Abstract
The invention discloses a preparation method of a DGP substance, which comprises the following steps: acidifying the substrate, carrying out high-temperature cracking under an anaerobic condition, dissolving a cracking product, and separating by adopting chromatographic column chromatography to obtain the DGP substance. The invention also discloses application of the DGP substance obtained by the preparation method in improving the mild and sweet taste characteristics of cigarette smoke in cigarettes.
Description
Technical Field
The invention belongs to the technical field of tobacco flavors, and particularly relates to a preparation method and application of a flavor 1,4:3, 6-dehydrate-alpha-D-glucopyranose.
Background
Sweetening is one of the important qualities sought for cigarettes. The sugar substances are added into the cigarettes, so that the generated volatile sweet compounds can be cracked, but the content is extremely low; if the adding amount of the saccharides is too large, the generated formaldehyde, acrolein and the like can influence the acid-base balance of the smoke, so that the astringency is increased, and the smoking experience is influenced.
1,4:3, 6-dianhydro-alpha-D-glucopyranose (abbreviated DGP) is ubiquitous in cigarette smoke and sugar cleavage products, a substance that can be transferred directly into mainstream smoke. Related researches show that the DGP has great influence on the cigarette sweetness, can improve the sweet return feeling of smoke and shows the mild and sweet taste characteristics of the cigarette.
The DGP substance contributes to the sweet taste of the cigarette, but the content of the DGP substance in the conventional cigarette is low. At present, no commercial DGP substance exists, so that the DGP substance is not generally used as a cigarette sweet taste additive until now, and the preparation and purification of the DGP substance are difficult. Therefore, research on the preparation method of DGP is the key content of tobacco flavor research.
Disclosure of Invention
The invention aims to provide a preparation method of a DGP substance, which has the characteristics of simple steps, high yield of the DGP substance and easy popularization. The obtained DGP substance is used in a cigarette filter stick to improve the mild and sweet taste characteristics of cigarette smoke.

The invention discloses a preparation method of a DGP substance, which comprises the following steps: acidifying the substrate, carrying out high-temperature cracking under an anaerobic condition, dissolving a cracking product, and separating by adopting chromatographic column chromatography to obtain the DGP substance.
Preferably, the method comprises the following specific steps:
adding a substrate into a sodium phosphate-methanol or phosphoric acid-methanol solution system to acidify the substrate, and then distilling the substrate under reduced pressure at a certain temperature to evaporate methanol to obtain an acidified substrate;
cracking the acidified substrate at high temperature for a period of time under the protection of inert gas to obtain a cracked product;
dissolving the cracked product with methanol, filtering with microfilm, loading C18 bonded silica gel filler to prepare chromatographic column, isocratic eluting with methanol solution, collecting fraction at 270nm for 15-18min, concentrating and drying to obtain DGP.
Preferably, the substrate of the step (I) is monosaccharide or glycan; the monosaccharide is D-glucose; the polysaccharide is cellulose or starch.
Preferably, the reduced pressure distillation temperature of the step (i) is 30-50 ℃.
Preferably, the inert gas in the second step is nitrogen, the cracking temperature is 300-.
Preferably, the aperture of the micro-membrane in the step (c) is 0.40-0.50 μm; the concentration of the methanol solution is 80-90 wt%.
7. The method according to claim 2, wherein the filler is C18 bonded silica gel, has a diameter of 10-15 μm, and has a column efficiency of 2600 or more.
Preferably, the chromatography column is a reverse phase high performance preparative chromatography column.
The invention also discloses application of the DGP substance obtained by the preparation method in improving the mild and sweet taste characteristics of cigarette smoke in cigarettes.
Compared with the prior art, the invention has the following obvious advantages:
1. the preparation method of the DGP substance is reported for the first time. Has the characteristics of simple preparation method, high yield and easy popularization. The process flow is short, the cost is low, and the process feasibility and the popularization are good.
2. The preparation method of the invention greatly improves the capability of substrate cracking and converting into DGP under the acidification condition, and the yield is about 30%.
3. The preparation method of the invention takes the reverse phase high-efficiency preparation of liquid phase as a means, and can prepare high-purity DGP with the purity of more than 95 percent. The self-filling of the preparation column is carried out by selecting different fillers, so that the selectivity of DGP is effectively improved.
4. When the DGP substance prepared by the method is used in cigarettes, the mild and sweet taste characteristics of smoke are obviously improved, and the fullness of aroma and the comfort of the smoke are also improved.

Detailed Description
In order to make the objects, technical solutions and advantages of the present invention more apparent, the present invention is further described in detail with reference to the following embodiments. It should be understood that the specific embodiments described herein are merely illustrative of the invention and are not intended to limit the invention. The examples do not specify particular techniques or conditions, and are performed according to the techniques or conditions described in the literature in the art or according to the product specifications. The reagents or instruments used are not indicated by the manufacturer, and are all conventional products available by purchase.
Example 1
The preparation method of DGP of this embodiment comprises the following steps:
1. substrate acidification treatment
The substrate was acidified by placing 20g of cellulose in 160g of sodium phosphate-methanol solution system. After the substrate was filtered of the solvent, the acidified substrate was obtained after evaporation of methanol at 40 ℃ under reduced pressure.
2. And (4) anaerobic high-temperature cracking.
Putting the acidified substrate into a thermal cracking glass tube, continuously introducing nitrogen to ensure an oxygen-free environment, and performing high-temperature cracking at 400 ℃ for 30min to obtain a cracked product. The cleavage products were analyzed as shown in FIG. 1, and the peaks marked with black boxes were DGP substances.
3. And (4) preparing a pure DGP substance.
Dissolving 5g of cracked product with 5g of methanol, filtering by a 0.45-micron microfilm, loading a reversed-phase high-efficiency preparation chromatographic column with 12-micron diameter C18 bonded silica gel filler, isocratically eluting by using 85% methanol water solution, collecting fractions at the retention time of 15.5-16.9min, concentrating and drying to obtain the DGP substance. The analysis chromatogram is shown in FIG. 2, and the purity is more than 98%.
Example 2
The DGP substance is used in the cigarette filter stick.
Firstly, the pure DGP substance prepared in the embodiment 1 is dissolved and diluted to 0.05 wt% by ethanol, and the solution is injected and added into a cigarette filter stick with a certain specification for sensory evaluation, and the cigarette without the pure DGP substance is used as a control sample. Sensory evaluation results show that the sweetness of the smoke of the cigarette added with the DGP substance is improved, and the fullness of the aroma and the comfort of the smoke are obviously improved.
The DGP substance can also be used in tobacco shreds, and can also be used in novel cigarettes, such as electronic cigarettes, heating non-combustible cigarettes, and the like, and the effects are similar.

A preparation method of a DGP substance is characterized by comprising the following steps: acidifying the substrate, carrying out high-temperature cracking under an anaerobic condition, dissolving a cracking product, and separating by adopting chromatographic column chromatography to obtain the DGP substance.
2. The preparation method according to claim 1, comprising the following steps:
adding a substrate into a sodium phosphate-methanol or phosphoric acid-methanol solution system to acidify the substrate, and then distilling the substrate under reduced pressure at a certain temperature to evaporate methanol to obtain an acidified substrate;
cracking the acidified substrate at high temperature for a period of time under the protection of inert gas to obtain a cracked product;
dissolving the cracked product with methanol, filtering with microfilm, loading C18 bonded silica gel filler to prepare chromatographic column, isocratic eluting with methanol solution, collecting fraction at 270nm for 15-18min, concentrating and drying to obtain DGP.
3. The process according to claim 2, wherein the substrate of step (i) is a monosaccharide or a polysaccharide; the monosaccharide is D-glucose; the polysaccharide is cellulose or starch.
4. The process according to claim 2, wherein the temperature of the vacuum distillation in the step (i) is from 30 to 50 ℃.
5. The method as claimed in claim 2, wherein the inert gas in step (II) is nitrogen, the cracking temperature is 300-600 ℃, and the cracking time is 20-60 min.
6. The method according to claim 2, wherein the pore size of the micro-membrane of step (c) is 0.40-0.50 μm; the concentration of the methanol solution is 80-90 wt%.
7. The method according to claim 2, wherein the filler is C18 bonded silica gel, has a diameter of 10-15 μm, and has a column efficiency of 2600 or more.
8. The method of claim 2, wherein the column is a reverse phase high performance preparative column.
9. Use of a DGP material obtained by the preparation method according to any one of claims 1 to 8 in cigarettes to improve the mild and sweet taste characteristics of cigarette smoke.
