Abstract
本发明涉及一种染色体DNA中含有cytlAa和cryllAa基因的苏云金杆菌(Bacillusthuringiensis)杀虫工程菌及其构建方法。该工程菌是利用转座子衍生载体pTV1TS将cytlAa和cryllAa基因导入受体染色体中,并使之缺失转座酶而获得的。室内外试验证实该工程菌株具有高毒、广谱和遗传稳定的特点,对鳞翅目害虫有更高毒效并能延缓或克服害虫抗性,同时扩大杀虫谱即兼杀双翅目害虫。
Description
本发明属于苏云金杆菌(Bacillus thuringiensis,简称Bt)基因工程技术领域,具体涉及一种苏云金杆菌杀虫工程菌及其构建方法。
苏云金杆菌杀虫剂是目前国内外生产量最大、应用面积最广的一种微生物杀虫剂,具有无污染、无残毒、对靶害虫专一和对人畜、植物等非目标生物安全等优点。但利用天然Bt菌株生产的杀虫剂也存在缺点,如杀虫谱窄、毒力不够强等,而且害虫也会对Bt产生抗性。斜纹夜蛾(Spodoptera litura)和甜菜夜蛾(Spodoptera exigua)都是杂食性害虫,食量大且分布广,对多种农作物尤其是蔬菜危害严重,而且已对许多化学杀虫剂产生了抗性,同时也是典型的对Bt杀虫剂不够敏感的害虫。自八十年代中期以来,已有大量实验数据与统计资料证明,昆虫会象对付化学农药一样发展自己对Bt的抗性。在我国深圳地区连续使用Bt的田间,已经发现了高抗性的小菜蛾(Plutella xylostella)。在夏威夷连续两年使用Bt杀虫剂后,小菜蛾种群对B.t.subsp.kurstaki的敏感性下降了25-30倍。在Bt广泛和大量使用的台湾和菲律宾地区,也有小菜蛾对Bt敏感性降低的报道。其他一些抗性昆虫亦陆续见诸报道,如埃及伊蚊(Aedes aegypti)、干果斑螟(Cadracautella)、烟芽夜蛾(Heliothis virescens)、向日葵同斑螟(Homoeosoma electellum)及马铃薯叶甲(Lepinotarsa decemlineata)等。最近,Monsanto公司推出不久的转Bt-ICP基因的棉花已完全失去抗棉铃虫的能力。因此对Bt进行遗传改良,扩大杀虫范围,提高晶体蛋白的产量和毒力,延缓害虫产生抗性等,对于充分发挥Bt制剂综合效能具有特别重要的意义。
苏云金杆菌对昆虫的毒性主要靠菌体产生的各种杀虫晶体蛋白(Insecticidal CrystalProteins,ICPs),这些ICPs具有杀虫的专一性,研究已证明大多数ICPs的编码基因位于菌体内的大质粒上。Bt在形成芽孢时能形成由杀虫晶体蛋白构成的伴孢晶体,当敏感害虫吞食后,在昆虫中肠内晶体蛋白溶解、一级结构发生变化而被激活为活性毒素并与特异性受体结合,最终导致膜穿孔使取食昆虫感染并逐渐死亡(Adang,1991;龙綮新、庞义,1994)。目前已报道的ICP基因有135个,根据其编码蛋白的氨基酸序列同源性的大小被划分为24群48类71亚类,具有溶细胞作用的ICP基因归为cyt系列如cyt1、cyt2,其余都归入cry系列如cry1、cry2等(Crickmore,1998)。ICP基因编码的蛋白,其N-端为毒性区,决定ICPs对靶昆虫的毒力,C-端为保守区,与其对靶昆虫的专一性(specificity)有关。
cry11Aa基因(EMBL Accession NO.X03182)和cyt1Aa基因(EMBL Accession No.M31737或J03510)克隆自苏云金杆菌莫里逊亚种(B.t.subsp.morrisoni),cyt1Aa基因原名cytA,表达27kDa蛋白,在靶昆虫中肠消化液中被激活为25kDa的核心毒素;cry11Aa基因原名cryIVD,表达67kDa蛋白,在靶昆虫中肠消化液中被激活为40kDa的核心毒素。两者对双翅目的蚊子及黑蝇具高毒效,前者还有广谱溶细胞特性;此外,在研究害虫对Bt抗性过程中人们发现广谱溶细胞的Cyt1Aa蛋白,不但对其它ICPs有协同增效作用,而且可能有克服或延缓目标害虫对Bt制剂产生抗性的作用。但目前对鳞翅目、鞘翅目害虫有效的Bt制剂中均不含Cyt1Aa蛋白。另外,现有含有cyt1Aa和cry11Aa基因的苏云金杆菌其所含这两个基因均位于大质粒上。而通过质粒转移或ICPs的共表达构建苏云金杆菌重组菌株,以期扩大Bt杀虫谱和增强毒力,常因质粒的不稳定性和质粒间的排斥性而受到一定限制(庄美宝、庞义,1995)。
本发明的目的是通过遗传工程方法提供一种含有cyt1Aa和cry11Aa基因的杀虫苏云金杆菌工程菌及其构建方法,该工程菌具有高毒、广谱、遗传稳定且害虫不易产生抗性等优点,从而解决已有Bt制剂所存在的上述问题。
本发明的苏云金杆菌杀虫工程菌的特征是其染色体DNA中含有cyt1Aa和cry11Aa基因。
上述工程菌的代表菌株Bt-TnY已保藏于中国典型培养物保藏中心(CCTCC,中国,武汉大学校内),保藏编号:CCTCC M98017,保藏日:1998年11月26日。该菌株Bt-TnY的染色体DNA中含有cyt1Aa和cry11Aa基因,主要产生130、60、67和27 kDa的4种杀虫晶体蛋白(如图6)。
本发明的苏云金杆菌杀虫工程菌的构建方法是:把cyt1Aa和cry11Aa基因分别克隆到链球菌(Streptococcus faecalis)的转座子Tn917(Youngman,1983;1984)(如图1)载体pTV1TS(Sandman&Youngman,1987)上,然后同时转入出发菌中,使这两个基因整合到出发菌染色体上,得到第一代工程菌,再将这工程菌与出发工程菌进行原生质体融合,从而获得目标工程菌。其具体步骤如下:
1.分别在Tn917的tnpA序列区中,以HindIII酶切去掉1.3kbp的DNA片段,再插入8kbp左右的目标基因(cyt1Aa和cry11Aa基因)DNA片段,从而分别得到含cyt1Aa基因和含cry11Aa基因的转座子衍生载体pTYP45和pTYP26。这两个目标基因直接来源于质粒pWF45和pWF25,质粒pWF45含有cyt1Aa基因和20 kDa蛋白基因及启动子(Wu,etal.,1993),而质粒pWF26含有cry11Aa和20 kDa蛋白基因(Wu,et al.,1995)。将tnpA基因克隆到B.t-E.coli穿梭质粒pHT3101(D.Lereclus,et al.,1989)上,得到一个非转座载体质粒pHtnpA。上述载体pTYP45、pTYP26和pHtnpA的构建过程分别如图2、图3和图4所示。
由于在Tn917的tnpA序列区中,以HindIII酶切去掉1.3kbp的DNA片段,再插入8kbp左右的目的基因DNA片段,这样,重组的转座子衍生载体pTYP26和pTYP45实际上转座酶基因已失活,不能再表达TnpA蛋白,理论上转座作用不能发生,需要依靠一个非转座载体质粒pHtnpA的帮助。由于整合进染色体的转座酶基因tnpA已失活,因而可避免在自然界中二次转座作用的发生;外源基因随染色体DNA的复制而复制,遗传稳定,不会扩散于环境中,安全性高。
2.用高压电激法同时把pTYP26、pTYP45和pHtnpA(按1∶1∶1)转入出发菌,在含红霉素的培养基上筛选转化子。分别提取质粒DNA和总DNA,用目标DNA片段标记为探针,经Dot blot和Southern blot鉴定,并经PCR验证,获得目的基因整合进染色体的第一代工程菌。所得到的转座频率为5.9×10-5,共合体折分率为97%。上述得到的第一代工程菌能高水平表达Cyt1Aa和Cry11Aa蛋白,对双翅目昆虫(如蚊子和黑蝇等)幼虫有高毒,如对库蚊的毒力LC50达10~80ng/ml。
由于在筛选工程菌过程中,多次进行42℃高温处理,因高温处理可促进转座作用的进行及富集染色体上多点插入突变菌,但也极易使存在于菌体中的质粒丢失。第一代工程菌基本上丢失了原出发菌株的大部分质粒。苏云金杆菌大多数伴孢晶体的结构基因及调节基因位于较大的质粒上,其大小为33-150MD,目前尚未在小于30MD的质粒上发现有毒素基因。
3.将第一代工程菌与出发菌与进行原生质体融合,融合子重新获得大质粒。对融合子进行晶体镜检、质粒图谱分析、PCR、SDS-PAGE及生物测定等,最后获得目标工程菌。
本发明的杀虫苏云金杆菌的具体构建过程如图5所示。
上述方法中所用的出发菌通常选用较高杀虫活性的Bt菌株。本发明所用的优选菌株是从德国土壤中分离的菌株Bt-S184(用特定引物对其PCR检测,可检出cry1和cry2型ICP基因)所得到的目标工程菌即为Bt-TnY(CCTCC M98017)。经SDS-PAGE蛋白电泳分析、PCR检测等表明,Bt-TnY产生的伴孢晶体除含有出发菌株Bt-S184的130kD和60kD两个主要蛋白外,还含有外源基因cyt1Aa和cry11Aa的表达产物27kD和67kD蛋白。进一步的生物测定证实,该工程菌不但对鳞翅目的斜纹夜蛾(Spodoptera litura)和甜菜夜蛾(Spodoptera exigua)等难治的夜蛾科害虫以及棉铃虫有高毒,而且对双翅目的蚊子也有强烈毒杀作用;初步实验证明该菌还有延缓害虫产生抗性的作用;室内试验证实了工程菌株Bt-TnY有良好的遗传稳定性。Bt-TnY是一株高毒、广谱的杀虫Bt工程菌。
在工程菌Bt-TnY中,目标基因cyt1Aa和cry11Aa通过非转座载体介导整合进染色体基因组中,并获得稳定的高效表达;在光学显微镜和电子显微镜下,可看见菱形、方形、不规则形和卵圆形伴孢晶体(如表四)。SDS-PAGE分析显示有四条主带,分子量分别为130、60、67和27kDa(如图6)。由于非转座载体pHtnpA上的转座酶基因随着热处理(42℃)质粒的丢失而丢失,发生二次转座的可能性很小,目的基因能随受体菌的分裂、传代而稳定遗传,连续培养100代,丢失频率小于10-5。
本发明所采用的转座子Tn917的衍生载体pTV1TS如图1所示。Tn917作为特别有效的插入突变介导具有如下特征:(a)Tn917插入染色体、质粒或噬菌体具有相当高的随机性。Tn917可在不同染色体或染色体不同位点上形成插入突变子,但Tn917的插入在染色体上具有区域优先性。(b)Tn917转座频率相当高,转座染色体的频率为10-5,质粒为10-4。(c)Tn917插入突变非常稳定,回复率小于10-10。(d)Tn917允许容纳8kbp的外源DNA而对其转座频率无影响。(e)在Bs或其它许多G+菌中,可利用Tn917携带的Emr作选择压力(余健秀、庞义,1998)。pTV1TS由转座子Tn917、温度敏感复制子Ts-Rep和存在于转座子序列之外的cat基因三部分构成。该载体具有如下两大优点:(a)较低温(38~48℃)可以富集染色体上有转座插入的菌落。(b)染色体上含有转座插入的菌落可以不必在选择压力温度而在32℃下直接铺平板而得到。pTV1TS不带有任何危险的标记基因,对人类和生态环境没有或危害的可能性极小。

图6是本发明的野生菌株Bt-S184和工程菌Bt-TnX、Bt-TnY的SDS-PAGE分析。图中M代表标准蛋白,X代表Bt-TnX,Y代表Bt-TnY,S代表Bt-S184。Bt-TnY主要含有Cry11Aa(67 kDa)、Cyt1Aa(27kDa)、Cry1(130kDa)和Cry2(60kDa)四种蛋白;Bt-TnX主要含有Cry11Aa(67kDa)、Cyt1Aa(27kDa)和Cry1(130kDa)三种蛋白;Bt-S184主要含有Cry1(130kDa)和Cry2(60kDa)两种蛋白。
以下通过工程菌Bt-TnY的构建实例及应用实例对本发明作进一步说明。
一.Bt-TnY的构建
(1)在液体培养基富集衍生转座子整合进Bt染色体的阳性菌落:利用高压电激法同时把pTYP26、pTYP45和pHtnpA(按1∶1∶1)转入Bt-S184受体菌,电激后静置30分钟,在32℃25ml LB液体培养基(含1μg/ml Em、5μg/ml Cm及25μg/ml Lm)在250ml锥瓶振摇至中对数生长期。按1∶20稀释上述菌液,42℃继续培养,只加入针对转座子的抗生素Em(1μg/ml)。再按1∶20稀释,置42℃,1μg/ml Em继续培养至晚对数期(3-5h)。以4000rpm离心5分钟收集菌体。用原体积数的1/25的10%甘油悬浮沉淀,然后分装于EP管中,迅速放入到液氮中保存备用。
(2)初选阳性菌落:取晚对数期培养液100-200μl在含1μg/ml Em的LB固体平板上划线,12-16小时培养。挑取单菌落接种到含1μg/ml Em的LB固体平板上,待生长成2-3mm。同时计算转座子衍生质粒的转座频率及共合体拆分率。
(3)挑选有意义的阳性菌落:上述单菌落在含Em的LB液体培养基中32℃摇床培养,3天收获菌体,进行SDS-PAGE鉴定外源基因表达与否。同时分别进行总DNA(包括染色体、质粒)和质粒DNA的酶切后,进行Southern分析,探针标记采用Dig法。这样得到了第一代工程菌Bt-TnX。
(4)已知大多数ICPs的编码基因位于菌体内的大质粒上,但42℃高温处理使存在于菌体中的大质粒极易丢失。所以必须进行出发菌与阳性菌的原生质体融合,采取如下路线:种的复壮一抗生素梯度试验--原生质体形成及再生曲线--原生质体融合--融合子的筛选。这样得到了目标工程菌Bt-TnY。
(5)工程菌遗传稳定性试验:在无选择压力下连续继代培养50-100代,稀释涂板,进行抗性检测和生产检测,稳定系数达到98.1%。
将目的基因整合到不同Bt菌株染色体上的方法与上述相同。
二.工程菌Bt-TnY应用实例
a.室内外试验证实该工程菌株具有高毒、广谱和遗传稳定的特点,对鳞翅目害虫如斜纹夜蛾(Spodoptera litura)、银纹夜蛾(Argyrogramma agnata)、甜菜夜蛾(Spodopteraexigua)、小菜蛾(Plutella xylostella)、小菜粉蝶(Pieris rapae)、玉米螟(Ostrinianubilalis)和棉铃虫(Heliothis armigera)等幼虫有更高毒效并能延缓或克服害虫抗性,同时扩大杀虫谱即兼杀双翅目害虫(如蚊子)。

b.在我国深圳地区连续使用Bt的菜田间,已经发现了对目前市场上主要Bt产品HD-1高抗性的小菜蛾(Plutella xylostella)。生物测定表明,Bt-TnY能有效地毒杀深圳地区高抗性的小菜蛾。工程菌Bt-TnY的摇瓶培养液原液稀释至1000-10000倍,在24小时内能有效杀灭高抗性的小菜蛾。
c.小规模田间应用及实验室试验:将工程菌Bt-TnY接种于5个250ml三角瓶的液体LB培养基28℃摇床培养3~4天,至孢子囊成熟晶体脱落,收集瓶装,即时使用或加防腐剂在常温下(25℃)避光保存备用。
d.大规模田间应用:可以用工厂化的大型发酵罐发酵培养,可制成粉剂、颗粒剂、水剂、乳剂和油剂。

Claims (2)
1.一种苏云金杆菌杀虫工程菌,其特征在于该菌株为Bt-TnY,其保藏号为:CCTCC M98017。
2.权利要求1所述苏云金杆菌杀虫工程菌的构建方法,其特征是把cyt1Aa和cry11Aa基因分别克隆到链球菌的转座子Tn917载体pTV1Ts上,然后同时转入出发菌中,得到cyt1Aa和cry11Aa基因整合进染色体的第一代工程菌,再将该工程菌与出发菌进行原生质体融合,从而获得目标工程菌。
