Abstract
本发明提供了一种仿生细胞膜多聚囊泡纳米材料及其制备方法和应用,属于纳米医学技术领域。本发明所述囊泡包括亲水性外壳和疏水性内核,所述亲水性外壳连接在疏水性内核的两端,二者形成类似于磷脂双分子层结构;其中,所述疏水性内核由金纳米棒AuNRs表面的十六烷基三甲基溴化铵与吲哚菁绿配位后,再与PCL片段共同组成,所述亲水性外壳由PEG片段组成,所述AuNRs呈垂直状态竖立在囊泡外层。本发明提供的仿生细胞膜多聚囊泡结构能够促进囊泡内的纳米药物释放和随后的细胞质内移位,增强了PTT/PDT联合治疗效果;并能屏蔽血液循环中蛋白质的吸附,避免蛋白质冠的产生,以利于纳米药物在肿瘤部位更好地蓄积,减少脱靶效应。

技术领域
本发明属于肿瘤医学技术领域,涉及一种囊泡纳米材料,具体涉及一种仿生细胞膜多聚囊泡纳米材料及其制备方法和应用。
背景技术
在过去的二十年中,基于光学的纳米医学在精准诊断、成像、药物递送、治疗等生物医学应用的各个方面被广泛研究。光热治疗(PTT)和光动力治疗(PDT),因具有无创、高效、有效的肿瘤内蓄积、出色的组织渗透性和更好的患者依从性等优良特性,非常有希望成为传统治疗方法和手术的替代治疗方法。光热治疗依赖于受近红外光(NIR)辐照后,肿瘤细胞内的光热转导剂(PTA)产生的热。光热转导剂可以从外界光源获得能量,并将其转化为热能,以提高肿瘤微环境的温度从而引起肿瘤细胞的死亡。光动力治疗依赖于响应一定波长光的光敏剂和分子氧,光敏剂通过将光能传递至周围的氧分子,从而生成活性氧(ROS),例如单线态氧(SO),以杀死肿瘤细胞。
目前,许多关于纳米材料的治疗研究已从单一治疗方法转向了联合光热及光动力的治疗方案,并且业已证实联合方案治疗肿瘤的重要优势。迄今为止,大部分联合治疗方案需要采用两种波长光分别激发光热和光动力治疗,这大大增加了临床操作的难度并限制了其临床应用。另外,联合治疗面临的另一个问题是低的光转化率。因此,选择合适的纳米材料以最大化联合治疗效果,是肿瘤治疗的关键步骤。
在金属纳米材料中,金纳米材料由于其独特的性能而备受关注,适合于标记、传递、加热和传感等多种应用。相比于其他类型的金纳米材料,金纳米棒(AuNRs)由于其重要的理化特性而被证实为有效的光热转导剂,它们可以强烈地吸收和散射波长为650-900nm的近红外辐射光,并具有合适的纵横比(长除以宽)。由于AuNRs可以产生长波长的辐射光,具有很强的组织穿透力,因此适用于治疗具有复杂周围组织的深部肿瘤。然而,高剂量的纳米材料,或者说,从其表面脱离的表面活性剂具有“毒性”。因此,最佳使用剂量和更好的AuNRs纯化方法对于最大程度降低治疗过程中的毒副作用非常重要。目前迫切需要开发一种有效策略,以最大程度地提高光热治疗作用,并最小化其临床转化导致的毒副作用。
同时,作为常用的多功能光敏剂,吲哚菁绿(ICG)不仅可以产生光热效应,而且还可以通过高强度的NIR辐射(波长>750nm)激发,将氧气转化为ROS,以达到不可逆转地破坏肿瘤细胞和组织的目的。尽管ICG克服了大部分光敏剂的明显缺陷,例如在正常组织中积累,特别是在皮肤和眼结膜上,以及因日光造成的严重副作用,例如长期的皮肤光过敏,但是热或光导致的ICG降解会引起其在水溶液中的不稳定。联合应用有机和无机的光疗材料可以弥补各自的缺点,并利用其优势实现协同治疗的效果。早期研究发现,将ICG与作为传递载体的金纳米材料联合,可以增强ICG的光稳定性,提高其光破坏性的效果。然而,大多数与ICG或其他光敏剂联合使用的金纳米材料都是亲水性的,而ICG在水性介质中容易降解,从而导致其吸收率和荧光的同时丧失。另一缺陷是,ICG易与人血清白蛋白非特异性结合,导致其在肝脏中被快速清除。故而关键挑战仍然在于合理设计金纳米材料与ICG的结合,以促进协同治疗效果。
前列腺癌是最常见的癌症之一,也是美国男性癌症死亡的第二大主要病因。目前的常规治疗方法虽然在治疗开始时有效,但随着疾病进展往往失效。目前的临床试验显示,对于那些PSMA阳性的晚期前列腺癌,放射性核素标记的PSMA抗体(177Lu-PSMA)可能是一种有前途的治疗方法。但是,对于那些PSMA阴性的雄激素非依赖性晚期前列腺癌,需要探索其他有效的治疗方法。
能否提供一种金纳米棒与ICG结合的纳米材料,以实现增强PTT/PDT协同治疗效果,并能很好用于PSMA阴性的雄激素非依赖性晚期前列腺癌的治疗,成为一种亟待解决的技术问题。
发明内容
本发明的目的就是为了解决上述技术问题,而提供一种仿生细胞膜多聚囊泡纳米材料及其制备方法和应用。本发明提供的仿生细胞膜多聚囊泡结构能够促进囊泡内的纳米药物释放和随后的细胞质内移位,增强了PTT/PDT联合治疗效果;并能屏蔽血液循环中蛋白质的吸附,避免蛋白质冠的产生,以利于纳米药物在肿瘤部位更好地蓄积,减少脱靶效应。
本发明的目的之一是提供一种仿生细胞膜多聚囊泡纳米材料,所述囊泡包括亲水性外壳和疏水性内核,所述亲水性外壳连接在疏水性内核的两端,二者形成类似于磷脂双分子层结构;其中,所述疏水性内核由金纳米棒AuNRs表面的十六烷基三甲基溴化铵与吲哚菁绿配位后,再与PCL片段共同组成,所述亲水性外壳由PEG片段组成,所述AuNRs呈垂直状态竖立在囊泡外层。
本发明提供的上述囊泡结构,其中AuNRs表面的CTAB用于与PEG-PCL和ICG的共聚物配位以制备囊泡。囊泡是通过亲水性和疏水性的作用形成的,类似于细胞膜的磷脂结构。因此,疏水性组分被完全包裹在囊泡内部,以改善AuNRs和ICG在血液中的生物相容性,并减少纳米组分的疏水性对非靶器官的损害。其中AuNR呈垂直状态竖立在囊泡外层。囊泡的形状可根据周围空间而变化,这便于纳米药物的体内运输。
本发明通过AuNRs表面的CTAB与PEG-PCL和ICG的共聚物配位,成功构建了包载AuNRs和ICG的仿生细胞膜聚合物囊泡,从而实现通过一个波长的NIR激发发挥纳米药物光热/光动力协同治疗前列腺癌的构想。经试验证明,这种纳米平台表现出良好的细胞摄取,高效的肿瘤聚集,更高的光热转化效率和优异的抗光漂白性,有利于进行基于ICG的荧光肿瘤成像。由于使用相同波长的近红外光激发,采用本发明的囊泡用于肿瘤治疗时可以降低皮肤光损伤副作用的发生率。PC3肿瘤细胞作为一种雄激素非依赖性晚期前列腺癌的细胞系,具有PSMA表达阴性的特征(PSMA,前列腺特异性膜抗原),本发明的囊泡在NIR照射下可通过产生ROS破坏其胞质溶酶体膜的完整性来促进PC3肿瘤细胞发生凋亡,达到杀灭肿瘤的目的。体外及体内实验均充分证实了近红外光照射下,自组装的囊泡对PC3肿瘤的PTT/PDT联合治疗作用显著,这提示了该纳米平台对晚期前列腺癌治疗的临床应用价值,尤其是作为针对PSMA阴性去势抵抗前列腺癌的治疗补充策略,具有实践意义。
进一步的是,所述囊泡的内径为100~200nm,外径为200~300nm。
本发明的目的之二是提供上述仿生细胞膜多聚囊泡的制备方法,所述制备方法为:将吲哚菁绿、AuNRs和PEG-PCL共聚物溶于有机溶剂中,然后在超声作用下将所得物分散到蒸馏水中,透析24小时后获得囊泡的溶液,记为AuNR/ICG。
进一步的是,所述吲哚菁绿、AuNRs和PEG-PCL共聚物的质量比为1:0.2:4。

进一步的是,所述有机溶剂包括二甲基甲酰胺或四氢呋喃。
进一步的是,所述有机溶剂的用量占AuNRs的质量体积比为1mg AuNRs:3mL有机溶剂。
本发明利用AuNRs疏水性表面的十六烷基三甲基溴化铵(CTAB)强配位能力的优势,通过CTAB与ICG配位,并和PCL片段一起形成囊泡的疏水层,而亲水性PEG片段连接在疏水性片段的两端,利用其亲疏水性自组装形成类似于磷脂双分子层结构的聚合物囊泡,从而赋予它们极好的化学稳定性和载药能力。囊泡内部的疏水层能够使光敏剂积聚,从而增强PTT/PDT协同治疗效果。而PEG段外层因亲水性可屏蔽血液循环中蛋白质的吸附,避免蛋白质冠的产生,以利于纳米药物在肿瘤部位更好地蓄积,减少脱靶效应。
本发明的目的之三是提供上述仿生细胞膜多聚囊泡的应用,所述应用包括将该囊泡用于制备纳米材料应用于前列腺癌的治疗中,特别是应用于晚期前列腺癌的治疗。
进一步的是,所述应用还包括将该囊泡应用于荧光肿瘤成像。
早期研究显示,基因治疗或其他组合治疗策略可抑制人源性前列腺癌细胞株PC3的增殖,该细胞来源于PSMA阴性的雄激素非依赖性晚期前列腺癌患者。我们目前的研究结果表明,在近红外光辐照下,构建的仿细胞膜聚合物囊泡纳米药物可以使包载的AuNRs和ICG产生较游离态更明显的光热效应,从而对PC3肿瘤细胞造成更大的杀伤。有趣的是,由ICG产生的ROS不仅产生PDT功效,而且还能引起溶酶体破裂,进一步促进囊泡内的纳米药物释放和随后的细胞质内移位,从而增强了PTT/PDT联合治疗效果。另外,自组装AuNR/ICG囊泡具有优异的抗光漂白性能、良好的细胞摄取能力和有效的肿瘤聚集特性,这些特性有利于ICG的荧光肿瘤成像,以及增强PTT和PDT的协同治疗效果达到肿瘤消融的目的。
本发明的有益效果如下:
(1)本发明提供了一种新型的包载AuNRs和ICG的仿生细胞膜聚合物囊泡结构,表现出良好的细胞摄取,高效的肿瘤聚集,更高的光热转化效率和优异的抗光漂白性,有利于进行基于ICG的荧光肿瘤成像;
(2)由于使用相同波长的近红外光激发,本发明提供的AuNR/ICG囊泡可以降低皮肤光损伤副作用的发生率;
(3)本发明提供的AuNR/ICG囊泡在NIR照射下可通过产生ROS破坏其胞质溶酶体膜的完整性来促进PC3肿瘤细胞发生凋亡,达到杀灭肿瘤的目的。自组装AuNR/ICG囊泡对PC3肿瘤的PTT/PDT联合治疗作用显著。

具体实施方式
为了使本发明的目的、技术方案及优点更加清楚明白,以下结合实施例对本发明进行具体描述,有必要指出的是,以下实施例仅仅用于对本发明进行解释和说明,并不用于限定本发明。本领域技术人员根据上述发明内容所做出的一些非本质的改进和调整,仍属于本发明的保护范围。
实施例1
AuNR/ICG囊泡的合成:
AuNRs购自江苏XFNANO材料。将1mg ICG,0.2mg AuNRs和4mg PEG114PCL60混合在0.6mL二甲基甲酰胺(DMF)中,然后在超声作用下将溶剂分散到5mL蒸馏水中,透析24小时后获得AuNR/ICG囊泡的溶液。
以游离AuNR/ICG溶解在0.5%DMF中作为对照。
上述AuNR/ICG囊泡的合成示意图如图1(a)所示,该囊泡通过PTT/PDT协同用于增强前列腺癌治疗的示意图如图1(b)所示。
测试例1
实验方法如下:
(一)AuNR/ICG囊泡的表征
通过透射电子显微镜(TEM)成像(Hitachi H-600)观察样品的形态,使用酶标仪(Multiskan GO)测量吸收光谱。
(二)化学稳定性和光稳定性
将AuNR/ICG囊泡分散在不同pH值的液体中以检测其化学稳定性,包括pH 5.0缓冲液,pH 7.4缓冲液,培养基和血清。然后在0、2、4、6、24和48h时测量其吸收光谱。用785nmNIR照射AuNR/ICG囊泡和游离AuNR/ICG纳米药物0、0.5、1、2、3、5和10min后评估其光稳定性,照射后的样品用酶标仪进行吸光度测量判定。
(三)SO的监控
通过使用1,3-二苯基异苯并呋喃(DPBF)作为探针测量SO。将浓度为1.0μg·mL-1的ICG样品与3mM DPBF混合,然后照射10分钟(785nm,1.0W·cm-2)。在照射期间监测DPBF在415nm的吸光度。
(四)溶酶体膜破裂检测
将PC3细胞(5.0×104/孔)接种在24孔板上,在含有10%FBS的RPMI 1640培养基中孵育过夜。将浓度为0、1.0、2.0和5.0(μg·mL-1)的AuNR/ICG囊泡加入平板各孔中。孵育6小时后,以785nm激光在1.0W·cm-2下照射细胞3分钟。再孵育1小时后,将细胞用吖啶橙(AO)(20μg·mL-1,0.5mL)染色0.5h。使用荧光显微镜对细胞进行成像。
(五)二氢乙啶(DHE)染色
将PC3细胞(5.0×104/孔)接种在24孔板上,在含有10%FBS的RPMI 1640培养基中孵育过夜。将浓度为0、1.0、2.0和5.0μg·mL-1的AuNR/ICG囊泡加入平板各孔中。孵育6小时后,以785nm激光在1.0W·cm-2下照射细胞3分钟。然后将细胞用DHE(5μM,0.5mL)染色0.5小时。使用荧光显微镜对细胞进行成像。
(六)光热转换效率
将AuNR/ICG囊泡和游离的AuNR/ICG置于0.5mL玻璃小瓶中,用785nm的1.0W·cm-2激光照射以监测光热转换效率。当温度达到其平稳状态时,将激光器移开以冷却至室温。在整个过程中记录样品的温度。计算描述如下:
其中,η是光热转换效率,ΔTmax是温度最大变化,Q是溶剂的每单位时间的热量增加,I是激光功率,Aλ代表在785nm波长下囊泡的吸收率。hs可以通过以下公式计算:
在照射下DPBF在415nm处的归一化吸光度如图2(g)所示;游离AuNR/ICG的光热转化效率如图2(h)所示;AuNR/ICG囊泡(785nm,1.0W·cm-2)的光热转化效率如图2(i)所示。
(七)细胞摄取
将PC3肿瘤细胞(1.0×106/孔)接种在6孔板上,在含有10%胎牛血清(FBS)的RPMI1640培养基中孵育过夜。将AuNR/ICG囊泡和游离的AuNR/ICG加入板中。孵育24小时后,计数细胞并超声处理。随后,通过DMF提取细胞中的ICG,用流式细胞仪测定ICG的浓度。
(八)体外细胞毒性
将PC3细胞(5.0×103/孔)接种在96孔板上,在含有10%FBS的RPMI 1640培养基中孵育过夜。将AuNR/ICG囊泡和游离的AuNR/ICG以ICG浓度为1、2、5、10、25和50μg·mL-1添加到平板中,然后以785nm波长的NIR以1.0W·cm-2照射平板3分钟。用3-(4,5-二甲基噻唑-2-基)-2,5-二苯基溴化四唑(MTT)测定法检测细胞活力。
(九)凋亡测定
将PC3细胞(5.0×104/孔)接种在24孔板上,在含有10%FBS的RPMI 1640培养基中孵育过夜。将AuNR/ICG囊泡,游离AuNR/ICG和PBS加入板中。孵育24小时后,以785nm波长的NIR以1.0W·cm-2照射平板3分钟。24小时后使用AnnexinV-FITC/PI细胞凋亡检测试剂盒对细胞染色,并用流式细胞仪进行测定。
(十)体内近红外荧光(NIRF)成像和生物分布
通过将PC3细胞(5.0×106/只裸鼠)皮下注射到侧面,构建荷PC3移植瘤的雄性Balb/c裸鼠。然后尾静脉注射AuNR/ICG囊泡和游离AuNR/ICG,剂量为含7.5mg·kg-1的ICG。使用IVIS Lumina II在24、48和72h在745nm NIR激发下对裸鼠成像。为了了解纳米材料在裸鼠体内的生物学分布,纳米药物注射24小时后提取裸鼠的各个脏器,包括心脏,肝脏,脾脏,肺,肾和肿瘤,进行IVIS Lumina II成像。
(十一)体内红外热像仪
将AuNR/ICG囊泡和游离AuNR/ICG纳米药物分别以含5.0、7.5和10.0mg·kg-1的ICG剂量,尾静脉注射至荷PC3移植瘤的裸鼠体内。以785nm波长的NIR以1.0W·cm-2照射不同组裸鼠的移植瘤5分钟。用红外照相机监测肿瘤区域的温度。
(十二)体内抗癌功效
分别给各组荷瘤裸鼠尾静脉注射PBS、游离AuNR/ICG,ICG囊泡和AuNR/ICG囊泡,剂量为7.5mg·kg-1ICG。注射24小时后,以785nm波长的NIR以1.0W·cm-2照射移植瘤3分钟。然后每两天用游标卡尺测量肿瘤的长度和宽度,计算并记录移植瘤体积。注射24天后,安乐死处死裸鼠并提取肿瘤组织,拍照并进行后续组织水平相关实验。
(十三)免疫组织化学
分别给各组荷瘤裸鼠尾静脉注射PBS、游离AuNR/ICG,ICG囊泡和AuNR/ICG囊泡,剂量为含有7.5mg·kg-1的ICG。注射24小时后,用785nmNIR在1.0W·cm-2下照射肿瘤3min。注射24天后,安乐死裸鼠并取出肿瘤组织,固定于福尔马林液体中24小时,然后将肿瘤组织切片,于封闭液中孵育20分钟后,与Caspase3和Ki-67抗体在4℃孵育过夜。洗涤载玻片,并在室温下与二抗一起孵育。载玻片用DAB溶液染色并用荧光显微镜观察。

具体结果分析为:
制备和表征
AuNRs表面的CTAB用于与PEG-PCL和ICG的共聚物配位以制备囊泡(图4a)。囊泡是通过亲水性和疏水性的作用形成的,类似于细胞膜的磷脂结构。因此,疏水性组分被完全包裹在囊泡内部,以改善AuNRs和ICG在血液中的生物相容性,并减少纳米组分的疏水性对非靶器官的损害。囊泡中ICG和AuNRs的载药量分别为16.13%和3.2%。透射电镜TEM图像显示,AuNR的长径和短径分别为80.46±8.67和25.17±0.86nm(图4b);AuNR/ICG囊泡的内径为148.71±6.21nm,外径为238.46±32.95nm(图4c)。由于聚合物的掩蔽效应和放大后拍摄的TEM图像像素的减少,在图4c中不容易看到AuNRs。因此,我们还以1μm的比例拍摄了囊泡图像,如图2所示。图像所示,箭头指示了AuNR的部分位置,呈垂直状态竖立在囊泡外层。囊泡的形状可根据周围空间而变化,这便于纳米药物的体内运输。
进一步实验证实,在不同pH值的水溶液中,特别是在血清中,AuNR/ICG囊泡的化学稳定性优于游离态(图4d,n=3)。这表明该纳米平台对之后的体内实验具备足够的稳定性。在785nmNIR激发3,5和10min时检测AuNR/ICG囊泡的吸光度值,我们发现该纳米平台组的吸光度值要明显高于游离ICG组(图4e和图4f),说明ICG经囊泡组装后其光稳定性显著提高。因此,经囊泡组装可以延长纳米药物被激发后的PTT和PDT治疗效果,这可能是体内实验显示该纳米平台可有效抗肿瘤的机制之一。为了最大程度减少NIR的副作用,在之后的实验中我们将785nmNIR的激发时间设置为3分钟。
ROS的产生和光热效应
ROS的产生可以通过使用1,3-二苯基异苯并呋喃(DPBF)作为指示剂的化学测定法检测,表现为410nm处的明显吸收带。DPBF可与ROS发生不可逆反应,从而被漂白,因此通过检测DPBF吸光度值的降低可实时监测ROS的产生。DPBF实验证实了AuNR/ICG纳米囊泡经785nmNIR激发后,比游离AuNR/ICG可产生更多的ROS(图4g),并且使光热转化效率从游离态的15.6%增加到18%(图4h和4i)。这说明,这种简单、方便且稳定的纳米自组装技术可以有效地提高AuNR/ICG纳米药物的光动力转化和光热转化效率。
细胞摄取和细胞活力
体外细胞摄取实验(n=3)显示,PC3细胞对AuNR/ICG囊泡组的摄取率高于AuNR/ICG游离组(图5a),其原因在于PEG-PCL的包封和自组装形成亲水性囊泡。
MTT实验(n=4)显示,在没有NIR激发的状态下,25μg·mL-1的AuNR/ICG囊泡对PC3肿瘤细胞具有暗毒性,浓度越高,细胞活力越低,呈浓度依赖性(图5b)。游离的AuNR/ICG纳米材料在50μg·mL-1的浓度下具有暗毒性(图5c)。究其原因,可能是由于PEG-PCL的包载使PC3细胞对AuNRs和ICG的摄取率增加,从而降低了细胞活力。在3分钟785nmNIR激发下,仅5μg·mL-1的浓度,AuNR/ICG囊泡组便可使PC3细胞活力下降,并呈浓度依赖性,而游离AuNR/ICG纳米材料组浓度须在25μg·mL-1以上才会引起PC3细胞活力下降(图5b和图5c)。该测定重复三次,每次三复孔,得到相似的结果。这说明,PEG-PCL包封后,可增加纳米药物的细胞摄取率及ICG的光稳定性,从而增加了肿瘤细胞内ROS的生成、增强了对肿瘤细胞的损伤,改善了光热转换效率,从而表现为肿瘤细胞活力的下降。
细胞凋亡,溶酶体膜破坏和DHE染色
通过流式细胞仪检测PC3细胞的凋亡,以进一步了解AuNR/ICG囊泡对PC3细胞的杀伤机制(图5d-f,n=3)。该测定重复三次,每次三复孔,得到相似的结果。根据膜联蛋白V的表达差异可以判定,首先,PBS/照射组的肿瘤细胞发生凋亡的百分率很低,可认为785nmNIR照射3min对细胞没有造成损伤;其次,AuNR/ICG囊泡处理后PC3细胞经785nmNIR照射3min,发生凋亡的比例为14.27%±1.42%,显著高于在游离组(10.70%±0.61%,P<0.0001)和PBS组(4.7%±0.56%,P<0.0001)。细胞凋亡包括早期凋亡与晚期凋亡。经分析,这种显著差异(P<0.0001)主要是由于AuNR/ICG囊泡组的PC3肿瘤细胞发生了明显的晚期凋亡,各组的百分比分别为AuNR/ICG囊泡组12.57%±0.81%,游离AuNR/ICG组9.03%±0.91%,和PBS组4.2%±0.62%。各组处于早期凋亡状态的肿瘤细胞百分比分别为,AuNR/ICG囊泡组1.7%±0.62%,游离AuNR/ICG组1.7%±0.31%,亦显著高于PBS组0.17%±0.12%)(P<0.05)。在这三个不同的治疗组中,坏死状态的PC3肿瘤细胞百分比没有太大差异(图5g)。由此说明,AuNR/ICG囊泡在785nmNIR激发下,可促使更多的PC3肿瘤细胞发生凋亡,主要是诱导细胞进入晚期凋亡状态。
许多情况会导致细胞发生凋亡。溶酶体膜通透性增加可导致溶酶体酶释放,从而触发线粒体上游的细胞凋亡级联反应,这是促进细胞凋亡的重要原因。吖啶橙染色(AO染色)可测定PC3细胞溶酶体膜的完整性。完整的溶酶体膜可呈现绿色荧光核和周围的红色荧光细胞质,若溶酶体膜破裂则仅显示绿色荧光核。AO染色实验显示,尽管AuNR/ICG囊泡的浓度增加至5.0μg·mL-1,在没有被光激发的情况下,PC3细胞的溶酶体膜保持完整,显示红色和绿色荧光;一旦被NIR辐照3分钟,仅1.0μg·mL-1的AuNR/ICG囊泡也可使PC3细胞的溶酶体膜发生破裂,仅显示绿色荧光(图6a,n=3)。
细胞溶酶体膜的完整性与胞内ROS的积累有关。胞内ROS积累越多,溶酶体膜越容易被破坏。我们通过细胞二氢乙啶(DHE)染色试验(n=3)检测肿瘤细胞内ROS的生成,可见随着AuNR/ICG囊泡浓度的增加,被NIR激发后PC3细胞的红色荧光变得越来越明显,这说明胞内ROS的产生呈浓度依赖性(图6b),与AO染色结果一致,由此确定了AuNR/ICG囊泡在NIR激发后引起PC3肿瘤细胞溶酶体膜破坏的原因。
NIRF成像,生物分布和红外热成像
通过PC3荷瘤裸鼠模型来验证AuNR/ICG囊泡在体内的治疗效果,首先通过小动物无创NIR荧光(NIRF)活体成像来评估AuNR/ICG囊泡的体内分布(n=3)。通过NIRF成像显示,经尾静脉给予AuNR/ICG囊泡24小时后,裸鼠体内肿瘤部位的荧光强度最高。而游离AuNR/ICG组的体内荧光分布非常分散。不管是囊泡组还是游离组,荧光强度都会随着时间延长而衰减,但是相对于游离AuNR/ICG组,AuNR/ICG囊泡组在裸鼠肿瘤部位的滞留时间明显延长(图7a和图7b)。
进一步检测各个器官中的荧光强度(n=3),可见游离组的荧光强度在肝脏组织最高,说明AuNR/ICG囊泡具有更好的肿瘤靶向作用(图7c和图7d),可见AuNR/ICG囊泡在体内达到静态平衡时,主要集中在肿瘤部位,这是由于增强的渗透和保留效应(EPR)所致。尽管荧光强度会随时间延长持续减弱,但在给药的早期(≤24小时)进行定位成像,可作为肿瘤跟踪成像剂用于体内定位肿瘤。
通过红外热成像实验验证AuNR/ICG囊泡的体内光热转换效率(n=3)。实验表明,随着注射浓度的增加,给予AuNR/ICG囊泡的裸鼠肿瘤部位的温度迅速上升,呈浓度依赖性。当注射浓度为5.0mg·kg-1时,肿瘤部位在100s时可达到峰值温度,并维持在10℃;当注射浓度为7.5mg·kg-1时,肿瘤部位仍在100s时达到峰值温度,但温度可保持在15℃至16℃之间。随着注射浓度的增加,肿瘤部位峰值温度越高。但注射浓度为10.0mg·kg-1时,NIR激发后肿瘤部位温度迅速升高,25s内即可超过16℃,然后缓慢升至17℃至18℃之间,并保持300s以上(图8a和图8b)。然而,游离AuNR/ICG组并未显示相似的时间-肿瘤部位温度曲线,相同剂量下游离组的肿瘤部位达到峰值温度的时间明显延迟,且峰值温度低于相同剂量囊泡组的一半,高峰维持时间也大大缩短(图8c和图8d)。这说明,滞留在肿瘤部位的AuNR/ICG囊泡具有很强的加热能力,考虑与其良好的肿瘤靶向性和较高的光热转化效率有关。以上结果与体外实验结果基本一致。
抗肿瘤功效和免疫组化
为了验证AuNR/ICG囊泡的PTT和PDT联合治疗对PC3异种移植瘤的协同杀伤作用,我们构建裸鼠PC3异种移植瘤模型,并设立AuNR/ICG囊泡组和不同类型的对照组,包括单个ICG囊泡组、游离AuNR/ICG组和PBS组,比较疗效差异。经尾静脉给荷瘤裸鼠注射7.5mg·kg-1的纳米药物,给药24小时后,四组不同处理组裸鼠均接受785nmNIR辐照3分钟,将未接受NIR辐照的各组裸鼠作为各自的对照组。于指定的时间点连续记录肿瘤的生长。以给药日的移植瘤体积作为参考,设为1,以每次记录的移植瘤体积与其相比的值作图,绘制成时间-相对体积曲线(图9a,n=5)。分析该曲线,可见NIR激发后的ICG囊泡组和游离AuNR/ICG组,以及没有被激发的AuNR/ICG囊泡组在实验后期均显示一定的抑制肿瘤生长的作用。这说明了AuNRs和单个ICG的治疗作用,以及AuNR/ICG囊泡的暗毒性。令人兴奋的是,AuNR/ICG囊泡组经NIR激发后,表现出显著的肿瘤杀伤作用,给药后第三天肿瘤体积即明显减小,第五天肿瘤完全消融。虽然5只裸鼠中有2只在第12天出现肿瘤复发,但是直到实验结束(给药后第24天),肿瘤的体积均明显小于其他各组。至实验结束,经NIR激发的AuNR/ICG囊泡组另外3只裸鼠均保持无肿瘤状态,肿瘤治愈率为60%。因此,AuNR/ICG囊泡经NIR激发后其PTT和PDT的协同作用比单独ICG囊泡或游离AuNR/ICG纳米材料经NIR激发后明显增强,具有更好的协同治疗效果。究其原因,在于ICG结合了AuNRs和经PEG-PCL的封装可消除ICG的光漂白(图9b);其次,该纳米平台的EPR效应提高了肿瘤的靶向性摄取,因此,囊泡结构和两种纳米药物的结合方式增强了AuNR/ICG纳米材料的PTT和PDT的协同治疗作用。
为了进一步证实NIR激发的AuNR/ICG囊泡对肿瘤细胞的杀伤作用,我们在组织水平比较不同处理组肿瘤组织的增殖和凋亡情况(n=3),通过IHC检测肿瘤组织切片中Ki-67和Caspase3蛋白的表达水平。经NIR辐照的PBS组、ICG囊泡组和游离AuNR/ICG组切片均显示了形态异常的PC3肿瘤细胞排列紧密,布满整个视野;而经NIR辐照的AuNR/ICG囊泡组的切片中肿瘤细胞明显减少,仅在切片中央可发现数个被成纤维细胞包围的PC3移植瘤细胞。在蓝染的细胞核中,Ki-67阳性的细胞核呈棕色。成纤维细胞核也可被非特异性地染上棕色(图9c)。排除成纤维细胞核的非特异性染色,经NIR辐照的PBS组、ICG囊泡组、游离AuNR/ICG组和AuNR/ICG囊泡组的肿瘤细胞表达Ki-67的阳性百分比分别为40.65%、27.98%、33.42%和4.606%(图9d)。这说明,NIR激发下AuNR/ICG囊泡PTT和PDT的协同杀伤作用几乎完全抑制了PC3肿瘤细胞的增殖。Caspase3阳性表现为细胞质中出现褐色斑点。在NIR激发的AuNR/ICG囊泡组处理的肿瘤组织切片中,可见成纤维细胞中聚集了明显的核固缩的Caspase3阳性的肿瘤细胞;而在其他三组的肿瘤组织切片中仅发现了散在的阳性细胞(图9c)。Caspase3的阳性比率在经NIR辐照的PBS组、ICG囊泡组、游离AuNR/ICG组和AuNR/ICG囊泡组的切片中分别为21.47%,31.13%,21.92%和55.2%(图9e)。这说明,在NIR激发下AuNR/ICG囊泡的PTT和PDT协同杀伤作用可促进细胞凋亡以杀死PC3肿瘤细胞。与体外纳米药物治疗实验相比,在体内试验过程中,AuNR/ICG囊泡在NIR激发下对肿瘤增殖抑制和凋亡促进的作用得到了扩大,这表明AuNR/ICG囊泡在NIR激发下的治疗作用可能会越来越明显。
另外,苏木精和伊红染料(HE)染色是目前评估纳米药物生物安全性检测的常用标准方法,因此我们检查了注射有该纳米平台的裸鼠的重要脏器,包括心脏,肝脏,脾脏,肺脏,肾脏,进行HE染色(图10)。组织病理学分析提示没有明显证据表明该纳米平台可引起明显组织损伤。
Claims (9)
1.一种仿生细胞膜多聚囊泡,其特征在于,所述囊泡包括亲水性外壳和疏水性内核,所述亲水性外壳连接在疏水性内核的两端,二者形成类似于磷脂双分子层结构;其中,所述疏水性内核由金纳米棒AuNRs表面的十六烷基三甲基溴化铵与吲哚菁绿配位后,再与PCL片段共同组成,所述亲水性外壳由PEG片段组成,所述AuNRs呈垂直状态竖立在囊泡外层。
2.根据权利要求1所述的仿生细胞膜多聚囊泡,其特征在于,所述囊泡的内径为100~200nm,外径为200~300nm。
3.一种如权利要求1所述仿生细胞膜多聚囊泡的制备方法,其特征在于,所述制备方法为:将吲哚菁绿、AuNRs和PEG-PCL共聚物溶于有机溶剂中,然后在超声作用下将所得物分散到蒸馏水中,透析24小时后获得囊泡的溶液,记为AuNR/ICG。
4.根据权利要求3所述的制备方法,其特征在于,所述吲哚菁绿、AuNRs和PEG-PCL共聚物的质量比为1:0.2:4。
5.根据权利要求3所述的制备方法,其特征在于,所述PEG-PCL共聚物为PEG114PCL60。
6.根据权利要求3所述的制备方法,其特征在于,所述有机溶剂包括二甲基甲酰胺或四氢呋喃。
7.根据权利要求3所述的的制备方法,其特征在于,所述有机溶剂的用量占AuNRs的质量体积比为1mgAuNRs:3mL有机溶剂。
8.一种如权利要求1-2任一项所述的仿生细胞膜多聚囊泡或如权利要求3-7任一项所述方法制备得到的仿生细胞膜多聚囊泡的应用,其特征在于,所述应用包括将该囊泡用于制备纳米材料应用于前列腺癌的治疗中,特别是应用于晚期前列腺癌的治疗。
9.根据权利要求8所述的应用,其特征在于,所述应用还包括将该囊泡应用于荧光肿瘤成像。
