s200: adding water wetting solution and lysis solution into a first extraction tank 9 to soak ginseng slices, keeping the first extraction tank 9 sealed, and vacuumizing the first extraction tank 9 through a vacuumizing device 10 to ensure that the pressure in the first extraction tank 9 is greater than 0.3 atmospheric pressure and less than 0.5 atmospheric pressure; the water-moistening liquid consists of the following raw materials in parts by weight: 19 parts of ethanol, 30 parts of azone, 21 parts of thiaketone and 25 parts of organic fluorine; the water-moistening solution can accelerate the absorption of ginseng to various medicaments, and is favorable for quickly extracting the DNA of the ginseng.
S300: heating the first extraction tank 9 by a heating base 18, raising the temperature of the liquid in the first extraction tank 9 to 40-50 ℃, and stirring the liquid in the first extraction tank 9 by a driving device 19 and a stirring rod 22;
s400: extracting the supernatant of the liquid in the first extraction tank 9 and introducing the supernatant into the second extraction tank 17;
s500: introducing the buffer solution, the binding solution, the crude washing solution, the refined washing solution, the eluent and the suspension in the first medicament bottle 11, the second medicament bottle 12, the third medicament bottle 13, the fourth medicament bottle 14, the fifth medicament bottle 15 and the sixth medicament bottle 16 into a second extraction tank 17, stirring and standing for 10 minutes; the buffer solution, the binding solution, the crude washing solution, the refined washing solution, the eluent and the suspension solution adopt the conventional medicaments. Such as: buffer solution: 11 parts of urea, 6 parts of sodium acetate and 13 parts of beta-mercaptoethanol; binding liquid: guanidine hydrochloride 9, diamine tetraacetic acid 13; crude washing liquid: isopropanol 5, guanidinium isothiocyanate 9; washing liquid: glycine 8 and deionized water 19; eluent: 11m tris-HCl buffer 11, MEDTA 17; the suspension is a suspension of magnetic beads.
S600: the supernatant in the second extraction tank 17 was taken and subjected to fluorescent PCR detection.
Claims (5)
1. A kit for identifying the authenticity of ginseng is characterized by comprising a base (1), wherein a liquid nitrogen storage tank (2), a first extraction tank (9), a vacuumizing device (10) and a second extraction tank (17) are arranged on the base (1); the first extraction tank (9) is of a sealing structure, a cover body of the first extraction tank (9) is provided with a first connecting pipe (4), a second connecting pipe (5) and a third connecting pipe (6), and the first connecting pipe (4), the second connecting pipe (5) and the third connecting pipe (6) are communicated with the first extraction tank (9); the liquid nitrogen storage tank (2) is communicated with the first connecting pipe (4), and a first valve body (3) is arranged on a pipeline between the liquid nitrogen storage tank (2) and the first connecting pipe (4); the second extraction tank (17) is communicated with the second connecting pipe (5), and a second valve body (8) is arranged on a pipeline between the second extraction tank (17) and the second connecting pipe (5); the vacuumizing device (10) is communicated with the third connecting pipe (6), and a third valve body (7) is arranged on a pipeline between the vacuumizing device (10) and the third connecting pipe (6); a heating base (18) is arranged in the base (1), and the bottom of the first extraction tank (9) is attached to the heating base (18); a driving device (19) is arranged below a first extraction tank (9) in the base (1), a stirring rod (22) is arranged in the first extraction tank (9), and the driving device (19) can drive the stirring rod (22) to rotate in the first extraction tank (9); this kit structure still includes first medicament bottle (11), second medicament bottle (12), third medicament bottle (13), fourth medicament bottle (14), fifth medicament bottle (15) and sixth medicament bottle (16), and first medicament bottle (11), second medicament bottle (12), third medicament bottle (13), fourth medicament bottle (14), fifth medicament bottle (15) and sixth medicament bottle (16) all draw jar (17) intercommunication with the second.

The kit for authenticating ginseng according to claim 1, wherein the driving means (19) comprises a driving motor (20) and a magnetic strip (21), the magnetic strip (21) is mounted on an output shaft of the driving motor (20), the stirring rod (22) has magnetism, and the stirring rod (22) is positioned at a position corresponding to the magnetic strip (21) at the bottom of the first extraction pot (9).

The kit for verifying ginseng authenticity according to claim 2, wherein the heating base (18) has a first groove body having a cylindrical shape, and the bottom of the first extraction tank (9) is located in the first groove body; the center position of the bottom of the first groove body is provided with a cylindrical second groove body, and the driving device (19) is positioned in the second groove body.

The kit for verifying the authenticity of ginseng according to claim 3, wherein the following methods are used: sequentially filling a buffer solution, a binding solution, a coarse washing solution, a fine washing solution, an eluent and a suspension into a first medicament bottle (11), a second medicament bottle (12), a third medicament bottle (13), a fourth medicament bottle (14), a fifth medicament bottle (15) and a sixth medicament bottle (16); the using method comprises the following steps:
s100: slicing ginseng and placing the sliced ginseng into a first extraction tank (9), filling liquid nitrogen in a liquid nitrogen storage tank (2) into the first extraction tank (9), and then smashing the sliced ginseng through a driving device (19) and a stirring rod (22);

s600: taking the supernatant in the second extraction tank (17), and carrying out fluorescence PCR detection.
5. The kit for identifying the authenticity of the ginseng according to claim 4, wherein the water-based moistening liquid is prepared from the following raw materials in parts by weight: 19 parts of ethanol, 30 parts of azone, 21 parts of thiaketone and 25 parts of organic fluorine.
