Description
Technical Field
The invention relates to the technical field of bioactive substance extraction, in particular to a method for extracting coix seed active substances.
Background
The coix seed is taken as a traditional medicinal and edible plant in China, has rich nutritional value and medicinal value, has the effects of promoting diuresis, removing dampness, strengthening spleen, relieving diarrhea, removing arthralgia, expelling pus, detoxicating and resolving hard mass, and can be used for treating diseases such as edema, beriberi, dysuria, spleen deficiency diarrhea, damp arthralgia, pulmonary abscess, intestinal abscess, wart, cancer and the like. In recent years, students at home and abroad research on chemical components of coix seeds by TLC, HPLC, GC and the like, and find that the coix seeds contain various active substances, mainly comprising coix seed esters, triglycerides, fatty acids, lactams, coix seed lactones, saccharides, sterols, polyphenols, polysaccharides, proteins and the like, and further comprise other components, such as flavonoids, triterpenes, alkaloids and the like. Is beneficial to modern scientific technology, and coix seed active substances are widely applied in the industries of foods, nourishments, cosmetics, medicines and the like.
At present, the extraction method of the active ingredients of coix seeds is usually a traditional water extraction method, and in order to improve the extraction efficiency, related researchers propose a plurality of auxiliary methods based on the water extraction method, such as an enzyme auxiliary method, an ultrasonic auxiliary water extraction method, a microwave auxiliary method and an ethanol water extraction method, but the effect of the auxiliary methods on improving the extraction rate is still not obvious.
Disclosure of Invention
The invention aims to provide the extraction method of the coix seed active substance, which improves the extraction efficiency of the coix seed active substance, and the added auxiliary agents are all conventional raw materials of cosmetics, so that the dosage is small, the content in the finished product is small, separation is not needed, and the extracted solid can be directly used for the production of cosmetics.
The aim of the invention is achieved by the following technical scheme:
a method for extracting coix seed active substances comprises the following steps:
s1: taking coix seed raw material, and preprocessing to obtain coix seed powder;
s2: mixing the coix seed powder obtained in the step S1 with water and an auxiliary agent to obtain an extracting solution, and then extracting to obtain the coix seed active substance, wherein the auxiliary agent contains amino.
In step S1, the pretreatment process specifically includes: pulverizing Coicis semen, sieving, and drying to constant weight.
Sieving with 40 mesh sieve.
In step S2, the auxiliary agent includes one or more of betaine, triethanolamine or urea.
In the step S2, the dosage of the auxiliary agent is 0.1-20% of the weight of the coix seed powder.
In the step S2, the liquid-to-material ratio of water to coix seed powder is 5:1 (mL/g).
In step S2, extraction is performed in a thermostatic water bath.
In step S2, the extraction temperature is 80deg.C, and the extraction time is 0-120min, but not 0.
In the step S2, after the extraction is completed, extracting solution is filtered, upper liquid phase is collected, and coix seed active substances are obtained after drying.
In the step S2, after the extraction is completed, centrifuging the extracting solution, collecting an upper liquid phase, and drying to obtain the coix seed active substance.
The invention is based on the traditional water extraction method (the method takes water as a solvent, heats or boils substances, dissolves active ingredients in the water, finally separates out the substances stable to humidity and heat), adds proper amino-containing auxiliary agents (which are common additives in the fields of cosmetics and foods), and then obtains coix seed active substances through centrifugation or filtration, thereby effectively improving the extraction efficiency of the active substances. In the extraction process, the amino group on the amino group-containing auxiliary agent plays the following roles: 1, hydrogen bonding. Can form hydrogen bond with hydroxyl groups on cell walls and cell membranes, and promote dissolution of polyphenol structures. 2. The amino group has positive electrical properties. Amino groups can bind to cellulose or the like having a negative charge, changing its electrical properties. Similar to the principle of eutectic extraction. According to the types of the auxiliary agents, the auxiliary agents with different dosages are added to obtain the maximum yield of the active substances of the coix seeds.
