Abstract
The invention provides a product containing cecropin and a method for detecting the content of the cecropin in the product, and the product containing the cecropin comprises the cecropin and a water-insoluble substance, wherein the water-insoluble substance comprises corncob powder. The method for detecting the content of cecropin in the product comprises the following steps of (1) dissolving cecropin in the product by using a dissolving solution; (2) filtering or centrifuging to obtain solution containing cecropin; (3) and (5) measuring the content of cecropin in the solution. The product containing cecropin can be used as feed additive for improving animal resistance and promoting animal growth and development. By the method for detecting the content of the cecropin in the product, whether the addition amount of the cecropin in the product is qualified or not can be quickly detected, and delivery of unqualified products is reduced.
Description
Technical Field
The invention relates to cecropin, in particular to a product containing cecropin and a method for detecting the content of the cecropin in the product.
Background
In the prior art, animals, particularly poultry and livestock are prevented from diseases by using antibiotics, but the antibiotics can improve the drug resistance, and along with the requirement of people on antibiotic-free products, people start to prevent diseases of the animals by using traditional Chinese medicine additives, but because the planting amount of traditional Chinese medicines is small, the traditional Chinese medicines are different in different planting conditions and different in drug properties of the traditional Chinese medicines in different years, the difference of the drug properties of the traditional Chinese medicines is large, and the disease prevention and the prevention of the animals are influenced. The application provides a product containing cecropin, which can be used as a feed additive to improve the resistance of animals and promote the growth and development of the animals. However, when the product is subjected to quality detection and quantitative detection, the product is dissolved by water, and then the supernatant is centrifuged to be detected, so that the detection result is far lower than the theoretical content, only 40% -60% of the theoretical content can be detected, the daily quantitative detection requirement on the product cannot be met, and the experimenters continuously improve the method, so that the content detection accuracy cannot be improved all the time. In the step (3), the content of cecropin in the solution is determined by using an ultraviolet spectrophotometry.
In the step (3), the content of cecropin in the solution is measured by using a high performance liquid chromatograph.
The dissolving solution is a mixed solution of water and an additive for reducing the tension of the water.
The additive for reducing water tension is at least one of ethanol, trifluoroacetic acid and tween 80.
The dissolving solution is a mixed solution of water and ethanol, and the volume fraction of the ethanol in the mixed solution is 30-60%.
The dissolving process of the step (1) also comprises vortex oscillation and ultrasonic treatment.
Vortex and shake for 30min, and sonicate for 30 min.
Compared with the prior art, the product containing cecropin and the method for detecting the content of the cecropin in the product can be used as a feed additive to improve the resistance of animals and promote the growth and development of the animals. Detailed Description
A product containing cecropin comprises cecropin and water insoluble substance, wherein the water insoluble substance comprises corncob powder.
The water-insoluble substance also comprises at least one of Maifanitum and rice hull powder. The method for detecting the content of cecropin in the product comprises the following steps of (1) dissolving cecropin in the product by using a dissolving solution; (2) filtering or centrifuging to obtain solution containing cecropin; (3) and (5) measuring the content of cecropin in the solution.
In the step (3), the content of cecropin in the solution is determined by using an ultraviolet spectrophotometry.
In the step (3), the content of cecropin in the solution is measured by using a high performance liquid chromatograph.
The dissolving solution is a mixed solution of water and an additive for reducing the tension of the water.
The additive for reducing water tension is at least one of ethanol, trifluoroacetic acid and tween 80.
The dissolving solution is a mixed solution of water and ethanol, and the volume fraction of the ethanol in the mixed solution is 30-60%.
The dissolving process of the step (1) also comprises vortex oscillation and ultrasonic treatment.

Randomly selecting 5 batches of products, dissolving by adopting the following different dissolving solutions, centrifuging, taking supernate, and detecting the cecropin content in the products by using a high performance liquid chromatograph (Agilent 1260).
(1) Dissolving the product containing cecropin by using a mixed solution of water and ethanol, performing vortex oscillation for 30min and ultrasonic treatment for 30min in the dissolving process, wherein the content measurement results of the cecropin in the product containing cecropin are shown in the following tables 1 and 2 under the condition that the ethanol in the mixed solution has different volume percentage contents.

From the results of tables 1 and 2, 30% ethanol was found to be the most preferable solvent. The solvent has good dissolving effect, the recovery rate can reach 100%, and the cost of 30% ethanol is the lowest compared with other solvents with the recovery rate of 100%.
(2) Dissolving the product containing cecropin by using a mixed solution of water and ethanol, wherein the volume percentage of the ethanol in the mixed solution is 30%, performing vortex oscillation for 30min and ultrasonic treatment for 30min during the dissolving process, and the measurement result of the content of the cecropin in the product containing the cecropin is shown in the following table 3.

The results are given in table 3: vortex oscillation for 30min, and ultrasonic treatment for 30min are very necessary in the dissolving step, and the cecropin can be fully dissolved only by treating the two steps in place.
(3) The original dissolving method adopts water as dissolving solution; in the application, a product containing cecropin is dissolved by using a mixed solution of water and ethanol, wherein the volume percentage of the ethanol in the mixed solution is 30%, and whether vortex oscillation and ultrasonic treatment are performed for 30min or not in the dissolving process is performed for 30 min; the results of measuring the content of cecropin in the cecropin-containing product are shown in table 4 below.

The results in table 4 show that the detection method greatly improves the dissolution rate of cecropin, so that the cecropin in the cecropin-containing product can be quickly and accurately detected.

7. The method for detecting cecropin content in a product according to claim 6, wherein: the additive for reducing water tension is at least one of ethanol, trifluoroacetic acid and tween 80.
8. The method for detecting cecropin content in a product according to claim 7, wherein: the dissolving solution is a mixed solution of water and ethanol, and the volume fraction of the ethanol in the mixed solution is 30-60%.
9. The method for detecting cecropin content in a product according to claim 3, wherein: the dissolving process of the step (1) also comprises vortex oscillation and ultrasonic treatment.
