Abstract
本发明涉及生物医药领域,特别涉及FRT细胞株在制备筛选TRPV4调节剂的制剂或试剂盒中的应用。RT‑PCR和Western blot证实FRT细胞内源性表达TRPV4;倒置荧光显微镜下可观察到ANO1清晰表达在FRT细胞膜上,YFP‑H148Q/I152L清晰表达在FRT细胞的胞浆中,成功构建了共表达ANO1和YFP‑H148Q/I152L的FRT细胞模型;荧光淬灭动力学实验证实该模型可以筛选TRPV4调节剂;该模型可敏感检测细胞内钙浓度变化;Z'因子为0.728,该模型可高通量筛选TRPV4调节剂。综上,本发明成功构建了一种可以敏感高效筛选TRPV4调节剂的高通量模型。

技术领域
本发明涉及生物医药领域,特别涉及FRT细胞株在制备筛选TRPV4调节剂的制剂或试剂盒中的应用。
背景技术
瞬时受体电位离子通道(transient receptor potential,TRP)是一种在生物体内分布广泛的非电压依赖的、非选择性阳离子通道,TRP家族分为7个亚族:TRPA、TRPC、TRPM、TRPML、TRPN、TRPP及TRPV。瞬时受体电位香草素受体4型通道蛋白(transientreceptor potential vanilloid receptor 4,TRPV4)是TRPV亚家族的一个成员,广泛分布于心脏、大脑、肾脏、肝脏、肺、胰腺、卵巢、骨组织以及皮肤表面。TRPV4调节剂在许多生理和病理生理过程中都起着重要作用,如:TRPV4激动剂可以减少动脉粥样硬化斑块的形成,促进成年海马齿回神经干细胞的增值,促进血管和动脉的生成;TRPV4拮抗作用可治疗水肿,疼痛,胃肠道疾病和肺部疾病,例如咳嗽,支气管收缩,肺动脉高压和急性肺损伤。目前TRPV4调节剂作用研究还处于初始阶段,因其参与许多生理病理过程,有可能是多种疾病的潜在治疗靶点。目前,虽然市面上已有TRPV4的激活剂和抑制剂的出现,但是其中大多数存在选择性不强、效力不佳等问题。因此,TRPV4的相关研究成为了当今的研究热点,筛选TRPV4特异性调节剂具有重要研究价值和科研意义。
目前,筛选离子通道调节剂的方法主要有电生理技术和荧光染料法等。电生理技术因其直接、灵敏的优点而被公认为离子通道功能检测的“黄金标准”,但该方法操作繁琐,成本昂贵,对操作人员技术要求高,无法适用于高通量药物筛选。而荧光染料法如Fluo-3、Fluo-4染料等,与Ca2+结合后,染料荧光强度增强,该方法虽然能够直观、实时检测细胞内Ca2+浓度变化,但其缺点是操作复杂,试剂费用高,实验周期长,限制了TRPV4调节剂的高通量筛选。
发明内容
有鉴于此,本发明构建了基于钙激活氯离子通道(calcium-activated chloridechannels,CaCC)的瞬时受体势V4(transient receptor potential vanilloid receptor4,TRPV4)通道调节剂的高通量筛选模型。
为了实现上述发明目的,本发明提供以下技术方案:
本发明提供了FRT细胞株在制备筛选TRPV4调节剂的制剂或试剂盒中的应用;所述FRT细胞株中ANO1和YFP-H148Q/I152L共表达;
所述FRT细胞株通过荧光信号反应细胞内Ca2+浓度的变化进而筛选TRPV4调节剂;

TRPV4对Ca2+具有选择性渗透性,其对机械刺激、低渗透压及热能等多种刺激敏感,其激活会诱导Ca2+流入,从而增加细胞内游离Ca2+浓度。钙激活氯离子通道蛋白1(anoctamin 1,ANO1)是钙激活氯离子通道(calcium-activated chloride channels,CaCC)之一,在细胞内Ca2+升高的作用下开放并向细胞内转运Cl-,I-等阴离子。YFP-H148Q/I152L是对卤族元素碘十分敏感的黄色荧光蛋白双突变体,遇I-会发生荧光淬灭。因此,本研究利用稳定共转染ANO1和YFP-H148Q/I152L的Fischer大鼠甲状腺滤泡上皮(Fischerrat thyroid,FRT)细胞系作为基于CaCC靶向TRPV4筛选细胞模型,测定原理如图1。当TRPV4通道被激活时,Ca2+内流增加细胞内Ca2+浓度,从而引起CaCC的开放,细胞外I-被转运到胞浆内,使黄色荧光蛋白YFP-H148Q/I152L发生荧光淬灭。本方法可以快速敏感的检测细胞内钙信号的变化,实现对TRPV4调节剂的高通量筛选,具有稳定性强,灵敏度高,可反复传代的优点。同时,本模型不但为进一步研究和开发TRPV4新型药物提供参考,还为其他TRP通道筛选调节剂提供了初筛模型,具有相当广阔的应用前景。
在本发明的一些具体实施方案中,所述FRT细胞株的构建方法为:构建钙激活氯离子通道ANO1和对卤族元素敏感的黄色荧光蛋白双突变体YFP-H148Q/I152L真核表达载体,经脂质体转染、抗生素筛选和稀释,获取共表达ANO1和YFP-H148Q/I152L的FRT细胞。
在本发明的一些具体实施方案中,所述TRPV4调节剂包括激活剂或抑制剂;所述激活剂包括GSKl016790A、4α-PDD或RN-1747中的一种或多种;所述抑制剂包括GSK2193874、HC-067047、RN-1734。
本发明还提供了FRT细胞株在检测细胞内游离钙离子浓度中的应用;所述FRT细胞株中ANO1和YFP-H148Q/I152L共表达。

本发明中,RT-PCR检测FRT细胞中内源性表达TRPV4,所得PCR产物切胶回收后测序;Western blot检测FRT细胞中TRPV4蛋白的表达情况;应用脂质体转染法构建共表达ANO1和YFP-H148Q/I152L的FRT细胞模型,利用倒置荧光显微镜观察ANO1和YFP-H148Q/I152L在细胞的表达情况,应用荧光淬灭动力学实验测定细胞模型的有效性;加入TRPV4激活剂和抑制剂,应用荧光淬灭动力学实验检测模型能否筛选TRPV4调节剂;Fura-2荧光探针法检测加入TRPV4激活剂后细胞内的钙浓度;Z'因子评估细胞模型是否适用于高通量筛选。

本发明公开了FRT细胞株在制备筛选TRPV4调节剂的制剂或试剂盒中的应用,本领域技术人员可以借鉴本文内容,适当改进工艺参数实现。特别需要指出的是,所有类似的替换和改动对本领域技术人员来说是显而易见的,它们都被视为包括在本发明。本发明的方法及应用已经通过较佳实施例进行了描述,相关人员明显能在不脱离本发明内容、精神和范围内对本文所述的方法和应用进行改动或适当变更与组合,来实现和应用本发明技术。
本发明首次提出了相对荧光强度变化值/slope值/斜率值的概念:
本方法通过动态检测相对荧光强度的变化,并进一步量化相对荧光强度的变化,
每一个荧光强度变化值(也称作slope值/斜率值)对应一个细胞内的Ca2+浓度,结果显示相对荧光强度变化值/slope值/斜率值与细胞内Ca2+浓度呈现良好的正相关。

近年来,随着药物研发技术的迅速发展,离子通道成为了重要的药物靶点而备受关注。TRPV4作为离子通道之一,广泛分布在消化、神经、呼吸和心脑血管等系统,可被温度、机械刺激、渗透压等因素激活,可引起钙离子的流动,并主要通过对钙离子的调控和细胞内信号转导通路,影响细胞的增殖、分化、迁移和凋亡等,进而在疾病的发生发展中发挥重要作用。研究表明,TRPV4调节剂与动脉粥样硬化、纤维化、疼痛、炎症、癌症等疾病的治疗有着密切的联系。现已发现的TRPV4通道的调节剂均存在着选择性差,产生明显激活或抑制作用所需浓度高等问题。目前尚无高通量筛选TRPV4通道调节剂的方法,因此研究筛选TRPV4通道调节剂具有重大意义。
常用筛选离子通道调节剂的方法大多不适用于TRPV4通道大量化合物库的高通量筛选。因此,本研究建立了一种基于CaCC的高通量筛选TRPV4调节剂的方法,利用TRPV4可以升高细胞内钙离子浓度的进而开放CaCC,细胞外I-转运至细胞内,胞内黄色荧光蛋白YFP-H148Q/I152L的荧光发生淬灭的原理,检测荧光信号就可以实现对TRPV4通道调节剂的高通量筛选。Fisher大鼠甲状腺滤泡上皮细胞,具有贴壁紧,胰酶不易消化,生长速度适中,特别与CHO和COS-7等细胞比较具有更易使膜蛋白表达的特点,常用于研究氯离子通道及其调节剂高通量筛选的细胞模型,因此本研究选择Fischer大鼠甲状腺滤泡上皮细胞系作为目的细胞进行建模。同时,本模型可敏感检测细胞内钙离子浓度,用荧光变化slope值表征细胞内钙浓度,比Fura-2法具有更大的信号窗口且可以更直观反映细胞内钙浓度。本模型经传反复传代至30代以上,依然保持其稳定性,且Z'因子为0.73,完全符合高通量筛选。
此外,由于本模型只是TRPV4调节剂的初筛方法,存在一系列不足。模型在用于TRPV4激活剂的筛选时,可能会筛选出CaCC通道或其他内源性Ca2+通道的激活剂,筛选之后还需要通过后续实验会区分出假阳性结果,可应用不同激活剂和抑制剂组合在一定程度上区分结果的真伪。而利用本模型进行抑制剂筛选时则没有此问题。由于本模型内源性表达TRPV4,并不表达其他TRPV通道的其他蛋白,说明本研究筛选的激活剂和抑制剂均作用于TRPV4,但对于筛选的激活剂和抑制剂是否作用于TRPV通道的其他蛋白还需后续实验进一步验证。
综上,本模型是高通量筛选TRPV4通道调节剂的新方法,本模型具有筛选周期短,操作简便,大大降低了科研成本的优点。本模型不但为进一步研究和开发TRPV4新型药物提供初筛方法,还为其他离子通道筛选调节剂提供了新思路,具有相当广阔的应用前景。
数据处理所有实验均经过3次重复试验,采用GraphPad Prism 8.0软件对激活剂和抑制剂的剂量依赖关系进行非线性曲线拟合分析,并计算半数有效浓度EC50、半数抑制浓度IC50值,统计学分析采用单因素方差分析和Mann-Whitney检验,以P<0.05为差异有统计学意义。

本发明提供的FRT细胞株在制备筛选TRPV4调节剂的制剂或试剂盒中的应用中所用原料及试剂均可由市场购得。
下面结合实施例,进一步阐述本发明:
实施例1 RT-PCR检测FRT细胞中TRPV4的表达情况
取生长状态良好的FRT细胞株,按照TRIzol说明书提取FRT细胞的总RNA,Nanodrop2000检测总RNA浓度,测定RNA溶液的A260/A280的比值范围。对提取的RNA进行琼脂糖凝胶电泳,检测总RNA完整性。
本研究设计合成了8对特异性引物:TRPV1~TRPV6,其中TRPV4设计了2对引物:TRPV4-1和TRPV4-2。TRPV4-1、TRPV4-2及管家基因β-actin的特异的引物序列如表1所示。按照2×EasyTaq PCR SuperMix试剂盒说明书进行PCR扩增,将所得PCR产物进行琼脂糖凝胶电泳,利用凝胶成像仪进行曝光成像。用干净的手术刀将目的条带切下,采用切胶回收试剂盒对目的条带进行切胶回收,回收后的DNA溶液进行核酸测序。

实施例2 Western blot检测FRT细胞中TRPV4蛋白的表达情况
(1)提取蛋白
①取生长状态良好的FRT细胞500万个,用胰酶消化,待消化好后把细胞转移至1.5mL离心管中2000rpm离心5分钟,弃上清,细胞沉淀再用PBS清洗一遍后同样条件离心,弃上清。
②向细胞中加入RIPA裂解液100μL并充分混匀置于4℃层析柜中,放置10分钟。之后,将裂解好的细胞放入4℃预冷离心机中12000rpm离心15分钟,保留上清,即为提取的总蛋白液。
③采用BCA定量试剂盒对提取的蛋白质进行定量。首先将牛血清白蛋白(BSA)进行梯度稀释成终浓度分别为0、0.125、0.25、0.5、1和2mg/mL来制备蛋白标准曲线。为保证待测蛋白浓度测定值位于标准蛋白曲线范围内,将待测蛋白溶液稀释5倍;取一个96孔板将标准蛋白和待测定蛋白加入孔中,每个孔上样10μL。随后向每个孔中加入BCA工作液200μL(工作液:试剂A与试剂B按50:1比例混合而成),然后放置到水平摇床上37℃恒温孵育30分钟。
④孵育结束后,用酶标仪在562nm处读出吸光度并绘制标准曲线,算出蛋白浓度,然后待测蛋白溶液中加入适量5×loading buffer,并置于金属浴中100℃煮样5分钟,加热后的样品可以直接用于SDS-PAGE电泳。
(2)SDS-PAGE电泳
①配胶:配制SDS-PAGE分离胶和浓缩胶;根据待检测蛋白分子量大小选择分离胶所含丙烯酰胺浓度8%-15%(w/v)不等,浓缩胶一般都为5%(w/v)丙烯酰胺浓度。按照下表的配方配置胶,先配制分离胶,待分离胶凝固之后再加入浓缩胶并插入梳子,待浓缩胶完全凝固后拔掉梳子,胶的制备完成。

②电泳:将煮好的适量的蛋白样品加入孔中,电泳条件:浓缩胶阶段恒压80V运行30分钟,分离胶阶段恒压120V运行60分钟。
③转膜:将胶取出并弃掉浓缩胶部分,将分离胶和PVDF膜之间叠在一起固定好(上面是PVDF膜下面是分离胶,转膜前先将孔径大小是0.22μm PVDF膜用甲醇活化1分钟),进行转膜,转膜条件为恒流250mA运行90分钟。
④封闭:待转膜结束后把PVDF膜取出放入含有5%(w/v)脱脂奶粉封闭液中进行室温封闭1小时。
⑤孵育一抗与二抗:封闭好的膜加入含TRPV4的兔多克隆一抗[比例一般为1:1000(v/v)配制]的5%(w/v)脱脂奶粉溶液并转移至摇床上轻轻振摇4℃孵育过夜。第二天取出膜,加入5mL TBST在摇床上洗3遍,每遍5分钟。之后用羊抗兔二抗[比例一般为1:500(v/v)配制]的TBST避光孵育1小时,之后再用TBST避光洗膜3次,每次5分钟。
⑥显影:将结合了一抗与二抗的膜放到免疫印迹成像系统中进行扫描成像,最后得到相应的蛋白条带。
实施例3构建共表达ANO1和YFP-H148Q/I152L的FRT细胞模型
最适抗生素浓度的选择取状态良好的FRT细胞,分别加入浓度为1500μg/mL、1000μg/mL、800μg/mL、600μg/mL、400μg/mL、200μg/mL、100μg/mL的zeocin抗生素,培养两周后细胞全部死亡的最低浓度则为zeocin抗生素筛选的最适浓度。G418抗生素同理检测最适浓度。
构建共表达ANO1和YFP-H148Q/I152L的细胞株取状态良好的FRT细胞,按照Lipofectamine 3000说明书将ANO1转染到FRT细胞中,2天后利用倒置荧光显微镜下观察。利用最适浓度的zeocin抗生素进行筛选,3周后挑取倒置荧光显微镜下可见细胞膜上表达绿色荧光的细胞进行有限稀释。经过多次有限稀释得到稳定转染ANO1的单克隆细胞并进行扩大培养,获得表达ANO1的FRT细胞株。同理,将YFP-H148Q/I152L转染到已稳定表达ANO1的FRT细胞中,应用最适浓度G418抗生素筛选,挑取倒置荧光显微镜下细胞膜和胞浆均可见绿色荧光的单细胞克隆团,扩大培养后获得共表达ANO1和YFP-H148Q/I152L的FRT细胞株。
荧光淬灭动力学实验验证细胞模型的有效性取生长状态良好的共表达ANO1和YFP-H148Q/I152L的FRT细胞。将细胞分为两组:实验组和对照组,每组三个复孔。向实验组加入120μL含有ANO1特异性激活剂Eact(10μmol/L)的高浓度碘离子的PBS溶液,对照组加入钙激活氯离子通道抑制剂NFA(300μmol/L)孵育10min,再加入120μL高浓度碘离子的PBS溶液,采用Fluo star多功能酶标仪检测相对荧光强度动态变化。具体设置如下:发射光波长540nm,激发光波长500nm。以5个点/s的速度动态检测相对荧光强度,其中前2s为基线,2s后以200μL/s的速度向实验组孔中加入120μL含有Eact的高浓度碘离子的PBS溶液,向对照组孔中加入120μL高浓度碘离子的PBS缓冲液。
实施例4荧光淬灭动力学实验验证细胞模型功能
为了验证细胞模型可筛选TRPV4调节剂,将生长状态良好的共表达ANO1和YFP-H148Q/I152L的FRT细胞分为4组,每组三个复孔。其中3组加入TRPV4激活剂GSKl016790A、4α-PDD、RN-1747的高浓度碘离子的PBS溶液,另外1组加入TRPV4抑制剂HC-067047孵育10min,再加入高浓度碘离子的PBS溶液,采用Fluostar多功能酶标仪检测相对荧光强度动态变化,利用Excel软件对原始数据进行宏计算,得到荧光变化的斜率值(slope)。
将生长状态良好的共表达ANO1和YFP-H148Q/I152L的FRT细胞分为三组,采用倍比稀释的方法获得不同浓度的激活剂和抑制剂。其中三组分别加入不同浓度的GSKl016790A、4α-PDD、RN-1747三种TRPV4激活剂,采用多功能酶标仪进行检测,加入高浓度碘离子PBS缓冲液,记录相对荧光强度动态变化。另外三组加入不同浓度的GSK2193874、HC-067047、RN-1734三种TRPV4抑制剂孵育10min,同样采用多功能酶标仪进行检测,加入含有GSK1016790A的高浓度碘离子的PBS缓冲液,记录相对荧光强度动态变化。计算slope值,使用GraphPadPrism 8.0软件绘制模型对不同激活剂和抑制剂的浓度依赖曲线。
实施例5 Fura-2荧光探针法验证细胞内钙离子变化
将稳定共表达ANO1和YFP-H148Q/I152L的FRT细胞制成细胞悬液,加入Fura-2/AM,使其终浓度为5μmol/L,于37℃孵育30min。洗涤细胞,离心后制成细胞悬液。上机采用Fluostar多功能酶标仪检测,使用340和380nm双激发源在510nm处记录荧光强度。测定时记录静息时和加入不同浓度的GSK1016790A后的340nm/380nm荧光比值。加入TritonX-100及EGTA测定最大荧光和最小荧光。根据荧光比值计算Ca2+浓度。
实施例6 Z'因子评估
Z'因子是评估高通量筛选的一种重要指标。计算公式如下:Z'=1-3×(|SDpositive|+|SDnegtive|)/(|Meanpositive|-|Meannegative|。
将稳定共转染ANO1-YFP-H148Q/I152L的FRT细胞接种于到黑壁透明底的96孔板中,培养18h。以含钙镁离子的PBS缓冲液洗涤两组细胞3次,加入50μL含钙镁PBS缓冲液,向96孔板的6列加入10μmol/L GSK1016790A作为阳性对照(positive),同时将另外6列加入PBS溶液作为阴性对照(negative),检测96孔板的Z'因子值。
效果例1
1 RT-PCR检测TRPV4表达情况
提取总RNA浓度为413.2ng/μL,测定总RNA溶液的A260/A280的比值为1.83。琼脂糖凝胶电泳结果显示,如图2A,总RNA完整性良好可用于进一步cDNA的合成。
RT-PCR结果显示,TRPV4-1、TRPV4-2和β-actin分别在395bp、455bp和260bp处出现特异性条带,与预期的目的产物大小一致,如图2B。TRPV1、TRPV2、TRPV3、TRPV5、TRPV6特异性引物均未扩增出条带。将TRPV4条带的切胶回收产物送生工公司测序,测序结果在chromas软件上进行分析,峰图结果如图2C,无重叠峰,其中竖线位置为内含子。所测核苷酸序列在NCBI-BLAST进行比对,与GenBank数据库收录的TRPV4的基因序列相似性为100%,证明所克隆DNA片段即为目的基因片段,如图2D。结果表明,FRT细胞在mRNA水平上内源性表达TRPV4。
2Western blot检测TRPV4蛋白表达水平
Western blot结果分析显示:FRT细胞中有相对分子量为98KDa的TRPV4蛋白表达,如图3。结果表明,FRT细胞在蛋白水平上内源性表达TRPV4蛋白。因此,FRT细胞内源性表达TRPV4,可利用FRT细胞进行TRPV4通道调节剂的筛选进行建模。
3共表达ANO1和YFP-H148Q/I152L的FRT细胞模型构建结果
zeocin抗生素筛选的最适浓度为1000μg/mL,G418抗生素筛选的最适浓度为1000μg/mL。
通过倒置荧光显微镜清晰可见FRT细胞膜上呈绿色荧光,结果表明ANO1表达在细胞膜上,如图4A;FRT细胞胞浆内和细胞膜上均可见绿色荧光,结果表明YFP-H148Q/I152L表达在胞浆中,如图4B。结果证明,成功构建稳定共表达ANO1和YFP-H148Q/I152L的FRT细胞株。
酶标仪结果显示,实验组的相对荧光强度显著下降,表明ANO1开放,I-内流,黄色荧光蛋白发生淬灭,对照组的相对荧光强度无明显变化,表明NFA抑制了ANO1的开放,如图4C所示。如果表达位置错误,FRT细胞将无法转运细胞外I-且不具备荧光淬灭的生物学功能,通过荧光淬灭动力学试验进一步验证了ANO1表达在细胞膜上而非细胞外。上述结果表明,共表达ANO1和YFP-H148Q/I152L的FRT细胞具备转运细胞外I-致使荧光淬灭的生物学功能,从另一个方面验证了ANO1定位和功能的正确性,因此细胞模型构建成功。

4细胞模型的功能验证
加入TRPV4激活剂GSKl016790A、4α-PDD、RN-1747后,荧光迅速淬灭。而加入TRPV4抑制剂HC-067047后,荧光不淬灭,如图5A。加入激活剂组荧光斜率值显著高于加入抑制剂组,各激活剂组与抑制剂组具有显著性差异(P<0.001),如图5B,表明模型具有筛选TRPV4调节剂功能。

在加入不同浓度的TRPV4激活剂和抑制剂后,荧光信号呈现不同的变化。激活剂和抑制剂浓度与荧光slope值呈现剂量-效应依赖关系。结果采用GraphPad Prism 8.0软件进行非线性曲线拟合分析,浓度取对数作图,得到浓度效应曲线结果显示,GSKl016790A、4α-PDD、RN-1747的EC50分别为9.17μmol/L、50.06μmol/L、147.50μmol/L,见图6A。GSK2193874、HC-067047、RN-1734的IC50分别为5.74μmol/L、10.91μmol/L、72.49μmol/L,见图6B。结果表明,slope值反映激活剂和抑制剂对TRPV4通道的作用具有浓度依赖性,证实了本模型可用于TRPV4通道调节剂的筛选。

5Fura-2荧光探针法的检测
加入不同浓度的GSKl016790A后,GSKl016790A浓度越大,相对荧光强度下降的幅度越大,荧光变化slope值越大,如图7A所示,结果表明荧光变化slope值与GSKl016790A浓度呈剂量依赖关系。同时细胞内Ca2+浓度瞬时升高,随着GSKl016790A浓度的升高,胞浆内Ca2+浓度越高,其浓度与GSKl016790A浓度呈剂量依赖关系,如图7B所示。分析结果表明,荧光变化slope值随着胞浆内Ca2+浓度增加而增加,Ca2+浓度越大,荧光变化slope值越大,如图7C所示。结果表明,FRT细胞模型荧光淬灭slope值与细胞内Ca2+浓度呈正相关,通过荧光变化slope值可反映细胞内Ca2+浓度,且细胞荧光淬灭slope比细胞内Ca2+浓度值信号窗口更大。因此该细胞模型可敏感检测细胞内Ca2+浓度的变化。

6Z'因子评估
Z'因子结果值为0.728,如图8。SDpositive值为5.06,SDnegtive值为1.57,Meanpositive值为78.2,Meannegative值为5.07,根据公式计算结果,模型的Z'因子为0.728,如图8。一般认为当Z'因子值大于0.5时,该高通量方法比较理想,因此该细胞模型适用于TRPV4调节剂的高通量筛选。

详细说明如下:价格:invitrogen公司的检测细胞内钙浓度的荧光探针(Fluo4)市场价格为4196.00人民币。
CaCC法灵敏度高于荧光探针法的原因:
(1)细胞内Ca2+浓度仅仅为nM级别或者μM级别,很少能达到mM级别,也就是说细胞内Ca2+浓度的微量性决定了其直接检测的难度;
(2)CaCC法是通过YFP双突变体的相对荧光信号的变化间接反映细胞内Ca2+浓度,YFP双突变体具有极强的碘离子敏感的特性;CaCC单个通道在碘离子依次通过的情况下,每秒钟可转运106个碘离子;而且通过稳定转染,本细胞模型具有高表达CaCC的特性(也就是说每个细胞上表达多个CaCC);此外YFP的荧光信号强(YFP的荧光强度是普通荧光信号如GFP即绿色荧光蛋白的数倍)。
如图9所示,结果表明:在细胞内Ca2+浓度在40、50、100、200、400和800nM时CaCC法灵敏度均显著优于荧光探针法。
综上所述,CaCC灵敏度高于荧光探针法。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。

1.FRT细胞株在制备筛选TRPV4调节剂的制剂或试剂盒中的应用;
所述FRT细胞株中ANO1和YFP-H148Q/I152L共表达;
所述FRT细胞株通过荧光信号反应细胞内Ca2+浓度的变化进而筛选TRPV4调节剂;
所述荧光信号为相对荧光强度变化值/slope值/斜率值,所述相对荧光强度变化值/slope值/斜率值与TRPV4调节剂浓度成剂量依赖关系;
其中,相对荧光强度变化值/slope值/斜率值与细胞内Ca2+浓度呈现良好的正相关;

4.如权利要求3所述的应用,其特征在于,所述与TRPV4通道相关疾病包括血管疾病、动脉粥样硬化、神经疾病、水肿、疼痛、胃肠道疾病、肺部疾病中的一种或多种;所述肺部疾病包括咳嗽,支气管收缩,肺动脉高压和急性肺损伤中的一种或多种。
5.筛选TRPV4调节剂的制剂或试剂盒,其特征在于,包括FRT细胞株以及可接受的助剂;所述FRT细胞株中ANO1和YFP-H148Q/I152L共表达;
所述FRT细胞株通过荧光信号反应细胞内Ca2+浓度的变化进而筛选TRPV4调节剂;
所述荧光信号为相对荧光强度变化值/slope值/斜率值,所述相对荧光强度变化值/slope值/斜率值与TRPV4调节剂浓度成剂量依赖关系;
其中,相对荧光强度变化值/slope值/斜率值与细胞内Ca2+浓度呈现良好的正相关;
