Abstract

Description
技术领域
本发明涉及诱导多能干细胞,其由可条件永生化的细胞例如成体干细胞生成。特别地,本发明涉及由包含用于条件性永生化的可控转基因的细胞生成的诱导多能干细胞,以及那些诱导多能干细胞的子代。
背景技术
人多能干细胞(hPSC)由多能性(pluripotency)属性限定:能够分化为体内发现的任何细胞类型的属性。它们包括衍生自早期胚胎(胚泡,受精后大约第6.5天)的内细胞团的胚胎干细胞(hESC),以及通过借由某些转录因子的转导和外源表达将体细胞重编程为多能表型而生成的诱导多能细胞(hiPSC)。
能够重编程为多能性的此类转录因子的规范性集被称为OKSM(OCT4、KLF4、SOX2,C-MYC),但已知其他因子,其可替代O、K、S或M或者调节重编程(随机过程)发生的效率。
通常,低传代原代细胞是用于重编程以生成诱导多能干细胞(iPSC)的优选底物。此类细胞的优势是它们倾向于比高传代细胞分裂更快;不分裂的细胞难于重编程。它们也更可能是整倍体。成体干细胞(ASC)还为重编程至多能性提供有希望的底物,常常需要较少的转录因子和较适度的重编程事件,这是因为重编程因子(例如SOX2、KLF4)的内源性表达以及与(多/多个)能性相关联的更开放的染色质结构。
存在由永生哺乳动物细胞而非原代细胞生成的iPSC的一些实施例(大多数是EBV永生血细胞)。由于此类细胞通常由EBV或癌基因(诸如猿猴病毒40大T抗原)的稳定基因组整合而永生化,因此它们的临床实用性令人怀疑。293FT细胞系(例如稳定表达SV40大T抗原以及在转染重编程转录因子后的表型发生变化时),它会生成异常集落,而非真正的iPSC。因此,衍生自此类永生化细胞的iPSC系通常仅限于体外应用,诸如疾病建模、药物研发和发育研究。
此外,Skvortsova等人的一项研究(Oncotarget,2018,Vol.9(No.81),pp35241-35250)报告称,永生化的鼠成纤维细胞系难于重编程为多能状态,并且与永生化和体外选择相关联的非整倍体不太可能是此类不应性的原因。因此,当寻求标识适合于重编程为多能状态的细胞时,存在重大的技术挑战,并且至少一些永生化的细胞难于重编程。
多能干细胞稳定且可在体外无限期培养。然而,它们的临床应用存在挑战,诸如它们的畸胎瘤形成能力以及大多数分化方案引起对所期望终点的分化低于100%的问题。因此,仍然存在通过治疗性群体中的残留多能细胞的畸胎瘤形成的形式风险,以及将不期望的细胞类型共同转移给患者的问题。即使仅降低疗法的效率,此类不期望的亚群也可具有中性或负面作用。这种情况因以下事实而加剧:所期望治疗性细胞类型常常不是终末分化的细胞类型(其慢性或急性丢失导致患者病理性),而是在患者的适当组织体中产生最终细胞类型的晚期组织祖细胞/成体干细胞群体。晚期祖细胞群体在体外培养中常常不稳定,因此,除了上面提到的纯度问题外,它们在临床应用中以可接受的纯度水平可规模化制备也是一个非同寻常的挑战,即使在理论上由多能细胞进行的晚期祖细胞群体的制备也实际不受限制。
此类祖细胞群体通常也非常难以处理。如果它们在移植(例如骨髓细胞)前是从患者或另一个人中分离出来,则与材料可用性极为有限相关联的困难、两个人均可同时在相同时间和地点手术的要求、合适(例如免疫相容的)供体的可用性、缺乏转移材料纯度和QC均限制此类治疗的普遍性。理论上,通过分化hPSC诸如hiPSC生成此类细胞群体的可能性将改进此类问题,但它们也带来其他挑战。重要的是,祖细胞群体难以在体外处理,并且随着时间的推移不稳定。由此,即使有GMP(医疗)质量的iPSC可用,祖细胞的均质、符合GMP的可规模化制备通常也不可能。大多数分化方案并不是100%有效的,使得在治疗性细胞群体中可存在污染性亚群。
仍然需要生成具有临床实用性的干细胞。
发明内容
本发明基于令人惊奇的认识,即诱导多能干细胞(iPSC)技术可通过由条件永生性的细胞,特别是条件永生性的干细胞生成iPSC来改善。
本发明的第一方面提供诱导多能干细胞,其包含用于条件性永生化的可控转基因。可控转基因通常将能够有条件地使衍生自诱导多能干细胞的下游(分化程度更高)细胞永生化。这些下游细胞否则可难以处理,因此存在条件永生化的转基因是一种改善。
本发明的第二方面提供多能干细胞,其由条件永生性的细胞(通常是条件永生性的干细胞)而可获得或获得。
本发明的第三方面提供制备多能干细胞的方法,其包含重编程条件永生性的细胞(通常是条件永生性的干细胞)的步骤。该方法还可包含后续步骤,以由多能干细胞生成不同的细胞类型。
本发明的另一方面涉及通过本发明的任何方法制备的细胞,以及通过那些细胞中的任何一种制备的微粒。
发明人已发现,将条件永生性的细胞诸如干细胞(例如来自CTX0E03或STR0C05细胞系的细胞)重编程至多能性将允许生成其他成体干细胞或组织祖细胞群体。一旦生成多能细胞,常规分化方案就可用于提供任何所期望谱系的细胞,例如外胚层、内胚层或中胚层谱系。在某些实施方案中,将多能细胞指向与原始条件永生性的干细胞的谱系不同的谱系。在某些实施方案中,诱导多能细胞可分化为间充质干细胞、神经干细胞或造血干细胞,包括进一步分化为免疫系统的细胞,诸如T淋巴细胞、NK细胞和树突状细胞。在某些实施方案中,诱导多能细胞可分化为体(成体)干细胞、多能细胞、寡能细胞或单能细胞;或终末分化细胞。
下面的实施例论证,根据本发明由不同的条件永生性的细胞生成的iPSC是多能性的并能够进入内胚层、中胚层和外胚层谱系。实施例还示出,成体干细胞(MSC)可由本发明的iPSC生成。这些MSC被证明是多能性的并能够分化为软骨、脂肪和骨细胞。
还描述将本发明的诱导多能细胞分化为免疫系统的功能性细胞。这些免疫细胞包括T细胞、B细胞、自然杀伤(NK)细胞和树突细胞。如本领域技术人员将理解的,这些细胞经常将经由造血谱系细胞到达。
细胞例如干细胞可被认为通过c-myc-ERTAM转基因而可条件永生化。此转基因已被证明准许通过将4-羟基他莫昔芬添加到细胞培养基中来稳定、可规模化制备干细胞系诸如神经干细胞系CTX0E03和STR0C05,从而在无需表型的任何改变的情况下促进生长和细胞分裂。
条件永生性的干细胞通常是成体干细胞,也称为体干细胞。例如,它可为神经干细胞,诸如来自CTX0E03干细胞系的细胞。CTX0E03神经干细胞系已由申请人(ReNeuronLimited)保藏在英国波顿唐的欧洲认证细胞培养物收藏中心(European Collection ofAuthenticated Cell Cultures(ECACC),Porton Down,UK)并具有ECACC登录号04091601。在其他实施方案中,神经干细胞系可为“STR0C05”细胞系、“HPC0A07”细胞系(也由申请人在ECACC保藏)或在Miljan等人Stem Cells Dev.2009中公开的神经干细胞系。
将条件永生性的干细胞重编程为多能性。诱导多能表型通常涉及将与多能相关联基因的特定集的产物或“重编程因子”引入给定细胞类型中。重编程因子(也称为Yamanaka因子)的最初集是转录因子Oct4、Sox2、cMyc和Klf4。如本领域所公知的,通常使用病毒或附加体载体将重编程因子引入细胞中。
适合于将重编程因子引入细胞的病毒载体包括慢病毒、逆转录病毒和仙台病毒。引入重编程因子的其他技术包括mRNA转染。
已令人惊奇地观察到,仅需一个转录因子即可将某些条件永生性的干细胞诸如CTX0E03重编程为多能性。例如,图2B-D示出,仅OCT4就可诱导CTX0E03的多能性。观察到实现多能性的转录因子的组合包括:OCT4和SOX2;OCT、KLF4和SOX2;OCT4、KLF4、SOX2和MYC。因此,包含或由这些组合组成的重编程因子被提供用于本发明。在图2C中成功诱导多能性的因子的每种组合被提供为本发明的单独实施方案。用于诱导条件永生性的干细胞至多能性的重编程因子可包含例示性组合或由其组成。
其他永生因子在本领域中已知并对于本领域技术人员而言显而易见。可使用因子的任何合适的组合,这完全在本领域技术人员的能力范围内。例如,用小分子抑制剂重编程在本领域中已知,而NANOG和TET1被称为其他合适的转录因子。举例来说,Thomson及其同事使用NANOG、KLF4、SOX 2和LIN28作为OKSM的替代品。在另一个实施例中,TET1已被证明能够替代OCT4。
由于c-myc-ERTAM转基因的活性重演细胞MYC活性,因此在重编程c-myc-ERTAM诱导永生化的干细胞时,表达MYC癌基因的载体可有可无,因为如果需要,此活性可通过向培养基提供4-OHT以活化c-myc-ERTAM融合蛋白来提供。因此,在一些实施方案中,MYC(例如MYC重编程载体)不用作单独的重编程因子。技术人员当然会意识到,使用除MYC以外的基因的条件性永生化体系可需要外源MYC对其进行重编程。
诱导多能细胞可分化为任何所期望的细胞类型。确定细胞谱系或细胞类型的技术是本领域公知的。通常,这些技术涉及确定细胞表面(和/或不存在多能性标志物,诸如Oct4)或内部的分化标志物,诸如谱系特异性转录因子、细胞形态和功能的存在。例如,多能干细胞通常对规范性多能转录因子OCT4和细胞表面抗原TRA-1-60和SSEA-4呈阳性,但不表达早期分化标志物SSEA-1。
内胚层谱系的标志物包括GATA6、AFP或HNF-α。其他内胚层标志物可包括Claudin-6、Cytokeratin 19、EOMES、SOX7和SOX17中的一种或多种。
中胚层谱系的标志物包括BMP2、Brachyury或VEGF。其他中胚层标志物可包括活化素A、GDF-1、GDF-3和TGF-β中的一种或多种。
外胚层谱系的标志物包括PAX6、巢蛋白或TubIII。其他外胚层标志物可包括Noggin、PAX2和chordin中的一种或多种。
例如在图3D中示出分化为不同谱系。实施例2(图7)和实施例3(图9)也论证CTX-iPSC和STR0C-iPSC向内胚层、中胚层和外胚层谱系的分化。实施例2使用以下标志物:
多能细胞可分化为任何所期望的细胞类型。这可包括间充质干细胞、神经干细胞或造血干细胞。在另一个实施方案中,体(成体)干细胞由本发明的诱导多能细胞的分化产生。在其他实施方案中,由该方法提供的细胞是多能细胞、寡能细胞或单能细胞。此实施方案的实施例是祖细胞,例如神经元祖细胞的制备。Nistor及其同事在PloS One(2011),vol.6e20692中描述神经元祖细胞在神经退行性疾病治疗中的潜在用途。使用已知分化技术,也可将产生的细胞完全分化为终末分化细胞。如Carri及其同事(2013)所描述,此实施方案的实施例是向中棘神经元(如在亨廷顿舞蹈症中所丢失的)的分化及其可规模化制备;参见Stem Cell Review and Reports,DOI 10.1007/s12015-013-9441-8。在具体实施方案中,造血干细胞可分化为T细胞、NK细胞和/或树突细胞。因此,作为一个实施方案,提供T细胞。提供天然杀伤细胞作为另一个实施方案。还提供树突状细胞。
实施例1和图5论证CTX-iPSC向间充质干细胞的分化。在此实施例中,通过标志物CD73、CD90和CD105而不是CD14、CD20、CD34或CD45的存在来标识MSC表型。
实施例3和图10论证CTX-iPSC-MSC向软骨、脂肪和骨细胞的分化。
重活化(如有必要)c-myc-ERTAM转基因及然后向培养基中添加4-OHT应准许衍生的细胞群体无限期生长。通过与CTX0E03本身的类比,预期这将允许在体外可规模化制备有效无限量的治疗上有用的细胞群体,这对于以急性或慢性细胞丢失为特征的任何病况而言可用作“现成”疗法,对于该病况,细胞疗法要么不存在,要么受限于组织供体可用性或由hPSC分化方案的技术限制。

条件永生性的干细胞、由重编程产生的多能细胞以及可从多能细胞获得的分化程度更高的细胞通常被分离或纯化。由这些细胞中的任何一种制备的微粒(细胞外囊泡)例如外泌体通常也被分离或纯化。
分化后提供的细胞以及由它们制备的微粒可用于疗法中。疗法通常是对其有需要的个体的疾病或病症。患者通常是人类。
在另一方面,本发明提供组合物,其包含:条件永生性的干细胞;由重编程产生的多能细胞;由多能细胞可获得的分化程度更高的细胞;或由这些细胞中的任何一种制备的微粒;和药学上可接受的赋形剂、载体或稀释剂。

具体实施方式
本发明人惊奇地发现,条件永生性的细胞可被重编程为多能干细胞表型。与现有的诱导多能干细胞相比,这具有优势。特别地,尽管在体外永生和长期培养情况下,但令人惊奇地示出,神经干细胞系CTX0E03和STR0C05可被外源转录因子重编程。此外,令人惊奇的是,像先前一样,在重编程后,有条件地控制的基因仍然能够被活化和沉默。
有利地,与标准(无条件永生化)细胞相比,条件永生性的细胞的重编程常常可用更少重编程因子来实现。在某些实施方案中使用成体干细胞提供类似优势。
此外,细胞的条件永生性的性质提供对细胞和永生化体系的有益的可控制性。在一些实施方案中,这些益处由C-MYC-ERTAM条件性永生化体系提供。不希望受理论的束缚,与使用永生化(即永久永生化)细胞的先前尝试相比,认为使用条件永生性的细胞作为重编程为多能性的来源有助于所观察的益处。
本发明的诱导多能细胞诸如实施例中的CTX-iPSC和STR0C-iPSC代表非常有用的临床资源。它们可沿期望的谱系分化以生成靶标群体,诸如组织祖细胞类型或成体干细胞群体。然后,提供永生化试剂(例如4-OHT)以促进连续生长并防止细胞周期退出,且相关联的另外分化可允许常规可规模化制备先前无法获得的临床相关亚群,而无需每当需要新一批细胞疗法产品时从原材料中反复分离细胞或重新反复从诱导多能干细胞开始的分化方案。这提供现成的细胞资源的可能性,例如用于同种异体细胞疗法。
重新应用诱导永生化以规模化制备同种异体、现成成体干细胞治疗性群体的能力预期将特别有益。
克隆或纯化步骤可用于由或多或少的异质分化培养物中生成所期望治疗性类型的纯群体,以大规模制备原始条件永生性的细胞(例如CTX0E03)本身不合适的病况的现成治疗,避免在分化方案效率不高情况下的现有技术中存在的缺点。这既适用于细胞本身,也适用于由不同细胞类型制备的微粒级分(例如,外泌体级分),其中有效载荷分子的受体特征与由CTX细胞本身制备的那些分子不同。
此外,由于这些CTX-iPSC衍生型亚系衍生自已通过临床阶段安全性试验(CTX)的细胞系,因此有可能加快其进入新适应症临床功效试验的进程。
在某些方面,本发明涉及由不同神经干细胞生成的诱导多能干细胞,其包含用于条件性永生化的可控转基因,诸如分别衍生自皮质和纹状体组织且各自来自不同人类供体的CTX0E03或STR0C05神经干细胞系;以及那些诱导多能干细胞的子代。
多能性和iPS细胞的诱导
诱导多能细胞的生成是本领域已知的,因为Takahashi和Yamanaka示出通过同时引入四个基因可从小鼠成纤维细胞中生成具有与胚胎干细胞相似属性的干细胞(Cell.2006;126:663-676)。该原理于2007年应用于人细胞(Takahashi等人,Cell.2007;131:861-872;Yu等人,Science.2007;318:1917-1920)。Shi等人在Nature ReviewsDrugDiscovery,第16卷第115-130页(2017年)中提供最新综述。
iPSC通常是通过将特定集的多能性相关基因的产物或“重编程因子”引入给定细胞类型而获得的。重编程因子(也称为Yamanaka因子)的最初集是转录因子Oct4、Sox2、cMyc和Klf4。
iPS细胞的生成取决于用于诱导的转录因子。Oct-3/4和Sox基因家族的某些产物(Sox1、Sox2、Sox3和Sox15)已被标识为参与诱导过程的关键转录调节因子,缺乏该转录调节因子使诱导无法进行。然而,附加基因(包括Klf家族的某些成员(Klf1、Klf2、Klf4和Klf5)、Myc家族(c-myc、L-myc和N-myc)、Nanog和LIN28)已被标识为提高诱导效率。
“POU5F1”、“OCT4”和“OCT3/4”是相同转录因子的同义词。这是本领域中常称为OCT4的转录因子,但最近更名为POU5F1(POU类5同源盒1)。如本领域技术人员将显而易见的,这些名称在本文中可互换使用。
如本领域所公知的,通常使用病毒或附加体载体将重编程因子引入细胞中。适合于将重编程因子引入细胞的病毒载体包括慢病毒、逆转录病毒和仙台病毒。引入重编程因子的其他技术包括mRNA转染。
非整合重编程方法是本领域已知的,例如,如Schlaeger等人,NatBiotechnol.2015年1月;33(1):58–63。在仙台病毒重编程中,仙台病毒颗粒通常用于转导具有编码该组重编程因子的可复制型RNA的靶细胞。在附加体重编程中,延长的重编程因子表达通常是通过有利于附加体质粒DNA在分裂细胞中的复制的艾巴氏病毒衍生的序列实现的。在mRNA重编程中,通常用编码重编程因子的体外转录mRNA来编码细胞,并且常常采用化学措施来限制外源核酸对先天免疫系统的活化。由于mRNA的半衰期很短,常常需要每天转染来诱导hiPSC。
重编程因子的转染可通过本领域已知的多种方式来实现,诸如通过脂质转染、核转染或电穿孔。
在下面的一个实施例中,使用编码“Yamanaka因子”OCT4、L-MYC、KLF4和SOX2以及LIN28的标准非整合附加体载体,将条件永生性的CTX0E03细胞重编程为多能性。在另一个实施例中,示出仅OCT4诱导CTX0E03的多能性。还观察到实现多能性的转录因子组合包括:OCT4和SOX2;OCT、KLF4和SOX2;OCT4、KLF4、SOX2和MYC。
在某些实施方案中,OCT4、L-MYC、KLF4和SOX2以及LIN28中的一个、两个、三个或四个用于将条件永生性的细胞重编程为多能性。在某些实施方案中,使用OCT4和L-MYC、KLF4和SOX2以及LIN28中的一个或多个。在一些实施方案中,这些因子与cMYC-ERTAM条件性永生化体系组合使用。
在下面的另一个实施例中(实施例3),STR0C05细胞用重编程质粒pCE-hOCT3/4、pCE-hSK、pCE-hUL和pCEmP53DD重编程,从而表达转录因子POU5F1、SOX2、KLF4、L-MYC、LIN28和p53的显性阴性抑制剂。因此,在某些实施方案中,根据本发明使用的转录因子可包含或由POU5F1、SOX2、KLF4、L-MYC、LIN28和p53的显性阴性抑制剂组成。如本领域技术人员将显而易见的,这些中的一个、两个、三个或更多个可被去除或替换。在某些实施方案中,POU5F1、SOX2、KLF4、L-MYC、LIN28和p53的显性阴性抑制剂中的一个、两个、三个、四个或更多个用于将条件永生性的细胞重编程为多能性。在一些实施方案中,这些因子与c-myc-ERTAM条件性永生化体系组合使用。
在一些实施方案中,通过在培养基中提供4-OHT以活化待重编程的干细胞中的c-myc-ERTAM转基因,提供MYC活性以促进重编程过程。因此,在某些实施方案中,不需要单独添加的MYC。
条件永生性的细胞
本发明获取条件永生性的细胞并诱导它们具有多能表型。条件永生性的细胞通常是条件永生性的干细胞,例如条件永生性的成体干细胞。
条件永生性的细胞通常属于哺乳动物,更通常是人类。
干细胞是本领域已知的。干细胞是具有在生物体整个生命周期内增殖、表现出自我维持或更新,且生成克隆相关子代的能力的细胞。根据本发明重编程的干细胞通常是多能细胞。根据本发明重编程的干细胞通常是成体(体)干细胞。
用于本发明的干细胞经分离。术语“分离”指示其所指代的细胞或细胞群体不在其自然环境中。细胞或细胞群体已与周围组织充分分开。在一些实施方案中,如果样品含有至少约75%、在一些实施方案中至少约85%、在一些实施方案中至少约90%并且在一些实施方案中至少约95%的干细胞,则细胞或细胞群体与周围组织基本上分开。换句话说,如果样品含有小于约25%,在一些实施方案中小于约15%、在一些实施方案中小于约5%的除干细胞以外的材料,则样品与周围组织基本上分开。此类百分比值指重量百分比。该术语还涵盖已从其起源的生物体中去除并存在于培养物中的细胞。该术语还涵盖已从其起源的生物体中去除并随后重新插入生物体中的细胞。含有重新插入的细胞的生物体可为从中去除细胞的相同生物体,也可为不同的生物体。
对于根据本发明制备的子代细胞的任何未来受体,干细胞通常是同种异体的。
本发明使用条件永生性的干细胞诸如干细胞系,其中可调节永生因子的表达而不会不利地影响制备治疗上有效的干细胞。这可通过引入永生因子来实现,如果不向细胞供应活化剂,则永生因子无活性。此类永生因子可为基因诸如c-mycER。c-MycER基因产物是融合蛋白,其包含与突变雌激素受体的配体结合结构域融合的c-Myc变体。C-MycER仅在合成类固醇4-羟基他莫昔芬(4-OHT)存在的情况下驱动细胞增殖(Littlewood等人,1995)。此方法允许在体外神经干细胞的受控扩增,同时避免由于在神经干细胞系中存在c-Myc或编码其的基因而对宿主细胞增殖的不期望的体内影响(例如肿瘤形成)。
在某些实施方案中,条件永生性的干细胞可为:
间充质干细胞,其任选地选自衍生自骨髓的干细胞、子宫内膜再生细胞、间充质祖细胞或多能成体祖细胞;
神经干细胞;
造血干细胞,任选地为CD34+细胞和/或从脐带血中分离,或任选地为CD34+/CXCR4+细胞;
非造血脐带血干细胞;或
衍生自脂肪组织的间充质干细胞。
在这些实施方案的每一个中,细胞通常属于哺乳动物,更通常是人类。
通常,条件永生性的干细胞是神经干细胞,例如人神经干细胞。
神经干细胞在发育期间会产生神经元、星形胶质细胞和少突胶质细胞并可替代成体大脑中的许多神经细胞。根据本发明用于某些方面的典型神经干细胞表现出一种或多种神经表型标志物Musashi-1、巢蛋白、NeuN、III类β-微管蛋白、GFAP、NF-L、NF-M、微管细胞相关联蛋白(MAP2)、S100、磷酸二酯酶、磷脂酰肌醇聚糖(尤其是磷脂酰肌醇聚糖4)、神经元穿透素II、神经元PAS 1、神经元生长相关联蛋白43、神经突起生长延伸蛋白、波形蛋白、Hu、丝联蛋白、04、髓磷脂碱性蛋白和多效蛋白等等。
神经干细胞可来自干细胞系,即稳定分裂的干细胞的培养物。可使用单个限定来源使干细胞系大量生长。
优选条件永生性的神经干细胞系包括CTX0E03、STR0C05和HPC0A07神经干细胞系,它们已由本专利申请的申请人ReNeuron Limited在欧洲动物培养物保藏中心(ECACC);疫苗研究与制备实验室(VaccineResearch and Production laboratories);威尔特郡,索尔兹伯里SP4 OJG,波顿唐的公共卫生实验室服务(Public Health Laboratory Services,Porton Down,Salisbury,Wiltshire,SP4 0JG)中保藏,其中登录号04091601(CTX0E03);登录号04110301(STR0C05);和登录号04092302(HPC0A07)。这些细胞的衍生和起源描述于EP1645626 B1和美国专利7416888中,两者均以全文引用的方式并入本文。
CTX0E03(ECACC保藏#04091601)
CTX0E03是在临床试验中作为用于缺血性中风和肢体损伤的疗法的神经干细胞系。它通过整合C-MYC-ERTAM融合蛋白而可控制地永生化,该融合蛋白在ERTAM域与合成雌激素衍生物4羟基他莫昔芬(4-OHT)结合后转移到核内,在其中C-MYC域促进无限期细胞循环。C-MYC-ERTAM的表达显然不会影响细胞表型。因此,无限多数量的患者可用作为“现成”同种异体疗法的CTX来治疗。转基因已被证明在去除4-OHT和/或转移到患者后被沉默。
可在以下培养条件下培养CTX0E03细胞系的细胞:
·人血清白蛋白0.03%

加碱性成纤维细胞生长因子(10ng/ml)、表皮生长因子(20ng/ml)和4-羟基他莫昔芬(100nM)用于细胞扩增。可通过去除4-羟基他莫昔芬来分化细胞。通常,可在5%CO2/37℃或5%、4%、3%、2%或1%O2的低氧条件下培养细胞。这些细胞系不需要成功培养血清。血清是成功培养许多细胞系所必需的,但其中含有许多污染物。CTX0E03、STR0C05或HPC0A07神经干细胞系或任何其他不需要血清的细胞系的另一优势是避免由血清引起的污染。在本发明的一些实施方案中,可维持体系中血清的缺乏,例如通过将E8培养基用于重编程和培养诱导多能干细胞的步骤。
根据需要,CTX培养基可添加或不添加4-OHT,以通过c-myc-ERTAM转基因提供MYC活性。
CTX0E03细胞系的细胞是多能细胞,其最初衍生自12周人胚胎皮层。Sinden等人详细描述CTX0E03细胞系的分离、制造和方案(美国专利7,416,888和EP1645626B1)。CTX0E03细胞不是“胚胎干细胞”,即它们不是衍生自胚泡的内细胞团的多能细胞;分离原始细胞不会引起胚胎的破坏。在生长培养基中,CTX0E03细胞为巢蛋白阳性,而GFAP阳性细胞(即,该群体对GFAP阴性)的百分比较低。
CTX0E03是克隆细胞系,其含有单拷贝的c-mycER转基因,其通过逆转录病毒感染传递且受4-OHT(4-羟基他莫昔芬)有条件地调节。C-mycER转基因表达在4-OHT存在下刺激细胞增殖的融合蛋白且因此允许在4-OHT存在下培养时受控扩增。此细胞系是克隆的,在培养中迅速扩增(倍增时间50-60小时)并具有正常人类核型(46XY)。它是遗传稳定的且可大量生长。这些细胞安全且无致瘤性。在没有生长因子和4-OHT的情况下,细胞会经历生长停滞并分化为神经元和星形胶质细胞。一旦植入缺血损伤的大脑,这些细胞仅迁移至组织损伤的区域。
CTX0E03细胞系的开发已允许用于临床应用的一致产品得以规模化。由堆积材料制备细胞允许大量生成大量用于商业应用的细胞(Hodges等人,2007)。
CTX0E03药物产品可以新鲜的形式(如PISCES试验中的情况)或冷冻的活细胞悬浮液的形式提供,如US9265795中所述并用于PISCES II试验中。药物产品通常包含≤37传代的CTX0E03细胞。
CTX临床药物产品通常在无溶剂的赋形剂中配制为“现成”冷冻保存产品(例如,如美国专利9265795中所述),保质期为许多月。此制剂通常包含Trolox(6-羟基-2,5,7,8-四甲基苯并二氢吡喃-2-羧酸)、Na+、K+、Ca2+、Mg2+、Cl-、H2PO4-、HEPES、乳糖酸、蔗糖、甘露醇、葡萄糖、右旋糖酐-40、腺苷和谷胱甘肽。这些赋形剂中的一种或多种例如两、三或四种可任选地去除或替换。通常,制剂不包含偶极非质子溶剂,特别是DMSO。
干细胞产品的临床放行基准通常包括无菌性、纯度(细胞数量、细胞活力)的测量结果,以及临床产品放行或如监管机构要求的信息所需的确定度、稳定性和效能的许多其他测试。下表1总结CTX0E03所采用的测试。

先前已使用人PBMC检验来论证CTX0E03细胞系非免疫原性。缺乏免疫原性使细胞避免被宿主/患者免疫系统清除,从而发挥其治疗性作用而没有有害免疫和炎症反应。
Pollock等人在2006年描述,在大鼠中风模型(MCAo)中移植CTX0E03在移植后6至12周在感觉运动功能和总体运动不对称性方面均具有统计学上的显著改善。这些数据指示CTX0E03具有开发为治疗性细胞系所必需的适当生物学和制造特性。
Stevanato等人2009证实CTX0E03细胞在体外提取EGF、bFGF和4-OHT后以及体内植入MCAo大鼠脑后均下调c-mycERTAM转基因表达。体内c-mycERTAM转基因的沉默为潜在临床应用提供CTX0E03细胞的附加安全特征。
Smith等人2012年描述CTX0E03在中风(短暂性中脑动脉阻塞)大鼠模型中的临床前功效测试。结果指示,CTX0E03植入物可在3个月的时间范围内稳固地恢复行为功能障碍并且此作用专用于其植入部位的。病变拓扑结构是恢复的潜在重要因素,与较大面积的损伤相比,局限于纹状体的中风示出更好的效果。
STR0C05(ECACC保藏#04110301)
此c-MycERTAM转导的神经干细胞系衍生自12周胎纹状体。在bFGF、EGF和4-羟基他莫昔芬存在的情况下,使用限定无血清“人类培养基”将细胞系维持在层粘连蛋白包被的培养瓶上。在常规培养中,细胞系的倍增时间为3-4天,尽管在短期培养中,其倍增时间为20-30小时。
在生长培养基中,细胞为巢蛋白阳性、β-III微管蛋白阴性,而GFAP阳性细胞百分比较低。分化7天后,存在巢蛋白下调,βIII微管蛋白低水平表达,和GFAP强烈表达,表明该细胞系主要是星形胶质细胞。
此细胞系在遗传上正常,雄性XY,且在50倍以上群体倍增中稳定。
此处描述的细胞系是在质量保证条件下衍生出的,适合于重编程临床用途的指定细胞系。通过用胰蛋白酶的酶消化并结合机械研磨技术,在死后从12周妊娠胎GS006的纹状体中分离作为来源材料的人神经干细胞。一旦在培养物中建立,这些原代神经细胞就可用c-MycERTAM癌基因通过逆转录病毒来转化(如上述CTXOEO3细胞系所述),并分离出一系列克隆和混合群体细胞系。此系列的所有细胞系均衍生自层粘连蛋白包被的培养皿,并使用人类培养基(HM);DMEM:F12加上如下所述的指定补充剂。

STR0C05生长特性
在常规培养条件下,细胞从冷冻原种(经常在T180培养瓶中有2-4百万个细胞)中扩增。几种培养基改变后,汇合时使细胞传代。根据过程记录,STR0C05的群体倍增时间估计为3-4天,如下图所示。此倍增时间比对数期生长慢并且还包括传代期间的细胞丢失。
作为对STR0C05的对数期生长的更具代表性的评估,使用Cyquant荧光染料(分子探针)建立细胞增殖检验。使用Tecan Magellan荧光板读数器;ex.480nm;em 520nm测量细胞数量。
使STR0C05细胞传代,重悬于HM加生长因子中,并以5000细胞/孔的速度接种在层粘连蛋白包被的96孔带状孔板上。进行时程研究,通过每天从板上去除条带,每个时间点n=16个孔,去除培养基并将细胞冷冻在-70℃。
在此时程结束时,将所有冷冻的条带放回板上,并用Cyquant检验进行分析。简要地说,将细胞在裂解缓冲液中裂解,然后加入Cyquant试剂并在黑暗中放置5分钟。然后将每个孔的150ul样品转移到黑色Optilux板上,以在Tecan Magellan板读数器上读数。数据已导出到Excel电子表格以进行数值平均,且然后进一步导出到GraphPad Prism以进行分析。
结果示出,细胞在7天内稳定生长,估计倍增时间为20-30小时。
STROC05表型
已使用免疫细胞化学术对STR0C05的表型进行概述,以对神经干细胞标志物巢蛋白染色并对成熟的分化标志物(β-III微管蛋白(神经元)和GFAP(星形细胞))染色。
在存在和不存在生长因子与4-OHT的情况下确定STR0C05表型。细胞最初来源于STR0C05工作库存。使细胞传代并接种在96孔板中。
在室温下将细胞在4%多聚甲醛中固定15分钟,用PBS洗涤并用0.1%TritonX100/PBS渗透15分钟。然后在室温下用10%正常山羊血清(NGS)的PBS溶液阻断非特异性结合1小时。然后在室温下对细胞用对巢蛋白的抗体(1:200,日本化工(Chemicon))、β-III微管蛋白(1:500;西格玛(Sigma))和GFAP(1:5000;DAKO)进行探针检测过夜。用PBS洗涤后,然后将它们用溶解在1%NGS/PBS中的过滤的Alexa GoatαMouse 488(1:200;MolecularProbes)和Alexa GoatαRabbit 568(1:2500;Molecular Probes)在室温下处理1小时。然后将它们用PBS洗涤,并用Hoechst 33342(西格玛)复染2分钟,然后在荧光显微镜上分析。
从培养基中去除生长因子和4-OHT诱导细胞的形态学和表型变化,其伴随巢蛋白的下调。具体而言,一小部分细胞对神经元标志物β-III微管蛋白呈阳性,并获取具有延伸成树突/轴突外生长物的圆形细胞体的神经元形态。然而,更显著的表型变化是GFAP的上调,表明星形细胞谱系占优势。

在两个单独的实验中,与在其他细胞系情况下所见的清晰环带相比,没有探针杂交的证据。
细胞群体
本发明使用并涉及分离的干细胞群体,其中该群体基本上仅包含本发明的干细胞,即该干细胞群体是基本上纯的。在许多方面中,相对于构成总细胞群体的其他细胞,干细胞群体包含至少约75%或至少80%(在其他方面,至少85%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、99.5%、99.9%或100%)本发明的干细胞。例如,关于神经干细胞群体,与构成总细胞群体的其他细胞相比,此术语意指存在至少约75%、在一些实施方案中至少约85%、在一些实施方案中至少约90%并且在一些实施方案中至少约95%的纯神经干细胞。因此,术语“基本上纯的”指本发明的干细胞群体,其含有少于约25%、在一些实施方案中少于约15%并且在一些实施方案中少于约5%的非神经干细胞的细胞。
分离的干细胞可由某些标志物的独特表达概况来表征并与其他细胞类型的干细胞区分。当本文描述标志物时,其存在或不存在可用于区分神经干细胞。
在一些实施方案中,神经干细胞群体的特征可在于群体的细胞表达例如所有标志物巢蛋白、Sox2、GFAP、βIII微管蛋白、DCX、GALC、TUBB3、GDNF和IDO中的一个、二个、三个、四个、五个或更多个。
通常,神经干细胞为巢蛋白阳性。
“标志物”指生物分子,其存在、浓度、活性或磷酸化状态可经检测且用于标识细胞表型。
如果细胞群体中的至少约70%示出可检测水平的标志物,则通常认为本发明的干细胞带有标志物。在其他方面,至少约80%、至少约90%或至少约95%或至少约97%或至少约98%或更多的群体示出可检测水平的标志物。在某些方面中,至少约99%或100%的群体示出可检测水平的标志物。标志物的定量可通过使用定量RT-PCR(qRT-PCR)或通过荧光活化细胞分选术(FACS)来检测。应理解,仅以实施例的方式提供此列表,并不旨在进行限制。通常,如果群体中至少约90%的细胞示出如通过FACS检测的可检测水平的标志物,则认为本发明的神经干细胞带有标志物。
术语“表达的”用于描述细胞内标志物的存在。为了被认为是表达的,标志物必须以可检测的水平存在。“可检测水平”意指可使用标准实验室方法中的一种诸如qRT-PCR或RT-PCR、分子印迹、质谱或FACS分析来检测标志物。如果可在低于或等于35的交叉点(cp)值(在qRT-PCR阵列上的标准截止值)处合理检测到表达,则认为基因由本发明的细胞群体表达。Cp代表其中扩增曲线与检测阈值相交的点,且也可被报告为交叉阈值(ct)。
术语“表达(express/expression)”具有相应的含义。在低于此cp值的表达水平时,认为标志物不表达。本发明的干细胞中的标志物的表达水平与另一细胞例如间充质干细胞中的相同标志物的表达水平之间的比较可优选地通过比较从相同物种中分离出来的两种细胞类型来进行。优选地,物种是哺乳动物,并且更优选地,该物种是人类。此类比较可使用逆转录酶聚合酶链反应(RT-PCR)实验方便地进行。
如本文所用,术语“显著表达”或其等同术语“阳性”和“+”当用于标志物时应理解为在细胞群体中大于20%,优选地大于30%、40%、50%、60%、70%、80%、90%、95%、98%、99%或甚至细胞中的所有表达所述标志物。
如本文所用,相对于标志物所使用的“阴性”或“-”应理解为意指在细胞群体中少于20%、10%,优选地少于9%、8%、7%、6%、5%、4%、3%、2%、1%或没有细胞表达所述标志物。
细胞表面标志物的表达可使用常规方法和仪器(例如,Beckman Coulter EpicsXL FACS系统,其与可商购的抗体和本领域已知的标准方案一起使用)例如借助于特定细胞表面标志物的流式细胞术和/或荧光活化细胞分选术(FACS)来确定,以确定特定细胞表面标志物的信号是否大于背景信号。背景信号限定为由与用于检测每个表面标志物的特异性抗体相同亚型的非特异性抗体生成的信号强度。对于被认为是阳性的标志物,观察到的特定信号通常大于20%,优选地大于30%、40%、50%、60%、70%、80%、90%、100%、500%、1000%、5000%、10000%或以上,相对于背景信号强度更大。用于分析感兴趣的细胞表面标志物表达的替代方法包括使用针对感兴趣的细胞表面标志物的抗体通过电子显微镜进行的视觉分析。
干细胞培养和制备
用于干细胞培养的简单生物反应器是单隔室烧瓶,诸如常用的T-175烧瓶(例如BDFalconTM175cm2细胞培养瓶,750ml,经组织培养处理的聚苯乙烯直颈蓝色塞式密封螺钉盖,BD产品代码353028)。
条件永生性的干细胞通常可取自在T-175或T-500烧瓶中培养的增殖干细胞。
如本领域中已知的,生物反应器也可具有多个隔室。这些多隔室生物反应器通常含有由一个或多个膜或屏障隔开的至少两个隔室,该膜或屏障将容纳细胞的隔室与容纳气体和/或培养基的一个或多个隔室分开。多隔室生物反应器在本领域中是公知的。多隔室生物反应器的实施例是Integra CeLLine生物反应器,其含有培养基隔室和借助于10kDa半透膜分开的细胞隔室;此膜可允许营养素不断扩散到细胞隔室内,同时去除任何抑制性废物。隔室的各个可及性允许在不机械干扰培养物的情况下为细胞提供新鲜的培养基。有机硅膜形成细胞隔室的基底并通过提供通向细胞隔室的短扩散路径来提供最佳氧气供应和二氧化碳水平控制。根据本发明,可使用任何多隔室生物反应器。
术语“培养基(culture medium/medium)”在本领域中是公认的并且一般指用于培养活细胞的任何物质或制剂。如用于提及细胞培养的术语“培养基”包括细胞周围环境的组分。培养基可为固体、液体、气体的或相和材料的混合物。培养基包括液体生长培养基以及不维持细胞生长的液体培养基。培养基还包括凝胶状培养基,诸如琼脂、琼脂糖、明胶和胶原蛋白基质。示例性气态培养基包括气相,在培养皿或其他固体或半固体支持物上生长的细胞暴露于该气相。术语“培养基”还指旨在用于细胞培养的材料,即使该材料尚未与细胞接触。换句话说,准备用于培养的富含营养的液体是培养基。类似地,当与水或其他液体混合时变得适合于细胞培养的粉末混合物可被称为“粉状培养基”。“限定的培养基”指由化学组分限定的(经常纯化)组分制成的培养基。“限定的培养基”不含有特性较差的生物提取物,诸如酵母提取物和牛肉汤。“完全培养基”包括旨在支持特定物种的大多数或所有可行形式的生长的培养基。完全培养基常常包括复杂生物提取物。“适合于高密度培养物生长的培养基”是当其他条件(诸如温度和氧气传输速率)准许此类生长时允许细胞培养物达到3或更高的OD600的任何培养基。术语“基础培养基”指促进不需要任何特殊营养补充剂的许多类型微生物生长的培养基。大多数基础培养基一般包含四种基本化学组:氨基酸、碳水化合物、无机盐和维生素。基础培养基一般用作更复杂培养基的基础,向其中添加补充剂,诸如血清、缓冲液、生长因子、脂质等。在一方面中,生长培养基可为具有必需生长因子的复合培养基,以支持本发明细胞的生长和扩增,同时维持其自我更新能力。基础培养基的实施例包括但不限于Eagle基础培养基、最低必需培养基、DuIbecco改良的Eagle培养基、培养基199、营养混合物Ham’s F-10和Ham’s F-12、McCoy’s 5A、DuIbecco’s MEM/F-I 2、RPMI1640和Iscove改良的DuIbecco培养基(IMDM)。
由本发明的多能细胞及其子代制备的微粒
本发明的多能干细胞以及由那些细胞生成的分化细胞将制备微粒。在一方面,本发明提供由本发明的诱导多能干细胞或由那些iPS细胞生成的分化细胞而可获得的微粒。这些微粒可用于疗法。
由本发明的细胞获得的微粒也可用作外来运输物的递送媒介。在一些实施方案中,运输物可为外源核酸(例如DNA或RNA,特别是RNAi试剂,诸如siRNA或化学修饰的siRNA)、外源蛋白质(例如抗体或抗体片段、信号蛋白或蛋白药品)。在该技术领域中已知的是,可例如通过转染或电穿孔将运输物直接装载到微粒中。还已知操纵制备微粒的细胞可改变微粒的含量。
微粒的性质、含量和特性受制备它们的细胞的影响。因此,本发明有利地提供从单一充分表征的起始原料(即条件永生性的细胞)制备的各种各样微粒。例如,微粒可从iPS细胞或衍生自该细胞的任何更分化的细胞(例如已进入内胚层、中胚层或外胚层谱系的细胞)中分离。这允许由单个已知起始细胞提供许多不同微粒。
“微粒”是从细胞释放的直径为30至1000nm的细胞外囊泡。它受到包裹生物分子的脂质双层的限制。术语“微粒”在本领域中已知并且涵盖许多不同种类的微粒,包括膜颗粒、膜囊泡、微囊泡、外泌体样囊泡、外泌体、外泌体样囊泡、外泌体或外囊泡。不同类型的微粒根据直径、亚细胞来源、它们在蔗糖中的密度、形状、沉降速率、脂质组成、蛋白质标志物和分泌模式(即,遵循信号(可诱导型)或自发(组成型))来区分。下表1中描述四种常见的微粒及其区别特征。在某些实施方案中,微粒是外泌体。

人们认为微粒通过直接和间接机制充当供体和受体细胞之间的媒介而在细胞间通讯中发挥作用。直接机制包括受体细胞摄取微粒及其供体细胞衍生的组分(例如蛋白、脂质或核酸),这些组分在受体细胞中具有生物活性。间接机制包括微囊泡-受体细胞表面相互作用并引起受体细胞的胞内信号传导的调节。因此,微粒可介导受体细胞获得一种或多种供体细胞衍生的属性。已观察到,尽管在动物模型中干细胞疗法有效,但是干细胞似乎没有移植到宿主中。因此,干细胞疗法有效的机制尚不清楚。不希望受理论的束缚,发明人相信由神经干细胞分泌的微粒在这些细胞的治疗性效用中起作用且因此本身在治疗上有用。
如本文针对细胞所限定的,本发明的微粒经分离。
本发明提供由本发明的细胞制备的经分离干细胞微粒的群体,其中该群体基本上仅包含本发明的微粒,即该微粒群体是纯的。在许多方面,微粒群体包含至少约80%(在其他方面,至少85%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、99.5%、99.9%或100%)本发明的微粒。
在某些实施方案中,微粒是外泌体。外泌体的脂质双层通常富含胆固醇、鞘磷脂和神经酰胺。外泌体还表达一种或多种四跨膜蛋白标志物蛋白。四跨膜蛋白包括CD81、CD63、CD9、CD53、CD82和CD37。CD63是典型外泌体标志物。外泌体还可包括生长因子、细胞因子和RNA,特别是miRNA。外泌体通常表达标志物TSG101、Alix、CD109、thy-1和CD133中的一种或多种。Alix(蛋白质数据库登录号Q8WUM4)、TSG101(蛋白质数据库登录号Q99816)和四跨膜蛋白CD81(蛋白质数据库登录号P60033)和CD9(蛋白质数据库登录号P21926)是特征性外泌体标志物。
Alix是胞内体途径标志物。外泌体是内体来源的,且因此对此标志物呈阳性的微粒被表征为外泌体。本发明的外泌体通常对Alix呈阳性。微囊泡通常对Alix呈阴性。
在一些实施方案中,微粒诸如外泌体可装载有外源运输物。外源运输物可为蛋白质(例如抗体)、肽、药物、前药、激素、诊断剂、核酸(例如RNAi剂,诸如miRNA、siRNA或shRNA,或DNA或RNA载体)、碳水化合物或其他感兴趣的分子。运输物可例如通过电穿孔或转染直接装载到外泌体中,或者可通过对制备外泌体的细胞进行改造以使细胞在外泌体释放之前将运输物包裹入外泌体中而将运输物装载到外泌体中。将运输物装载到微粒诸如外泌体中在本领域中已知。
药物组合物
本发明的多能干细胞可经分化以生成可用于疗法中的细胞,且因此可被配制成药物组合物。本发明的多能干细胞以及从那些细胞生成的分化细胞将制备如本文别处所述的微粒,其也可用于疗法中且因此可被配制成药物组合物。
除了治疗性细胞或微粒外,药学上可接受的组合物通常还包括至少一种药学上可接受的载体、稀释剂、媒介和/或赋形剂。合适载体的实施例是林格氏乳酸溶液。此类组分的全面论述在Gennaro(2000)Remington:《药学的科学和实践(The Science andPractice ofPharmacy)》,第20版,ISBN:0683306472中提供。
短语“药学上可接受的”在本文用于指在合理医学判断范围内,适合于与人类和动物的组织接触而无过量毒性、刺激性、过敏反应或其他问题或并发症、与合理获益/风险比相称的那些化合物、材料、组合物和/或剂型。

本发明的药物组合物可呈多种形式。这些包括例如半固体和液体剂型,诸如冻干制剂、冷冻制剂、液体溶液或悬浮液、可注射和可输注溶液。药物组合物优选地是可注射的。
药物组合物一般将呈水性形式。组合物可包括防腐剂和/或抗氧化剂。
为了控制张力,药物组合物可包含生理盐,诸如钠盐。优选氯化钠(NaCl),其可以1至20mg/ml存在。可存在的其他盐包括氯化钾、磷酸二氢钾、脱水磷酸二钠、氯化镁和氯化钙。
组合物可包括一种或多种缓冲液。典型的缓冲液包括:磷酸盐缓冲液;三羟甲基氨基甲烷缓冲液缓冲液;硼酸盐缓冲液;琥珀酸盐缓冲液;组氨酸缓冲液;或柠檬酸盐缓冲液。缓冲液的浓度通常包括在5-20mM范围内。组合物的pH一般将在5至8之间,并且更通常在6至8之间,例如在6.5和7.5之间或在7.0和7.8之间。
组合物优选地是无菌的。组合物优选是非热原性的。
在典型的实施方案中,将细胞或微粒悬浮在组合物中,该组合物包含选自6-羟基-2,5,7,8-四甲基苯并二氢吡喃-2-羧酸Na+、K+、Ca2+、Mg2+、Cl-、H2PO4 -、HEPES、乳糖酸、蔗糖、甘露醇、葡萄糖、右旋糖-40、腺苷和谷胱甘肽中的1种、2种、3种、4种、5种、6种、7种、8种、9种、10种或更多种赋形剂。在一个实施方案中,组合物包含这些赋形剂中的所有。通常,组合物将不包括偶极非质子溶剂,例如DMSO。合适组合物可商购获得,例如此类组合物是有利的,因为它们允许将细胞在4℃至25℃下保存更长的时间(数小时至数天)或在低温下(即低于-20℃的温度下)保存。然后,干细胞可在解冻后以此组合物施用。

CTX-iPSC共享表征人类iPSC和ESC的许多特征。在重编程后,细胞形态从具有表征CTX细胞的延长过程的神经元表型显著变化到具有明显核仁且密集封装到表征人多能干细胞的“岛”中的细胞之间难以区分的分裂的小而圆的未分化细胞中的一个(图1C,图2)。CTX-iPSC在第21天终点表达组织非特异性碱性磷酸酶的酶标志物(图1D,图3)。
剖析CTX重编程要求的变化的转录因子组合。

CTX-iPSC与经典hPSC共享许多特征
CTX-iPSC的多能表型如图3所示。
(A)通过转染OKSML转录因子集来重编程为多能性对衍生自CTX细胞的两种不同细胞系(ii,iii)的CTX-iPSC的细胞和集落形态评估,示出这些重编程细胞系重演具有hPSC明显核仁特性的小而紧密排列的细胞的密集集落,与亲本CTX细胞的神经元表型(i)明显不同。
CTX-iPSC系表达酶标志物碱性磷酸酶(粉红色染色),如图3B所示。
如针对人多能干细胞所预期,流式细胞术示出CTX-iPSC对规范性多能转录因子OCT4和细胞表面抗原TRA-1-60和SSEA-4呈阳性,但不表达早期分化标志物SSEA-1(图3C)。
(D)RT-qPCR,其示出谱系特异性标志物在体外分化为内胚层、中胚层和外胚层后上调(各个CTX-iPSC系用阴影指示)。
c-myc-ERTAM转基因在CTX-iPSC中的状态
CTX-iPSC中转基因基因座的评估如图4所示。
(A)在G418中亲本CTX0E03细胞(顶部,第4天,第2行,第10天)和第4天时5个CTX-iPSC系(第3至第7行)的吉姆萨染色指示与c-myc-ERTAM相关联的NeoR基因的表达活性。
(B)驱动c-myc-ERTAM转基因的CMV-IE启动子的亚硫酸氢盐转化示出在基因座处的胞嘧啶甲基化状态(白色圆圈,未甲基化的CpG;黑色圆圈,甲基化的CpG;逗号,不确定的读数)。
由CTX-iPSC衍生治疗性细胞群体
使用RT-qPCR可示出,实现沿三种生殖谱系(内胚层、中胚层、外胚层)的分化。还可证实CTX-iPSC向治疗上相关细胞类型的分化。对于成体干细胞类型(间充质干细胞),已论证这一点。如技术人员将显而易见的,其他细胞类型可通过适当的培养条件来生成。特别地,免疫系统的细胞诸如T淋巴细胞、NK细胞和树突状细胞可通过Themeli等人(2013)在Nature Biotechnology(31),928-933中公开的方法进行分化。
图5示出衍生自CTX-iPSC的治疗性细胞群体的制备。(A)在mTeSR1培养基中的层粘连蛋白521上的CTX-iPSC。(B)在MSC培养基(α-MEM,10%FCS,25mM HEPES)中衍生自(A)中的细胞的塑料粘附候选间充质干细胞(MSC)。(C)根据ISCT基准,CTX-iPSC-MSC的流式细胞术示出它们表达MSC标志物CD73、CD90和CD105,但不表达CD14、CD20、CD34或CD45(蓝色,染色;红色,同型对照)。
结论
尽管体外永生和长期培养,但令人惊奇地示出,神经干细胞系CTX0E03可被外源转录因子重编程。
如细胞形态、细胞表面表达、转录因子和酶标志物以及多能性所限定的,CTX-iPSC与低传代原代细胞生成的常规iPSC显然没有区别。
CTX-iPSC中的c-myc-ERTAM基因座在至少某些细胞系中保持有活性。
临床相关细胞类型(例如MSC、免疫细胞,诸如T细胞、NK细胞和树突状细胞)可由CTX-iPSC生成。
在CTX-iPSC-MSC中经由4-OHT/C-MYC-ERTAM体系诱导细胞周期可准许CTX-iPSC-MSC可大规模制备用于同种异体疗法。
因此,CTX-iPSC代表非常有用的临床资源。它们可沿期望的谱系分化以生成靶标群体,诸如组织祖细胞类型或成体干细胞群体,且然后提供4-OHT以促进连续生长并防止细胞周期退出,并且相关联的另外分化可允许常规可规模化制备先前无法获得的临床相关亚群,而无需从原材料中反复分离细胞。
克隆或纯化步骤可用于由或多或少的异质分化培养物中生成所期望治疗性类型的纯群体,以大规模制备针对CTX本身不适合的病况的现成治疗,从而消除分化方案效率不高的现有技术中所见的缺点。这既适用于细胞本身,也适用于由不同细胞类型制备的外泌体级分,其中有效载荷分子的受体特征与由CTX细胞本身制备的那些分子不同。
此外,由于这些CTX-iPSC衍生物亚系衍生自已通过临床阶段安全性试验(CTX)的细胞系,因此有可能加快其进入新适应症临床试验的进程。
实施例2:重编程的CTX-iPSC的表征
示出重要基因表达的重编程诱导调节,证实CTX细胞证据已被正确地重编程。
结果提供在图6中。每个小图是从CTX创建的单细胞转录组数据的“tSNE”图。左上角的键指示绿色“云”是CTX,CTX-iPSC是蓝色并且已经受皮质分化方案且然后在其与CTX本身最接近时分析其转录组的CTX-iPSC是红色。每个云由代表单个细胞的点组成。灰色:无表达,橙色:中等表达;红色:高表达。这些图示出,在CTX中无活性的多能性基因已在重编程的细胞中被活化:POU5F1、NANOG、UTF1、TET1、DPP4、TDGF1、ZSCAN10和GAL。重要的是,在这些基因中,只有POU5F1是在重编程期间由外源提供的。相比之下,重编程为多能性时,由CTX表达的若干神经基因(NOGGIN、ADAM12、OCIAD2、NTRK3、PAX6)被下调。最后,在多能细胞的皮质分化时,GLI3(且很大程度上是PAX6)被上调。
诱导多能干细胞的生殖谱系分化及其染色(图7和图9)
方法
1.将CTX-iPSC或STR0C05-iPSC适当地平板接种在人层粘连蛋白521包被的8孔腔载玻片上。然后将它们适当地用适当的分化培养基(干细胞技术有限公司(StemCellTechnologies),目录号05230)处理5-7天,然后固定在4%甲醛的磷酸盐缓冲盐水(PBS)中,并在4℃下储存直至免疫染色。
2.对孔进行免疫染色如下:
1.通过在室温下于10%NGS/PBS中温育30分钟来用正常山羊血清(NGS)封闭。
2.将孔与一级抗体一起温育:在室温下或在4℃下过夜,在0.1%PBST(0.1%Triton-X-100/PBS)中适当稀释小鼠抗X抗体和兔抗Y抗体(见下表)2-4小时。
3.将孔用PBS洗涤3次,持续10分钟,或在PBS中于4℃下放置过夜。
4.将孔与二级抗体温育:山羊抗小鼠IgG-Alexafluor-488(1:300稀释)和/或山羊抗兔IgG Alexafluor-568(1:2000稀释)在PBS中于室温下温育2小时。
5.在室温下用PBS将孔洗涤3次。
6.将孔用在PBS中以1:10,000稀释的Hoechst 33342染色5分钟。
7.将孔用PBS洗涤3次,持续5分钟。
8.从载玻片上去除孔,加入2滴Vectashield,且将玻璃盖玻片置于顶部,然后通过荧光显微镜检查。
3.所采用抗体如下表所示。
结果:
CTX-iPSC多能性的进一步证实是通过提供向内胚层、中胚层和外胚层分化的证据,这是通过标识三个主要的胚胎层的蛋白质标志物(主要是转录因子)共同表达来示出。图7中的这些数据是对先前示出的RT-qPCR数据的补充。
实施例3:胎纹状体细胞的重编程
成功重编程另一种条件永生性的成体干细胞类型。此细胞系是STR0C05,衍生自胎纹状体细胞。
方法-将STR0C05细胞重编程为多能性
1.使用赛默飞世尔(Thermofisher.com)提供的Neon电穿孔仪,标识STR0C05细胞特异的转染条件的最佳范围。使用由仪器制造商建议的一系列不同参数诸如电压、脉冲持续时间等)将GFP表达质粒转染到细胞中时,评估获得的活细胞和绿色细胞的频率,以标识适合此细胞系的转染条件。
2.然后使用(1)中标识的条件,用Epi5重编程试剂盒(赛默飞世尔,目录号A15960;含有重编程质粒pCE-hOCT3/4、pCE-hSK、pCE-hUL和pCEmP53DD,表达转录因子POU5F1、SOX2、KLF4、L-MYC、LIN28和p53的显性阴性抑制剂)电穿孔STR0C05细胞,并将该细胞平板接种在人层粘连蛋白521上。每天使用在培养箱内运行的Incucyte Zoom自动相差显微镜监测孔。
3.一周后,将细胞再平板接种或保留在相同孔中,并将培养基换成mTeSR1(干细胞技术有限公司,目录号85850)。
4.监测孔,直到出现多能表型集落。
5.一旦足够大,用移液器吸头将单个集落挑入同样包被有hLn-521的24孔板的一个孔中,并扩增,直至冷冻或分析。
6.与先前的工作一样,使用Stemgent碱性磷酸酶染色试剂盒(目录号00-0055)进行碱性磷酸酶染色,并用补充有FITC偶联的小鼠抗人TRA-1-60抗体(BD目录号560380)的Becton Dickinson Stemflow抗体试剂盒(目录号560477)对多能干细胞标志物诸如SSEA1和SSEA4执行流式细胞术,以上试剂均根据制造商的说明。流式细胞仪样品在MiltenyiMACSQuant 10流式细胞仪上进行分析。
结果
结果示出在图8中,其中:
小图A示出用重编程因子转染后24天的重编程的STR0C05细胞的集落;
小图B示出在重编程的早期阶段碱性磷酸酶(红色)染色的STR0C05细胞,示出一些表达多能性标志物碱性磷酸酶的细胞;
小图C示出已建立的STR0C05-iPSC系;
小图D示出AP阳性集落在经受不同转染条件的孔中出现的频率不同;将没有集落的孔1用作为对照的GFP非重编程质粒转染,并且没有重编程的细胞,孔4和孔6几乎没有存活细胞;
小图E示出已建立的STR0C05-iPSC系是碱性磷酸酶阳性;和
小图F示出对于多能性标志物SSEA4也呈阳性,而对于早期分化标志物SSEA1则呈阴性。
还使用上述实施例2中所述的生殖谱系分化方法证实STR0C05-iPSC的多能性,并在图9中示出结果。通过标识三个主要胚胎层的蛋白标志物(主要是转录因子)的共表达论证内胚层、中胚层和外胚层的分化,如针对CTX的图7所示。
实施例4:衍生自重编程的iPSC的成体干细胞是多能性的
证实衍生自CTX-iPSC的成体干细胞的多能性。先前,我们已示出实施例流式细胞仪概况,其示出适当标志物表达以及候选CTX-iPSC-MSC(间充质干细胞)粘附于塑料的能力。此实验证实CTX-iPSC-MSC分化为若干不同细胞类型的能力。
方法-分化CTX-iPSC-MSC以证实多能性
1.为了评估脂肪和骨细胞形成,将CTX-iPSC-MSCs平板接种在6孔经组织培养处理的板中,并与促进脂肪形成和骨形成的市售培养基温育长达28天(脂肪形成:干细胞技术有限公司,目录号05412,骨形成:干细胞技术有限公司,目录号05465或R&D系统公司目录号CCMN007和CCM008),然后固定和染色。为了评估软骨形成,将CTX-iPSC-MSC在15ml试管的底部以团块形式成丸,并用软骨形成培养基(干细胞技术有限公司,目录号05455)培养,然后使用标准方法进行甲醛固定、石蜡包埋和切片。
2.阿尔辛蓝染色(软骨形成):将载玻片上的切片水合至蒸馏水中,用3%乙酸处理3分钟,然后用1%阿尔辛蓝的3%乙酸(pH 2.5)溶液中染色30分钟。然后将载玻片在流水中洗涤5分钟,在蒸馏水中漂洗,并在成像前用0.1%核固红的5%硫酸铝溶液中复染5分钟。
3.油红O染色(脂肪形成):将6孔板中的细胞用PBS洗涤,在室温下用10%甲醛固定10分钟,并用PBS洗涤两次。将它们在0.3%油红O的60%异丙醇/40%水的溶液中染色15分钟,并在成像前用重蒸馏水洗涤。
4.茜素红色S染色(骨形成):将6孔板中的细胞用PBS洗涤,在室温下用10%甲醛固定10分钟,并用PBS洗涤两次。将它们在室温下用2%pH 4.2的茜素红色S溶液染色15分钟,用水洗涤并成像。
结果
图10示出iPSC衍生的MSC分化为软骨(由唾液聚糖的阿尔辛蓝色染色示出)、脂肪(由油红色O染色细胞内脂质小滴示出)和骨(由沉积钙的茜素红色染色示出)的能力。
然后获得在存在或不存在4-OHT的情况下培养至高传代(20传代)的CTX-iPSC-MSC的流式细胞术概况。结果示出在图11中。被测细胞系是先前生成的亚硫酸氢盐数据指示的具有去甲基化的C-MYC-ERTAM启动子的细胞系,继而表明启动子在这些细胞中仍应有活性。有趣的是,当4-OHT诱导细胞周期时,该细胞系似乎可更好地维持其标志物分布:CD90和CD105表达更均匀且更高,而阴性标志物CD14、20、34和45更紧密地“关闭”。(此细胞系总是示出较低CD73表达,可能是抗体假象。)在第二小图中,经4-OHT处理的细胞似乎在分化时在生成骨方面更有效,表明4-OHT介导强迫的细胞周期改进从循环中的退出和效能丢失。
实施例5:重编程的CTX-iPSC-MSC的进一步表征
在不存在或存在4-OHT的情况下培养CTX-iPSC-MSC系。图12和13中用两种不同CTX-iPSC-MSC细胞培养物进行的实验的结果示出存在4-OHT/C-MYC-ERTAM并具有活性的情况下长期生长得到改善且更加一致。
此实施例示出,条件永生性的iPSC-ASC可更可靠地传播并持续更长时间。

13.一种制备多能干细胞的方法,其包含重编程条件永生性的干细胞的步骤。
14.根据权利要求13所述的方法,其中所述重编程包含将所述转录因子OCT4、L-MYC、KLF4和SOX2中的一种或多种以及任选地结合RNA的LIN28引入所述条件永生性的干细胞中。
15.根据权利要求14所述的方法,其中:
所述引入的转录因子包含或由OCT4组成;
所述引入的转录因子包含或由OCT4和SOX2组成;
所述引入的转录因子包含或由OCT、KLF4和SOX2组成;
所述引入的转录因子包含或由OCT4、KLF4、SOX2和MYC组成;或者
提供MYC活性,以通过在培养基中提供4-OHT以活化待重编程的所述干细胞中的c-myc-ERTAM转基因来促进所述重编程过程。
16.根据权利要求14或权利要求15所述的方法,其中使用一种或多种附加体质粒,任选地选自慢病毒、逆转录病毒或仙台病毒的一种或多种病毒载体或通过mRNA转染,将所述转录因子和任选的LIN28引入所述条件永生性的干细胞中。
17.根据权利要求13至16中任一项所述的方法,其还包含向所述内胚层、中胚层或外胚层谱系分化所述多能干细胞的步骤。
18.根据权利要求17所述的方法,其中所述内胚层、中胚层或外胚层谱系与所述被重编程的条件永生性的干细胞的谱系不同。
19.根据权利要求17或权利要求18所述的方法,其中所述多能干细胞分化为:
间充质干细胞、神经干细胞或造血干细胞;
体(成体)干细胞;
多能细胞、寡能细胞或单能细胞;
终末分化细胞;
免疫细胞,其任选地选自由T细胞、NK细胞、B细胞或树突细胞组成的列表;
软骨细胞;
脂肪细胞;或
骨细胞。
20.根据权利要求17至19中任一项所述的方法,其包含重活化由所述方法产生的所述细胞的所述条件永生性表型的步骤。
21.根据权利要求13至20中任一项所述的方法,其中所述被重编程的条件永生性的干细胞如权利要求3至8中任一项所限定。
22.根据权利要求13至21中任一项所述的方法,其包含选自以下项的一个或多个步骤:
培养由所述方法产生的所述细胞;
使由所述方法产生的所述细胞传代;
收获或收集由所述方法产生的所述细胞;
将由所述方法产生的所述细胞封装到一个或多个容器中;和/或
用一种或多种赋形剂、稳定剂或防腐剂配制由所述方法产生的所述细胞。
23.一种多能干细胞,所述多能干细胞通过权利要求13至16、21或22所述的方法而获得或可获得。
24.一种细胞,所述细胞通过权利要求17至22中任一项所述的方法而获得或可获得。
25.一种微粒,所述微粒通过权利要求1至12、23或24中任一项所述的细胞制备。
26.根据权利要求25所述的微粒,其是外泌体。
27.一种药物组合物,其包含根据权利要求1至12、23或24中任一项所述的细胞或根据权利要求25或权利要求26所述的微粒,以及一种或多种药学上可接受的赋形剂。
