Abstract
本发明提供了一种多金属氧酸盐POM,其化学式为:[HL]8[Cu(L)4]2H4[Cu(L)3(P2Mo5O23)]4·8H2O,其中L=咪唑。本发明还提供了掺杂其的共混水凝胶以及其制备方法,其利用POM片段的高负电荷特性能够与带正电荷的表面活性剂静电结合,得到表面活性剂修饰的多金属氧酸盐(CTAB/POM)阳离子胶束,该胶束与裸水凝胶基体通过多基团的共聚合反应生成共混水凝胶,其具有显著抑菌效力以及良好的阳离子吸附效果。

技术领域
本发明属于功能化多酸技术领域,具体涉及一种多金属氧酸盐、及其掺杂的共混水凝胶制备方法及在制备抗菌药物中的应用,其将具有良好抑菌效力的POM化合物,与表面活性剂结合形成以POM为核心的阳离子胶束,随后与具有独特结构、高溶胀率、良好局部作用、能够保持/释放药物能力的凝胶前体溶液混合形成。该类体系不仅可以在生理pH条件下保护POM分子不被分解,同时还可以提高其循环利用效率和生物效能;此外又对其实际应用潜力进行了探索,这为未来进一步的应用提供了有价值的参考。
背景技术
多金属氧酸盐(简称POMs)是一类新兴的无机金属氧化物,大多呈阴离子构型,由于其广泛的可调谐的结构和生物效应,被认为是有应用前景的金属类药物。特别是,POMs在治疗癌症、阿尔茨海默病、糖尿病等方面具有突出的生物学应用潜力。最近的研究表明,POMs可以改变与生物目标大分子的相互作用,以提高它们对特定生物系统的有利活动。然而,遗憾的是,到目前为止在临床试验中POMs还没有得到实际应用,阻碍其应用的原因在于其稳定性低,即在生理pH值下易降解为无机物质的混合物。此外,其他原因也阻碍了它们的临床应用,包括与生物分子的非特异性相互作用以及对正常组织不可避免的细胞毒性等。目前,将生物活性POMs封装到新的药物载体中,如壳聚糖、氨基酸或肽以及特定的纳米粒子(磁性Fe3O4、Au、Ag)是生物医学研究领域的主要方向。与纯无机POMs相比,新的杂化组装材料不仅能使POMs具有较高的稳定性,以保护其不被降解,能降低其对正常细胞的生理毒性,同时增强它们与生物靶点的相互作用,从而赋予了它们更有前途的治疗效力。然而,合成这样一种载体材料,同时具备可调节的尺寸、适当分子量、高载药量、在酸性/弱碱性环境下具有较好的稳定性、能以缓慢可控的方式缓释药物以及良好的生物相容性等一系列特性,已经被证明具有一定的挑战性。
水凝胶是由聚合物链通过物理相互作用或化学键交联形成的三维(3D)结构。由于其高的水溶胀性、良好的生物相容性、灵活的加载和释放药物能力以及多样性的结构,近年来在生物医学应用领域激发了人们广泛的研究兴趣。在过去的几年中,水凝胶作为药物载体得到了广泛的研究。到目前为止,已经开发了一系列改性水凝胶系统用于药物分子的靶向传递,包括无机抗菌剂,常规抗生素以及生物提取物,例如金属纳米粒子(Ag和ZnO)、氨苄青霉素钠、左氧氟沙星、海藻提取物等。将生物活性治疗药物载入水凝胶不仅可以提高药物的生物利用度,还可以控制药物的释放速率,并保持其长期生物活性,从而提高药物的局部治疗水平,降低其产生耐药的可能性。此外,药物包封的方法直接影响药物在释放过程中的可利用度。到目前为止,已经开发了几种不同的药物掺杂方法,主要包括:(一)将药物分子直接加载到水凝胶体系中,在此过程中,聚合物网络被药物分子缠绕,或者将药物分子直接分散到水凝胶交联的多孔结构中;(二)将单个药物释放系统负载到水凝胶中,即包封药物的扩散由凝胶体系和另一个释放系统共同调节,这可能是含有药物的微胶囊或纳米胶囊,目的是减缓释放速率;(三)将药物分子共价连接到水凝胶基体上,该类体系的药物释放主要是通过聚合物-药物键裂解速率来控制。为了避免生物屏障以及更有效地触发药物释放,迫切需要开发具有多反应性的聚合物材料,特别是在药物运输的过程中。能同时对一系列刺激做出响应的聚合物材料,包括物理和化学刺激,例如光、温度、pH和盐等已经得到了广泛的研究。例如,Wang等人开发了第一例具有双pH响应的聚合物颗粒,其中共聚物是通过将2,3-二甲基马来酸酐(DMMA)和阿霉素(DOX)与聚乙二醇-b-聚(烯丙基乙烯磷酸酯) (PEG-b-PAEP)结合而得到的。通过对细胞外肿瘤pH值的响应,所制备的聚合物药物颗粒能够促进药物分子在肿瘤组织的优先积累。陈和他的同事通过将聚(L-谷氨酸-co-L-赖氨酸)和1-乙基-3-(3-二甲基氨基丙基)碳二亚胺盐酸盐进行聚合得到了具有三重刺激响应(pH、离子和蛋白酶)的聚(L-谷氨酸-L-赖氨酸)型水凝胶,并研究了其对胰蛋白酶的释放行为。在这两种情况下,水凝胶的溶胀比可因pH值变化引起的电荷排斥而变化。因此,设计和合成能“感知”生理环境并按需储存/释放封装治疗药物的功能材料是非常有趣的。
发明内容
本发明目的在于克服现有技术缺陷,提供一种多金属氧酸盐及其掺杂的共混水凝胶,其将具有良好抑菌效力的POM化合物,与表面活性剂结合形成以POM为核心的阳离子胶束,随后与具有独特结构、高溶胀率、良好局部作用、能够保持/释放药物能力的凝胶前体溶液混合形成,该类体系不仅可以在生理pH条件下保护POM分子不被分解,同时还可以提高其循环利用效率和生物效能;同时又对其实际应用潜力进行了探索,这为未来进一步的应用提供了有价值的参考。
本发明还提供了上述多金属氧酸盐共混水凝胶的制备方法以及其在制备抗菌药物中的应用。
为实现上述目的,本发明采用如下技术方案:
一种多金属氧酸盐POM,其化学式为:[HL]8[Cu(L)4]2H4[Cu(L)3(P2Mo5O23)]4·8H2O,其中L = 咪唑;该多金属氧酸盐属于单斜晶系,C2/c空间群,晶胞参数为:a = 43.679(3)Å,b = 8.7794(6) Å,c = 24.3844(18) Å,α = 90°,β = 112.7320(10)°,γ= 90°。
本发明提供了上述多金属氧酸盐的制备方法,其将Cu(ClO4)2·6H2O和咪唑溶于水中,50-70℃条件下搅拌反应30-60 min,冷却至室温后,加入到Na2MoO4·2H2O水溶液中,在连续搅拌条件下滴加浓H3PO4调节pH值至3.0-4.0,继续搅拌反应30-60 min,过滤,滤液室温静置挥发,析出的晶体(一般需3-10天)即为多金属氧酸盐POM。
具体的,所述Cu(ClO4)2·6H2O、咪唑和Na2MoO4·2H2O的摩尔比为1:2-4:4-6。
本发明提供了一种掺杂上述多金属氧酸盐的共混水凝胶的制备方法,其包括下述步骤:
1)制备表面活性剂修饰的多金属氧酸盐胶束分散液(CTAB/POM):
将1.0-10 mg的多金属氧酸盐均匀分散于1 mL去离子水中得到悬浊液,然后与2mL浓度0.6 M的十六烷基三甲基溴铵(CTAB)溶液混合,超声30-60 min以形成CTAB包覆的POM胶束(核心是POM分子),获得多金属氧酸盐胶束分散液;
2)采用共聚合法制备多金属氧酸盐掺杂的共混水凝胶:首先,以过硫酸铵(APS)为引发剂,进行了甲基丙烯酸(MAA)改性β-环糊精(β-CD)获得MAA/β-CD,然后与N,N-亚甲基双丙烯酰胺(MBA)进行自由基聚合反应。具体如下:
将2.8 mmol N,N'-羰基二咪唑(CDI)、2.2-2.6 mmol MAA和2.2-2.6 mmol β-环糊精(β-CD)加入到30 mL水溶液中,室温下搅拌反应12-15 h,然后在异丙醇中沉淀,洗涤、干燥得到MAA/β-CD;
将3 mL多金属氧酸盐胶束分散液加入到等体积量的凝胶前驱体溶液中,然后在N2气氛下于50-70℃搅拌反应1-2 h即得共混水凝胶。将所得共混水凝胶置于冷冻干燥器中,得到冻干产物并用于后续实验。
具体的,步骤2)中,所述凝胶前驱体溶液中含有0.15-0.18 g MAA/β-CD、5.0-5.5mmol MAA、0.2-0.3 mmol MBA和0.06-0.10 mmol 过硫酸铵。
本发明提供了采用上述制备方法制备得到的掺杂多金属氧酸盐的共混水凝胶,POM分子有望均匀地分散到水凝胶的孔隙中。
本发明还提供了上述掺杂多金属氧酸盐的共混水凝胶在制备抗炎药物中的应用。
本发明还提供了上述掺杂多金属氧酸盐的共混水凝胶在作为阳离子染料吸附剂中的应用。具体的,所述阳离子染料吸附剂为亚甲基蓝(MB)、碱性品红 (FB)、龙胆紫(GV)、罗丹明B(RhB)、刚果红(CR)等。
上述共混水凝胶制备过程中,第一个步骤包括将前驱体POM悬浮液在超声作用下滴加到CTAB的水溶液中,溶液中带高负电荷的POM片段能够静电结合到带正电荷的表面活性剂上,促使载有POM胶束的最终形成;第二个步骤是在搅拌和N2气氛下,将上述得到的POM阳离子胶束加入到等体积的裸水凝胶基体溶液中,通过多个交联基团的共聚反应合成了共混水凝胶,其中CTAB/POM胶束被认为均匀地分布到共混水凝胶结构中。由于凝胶材料的固有吸水性能,将材料加入菌悬液中,材料从周围环境中吸收水分,从而使细菌细胞失去生存所需的营养介质。其次,所制备的共混水凝胶由于阳离子CTAB/POM胶束的引入而带正表面,可与带负电荷的细胞壁/膜相互作用,促使细菌细胞在初始阶段的粘附。
本发明中,负电荷的POM片段能够静电结合到带正电荷的表面活性剂上,形成CTAB/POM阳离子胶束,该阳离子胶束通过共聚反应能够与裸水凝胶基体溶液反应合成共混凝胶。共混水凝胶具有吸水特性,能够剥夺细菌细胞生存所必须的环境,其次在表面正电荷的作用下能与带负电荷的细菌细胞壁/膜作用,对细菌进行捕获,从而实现此共混水凝胶的合成条件、抗菌效力和实际应用潜能。
本发明提供了一例凝胶前体用于负载和控制生物活性POM分子的包封和释放,以提高生理稳定性,降低毒性效应,从而提高POM组分的生物利用度。考虑到POM的优良生物学效应以及水凝胶的控释特性,本申请合理地假设所得的共混水凝胶具有双重优势,即能够保持同等的治疗效力,同时相比于纯POM又能提高其生理稳定性。具体来说,将POM分子封装到表面活性剂十六烷基三甲基溴化铵中形成以POM为核心的阳离子胶束,然后再将添加到裸水凝胶基体溶液中,通过多个交联基团间的共聚合反应获得共混材料。值得注意的是,甲基丙烯酸(MAA)及其衍生物基pH响应型水凝胶处于紧致的收缩状态,可以避免酸性/中性pH环境中的药物泄漏。通过这样的设计,水凝胶不仅可以在生理pH条件下保护POM分子,同时还可以提高其循环利用效率和生物效能。
本发明围绕严重的细菌感染问题,开发高效、可循环利用的抗菌试剂。本发明公开了一类具有优良抑菌效力的共混水凝胶。将表面活性剂CTAB修饰的POM阳离子胶束加入到等量体积的裸水凝胶基体溶液中,通过多个交联基团的共聚反应合成共混水凝胶,所得的杂化体系不仅可以在生理pH条件下保护生物活性POM分子不被分解,同时兼具凝胶材料高溶胀率、良好局部作用、控/释药性能,还可以提高材料的循环利用效率。此外,还对所得材料的实际应用潜力进行探索,这为其未来进一步的应用提供有价值的参考。本发明制备的共混水凝胶的合成路线如下:
裸凝胶基体溶液 + CTAB/POM →共混凝胶,其中,POM为Strandberg型多酸阴离子。即利用POM片段的高负电荷特性能够与带正电荷的表面活性剂静电结合,得到阳离子CTAB/POM胶束,该胶束与裸水凝胶基体通过多基团的共聚合反应生成共混水凝胶,其可能的抗菌机制见图11。由于凝胶的优越特性以及POM的固有抗菌性能,共混水凝胶被赋予更强的抗菌效果,其产生显著抑菌效力的原因如下:(1)凝胶材料具有吸水性能,将材料加入菌悬液中,材料从周围环境中吸收水分,从而使细菌细胞失去生存所需的营养介质;(2)所制备的共混水凝胶由于大量阳离子CTAB/POM胶束的存在而带正表面,可与带负电荷的细胞壁/膜相互作用,促使细菌细胞在初始阶段的粘附;(3)POM的广谱生物活性(使细胞壁/膜破裂、胞内物质泄漏以及生物靶点干扰等)赋予共混水凝胶更强的杀菌作用,表面活性剂CTAB的共聚合进一步加强这一作用;(4)总之,细菌与样品表面电荷的协同作用、生物活性成分的存在以及凝胶的自身特性都是本申请制备共混水凝胶所具备的重要优势,这正是抗菌生物材料所必备的特性。
本发明通过将POM分散到CTAB溶液中,产生了POM包埋的阳离子胶束。随后,采用直接加载策略,将得到的胶束封装到水凝胶的前驱体溶液中制备共混材料。分别对革兰氏阴性大肠杆菌和革兰氏阳性金黄色葡萄球菌两种菌株的抑菌效力进行了评价,同时对其作用机制进行了探究。此外,还对其实际应用潜力进行了评估。和现有技术相比,本发明具有如下优点:
1)合成的共混水凝胶在弱酸性/中性pH条件下具有较好的pH响应行为,这是避免酸性阴离子POM组分沉积时分解所必需的一个特征;
2)选择的凝胶前体具有大孔结构,这为POM组分的充分扩散提供了足够的空间;
3)制备的共混水凝胶具有较高杀菌效率和良好的局部作用,能够在一定时间内保持治疗效果;
4)所制备的共混水凝胶材料即使经过六次循环,仍具有一定的抗菌效力。此外,本申请还对该材料的可能作用机制进行了阐明,坚信所得到的杂化水凝胶材料在生物医学领域有着很大的应用前景。

具体实施方式
以下结合实施例对本发明的技术方案作进一步地详细介绍,但本发明的保护范围并不局限于此。
实施例1:
一种多金属氧酸盐POM,其化学式为:[HL]8[Cu(L)4]2H4[Cu(L)3(P2Mo5O23)]4·8H2O,其中L = 咪唑。
本发明提供了上述多金属氧酸盐的制备方法,具体为:
将Cu(ClO4)2·6H2O (0.056 g,0.15 mmol)和咪唑(0.02 g,0.3 mmol)溶于水中获得水溶液,在50℃条件下搅拌反应30 min,冷却至室温后,加入到10 mL Na2MoO4·2H2O(0.145 g,0.6 mmol)水溶液中,在连续搅拌条件下滴加浓H3PO4调节pH值至3.0,继续搅拌反应30 min,过滤,滤液室温静置挥发,7天后析出了蓝色条状的晶体即为多金属氧酸盐POM。
X-射线单晶衍射的分析结果表明,上述制备得到的多金属氧酸盐POM的晶体结构类型属于单斜晶系,C2/c空间群,晶胞参数为:a = 43.679(3) Å,b = 8.7794(6) Å,c =24.3844(18) Å,α = 90°,β = 112.7320(10)°,γ= 90°。配合物POM (图4A) 结构单元中存在由[Cu(1)(L)4]连接的两个[P2Mo5O23]6–阴离子结构,另外两个[Cu(2)(L)3(H2O)]小单元分布在左右两端,两个{[Cu(2)(L)3(H2O)] [P2Mo5O23]}4–关于[Cu(1)(L)4]呈中心对称。图4B呈现了沿b轴的三维网络多面体,可以观察到相邻两个单元的相互作用,连接成一维单链结构,然后相邻的两条单链进一步生成一个具有一维阶梯链的结构。这些结果与X-射线衍射结构分析的结果一致。图5给出了POM的单晶XRD模拟与粉末XRD对比图,通过对比衍射峰的位置可以看出两者之间有着很好的吻合,表明所得的POM具有较好的相纯度。图6给出了POM的SEM及对应的mapping图谱,从图中可以看出POM呈现棒状结构和光滑的表面(部分因外部碾压而破坏),元素映射清楚地显示了P(青色)、Mo(紫色)、Cu(白色)和N(绿色)的存在,且磷,钼的含量要大于铜和氮的含量,这与上述元素分析结果相一致,也证实了所制备样品的规则均匀性。
一种掺杂上述多金属氧酸盐的共混水凝胶的制备方法(合成路线见图1),其包括下述步骤:
1)制备表面活性剂修饰的POM胶束分散液(CTAB/POM):
预先制备2 mL、0.6 M的十六烷基三甲基溴铵(CTAB)溶液,将10 mg的多金属氧酸盐POM均匀分散于1 mL去离子水中得到悬浊液,随后将其加入上述制得的CTAB溶液中,超声作用30 min以形成CTAB包覆的POM胶束,即得CTAB修饰的多金属氧酸盐胶束分散液;
2)采用共聚合法制备多金属氧酸盐掺杂的共混水凝胶:首先,以过硫酸铵(APS)为引发剂,进行了甲基丙烯酸 (MAA)改性β-环糊精(β-CD)获得MAA/β-CD,然后与N,N-亚甲基双丙烯酰胺(MBA)进行自由基聚合反应。具体如下:
将2.8 mmol N,N'-羰基二咪唑(CDI)、2.4 mmol MAA和2.4 mmol β-环糊精(β-CD)加入到30 mL水溶液中,室温下剧烈搅拌反应12 h,然后在100 mL异丙醇中沉淀,沉淀用乙醇洗涤、干燥得到MAA/β-CD;
将3 mL CTAB修饰的多金属氧酸盐胶束分散液加入到等体积量的凝胶前驱体溶液(所述凝胶前驱体溶液中含有0.16 g MAA/β-CD、5.3 mmol MAA、0.25 mmol MBA和0.08mmol 过硫酸铵)中,然后在N2气氛下于60℃搅拌反应1 h即得共混水凝胶。将所得共混水凝胶置于冷冻干燥器中,得到冻干产物并用于后续实验;
另外,在上述所述步骤2)中,将不添加多金属氧酸盐POM的胶束分散液与凝胶前驱体溶液直接反应获得裸水凝胶,将得到的裸水凝胶同样置于冷冻干燥器中,得到冻干产物用于对照实验。
图1给出了共混水凝胶的合成路线及简要的抗菌实验过程。图1中可以看出:水凝胶被用作药物载体,用于构建基于POM的药物混杂体系。采用共聚合方法,将表面活性剂用作阳离子底物来捕获POM分子以形成POM改性的阳离子胶束,然后再将其添加到裸水凝胶基体溶液中形成POM负载的共混材料。简单地说,第一个步骤是在超声作用下将前驱体POM悬浮液滴加到CTAB水溶液中,最终形成以POM为核心的胶束。在溶液中带高负电荷的POM分子能够以静电结合的方式键合到带正电荷的表面活性剂上,以下Zeta电位测量值也证明了这一点。其次,在搅拌和N2气氛下,将上述得到的阳离子胶束加入到等体积量的凝胶前驱体溶液中,成功地合成了共混水凝胶,其中阳离子胶束被认为均匀地分布到凝胶结构中。最后,对所得材料进行收集、冷冻干燥、表征并进一步应用。
图2给出了上述所得水凝胶材料的SEM及EDS能谱图。图中可以看出:裸水凝胶是由许多小的玫瑰花状结构组合而成的,还可以观察到样品在整体上有大量典型的互穿孔结构。此外,放大图像的观察表明,小玫瑰结构是由几十个花瓣组成的,它们随机地相互结合,形成玫瑰状的花朵形状。并且每个花瓣层都很薄。这些玫瑰状结构与大的比表面积密切相关,可以促进大量溶剂的吸收以及多种药物的负载。相反,与CTAB/POM组分聚合后,共混水凝胶样品形貌发生了显著的变化,其中薄片状结构变得支离破碎,表面更加无序和粗糙。这一结果归因于CTAB/POM胶束的引入干扰了裸水凝胶的正常形成,从而破坏了原始有序的薄层状结构。对结构进一步放大后,观察到在单个花瓣层中存在POM分子,还可以观察到破裂碎片表面相对均匀、平坦。EDS能谱中可以观察到P、Mo和Cu元素的存在,表明POM组分成功地结合到共混水凝胶中。
图3给出了上述制备所得水凝胶材料的光学图像。图中可以看出:与裸水凝胶相比,共混合水凝胶的颜色由纯白色变为浅黄色。上述所有这些颜色变化主要归因于POM分子的掺入,从而影响了杂化材料的最终颜色和结构。
图7给出了上述所制备材料以及前驱体材料(β-CD、CTAB和POM)的红外图谱。β-CD图谱中3377和2925 cm‒1处的振动峰是O‒H和C‒H的拉伸振动所致。葡萄糖单元中的C‒O,C‒O‒C以及环CD中的C‒O‒C的特征峰在1642、1155和1034 cm‒1处也可以观察到。在裸水凝胶和共混水凝胶的图谱中,能够清楚地观察到这些特征振动峰,证实了β-CD的存在。值得一提的是,随着水凝胶的形成,由于MAA的交联相互作用,这些基团的峰位置发生了轻微的红移。此外,游离POM组分在1107、1048、907和683 cm‒1处强的拉伸分别对应于P‒O、Mo=Od,以及Mo‒O‒Mo的拉伸振动。CTAB图谱中2922、2854和1475 cm‒1处的尖峰分别归属于‒CH2和RN(CH3)3 +拉伸振动。所有这些特殊的拉伸和振动峰都可以在CTAB/POM胶束以及最终的共混材料中检测到,证实了共混水凝胶中这些组分存在。
实施例2:
Zeta电位样品处理步骤:为了更好地理解上述合成过程,通过Zeta电位分析仪测量了所得凝胶材料和多金属氧酸盐胶束分散液的Zeta电位。10 mg样品均匀地分散在10 mL去离子水中进行分析。
图8为样品的Zeta电位测量值作图,(a‒c) 分别代表裸水凝胶、CTAB/POM胶束及共混水凝胶。图中可以观察到一个显著的现象,即共混水凝胶与裸水凝胶(‒35.4±0.7 mV)相比,其正电荷显著增加。这一趋势可以通过多金属氧酸盐胶束的表面强的正电荷(+46.8±0.4 mV)来解释,由于合成过程中聚合物基体间静电相互作用,从而使得最终水凝胶产物的表面电位发生转变(+29.1±0.6 mV)。总的来说,这一结果为CTAB/POM组分在共混水凝胶材料中的掺入提供了证据,并强烈地说明本发明合成策略的可行性。除此之外,封装的阳离子生物聚合物被认为与用于制备水凝胶的带电聚合物基质静电相互作用,从而影响材料的性能,例如,负载、保留、释放以及生物学效应。
实施例3:
凝胶溶胀能力评价步骤:200 mg的冻干共混水凝胶样品浸泡在不同的溶液中(0.75% NaCl溶液、纯水以及pH为6‒8的生理缓冲液)。在一定的时间间隔内(5、10、20、40、60、80、100、120、150 min),通过滤纸缓慢吸除表面过量的溶剂来测量样品的重量。随后,再将其放入溶剂中,直至达到溶胀平衡。根据方程(Eq 1)确定凝胶的溶胀比q。
溶胀比q (%) = (样品溶胀态重量W ‒ 冻干态下的重量Wo) / 冻干态下的重量Wo× 100% (Eq 1)。
图9A给出了不同溶剂体系对共混水凝胶溶胀性能的影响。溶胀比是凝胶材料的一个基本性质,以更好地说明溶剂的可能摄取,特别是在生物医学领域,如伤口敷料。根据先前的报道,溶胀响应与负载材料的性质、外介质中的离子浓度、pH以及孔径和分布等密切相关。在此,对共混水凝胶在非缓冲液(0.75% NaCl溶液、纯水)和缓冲溶液(pH为6-8的生理缓冲液)中的溶胀性能进行了研究。从图中可以清楚地观察到,所有实验组都表现出相似的溶胀反应,包括快速增长阶段和相对稳定的上升期。总的来说,前5 min时间段内,溶胀比快速的增强可能是水凝胶表面快速吸收的结果,其中以渗透压的方式混合的自由能使溶剂扩散到干燥的凝胶基质中。随着时间的延长,溶胀比增加,但总体上呈稳定的趋势,这一阶段可以归结为溶剂在材料内部的渗透,导致渗透压的增加,阻止更多的溶剂分子进入,因此,整体吸收趋势较前5 min明显下降。此外,材料在缓冲pH溶液(pH 6.0除外)体系比非缓冲溶液体系具有更大的溶胀性,其中材料在缓冲pH 8.0中有着最大溶胀比为229%,这是由于凝胶网络中羧基的电离所致。NaCl溶液中材料的溶胀比为205%,低于纯水中的溶胀比,这可能是凝胶内部渗透压的增加所致,导致溶剂的内向运动最小化。
实施例4:
药物释放性能评价步骤:将得到的冻干共混水凝胶样品200 mg置于100 mL的溶液(纯水和pH = 8.0的生理缓冲液)中,在室温下轻轻搅拌。在选定的时间点(5、10、20、40、60、80、100、120、150 min),从系统中吸取5.0 mL的溶液,并通过UV-Vis在258 nm (POM) 处检测溶液中含有的POM量。以冻干样品中所含的POM量为主要标准,根据Eq 2计算POM的累计释放百分比。
累计释放量 (%) = (实验组峰强度值I) / 冻干样品峰强度值Io × 100% (Eq2)。
图9B给出了共混凝胶中POM组分随着时间延长的线性释放曲线,实验研究了具有良好溶胀行为的pH 8.0(缓冲溶液)和纯水(非缓冲溶液)这两个具有代表性的溶液体系。根据先前的研究报道,聚合物体系中药物释放的最常见的三种机制,包括侵蚀、扩散和溶胀。侵蚀是水解过程,聚合物基体从边缘开始降解,并导致药物释放。扩散发生在聚合物基体内,其中存在着浓度差异,样品与溶剂直接接触,因此它与水凝胶的溶胀比直接相关。从图中可以看出,共混水凝胶在初始阶段有着较大的药物释放速率,特别是在前5 min时间段内,在随后的时间段内表现出稳定的上升。缓冲溶液体系中,在最初的5 min内,共混水凝胶的POM组分累积释放率约为63%。随着释放时间从5 min延长到120 min,POM组分从凝胶内部缓慢扩散到外部介质中,在120 min时,释放率接近100%。同样,对于纯水介质,凝胶在整个阶段的释放速率比上述体系慢。在最初5 min内有40%的POM组分释放,随后释放比率保持稳定增长,并在150 min的时间点处建立平衡。总的来说,在早期阶段,POM组分的缓释遵循溶胀控释机制。当达到平衡点时,释放以扩散控制的方式进行,达到最大释放量100%。在整个阶段,释放归因于材料与宿主凝胶的分子间和分子内相互作用,包括凝胶与POM官能团之间氢键、范德华尔相互作用以及与介质溶剂分子间的缩合反应。
实施例5:
抑菌效力评价方法1圆盘扩散法:
实验前将得到的冻干共混水凝胶样品切成直径为8 mm,厚度为2 mm的圆柱体。将所有样品都在紫外线照射下进行灭菌处理。新鲜菌株(革兰氏阳性菌:金黄色葡萄球菌S. aureus;革兰氏阴性菌:大肠杆菌E.coli)分布到营养琼脂板上,随后将材料放置在上面,将琼脂板放置在隔水培养箱中37℃培养12 h,最后测定样品的抑菌圈(ZOI)外径并记录光学图像。将样品取下处理后转移到另一新鲜的酵母蛋白胨培养基(酵母浸粉10 g、蛋白胨5 g、NaCl 10 g、琼脂15 g、高纯水1000 mL、pH 7.0,培养基采用0.11 Mpa 121℃蒸汽灭菌20min)上,实验循环进行7次。实验以裸水凝胶为对照组,以共混水凝胶为实验组。
图10A给出了材料作用后的直观抑菌圈图像,物质的抗菌能力是通过测量抑菌圈的直径来评估的,其中圈的大小代表抗菌效果的强弱。从图中可以观察到,与共混水凝胶相比,裸水凝胶的抑制区几乎不可观察,即抑制圈非常小。而对于共混水凝胶,可以观察到明显的抑制区。上述抑菌圈大小的巨大差异充分地表明POM组分在杂化材料的抗菌作用中发挥着至关重要的作用(单独CTAB无抑制活性)。此外,还必须强调的是,共混水凝胶对金黄色葡萄球菌的活性更强,这一趋势可归因于金黄色葡萄球菌的细胞壁完全由肽聚糖原组成,而大肠杆菌由肽聚糖原和外脂多糖层组成,从而削弱了材料的生物学效应。
实施例6:
抑菌效力评价方法2光密度值(OD)测量法:
配置菌悬液:用接种环挑取一环培养好的菌体细胞(大肠杆菌/金黄色葡萄球菌)放入20 mL生理盐水里,充分振摇10 min,制成菌体悬液备用。然后对每一菌体悬液测OD600值。将得到的冻干共混水凝胶样品灭菌处理后浸入相对应的菌悬液中,在37℃下共培养12h,随后对菌悬液进行稀释并记录其在600 nm处的OD值,以无样品作用的菌悬液视为100%细胞存活率。用Eq 3计算细菌细胞的存活率。循环实验中,对材料进行回收和再处理,并按照如上所述的步骤重复6个循环。
细菌细胞存活率 (%)= (实验组OD值) / 对照组细胞OD值 × 100% (Eq 3)。
图10B给出了凝胶材料处理12 h后的菌株的光密度测量值(方框内的是循环试验结果,其变化趋势用黑色箭头表示)。从图中可以看出,材料对两种菌株细胞活力的影响呈现出相似的趋势,其中,共混水凝胶样品致使细菌细胞存活率显著降低(材料作用后大肠杆菌和金黄色葡萄球菌存活率分别为3.5%和4.5%)。而裸水凝胶对两株菌株的生长抑制作用较小。这些结果进一步证实了POM组分在提升抗菌效力中的重要性。此外,还对共混水凝胶的可回收性进行了研究。循环试验结果表明即使在六次重复使用后,材料仍有51%的杀菌率,清楚地表明该聚合物材料潜在的生物应用前景。
实施例7:
抑菌效力评价方法3细胞染色法:
将得到的冻干共混水凝胶样品与1 mL OD600值为0.1的大肠杆菌悬浮液在37℃下进行共培养12 h,去除样品后用染料碘化丙啶(PI, 5 μL,1 mg/mL)和二乙酸荧光素(FDA,2 μL,5 mg/mL)对上述菌体悬浮液进行染色,于黑暗条件下作用15 min。用共聚焦荧光显微镜记录图像,其中死亡的细菌细胞与PI结合,在λex/λem = 561/620 nm波长处发出红色荧光,活细胞与FDA结合后在488/530 nm波长处显示出绿色荧光。
图10C给出了细胞荧光染色图像,对于裸水凝胶,三维图像内的大多数细胞发出绿色荧光。相反,用共混水凝胶培养后,红色斑点的分布较多,表明POM组分的加入增强了材料的杀菌作用。此外,三维图像中的细菌细胞处于簇状聚集体状态,而不是均匀分布的斑点,这可能是由于悬浮液的不均匀分散以及细胞与实验过程中释放的凝胶基质之间的相互作用所致。
实施例8:
扫描电镜观察样品-细菌相互作用:用扫描电镜对样品上大肠杆菌细胞的形态变化进行了表征。将预浸入1 mL大肠杆菌悬浮液(106 CFU/mL磷酸盐缓冲盐水(PBS)中的冻干共混水凝胶样品用2.5%戊二醛溶液固定4 h,然后依次用梯度浓度(0、30、50、70、90、100%、100%)的乙醇溶液进行组织脱水。冷冻干燥处理后,用SEM监测粘附细菌细胞与样品间的相互作用,测试前将金溅射到样品表面。
图10D给出了细胞与水凝胶材料间相互作用显微图像,从图中可以看出,一定数量的菌体细胞沉积在材料中,这细胞在材料表面的初始相粘附和附着提供了证据。此外,还可以观察到,裸水凝胶上的细菌基本上保持了完整的细胞结构,呈棒状,其中,不光滑的表面以及小孔洞可能是裸水凝胶作用的结果。相反,在共混水凝胶材料表面几乎没有完整的细胞出现,它们都处于严重的凋亡状态,以白色圆圈标记,包括细胞骨架破坏、细胞内成分泄漏和细胞的整体碎裂。这些结果与上述抗菌活性数据一致,证实了共混水凝胶材料组分间的协同作用。
实施例9:
体外细胞毒性评价方法:采用MTT(3-[4,4,5-二甲基硫氮醇-2-基]-2,5-二苯基四碘化)法评价样品对人正常肝细胞HL-7702细胞存活率的影响。将细胞接种在96孔板中,并在37℃培养箱(5%CO2)中培养12 h。将样品(裸水凝胶和冻干共混水凝胶)放入孔中替代原始细胞培养基并孵育24 h。最后,去除样品,用MTT法测定作用24 h后的细胞OD值,记录每个孔在570 nm处的OD值。用Eq 4计算细胞活性 (%) = (OD样品) / OD对照× 100% (Eq 4),以磷酸盐缓冲液处理的细胞被认为是具有100%的细胞存活率。
图12给出了不同样品的细胞毒性结果,与空白对照组相比,裸水凝胶组的细胞存活率略有下降。而在共混水凝胶组,细胞存活率出现了显著的下降(50%的存活率),这表明共混水凝胶的局部、持续作用导致了一定的毒性,但不会造成严重的损伤。
实施例10:
凝胶材料为水处理提供了替代的策略,因为它们的溶剂吸收能力以及可逆的溶胀/脱水性能。为了评估其实际应用潜力,已经开发了两种评价方法:(1)微生物抑制实验;(2)污染物的吸附。所有这些策略都取决于水凝胶吸收物质(例如水和小分子)和/或捕获/杀死其他微生物的能力。本申请采集多种水样(雨水、湖水和自来水)进行样品的微生物抑制测试,通过评估共混水材料对微生物的抑制能力,评价了材料的实际应用性能。
图13(左)给出了样品的抑制微生物实验结果,从图中可以看出样品对所采集的水样都表现出显著的抗微生物效力。此外,还通过监测染料(亚甲基蓝(MB)、碱性品红 (FB)、龙胆紫(GV)、罗丹明B(RhB)、刚果红(CR)和甲基橙(MO)从水溶液中的去除情况,评估材料的污染物吸附能力。图13(右)给出了样品在各类染料介质中的吸附结果,从图中可以观察到共混水凝胶能够选择性地吸附阳离子染料(MB、FB、GV、RhB和CR),这依赖于静电相互作用。而对阴离子染料MO无特异性吸附作用。这可能是由于长时间的作用使其中的POM组分完全释放所致,从而使系统恢复到原来的负电荷状态。目前正在进行详细的实验探究,以进一步准确地确定这一杂化材料吸附行为的机制。
综上,本申请给出了一个有前景的水处理应用实例,为进一步的实际应用提供了有价值的参考。此外,还对该材料的未来应用前景进行了预测,主要包括以下领域:(一)废水处理:作为净水的过滤材料;(二)生物医学领域:伤口敷料、组织相关感染和治疗炎症反应;(三)医疗保健,例如设备材料涂层;(四)食品工业或其他领域。所有这些未来应用方向预测将为设计和合成具有预期性能的下一代混合材料开辟一条新途径。
Claims (5)
1.一种掺杂多金属氧酸盐的共混水凝胶的制备方法,其特征在于,包括下述步骤:
1)制备表面活性剂修饰的多金属氧酸盐胶束分散液:
将1.0-10 mg的多金属氧酸盐均匀分散于1 mL去离子水中得到悬浊液,然后与2 mL浓度0.6 M的十六烷基三甲基溴化铵溶液混合,超声30-60 min,获得多金属氧酸盐胶束分散液;
所述多金属氧酸盐化学式为:[HL]8[Cu(L)4]2H4[Cu(L)3(P2Mo5O23)]4·8H2O,其中L = 咪唑;该多金属氧酸盐属于单斜晶系,C2/c空间群,晶胞参数为:a = 43.679(3) Å,b =8.7794(6) Å,c = 24.3844(18) Å,α = 90°,β = 112.7320(10)°,γ= 90°;
2)制备多金属氧酸盐掺杂的共混水凝胶:
将2.8 mmol N,N'-羰基二咪唑、2.2-2.6 mmol 甲基丙烯酸和2.2-2.6 mmol β-环糊精加入到30 mL水溶液中,室温下搅拌反应12-15 h,然后在异丙醇中沉淀,洗涤、干燥得到MAA/β-CD;
将3 mL多金属氧酸盐胶束分散液加入到等体积量的凝胶前驱体溶液中,然后在N2气氛下于50-70℃搅拌反应1-2 h即得;
步骤2)中,所述凝胶前驱体溶液中含有0.15-0.18 g MAA/β-CD、5.0-5.5 mmol 甲基丙烯酸、0.2-0.3 mmol N,N' -亚甲基双丙烯酰胺和0.06-0.10 mmol 过硫酸铵。
2.采用权利要求1所述制备方法制备得到的掺杂多金属氧酸盐的共混水凝胶。
3.权利要求2所述掺杂多金属氧酸盐的共混水凝胶在制备抗菌药物中的应用。
4.权利要求2所述掺杂多金属氧酸盐的共混水凝胶在作为阳离子染料吸附剂中的应用。
5.如权利要求4所述掺杂多金属氧酸盐的共混水凝胶在作为阳离子染料吸附剂中的应用,其特征在于,所述阳离子染料为亚甲基蓝、碱性品红、龙胆紫、罗丹明B或刚果红。
