adding 90% ethanol 13 times the weight of Oletum Trogopterori, heating, reflux-extracting, cooling to room temperature, filtering, collecting ethanol extractive solution, concentrating to 10% of the original volume, and spray drying to obtain Oletum Trogopterori extract;
and mixing 190g of the prepared sweet wormwood herb extract, 250g of perilla extract and 110g of trogopterus dung extract to obtain the traditional Chinese medicine composition for preventing and treating the dengue fever virus.

Compared with example 5, the traditional Chinese medicine composition for preventing and treating dengue fever virus provided in comparative example 1 has the same raw materials and preparation method as example 5 except that the artemisia annua extract and the content thereof are omitted.
Comparative example 2
Compared with example 5, the traditional Chinese medicine composition for preventing and treating dengue fever virus provided in comparative example 2 is identical to example 5 in all raw materials and preparation method except that the perilla herb extract and the content thereof are omitted.
Comparative example 3
Compared with example 5, the traditional Chinese medicine composition for preventing and treating dengue fever virus provided in comparative example 3 is the same as example 5 except that trogopterus dung and the content thereof are omitted.
Comparative example 4
Compared with example 5, the artemisia apiacea extract in the traditional Chinese medicine composition for controlling dengue fever virus provided by comparative example 4 is prepared by the following steps: grinding fresh herba Artemisiae Annuae, pulping, filtering, and evaporating at 35 deg.C to obtain herba Artemisiae Annuae extract. The remaining raw materials and the preparation method were the same as in example 5.
Comparative example 5
Compared with example 5, the perilla extract liquid in the traditional Chinese medicine composition for preventing and treating dengue fever virus provided by comparative example 5 is prepared by the following steps: pulping whole perilla, filtering, performing ultrasonic extraction with ultrasonic power of 450w, and filtering with diatomite filter at 90 deg.C for 110min, wherein the weight ratio of crushed perilla to water is 1: 11. The remaining raw materials and the preparation method were the same as in example 5.
Comparative example 6
Compared with the example 5, in the comparative example 6, when the traditional Chinese medicine composition for preventing and treating dengue fever virus is prepared, trogopterus dung is directly pulverized and added for use, and the rest raw materials and the preparation method are the same as those in the example 5;
first, cytotoxicity test
1. Test samples: the traditional Chinese medicine composition for preventing and treating dengue fever virus provided by the embodiment 5 of the invention.
2. Test methods and test results: preparing a single cell suspension by using a DMEM medium containing 5% fetal bovine serum, and inoculating 2000-3000A 549 cells in each hole to a 96-hole cell culture plate, wherein the inoculation volume of each hole is 100 uL; at 37 ℃ 5% CO2Culturing for 1 day under the culture condition; the DMEM medium in each well was aspirated and 100uL of the Chinese medicinal composition of example 5 diluted to the corresponding concentration (10UM, 25UM, 50UM) in DMEM medium containing 5% fetal bovine serum was added to each well, and 100uL of DMEM medium containing 5% fetal bovine serum without drug was added to the control wells; after 24 hours of incubation, 10uL of MTT solution was added to each well at 37 ℃ in 5% CO2Continuously incubating for 4 hours under the culture condition, then terminating the culture, removing culture supernatant in each hole by suction, adding 150uL DMSO in each hole, and oscillating for 10min to fully melt crystals; the 490nm wavelength was selected, the absorbance of each well was measured on an enzyme linked immunosorbent assay, and the results are reported in Table 1.

As can be seen from the above tests, the survival rates of a549 cells at the concentrations of the Chinese medicinal composition provided in examples 5 of 0UM, 10UM, 25UM and 50UM were 100%, 96.7%, 95.3% and 92.4%, respectively.
The cytotoxicity test of the traditional Chinese medicine composition prepared in the embodiments 1 to 4 of the invention is carried out by adopting the test method, and the test results are shown in the following table 2.

From the test results in table 2 above, the survival rates of a549 cells at the concentrations of the traditional Chinese medicine compositions provided in examples 1 to 4 of 0UM, 10UM, 25UM and 50UM are respectively 100%, 94.5% to 96.3%, 93.7% to 95% and 91.3% to 92.1%.
Second, inhibition of dengue II virus
DENV2 MOI =1 infected a549 cells, experimental procedure as follows:
1. planting cells: a549 cells are inoculated in a 24-hole cell culture plate, after 24 hours, the cells grow to a monolayer (the area of the cell covering the bottom of a hole is about 80-90%), a culture medium is sucked out, a virus sample is inoculated in 100uL, and the virus sample is adsorbed at 37 ℃ for 1 hour. 2. TCID50 experiments:
determination of the minimum viral load causing 50% of the lesions after incubation of dengue type II virus infected susceptible cells C6/36, namely: c6/36 cells are inoculated in a 96-well plate → 24h, and then 100uL dengue fever type II virus diluted by 10 times in a culture medium containing 2% FBS is sequentially inoculated, the dilution is 10-1 to 0-10, each dilution has 6 auxiliary wells → is placed in a 37-welloC,5%CO2Adsorbing for 1 hr in incubator, adding 100uL of 2% FBS-containing medium at 35 deg.C and 5% CO2And (5) culturing in an incubator, observing and recording results after about 5-6 days, wherein the results are shown in the following table 4.

As shown in the test results of tables 2 to 4 above, the traditional Chinese medicine composition for preventing and treating dengue fever virus provided by embodiments 1 to 5 of the invention has low toxicity and high efficiency, can effectively inhibit dengue fever type II virus, and has an inhibition effect obviously superior to that of comparative examples 1 to 6. This indicates that the inhibition effect of the extract of Artemisia annua, the extract of Perilla frutescens and Oletum Trogopterori (extract) is better than that of the extract of Artemisia annua, the extract of Perilla frutescens and Oletum Trogopterori (extract) which are used alone. And when preparing the sweet wormwood extract, the sweet wormwood extract is prepared by the process of soaking the sweet wormwood extract by a sodium carbonate solution and then pulping and extracting; when preparing the perilla extracting solution, concentrating the filtered perilla filtrate, and adding ethanol for adjustment to obtain the perilla extracting solution; and the trogopterus dung extract prepared by the alcohol extraction process is beneficial to improving the inhibiting effect of the composition on dengue fever II viruses.

Claims (10)

1. The traditional Chinese medicine composition for preventing and treating dengue fever virus is characterized by comprising the following components in parts by weight:
18-30 parts of sweet wormwood herb extract, 20-33 parts of perilla extract and 7-11 parts of trogopterus dung.
2. The Chinese medicinal composition for preventing and treating dengue fever virus according to claim 1, wherein the artemisia apiacea extract is prepared by the following steps:
grinding and crushing whole fresh or dry sweet wormwood herb, adding an alkaline soak solution with the mass being 2-3 times of that of the fresh or dry sweet wormwood herb, soaking for 5-6 hours, pulping, standing and filtering to obtain sweet wormwood herb filtrate;
and carrying out evaporative crystallization on the sweet wormwood herb filtrate, wherein the evaporative crystallization temperature is 30-40 ℃, and obtaining the sweet wormwood herb extract.
3. The traditional Chinese medicine composition for preventing and treating dengue fever virus according to claim 2, wherein the alkaline soak solution is a sodium carbonate solution with a concentration of 0.3-0.4%.
4. The traditional Chinese medicine composition for preventing and treating dengue fever virus according to claim 1, wherein the perilla extract is prepared by the following steps:
drying the whole perilla herb or fresh perilla leaf at 50-60 ℃ for 36-48 hours, taking out and crushing to obtain crushed perilla;
performing ultrasonic extraction by using water as an extractant, wherein the ultrasonic power is 360-500 w, the weight ratio of the crushed perilla to the water is 1: 10-15, the temperature is 85-90 ℃, the time is 90-120 min, filtering the crushed perilla by using a diatomite filter, and then performing reduced pressure concentration at the temperature of below 60 ℃ until the relative density is 1-1.15 to obtain a perilla extract concentrated solution;
adjusting the ethanol concentration of the concentrated perilla extract liquid with 70% ethanol to ensure that the ethanol volume is 6-7% to obtain the perilla extract liquid.
5. The traditional Chinese medicine composition for preventing and treating dengue fever virus as claimed in claim 4, wherein the ultrasonic extraction is carried out by using water as an extractant, the ultrasonic power is 360-500 w, the weight ratio of the crushed perilla to water is 1: 10-15, the temperature is 85-90 ℃, the time is 90-120 min, the crushed perilla is filtered by a diatomite filter and then is concentrated under reduced pressure at the temperature below 60 ℃ until the relative density is 1-1.15, and the step of obtaining the concentrated perilla extract solution specifically comprises the following steps:
performing ultrasonic extraction with water as extractant at ultrasonic power of 450w, the weight ratio of the crushed perilla to water is 1:12, the temperature is 90 ℃, the time is 100min, filtering with a diatomite filter, and concentrating under reduced pressure at below 60 ℃ until the relative density is 1-1.15 to obtain the concentrated perilla extract.
6. The traditional Chinese medicine composition for preventing and treating dengue fever virus according to claim 1, which comprises the following components in parts by weight:
18-25 parts of sweet wormwood herb extract, 20-27 parts of perilla extract and 10-11 parts of trogopterus dung.
7. The traditional Chinese medicine composition for preventing and treating dengue fever virus according to claim 6, which comprises the following components in parts by weight:
19 parts of sweet wormwood herb extract, 25 parts of perilla extract and 11 parts of trogopterus dung.
8. The traditional Chinese medicine composition for preventing and treating dengue fever virus according to claim 1, wherein the traditional Chinese medicine composition for preventing and treating dengue fever virus is prepared into a tablet, an oral liquid or a capsule preparation.
9. The method for preparing the Chinese medicinal composition for preventing and treating dengue fever virus according to any one of claims 1 to 8, which comprises the following steps:
weighing the components according to the formula of the traditional Chinese medicine composition for preventing and treating dengue fever virus as claimed in any one of claims 1 to 8 for later use;
adding 90-95% ethanol in an amount which is 13-14 times the mass of the trogopterus dung, heating, refluxing, extracting, cooling to room temperature, filtering, collecting an ethanol extract, concentrating to 10-12% of the original volume, and performing spray drying to obtain a trogopterus dung extract;
mixing the sweet wormwood herb extract, the perilla herb extract and the trogopterus dung extract to obtain the traditional Chinese medicine composition for preventing and treating the dengue fever virus.
10. The method for preparing a Chinese medicinal composition for preventing and treating dengue fever virus according to claim 9, wherein the step of adding ethanol with a mass concentration of 90-95% in an amount 13-14 times the mass of trogopterus dung into the trogopterus dung, heating and refluxing for extraction, cooling to room temperature, filtering, collecting an ethanol extract, concentrating to 10-12% of the original volume, and spray-drying to obtain the trogopterus dung extract comprises the following steps:
