"humanized antibody" refers to an antibody that comprises CDR regions derived from a non-human antibody, and the remainder of the antibody molecule is derived from one (or more) human antibody. Furthermore, to preserve binding affinity, some residues of the backbone (referred to as FR) segment may be modified.
The term "monoclonal antibody" refers to a preparation of antibody molecules having a single molecular composition. Monoclonal antibody compositions exhibit a single binding specificity and affinity for a particular epitope.
The medicament contains at least one functional component and also comprises a medicinal carrier. Preferably, the pharmaceutically acceptable carrier is water, aqueous buffered solutions, isotonic saline solutions such as PBS (phosphate buffered saline), glucose, mannitol, dextrose, lactose, starch, magnesium stearate, cellulose, magnesium carbonate, 0.3% glycerol, hyaluronic acid, ethanol, or polyalkylene glycols such as polypropylene glycol, triglycerides, and the like. As used herein, "CDR regions" or "CDRs" refer to the hypervariable regions of the heavy and light chains of an immunoglobulin, as defined by Kabat et al (Kabat et al, Sequences of proteins of immunological interest,5th ed., u.s.department of Health and Human Services, NIH,1991, and later versions). There are three heavy chain CDRs and three light chain CDRs. As used herein, the term CDR or CDRs is intended to indicate one of these regions, or several or even all of these regions, which comprise the majority of the amino acid residues responsible for binding by the affinity of the antibody for the antigen or its recognition epitope, as the case may be.
The invention provides an antibody humanization transformation method, which carries out reasonable antibody humanization design by FR transplantation on a reference multi-template to obtain a humanized antibody with the affinity equivalent to that of a murine antibody.
The preparation method of the humanized anti-A beta monoclonal antibody provided by the invention comprises the following steps:
step 1: preparing mouse hybridoma, and obtaining an antibody sequence through 5' RACE;
step 2: humanization of the antibody, sequence alignment on an NCBI tool, completion of humanization modification, and screening of the modified antibody.
Specifically, the method comprises the following steps:
the preparation method of the humanized anti-Abeta monoclonal antibody comprises the following steps: using a mouse antibody 066-7.17.2 as a template, performing PCR amplification to obtain an antibody heavy chain variable region gene VH and a light chain variable region gene VL, translating the genes into amino acid sequences, comparing the amino acid sequences with human antibody sequences in an NCBI database, respectively selecting 5 human antibody sequences with the highest similarity to the variable region VH and VL, using the sequences as a humanized transformation reference template, determining a CDR region of the mouse antibody 066-7.17.2, keeping the CDR region unchanged, transplanting FR regions in the 5 reference templates of the VH and VL respectively to 066-7.17.2 to obtain humanized sequences, and respectively constructing transient expression vectors after codon optimization; transferring the expression vector into 293E cells for expression to obtain a humanized antibody specifically combined with A beta; the humanized A beta antibody is finally obtained by affinity determination, determination of the EC50 value bound with the antigen, activity detection of macrophage phagocytosis A beta and cytotoxic protection detection of the humanized antibody.

The invention discloses a humanized anti-Abeta monoclonal antibody and application thereof, and a person skilled in the art can realize the humanized anti-Abeta monoclonal antibody by properly improving process parameters by referring to the content. It is expressly intended that all such similar substitutes and modifications which would be obvious to one skilled in the art are deemed to be included in the invention. While the methods and applications of this invention have been described in terms of preferred embodiments, it will be apparent to those of ordinary skill in the art that variations and modifications in the methods and applications described herein, as well as other suitable variations and combinations, may be made to implement and use the techniques of this invention without departing from the spirit and scope of the invention.

2. Positive control antibody expression vector construction
pGS003-hIgG1CH and pGS003-hIgKCL are respectively selected as expression vectors for constructing heavy chains and light chains of anti-human Abeta positive antibodies (066-P01: Solanezumab, Eli lily, 066-P02: Aducanum ab, Biogen), after codon optimization of amino acid sequences of variable regions of the positive antibodies, genes of the VH and VL of the positive antibodies are respectively cloned to pGS003-hIgG1CH and pGS003-hIgKCL by using an enzyme digestion method to obtain the transient expression vectors of the heavy chains and the light chains of the positive antibodies, namely pGS003-066-P02VH-hIgG1CH and pGS-066-P02 VL-hIgKCL. 3. instantaneous expression
pGS003-066-P01VH-hIgG1CH and pGS003-066-P01 VL-hIgKCL;
pGS003-066-P02VH-hIgG1CH and pGS003-066-P02VL-hIgKCL were transiently expressed.
Using FreeStyleTM293E cells in Freestyle medium for transient expression. 24 hours before transfection, 0.5X 10 inoculum was inoculated in 125ml Erlenmeyer flasks630ml of 293E cells per ml, 5% CO at 37 ℃2Shaking the culture in an incubator at 130 rpm. 4. Protein purification

EXAMPLE 2 anti-A.beta.monoclonal hybridoma preparation
1. Immunization of BALB/c mice
Mixing A beta1-42The polypeptide antigen and Freund's complete adjuvant are mixed by vortex according to dosage, and BALB/c female mice of 6 weeks old are immunized for the first time after complete emulsification. Each mouse was injected intraperitoneally with 200. mu.g of antigen, and 3 groups of mice were immunized in total, with 5 mice each. Two weeks after the first immunization, the mice were subjected to a second intraperitoneal immunization using Freund's incomplete adjuvant, and the immunization antigen doses were the same for the first immunization. Mice were immunized intraperitoneally twice a month thereafter, with the same adjuvant and antigen doses as for the second immunization.
And (3) after the first immunization, performing orbital small-amount blood collection on the mice every six weeks, performing serum titer detection, detecting the serum titer reaches 1:200000 or more by an indirect ELISA method, and performing boosting immunization on the mice for fusion.
2. Preparation of myeloma cells for fusion
Myeloma cells P3X63Ag8.653 for fusion were recovered three weeks in advance and cultured for two weeks in DMEM medium containing 1X 8-azaguanine and 10% fetal bovine serum, and cultured in DMEM medium containing 10% fetal bovine serum for one week of fusion, maintaining the density of P3X63Ag8.653 at 70% -80% until the day of fusion.
3. Cell fusion and HA screening
Acquisition and preparation of splenocytes: 2 mice after boosting were taken, sacrificed after immune serum was collected and soaked in 75% alcohol for 2-3 minutes. The skin and peritoneum were cut from the ventral side of the immunized mice, exposing the spleen. Removing peripheral tissues by using a tip to obtain a spleen, grinding by using a grinding rod, and filtering by using a cell screen to prepare a single cell suspension. Centrifuging and then discarding the supernatant,
cell fusion pretreatment: P3X63Ag8.653 in the culture flask was collected, centrifuged at 1000rpm/5min, the supernatant was discarded, and viable myeloma cells were counted after resuspension. Centrifuging the spleen cell suspension, removing supernatant, adding ACK lysate, incubating, centrifuging, removing red blood cells, resuspending with DMED, and counting spleen viable cells.
Cell fusion: according to the ratio of splenocytes: mixing cells at a ratio of P3X63Ag8.653 to 1:2, centrifuging at 2000rpm/5min, discarding supernatant, shaking off cell pellet, and mixing at 400 μ l/1 × 108Adding 1mg/ml Pronase into splenocytes, incubating for 15 seconds, adding 10ml fetal calf serum to stop reaction, adding electrotransfer solution (ECF) to 50ml, centrifuging at 2500rpm for 5 minutes, removing supernatant, resuspending ECF, counting viable cells, adjusting the density of splenocytes to 2 × 106And/ml. Adding the cell suspension with the adjusted density into an electric fusion tank, operating an electric transfer instrument to perform cell fusion, transferring the cell suspension from the fusion tank into 1/2HA culture medium after the fusion is finished, and performing cell plating after standing for 3 hours.
HA medium screening: AT selection medium containing 1/2HA, 1 × QINGNIN-streptomycin, 20% fetal bovine serum and 80% DMEM medium was prepared. The murine hybridoma cells were resuspended in the 1/2HA screening medium and mixed well. At a volume of 200. mu.l/well, 1X 106Spleen cells/plate, add cell suspension to 96-well cell culture plate, and culture in 37 ℃ cell culture box. After 1 week of culture, the cells were first changed with 1/2HA medium and placed in a 37 ℃ cell culture box for further culture, and after 3 days of culture, the cells were changed with 1/2HA medium for the second time.
4. Screening of Positive cell lines
2 weeks after the fusion, cell supernatants were taken for ELISA assay to detect cell supernatants and human Abeta1-42Screening out cells with positive ELISA result, and retesting the ELISA result for the second time. And taking the cell supernatant with positive retest result for subcloning and expanding culture.
5. Expanding culture
Transferring the cell strain with positive ELISA detection into 24-hole from 96-hole for culture, and transferring into 25cm after full growth2Culturing in a culture flask.
6. Subcloning by limiting dilution method
Positive of mixing by blowing and beatingCell lines and small amounts were aspirated for viable cell counting. About 200 cells were pipetted into 80ml of complete medium and mixed well and plated in 4 plates. About 400 additional cells were pipetted into 80ml of complete medium and mixed well and plated in 4 plates. About 1000 additional cells were pipetted into 20ml of complete medium and mixed well and plated in 1 block. 7. Clone detection and expanded culture

Antigen: after washing the plate three times with PBST, biotinylated Abeta was taken separately1-42、Aβ1-16、Aβ14-29The polypeptides, diluted to 1. mu.g/ml with PBS (pH 7.2), were added to 96 wells of an ELISA plate at 50. mu.l per well and incubated at room temperature for 1 hour.
Adding a primary antibody: after washing the plate three times with PBST, the murine candidate antibody was added at 50. mu.l/well and incubated at room temperature for 2 hours.
Adding a secondary antibody: after washing the plate three times with PBST, anti-mouse IgG Fc-HRP antibody, 1:5000 dilution, 50. mu.l/well was added and incubated at room temperature for 1 hour.
Color development: after washing the plate six times with PBST, 50. mu.l of TMB developing solution was added to each well, and the plate was developed in the dark at room temperature for 10 minutes.
And (4) terminating: stop solution was added directly to stop the reaction, 50. mu.l per well.
And (3) detection: immediately placing the ELISA plate into an ELISA reader after the reaction is terminated, measuring the OD value at 450nm, and storing the original data.
Data processing: the raw data were input into the software SoftMaxPro 6.2.1 for data processing. Specific data are shown in the table1. The results showed that 12 murine candidate antibodies contained three different epitopes, N-terminal (A β) respectively1-16) Terminal C (Abeta)30-42) Intermediate (Abeta.) of14-29) A peptide fragment. 8. Identification of subclasses
Coating goat anti-mouse IgG1, IgG2a, IgG2b, IgG2c, IgG3, IgM and IgGA, 50ng/100 mu l/hole, keeping overnight at 4 ℃, blocking BSA at room temperature, adding cell supernatant to be detected, incubating for 2 hours at room temperature, adding enzyme-labeled secondary goat anti-mouse IgG, kappa and lambda, developing, stopping, reading at 450nm, judging the subclass of the cell strain to be IgG1, kappa or IgG2a, kappa or IgG2b, and the result is shown in Table 2. Wherein, the heavy chain constant region of 066-7.17.2 is mouse IgG1, and the light chain constant region is the constant region of mouse kappa chain.

9. Cell cryopreservation
Preparing a frozen stock solution: 90% fetal bovine serum, 10% DMSO.

11. Antibody preparation by in vitro culture method
Reviving the prepared hybridoma cell strain by reviving a DMEM medium containing 10% fetal calf serum and 1% streptomycin, culturing by using a small bottle, carrying out subculture expansion after the confluence degree of the cells is about 90%, expanding the cells until the cell culture supernatant is about 200ml, collecting the supernatant, carrying out centrifugal filtration, and purifying.
Example 3 anti-Abeta monoclonal antibody inhibits Abeta polymerization assay
The A beta dry powder was dissolved with 8.2% DMSO/DPBS solution (DMSO: sigma; DPBS: Hyclone) to 1mg/ml, the DPBS diluted the A beta solution to 33. mu.g/ml, the anti-A beta monoclonal antibodies 066-4.6.8, 066-4.18.2, 066-4.22.1, 066-4.26.14, 066-5.4.1, 066-6.1.1, 066-6.1.3, 066-6.2.1, 066-6.7.2 to 450. mu.g/ml (IC100), and the ThT (sigma) diluted with ultrapure water to 20. mu.M. 50 μ l of the antibody dilution was added to a 96-well blackboard (corning), and then 50 μ l A β dilution was added thereto, and finally 100 μ l ThT was added thereto, and incubated at room temperature in the dark for 24 hours, and the fluorescence intensity was measured with a multifunctional microplate reader (Ex/Em ═ 440/485). The abscissa represents different sample application groups, and the ordinate represents relative fluorescence intensity, and the results are shown in FIG. 3. In FIG. 3(A), when the relative fluorescence intensity of the IgG group was 1.0, the relative fluorescence intensity of the anti-A β monoclonal antibody 066-5.4.1 group was 0.59; in FIG. 3(B), when the relative fluorescence intensity of the PBS group was 1.0, the relative fluorescence intensity of the anti-A β monoclonal antibody 066-4.22.1 group was 0.44, and the relative fluorescence intensity of the anti-A β monoclonal antibody 066-4.26.14 group was 0.46; it can be seen that anti-A beta monoclonal antibodies 066-4.22.1, 066-4.26.14, 066-5.4.1, etc. all inhibit A beta polymerization.
Example 4 detection of macrophage phagocytosis of A.beta.Activity by anti-A.beta.monoclonal antibodies

Wherein, when the relative value of LDH release of the Vehicle group is 1.0, the relative fluorescence intensity of the anti-A beta monoclonal antibody 066-4.26.14 group is 1.2, the relative value of LDH release of the anti-A beta monoclonal antibody 066-5.4.1 group is 1.37, the relative value of LDH release of the anti-A beta monoclonal antibody 066-6.1.1 group is 1.43, the relative value of LDH release of the anti-A beta monoclonal antibody 066-6.1.3 group is 1.35, the relative value of LDH release of the anti-A beta monoclonal antibody 066-6.2.1 group is 1.34, the relative value of LDH release of the anti-A beta monoclonal antibody 066-6.7.2 group is 1.44, and the relative values of LDH release of the positive control antibody 066-P02 group are 1.26(A) and 1.53 (B). It can be seen that the 066-4.26.14, 066-5.4.1, 066-6.1.1, 066-6.1.3, 066-6.2.1, 066-6.7.2 and 066-7.17.2 antibodies all have cytotoxic protection effects, and the protection effects are equivalent to those of 066-P02.
Example 6Morris Water maze experiment
1. The experimental method and the steps are as follows:
experimental animals 3 × Tg mice were purchased from Experimental animals technical Co., Ltd, Kyoto Jilin university medical college, Peking. The grouping situation is as follows:
according to different injection medicines, the injection medicines are divided into an antibody 066-4.26.14 treatment group, an antibody 066-5.4.1 treatment group, an antibody 066-7.17.2 treatment group, a 3 XTg blank control group, a wild type PBS injection group and a positive antibody 066-P02 control group, wherein each group contains 8 antibodies.
Reference (Nabeshima, 2007) was made to the method of intraperitoneal injection of 6-month-old male 3 × Tg mice (500 g/mouse) once a week for 10 weeks into 6-month-old male 3 × Tg mice (mab 066-4.26.14, 066-5.4.1, 066-7.17.2), and after 8 weeks of injection, Morris water maze assay was performed.
Morris water maze detection step:
(1) a specially-made water maze is designed, and mainly comprises a cylindrical pool and a platform with a movable position. The height of the water pool is 45cm, the diameter of the water pool is 100cm, the diameter of the platform is 9cm, the height of the platform is 15-40cm, and the water pool is connected with a computer through a digital camera.
(2) The method comprises the steps of injecting clear water into a water pool in advance, wherein the wall and the bottom of the water pool are black, adding white pigment for food into the water of the water pool, so that a platform below the water surface cannot be seen by a mouse, the water depth is 30cm, and the water surface is 1cm higher than the platform.
(3) The water temperature was controlled at 19 + -1 deg.C and the water entry point was marked in the quadrant outside the quadrant of the platform on the basin. And different-shaped markers are stuck on the corresponding side walls of each quadrant. The platform position was kept constant during the experiment.
(4) Each experiment is carried out in a sound-proof room, and the positions of objects in a laboratory such as a water pool, a light source, a mouse cage and the like are kept unchanged.
(5) Training was started on day 3 after week 8 dosing, and the experiment was continued for a total of 5 days (water maze-Hidden platform test), with 4 measurements per day. When the mouse is put into water, the face of the mouse faces the wall surface of the pool, and the mouse is gently put into the water. The 5 training (experiments) per day were performed randomly in the area outside the set-up platform quadrant, with the first two times of the first two days of the experiment as exercises. The platform was found within 60 seconds by the mouse and left on the platform for 15 seconds, and the platform was not found within 60 seconds by the mouse (latency was recorded as 60 seconds), and then the experimenter led it to the platform and left on the platform for 15 seconds. The average of four incubation periods of the mice was taken as the performance of the mice on the day.
2. Experimental results (see fig. 6):
on day 2 of the Hidden platform test, the time for all antibody administration groups to find the Hidden platform under the water surface (escape latency) was significantly shortened in comparison with the 3 × Tg blank group, which was statistically significant. Therefore, the anti-Abeta monoclonal antibodies 066-7.17.2 and 066-4.26.14 have certain functions of improving the cognitive learning and memory ability of the mouse model with the Alzheimer dementia.
EXAMPLE 7 monoclonal antibody Gene sequencing and chimeric antibody preparation
1. Monoclonal antibody gene sequencing
After immunization, fusion and monoclonality, a 066-7.17.2 monoclonal antibody cell strain is selected based on the experimental results of binding epitope, macrophage phagocytosis Abeta activity detection, cytotoxic protection activity detection, Morris water maze and the like to extract total RNA, and is reversely transcribed into cDNA, and then the heavy chain variable region and the light chain variable region of the antibody are amplified by PCR by taking the cDNA as a template.
Total RNA extraction of the 066-7.17.2 monoclonal antibody cell line was performed using the TRIZOLREAgent kit (15596-026) from Invitrogen, as described in the manual, and the results are shown in FIG. 7.
Then, 5 ' RACE FULL kit (D315) from Takara was used, the total RNA was used as a template, random primers in the kit were reverse-transcribed to first strand cDNA, then heavy chain primers mIgGR (5 ' -CTCAGGGAARTARCCYTTGAC-3 ', SEQ ID NO:42) designed as a constant region and linker primers in the kit were PCR-amplified, and light chain primers mIgKR (5'-TCACTGCCATCAATCTTCCAC-3', SEQ ID NO:43) designed as a constant region and linker primers in the kit were PCR-amplified, and the results are shown in FIG. 8.
PCR fragments recovered by an agarose gel recovery kit are subjected to TA cloning, then single clones are selected for PCR identification, identification primers are M13F (5'-TGTAAAACGACGGCCAGT-3', SEQ ID NO:44) and M13R (5'-CAGGAAACAGCTATGACC-3', SEQ ID NO:45), and a part of samples selected from correct strains are identified and sent to Invitrogen for sequencing. pGS003-066-7.17.2-chAbVH-hIgG4CH and pGS003-066-7.17.2-chAbVL-hIgKCL were transiently expressed.
Using FreeStyleTM293E cells in Freestyle medium for transient expression. 24 hours before transfection, 0.5X 10 inoculum was inoculated in 125ml Erlenmeyer flasks630ml of 293E cells per ml, 5% CO at 37 ℃2Shaking the culture in an incubator at 130 rpm. Example 8 antibody humanization
Selecting a murine antibody 066-7.17.2 for humanization, wherein the humanization process comprises searching and transplanting mainly for a human template.
The main goal of humanization engineering is the FR sequence in the variable region. The amino acids of the VH and VL of the murine antibody 066-7.17.2 are used as templates, sequence alignment is carried out on NCBI website, 5 humanized reference sequences are found, and the humanized modified sequences are designed as the reference templates for humanized modification of the FR regions of the antibody.
The specific sequences of the CDR regions are shown in Table 4, and the sequences of the humanized antibody after grafting of the remaining FR regions are shown in Table 5.

Example 9 preparation of humanized full-length antibody of A.beta.
1. Construction of full-Length antibody transient transfection expression vector
pGS003-hIgG1CH and pGS003-hIgKCL were selected as expression vectors for constructing heavy and light chains of anti-human Abeta full-length antibodies, respectively. The 066-7.17.2 humanized antibody sequence was codon optimized, PCR amplified, with Hind III and Nhe I for the heavy chain and Hind III and Nar I for the light chain, and 5 VH and 5 VL antibody genes cloned into pGS003-hIgG1CH and pGS003-hIgKCL, respectively, as shown in Table 6. After sequencing to identify the correct insertion of the antibody gene, the recombinant expression vector was transformed into E.coli TOP 10F', and a single colony was picked and inoculated into LB medium containing 100. mu.g/ml ampicillin and cultured with shaking at 37 ℃ for 16 hours. 3. Purification and detection of antibodies

The supernatant was filtered through a 0.22 micron filter, then passed through a MabSelect Sure affinity chromatography column (GE), 20mM citric acid-sodium citrate, pH 3.0, eluted, pH adjusted to neutral with 1M Tris base, and adjusted to an isotonic solution by addition of 10 XPBS. Protein was purified by SDS-PAGE using 4-20% gradient gel (Biotech, GmbH, King-Shirui, Nanjing), and the results are shown in FIG. 10.
Example 10 determination of humanized candidate antibody EC50 values
Coating: the mixture of human Abeta 42 monomer and polymer is diluted to 1 mu g/ml by CBS (pH 9.4), added into 96 wells of an ELISA plate with 50 mu l per well, and incubated overnight at 2-8 ℃.
And (3) sealing: after three washes with PBST, the plates were blocked with 3% BSA and incubated at 200. mu.l/well for 1 hour at 25 ℃.
Sample treatment: humanized candidate antibody and chimeric antibody were each diluted 2-fold with a gradient of 10. mu.g/ml as the starting concentration (2)0~2-11) 50 μ l/well, incubated for 1h at 25 ℃.
Adding an antibody: after washing the plate four times with PBST, anti-human IgG (H + L) -HRP antibody, 1:5000 dilution, 50. mu.l/well, was added and incubated for 1H at 25 ℃.
Color development: after washing the plate four times, adding TMB developing solution, 50 μ l per well, and developing for 3 minutes in the dark at room temperature.
And (4) terminating: stop solution was added directly to stop the reaction, 50. mu.l per well.
And (3) detection: immediately placing the ELISA plate into an ELISA reader after the reaction is terminated, measuring the OD value at 450nm, and storing the original data.
Data processing: the raw data were input into the software SoftMax Pro 6.2.1 for data processing. The data are shown in Table 9. The results showed that 6 humanized candidate antibodies had binding ability to human a β comparable to that of the chimeric antibody.

EXAMPLE 11 determination of KD values for humanized candidate antibodies
Biacore-T200 detection is applied, a ProteinA chip is used for capturing candidate antibodies or positive antibodies, human Abeta antigens with different concentrations flow through the chip, and fitting analysis is carried out according to collected data. Antigen samples were diluted in a 2-fold concentration gradient using HBS-EP + Buffer to 400nmol/L, 200nmol/L, 100nmol/L, 50nmol/L, 25nmol/L, 12.5nmol/L, 6.25nmol/L, 3.125nmol/L, 1.56nmol/L, 0.78nmol/L, 0nmol/L concentration gradient solutions. Samples were tested at a repeat concentration of 25 nmol/L. Example 12 detection of macrophage phagocytosis of A.beta.Activity by humanized A.beta.antibody
Mouse primary abdominal cavity macrophage with good adherence for 3 days, digesting with 0.25% trypsin, counting, and adjusting cell density to 2 × 10 with DMEM culture medium (Gibco) containing 10% serum fetal bovine serum5Seed 96-well cell culture plate in 100. mu.l/well; a working solution was prepared by diluting 066-7.17.2 humanized candidate antibody to 20. mu.g/ml in DMEM medium containing 1% fetal bovine serum, diluting A.beta.to 240. mu.g/ml, and diluting ThT (sigma) to 20. mu.M in ultrapure water. Discarding the culture medium in the culture plate, adding 50 μ l of candidate antibody diluent, adding 50 μ lA β diluent, and setting multiple wells; 37 ℃ and 5% CO2Culturing for 6 hours in an incubator; 50. mu.l of the supernatant was put into a 96-well blackboard, and 50. mu.l of ThT was added thereto, and the fluorescence intensity was measured by a multifunctional microplate reader (Ex/Em. 440/485). The abscissa represents the different sample addition groups, and the ordinate represents the fluorescence intensity, and the results are shown in FIG. 11. The relative fluorescence intensity of hIgG was 1.00, that of 066-7.17.2-mAb group was 0.47, that of 066-7.17.2-chop group was 0.54, that of 066-7.17.2H1L5 group was 0.51, that of 066-7.17.2H2L3 group was 0.55, that of 066-7.17.2H2L5 group was 0.63, that of 066-7.17.2H3L5 group was 0.44, that of 066-7.17.2H4L5 group was 0.44, and that of 066-7.17.2H5L5 group was 0.42. It can be seen that the 066-7.17.2 humanized candidate antibodies all have the activity of promoting macrophage phagocytosis of A beta.
Example 13 assay of humanized A β antibody cytotoxic protective Activity
Logarithmic growth phase SHSY5Y cells, digested with 0.25% trypsin, counted, and adjusted to 3X 10 cell density with EMEM culture medium (ATCC) containing 10% fetal bovine serum4Seed 96-well cell culture plate in 100. mu.l/well; the humanized candidate antibody was diluted 066-7.17.2 to 200. mu.g/ml (IC100) in EMEM medium with 1% fetal bovine serum as a working solution, and A.beta.was diluted to 240. mu.g/ml. Discarding the culture medium in the culture plate, adding 50 μ l of candidate antibody diluent, adding 50 μ l A β diluent, and setting multiple wells; culturing at 37 deg.C in 5% CO2 incubator for 48 hr; adding 50 μ l of supernatant into a new 96-well plate, and addingAdding 50 μ l LDH assay buffer, keeping out of the sun at room temperature for 30min, adding 50 μ l stop solution, and detecting the light absorption value with a multifunctional microplate reader. The abscissa is the different loading groups and the ordinate is the relative value of LDH release, and the results are shown in FIG. 12. The relative value of LDH release of Vehicle is 1.00, the relative value of LDH release of 066-P02 group is 1.47, the relative value of LDH release of 066-7.17.2-mAb group is 1.43, the relative value of LDH release of 066-7.17.2-ChAb group is 1.40, the relative value of LDH release of 066-7.17.2H1L5 group is 1.30, the relative value of LDH release of 066-7.17.2H2L3 group is 1.52, the relative value of LDH release of 066-7.17.2H2L5 group is 1.29, the relative value of LDH release of 066-7.17.2H3L5 group is 1.00, the relative value of LDH release of 066-7.17.2H4L5 group is 1.31, and the relative value of LDH release of 066-7.17.2H5L5 group is 1.41. It can be seen that the 066-7.17.2 humanized candidate antibodies all have cytotoxic protection effects, and the protection effects are equivalent to 066-P02, wherein 066-7.17.2H3L5 performs optimally.

Claims (19)

1.A humanized anti-A beta monoclonal antibody characterized in that,
the amino acid sequences of CDR1-3 of the heavy chain of the monoclonal antibody are respectively shown as SEQ ID NO 1, 2 and 3; and the amino acid sequences of CDR1-3 of the light chain of the monoclonal antibody are respectively shown as SEQ ID NO. 4, 5 and 6.
2. The monoclonal antibody of claim 1,

17. Medicament comprising a monoclonal antibody according to any one of claims 1 to 8, a conjugate according to claim 13 and/or a conjugate according to claim 14.
18. Use of a monoclonal antibody according to any one of claims 1 to 8, a conjugate according to claim 13 and/or a conjugate according to claim 14 for the preparation of a product for detecting expression of a β.
19. A kit comprising a monoclonal antibody according to any one of claims 1 to 8, a conjugate according to claim 13 and/or a conjugate according to claim 14.
