本发明公开了一种纳米胶囊型聚多巴胺药物材料及其制备方法与应用,属于纳米药物及其制备领域。本发明采用一步合成法的方法制得,该方法是将有机金属框架作为模板,将药物固定,大大提高了药物负载量;本发明所提供的纳米胶囊型聚多巴胺药物具有较高的药物负载量,且其在近红外光介导下可以促进药物的释放,达到联合治疗的效果;同时具有制备方法条件温和,对设备和操作、环境等要求不高,原料经济易得,易于实施且稳定性、重现性好。

技术领域
本发明属于纳米药物及其制备领域,具体地,涉及一种纳米胶囊型聚多巴胺药物材料及其制备方法与应用。
背景技术
纳米聚多巴胺颗粒因其较好的水溶性、生物相容性及良好的光热升温效果,在肿瘤光热治疗及联合治疗方面具有诸多应用。其表面所具有的易修饰性也为其拓宽了应用,可将小分子药物通过化学键偶联的方式修饰在纳米聚多巴胺颗粒表面达到联合治疗的作用。但这种方面的药物负载量相对较低,且难以到达细胞亚细胞器从而进行化学治疗。
纳米胶囊型聚多巴胺是一种新型的纳米聚多巴胺材料,其具有中空内壳。因胶囊型聚多巴胺表面含有大量微孔,其可对小分子药物进行吸附与释放,从而达到药物搭载与释放的目的,从而提高联合治疗的效果。但目前中空型聚多巴胺的制备多靠刻蚀法得到,其通过将聚多巴胺修饰在牺牲模板表面后将牺牲模板刻蚀掉的方法制得。该方法的制备过程中往往要使用强酸或强碱作为刻蚀剂,且所用牺牲模板多为实心材料,且后搭载的药物载药率也普遍仅为10%,难以实现一步法制备高载药率纳米胶囊型聚多巴胺药物。因此本领域急需一种新型方法通过一步法得到高载药率纳米胶囊型聚多巴胺药物,这将在纳米聚多巴胺进行协同治疗方面取得新的突破。
发明内容
针对现有技术的不足,本发明提供了一种纳米胶囊型聚多巴胺药物材料及其制备方法与应用,所述药物材料具有高载药率,并且可以有效的进行光热与化疗的协同治疗;所述药物材料的制备方法简单易行、稳定可靠,所述药物材料能够用于多种肿瘤的光热与化疗的联合治疗。
本发明是通过以下技术方案实现的:一种纳米胶囊型聚多巴胺药物材料的制备方法,具体包括以下步骤:
(1)将搭载化疗药物的有机金属框架材料与多巴胺按照质量比为1:0.5~5混合,随后加入Tris-HCl(pH=8.8)溶液,使得多巴胺的浓度为0.2~10mg/mL,超声后得到母液。
(2)将步骤1中得到的母液置于磁力搅拌器上,在10~50℃条件下,反应24~120h。
(3)将步骤2中反应后的溶液离心分离、冷冻干燥,得到纳米胶囊型聚多巴胺药物。
进一步地,步骤1中所述的有机金属框架材料为ZIF-8、ZIF-90、UIO-66、MIL-88A、MIL-100、MIL-101中的一种。
进一步地,所述药物材料包括搭载化疗药物的药物部分和纳米胶囊型聚多巴胺的外层载体部分。
进一步地,所述药物材料的大小为50~300nm,聚多巴胺层厚度为5~30nm,药物负载量为20~60%。
进一步地,所述化疗药物为阿霉素、顺铂、去甲长春碱、紫杉醇、丝裂霉素、长春地辛中的至少一种。
进一步地,一种纳米胶囊型聚多巴胺药物材料在癌症联合治疗中的应用,其特征在于,包括乳腺癌、胰腺癌、肾癌、肝癌。
与现有技术相比,本发明产生的有益效果包括:
1)本发明所提供的纳米胶囊型聚多巴胺药物易于合成,采用一步合成法的方法制得,该方法是将有机金属框架作为模板,将药物固定,大大提高了药物负载量;
2)本发明所提供的纳米胶囊型聚多巴胺药物具有较高的药物负载量,且其在近红外光介导下可以促进药物的释放,达到联合治疗的效果;
3)本发明的制备方法条件温和,对设备和操作、环境等要求不高,原料经济易得,易于实施且稳定性、重现性好。

实施例1
按照如下步骤合成纳米胶囊型聚多巴胺药物
按照ZIF-8@阿霉素与多巴胺的质量比为1:2,随后加入Tris-HCl(pH=8.8)溶液,多巴胺最终浓度为3mg/mL,超声后得到母液。将母液置于30℃搅拌反应台上搅拌48h后取出,离心冷冻干燥,得到纳米胶囊型聚多巴胺药物,药物大小为230nm,聚多巴胺层厚度为18nm,药物负载量为38%。
实施例2
按照如下步骤合成纳米胶囊型聚多巴胺药物
按照ZIF-8@顺铂与多巴胺的质量比为1:1,随后加入Tris-HCl(pH=8.8)溶液,多巴胺最终浓度为2mg/mL,超声后得到母液。将母液置于25℃搅拌反应台上搅拌72h后取出,离心冷冻干燥,得到纳米胶囊型聚多巴胺药物,该药物大小为170nm,聚多巴胺层厚度为15nm,药物负载量为45%。
实施例3
按照如下步骤合成纳米胶囊型聚多巴胺药物
按照ZIF-8@紫杉醇与多巴胺的质量比为1:0.5,随后加入Tris-HCl(pH=8.8)溶液,多巴胺最终浓度为0.5mg/mL,超声后得到母液。将母液置于10℃搅拌反应台上搅拌72h后取出,离心冷冻干燥,得到纳米胶囊型聚多巴胺药物,该药物大小为150nm,聚多巴胺层厚度为5nm,药物负载量为43%。

实施例13
按照如下步骤合成纳米胶囊型聚多巴胺药物
按照MIL-101@阿霉素与多巴胺的质量比为1:4,随后加入Tris-HCl(pH=8.8)溶液,多巴胺最终浓度为3mg/mL,超声后得到母液。将母液置于45℃搅拌反应台上搅拌108h后取出,离心冷冻干燥,得到纳米胶囊型聚多巴胺药物,该药物大小为207nm,聚多巴胺层厚度为27nm,药物负载量为41%。

按照如下步骤合成纳米胶囊型聚多巴胺药物
按照MIL-101@顺铂与多巴胺的质量比为1:2,随后加入Tris-HCl(pH=8.8)溶液,多巴胺最终浓度为3mg/mL,超声后得到母液。将母液置于25℃搅拌反应台上搅拌72h后取出,离心冷冻干燥,得到纳米胶囊型聚多巴胺药物,该药物大小为198nm,聚多巴胺层厚度为24nm,药物负载量为39%。
实施例15
按照如下步骤合成纳米胶囊型聚多巴胺药物
按照MIL-101@紫杉醇与多巴胺的质量比为1:1.5,随后加入Tris-HCl(pH=8.8)溶液,多巴胺最终浓度为4.5mg/mL,超声后得到母液。将母液置于20℃搅拌反应台上搅拌72h后取出,离心冷冻干燥,得到纳米胶囊型聚多巴胺药物,该药物大小为137nm,聚多巴胺层厚度为28nm,药物负载量为41%。
实施例16
按照如下步骤合成纳米胶囊型聚多巴胺药物
按照ZIF-90@阿霉素与多巴胺的质量比为1:0.8,随后加入Tris-HCl(pH=8.8)溶液,多巴胺最终浓度为7.5mg/mL,超声后得到母液。将母液置于35℃搅拌反应台上搅拌48h后取出,离心冷冻干燥,得到纳米胶囊型聚多巴胺药物,该药物大小为204nm,聚多巴胺层厚度为30nm,药物负载量为37%。
实施例17
按照如下步骤合成纳米胶囊型聚多巴胺药物
按照MIL-88A@阿霉素与多巴胺的质量比为1:5,随后加入Tris-HCl(pH=8.8)溶液,多巴胺最终浓度为1mg/mL,超声后得到母液。将母液置于35℃搅拌反应台上搅拌60h后取出,离心冷冻干燥,得到纳米胶囊型聚多巴胺药物,该药物大小为180nm,聚多巴胺层厚度为16nm,药物负载量为43%。
实施例18
按照如下步骤合成纳米胶囊型聚多巴胺药物
按照MIL-88A@去甲长春碱与多巴胺的质量比为1:4,随后加入Tris-HCl(pH=8.8)溶液,多巴胺最终浓度为1.5mg/mL,超声后得到母液。将母液置于50℃搅拌反应台上搅拌36h后取出,离心冷冻干燥,得到纳米胶囊型聚多巴胺药物,该药物大小为241nm,聚多巴胺层厚度为12nm,药物负载量为46%。
实施例19
按照如下步骤合成纳米胶囊型聚多巴胺药物
按照MIL-88A@长春地辛与多巴胺的质量比为1:4,随后加入Tris-HCl(pH=8.8)溶液,多巴胺最终浓度为9mg/mL,超声后得到母液。将母液置于45℃搅拌反应台上搅拌96h后取出,离心冷冻干燥,得到纳米胶囊型聚多巴胺药物,该药物大小为169nm,聚多巴胺层厚度为30nm,药物负载量为40%。
实验例1
结构表征
对实施例1中制备的各复合物进行结构表征
表征方法:透射电子显微扫描仪器:FEITecnai F20
图1给出了实施例1所得到产物的TEM图。
对本发明的其他实施例中制备的各复合物进行同样的结构表征,证明通过本发明的方法确实能够得到纳米胶囊型聚多巴胺药物。
实验例2
纳米胶囊型聚多巴胺药物的光热升温实验
配置实施例1所得到的纳米胶囊型聚多巴胺药物PBS(pH=7.4)的溶液,浓度为0~400微克每毫升。取一毫升溶液在808纳米激光器光照,激光功率为1.5W/cm2。每组光照600秒,溶液升温效果由红外温度记录仪进行记录。升温结果如图2所示。
该结果说明所制备得到的纳米胶囊型聚多巴胺药物具有较好的升温效果,有望用于癌症的光热治疗。
实验例3
纳米胶囊型聚多巴胺药物的药物释放实验
将1mL阿霉素浓度为1mg/mL的实施例1得到的纳米胶囊型聚多巴胺药物PBS(pH=7.4)溶液置于截留分子量为2000的透析袋中,分别置于49mL的PBS(pH=7.4)与PBS(pH=5.0)中。在37摄氏度摇床中以100rpm速度均匀摇晃,在特定时刻取样1mL后补充1mLPBS。另设一组置于PBS(pH=5.0)中,并每隔一小时光照5分钟,光照4次使用808nm激光器,激光功率为1.5W/cm2。图3为纳米胶囊型聚多巴胺药物的药物累积释放曲线。
可见本发明所得到的药物在激光照射下能够快速进行药物释放,从而达到近红外光介导下的药物可控释放。
对本发明其他实施例中制备的各复合物进行相同的药物释放实验,由于结构类似,因此释放曲线均与图3近似。
实验例4
5000个MCF-7乳腺癌细胞接种于96孔板中37摄氏度,5%CO2赋予过夜,分别加入1.25~40μg/mL阿霉素计量的实施例1所制备得到的纳米胶囊型聚多巴胺药物,共孵育4小时进行光照,使用808nm激光器,激光功率为1.5W/cm2,再孵育16小时后加入10μL CCK-8,后再孵育2-4小时,在450nm测读取吸光度,测定细胞存活率。不同浓度下及对照组的细胞抑制率如图4所示。
所得到的纳米胶囊型聚多巴胺药物能够实现光热及药物的联合治疗,从而达到联合治疗的目的。
实验例5
5000个PANC-1胰腺癌细胞接种于96孔板中37摄氏度,5%CO2赋予过夜,分别加入10μg/mL阿霉素计量的实施例1所制备得到的纳米胶囊型聚多巴胺药物,共孵育4小时进行光照,使用808nm激光器,激光功率为1.5W/cm2,再孵育16小时后加入10μL CCK-8,后再孵育2-4小时,在450nm测读取吸光度,测定细胞存活率。DOX组、PDA@DOX组、PDA+NIR组和PDA@DOX+NIR组的细胞抑制率分别为58%、69%、85%和61%。
所得到的纳米胶囊型聚多巴胺药物能够实现光热及药物的联合治疗,从而达到联合治疗的目的。
实验例6
5000个OS-RC-2-肾癌细胞接种于96孔板中37摄氏度,5%CO2赋予过夜,分别加入10μg/mL阿霉素计量的实施例1所制备得到的纳米胶囊型聚多巴胺药物,共孵育4小时进行光照,使用808nm激光器,激光功率为1.5W/cm2,再孵育16小时后加入10μL CCK-8,后再孵育2-4小时,在450nm测读取吸光度,测定细胞存活率。DOX组、PDA@DOX组、PDA+NIR组和PDA@DOX+NIR组的细胞抑制率分别为56%、66%、81%和59%。
所得到的纳米胶囊型聚多巴胺药物能够实现光热及药物的联合治疗,从而达到联合治疗的目的。
实验例7
5000个LM-3肝癌细胞接种于96孔板中37摄氏度,5%CO2赋予过夜,分别加入10μg/mL阿霉素计量的实施例1所制备得到的纳米胶囊型聚多巴胺药物,共孵育4小时进行光照,使用808nm激光器,激光功率为1.5W/cm2,再孵育16小时后加入10μL CCK-8,后再孵育2-4小时,在450nm测读取吸光度,测定细胞存活率。DOX组、PDA@DOX组、PDA+NIR组和PDA@DOX+NIR组的细胞抑制率分别为63%、72%、84%和65%。
所得到的纳米胶囊型聚多巴胺药物能够实现光热及药物的联合治疗,从而达到联合治疗的目的。
以上所述,仅是本申请的几个实施例,并非对本申请做任何形式的限制,虽然本申请以较佳实施例揭示如上,然而并非用以限制本申请,任何熟悉本专业的技术人员,在不脱离本申请技术方案的范围内,利用上述揭示的技术内容做出些许的变动或修饰均等同于等效实施案例,均属于技术方案范围内。
Claims (5)
1.一种纳米胶囊型聚多巴胺药物材料的制备方法,其特征在于,具体包括以下步骤:(1)将搭载化疗药物的有机金属框架材料与多巴胺按照质量比为1:0.5~5混合,随后加入Tris-HCl溶液,所述Tris-HCl溶液pH=8.8,使得多巴胺的浓度为0.2~10mg/mL,超声后得到母液;
(2)将步骤1中得到的母液置于磁力搅拌器上,在10~50℃条件下,反应24~120h;
(3)将步骤2中反应后的溶液离心分离、冷冻干燥,得到中空型纳米胶囊型聚多巴胺药物;聚多巴胺层厚度为5~30nm。
2.根据权利要求1所述的制备方法,其特征在于,步骤1中所述的有机金属框架材料为ZIF-8、ZIF-90、UIO-66、MIL-88A、MIL-100、MIL-101中的一种。
3.一种如权利要求1所述制备方法得到的纳米胶囊型聚多巴胺药物材料,其特征在于,所述药物材料包括搭载化疗药物的药物部分和纳米胶囊型聚多巴胺的外层载体部分。
4.根据权利要求3所述药物材料,其特征在于,所述药物材料的大小为50~300nm,药物负载量为20~60%。
5.根据权利要求3所述药物材料,其特征在于,所述化疗药物为阿霉素、顺铂、去甲长春碱、紫杉醇、丝裂霉素、长春地辛中的至少一种。
