The DNA Oligos were annealed to double strands and ligated to pUC57-sgRNA expression vector (Addgene ID:51132) linearized with Bsa I endonuclease to construct a sgRNA expression vector. pST1374-NLS-flag-linker-Cas9(Addgene ID:44758) was selected as the Cas9 expression vector. sgRNA and Cas9 expression vectors were transcribed into injectable sgRNA and Cas9-RNA using an in vitro transcription kit (Ambion Am 1345).
(3) Preparation of F0-generation rats
Selecting 15 SD female mice of 4-6 weeks old, injecting 20 IU/mouse of Pregnant Mare Serum Gonadotropin (PMSG) into abdominal cavity, and injecting 20 IU/mouse of human chorionic gonadotropin (hCG) after 48h, and performing superovulation. According to the following steps: 1 ratio and adult male mice, collecting about 150 fertilized eggs for microinjection. 30 vasectomized male mice of 8-10 weeks are prepared at the same time, and the male mice are combined with adult female mice 1d before ovulation to prepare pseudopregnant receptor female mice.
Cas9-RNA obtained by in vitro transcription and sgRNA are mixed, after the concentration is adjusted, the RNA mixture is injected into the male pronucleus and cytoplasm of fertilized eggs of SD rats by a microinjection method (TE2000U microinjector), and then the fertilized eggs are implanted into bodies of pseudopregnant SD rats.
(4) Genotype identification of F0 generation PLS3 mutant rats
Taking F0 newborn suckling mouse toes from 7-14d, extracting with genome DNA extraction kit (

Genomic DNA Kit EE101-12, Beijing Pan-Kagaku Co.) was extracted from rat Genomic DNA. Based on the detection of WT primers for the wild-type gene and KO primers for the mutant-type gene (Table 3) designed based on the sequence information, the target gene was amplified by PCR, the amplified products were separated by electrophoresis on 1.0% agarose gel, and the specific information of the knock-out fragment was determined by gene sequencing.

(5) F1 generation PLS3 mutant rat breeding and genotype identification
The 6F 0 generation mutant rats are cultured according to 6 strains respectively, and the rats without Specific Pathogen (SPF) are raised in the barrier environment of experimental animals under the condition of 12h light/12 h dark light cycle, the temperature is 20-25 ℃, the relative humidity is 50-65%, and the supply of sufficient feed and water is ensured. And breeding the female F0 generation mutant rats and the male F0 generation mutant rats to wild type SD rats in a ratio of 1:1 and a ratio of 1:1 or 1:2 when the female rats and the male F0 generation mutant rats are bred to 2-3 months of age, wherein female rats of male F0 mutant rat offspring are heterozygous mutations of the PLS3 gene, and male rats are wild type.
In the offspring of female F0 generation mutant rats, F1 generation female rats have two genotypes of wild type and heterozygous mutant, and F1 generation male rats have two genotypes of hemizygous mutant and wild type. The above F1 generation was cultured for 2-3 weeks, rat genome was extracted as described above, PCR-amplified with KO primer, and then the genotype of F1 generation rat was identified by gel electrophoresis.
2. Results of the experiment
The results of amplification using KO primers for mutant genes are shown in FIG. 3, and show that the edited types of 6 target genes were co-detected, wherein the female mouse heterozygous for the F0 generation PLS3mutation (the genotype is PLS 3)+/-) The male mouse is numbered 12#, 13#, 26#, F0 generation PLS3 hemizygous mutant (the genotype is PLS 3)-/0) No.4, No.7 and No. 22, 6 positive F0 generation rats were obtained, of which 3 female rats (genotype Pls 3)+/-12, 13, 26) and 3 male mice (genotype Pls 3)-/0And the numbers are 4, 7 and 22), the sizes of the first mouse knockout fragments are shown in a table 4. The results of amplification using KO primers for mutant genes are shown in FIG. 4, in which female mouse progeny No. 26, No. 1-13F 0, female mouse progeny No. 13, No. 14-23F 0, and genotype identification results show male hemizygous mice No.1, 9, and 14F 1 (the genotype is PLS 3)-/0) The numbers 3, 5, 13, 15 and 21 are heterozygous female mice (the genotype is PLS 3)+/-) The results show the successful construction of a PLS3 gene targeted mutation rat model.

Claims (10)

1. A construction method of a novel PLS3 gene knockout rat animal model is characterized by comprising the following steps:
(1) carrying out targeted knockout on a specific fragment of the PLS3 gene in a cell to prepare a PLS3 gene mutant cell;
(2) and (3) constructing a novel PLS3 gene knockout animal model by using the PLS3 gene mutation cell obtained in the step (1).
2. The method of constructing a recombinant vector according to claim 1, wherein the specific fragment in step (1) is an E10-E16 fragment of PLS3 gene.
3. The method of claim 1, wherein the animal in step (2) is a mammal, preferably a rat.
4. The method of construction according to claim 1, comprising a knock-out of a specific fragment of PLS3 gene in a cell.
5. Construction method according to claim 4, characterized in that the CRISPR/Cas9 gene editing system is adopted in the knockout method, and the target mutation of PLS3 gene is realized by introducing sgRNA and Cas9 into a receptor cell, preferably the receptor cell is a fertilized egg.
6. The construction method according to claim 5, characterized in that the sgRNA expression vector includes sgRNA1, sgRNA2, sgRNA3 and/or sgRNA 4;
preferably, the sequence of the sgRNA1 is shown as SEQ ID No. 5-6, the target sequence is shown as SEQ ID No.1, the sequence of the sgRNA2 is shown as SEQ ID No. 7-8, the target sequence is shown as SEQ ID No.2, the sequence of the sgRNA3 is shown as SEQ ID No. 9-10, the target sequence is shown as SEQ ID No.3, the sequence of the sgRNA4 is shown as SEQ ID No. 11-12, and the target sequence is shown as SEQ ID No. 4.
7. The construction method according to claim 5 or 6, characterized in that the sgRNAs comprise sgRNAs and sgRNA-associated biomaterials.
8. The construction method according to claim 7, wherein the sgRNA-associated biomaterial is selected from any one of the following (1) to (7):
(1) a nucleic acid molecule encoding the sgRNA;
(2) an expression cassette comprising the nucleic acid molecule of (1);
(3) a recombinant vector comprising the nucleic acid molecule of (1), or a recombinant vector comprising the expression cassette of (2);
(4) a recombinant microorganism containing (1) the nucleic acid molecule, or a recombinant microorganism containing (2) the expression cassette, or a recombinant microorganism containing (3) the recombinant vector;
(5) a transgenic animal cell line containing the nucleic acid molecule of (1) or a transgenic animal cell line containing the expression cassette of (2);
(6) a transgenic animal tissue containing the nucleic acid molecule of (1) or a transgenic animal tissue containing the expression cassette of (2);
(7) a transgenic animal organ containing the nucleic acid molecule of (1) or a transgenic animal organ containing the expression cassette of (2).
9. The use of the production method according to claim 1 in any one of the following (1) to (3):
(1) use in the study of osteoporosis and/or other bone diseases;
(2) the application in the research of tumor diseases;
(3) use in the study of spinal muscular atrophy and/or other motor neuron diseases.
10. The sgRNA or sgRNA-related biomaterial of any one of claims 5 to 8, for any one of the following (1) to (7):
(1) the application of the mutant in constructing PLS3 gene cells;
(2) the use thereof for the preparation of a cell product for the construction of a PLS3 gene mutation;
(3) the application in preparing PLS3 gene mutation animals;
(4) the use thereof for the preparation of an animal product for the construction of a PLS3 gene mutation;
(5) use in the study of osteoporosis and/or other bone diseases;
(6) the application in the research of tumor diseases;
(7) use in the study of spinal muscular atrophy and/or other motor neuron diseases.
