As an essential tool in optogenetic technology, photoreceptors have been discovered or synthesized in many ways so far, and have been successfully used for studies on cellular functions, signal transduction, molecular functions at the sub-cellular level, and the like. Among them, light-oxygen-voltage sensing domain (LOV) domain protein is a photo receptor which is currently being researched more, has small relative molecular mass and solubility, and has many ways of regulating signal molecules because its required cofactors exist in most types of cells, which make LOV an ideal molecule for exploring photo-controlled ion channels. Previous studies have used various means to embed or attach different polypeptides and enzymes in proximity to the LOV domain (LOV regulation).
Ion channels are involved in regulating intracellular signaling and information integration in neurons and glial cells, and are also important drug targets for many neurological diseases. To assess the role of ion channels in the complex nervous system, gene-regulated ion channels are a desirable model. In the context of gene regulation of ion channels, there are several strategies: one aspect is the expression of genetically encoded membrane-targeted peptide toxins in different types of cells. The peptide toxin is composed of a series of polypeptides from toxic animal genetic code regulation ion channels, and can specifically recognize various ion channels. These toxins induce a reversible response over a period of hours to days without the addition of exogenous chemicals, and have been shown to be effective in the living brain of mammals. The second strategy is to use a photosensitive compound that anchors at specific positions in the ion channel (e.g., SPARK, LiGluR, HyLighter) and can induce a rapid and reversible light sensing response.
