Abstract
本发明提供了卷曲乳酸杆菌用于制备女性避孕药物的应用。该技术方案首先研究了卷曲乳酸杆菌对精子的影响,分别采用体外共孵育和动物阴道给药的方式进行实验;实验结果表明,卷曲乳酸杆菌对精子具有明显的粘附能力,使精子活力及泳动能力得到显著下降。基于以上所发现的卷曲乳酸杆菌的新性质,确定了利用其制备女性避孕药物的新用途。动物实验表明,将含有卷曲乳酸杆菌菌液的阴道栓剂作用于雌鼠阴道,可确切降低其受孕率,避孕率可达50%左右。本发明中卷曲乳酸杆菌菌悬液安全无毒,将其益生效果与生殖避孕相结合,突破了传统避孕方式的局限性,安全、有效且可逆,不仅可以帮助调节妇女的生殖道微生态平衡,对于女性生殖健康更具重要意义。

避孕是通过理化方法暂时避免女性受孕的技术手段。传统的避孕方法包括输精管结扎、激素类避孕药以及避孕套等,以上方法都存在一定的缺陷,例如降低性生活的体验感、避孕不彻底、影响人体激素水平等,因此寻找一种更加安全、有效、可逆、非激素类的避孕方法具有十分重要的意义。
人类微生物群与许多健康影响有关,最近研究重点已从肠道扩展到许多其他器官系统。已发表的研究表明,阴道微生物群不仅在人类健康和微生物失调方面发挥着重要作用,而且在成功受精和健康怀孕方面也发挥着重要作用。阴道微生物群以乳酸菌种类的菌群为主,乳酸菌的缺失与一些不良病症有关,包括异位妊娠、盆腔炎、不孕等影响生活质量的众多症状。而作为阴道中的重要微生物群卷曲乳酸杆菌是健康妇女生殖道的一种占主导地位的乳杆菌,对维持生殖道菌群平衡以及健康起关键作用。它能抑制多种生殖道病原细菌和真菌,例如加德纳氏菌和白色念珠菌等,也能降低HIV感染和性传播疾病的风险,是一种理想的生殖道益生菌。
此外,分子分析技术表明:精液中以厌氧菌、棒状杆菌、加德那拉菌、乳酸菌、单胞菌、链球菌和细孔菌最多。厌氧菌和假单胞菌的比例与精液健康和生育能力相关,乳酸菌的比例与精液健康相关。然而,其他类型的阴道益生菌对精子活力的作用却鲜少被研究。
发明内容
本发明旨在针对现有技术的技术缺陷,提供卷曲乳酸杆菌用于制备女性避孕药物的应用,以解决现有技术中常规避孕方法存在降低性生活的体验感、避孕不彻底、影响人体激素水平等缺陷的技术问题。
本发明要解决的另一技术问题是,如何通过微生物菌剂实现女性避孕作用。
本发明要解决的再一技术问题是,现有技术中卷曲乳酸杆菌对精子活性的影响尚不明确。
为实现以上技术目的,本发明采用以下技术方案:
卷曲乳酸杆菌用于制备女性避孕药物的应用。
作为优选,所述药物的剂型为阴道栓剂。
作为优选,所述药物中卷曲乳酸杆菌的给药浓度为该药物所处环境中精子浓度的10倍。
作为优选,所述药物与精子的作用时间不少于2h。
作为优选,所药物黏附该药物所处环境中的精子。
作为优选,所述药物降低该药物所处环境中精子的活力。
作为优选,所述药物对精子活力的降低幅度不低于95%。
作为优选,所述药物降低该药物所处环境中精子的泳动能力。
作为优选,所述药物的避孕率不低于50%。
作为优选,该应用包括以下步骤:
1)将卷曲乳酸杆菌,按新鲜培养基体积的5%接种于5mL MRS培养基中,于37℃,5%CO2恒温培养箱中培养过夜,第二日进行二次活化,计算菌液浓度;从已知浓度的卷曲乳酸杆菌菌液中吸取1mL用HS溶液重复洗涤3次后按101~109CFU/mL的比例进行梯度稀释制成卷曲乳酸杆菌悬液;
2)向作为阴道栓剂的医用吸收性明胶海绵中吸入所述卷曲乳酸杆菌悬液,即得到所述女性避孕药物。
在以上技术方案中,所述HS溶液包括以下成分:135mM NaCl,5mM KCl,1mM MgSO4,2mM CaCl2,20mM乙磺酸,5mM葡萄糖,10mM乳酸,1mM钠-丙酮酸盐;所述HS溶液的pH为7.4。
本发明提供了卷曲乳酸杆菌用于制备女性避孕药物的应用。该技术方案首先研究了卷曲乳酸杆菌对精子的影响,分别采用体外共孵育和动物阴道给药的方式进行实验;实验结果表明,卷曲乳酸杆菌对精子具有明显的粘附能力,使精子活力及泳动能力得到显著下降。基于以上所发现的卷曲乳酸杆菌的新性质,确定了利用其制备女性避孕药物的新用途。动物实验表明,将含有卷曲乳酸杆菌菌液的阴道栓剂作用于雌鼠阴道,可确切降低其受孕率,避孕率可达50%左右。
本发明以体外卷曲乳酸杆菌对精子的活力的降低作用为基础,在性成熟母鼠体内以卷曲乳酸杆菌栓塞载体给予阴道给药,同样发现较好的避孕效果。为研发卷曲乳酸杆菌避孕药物的提供了一定的实验依据。
本发明摸索了卷曲乳酸杆菌与精子的共孵育条件,在体外环境下考察了卷曲乳酸杆菌对精子的粘附性及其对精子活力和泳动能力的影响,同时通过动物实验探究了体内避孕效果。在体外实验中,将卷曲乳酸杆菌与精子共孵育,探究其粘附能力与对精子活力的影响。动物实验中,利用阴道给菌的方式对卷曲乳酸杆菌的避孕效果进行测定与评价,并考察了对怀孕率的影响。
本发明的实验方法如下:
1、以MRS培养基活化卷曲乳酸杆菌:
从-80℃冰箱取出卷曲乳酸杆菌甘油保藏管,按新鲜培养基体积的5%接种于5mLMRS培养基中,于37℃,5%CO2恒温培养箱中培养过夜,第二日进行二次活化,测OD计算菌液浓度(CFU/mL)。从已知浓度的卷曲乳酸杆菌菌液中吸取1mL用HS溶液重复洗涤3次后按101~109的比例进行梯度稀释制成菌悬液备用。
2、体外探究卷曲乳酸杆菌对精子活力及泳动能力的影响:
(1)卷曲乳酸杆菌对精子的粘附能力评价:
将精子与上述卷曲乳酸杆菌的菌悬液按照1:10的浓度比在HS溶液中混匀,于37℃、5%CO2的培养箱中共孵育2h。加入1mL无菌PBS以离心-震荡方式洗涤3次后再加入1mL无菌PBS重悬。吸取10μL液体滴于载玻片上,革兰氏染色,显微镜统计每100个精子上粘附的卷曲乳酸杆菌数目。
(2)卷曲乳酸杆菌对精子的活力影响检测:
同步骤(1)完成精子与卷曲乳酸杆菌的共孵育及洗涤,再使用CASA(computer-assisted sperm analysis system)仪器检测共孵育2h后的精子活力。各项检测参数包括:前向运动能力、总精子活力、平均路径速度。
(3)卷曲乳酸杆菌对精子在模拟阴道中泳动能力的影响:
同步骤(1)完成精子与卷曲乳酸杆菌的共孵育及洗涤,再以内有甲基纤维素的毛细管模拟女性阴道,将毛细血管插入共孵育体系中,于37℃、5%CO2的培养箱中继续共孵育1h。取出毛细管,于显微镜下观察距离毛细血管底部1cm和2cm处的两个区域多个视野下的平均精子数目。
3、小鼠实验体内探究卷曲乳酸杆菌在避孕中的作用:
通过阴道栓塞给药的方式,利用医用吸收性明胶海绵蘸取50μL的卷曲乳酸杆菌菌悬液塞入性成熟SD雌鼠阴道口,使卷曲乳酸杆菌进入大鼠阴道并在精子通过阴道时发挥粘附作用,以实现细菌性避孕效果。
本发明在体外将卷曲乳酸杆菌与精子进行共孵育,经测定粘附至精子上的卷曲乳酸杆菌可大大降低精子活力(下降95%左右),同时显著降低精子在模拟女性阴道中的泳动能力。此外,在小鼠实验中,以阴道给菌方式为性成熟母鼠提供卷曲乳酸杆菌,其避孕率可达50%左右。
本发明利用卷曲乳酸杆菌对精子活力的降低作用开拓了避孕的新方法。本发明中卷曲乳酸杆菌菌悬液安全无毒,将其益生效果与生殖避孕相结合,突破了传统避孕方式的局限性,安全、有效且可逆,不仅可以帮助调节妇女的生殖道微生态平衡,对于女性生殖健康更具重要意义。

具体实施方式
以下将对本发明的具体实施方式进行详细描述。为了避免过多不必要的细节,在以下实施例中对属于公知的结构或功能将不进行详细描述。以下实施例中所使用的近似性语言可用于定量表述,表明在不改变基本功能的情况下可允许数量有一定的变动。除有定义外,以下实施例中所用的技术和科学术语具有与本发明所属领域技术人员普遍理解的相同含义。
以下实验中所采用的MRS液体培养基、实验动物均为常规生化实验材料,可自市面购得。
实施例1(卷曲乳酸杆菌与精液的获取)
一、卷曲乳酸杆菌的获取
取健康女子阴道分泌物,要求受试者近期无阴道激素、抗生素用药记录,一周内无宫颈治疗记录,5天内无冲洗记录,48小时内无性生活记录,通过涂平板、测序等方法筛选出阴道卷曲乳酸杆菌进行后续实验。
二、精液的获取
取健康男子精液,要求受试者精子各项参数正常(包括精子大小、精子数量、活动精子、非活动精子、精子形态)且核型正常。取样前,用肥皂和水清洗龟头,精液通过受试者手淫获得,射入无菌收集管中,37℃孵育25-45分钟液化。
实施例2(体外探究卷曲乳杆菌对精子的影响)
一、以MRS培养基活化卷曲乳酸杆菌
(1)从-80℃冰箱取出卷曲乳酸杆菌甘油保藏管,按新鲜培养基体积的5%接种于5mL MRS培养基中,于37℃,5%CO2恒温培养箱中培养过夜;
(2)吸取(1)中培养液,按新鲜培养基体积的5%接种5mL MRS培养基中,进行卷曲乳酸杆菌的二次活化以确保细菌良好的生理状态;
(3)将步骤(2)中已经二次活化的卷曲乳酸杆菌混匀取1mL菌液,6000rpm/min离心3min后弃上清,加入1mL HS溶液重悬菌泥,充分震荡后再次离心弃上清,再以HS溶液重悬。重复洗涤3次,最后以1mL HS溶液按101~109的比例进行梯度稀释制成菌悬液备用。
二、卷曲乳酸杆菌与精子共孵育条件的确定
取不同浓度梯度的卷曲乳酸杆菌的菌悬液分别与精子按照1:1的体积比进行共孵育,每隔30min使用CASA仪器检测精子的活性,确定其最佳共孵育时间以及菌悬液的浓度,最终得到最佳共孵育时间为2h,卷曲乳酸杆菌与精子浓度比为1:10。
三、体外探究卷曲乳酸杆菌对精子活力及泳动能力的影响
(1)卷曲乳酸杆菌对精子的粘附能力评价:
将精子与上述卷曲乳酸杆菌的菌悬液按照1:10的浓度比在HS溶液中混匀,于37℃、5%CO2的培养箱中共孵育2h。加入1mL无菌PBS,3000rpm/min离心5min,弃掉上清。重复操作洗涤3次以洗去未粘附至精子上的卷曲乳酸杆菌,最后再加入1mL无菌PBS重悬沉淀。洗取10μl液体滴于载玻片上,甲醇固定30min,革兰氏染色,显微镜下随机挑选20个视野,记录每个视野中每100个精子上粘附的卷曲乳酸杆菌数目并做记录,同时拍照。实验结果如图1所示。
(2)卷曲乳酸杆菌对精子的活力影响检测:
将精子与上述卷曲乳酸杆菌的菌悬液按照1:10的浓度比在HS溶液中混匀,于37℃、5%CO2的培养箱中共孵育2h。加入1mL无菌PBS,3000rpm/min离心5min,弃掉上清。重复操作洗涤3次以洗去未粘附至精子上的卷曲乳酸杆菌,最后再加入1mL无菌PBS重悬沉淀。使用CASA仪器检测共孵育2h后的精子活力,各项检测参数包括:前向运动能力、总精子活力、平均路径速度。
(3)卷曲乳酸杆菌对精子在模拟阴道中泳动能力的影响:
将精子与上述卷曲乳酸杆菌的菌悬液按照1:10的比例在HS溶液中混匀,于37℃、5%CO2的培养箱中共孵育2h。加入1mL无菌PBS,3000rpm/min离心5min,弃掉上清。重复操作洗涤3次以洗去未粘附至精子上的卷曲乳酸杆菌,最后再加入1mL无菌PBS重悬沉淀。以内有甲基纤维素的毛细管模拟女性阴道,将毛细血管插入上述共孵育体系中,于37℃、5%CO2的培养箱中继续共孵育1h。取出毛细管,于显微镜下观察距离毛细血管底部1cm和2cm处的两个区域多个视野下的精子游动情况并记录视野内平均精子数目。实验结果如图2所示。
实施例3(卷曲乳酸杆菌在动物避孕实验中的作用)
一、大鼠生殖模型的建立
(1)实验用雌性SD大鼠分组为:阴性对照组、卷曲乳酸杆菌组,每组8只,雄性SD大鼠共10只;
(2)将医用吸收性明胶海绵剪成0.5cm×0.5cm大小,吸取50μL浓度为1×105CFU/mL菌液使明胶海绵充分吸收菌液后放入大鼠阴道内。完成阴道给菌后,将雄性大鼠与雌性大鼠按照1:4的比例同笼放置使其交配,并在隔天早上九时左右检查雌鼠阴栓的有无,将有阴栓的雌鼠分隔饲养,停止给菌。
(3)连续给菌14天后,取大鼠阴道分泌物,提取阴道菌群,利用高通量测序技术分析给菌后大鼠阴道微生物的种类和数量变化。之后摘除眼球取血后颈椎脱臼处死实验大鼠,解剖计数孕鼠怀孕数,取大鼠子宫与阴道组织,部分置于4%多聚甲醛溶液保存,以进行HE染色及免疫组化检测,部分置于液氮速冻后转移至-80℃冰箱保存,以进行Western-blot、ELISA、q-PCR等检测。
二、动物实验给菌后各项指标检测
1、大鼠子宫HE染色
(1)脱水:将已经固定好的子宫取出,用PBS充分冲洗,然后乙醇脱水,分别用70%乙醇,80%乙醇,90%乙醇,95%乙醇,95%乙醇脱水150min;然后用无水乙醇脱水两次,分别为60min;
(2)包埋:二甲苯处理脱水后的组织30min,石蜡进行包埋;
(3)切片和封片:轮转机切出平整平面,设置切面厚度为55μm,小自挑起切好的拉片,放置于水温为45℃的展片机水槽中,将有褶皱的蜡片展平;将蜡片置于载玻片上,放置在60℃烤片机上,烘烤4h;
(4)脱蜡:分别用二甲苯(Ⅰ)、(Ⅱ)脱蜡15min,然后用二甲苯:无水乙醇=1:1溶液处理5min,分别用浓度递减的乙醇溶液,100%乙醇,95%乙醇,80%乙醇,70%乙醇处理5min。
(5)染色:Harris苏木素染色5min,分别用浓度递减的乙醇溶液,70%乙醇,80%乙醇,95%乙醇,100%乙醇处理5min;0.5%伊红染色1min;依次用100%乙醇(Ⅰ)、100%乙醇(Ⅱ)处理5min;分别加入二甲苯(Ⅰ)、二甲苯(Ⅱ)中透明10min;中性树胶封片;
(6)观察:将封片置于光学显微镜下,分别在200倍与400倍镜下分别拍照观察,并作病理生理学分析。
2、大鼠子宫组织免疫组化
(1)同HE染色常规脱蜡,脱水处理;
(2)为灭活内源性酶用3%新鲜配置的过氧化氢水溶液室温处理5-10min,用去离子水洗3次;
(3)将切片浸入浓度为0.01mol/L pH=6.0构椽酸盐缓冲液中,加热至沸腾后停止加热,自然冷却5-10min后,在重复此操作1-2次,冷却后用pH=7.2-7.6的PBS,洗涤1-2次;
(4)用抗原修复液1室温条件下孵育切片10min,抗原修复液1滴加在切片上并用pH=7.2-7.6的PBS,洗涤1-2次;
(5)室温条件下用5%小牛血清封闭切片20min,除尽多余的水分;
(6)分别滴加1:400兔抗FasL一抗,37℃孵育30min,4℃过夜,pH=7.2-7.6的磷酸盐缓冲液,洗涤2min,洗3次;
(7)滴加1:150生物素二抗(山羊抗兔FasL),37℃温育30min,pH=7.2-7.6的磷酸盐缓冲液洗3次;
(8)滴加1:150SP复合物,37℃孵育30min,pH=7.2-7.6的磷酸盐缓冲液,洗涤5min,洗3次;
(9)微量移液枪吸取1mL去离子水,加DAB显色剂A,B,C各1滴,混匀;
(10)室温条件下吸取50μL显色剂,镜下控制反应时间为5-30min,显色完毕后用去离子水充分洗涤以终止反应;
(11)苏木素复染1min、按照HE染色常规步骤脱水、脱错、透明、封片,镜下观察,分别在200倍与400倍镜下分别拍照观察,并作病理生理学分析。
3、卷曲乳酸杆菌调控炎症信号通路相关蛋白的表达
检测阴道给菌后子宫组织中炎症相关信号通路TLR4-MyD88-NF-KB的蛋白表达情况,以分析卷曲乳酸杆菌对相关信号通路蛋白表达的影响。此项评价通过Western-blot显示。
大鼠处死后,提取子宫组织蛋白,进行Western-blot。
(1)大鼠子宫组织蛋白提取
①组织用含PMSF(1mM)的无菌冰去离子水冲洗数次;
②将组织放置匀浆器中,加入适量裂解液缓冲液RIPA及蛋白酶抑制剂,冰水混合浴中匀浆;
③裂解产生的蛋白溶液全部转移至1.5mLEP管中;
④1.5mL管置于冰上,漩涡震荡10s,放置5min,重复3-4次;
⑤4℃,13000rpm离心10min;
⑥吸取上清至全新1.5mLEP管中,-80℃冰箱保存,上清即为全蛋白。
(2)Western-blot
①在收集的蛋白样品中加入适量的5x蛋白上样缓冲液,沸水浴加热10min,以充分变性蛋白;
②配制5%的浓缩胶,10%或12%分离胶,待胶凝固后,将胶浸入1×电泳缓冲液中,上样电泳;
③电泳完成后,将胶剥离玻璃板,转膜待用。将PVDF膜用1×转膜缓冲液完全浸润后,胶和膜用夹板夹好,进行转膜;
④转膜完成后,取出PVDF膜,用5%脱脂乳或5%BSA封闭后,添加目标蛋白的一抗后4℃孵育过夜,再添加HRP即标记的二抗孵育2h,进行显色曝光;
⑤对显色内容进行拍照,使用软件分析灰度,作图分析。
4、卷曲乳酸杆菌调控炎症因子的表达
检测阴道给菌后子宫组织中TNF-α、IL-1β、IL-6、IL-2及血清中TNF-α、IL-1β、IL-6、IL-2的表达量,以分析卷曲乳酸杆菌对免疫应答的影响。抑炎和促炎体现在mRNA水平,影响促炎及抑炎性因子的转录,下调、上调炎症介质mRNA的表达量,此项评价通过q-PCR显示。也体现在蛋白水平,影响促炎性及抑炎因子的翻译,下调、上调炎症介质蛋白的表达量,此项评价通过ELISA显示。
大鼠处死后,提取阴道组织RNA,反转录成cDNA后进行q-PCR,检测阴道组织中炎症因子TNF-α、IL-1β、IL-6、IL-2转录水平的表达情况。
(1)大鼠子宫组织RNA的提取
①取相同质量的大鼠子宫内膜组织,加入1mL Trizol,用剪刀剪碎,并用组织匀浆机匀浆,5000rpm 10min,转移上清至新的EP管中;
②加入氯仿160μL,充分振荡混匀。4℃13000rpm 15min,离心完毕,吸取上清,注意不要吸到蛋白层;
③加入等体积异丙醇,充分混匀,4℃13000rpm 10min,倒去上清;
④加入1mL 75%乙醇洗涤,振荡,充分溶解后,4℃13000rpm 10min,弃上清;
⑤加入1mL无水乙醇洗涤,振荡,充分溶解后,4℃13000rpm 10min,弃上清;
⑥常温晾干30min;
⑦加160μLRNase Free Water,55℃促溶;
⑧测定RNA浓度。
(2)q-PCR
参照TAKARA试剂盒说明书进行操作。
大鼠处死前进行摘除眼球取血,离心分离血清,ELISA检测血清中炎症因子TNF-α、IL-1β、IL-6、IL-2蛋白水平的表达情况。
(1)ELISA实验步骤
①使用前讲试剂盒在室温下平衡半小时;
②空白孔不加样,只加A,B和终止液用于调零;
③标准品孔:每孔加好稀释好的标准品50μL,零孔加入标准品/样品稀释液50μL,然后加入生物素抗原工作液50μL;
④样品孔:加入稀释好3倍的样品50μL,然后加入生物素抗原工作液50μL;
⑤轻轻摇晃,盖上封膜板,37℃培养60min;
⑥将各25倍浓缩洗涤液用蒸馏水25倍稀释后备用;
⑦第一次洗涤:小心揭开封膜板,弃去液体,甩干,每孔加入200μL,静置30s后弃去,拍干,如此重复3次;
⑧加入50μL亲和素-HRP即到标准品孔和样品孔中,轻轻摇晃,盖上封膜板,37℃培养60min;
⑨第二次洗涤:小心揭开封膜板,弃去液体,甩干,每孔加入200μL,静置30s后弃去,拍干,如此重复3次;
⑩显色:每孔加入显色剂A 50μL,再加入显色剂B 50μL,轻轻震荡混匀,37℃避光显色10min;
以上实验结果表明,卷曲乳杆菌菌悬液能够在不影响女性生殖道菌群平衡以及健康的情况下,通过降低精子活性达到细菌性避孕的目的,从而为其制备女性避孕药物的用途提供了实验依据。
