First, bone mineral density experiment
40 SPF female Wistar rats with the weight of about 300g are divided into a control group and an experimental group after being adaptively fed for one week; wherein after the experimental group is fed adaptively, the formula milk powder obtained in examples 1 to 3 and comparative examples 1 to 2 is fed, and the formula milk powder (the formula milk powder given in the specific embodiment in the patent with the publication number of CN 11296577A) is fed to the control group; each group of 10 animals was fed in a single cage, fed freely, dosed with deionized water (to avoid calcium pick-up from drinking), weighed weekly, and the feeding recorded for a 13 week period.
Animals were sacrificed after 13 weeks feeding, one side of the femur was peeled off, baked in an oven at 105 ℃ to constant weight, and the dry weight and length of the bones were weighed; measuring the bone density of the key point of the femur and the distal end of the femur by using a single photon bone densitometer; roasting thighbones on one side of a rat to constant weight, and then digesting the whole body by a wet method; the calcium content was determined by atomic absorption spectrophotometry (GB/T5009.92-2003).

Table 1 shows the effect of the formula milk powder on the body weight of rats, and it can be seen from table 1 that the formula milk powder of examples 1 to 3 of the present invention is selected to significantly increase the body weight of rats compared to the commercially available formula milk powder, and the lack of linolenic acid or N-acetylneuraminic acid has no significant effect on the body weight; table 2 shows the effect of the formula milk powder on the calcium content of the femur and femur of a rat, and it can be seen from table 2 that, compared with the formula of the commercially available milk powder, the milk powder of the present invention selected from the examples 1 to 3 can significantly improve the femur length, the femur weight and the calcium content of the femur of a rat, and the lack of linolenic acid or N-acetylneuraminic acid has no significant effect on the bone development of a mouse, thereby demonstrating that the formula milk powder of the present invention can significantly improve the height and the weight of a rat, and the sufficient calcium content in vivo can meet the requirement of the body development.
Water maze experiment
C57BL/6 mice were purchased 6 weeks old, acclimatized for 12 days, and then subjected to the water maze experiment.
The mouse Morris water maze online detection system consists of a stainless steel plastic-sprayed cylindrical water pool and an image acquisition and analysis system, wherein the diameter of the water pool is 100cm, the height of the water pool is 38cm, and the diameter of a platform is 6cm, and the height of the platform is 14 cm. The pool is divided into 4 quadrants (NE, SE, SW and NW) according to four directions of southeast, west and north, the middle point of the circular arc of the wall of the quadrant pool is an optional animal water inlet point, and the platform can be arranged in the center of any quadrant. The image acquisition and analysis system records animal swimming track data for index extraction and analysis.
Spatial search experiments: the NW quadrant is selected as the entry quadrant, and the spatial memory of the animal is evaluated by the number of times the animal passes through the original platform position within 60s, the original platform quadrant run length ratio and the time ratio (namely the ratio of the run length and time of the animal in the original platform quadrant to the total run length and total time).

Table 3 shows the results of the water maze test on mice, the effect of the milk powder on the mice is significantly different in different groups, the platform crossing times of the mice fed with the formula milk powder of examples 1 to 3 are significantly higher than those of the control group, and the quadrant run rate and time rate of the original platform are significantly higher than those of the control group, the difference is significant, which suggests that the spatial position memory ability of the mice fed with the formula milk powder of examples 1 to 3 is stronger than that of the control group, and in addition, as can be seen from comparative examples 1 and 2, the lack of linolenic acid or/and N-acetylneuraminic acid significantly affects the cognition of the mice.
Third, improving the fat blood of fat mice
The experimental process comprises the following steps: c57BL/6J mice are adaptively fed for about 8d, except for the control group mice which are fed with common feed, other mice are fed with high-fat feed for 3 months; after the model building is successful (the increase of the physical quality is more than 20 percent of that of a control group), mice fed with high-fat feed are randomly divided into an example 1 group, an example 2 group and an example 3 group according to the physical quality, 20 mice are fed with the formula milk powder of the example 1, the example 2 and the example 3 respectively for 2 months, the control group is fed with the same volume of commercial milk powder, the animal body quality and the food intake are monitored every week, and the mental state, the hair color, the diet, the drinking water, the activity and the like of the mice are observed.
And (3) detecting the state of the obese mouse: the results of the tests on the relative body mass growth rate and waist circumference (60d) of each group of mice are shown in table 4. from table 4, it can be seen that the formulas of examples 1 to 3 can significantly reduce the body mass growth rate and waist circumference of the mice relative to the control group.
Serological index detection: at 1d prior to testing, the pad was changed and the mice were fasted overnight. 2d, collecting plasma, standing for 2h at room temperature, centrifuging, subpackaging in a refrigerator at-80 ℃ for storage, and respectively detecting the levels of Triacylglycerol (TG) and Total Cholesterol (TC) in the serum, wherein the results are shown in Table 5, and as can be seen from the table 5, compared with a control group, the levels of TG and TC in the serum of the mice fed by the formula milk powder of the examples 1 to 3 are obviously reduced, which shows that the formula milk powder of the invention can improve the dyslipidemia of obese mice.

The production flow of the formula of example 1, example 2 and example 3 comprises the following steps:
(1) checking and accepting raw materials and auxiliary materials: all the used raw and auxiliary materials come from a supplier qualified by evaluation, products produced by enterprises obtaining production licenses must be selected, and the products have certification, imported raw materials must provide certification materials of entry and exit inspection and quarantine departments, quality departments and inspection departments inspect and accept the raw materials according to the inspection standards of companies, and the raw materials can be put into use after being qualified; all packing materials related to the production of the formula milk powder come from suppliers qualified by evaluation, all packing materials are required to be checked and accepted according to corresponding checking and accepting standards, the packing materials meet the checking and accepting standards of the packing materials specified by companies, and qualified packing materials can be put into use.
(2) Preparing materials: all raw and auxiliary materials are received and prepared according to the requirements of a production plan, and the names, specifications, production dates, batch numbers, quality guarantee periods, quantities, whether the raw and auxiliary materials are qualified, whether the external packages are pollution-free and the like of the raw and auxiliary materials are confirmed.
(3) Removing the outer package: the raw materials are taken off from the outer bag in a feeding workshop, before feeding, whether the variety and the quantity of various raw and auxiliary materials used conform to a production plan list is confirmed again, and the outer bag and the sealing line are stored in a centralized way.
(4) Dedusting and sterilizing the inner bag: after the outer package is removed, the inner package and the appearance of the content of the raw and auxiliary materials are checked to confirm that the inner package of the raw and auxiliary materials used in the production is intact. The inner package needs to be cleaned by a method of wiping 75 +/-1 percent (vol% at 20 ℃) of alcohol on the surface for disinfection;
sterilizing raw materials and auxiliary materials: adopt the tunnel that disinfects to disinfect, tunnel conveyer belt frequency: less than or equal to 30Hz, and the sterilization time is more than or equal to 60 s. Ultraviolet intensity is more than or equal to 70 mu w/cm2
(5) And (3) metering the ingredients: the batching personnel accurately weigh various raw materials and auxiliary materials by using the calibrated electronic scale according to the requirements of the formula table of the product, marks (names and quantities) are made on the various raw materials and the auxiliary materials, the raw materials and the auxiliary materials are conveyed to a goods shelf to be placed in order for the next procedure to use, and meanwhile, the records of batching and weighing are made.
(6) Premixing: pre-stirring calcium carbonate, DHA algae oil powder (7%), linolenic acid powder, N-acetylneuraminic acid (cubilose acid), hydrolyzed yolk powder, lactoferrin, animal bifidobacterium Bb-12, vitamin premix, mineral premix, bovine colostrum powder, lactase and the like. The obtained mixed powder is filled in a big sterilized inner bag, and a mark is made for standby.
(7) Feeding and sieving: checking the mixed powder information and the raw material information according to a production work order and a formula table, removing an inner package after confirming the correctness, putting the inner package into a vibrating sieving machine, carrying out physical sieving, and conveying the sieved mixture to a suspended weighing bin through a feed opening and a sealed pipeline.
(8) Final mixing: dry-mixing the premix with whole milk powder, fructo-oligosaccharide, xylitol, erythritol, 90% desalted whey powder and the like, fully stirring in a dry-mixing stirring cylinder to fully mix the materials uniformly, and conveying the mixed formula powder to a full-automatic packaging machine for quantitative packaging after passing through a vibrating screen and a filter screen.
(9) Metal detection: and the finished product materials to be packaged completely pass through a blanking pipeline provided with a metal detector before entering the filling machine. Before production, the standard block is used for verifying the equipment; in the production process, if the metal detector has an alarm, whether metal foreign matters exist or not is checked in time and the metal foreign matters are cleaned, so that the metal foreign matters are prevented from entering a product, and the detection limit of the metal detector is as follows: limit of iron detection:

Bottom cover, inner membrane bag: ozone sterilization is adopted, the ozone sterilization concentration is more than or equal to 10ppm, and the sterilization time is more than or equal to 60 min.
(11) Filling, vacuum nitrogen filling and packaging: the parameters of a filling machine and the net content of the product are set before starting production, the packing material is automatically supplied by equipment, and the semi-finished milk powder is continuously stirred by a spiral stirring paddle and filled into an iron can one by one. And conveying the semi-finished product to a can sealing machine through a conveying belt, automatically vacuumizing through the can sealing machine, and filling nitrogen. The pressure of the sealed tank is more than or equal to 0.2 MPa. When 800g and 400g of product are produced, the evacuation pressure P1 is set to be between-80 Kpa and-95 Kpa, and the vacuum degree P2 after nitrogen charging is set to be between-15 Kpa and-30 Kpa. When 85g and 50g of products are produced, the vacuum pressure P1 is set to be between-50 Kpa and-70 Kpa, and the vacuum degree P2 after nitrogen filling is set to be between-5 Kpa and-20 Kpa. Packaging requirements of canned products: the residual oxygen content is less than or equal to 3 percent, and the air leakage (vacuum pressure of-70 KPa, 30 s) cannot occur in the sealing property.
(12) Labeling, code spraying and boxing: and the canned product enters an outer packaging workshop through a conveyer belt, and is labeled and coded according to requirements. The code spraying should be clear and accurate. And (5) boxing according to the product requirements, printing the production date on the outer package of the box package according to the requirements, and checking whether the production date is correct or not.
(13) And (6) inspection and warehousing: and all finished products are inspected according to the product execution standard, and qualified inspection report sheets are issued after the finished products are inspected to be qualified and then are stored according to the finished product management regulations of the company.

The formula milk powder for promoting the overall development of children is characterized by comprising the following components in percentage by mass: 62-66% of full-cream milk powder, 18-22% of 90% desalted whey powder, 1-3% of xylitol, 6-10% of erythritol, 2-4% of fructo-oligosaccharide, 0.5-1% of calcium carbonate, 0.1-0.3% of DHA (docosahexaenoic acid), 0.05-0.06% of linolenic acid, 0.01-0.1% of bovine colostrum powder, 0.01-0.05% of N-acetylneuraminic acid, 0.01-0.1% of hydrolyzed yolk powder, 0.01-0.03% of lactoferrin, 0.01-0.06% of animal bifidobacterium, 0.1-0.3% of vitamin premix, 0.1-0.3% of mineral premix and 0.01-0.1% of lactase;
wherein the vitamin premix comprises the following components: vitamin C, taurine, lutein, vitamin A, vitamin D, nicotinic acid, vitamin K and vitamin B1; the mineral premix comprises the following components: magnesium, iron, zinc.

The formula milk powder for promoting the overall development of children according to claim 1, wherein the formula milk powder comprises the following components in percentage by mass: 64.75 percent of full-cream milk powder, 20 percent of 90 percent desalted whey powder, 2 percent of xylitol, 8 percent of erythritol, 3.5 percent of fructo-oligosaccharide, 0.85 percent of calcium carbonate, 0.2 percent of DHA, 0.056 percent of linolenic acid, 0.05 percent of bovine colostrum powder, 0.022 percent of N-acetylneuraminic acid, 0.06 percent of hydrolyzed yolk powder, 0.012 percent of lactoferrin, 0.05 percent of animal bifidobacterium, 0.2 percent of vitamin premix, 0.2 percent of mineral premix and 0.05 percent of lactase.
3. The formula milk powder for promoting the overall development of children according to claim 2, wherein the vitamin premix comprises the following components in percentage by mass: 19.12-25.87% of vitamin C, 17-23% of taurine, 9.95-13.46% of lutein, 1.54-2.09% of vitamin A, 1.17-1.95% of vitamin D, 1.20-1.62% of nicotinic acid, 0.46-0.62% of vitamin K, 0.20-0.28% of vitamin B1 and 31.11-49.36% of maltodextrin.
The mineral premix comprises the following components in percentage by mass: 12.54-15.34% of iron, 25.07-30.64% of magnesium, 3.64-4.45% of zinc and 49.57-58.75% of maltodextrin.
4. Use of a formula for promoting the overall development of children according to any one of claims 1 to 3 for improving obesity.
