01mM硫酸锌,第4组加入0.1mM硫酸锌,第5组加入0.01mM醋酸锌,第6组加入0.1mM醋酸锌,继续感染48小时后,收集细胞上清使用RD细胞测定病毒上清滴度,结果显示,氯化锌、硫酸锌和醋酸锌均能够显著抑制上清中EV-D68病毒滴度,并且呈剂量依赖性(见表1),说明不同锌盐对EV-D68病毒复制均具有抑制作用。

实施例7:
锌离子抗病毒有效浓度和细胞毒性测试:
本实施例中,对锌离子抑制EV-D68病毒复制的有效剂量和细胞毒性进行测试,在RD细胞中检测不同浓度的氯化锌对细胞内EV-D68病毒RNA含量的抑制情况,结果显示氯化锌的抗病毒作用半数有效浓度EC50为0.033mM(见图6中A部分);同时使用MTS方法测试不同浓度氯化锌的细胞毒性,结果显示氯化锌的IC50值为0.26mM(见图6中B部分),说明氯化锌的有效浓度远远小于其细胞毒性,因此,提示锌离子能够应用于预防和治疗EV-D68病毒感染。
以上所述仅为本发明的优选实例而已,并不用于限制本发明,对于本领域的技术人员来说,本发明可以有各种更改和变化。凡对本发明所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。
Claims (6)
1.一种锌离子在抗肠道病毒EV-D68中的应用。
2.根据权利要求1所述的锌离子在抗肠道病毒EV-D68中的应用,其特征在于:锌离子通过抑制病毒和感染靶细胞的表面吸附以及抑制病毒颗粒从感染细胞中释放的双重机制实现抗病毒作用。
3.根据权利要求1所述的锌离子在抗肠道病毒EV-D68中的应用,其特征在于:在细胞中加入锌离子载体吡咯烷二硫代氨基甲酸盐,促进锌离子的细胞内转运,锌离子载体通过促进锌离子细胞内流从而增强锌离子的抗EV-D68病毒活性。
4.根据权利要求1或2或3所述的锌离子在抗肠道病毒EV-D68中的应用,其特征在于:所述的锌离子为氯化锌、硫酸锌和醋酸锌。
5.根据权利要求4所述的锌离子在抗肠道病毒EV-D68中的应用,其特征在于:所述的锌离子有效浓度EC50。
6.一种锌补充剂在预防和治疗肠道病毒EV-D68感染的医药用途。
