Abstract
本发明公开了一种复合生物祛瘢因子的营养祛瘢美白霜及其制备方法。分离健康动物(绵羊、山羊、鹿)胎盘的羊膜与子叶,把羊膜和子叶制成富含祛瘢因子的复合精华乳液,再与基础美白霜按比例混合乳化,成为营养祛瘢美白霜,即复合了生物祛瘢因子的营养祛瘢美白霜。该品具有显著的促进创伤愈合、抑制纤维化的作用,因而产生明显的祛瘢功效。同时,由于羊膜与子叶丰富的营养与活性成分,辅以基础美白霜的美白功能,使本发明产品具备了三大美容功能:即祛瘢、营养、美白。

技术领域
本发明涉及美容护肤品及美容护肤品制备方法的技术领域,尤其涉及一种复合生物祛瘢因子的营养祛瘢美白霜及其制备方法。
背景技术
瘢痕是各种创伤后所引起的正常皮肤组织的外观形态和组织病理学改变的统称,它是人体创伤修复过程中必然的产物。瘢痕是影响人们外观的重要指标,严重时会影响人的身心健康,发生在面部的瘢痕尤其是这样。瘢痕生长超过一定的限度,就会发生各种并发症,诸如外形的破坏及功能活动障碍等,给患者带来巨大的肉体和精神痛苦,尤其是烧伤、烫伤、严重外伤后遗留的瘢痕更是如此。
有瘢痕体质的人,即使是很微小的外伤,如蚊子叮咬,也能导致皮肤产生很大的、向外生长的瘢痕,并且不会在短期内消退。瘢痕体质的人,发生手术、烧伤、外伤后,更易形成表观症状明显的增生性瘢痕或瘢痕疙瘩。
有鉴于此,防止瘢痕产生或消除已产生的瘢痕不仅是美容行业的重大课题,也是医学临床所面临的课题之一。
祛瘢化妆品包括专用面膜、霜类、乳液、膏类等均为功能性产品,但此类产品的功能并非专一性,多数产品即有祛瘢功能,但同时还兼有其他美容作用,如美白、补水保湿或洁面、润肤等等。有调查资料显示,许多祛瘢化妆品的祛瘢功能并不显著,即不能说明其功能成分及其结构,更不能阐明其祛瘢机理。因此,造成以下三种局面:⑴产品的祛瘢功能不确实,有的产品实际上没有祛瘢作用;⑵产品祛瘢的功能成分不明确,造成消费者难以选择,无从下手;⑶不正确的使用祛瘢产品或使用不合格的产品,不仅不能迅速祛瘢,还可能会加重瘢痕或形成瘢痕疙瘩,产生不同程度的副作用。
所以,开发一种具有确实祛瘢功能的新产品以及产品制备技术是改变这种不利局面的有效途径。本发明不仅提供了复合生物祛瘢因子营养祛瘢美白霜的产品设计与开发技术,还阐明了产品功能成分的名称、来源、结构、生物学活性,以及发挥此种祛瘢功能的机理所在。
发明内容
本发明的首要发明目的在于提出了一种复合生物祛瘢因子的营养祛瘢美白霜,本发明含有生物祛瘢因子等多种生物活性成分,具有促进创伤愈合、抑制纤维化的作用,同时兼具美白、营养皮肤等多重美容功效。
本发明的第二发明目的在于提出了该复合生物祛瘢因子的营养祛瘢美白霜的制备方法,本发明方法运用组织裂解等物理方法将羊膜和子叶中的生物活性成分释放出来,再与基础美白霜结合制成本发明产品,工艺先进,保证了各成分的生物活性,适合产业化发展。
为了实现本发明的目的,采用的技术方案为:
一种复合生物祛瘢因子的营养祛瘢美白霜,其特征在于,包括基础美白霜、羊膜子叶复合精华乳液;所述基础美白霜各组分质量分数如下:

进一步,所述营养祛瘢美白霜还包括透皮剂、防腐剂、香精。
本发明复合生物祛瘢因子的营养祛瘢美白霜的制备方法,其特征在于,包括基础美白霜的制备、羊膜子叶复合精华乳液的制备。
其制备方法,具体为:
1.所述基础美白霜的制备,按上述比例添加成分,包括以下步骤:
(1)A相的制备:按配方所述质量分数值分别取肉豆蔻酸异丙酯、对羟基苯甲酸甲酯溶于乙醇,搅拌均匀,即为A相;
(2)B相的制备:按配方所述质量分数值取橄榄油、甘油,搅拌均匀,即为B相;
(3)基础美白霜的制备:按配方所述质量分数值取非离子表面活性剂与去离子水混合,依次加入A相、B相,混匀,再加入氨基酪氨酸乳化成霜,即为所述基础美白霜。
2.所述羊膜子叶复合精华乳液的制备,包括以下步骤:
(1)羊膜、子叶分离:将胎盘清洗后钝性分离出羊膜和子叶,清洗干净;
(2)羊膜、子叶预处理:将分离出的羊膜和子叶溶血后浸渍于1%的TritonX-100PBS溶液中,4℃振荡24h;
(3)羊膜子叶复合精华乳液制备:预处理后的羊膜和子叶,经剪碎→组织匀浆→组织裂解→微生物灭活→分装,制备成所述羊膜子叶复合精华乳液。
所述步骤(1)中,羊膜、子叶分离过程为:胎盘先用清水洗净2~3次,再用4℃预冷的磷酸盐缓冲液清洗2~3次;分离羊膜、子叶,分离的羊膜、子叶用蒸馏水清洗2~3次。
所述步骤(2)中,羊膜、子叶预处理的具体步骤为:
A:溶血:洗净的羊膜与子叶置于洁净的容器中,加入10倍(1:9,V/V)蒸馏水,4℃条件下保持24小时振荡,去除残留血迹。
B:去污除脂:取1%TritonX-100PBS溶液于洁净的容器中,把羊膜与子叶浸渍其中,4℃条件下保持4小时振荡,之后取出用注射用水洗净。
其中,所述1%TritonX-100配制:先配制成30%的Triton X-100储备液,临用时稀释至所需浓度。取Triton X-100及PBS混合,置37~40℃水浴中2~3小时,使其充分溶解混匀即可。
所述步骤(3)的具体操作为:
A:羊膜、子叶剪碎:将羊膜、子叶组织剪(绞)成碎块,大小约1×1cm;
B:组织匀浆:绞碎的羊膜、子叶组织按1:1与注射用水混合,经胶体磨匀浆处理15~20min得到组织匀浆液;
C:组织裂解:羊膜、子叶组织匀浆液导入超高压均质机,4℃下、裂解(8000~12000rpm)30~45min;
D:微生物灭活:组织裂解液中加入终浓度为0.01%的β-丙内酯,混匀,在5℃下灭活24小时,灭活后置37℃下水解2小时。
E:分装:洁净条件下分装,备用。
3.所述复合生物祛瘢因子的营养祛瘢美白霜的制备方法为:取羊膜子叶复合精华乳液与基础美白霜按10~15:90~85的体积比混合,2~8℃下,500~1000rpm,乳化5~10min,即成。
所述复合生物祛瘢因子的营养祛瘢美白霜分装后于2~8℃保存。
本发明的有益效果在于:
第一,本发明产品含有生物祛瘢因子等多种生物活性成分,具有促进创伤愈合、抑制纤维化的作用,同时兼具美白、营养皮肤等多重美容功效。

本发明是通过以下技术方案实现的。
基础美白霜的制备:第一步:按配方先取肉豆蔻酸异丙酯、对羟基苯甲酸甲酯溶于乙醇;第二步:橄榄油、甘油混匀;第三步:取非离子表面活性剂与去离子水混合,依次加入第一步、第二步获取的乙醇溶液和橄榄油、甘油混合液,混匀,再加入氨基酪氨酸乳化成霜,即为基础美白霜。
羊膜子叶复合精华乳液的制备:分离的羊膜、子叶经净化、预处理后剪碎,制成组织均浆,再导入超高压均质机,4℃下、裂解(8000~12000rpm)45~60min,成羊膜子叶裂解液。后者用β-丙内酯(终浓度0.01%)微生物灭活,即成。
营养祛瘢美白霜:取羊膜子叶复合精华乳液与基础美白霜按10~15:90~85的体积比混合,50~100rpm,乳化5~10分钟,即为复合生物祛瘢因子的营养祛瘢美白霜。
由羊膜和子叶制备的复合精华乳液中,含有丰富的蛋白质、细胞因子、生物酶、核酸、活性肽、硫酸软骨素、氨基酸、维生素等成分,这些成分具有显著的促进组织修复,防止纤维化功能而统称为生物祛瘢因子。
生物祛瘢因子发挥防瘢、祛瘢作用的机理是:促进组织修复的同时,防止新生肉芽纤维化,减少纤维蛋白数量、软化肉芽。具体表现为:1.促进上皮细胞增殖与分化,刺激靶细胞生长,诱导上皮细胞形成单层集落,从而产生组织修复与细胞再生作用;2.生物祛瘢因子通过抑制转化生长因子β(TGF-β)的合成,抑制纤维母细胞的活性,从而减少纤维蛋白的合成,防止组织纤维化与瘢痕形成。因此,由生物祛瘢因子参与的组织修复可以完全恢复损伤组织的完整性,相同细胞的增生与分化不改变原有组织、细胞的结构与功能,因而不发生纤维化、也不产生瘢痕。
基础美白霜中含有多种美白成分,其中3-氨基酪氨酸,其化学结构与酪氨酸相似,是可与酪氨酸竞争酪氨酸酶的结构类似物,而酪氨酸是最终形成优黑素(Optimalmelanin)和褐黑素(Pheomelanin)的主要原料,氨基酪氨酸通过与酪氨酸竞争酶而有效地抑制了优黑素和褐黑素的生成,从而产生显著的美白作用。
橄榄油被誉为“液体黄金”,富含与皮肤亲和力极佳的角鲨烯和人体必需脂肪酸,吸收迅速,有效保持皮肤弹性和润泽;橄榄油中所含丰富的单不饱和脂肪酸和维生素E、K、A、D等及酚类抗氧化物质,能消除面部皱纹,防止肌肤衰老,有护肤护发和防治手足皴裂等功效,另外用橄榄油涂抹皮肤能抗击紫外线防止皮肤癌。
肉豆蔻酸异丙酯广泛应用于化妆品中,可以起到保湿和滋润皮肤的作用,皮肤对本品的吸收性较好,能在皮层内与毛囊有效接触,渗入皮层深处,并将化妆品中的活性组分带入,充分发挥有效成分的作用。作为化妆品溶剂及皮肤保湿剂,渗透剂。
由羊膜和子叶制备的复合精华乳液中,含有丰富的蛋白质、细胞因子(如表皮生长因子、转化生长因子、成纤维细胞生长因子、肝细胞生长因子等)、生物酶、核酸、活性肽、硫酸软骨素、氨基酸、维生素等成分,这些成分具有显著的促进组织修复,防止纤维化和组织细胞营养功能。
产品的营养功能源于羊膜子叶复合精华乳液中丰富的营养物质(如蛋白质、细胞因子、多肽、氨基酸、维生素等),而产品的美白功能是基于基础美白霜中的多种美白成分(如氨基酪氨酸、甘油、对羟基苯甲酸甲酯等),通过这样的成分组合,使本发明产品成为一种多用途功能性防瘢祛瘢产品。
实施例1
(一)基础美白霜制备
1.配方
2.工艺
(1)A相的制备:按配方所述质量分数值分别取肉豆蔻酸异丙酯、对羟基苯甲酸甲酯溶于乙醇,搅拌均匀,即为A相;
(2)B相的制备:按配方所述质量分数值取橄榄油、甘油,搅拌均匀,即为B相;
(3)基础美白霜的制备:按配方所述质量分数值取非离子表面活性剂与去离子水混合,依次加入A相、B相,混匀,再加入氨基酪氨酸乳化成霜,即为所述基础美白霜。
(二)羊膜子叶复合精华乳液的制备
(1)羊膜子叶分离:健康山羊胎盘先用清水洗净3次,再用4℃预冷的磷酸盐缓冲液清洗3次;分离羊膜、子叶,分离的羊膜、子叶用蒸馏水清洗3次。
(2)羊膜子叶预处理
A:溶血:洗净的羊膜与子叶置于洁净的容器中,加入10倍(1:9,V/V)蒸馏水,4℃条件下保持24小时振荡,去除残留血迹。
B:去污除脂:取1%TritonX-100PBS溶液于洁净的容器中,把羊膜与子叶浸渍其中,4℃条件下保持4小时振荡。之后取出用注射用水洗净。
其中,1%TritonX-100配制方法为:取TritonX-100原液及PBS按体积比3:7混合,置37~40℃水浴中3小时使其充分溶解混匀,配制成30%的TritonX-100储备液;将此储备液稀释30倍配制成1%的TritonX-100溶液。
(3)羊膜子叶复合精华乳液制备:预处理后的羊膜与子叶,按以下方法制备羊膜子叶复合精华乳液:羊膜子叶剪碎→组织匀浆→组织裂解→微生物灭活→分装。
A:羊膜、子叶剪碎:将羊膜、子叶组织剪(绞)成碎块,大小:1×1cm;
B:组织匀浆:绞碎的羊膜、子叶组织按1:1与注射用水混合,经胶体磨匀浆处理20min得到组织匀浆液;
C:组织裂解:羊膜、子叶组织匀浆液导入超高压均质机,4℃下、裂解(8000~12000rpm)40min;
D:微生物灭活:组织裂解液中加入终浓度为0.01%的β-丙内酯,混匀,在5℃下灭活24小时,灭活后置37℃下水解2小时。
E:分装:洁净条件下分装,备用。
(三)复合生物祛瘢因子的营养祛瘢美白霜的制备
取羊膜子叶复合精华乳液与基础美白霜按10:90的体积比混合,2~8℃下,500~1000rpm,乳化8min,即成。
(四)分装与贮存
洁净分装,于2~8℃保存。
实施例2:
羊膜子叶复合精华乳液与基础美白霜按12:88的体积比混合,其余制备方法同实施例1。
实施例3:
羊膜子叶复合精华乳液与基础美白霜按15:85的体积比混合,其余制备方法同实施例1。
瘢痕对外观的影响不仅是皮肤的凹凸程度,还有瘢痕一般伴随着色素沉淀,为了验证本发明祛瘢美白霜的性能,特找来30位脸部有痘痕或外伤致瘢痕的测试者进行测试,将其随机分成3组,每组10人。测试者每天早晚洁面后涂抹本实施例1所制祛瘢美白霜于全脸,瘢痕处着重按摩促进吸收,30天后用皮肤测试仪测试瘢痕处黑色素含量、全脸含水量、瘢痕的凹凸程度及皮肤整体光泽度。
黑色素下降率=(使用本发明前瘢痕处黑色素含量-连续使用本发明30天后瘢痕处黑色素含量)/使用本发明前瘢痕处黑色素含量×100%;
皮肤水分上升率=(连续使用本发明30天后脸部皮肤水分含量-使用本发明前脸部皮肤水分含量)/使用本发明前脸部皮肤水分含量×100%。测试结果见下表:

从上表可以看出,采用本发明制备的复合生物祛瘢因子的营养美白霜具有良好的美白皮肤、淡化瘢痕、补充水分及营养皮肤的功效。瘢痕的形成机理很复杂,需要长期使用有此功效的护肤品进行保养,本发明富含天然生物活性成分,适合长期作为祛瘢美白护肤品使用。

2.根据权利要求1所述的复合生物祛瘢因子的营养祛瘢美白霜,其特征在于,所述动物为山羊、绵羊、鹿。
4.根据权利要求1~3所述的复合生物祛瘢因子的营养祛瘢美白霜,其特征在于,所述非离子表面活性剂为聚氧丙烯硬脂酸酯、聚氧乙烯山梨醇羊毛脂衍生物、聚氧乙烯月桂醚、壬基酚聚氧乙烯醚中的一种或任意组合。
5.根据权利要求1~4所述的复合生物祛瘢因子的营养祛瘢美白霜,其特征在于,所述营养祛瘢美白霜还包括香精、防腐剂。
6.一种如权利要求1所述的复合生物祛瘢因子的营养祛瘢美白霜的制备方法,其特征在于,包括基础美白霜的制备、羊膜子叶复合精华乳液的制备。
7.根据权利要求6所述的复合生物祛瘢因子的营养祛瘢美白霜的制备方法,其特征在于,所述基础美白霜的制备,包括以下步骤:
(1)A相的制备:按配方所述质量分数值分别取肉豆蔻酸异丙酯、对羟基苯甲酸甲酯溶于乙醇,搅拌均匀,即为A相;
(2)B相的制备:按配方所述质量分数值取橄榄油、甘油,搅拌均匀,即为B相;
(3)基础美白霜的制备:按配方所述质量分数值取非离子表面活性剂与去离子水混合,依次加入A相、B相,混匀,再加入氨基酪氨酸乳化成霜,即为所述基础美白霜。
8.根据权利要求6所述的复合生物祛瘢因子的营养祛瘢美白霜的制备方法,其特征在于,所述羊膜子叶复合精华乳液的制备,包括以下步骤:
(1)羊膜、子叶分离:将胎盘清洗后钝性分离出含子叶的羊膜,清洗干净;
(2)羊膜、子叶预处理:将分离出的含子叶的羊膜溶血后浸渍于1%的TritonX-100PBS溶液中,4℃振荡24h;
(3)羊膜子叶复合精华乳液制备:预处理后的含子叶羊膜,经剪碎→组织匀浆→组织裂解→微生物灭活→分装,制备成所述羊膜子叶复合精华乳液。
9.根据权利要求6~8所述的复合生物祛瘢因子的营养祛瘢美白霜的制备方法,其特征在于,所述的复合生物祛瘢因子的营养祛瘢美白霜的制备方法为:取羊膜子叶复合精华乳液与基础美白霜按10~15:90~85的体积比混合,2~8℃下,500~1000rpm,乳化5~10min,即成。
10.根据权利要求6~9所述的复合生物祛瘢因子的营养祛瘢美白霜的制备方法,其特征在于,所述复合生物祛瘢因子的营养祛瘢美白霜分装后于2~8℃保存。
