本发明公开了一种水生动物饲料添加剂,包括如下组分:维生素A 1.52~2.24%、维生素D3 0.33~0.75%、维生素E 9.97~20.93%、维生素K3 1~7.97%、维生素B1 0.27~4.6%、维生素B2 3.43~4.6%、维生素B6 2.43~3%、维生素B12 0.25~0.35%、生物素0.69~1.35%、叶酸0.31~1.56%、烟酸8.97~13.46%、D‑泛酸5.69~8.94%、维生素C 12.78~33.27%、肌醇20.19~29.15%。使用本发明中饲料添加剂可以降低养殖成本,提高饵料系数0.84%,促进生长,提高强效免疫,存活率提高21.33%以上。

技术领域
本发明属于水产养殖技术领域,具体涉及一种水生动物专用维生素饲料添加剂及其应用。
背景技术
与陆生动物相比,只有虾和少数的肉食类鱼有胃,大部分的水生动物都没有胃,因而它们的肠道既要承担消化又要具备吸收的能力。和陆生动物相比,由于其简短的消化器官(消化道与体长之比为:猪14、鸡10、牛25、羊27、鲢鱼6、草鱼3、鲤鱼2.5和虾0.85),水生动物的食物在体内停留的时间更短,消化和吸收的效率更高。
对于水生动物,肠道不仅是消化吸收的器官,还是重要的免疫部位。因为所有的动物并不是生活在单一无菌的世界里,特别是水生动物,其消化道与外界环境一直相通。因此和陆生动物不同,水生动物没有相对稳定的内环境。由于水生动物吃的食物和所在的水环境都可能受到污染。长期在受到污染的环境生活,会影响水生动物正常生长,还会增加其发生各种疾病的风险。免疫系统的发展是为了防止外来物质损害多细胞生物。免疫系统在感染性微生物的压力下进化。在进化过程中,已发展出两种类型的免疫系统来抵抗外来物质,即先天(自然)免疫和适应性(后天)免疫。先天免疫系统在所有多细胞生物中都存在,能够识别非自身和危险信号,并通过摧毁入侵微生物和中和其毒性因子来保护宿主生物,是一个更古老的防御机制。利用生殖细胞编码受体来识别微生物病原体。先天免疫系统和脊椎动物的适应性免疫系统可通过此特征加以区分。因此,保持水生动物肠道菌群的稳定性特别重要。
培养健康的消化道,将直接影响水生动物的健康和生长。水环境和饵料极易影响水生动物肠道菌群,大部分细菌无法在消化道中定居(过路菌)。体内外环境会在水生动物处于健康状态时形成一个相对稳定的微生物菌群动态平衡;当出现饵料或环境变化等应激状态时,原始菌群平衡会受到影响,有害细菌会破坏宿主的正常防御系统。在某些条件下,病菌会迁移,定植和入侵其他组织器官,从而导致细菌性疾病的爆发。在肠道内的表现之一是厌氧菌与需氧菌比例明显下降。
维生素是水生动物生长与代谢中必要所需的微量有机物。在集约化与规模化的动物养殖生产中,复合维生素预混料是配合饲料中最关键的核心成分之一,对动物养殖行业的经济效益起着不可或缺的作用。维生素D作为一种神经内分泌-免疫调节激素,在调节细胞免疫中发挥重要作用。它不仅影响T淋巴细胞,B淋巴细胞,巨噬细胞和胸腺细胞的增殖和分化,缺乏维生素D还会影响这些细胞的功能。维生素D3可帮助健康肠道的有益菌黏附并在肠道壁上定植,形成保护性屏障。简而言之,在维生素D3的协助下,有益细菌与病原细菌竞争肠道黏膜黏附位点,从而导致致病菌无法与肠道黏膜相互作用,最终以肠道“过路菌”的形式被排出体外。
同时,维生素D3与钙磷协同,对消化道的菌群稳态有帮助,进而对胃肠道疾病有显著改善的影响。如维生素D3与钙磷缺乏会影响宿主体内代谢,极易引起机体免疫力降低,导致伤口处或与细菌接触的器官感染致病菌,特别是水生动物肠道,促使宿主发生炎症性肠道疾病或细菌性传染病的发生。维生素A在维持肠道菌群平衡中起重要作用,主要体现在其对肠道屏障功能和肠道黏膜免疫功能的调节。维生素A缺乏易影响肠上皮屏障的完整,降低肠道黏膜免疫能力,从而影响肠道菌群稳态,使致病菌群趁机而入,引起细菌感染性疾病的发生。维生素C通过促进胶原蛋白的形成可以参与表皮、黏液、鳞片的形成,在疾病预防和治疗过程中发挥着第一道屏障的作用。维生素C也是一种重要的抗氧化剂,可清除宿主体内的超氧离子、羟自由基和氢过氧化物等,保护生物膜免遭脂质过氧化的破坏。此外,维生素C具有很强的抗菌性能,可以消除肠道中的病原细菌。它具有强烈的消除和抑制荧光假单胞菌,副溶血性弧菌,嗜水气单胞菌和大肠杆菌生长的能力。维生素K3具有促进肝脏和胰腺中凝血酶合成凝血活素的功能。维生素K3缺乏时,容易引起宿主肠壁出血,引发肠壁炎症,容易导致肠道病原菌在炎症处的定植。维生素E可以改善氧的利用并促进组织细胞的呼吸。作为抗氧化剂,维生素E可以防止饲料、消化道和内源性代谢中易氧化的物质氧化。因此,维生素E间接保护了肠壁的受过氧化的破坏。
B族维生素的缺乏会导致水生动物营养物质匮乏,生长迟缓,从而导致水生动物肠道菌群之间的营养争夺。菌群间通过分泌生物菌素或改变自身代谢途径竞争性利用营养物质,导致肠道菌群稳态被打破。维生素B1以焦磷酸硫胺素(TPP)的形式存在于水生动物中,其主要参与宿主的糖代谢。硫胺素在体内可维持神经和肌肉的正常功能,维持食欲并参与消化液的分泌。若维生素B1缺乏,会引起宿主厌食、生长迟缓等症状,轻则降低饲料转化率,重则影响肠道菌群的平衡,甚至降低水生动物的存活率。生物素作为辅酶参与了脱羧-羧化反应和脱氨反应,在脂肪、氨基酸和碳水化合物的代谢过程中发挥着重要的作用。若缺乏生物素,不但影响肠道中的菌群稳态,还会导致水生动物出现生长迟缓等现象。若水生动物在幼龄时缺乏肌醇,严重者鳞、鳍、表皮剥落,暴露出宿主的骨骼和肌肉。此时容易导致病原菌侵入水生动物体内,争夺有益菌的养分,打破宿主体内肠道菌群稳态。
综上所述,维生素可通过影响肠道菌群结构来影响肠道健康。肠道菌群紊乱与吸收代谢,免疫功能及是否有炎症密切相关,因此保证肠道菌群稳态有利于机体的健康生长。本试验旨在研究专用型复合维生素对水生动物肠道微生物菌群稳态的影响,以此探讨最佳水生动物肠道微生物菌群稳态下专用型复合维生素的最适水平,为企业在此方面使用专用型复合维生素提供一个理论依据。
目前市场上有三大类控制水生动物菌群失调的产品:第一类产品使用抗生素直接杀菌,将有益菌和有害菌都杀灭,迅速控制宿主的细菌性疾病。但抗生素对动物菌群失调没有任何帮助,甚至是进一步破坏肠道微生物菌群的平衡,导致宿主进入亚健康状态;第二类产品是使用益生菌调节水生动物肠道微生物菌群平衡。由于发酵的工艺会降低益生菌的质量,导致市场上的益生菌质量参差不齐,甚者会有新的条件致病菌的摄入,引发水生动物的二次细菌性疾病的爆发;第三类产品是通过酸化剂降低肠道的pH值,给肠道中的有益菌(如乳酸菌或双歧杆菌)提供生存的环境,进而调节水生动物肠道微生物菌群平衡。此类物理性的酸化剂的质量也是良莠不齐,且需要每餐都添加,添加量在0.8-10%才有作用,此在一定程度上增加了养殖成本和风险。
中国水生动物通常以高密度繁殖方式进行养殖。水生动物疾病经常发生,长期的药物治疗已引起某些致病菌产生耐药性。且水产品中残留的药物通过食物转至人类体内,重则影响人类的健康。因此,寻找出一种安全、健康无毒的产品改善水生动物肠道微生物菌群平衡已成为目前水生动物研究的热点。

使用添加了本发明中的复合维生素的饲料可以改变水生动物肠道微生物的组成比例,提高益生菌数量,减少弧菌数量,更加有利于水生动物生长。
本发明的有益效果是:
1、使用本发明中的水生动物复合维生素饲料添加剂可以降低养殖成本,维生素具有多种生理活性功能并激活机体内代谢多种酶类,提高养分在体内沉积,提高饵料系数0.84%,促进生长,可提高强效免疫,增强抗病力,激活水生动物机体免疫细胞,提高免疫活性,可使存活率提高21.33%以上。
2、使用本发明中的水生动物复合维生素饲料添加剂可以调节肠道pH值,利于肠道菌群的平衡,可抑制弧菌、沙门氏菌等有害菌的生长繁殖,促进乳酸菌、芽孢杆菌等有益菌的生长,显著改善草鱼肠道的菌群结构,促进水生动物肠道健康。
3、使用本发明中的水生动物复合维生素饲料添加剂可以显著改善水生动物的肠黏液的分泌,增强上皮细胞的紧密连接,调节肠上皮细胞的增殖和分化,修复受损的肠道并维持良好的肠道黏膜形态结构和屏障功能的完整,并且增加杯状细胞的数量。

实施例1
一种水生动物饲料,包括基础日粮和饲料添加剂,基础日粮的组成及营养水平见表1,基础日粮和饲料添加剂的比例为999:1,即在999公斤的基础饲粮的基础上添加1公斤饲料添加剂。
其中,饲料添加剂包括复合维生素和载体,取复合维生素401.32g与载体598.68g配成1000g专用型复合维生素预混料,载体为脱脂稻谷壳。其中复合维生素包括如下组分:维生素A 2.24%、维生素D3 0.33%、维生素E 20.93%、维生素K3 1%、维生素B1 0.27%、维生素B2 3.43%、维生素B6 2.43%、维生素B12 0.25%、生物素0.69%、叶酸0.31%、烟酸8.97%、D-泛酸5.69%、维生素C 33.27%、肌醇20.19%,具体见表2。

实施例4
1.试验分组
随机挑选健康、大小均匀的草鱼1500尾(12g±1g),随机放入30个大小为2.5m×2.5m×1.6m的网箱里,每个网箱放入草鱼50尾。把30个网箱随机分为5个组,每组6个重复。
2.试验饲料及养殖管理
5个组中分别用实施例1~3和对比实施例1~2中的饲料进行投喂,每天的投喂时间固定在8:00和16:00。每日投喂量为鱼体重的3.5%。每15天根据鱼体体重调整投喂量。饲养期间,水体溶氧为5.5mg/L以上,水温为24~30℃,pH为6.9~7.2。养殖试验共进行95天。
3.生长性能检测
分别检测每周草鱼的成活率、增重、采食量以及饵料系数。
1)试验期间记录试验鱼死亡数,计算成活率;在试验开始和结束时称重,计算增重;计算每天的投饵量,分批收集残饵,试验结束后按绝干基础计算摄饵量和饵料系数。
2)存活率(%)=100×(试验开始时总尾数-试验期间死亡数)/试验开始时总尾数。
3)增重(GW)/(g/尾)=(试验鱼末重-试验鱼初始重)/尾数。
4)饵料系数=饲料消耗量/增重。
试验数据采用平均值±标准误表示(置信区间为0.05)。试验数据用SPSS 19.0软件进行方差分析和t检验。*代表P<0.05,**代表P<0.01,***代表P<0.001。
复合维生素对草鱼生长性能的影响见表3及图1~2,其中图1为复合维生素对草鱼饵料系数的影响,图2为复合维生素对草鱼存活率的影响。

结果表明,饲粮中不同水平的维生素对草鱼的饵料系数和成活率均有显著影响(P<0.05);由图1可知,对比实施例1~2与实施例1~3组间有显著差异性(P<0.01);说明不同比例的维生素配比会显著影响草鱼的存活率和饵料系数,相对于对比实施例1~2中的配比,实施例1~3中的草鱼存活率明显上升,饵料系数降低。提高饵料系数0.84%,促进生长,可提高强效免疫,增强抗病力,激活水生动物机体免疫细胞,提高免疫活性,使存活率提高21.33%以上。
维生素是鱼体内物质代谢中必不可少的特殊营养物质。它既不是构成身体结构的物质,又不能提供能量,但参与鱼体新陈代谢的调节,控制鱼的生长发育过程,提高机体的抗病能力,增强机体的抗应激能力,从而提高幼鱼的成活率。由本试验结果可得,在饲料中添加适宜配比的专用型复合维生素可显著降低草鱼的饵料系数,提高草鱼的存活率。且添加实施例2的专用型复合维生素可使草鱼的饵料系数在本试验中达到最低,存活率最佳。
4.肠道中微生物的数量测定
在实验期间的第30天,第60天,第95天分别取实验组和对照组各120尾鱼的肠道内容物,每10尾鱼的内容物合为一个样品。用无菌生理盐水将肠道内容物稀释至10、102、103、104、105、106倍。随机选择三个稀释倍数,并用移液枪吸取100μL溶液涂布在相应的筛选培养基上。每克肠道内容物中的菌落数(CFU)=平均菌落数×稀释倍数×10,其中微生物数量单位表示为CFU/mL。本试验中筛选用培养基及其对应微生物种类、培养条件和稀释倍数如表4和表5所示。

检测结果如表6和图3~5所示。图3为复合维生素对弧菌的影响情况,图4为复合维生素对芽孢杆菌的影响情况,图5为复合维生素对乳酸菌的影响情况。

结果表明,饲粮中不同水平的维生素对草鱼肠道中的弧菌、芽孢杆菌、乳酸菌的数量均有显著影响(P<0.05);弧菌数量中,30天、60天和95天的实施例1~3组的菌群数均显著低于对比实施例1~2组(P<0.01)。芽孢杆菌数量中,30天、60天和95天的实施例1~3组的菌群数量显著高于对比实施例1~2组(P<0.05),第30天和第95天实施例2的芽孢杆菌数量最高,第60天实施例3组的芽孢杆菌数量最高。乳酸菌数量中,第30天、第60天和第95天实施例1~3组数量相对对比实施例1~2均差异显著(P<0.01),第30天实施例3组乳酸菌数量最多,第60天和95天实施例2组乳酸菌数量最多。
水生动物的肠道菌群是一个处于动态平衡的整体,并由多种微生物组成,不同微生物之间以及微生物与宿主之间有非常紧密的联系。水生动物宿主的消化吸收、健康状况均受肠道菌群影响。肠道内有益菌属可通过改善宿主免疫调节功能、营养吸收能力来促进宿主生长,而肠道内的有害菌属则会增加肠道黏膜层的通透性,导致致病菌和其他大分子穿过黏膜屏障。所以,良好的肠道菌群稳态的建立对水生动物健康生长尤其重要。
极少量的弧菌是水生动物肠道中健康菌群的一员,当弧菌的量增多,在一定条件下成为致病菌而引起水生动物出现肠炎,重则引起肝胰腺坏死等疾病;乳酸菌属是水生动物的有益菌,主要是通过分泌乳酸等有机酸降低pH来抑制革兰氏致病菌扩散,可增强肠道免疫力、改善肠道结构、提高抗病力。由试验结果可得,在饲料中添加适宜量的专用型复合维生素可优化水生动物肠道微生物的组成比例,增加益生菌数量,减少弧菌数量。在草鱼饲料中添加实施例1~3组专用型复合维生素,弧菌数量都可极显著降低,芽孢杆菌和乳酸菌数量在第30天显著增多,芽孢杆菌在60天和95天极显著升高,乳酸菌数量在第95天显著上升,实施例1~3组可达到一致的效果,但从饲料成本而言,实施例2专用型复合维生素更佳;添加实施例3组专用型复合维生素,对第60天的芽孢杆菌数量可达到最佳效果。说明实施例1~3组的专用型复合维生素均可以显著改善草鱼肠道的菌群结构,效果旗鼓相当,但从饲料成本而言,实施例2组复合维生素效果更佳。
5.肠绒毛的形态观察和测量
在实验期间的第95天除去肠内容物后,将草鱼的前肠,中肠和后肠快速取出。每个肠段的长度约为1cm,将内容物浸入pH7.2~7.4的磷酸盐缓冲液中以将其洗净。卡诺氏固定剂固定12~24h,包埋并制成横切切片。常规苏木精-伊红(HE)染色和阿利西林-高碘酸盐席夫(AB-PAS)染色。使用光学显微镜观察和利用图像分析系统(IM50)测量肌层厚度和肠绒毛长度以及隐窝深度。从每个肠管中取出五个染色效果良好的切片并拍照,并为每个切片随机选择五根最长的肠绒毛进行测量。
对草鱼肠道肌层厚度和肠绒毛长度、隐窝深度的影响见表7及图6~8。其中图6为对草鱼肠道肌层厚度的影响,图7为对草鱼肠绒毛长度的影响,图8为对草鱼隐窝深度的影响。

由表7及图6可知,在前肠的实施例1~2组、后肠的实施例1~3组与对比实施例2组的肌层厚度间有统计学差异(P<0.05);在中肠中,对比实施例2组的肌层厚度与实施例1~3组之间有显著性差异(P<0.01)。此外,在同一试验组中,草鱼肠道肌层厚度中肠最大,后肠次之,前肠最小。
由表7及图7可知,各肠段中对比实施例1及前肠实施例1的绒毛长度与对比实施例2间均无显著差异(P>0.05);在前肠实施例2~3组和中肠实施例2组及后肠实施例2组的肠绒毛长度随复合维生素的增加逐渐增大,与对比实施例2组间有显著的统计学意义(P<0.05);中肠实施例1~2组和后肠的实施例1、3组的肠绒毛随复合维生素的增加,其长度逐渐增加,与对比实施例2组间有显著差异性(P<0.01)。在同一试验组中,草鱼肠绒毛长度从前肠、中肠到后肠逐步降低,前肠肠绒毛长度最长,中肠次之,后肠最短。
由表7及图8可知,前肠、后肠的试验实施例1~3组及中肠各试验组的隐窝深度显著低于对比实施例2组(P<0.05),此外,在前肠、中肠中,试验实施例2组的草鱼隐窝深度最浅,后肠中试验实施例3组的草鱼隐窝深度最浅。
肠道是机体消化吸收营养的重要场所。作为小肠的重要组成部分,肠绒毛不仅是吸收养分所必需的,而且它们的强而有规律的振荡还有助于排除有害细菌。另外,绒毛的表面积受绒毛形态变化的直接影响,从而影响机体吸收营养的能力。Caspary等报道指出,小肠对养分吸收的增强是由于肠绒毛长度的增加使小肠接触养分的面积增加而促使的,所以肠绒毛的形状与机体的生长发育有直接联系。隐窝深度可反映隐窝细胞的成熟度及增殖速率,隐窝越浅细胞成熟度越好,分泌功能越好。地穴细胞从绒毛的底部迁移到上部过程中逐渐分化形成具有吸收能力的柱状细胞。肠绒毛的高度和细胞数量密切相关。绒毛短时,吸收营养的能力会因成熟的绒毛细胞减少而降低。隐窝的深度反映了细胞产生的速率。细胞继续迁移并从隐窝底部分化至绒毛末端,形成吸收性绒毛以补充绒毛上皮的正常脱落。若这一过程变慢,则会降低基部的细胞形成速率,从而导致隐窝变浅。)绒毛长度/隐窝深度的比率(V/C值)全面反映了小肠的功能状态。率降低表明粘膜受到损害,消化吸收功能降低,动物的生长发育受到阻碍。
由试验结果可得,在增加中肠、后肠肌层厚度和降低前肠、后肠隐窝深度中,在饲料中添加实施例1~3组专用型复合维生素均有相同效果,均能显著增加肠道肌层厚度,降低隐窝深度;在增加前肠肌层厚度、中肠肠道绒毛长度,降低中肠隐窝深度中,实施例2~3组专用型复合维生素具有相同的效果;在增加前肠绒毛长度中,实施例2~3组专用型复合维生素均有显著效果;在增加后肠绒毛长度中,实施例2~3组专用型复合维生素均有显著效果。表明实施例1~3组专用型复合维生素均可作为专用型复合维生素,显著改善肠黏液的分泌,增强上皮细胞的紧密连接,调节肠上皮细胞的增殖和分化,进而修复受损的肠道并维持良好的肠道黏膜形态结构和屏障功能的完整性,但整体而言,实施例2组专用型复合维生素的作用最显著,实施例3组次之。
6.肠道杯状细胞的观察与计数
在实验期间的第95天从每个肠段中选择5块着色的AB-PAS染色切片,在光学显微镜下放大40×10倍观察、拍照并计数。每张染色切片随机选择5根最长的肠绒毛计算杯状细胞数量并取平均值。(从每个染色切片中随机选曲5条最长的肠绒毛,以计算杯状细胞的数量并取平均值。)结果见表8和图9。

备注:每个肠管随机取五根完整绒毛,在绒毛上皮层,从绒毛的顶部依次向下数100个柱状细胞,并记数其中的杯状细胞数,取平均值。
由表8及图9可知,虽然相对于对比实施例1~2,实施例1~3组的肠道杯状数量细胞无显著差异,但是,在前肠、中肠的试验中,实施例1~3组的草鱼肠道杯状数量细胞确明显高于对比实施例1~2组。而且前肠的试验实施例1组和中肠的试验实施例1~2组与对照组间有显著的统计学意义(P<0.05);前肠的试验实施例2组、后肠的试验实施例2组与对比实施例2间有显著差异性(P<0.01)。
作为典型的黏液细胞,杯状细胞主要分布在消化道和呼吸道的上皮中。杯状细胞是肠道黏膜免疫相关细胞的重要组成部分,是构成肠道黏膜免疫系统的第一道防御屏障,在抗感染、调节上皮细胞的完整性和对外源抗原的免疫反应中发挥着不可或缺的作用。肠道中杯状细胞数量的变化可以反映消化道的局部免疫情况。由试验结果可得,在饲料中添加实施例2组复合维生素均可有效增加各肠段杯状细胞的数量,促进杯状细胞的增殖与分化,且效果最佳。
综上所述,实施例1~3组复合维生素可显著提高水生动物饵料系数和存活率,改善水生动物肠道菌群平衡,且在本试验中,综合各肠道指标的最佳适用专用型维生素及其饲料添加剂的使用成本,在1吨基础饲粮的基础上添加1公斤实施例2专用型复合维生素预混料对改善水生动物肠道菌群平衡的效果最佳,实施例3组次之。
