Abstract
The invention discloses a method for detecting illegal addition of omeprazole in weight-losing health-care food, which comprises the following steps: weighing 0.2-1.0 g of a weight-reducing health-care product sample, adding 1.3-5.5 mL of an extraction reagent, performing ultrasonic extraction for 8-20 minutes, centrifuging after extraction is finished at a centrifugation speed of 10000 rpm for 2 minutes, and transferring 0.5mL of supernatant into a centrifuge tube after centrifugation; adding 1-5 mL of sodium chloride solution into the supernatant in the centrifuge tube, shaking, uniformly mixing, and standing to serve as a to-be-detected liquid sample for later use; and thirdly, taking 200-1500 muL of gold sol reagent and 8-50 muL of the liquid sample to be detected, uniformly mixing, and then placing in a detection bin of a Raman spectrometer for scanning detection. Through the mode, the invention provides the method for detecting the illegal addition of the omeprazole into the weight-reducing health-care food, so that the detection of the omeprazole in the weight-reducing health-care food can be realized, the method is suitable for on-site rapid detection, and the professional requirements of operators are obviously reduced.

Technical Field

Background
GB16740-2014 published in 2014 of China, namely the Standard for inspection of food (health food) Annationwide, item 2.1 defines the health food as follows: food claimed to have specific health-care function or for supplementing vitamins and minerals. The health food is suitable for specific people, has the function of regulating organism, does not aim at treating diseases, and does not produce any acute, subacute or chronic harm to human body.
In recent years, with the rapid development of equipment economy, the physical living conditions of people are continuously improved, more and more people are obese in China, some obese patients worry about the side effect caused by drug treatment, and the selection of weight-reducing health-care food is safer.
The thirty-eighth article of the food safety law of the people's republic of China also specifies that: "No medicine should be added to the food in production and management". Because the weight-losing health-care food has the characteristics of slow response, no toxic or side effect and the like, some illegal manufacturers illegally add chemical medicines into the weight-losing health-care food to increase the weight-losing efficacy by utilizing the psychology of the consumers in urgent need of losing weight and the propaganda of the health-care food without toxic or side effect.
The existing national standard detection method detects a small number of types of medicines and cannot meet the actual supervision requirement. The main method is to adopt high performance liquid chromatography or high performance liquid chromatography-tandem mass spectrometry to detect the illegal addition of chemicals in the weight-reducing health-care food. The method has expensive detection equipment, high requirement on operators, high instrument detection cost and relatively long detection speed. However, the existing quick detection method mostly adopts a chemical color development method, and the detection sensitivity of the method is low.
At present, most of the existing detection technologies for forbidden additives in health care products adopt liquid chromatography and liquid chromatography tandem mass spectrometry, and the detection technologies have the advantages of wide application range, good separation performance and high sensitivity. But the operation is more complicated, and the equipment volume is bigger, so the field use can not be realized.

The invention mainly solves the technical problem of providing a method for detecting the illegal addition of omeprazole in weight-reducing health-care food, can realize the detection of omeprazole in the weight-reducing health-care food, is suitable for on-site rapid detection, and obviously reduces the professional requirements of operators.

In a preferred embodiment of the invention, the extraction reagent is a 7:3 mixture of methanol and ethanol.
In a preferred embodiment of the present invention, the concentration of the sodium chloride solution is 1.6-4.5 mol/L.
The invention has the beneficial effects that: the method for detecting the illegal addition of omeprazole into the weight-reducing health-care food can realize the detection of omeprazole in the weight-reducing health-care food, is suitable for on-site rapid detection, and obviously reduces the professional requirements of operators.

and step three, taking 500 mu L of gold sol reagent and 20 mu L of sample to be detected, uniformly mixing, and then placing in a detection bin of a Raman spectrometer for scanning detection.
Wherein the extraction reagent is a 7:3 mixture of methanol and ethanol.
Further, the concentration of the sodium chloride solution is 2 mol/L.
Wherein the preparation process of the gold sol reagent comprises the following steps:
adding 100mL of ultrapure water into a 250mL three-neck round-bottom flask, quickly stirring and boiling, adding 1.2mL of 1% chloroauric acid aqueous solution, continuously heating to boil again, adding 1mL of 1% trisodium citrate aqueous solution, keeping stirring for about 25min, finishing the reaction, naturally cooling to room temperature, and sealing with a brown bottle for storage.
The Raman spectrum detection result shown in figure 1 is finally obtained through the processes, and the food safety can be rapidly judged according to the detection result.
In conclusion, the invention provides a method for detecting the illegal addition of omeprazole into weight-reducing health-care food, which can realize the detection of omeprazole in weight-reducing health-care food, is suitable for on-site rapid detection and obviously reduces the professional requirements of operators.
