技术领域
本发明属于材料和荧光成像技术领域,涉及一种藻蓝蛋白-酪蛋白/多孔淀粉微凝胶及其制备方法和应用。
背景技术
荧光蛋白—藻蓝蛋白(PC)是FDA在美国批准的唯一天然蓝色颜料。在欧盟,藻蓝蛋白已被列为有色食品原料,在食品中的使用没有含量限制。然而,由于藻蓝蛋白的稳定性较差,该蛋白质在高温环境下易于失活和降解,在食品中使用时容易褪色,这使得PC的价值无法得到充分利用。
微凝胶是一种内部交联的微观三维网络,由分散在合适溶剂中的交联聚合物分子组成。它们可以均匀地分散在溶剂中并溶胀。因此,它被广泛用作药物、营养、代谢产物和基因传递,可以显著改善药代动力学特性和靶向药物的生物分布。
通常,对于微凝胶在生物医学,组织工程和食品结构中的许多应用,生物降解性和生物相容性是非常重要的问题。因此,与传统化学材料相比,由天然大分子制备的微凝胶具有显著优势。另外,微凝胶与负载的生物活性物质之间的相互作用较弱,因此可以长时间维持药物的生物活性。然而,这些生物大分子载体仍然存在诸如低负载等问题。
发明内容
为了解决现有技术存在的问题,本发明提供一种藻蓝蛋白-酪蛋白/多孔淀粉微凝胶及其制备方法和应用,采用多孔淀粉和酪蛋白作为原料,制备得到微凝胶基体,在基体上负载藻蓝蛋白,得到高负载率且可用于荧光成像的微凝胶;本发明合成原理和过程简单,具有负载率高、缓释效果优良、荧光性能好、成本低等优点。
为了实现上述目的,本发明采用的技术方案是:
一种藻蓝蛋白-酪蛋白/多孔淀粉微凝胶的制备方法包括以下步骤:
1)制备酪蛋白/多孔淀粉微凝胶,备用;
2)在避光条件下,将藻蓝蛋白粉末溶解于酪蛋白/多孔淀粉微凝胶中,搅拌均匀,得到产物藻蓝蛋白-酪蛋白/多孔淀粉微凝胶。
进一步的,所述步骤2),产物中藻蓝蛋白浓度为0.40~0.60mg/mL;溶解温度20~25℃;搅拌时间1~2h。
进一步的,所述步骤1)中酪蛋白/多孔淀粉微凝胶的制备过程是:
1.1)将淀粉溶于水形成淀粉悬浮液;
1.2)向淀粉悬浮液中加入复合酶,再加入柠檬酸-柠檬酸钠缓冲液调节pH为5.8~6.0,经复合酶水解反应、分离后,将得到的沉淀物经洗涤、干燥、破碎和筛分,得到多孔淀粉;
1.3)将上述得到的多孔淀粉与酪蛋白混合,调节pH为6.8~7.2,经加热、均质,得到酪蛋白/多孔淀粉微凝胶。
进一步的,所述步骤1.2)中,淀粉悬浮液与复合酶液的体积比为9~11:1~2;所述淀粉悬浮液浓度为8~12mg/mL;所述复合酶液的酶活力比为糖化酶:α-淀粉酶=3000:2000~2400IU/mL;水解温度55~60℃;水解时间10~14h。
进一步的,所述淀粉为糯米淀粉、甘薯淀粉、玉米淀粉或马铃薯淀粉。
进一步的,所述步骤1.3)中,多孔淀粉与酪蛋白的质量比为6:3~6:5;加热温度25~35℃,加热时间10~15min;均质压力为80~100MPa。

本发明的有益效果是:
1、本发明采用酪蛋白和多孔淀粉两种大分子成分,制备得到酪蛋白/多孔淀粉微凝胶,进一步负载藻蓝蛋白得到产物藻蓝蛋白-酪蛋白/多孔淀粉微凝胶,合成工艺简单,成本低,易于推广,且制备的产物,缓释效果优良、荧光性能好。
2、本发明将淀粉在复合酶催化作用下水解形成多孔淀粉,然后多孔淀粉与酪蛋白混合,利用大分子间的静电相互作用力,得到酪蛋白/多孔淀粉微凝胶,由于水解后的多孔淀粉具有三维蜂窝状多孔结构,充分利用多孔淀粉的比表面积特性即其优异的吸附性能,用于功能性成分的递送,提高藻蓝蛋白的负载率。

参见图1,本发明提供的藻蓝蛋白-酪蛋白/多孔淀粉微凝胶,利用复合酶水解制备得到的多孔淀粉,与酪蛋白在适当条件下通过静电相互作用结合,得到高负载率的酪蛋白/多孔淀粉微凝胶(简称casein/PS MGs);然后在避光条件下将藻蓝蛋白PC溶解在制备的酪蛋白/多孔淀粉微凝胶中,并在室温下得到藻蓝蛋白-酪蛋白/多孔淀粉微凝胶(简称PC-casein/PS MGs)。本发明提供的制备过程简单、条件温和;原料易得、成本低,易于推广实现。
以具体的实施例对本发明提供的制备方法进行说明。
实施例1
本实施例提供的藻蓝蛋白-酪蛋白/多孔淀粉微凝胶PC-casein/PS MGs的制备过程包括以下步骤:
1)制备酪蛋白/多孔淀粉微凝胶
1.1)多孔淀粉的制备
将糯米淀粉与水混合,得到浓度10mg/mL的淀粉悬浮液;
1.2)向浓度10mg/mL的10mL淀粉悬浮液中加入1.5mL酶活力比为(糖化酶:α-淀粉酶=3000:2000IU/mL的复合酶液,水解温度60℃、水解时间12h,同时加入缓冲液(柠檬酸-柠檬酸钠)调节pH 6.0,最终制得水解率为40%的糯米多孔淀粉混合液,得到沉淀物;将沉淀物用蒸馏水充分洗涤,在40℃下真空干燥,压碎并筛分,得到多孔淀粉;
本实施例中,通过缓冲液(柠檬酸-柠檬酸钠)调节pH至复合酶水解所需的适当pH;
1.3)酪蛋白/多孔淀粉微凝胶的制备
按照质量比6:4分别称取多孔淀粉和酪蛋白,并均配制成浓度为0.5mg/mL的溶液;并将溶液混合后,调节混合溶液的pH为7.0,并于30℃下水浴加热15min,100MPa下高压均质3次,即可得到乳白色体系均一的酪蛋白/多孔淀粉微凝胶,简称casein/PS MGs;
2)藻蓝蛋白-酪蛋白/多孔淀粉微凝胶的制备
将粉末状的藻蓝蛋白PC溶于步骤2)制备好的casein/PS MGs中,在室温(温度25℃)避光、搅拌1h,得到产物藻蓝蛋白-酪蛋白/多孔淀粉微凝胶;本实施例中,PC与casein/PS MGs的质量体积比要保证最终得到的产物中PC终浓度为0.5mg/mL。
实施例2~实施例6
实施例2~实施例6提供的制备方法与实施例1相同,不同的是各个步骤中的反应条件参数不同,具体参见表1。

以上是本发明的几组实施例,本发明的原材料也可用下述进行替换。
本发明提供的实施例1~实施例6中,淀粉还可用甘薯淀粉,玉米淀粉或马铃薯淀粉替换;淀粉悬浮液与复合酶液的体积比为9~11:1~2内的任意值;淀粉悬浮液浓度为8~12mg/mL之间;复合酶液的酶活力比为糖化酶:α-淀粉酶=3000:2000~2400IU/mL之间;水解温度55~60℃之间;水解时间10~14h之间。多孔淀粉与酪蛋白的质量比在6:3~6:5之间;加热温度在25~35℃范围内,加热时间在10~15min之间;均质压力为80~100MPa之间。在此条件下,均能合成得到PC-casein/PS MGs。
进一步的,为了说明本发明制备的PC-casein/PS MGs的特征和性能优越性,进行表征试验。
试验1原料的SEM
选取原料糯米淀粉、多孔淀粉、酪蛋白、均质的酪蛋白以及实施例1制备的酪蛋白/多孔淀粉微凝胶,采用环境扫描电子显微镜(Quanta 200,FEI公司),检测条件:喷金处理后高真空模式下进行形貌表征,得到各原料不同放大倍数下的SEM图谱,结果如图2所示。
其中:图2A为糯米淀粉在5μm下的SEM图谱;图2B为糯米淀粉在1μm下的SEM图谱;图2C为多孔淀粉在1μm下的SEM图谱;图2D为酪蛋白在15μm下的SEM图谱;图2E为均质的酪蛋白在5μm下的SEM图谱;图2F为酪蛋白/多孔淀粉在5μm下的SEM图谱。
从图2的各个图谱中,可以看出,通过复合酶解成功得到蜂窝状的多孔结构,均质使得酪蛋白的粒度减小,且凝胶中淀粉受到高速剪切和挤压后变成棒状结构且表面变光滑,对本发明最终材料的高吸附性能具有明显的提升效果。
试验2
通过对去稳定剂、反应温度、质量比以及不同pH的考察,对多孔淀粉和酪蛋白之间作用机理进行验证。
(1)稳定剂
试验过程是在添加不同的化学试剂情况下,通过测定凝胶粒径及浊度的变化探索PS和casein间的结合作用力。分别在NGs中加入如下试剂:0.05mol/L NaCl、0.05mol/LUrea、0.5%SDS(0.05mol/L NaCl可屏蔽体系静电相互作用力;0.05mol/L Urea可破坏多糖蛋白等高分子之间的氢键相互作用,是一种氢键开裂剂;0.5%SDS可破坏高分子间的疏水相互作用),磁力搅拌30min后静止,测定体系粒径、PDI以及浊度变化;试验结果如图3A和图3B所示。图3A为去稳定剂对微凝胶粒径和PDI的影响,图3B为去稳定剂对微凝胶浊度影响。
从图3A和图3B得到:酪蛋白-PS微凝胶的形成主要依赖于分子间的静电相互作用和疏水相互作用,其中静电相互作用起主导作用。
(2)反应温度和摩尔比
试验组1:步骤1.2)的温度30℃;同时多孔淀粉与酪蛋白的质量比为0:1、6:6、6:5、6:4、6:3、6:2、6:1;
试验组2:步骤1.2)的温度50℃;同时多孔淀粉与酪蛋白的质量比为0:1、6:6、6:5、6:4、6:3、6:2、6:1;
试验过程是:试验组和对照组的得到的酪蛋白/多孔淀粉微凝胶(casein/PSMGs),采用荧光分光光度计(F-7000,日本日立公司),激发波长为280nm,狭缝宽度均为10nm,光谱条件下得到扫描范围:300~450nm对应的荧光光谱,如图3C(试验组,温度30℃)和图3D(对比组,温度50℃)所示。
从图3C和图3D得到结论:PS与酪蛋白相互作用过程是自发的,且是放热和熵驱动的过程,静电相互作用是主要的结合作用力。此外,表明也存在一定的疏水相互作用。
在图3C和图3D上分别选取激发波长在280nm时,不同质量分数对应的荧光强度变化值,然后得到双对数图,如图3E所示。双对数是摩尔浓度和荧光强度变化值(F0-F/F)做对数得到的,其中F0为多孔淀粉与酪蛋白的质量比是0:1时得到的荧光强度;F为加入不同浓度的淀粉后酪蛋白的荧光强度。
从图3E得到结论:多孔淀粉与酪蛋白在30℃和50℃的结合位点分别接近1.469和1.584。表明实验浓度下多孔淀粉与酪蛋白的比例约为(6:4,m/m),且受温度影响较小。并且PS-酪蛋白相互作用常数的Ka为106L/mol,远大于104L/mol,说明casein/PS MGs之间存在很强的结合力,有力证明本发明最终材料的易于结合和微凝胶合成工艺简单易行。
(4)圆二色谱和红外表征
1)试验过程:对酪蛋白、均质的酪蛋白和实施例1得到的酪蛋白/多孔淀粉微凝胶,采用圆二色光谱仪(Chirascan,英国应用光物理公司),检测条件:扫描速度50nm/min,狭缝宽度1nm,响应时间1s,扫描3次,分辨率为0.1nm,1mm石英样品池,记录190~260nm范围内酪蛋白CD图谱,参见图4A;
再采用傅里叶红外光谱仪(Tensor27,德国布鲁克),检测条件:以空气为背景,扫描范围为2000-1000cm-1,分辨率4cm-1,分别得到傅里叶变换红外光谱的变化,参见图4C;
2)试验过程:酪蛋白与多孔淀粉在混合时,调节pH分别为4.0、7.0、10.0,其余与实施例1的反应条件参数相同,分别得到3组酪蛋白/多孔淀粉微凝胶;采用圆二色光谱仪(Chirascan,英国应用光物理公司),检测条件:扫描速度50nm/min,狭缝宽度1nm,响应时间1s,扫描3次,分辨率为0.1nm,1mm石英样品池,记录190~260nm范围内酪蛋白CD图谱,得到3组微凝胶的圆二色光谱的变化趋势,参见图4B;
调节pH分别为4.0、7.0、10.0,其余与实施例1的反应条件参数相同,分别得到3组酪蛋白/多孔淀粉微凝胶。再采用傅里叶红外光谱仪(Tensor27,德国布鲁克),检测条件:以空气为背景,扫描范围为2000-1000cm-1,分辨率4cm-1,分别得到傅里叶变换红外光谱的变化,参见图4D。
从图4A~4D得到的结论是:均质化使酪蛋白分子更有序、更紧密,酪蛋白分子更稳定;多孔淀粉的加入对酪蛋白的球形结构有一定的保护作用,提高了其结构稳定性和有序性;此外,当调节pH为7.0时,与pH 4.0、10.0相比,α-螺旋含量大大增加,无规卷曲含量大大降低,体系更加有序和稳定。
试验3
(1)藻蓝蛋白PC、casein-PS MGs以及PC-casein/PS MGs的荧光变化
分别对藻蓝蛋白PC、实施例1制备的casein-PS MGs以及实施例1制备的PC-casein/PS MGs,分别采用荧光分光光度计(F-7000,日本日立公司),激发波长为600nm,狭缝宽度均为10nm光谱条件下得到扫描范围:620~800nm对应的荧光光谱,结果参见图5A;
从图5A得到结论:PC的负载与否并未使其荧光性能有明显损失,由此说明负载是可行的且理想的。
(2)体外缓释曲线
试验组1:实施例1制备得到的藻蓝蛋白-酪蛋白/多孔淀粉微凝胶;
试验组2:与试验组不同的是,直接采用糯米淀粉作为原料,选择与实施例1相同的反应条件参数,得到藻蓝蛋白-酪蛋白/糯米淀粉微凝胶;
试验组3:游离的藻蓝蛋白
对上述三组物质,具体过程:PC-MGs在超滤离心管中以4000r/min的速度离心30min,截留分子量为10KDa(游离PC可以通过超滤离心管的膜,而装载PC的MGs不能通过超滤膜。)然后在荧光分光光度计(F-7000,日本日立公司)下测定藻蓝蛋白在650nm处的荧光强度,计算得到0~50h内的缓释曲线,结果参见图5B所示。
从图5B得到结论:游离PC溶液在透析袋中始终保持突释状态,释放曲线明显陡峭短促。PC-casein/糯米淀粉MGs和PC-casein/PS MGs,由于MGs对PC的包埋作用,PC的释放过程相对缓慢,具有明显的缓释特性,证明本发明最终材料的包埋缓释性能特点。
(3)急性毒性试验
对照组:曝光三天的自来水;
试验组1:藻蓝蛋白PC;
试验组2:实施例1得到的casein/PS MGs
试验组3:实施例1得到的PC-casein/PS MGs
具体试验过程是:分别采用上述几组物质作为试样液,试样液0.5mg/mL,体积2000mL,并在每个试样液中投放10只斑马鱼,每24小时更换一次新的试样液,观察72h内不同时间下斑马鱼的死亡率,结果如图5C所示。
从图5C得到结论:与游离PC相比,负载PC的缓释特性成功使其72h急性毒性降低了50%,证明负载行为能够成功减低PC的毒性。
(4)Cu2+浓度对PC-casein/PS MGs的荧光强度影响
试验过程:准备7份浓度均为0.5mg/mL的藻蓝蛋白溶液10mL,分别7份溶液中对应加入浓度为0μM、2μM、4μM、6μM、8μM、10μM、20μM的Cu2+溶液,然后分别采用荧光分光光度计(F-7000,日本日立公司),激发波长为600nm,狭缝宽度均为10nm光谱条件下得到扫描范围:620~800nm对应的荧光光谱,得到对应的7份溶液的荧光光谱,结果参见图5D;
从图5D得到结论:当用2μM和10μM的Cu2+处理时荧光强度变化尤其明显,因此本试验采用2μM和10μM的Cu2+进行荧光抑制实验。
试验4荧光成像
试验组1:实施例1制备的casein/PS MGs
试验组2:藻蓝蛋白PC
试验组3:实施例1得到的PC-casein/PS MGs
试验组4:具有2μMCu2+的PC-casein/PS MGs
试验组5:具有10μMCu2+的PC-casein/PS MGs
试验过程是:
分别采用上述试验组的物质作为试样液,试样液浓度0.5mg/mL、体积100mL,在每个试样液中投放1只斑马鱼,培养12h后,在明场下、荧光场下观察斑马鱼头部和斑马鱼尾部的激光共聚焦显微镜照片,所有比例尺均为300μm,激发波长:559nm。结果参见图6。
图6中,A表示试验组1,B表示试验组2,C表示试验组3,D表示试验组4,E表示试验组5;(A1~E1)表示5个试验组在明场下的斑马鱼头的显微镜照片;(A2~E2)表示5个试验组在荧光场下的斑马鱼头的显微镜照片,(A3~E3)表示5个试验组在明场下的斑马鱼尾的显微镜照片,(A4~E4)表示5个试验组在荧光场下的斑马鱼尾的显微镜照片。
通过图6可以明显看出本发明制备的PC-casein/PS MGs具备荧光性能,体现了其优良的优良成像效果,具有可应用性。
Claims (7)
1.一种藻蓝蛋白-酪蛋白/多孔淀粉微凝胶的制备方法,其特征在于:所述制备方法包括以下步骤:
1)制备酪蛋白/多孔淀粉微凝胶,备用;
2)在避光条件下,将藻蓝蛋白粉末溶解于酪蛋白/多孔淀粉微凝胶中,搅拌均匀,得到产物藻蓝蛋白-酪蛋白/多孔淀粉微凝胶;
所述步骤1)中酪蛋白/多孔淀粉微凝胶的制备过程是:
1.1)将淀粉溶于水形成淀粉悬浮液;
1.2)向淀粉悬浮液中加入复合酶液,再加入柠檬酸-柠檬酸钠缓冲液调节pH为5.8~6.0,经复合酶水解反应、分离后,将得到的沉淀物经洗涤、干燥、破碎和筛分,得到多孔淀粉;
1.3)将上述得到的多孔淀粉与酪蛋白混合,调节pH为6.8~7.2,经加热、均质,得到酪蛋白/多孔淀粉微凝胶;
所述步骤1.2)中,所述复合酶液的酶活力比为糖化酶:α-淀粉酶=3000:2000~2400 IU/mL;水解温度55~60℃;水解时间10~14h;
所述步骤2),产物中藻蓝蛋白浓度为0.40~0.60 mg/mL;溶解温度20~25℃;搅拌时间1~2h。
2.根据权利要求1所述的藻蓝蛋白-酪蛋白/多孔淀粉微凝胶的制备方法,其特征在于:所述步骤1.2)中,淀粉悬浮液与复合酶液的体积比为9~11:1~2;所述淀粉悬浮液浓度为8~12mg/mL。
3.根据权利要求2所述的藻蓝蛋白-酪蛋白/多孔淀粉微凝胶的制备方法,其特征在于:所述淀粉为糯米淀粉、甘薯淀粉、玉米淀粉或马铃薯淀粉。
4.根据权利要求3所述的藻蓝蛋白-酪蛋白/多孔淀粉微凝胶的制备方法,其特征在于:所述步骤1.3)中,多孔淀粉与酪蛋白的质量比为6:3~6:5;加热温度25~35℃,加热时间10~15min;均质压力为80~100MPa。
5.一种如权利要求1-4任一项所述的藻蓝蛋白-酪蛋白/多孔淀粉微凝胶的制备方法所制备的藻蓝蛋白-酪蛋白/多孔淀粉微凝胶。
6.一种如权利要求5所述的藻蓝蛋白-酪蛋白/多孔淀粉微凝胶在荧光成像中的应用。
7.根据权利要求6所述的应用,其特征在于:所述藻蓝蛋白-酪蛋白/多孔淀粉微凝胶中藻蓝蛋白浓度为0.40~0.60 mg/mL。
