Abstract
一种含有板栗多糖和左旋肉碱的保健组合物,包括板栗多糖和左旋肉碱,所述板栗多糖和左旋肉碱质量比为(500~1200):(200~1000)。本发明本发明1、疗效较好,服用后能促进睾丸曲细精管间质细胞功能改善、增强性机能、促进精子生成、提高精子活力,有效提高生育能力;2、适用范围广,适用于各种年龄人群,尤其是对因年龄增大导致生育能力下降有较好疗效,且无明显副作用。

技术领域
本发明涉及保健食品领域,具体涉及含有板栗多糖和左旋肉碱的保健组合物。
背景技术
由于男性因素而导致的不孕不育症称男性不育症,男性不育症是一种发病率较高且较难治愈的病症。国内外目前研究资料表明:不孕不育症在育龄期夫妇中的发病率约为15%-20%,根据世界卫生组织统计:全球大约有600-8000万对夫妇患有不同程度的不育症,其中由男性因素导致的不育约占所有不育夫妇的50%。
中国人口基数大,且相对世界水平发病几率较高,近年来也呈上升趋势。不育症往往造成夫妻感情不和,婚姻家庭破裂,给社会带来不稳定因素。男性不育的病因多种多样,最常见且最直接的外在表现为精液品质下降。目前我国医药技术领域在治疗男性不育症方面积累了较为丰富的经验,有西医治疗,如采用药物、手术、局部用药等,也有许多有效的中成药,如补肾种子方、补肾生精丸、温肾丸、育精丸等,虽然这些治疗方法在治疗不育症方面各有其优点,但是每种方法都存在一定的局限性。且药物治疗持续时间长,并伴有众多副作用,手术治疗创伤大。且由于中华民族的传统,人们更愿意优先选择食补。因此,提供一种效果显著、无副作用、能改善精液质量、加快男性精子形成、提高生育能力的组合物及普通食品具有重要的现实意义。
发明内容
本发明针对现有技术的不足,提出一种无副作用,能改善精液质量,降低精子死亡率,提高生育能力的含有板栗多糖和左旋肉碱的保健组合物,具体技术方案如下:

下面结合附图对本发明的较佳实施例进行详细阐述,以使本发明的优点和特征能更易于被本领域技术人员理解,从而对本发明的保护范围做出更为清楚明确的界定。
实施例一:一种含有板栗多糖和左旋肉碱的保健组合物;
各成分按以下剂量制成粉剂:
每日1袋,晨起顿服,30天为一疗程,共服用3疗程。
实施例二:一种含有板栗多糖和左旋肉碱的保健组合物;各成分按以下剂量制成粉剂:
每日1袋,晨起顿服,30天为一疗程,共服用2疗程。
实施例三:一种含有板栗多糖和左旋肉碱的保健组合物;各成分按以下剂量制成粉剂:
每日1袋,晨起顿服,30天为一疗程,共服用4疗程。
在本发明中:
板栗多糖:板栗多糖是多种单糖的混合物。有研究显示板栗多糖具有多种生物学功能,能有效清除自由基,增强免疫系统功能,而且板栗多糖对雄性生殖也有一定的促进作用。
左旋肉碱:该物质是维持人体正常生理活动的必需成分。它具有非常重要且多样的作用。左旋肉碱是人体内转运供能物质的重要载体,一方面,附睾与精子中含有大量的左旋肉碱,它能将长链脂肪酸转运入精子线粒体内进行β氧化,为精子运动提供线粒体,另一方面,它还能够将代谢产生的乙酰基、乳酸等废物转运出精子,维持精子正常功能。此外,该物质还具有较强的还原性,能够降低精子内的氧化应激反应水平,保护精子的结构。有文献证实,培养基中添加该物质能够增加精子的活力、精子密度和精子DNA的完整性。
精氨酸:是精子蛋白的重要组成部分,适量补充精氨酸可以保持精子的正常功能。
雨生红球藻:具有抗氧化、消除自由基方面的能力,可以促进抗体的产生、增强动物的免疫功能。
锌:该物质大量存在于男性睾丸中,参与精子的整个生成、成熟和获能的过程。男性一旦缺锌,就会导致精子数量减少、活力下降、精液液化不良,最终导致男性不育。缺锌还会导致青少年没有第二性征出现、不能正常生殖发育。
硒:该物质是精浆中过氧化物酶的必需组成成份,男性不育症患者精液中硒水平普遍偏低,研究发现:精液中硒水平越高,精子数量越多,活力越强。硒具有强大的抗氧化作用,可清除过剩的自由基,抑制脂质过氧化作用。男性不育症患者精液硒水平低,自然会削弱机体自身对精液中存在的氧自由基的清除和脂质过氧化的抑制,从而导致患者精子活力低下,死亡率高,引发不育症。
叶酸:叶酸是一种水溶性维生素,对蛋白质合成及各种氨基酸的代谢有重要作用,并参与嘌呤和嘧啶合成,在细胞DNA合成中发挥作用。它能够预防胎儿低体重、唇腭裂、心脏缺陷和神经管发育缺陷等畸形。
维生素B12:以辅酶的形式存在,可以增加叶酸的利用率,促进碳水化合物、脂肪和蛋白质的代谢。
维生素D3:促进钙的吸收。
实验验证过程:
板栗多糖实验,三周龄公鼠注射白消安(B,40mg/kg),注射白消安后给小鼠进行灌胃处理,浓度梯度为CP 0mg/kg、CP 0.01mg/kg、CP 0.1mg/kg、CP 1mg/kg、CP 10mg/kg、B、B+CP 0.01mg/kg、B+CP 0.1mg/kg、B+CP 1mg/kg、B+CP 10mg/kg。连续灌胃5周,小鼠生长到8周时收取样品。相关指标变化及差异如表(1-1)。
从收样指标挑选出几个组进行深入的研究,CP 0mg/kg、CP 0.01mg/kg、CP0.1mg/kg、CP 1mg/kg、B、B+CP 0.01mg/kg、B+CP 0.1mg/kg、B+CP 1mg/kg。收样时发现睾丸的重量变化比较大,注射白消安后睾丸普遍缩小,睾丸指数如图1所示,不同浓度CP灌胃处理、注射B后不同浓度CP灌胃处理,睾丸指数的统计分析,a,b,c,d表示不同处理组之间差异显著,p<0.05;N>3
注射白消安后睾丸指数下降明显,0.1mg/kg的板栗多糖灌胃处理后,睾丸指数上升并且差异显著。
精子浓度与精子活力均发生了变化。精子浓度变化如图2,不同浓度CP灌胃处理与注射B后不同浓度CP灌胃处理,精子浓度变化的统计分析,a,b,c表示不同处理组之间差异显著,p<0.05;N>3;
与CP 0组相比,不同浓度的CP灌胃处理后,精子浓度上升,但是差异不显著。白消安处理后精子浓度明显下降,利用CP进行灌胃处理后,精子浓度有所上升,但是差异不显著。
注射白消安后精子活力也发生了变化,如图3所示,不同浓度CP灌胃处理与注射B后不同浓度CP灌胃处理,精子活力变化的统计分析,a,b,c表示不同处理组之间差异显著,p<0.05;N>3;注射白消安后小鼠的精子活力几乎为0,不同浓度的板栗多糖灌胃处理后精子活力上升,并且与白消安组相比差异显著。
HE染色从形态学上证实了,单独灌胃板栗多糖对小鼠睾丸曲细精管的形态和生精细胞的数量影响不明显。如图4所示,但是注射白消安后小鼠睾丸曲细精管中出现大量的空泡,并且生精细胞数量显著下降如图5所示。
注射白消安后小鼠睾丸曲细精管内出现了大量的空泡,但是板栗多糖灌胃处理后,曲细精管内生殖细胞增多。从小鼠各项指标、精子浓度、精子活力以及组织形态学观察发现板栗多糖0.1mg/kg对白消安注射的睾丸有一定的挽救作用。
继续探索板栗多糖是从哪些方面发挥了作用。继而从转录组、宏基因组、代谢组学三个大方面进行深入研究。
转录组学在整体水平上研究细胞中基因转录的情况及转录调控规律的学科。选取了CP 0mg/kg、CP 0.1mg/kg、B、B+CP 0.1mg/kg进行测序。目的从RNA水平研究白消安注射、板栗多糖灌胃处理后基因表达的变化情况,并据此推断相应未知基因的功能,揭示特定调节基因的作用机制。
宏基因组学通过直接从环境样品中提取全部微生物的DNA,构建宏基因组文库,研究样品所包含的全部微生物的遗传组成及其群落功能。目的通过宏基因组学的方法对肠道微生态方面进行研究,CP 0mg/kg、CP 0.1mg/kg、B、B+CP0.1mg/kg四个组,对单独板栗多糖灌胃以及白消安注射、板栗多糖灌胃的小鼠小肠内容物进行检测,从而从肠道微生物方面解释板栗多糖作用。
代谢组学主要研究的对象是各种代谢路径的底物和产物的小分子代谢物(MW<1000)。代谢组选取了CP 0.1mg/kg、B+CP 0.1mg/kg两个组进行检测。对血清中低分子量代谢产物同时进行定性和定量分析,从而解释板栗多糖对代谢产物的影响。
转录组学、宏基因组学、代谢组相互联系,主要是通过KEGG代谢通路与调控网络将几个组学的数据联合分析。从宏基因组分析关联到对肠道代谢关联到生殖内分泌最终关联到精子发生相关通路。从而解释板栗多糖是如何发挥作用的以及在哪些通路上发挥了作用。

1.一种含有板栗多糖和左旋肉碱的保健组合物,包括板栗多糖和左旋肉碱,其特征在于:所述板栗多糖和左旋肉碱质量比为(500~1200):(200~1000)。
