Abstract

背景技术
自然杀伤(Natural killer,NK)细胞是人体先天免疫的核心组成部分,在机体抗肿瘤免疫中发挥着重要作用。NK细胞具有区分正常细胞和肿瘤细胞的能力,可非特异性直接杀伤肿瘤细胞,这种天然杀伤活性既不需要预先由抗原致敏,也不需要抗体参与,具有主要组织相容性复合体(major histocompatibility complex,MHC)非限制性杀伤的特点。NK细胞的表型是CD3-CD56+,根据NK细胞表面CD16分子的表达情况,可将NK细胞进一步分为CD16neg、CD16dim和CD16bright三个亚群。CD16bright NK细胞可有效地发挥细胞毒作用,并能产生一定水平的细胞因子,而CD16neg和CD16dim NK细胞可产生高浓度的细胞因子,但是细胞毒性较低。
NK细胞对肿瘤细胞的杀伤主要包括以下四种途径:(1)穿孔素和颗粒酶介导的直接杀伤;(2)膜肿瘤坏死因子家族分子(如FasL、TRIAL)介导的肿瘤细胞凋亡;(3)通过分泌多种细胞因子,如γ-干扰素(γ-IFN)和肿瘤坏死因子(TNF)-α等,协同抑制或杀伤肿瘤细胞;(4)NK细胞表面表达免疫球蛋白Fc片段的低亲和力受体CD16分子(FcγRⅢ),当它与抗体结合后,会诱导抗体依赖性细胞毒性作用(antibody dependent cell cytotoxicity,ADCC),特异性杀伤与抗体IgG结合的肿瘤细胞。
CD137,又称4-1BB或肿瘤坏死因子受体9(TNFR9),是肿瘤坏死因子受体共刺激家族的一员,在活化的NK细胞和T细胞表面广泛表达。它的配体CD137L,或称为4-1BBL,通常表达于抗原提呈细胞(Antigen presenting cell,APC)表面。已知NK细胞中CD137传导信号的激活可以促进NK细胞的增殖,但对其具体作用机制仍知之甚少。
乳腺癌是女性中最常见的恶性肿瘤之一。近年来我国乳腺癌的发病率及死亡率均呈逐年上升的趋势。三阴性乳腺癌(trip-negative breast cancer,TNBC)是乳腺癌的一种特殊亚型,是指雌激素受体(estrogen receptor,ER)、孕激素受体(progesteronereceptor,PR)以及人表皮生长因子受体2(human epidermal growth factor receptor-2,Her2)均为阴性的乳腺癌。TNBC具有侵袭性强、易复发、进展快、生存期短、预后差等临床特点,乳腺癌的常规治疗手段,如内分泌治疗及抗Her-2靶向治疗对TNBC通常无效。
乳腺癌的转移是导致女性癌症死亡的主要原因。晚期转移性乳腺癌患者通常出现抗肿瘤免疫缺陷,如树突状细胞(Dendritic cell,DC)成熟度和功能受到抑制、调节性T细胞(Regulatory T cell,Treg)浸润增加、自然杀伤(Natural killer,NK)细胞对肿瘤细胞的细胞毒功能受到严重损害。其中NK细胞对乳腺癌的进展和预后尤为重要,NK细胞的功能受损可导致抗肿瘤免疫逃逸,一定程度上促进了乳腺癌的发展和转移。
发明内容
本发明的目的是提供CD137抗体在制备促进NK细胞表达CD16分子和/或促进NK细胞分泌细胞毒性因子的药物中的应用,旨在解决现有的肿瘤治疗药物、特别是乳腺癌治疗药物中存在的特异性差,进而导致治疗失败、预后差等问题。
为了实现上述发明目的,本发明采用的技术方案如下:
本发明一方面提供了CD137抗体在制备促进NK细胞表达CD16分子和/或促进NK细胞分泌细胞毒性因子的药物中的应用。
本发明另一方面提供了一种促进NK细胞表达CD16分子和/或促进NK细胞分泌细胞毒性因子的药物,其包括CD137抗体。
本发明再一方面提供了CD137抗体在制备促进NK细胞的细胞杀伤作用的药物中的应用。
本发明最后一方面提供了一种促进NK细胞的细胞杀伤作用的药物,其包括CD137抗体。
本发明通过将CD137抗体作用于NK细胞,既可以显著提高NK细胞中CD16的表达比例,又可显著促进NK细胞分泌更多的细胞毒性因子,因此,本发明CD137抗体可用于制备促进NK细胞表达CD16分子和/或促进NK细胞分泌细胞毒性因子的药物。
本发明促进NK细胞表达CD16分子和/或促进NK细胞分泌细胞毒性因子的药物,由于包括CD137抗体,该抗体具有提高NK细胞中CD16的表达比例及促进NK细胞分泌细胞毒性因子的双重作用,因此,包括CD137抗体的药物也具有促进NK细胞表达CD16分子和/或促进NK细胞分泌细胞毒性因子的作用。
CD137抗体可提高CD16的表达比例和促进NK细胞分泌更多的细胞毒性因子,由于CD16分子是介导NK细胞ADCC杀伤途径的关键分子,同时,NK细胞发挥杀伤作用的途径之一是通过分泌细胞毒性因子,因此,CD137可用于制备促进NK细胞的细胞杀伤作用的药物。
本发明促进NK细胞的细胞杀伤作用的药物,由于包括CD137抗体,且该抗体既可促进NK细胞表达CD16分子,又可促进NK细胞分泌细胞毒性因子,因此该药物可通过促进NK细胞表达CD16分子和分泌细胞毒性因子,提升NK细胞的细胞杀伤作用。

本发明实施例提供了CD137抗体在制备促进NK细胞表达CD16分子和/或促进NK细胞分泌细胞毒性因子的药物中的应用。
外周血来源NK细胞中存在部分CD16分子低表达或不表达的NK细胞,本发明实施例通过将CD137抗体制备促进NK细胞表达CD16分子的药物,可以显著提高NK细胞中CD16的表达比例。同时,CD137抗体又可显著促进NK细胞分泌更多的细胞毒性因子,可用于制备促进NK细胞分泌细胞毒性因子的药物。
经试验发现,CD137抗体处理后的NK细胞分泌IFN-γ和TNF-α的浓度显著提升。因此,在一些实施例中,将CD137抗体用于制备促进NK细胞分泌IFN-γ和TNF-α的药物。
本发明实施例还提供了一种促进NK细胞表达CD16分子和/或促进NK细胞分泌细胞毒性因子的药物,其包括CD137抗体。
本发明实施例的促进NK细胞表达CD16分子和/或促进NK细胞分泌细胞毒性因子的药物,由于包括CD137抗体,该抗体具有提高NK细胞中CD16的表达比例及促进NK细胞分泌细胞毒性因子的双重作用,因此,包括CD137抗体的药物也具有促进NK细胞表达CD16分子和/或促进NK细胞分泌细胞毒性因子的作用。
由于CD16可以介导NK细胞通过ADCC对肿瘤细胞进行杀伤,且NK细胞对肿瘤细胞发挥杀伤作用的途径之一是通过分泌细胞毒性因子,因此,在一些实施例中,将上述促进NK细胞表达CD16分子和/或促进NK细胞分泌细胞毒性因子的药物作为抗肿瘤药物使用,可对肿瘤细胞产生杀伤作用。
西妥昔单抗在临床上常用于EGFR阳性肿瘤的靶向治疗。在一些实施例中,将西妥昔单抗与上述促进NK细胞表达CD16分子的药物联用,可共同作用于NK细胞,一方面,CD137抗体促进NK细胞表达CD16分子;另一方面,西妥昔单抗的抗原结合片段(fragment ofantigen binding,Fab)可与肿瘤细胞表面的EGFR蛋白特异性结合,另一端Fc段可与NK细胞表面的CD16分子(FcγRIII)相结合,从而进一步促进NK细胞的活化及对肿瘤细胞的特异性杀伤。
在一些实施例中,将西妥昔单抗与上述促进NK细胞表达CD16分子的药物联用,可用于治疗乳腺癌,特别是三阴性乳腺癌。这是因为EGFR在大约60%的三阴性乳腺癌患者中高表达,西妥昔单抗可以与三阴性乳腺癌患者中高表达的EGFR蛋白特异性结合,通过促进NK细胞的活化,加强其对乳腺癌细胞的特异性杀伤效果。
在一些实施例中,将西妥昔单抗与上述促进NK细胞分泌细胞毒性因子的药物联用,可共同作用于NK细胞。一方面,CD137抗体促进NK细胞分泌细胞毒性因子作用并杀伤肿瘤细胞;另一方面,西妥昔单抗的抗原结合片段(fragment of antigen binding,Fab)可与肿瘤细胞表面的EGFR蛋白特异性结合,从而进一步促进NK细胞对肿瘤细胞的特异性杀伤。
在一些实施例中,将西妥昔单抗与上述促进NK细胞分泌细胞毒性因子的药物联用,可用于治疗乳腺癌,特别是三阴性乳腺癌。这是因为EGFR在大约60%的三阴性乳腺癌患者中高表达,西妥昔单抗可以与三阴性乳腺癌患者中高表达的EGFR蛋白特异性结合,通过促进NK细胞分泌细胞毒性因子,加强其对乳腺癌细胞的特异性杀伤效果。
在一些实施例中,将西妥昔单抗与上述促进NK细胞表达CD16分子的药物及上述促进NK细胞分泌细胞毒性因子的药物联用,可通过上述两种机制进一步促进西妥昔单抗对肿瘤细胞、特别是三阴性乳腺癌细胞的特异性杀伤效果。
本发明实施例还提供了CD137抗体在制备促进NK细胞的细胞杀伤作用的药物中的应用。
CD137抗体可提高CD16的表达比例和促进NK细胞分泌更多的细胞毒性因子,由于CD16分子是介导NK细胞ADCC杀伤途径的关键分子,同时,NK细胞发挥杀伤作用的途径之一是通过分泌细胞毒性因子,因此,CD137可用于制备促进NK细胞的细胞杀伤作用的药物。
在一些实施例中,由于CD16可以介导NK细胞通过ADCC对肿瘤细胞进行杀伤,且NK细胞对肿瘤细胞发挥杀伤作用的途径之一是通过分泌细胞毒性因子,因此,CD137用于制备促进NK细胞的细胞杀伤作用的药物时,所得药物尤其适用于促进NK细胞对肿瘤细胞的特异性杀伤作用。
本发明实施例还提供了一种促进NK细胞的细胞杀伤作用的药物,其包括CD137抗体。
本发明实施例的促进NK细胞的细胞杀伤作用的药物,由于包括CD137抗体,且该抗体既可促进NK细胞表达CD16分子,又可促进NK细胞分泌细胞毒性因子,因此该药物可通过促进NK细胞表达CD16分子和分泌细胞毒性因子,提升NK细胞的细胞杀伤作用。
在一些实施例中,由于CD16可以介导NK细胞通过ADCC对肿瘤细胞进行杀伤,且NK细胞对肿瘤细胞发挥杀伤作用的途径之一是通过分泌细胞毒性因子,因此,本发明实施例的促进NK细胞的细胞杀伤作用的药物尤其适用于促进NK细胞对肿瘤细胞的特异性杀伤作用。
在一些实施例中,将西妥昔单抗与促进NK细胞对肿瘤细胞的杀伤作用的药物联用,可提升西妥昔单抗对乳腺癌细胞的杀伤作用。
进一步地,由于西妥昔单抗可以与EGFR蛋白特异性结合,在一些实施例中,将西妥昔单抗与促进NK细胞对肿瘤细胞的杀伤作用的药物联用,可用于杀灭EGFR阳性的肿瘤细胞。同时,经实验发现,西妥昔单抗与促进NK细胞对肿瘤细胞的杀伤作用的药物联用联用时,两者会发生协同效应,对EGFR阳性的肿瘤细胞的杀伤作用显著高于西妥昔单抗以及促进NK细胞对肿瘤细胞的杀伤作用的药物联用单独对EGFR阳性的肿瘤细胞的杀伤作用。
此外,西妥昔单抗可以与EGFR蛋白特异性结合,三阴性乳腺癌患者中又高表达EGFR蛋白,因此,在一些实施例中,将西妥昔单抗与上述促进NK细胞对肿瘤细胞特异性杀伤的药物联用,可进一步提升NK细胞对三阴性乳腺癌细胞的特异性杀伤效果。
为使本发明上述实施细节和操作能清楚地被本领域技术人员理解,以及本发明实施例CD137抗体在制备促进NK细胞表达CD16分子和/或促进NK细胞分泌细胞毒性因子的药物中的应用的进步性能显著的体现,以下通过实施例来举例说明上述技术方案。
实施例1
(1)NK细胞的培养:用人淋巴细胞分离液从健康志愿者来源的肝素钠抗凝外周血中分离外周血单个核细胞(Peripheral blood mononuclear cell,PBMC),按照NK细胞处理试剂盒和NK细胞无血清培养基的说明书,培养NK细胞。后续根据NK细胞生长状况用NK细胞无血清培养基进行补液。在培养的第0、8、12、16天,取样计数。培养第8天和第16天时NK细胞的显微拍摄图如图1所示。经过16天的诱导培养,细胞数量由起始的3×106个细胞扩增到1.2×109个细胞,约扩增400倍,细胞生长曲线图如图2所示;
(2)NK细胞的表型检测:取1×106个诱导培养第16天的NK细胞,DPBS洗一遍后,参照流式抗体使用说明书用CD3-FITC、CD56-PE、CD16-APC或CD137-APC进行染色,常温避光孵育30min,DPBS洗一遍后,用0.5mL DPBS重悬细胞。流式细胞仪检测分析NK细胞表型,每个样本至少分析10000个细胞。流式检测散点图如图3所示,CD3-CD56+NK细胞比例为85.01﹪(图3-b),其中77.21﹪的NK细胞表面表达CD137分子(图3-c);
(3)CD137抗体处理NK细胞:在NK细胞培养体系中加入10μg/mL CD137抗体处理4h后,用流式细胞术检测NK细胞表面CD16分子表达比例,以未加抗体处理组作为阴性对照组,流式检测散点图见图4,详细检测结果如下:阴性对照组CD16+NK细胞比例为(79.57±0.92)﹪(图4-a),CD137抗体处理组CD16+NK细胞比例为(90.43±0.67)﹪(图4-b)。CD137抗体处理组CD16表达比例显著高于阴性对照组,差异有显著性(P<0.05),统计结果如图5所示。这一结果表明CD137处理NK细胞可促进NK细胞表面CD16分子的表达。
实施例2
(4)NK细胞的培养:用人淋巴细胞分离液从健康志愿者来源的肝素钠抗凝外周血中分离外周血单个核细胞(Peripheral blood mononuclear cell,PBMC),按照NK细胞处理试剂盒和NK细胞无血清培养基的说明书,培养NK细胞。后续根据NK细胞生长状况用NK细胞无血清培养基进行补液。在培养的第0、8、12、16天,取样计数。培养第8天和第16天时NK细胞的显微拍摄图如图1所示。经过16天的诱导培养,细胞数量由起始的3×106个细胞扩增到1.2×109个细胞,约扩增400倍,细胞生长曲线图如图2所示;
(5)NK细胞的表型检测:取1×106个诱导培养第16天的NK细胞,DPBS洗一遍后,参照流式抗体使用说明书用CD3-FITC、CD56-PE、CD16-APC或CD137-APC进行染色,常温避光孵育30min,DPBS洗一遍后,用0.5mL DPBS重悬细胞。流式细胞仪检测分析NK细胞表型,每个样本至少分析10000个细胞。流式检测散点图如图3所示,CD3-CD56+NK细胞比例为85.01﹪(图3-b),其中77.21﹪的NK细胞表面表达CD137分子(图3-c);
(6)取1×106个培养第16天的NK细胞,用10μg/mL CD137抗体处理4h后,1000rpm离心5min后,吸取上清,未加入CD137抗体的NK细胞培养上清作为阴性对照。根据使用说明书,利用ELISA检测试剂盒对两组NK培养上清中分泌的TNF-α、IFN-γ浓度进行检测,实验步骤如下:
包被:用包被缓冲液将捕获抗体稀释至建议浓度,在96孔酶标板中加0.1mL,4℃放置过夜;弃去孔内溶液,用PBST洗涤液(含0.5﹪Tween-20的PBS溶液)洗板3次;
封闭:每孔加入200μL的封闭液,置于室温封闭1h,洗板3次;
加样品:设立6个标准孔,每孔均设立3个复孔,用稀释缓冲液稀释标准品至浓度为2ng/mL,进行孔内稀释,终浓度分别为:500pg/mL,250pg/mL,125pg/mL,62.5pg/mL,31.25pg/mL,0pg/mL;同时把两组NK细胞培养上清液加入到设定的孔中,100μL/孔,设空白对照和阴性对照孔,置于℃过夜。弃去孔内溶液,用PBST洗涤液洗板5次;
加二抗:用稀释缓冲液将检测抗体稀释至建议浓度,每孔加入100μL,置于室温孵育1h,弃去孔内溶液,用PBST洗涤液洗板5次;
加Avidin-HRP:用稀释缓冲液将Avidin-HRP稀释至建议浓度,每孔加入100μL,置于室温孵育30min,弃去孔内溶液,用PBST洗涤液洗板5-7次;
显色:每孔加入100μL的TMB显色液,室温避光反应20分钟;
终止:每孔加入100μL的终止液。
结果显示:将96孔板放入全自动酶标仪中检测450nm波长处的OD值,根据公式计算两组NK细胞培养上清中TNF-α和IFN-γ的浓度。详细检测结果如下:阴性对照组NK细胞培养上清中IFN-γ浓度为(388.90±7.02)pg/mL,CD137抗体组NK细胞培养上清中IFN-γ浓度为(523.90±1.90)pg/mL,显著高于阴性对照组,差异有显著性(P<0.01),统计结果如图6所示;阴性对照组NK细胞培养上清中TNF-α浓度为(20.59±4.09)pg/mL,CD137抗体组NK细胞培养上清中TNF-α浓度为(47.22±2.14)pg/mL,显著高于阴性对照组,差异有显著性(P<0.05),统计结果如图7所示。
实施例3
(7)免疫荧光染色:利用EGFR抗体及绿色荧光二抗特异性标记乳腺癌细胞MDA-MB-453和MDA-MB-231中的EGFR蛋白,以检测EGFR蛋白的表达水平;同时利用Phalloidin-Atto565标记细胞中的肌动蛋白(Actin),分别在细胞爬片上接种5000个乳腺癌细胞MDA-MB-231、MDA-MB-453,37℃培养箱中继续培养过夜;吸弃培养孔中的培养基,DPBS洗涤一遍,加入4﹪甲醛溶液,室温孵育20min;吸弃4﹪甲醛溶液,加入DPBS洗涤2次;加入0.1﹪PBST(含0.1﹪Triton X-100的PBS溶液)室温处理5min后,用DPBS洗涤3次;加入5﹪BSA封闭液,室温孵育1小时;吸弃封闭液,加入200μL EGFR抗体稀释液,放入4℃冰箱孵育过夜;吸弃一抗溶液,加入0.1﹪PBST溶液洗涤3次;加入200μL的Alexa Fluor 488标记山羊抗兔IgG及Phalloidin-Atto 565稀释液,室温孵育1小时后,用0.1﹪PBST洗涤3次;用封片剂进行封片,避光放置,封片剂干燥后即可进行荧光拍摄。免疫荧光染色图(图8)结果显示,MDA-MB-231细胞中高表达EGFR蛋白(图8左下,实为绿色通道),而MDA-MB-453细胞中EGFR蛋白表达水平相对较低。图8中白色箭头所指处(实为绿色亮点)是MDA-MB-231细胞中高水平表达的EGFR蛋白,与Actin蛋白(图8中下,实为红色通道)的表达没有重合,说明该EGFR蛋白表达的检测是特异性的;
(8)Western Blot检测:对乳腺癌细胞MDA-MB-453和MDA-MB-231中的EGFR蛋白进行蛋白印迹检测,以β-Actin蛋白作为内对照。取适当量的裂解液,在使用前数分钟内加入PMSF,使PMSF的最终浓度为1mM;去除六孔板中乳腺癌细胞MDA-MB-231、MDA-MB-453的培养液,用PBS洗一遍。每孔加入200μL裂解液;充分裂解后,14000g离心5分钟,取上清,利用BCA法对蛋白浓度进行定量。取含相同质量蛋白质的上清液,加入蛋白上样缓冲液后95℃以上煮蛋白10min,使之变性后,-80℃冰箱保存备用;用Biofuraw Precast Gel预制胶进行上样,恒压80V条件跑胶30min,再更换成100V条件跑胶1h;使用已经预冷的转膜液替换电泳转膜装置中的电泳液,根据操作说明将上述实验完成后的分离胶转入转膜夹中,放入合适大小的PVDF膜,恒压100V条件下转膜1h;使用5﹪脱脂牛奶对PVDF膜进行封闭处理,室温放置1h;用5﹪脱脂牛奶按照1:1000的比例稀释EGFR抗体和β-actin抗体,充分混匀,4℃孵育PVDF膜并将其放置在水平摇床上(50rpm,1h);孵育结束后,用PBST溶液洗膜3次,每次10min;使用新配制5﹪脱脂牛奶以1:10000的比例稀释对应的荧光二抗,充分混匀,常温避光孵育PVDF膜1h;孵育结束后,用PBST溶液洗膜3次;洗膜完成后,使用Odyssey双色红外荧光扫描成像系统进行拍照。WB结果显示,MDA-MB-231细胞中高表达EGFR蛋白,而MDA-MB-453细胞几乎不表达EGFR蛋白(图9);
(9)NK细胞的培养:用人淋巴细胞分离液从健康志愿者来源的肝素钠抗凝外周血中分离外周血单个核细胞(Peripheral blood mononuclear cell,PBMC),按照NK细胞处理试剂盒和NK细胞无血清培养基的说明书,培养NK细胞。后续根据NK细胞生长状况用NK细胞无血清培养基进行补液。在培养的第0、8、12、16天,取样计数。培养第8天和第16天时NK细胞的显微拍摄图如图1所示。经过16天的诱导培养,细胞数量由起始的3×106个细胞扩增到1.2×109个细胞,约扩增400倍,细胞生长曲线图如图2所示;

(11)以NK细胞作为效应细胞,乳腺癌细胞系MDA-MB-231和MDA-MB-453作为杀伤靶细胞,用LDH法检测NK细胞在CD137抗体和(或)西妥昔单抗作用下对靶细胞的杀伤作用。将乳腺癌细胞系MDA-MB-453和MDA-MB-231细胞分别与NK细胞共孵育4小时,或单独与抗CD137单抗(10μg/mL)、西妥昔单抗(10μg/mL)或西妥昔单抗加抗CD137单抗(均为10μg/mL)孵育4小时。用LDH法检测各组条件下对乳腺癌细胞MDA-MB-453和MDA-MB-231的杀伤比例。实验分组如表1所示,实验步骤如下:在96孔细胞培养板中,按照NK细胞:乳腺癌细胞=2.5:1的效靶比设置反应体系,并按照分组加入相应抗体,同时设置无细胞的培养液孔(扣除背景)、只有靶细胞的样品对照孔以及只有靶细胞用于后续裂解的孔(样品最大酶活性孔),每组设置3个复孔。37℃、5﹪CO2培养箱中培养4h。在预定的检测时间点前1h,在样品最大酶活性孔中加入试剂盒提供的LDH释放试剂,反复吹打数次混匀,然后继续在细胞培养箱中孵育1h。将细胞培养板用多孔板离心机400g离心5min。分别取各孔的上清液80μL,加入到一新的96孔板相应孔中;各孔分别加入40μL LDH检测工作液,充分混匀,室温避光孵育30min后,将96孔板放入全自动酶标仪中检测490nm波长处的吸光度。细胞杀伤比例计算公式如下:细胞毒性或死亡率(﹪)=(实验组样品吸光度-样品对照孔吸光度)/(样品最大酶活性的吸光度-样品对照孔吸光度)×100﹪。检测结果如图10所示。在CD137抗体和西妥昔单抗同时存在的条件下,NK细胞对MDA-MB-231细胞的杀伤比例为(62.71±0.79)﹪(图10-h组右列),显著高于单独NK细胞组((15.34±0.08)﹪,图10-e组右列)、CD137抗体处理组((22.09±1.86)﹪,图10-f组右列)和西妥昔单抗处理组((45.34±0.44)﹪,图10-g组右列);但是,针对EGFR低表达乳腺癌细胞MDA-MB-453,CD137抗体对NK细胞的ADCC作用影响不明显,单独NK细胞的杀伤比例为(25.08±1.83)﹪(图10-e组左列),CD137抗体加NK细胞组的杀伤比例为(31.24±0.06)﹪(图10-f组左列),西妥昔单抗加NK细胞组的杀伤比例为(26.70±0.83)﹪(图10-g组左列),CD137抗体加西妥昔单抗加NK细胞组的杀伤比例为(32.20±0.23)﹪(图10-h组左列)。上述结果显示,经抗CD137单抗刺激后,NK细胞对EGFR高表达乳腺癌细胞MDA-MB-231发挥出更强的ADCC介导的杀伤作用。

本发明上述各实施例均采用Graphpad prism 5.0软件对实验数据进行统计分析,实验结果以表示。两组NK细胞的细胞表型、细胞因子分泌量及对乳腺癌细胞系的杀伤能力的比较采用两样本t检验。以P<0.05为差异有统计学意义。
以上所述实施例仅表达了本发明的几种实施方式,其描述较为具体和详细,但并不能因此而理解为对本发明专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进,这些都属于本发明的保护范围。因此,本发明专利的保护范围应以所附权利要求为准。
Claims (10)
1.CD137抗体在制备促进NK细胞表达CD16分子和/或促进NK细胞分泌细胞毒性因子的药物中的应用。
2.根据权利要求1所述的应用,其特征在于,所述细胞毒性因子为γ-干扰素和/或肿瘤坏死因子-α。
3.一种促进NK细胞表达CD16分子和/或促进NK细胞分泌细胞毒性因子的药物,其特征在于,所述促进NK细胞表达CD16分子和/或促进NK细胞分泌细胞毒性因子的药物包括CD137抗体。
4.根据权利要求3所述促进NK细胞表达CD16分子和/或促进NK细胞分泌细胞毒性因子的药物,其特征在于,所述促进NK细胞表达CD16分子和/或促进NK细胞分泌细胞毒性因子的药物包括西妥昔单抗。
5.CD137抗体在制备促进NK细胞的细胞杀伤作用的药物中的应用。
6.根据权利要求5所述的应用,其特征在于,所述促进NK细胞的细胞杀伤作用的药物为促进NK细胞对肿瘤细胞的杀伤作用的药物。
7.一种促进NK细胞的细胞杀伤作用的药物,其特征在于,所述促进NK细胞的细胞杀伤作用的药物包括CD137抗体。
8.根据权利要求7所述的促进NK细胞的细胞杀伤作用的药物,其特征在于,所述促进NK细胞的细胞杀伤作用的药物为促进NK细胞对肿瘤细胞的杀伤作用的药物。
9.根据权利要求8所述促进NK细胞的细胞杀伤作用的药物,其特征在于,所述促进NK细胞对肿瘤细胞的杀伤作用的药物包括西妥昔单抗。
10.根据权利要求8或9所述促进NK细胞的细胞杀伤作用的药物,其特征在于,所述肿瘤细胞为表皮生长因子受体阳性的肿瘤细胞。
