一种淀粉基大肠靶向双层益生菌片的制备方法,属于益生菌片技术领域。本发明以淀粉为原料,经盐酸酸解、预干燥、焙烤制得不同特性的芯层和外层材料;将芯层材料与益生菌干粉及菌体活性保护剂按一定比例混合后,经过压片得到片芯,再添加一定比例的外层材料压片得到淀粉基大肠靶向双层益生菌片。该片剂中益生菌能在消化道存活且能保持有效活菌数,进而在大肠及结肠中定植,调节宿主肠内微生物菌群平衡,发挥益生作用。

技术领域
本发明涉及一种淀粉基大肠靶向双层益生菌片的制备方法,属于益生菌片技术领域。
背景技术
益生菌是一类能够促进宿主肠内微生物菌群的生态平衡,提高食物消化率、制造营养物质,进而发挥增强营养、调节免疫、预防疾病等作用,对宿主健康和生理功能产生有益功效的活性微生物制剂。随着人们健康意识的日益提高,饮食习惯已经从精细转变为绿色、健康,有关益生菌的研究也不断深入,益生菌相关的功能性食品、药物受到了消费者的广泛关注和推崇。其类型主要为添加了益生菌的液体奶、酸奶、以及片剂、胶囊剂和颗粒剂等剂型的膳食补充剂。然而这些产品在生产、加工、进入消化道的过程中往往易受各种消化酶、胃酸和小肠碱性环境等不利因素的影响,导致菌体大量死亡,极大地降低了益生菌的效价。
专利CN101843337A采用海藻酸钠包埋益生菌片,提高菌片中的活菌数及菌体存放时间;专利CN102370119A采用乳清为凝胶基质,添加益生菌制成益生菌凝胶,延长益生菌的存活期。然而无论是包埋还是凝胶方法,都受限于包埋涂层材料及应用载体,制作复杂,且益生菌在胃肠道中都会不同程度的被人体消化酶降解,效果不显著,其生物利用度仍有待提高。
结肠靶向给药即通过药物传输系统,使药物口服后,在上消化道不释放,将药物输送至人体回盲部后开始崩解或蚀解并释放出来,而在大肠发挥局部或全身治疗作用。
抗性糊精,又称为难消化糊精,不被人体消化酶类降解、酸热稳定性好,且兼具降低血糖、调节血脂、调整肠道环境等益生功能,而被作为新型水溶性膳食纤维广泛应用于各种功能食品中,其作为一类新开发的大肠靶向材料,正引起越来越多的关注。抗性糊精在焦糊精的基础上制备得到,淀粉在高温酸解的过程中,生成α-1,2和α-1,3 糖苷键,并在部分还原末端上有分子内脱水的缩葡聚糖及β-1,6糖苷键的存在,新键的生成使其不被人体内消化酶类降解,因此可以进入大肠,被结肠微生物菌群中的β-葡萄苷酸酶、β-葡萄苷酶、纤维素酶等酶系降解。
因此,本发明基于焦糊精的耐酶解、耐酸特性,提供一种淀粉基大肠靶向双层益生菌片剂的制备方法,制得的益生菌片抗消化性能好、益生菌的生物利用度高、靶向性好。
发明内容
本发明的目的在于克服上述不足之处,提供一种淀粉基大肠靶向双层益生菌片的制备方法。该片剂的芯层及外层材料是一类对淀粉进行改性制得的糊精分子,不易被酸、酶等降解,溶解度可调控,且使用本发明制备的辅料压制的益生菌片的大肠靶向性和生物利用度得到了显著提高。
本发明的技术方案,一种淀粉基大肠靶向双层益生菌片的制备方法,以淀粉为原料,经盐酸酸解、预干燥、焙烤制得不同特性的芯层和外层材料;将芯层材料与益生菌干粉及菌体活性保护剂按一定比例混合后,经过压片得到片芯,再添加外层材料压片,即得到淀粉基大肠靶向双层益生菌片。
具体步骤如下:
(1)酸解:以原淀粉为原料,加水制备质量浓度为40%-50%的淀粉浆,用盐酸调节淀粉浆pH值,搅拌均匀;
(2)预干燥:将步骤(1)中所得淀粉浆抽滤,预干燥至含水量5%-10%,研磨过100目筛,得到淀粉颗粒;
(3)高温焙烤:将步骤(2)中干燥后的淀粉颗粒经不同高温焙烤后,分别获得芯层和外层材料;
(4)水洗:将步骤(3)所得芯层材料和外层材料,水洗至pH中性;
(5)干燥过筛:将水洗后的芯层材料和外层材料干燥至含水量10%-15%后,再次分别粉碎过100目筛,分别得到芯层材料和外层材料;
(6)益生菌干粉制备:将益生菌菌株液体培养完毕后离心,收集菌体沉淀;配置冷冻保护剂溶液,按菌体沉淀体积计添加2-3倍冷冻保护剂溶液后混合均匀,真空冷冻干燥,得到益生菌干粉;
(7)压片:将步骤(6)所得益生菌干粉、菌体活性保护剂菊芋粉与步骤(5)所得芯层材料以质量比2-4:1-2:15的比例混匀后,压制成片芯;再添加片芯2-4倍重量的步骤(5)所得外层材料,经压片制得益生菌片。
步骤(1)所述淀粉原料为大米淀粉、玉米淀粉、木薯淀粉、马铃薯淀粉中的一种或几种。
步骤(1)中的酸解是用0.5M HCl溶液调节淀粉浆pH值至2.0-3.5;随后以200-400rpm的搅拌速度搅拌30min。
步骤(2)和步骤(5)中的干燥温度为45-65℃,时间为12-24h。
步骤(3)所述制备芯层材料的高温烘烤温度为180-200℃,烘烤时间为1-5h;制备外层材料的烘烤温度为120-150℃,烘烤时间为1-4h。
步骤(6)所述益生菌干粉具体为植物乳杆菌、保加利亚乳杆菌、肠球菌属、嗜热链球菌、双歧杆菌及乳酸菌中的一种或几种;
所述冷冻保护剂溶液具体包括15%-25%的脱脂奶粉、2%-3%的葡萄糖、0.5%-1.5%的蔗糖和1%-3%的甘油的水溶液。
步骤(6)中的离心条件为4-6℃,4000-8000rpm/min,10-30min;真空冷冻干燥的条件为0.01mBar,-80℃。
所述步骤(7)中的压片所得益生菌片中的菌体密度为108-1011cfu/g。
本发明的有益效果:本发明克服了现有益生菌产品在经消化道后益生菌存活率低且不能保持有效活菌数的不足的缺陷,具有口服经过消化道后可有效保持益生菌活性且具有良好的大肠靶向性的特性;经过实验验证,与现有益生菌片相比,本发明具有耐酸、耐(消化)酶解、靶向性好以及良好的加工操作性和稳定性等特点。

(1)酸解:称取100g蜡质玉米淀粉,加水调浆至50%淀粉溶液,用0.5M HCI调节淀粉溶液pH至2.0,在磁力搅拌器上以200rpm搅拌30min;
(2)预干燥:将步骤(1)中淀粉浆抽滤,滤渣破碎成小块,45℃干燥24h,研磨过100目筛;
(3)高温焙烤:将步骤(2)中干燥后的淀粉颗粒平分为两份,其中1份于120℃下焙烤4h,得到样品记为外层材料K1;另一份于180℃下焙烤5h记为芯层材料K2;
(4)水洗:将上述焙烤的样品K1和K2,分别水洗至pH中性;
(5)干燥过筛:将水洗后的K1和K2,于45℃下干燥20h,粉碎后过100目筛;
(6)益生菌干粉:将保加利亚乳杆菌菌株液体培养完毕后在4℃,4000rpm/min,20min条件下离心,收集菌体沉淀,配置冷冻保护剂溶液,保护剂溶液具体包括15%脱脂奶粉、2%葡萄糖、0.5%蔗糖和1%甘油的水溶液,按菌体沉淀体积计添加2倍冷冻保护剂溶液后混合均匀,在0.01mBar,-80℃条件下真空冷冻干燥,得到保加利亚乳杆菌干粉;
(7)压片:将保加利亚乳杆菌干粉、菌体活性保护剂菊芋粉与上述的芯层材料K2以2:1:15的比例混匀后,压制成片芯,再添加片芯3倍重量的外层材料K1,经压片制得保加利亚乳杆菌益生菌片,保持菌片中菌体密度为108cfu/g。
实施例2
(1)酸解:称取100g蜡质玉米淀粉,加水调浆至50%淀粉溶液,用0.5M HCI调节淀粉溶液pH至3.0,在磁力搅拌器上以300rpm搅拌30min;
(2)预干燥:将步骤(1)中淀粉浆抽滤,滤渣破碎成小块,55℃干燥18h,研磨过100目筛;
(3)高温焙烤:将步骤(2)中干燥后的淀粉颗粒平分为两份,其中1份于130℃下焙烤3h,得到外层材料;另一份于190℃下焙烤3h得到芯层材料;
(4)水洗:将上述焙烤的芯层材料和外层材料,分别水洗至pH中性;
(5)干燥过筛:将水洗后的芯层材料和外层材料,于45℃下干燥20h,粉碎后过100目筛;
(6)益生菌干粉:将植物乳杆菌菌株液体培养完毕后在4℃,4000rpm/min,20min条件下离心,收集菌体沉淀,配置冷冻保护剂溶液,保护剂溶液具体包括20%脱脂奶粉、2.5%葡萄糖、1%蔗糖和2%甘油的水溶液,按菌体沉淀体积计添加2倍冷冻保护剂溶液后混合均匀,在0.01mBar,-80℃条件下真空冷冻干燥,得到植物乳杆菌干粉;
(7)压片:将植物乳杆菌干粉、菌体活性保护剂菊芋粉与上述的芯层材料以2:1:15的比例混匀后,压制成片芯,再添加片芯3倍重量的外层材料,经压片制得植物乳杆菌益生菌片,保持菌片中菌体密度为109cfu/g。
实施例3
(1)酸解:称取100g蜡质玉米淀粉,加水调浆至50%淀粉溶液,用0.5M HCI调节淀粉溶液pH至3.0,在磁力搅拌器上以300rpm搅拌30min;
(2)预干燥:将步骤(1)中淀粉浆抽滤,滤渣破碎成小块,60℃干燥14h,研磨过100目筛;
(3)高温焙烤:将步骤(2)中干燥后的淀粉颗粒平分为两份,其中1份于140℃下焙烤2h,得到外层材料;另一份于195℃下焙烤2h得到芯层;
(4)水洗:将上述焙烤的芯层和外层材料,分别水洗至pH中性;
(5)干燥过筛:将水洗后的芯层和外层材料,于45℃下干燥20h,粉碎后过100目筛;
(6)益生菌干粉:将乳酸菌菌株液体培养完毕后在4℃,4000rpm/min,20min条件下离心,收集菌体沉淀,配置冷冻保护剂溶液,保护剂溶液具体包括20%脱脂奶粉、2.5%葡萄糖、1%蔗糖和2%甘油的水溶液,按菌体沉淀体积计添加2倍冷冻保护剂溶液后混合均匀,在0.01mBar,-80℃条件下真空冷冻干燥,得到乳酸菌干粉;
(7)压片:将乳酸菌干粉、菌体活性保护剂菊芋粉与上述的芯层材料;以2:1:15的比例混匀后,压制成片芯,再添加片芯3倍重量的外层材料;,经压片制得乳酸菌益生菌片,保持菌片中菌体密度为1010cfu/g。
实施例4
(1)酸解:称取100g蜡质玉米淀粉,加水调浆至40%淀粉溶液,用0.5M HCI调节淀粉溶液pH至3.5,在磁力搅拌器上以400rpm搅拌30min;
(2)预干燥:将步骤(1)中淀粉浆抽滤,滤渣破碎成小块,65℃干燥12h,研磨过100目筛;
(3)高温焙烤:将步骤(2)中干燥后的淀粉颗粒平分为两份,其中1份于140℃下焙烤2h,得到外层材料;另一份于195℃下焙烤2h记为芯层;
(4)水洗:将上述焙烤的样品芯层和外层材料,水洗至pH中性;
(5)干燥过筛:将水洗后的芯层和外层材料,于45℃下干燥20h,粉碎后过100目筛;
(6)益生菌干粉:将双歧杆菌菌株液体培养完毕后在4℃,4000rpm/min,20min条件下离心,收集菌体沉淀,配置冷冻保护剂溶液,保护剂溶液具体包括20%脱脂奶粉、2.5%葡萄糖、1%蔗糖和2%甘油的水溶液,按菌体沉淀体积计添加2倍冷冻保护剂溶液后混合均匀,在0.01mBar,-80℃条件下真空冷冻干燥,得到双歧杆菌干粉;
(7)压片:将双歧杆菌干粉、菌体活性保护剂菊芋粉与上述的芯层材料以2:1:15的比例混匀后,压制成片芯,再添加片芯3倍重量的外层材料,经压片制得双歧杆菌益生菌片,保持菌片中菌体密度为1011cfu/g。
实施例5
(1)酸解:称取100g蜡质玉米淀粉,加水调浆至40%淀粉溶液,用0.5M HCI调节淀粉溶液pH至3.5,在磁力搅拌器上以400rpm搅拌30min;
(2)预干燥:将步骤(1)中淀粉浆抽滤,滤渣破碎成小块,65℃干燥12h,研磨过100目筛;
(3)高温焙烤:将步骤(2)中干燥后的淀粉颗粒平分为两份,其中1份于150℃下焙烤1h,得到外层材料;另一份于200℃下焙烤1h得到芯层;
(4)水洗:将上述焙烤的芯层和外层材料,水洗至pH中性。
(5)干燥过筛:将水洗后的芯层和外层材料,于45℃下干燥20h,粉碎后过100目筛。
(6)益生菌干粉:将嗜热链球菌菌株液体培养完毕后在4℃,4000rpm/min,20min条件下离心,收集菌体沉淀,配置冷冻保护剂溶液,保护剂溶液具体包括25%脱脂奶粉、3%葡萄糖、1.5%蔗糖和3%甘油的水溶液,按菌体沉淀体积计添加2倍冷冻保护剂溶液后混合均匀,在0.01mBar,-80℃条件下真空冷冻干燥,得到嗜热链球菌干粉;
(7)压片:将嗜热链球菌干粉、菌体活性保护剂菊芋粉与上述的芯层材料以2:1:15的比例混匀后,压制成片芯,再添加片芯3倍重量的外层材料,经压片制得嗜热链球菌益生菌片,保持菌片中菌体密度为1011cfu/g。
以实施例1为例对药物载体材料的特性进行测试,相关测试和结果如下:
1. 片剂芯层和外层材料结构表征:
1)颗粒结构的扫描电镜:采用JSM-6700F 型扫描电子显微镜(SEM)观察样品表面形态,测试条件:管压20 kV,样品测试前经过喷金预处理。放大倍数为500倍。
如图1所示,分别为原淀粉、片剂外层材料K1和片剂芯层材料K2的扫描电镜图,可见,经焙烤处理的样品K1和K2与淀粉一样,仍保持淀粉的颗粒结构。
2. 片剂芯层和外层材料的溶解性:
1)溶解性测试:称量0.4000g样品于50mL离心管中,加入40mL去离子水,混合均匀。悬浮液在25℃下搅拌30min,以9000rpm的转速离心10min。取10mL上清液移至已知重量的称量瓶中,干燥至恒重。平行三次。溶解度(S)计算公式:
a: 样品重量(0.4000g);b:干燥后残渣重量。
如图2所示,原淀粉和片剂外层材料K1的溶解性相似,而芯层材料K2的溶解性高达99%,有利于芯层到达大肠后快速释放。
Claims (9)
1.一种淀粉基大肠靶向双层益生菌片的制备方法,其特征在于:以淀粉为原料,经盐酸酸解、预干燥、焙烤制得不同特性的芯层和外层材料;将芯层材料与益生菌干粉及菌体活性保护剂按一定比例混合后,经过压片得到片芯,再添加外层材料压片,即得到淀粉基大肠靶向双层益生菌片。
2.根据权利要求1所述淀粉基大肠靶向双层益生菌片的制备方法,其特征在于具体步骤如下:
(1)酸解:以原淀粉为原料,加水制备质量浓度为40%-50%的淀粉浆,用盐酸调节淀粉浆pH值,搅拌均匀;
(2)预干燥:将步骤(1)中所得淀粉浆抽滤,预干燥至含水量5%-10%,研磨过100目筛,得到淀粉颗粒;
(3)高温焙烤:将步骤(2)中干燥后的淀粉颗粒经不同高温焙烤后,分别获得芯层和外层材料;
(4)水洗:将步骤(3)所得芯层材料和外层材料,水洗至pH中性;
(5)干燥过筛:将水洗后的芯层材料和外层材料干燥至含水量10%-15%后,再次分别粉碎过100目筛,分别得到芯层材料和外层材料;
(6)益生菌干粉制备:将益生菌菌株液体培养完毕后离心,收集菌体沉淀;配置冷冻保护剂溶液,按菌体沉淀体积计添加2-3倍冷冻保护剂溶液后混合均匀,真空冷冻干燥,得到益生菌干粉;
(7)压片:将步骤(6)所得益生菌干粉、菌体活性保护剂菊芋粉与步骤(5)所得芯层材料以质量比2-4:1-2:15的比例混匀后,压制成片芯;再添加片芯2-4倍重量的步骤(5)所得外层材料,经压片制得益生菌片。
3.根据权利要求2所述淀粉基大肠靶向双层益生菌片的制备方法,其特征在于:步骤(1)所述淀粉原料为大米淀粉、玉米淀粉、木薯淀粉、马铃薯淀粉中的一种或几种。
4.根据权利要求2所述淀粉基大肠靶向双层益生菌片的制备方法,其特征在于:步骤(1)中的酸解是用0.5M HCl溶液调节淀粉浆pH值至2.0-3.5;随后以200-400rpm的搅拌速度搅拌30min。
5.根据权利要求2所述淀粉基大肠靶向双层益生菌片的制备方法,其特征在于:步骤(2)和步骤(5)中的干燥温度为45-65℃,时间为12-24h。
6.根据权利要求2所述淀粉基大肠靶向双层益生菌片的制备方法,其特征在于:步骤(3)所述制备芯层材料的高温烘烤温度为180-200℃,烘烤时间为1-5h;制备外层材料的烘烤温度为120-150℃,烘烤时间为1-4h。
7.根据权利要求2所述淀粉基大肠靶向双层益生菌片的制备方法,其特征在于:步骤(6)所述益生菌干粉具体为植物乳杆菌、保加利亚乳杆菌、肠球菌属、嗜热链球菌、双歧杆菌及乳酸菌中的一种或几种;
所述冷冻保护剂溶液具体包括15%-25%的脱脂奶粉、2%-3%的葡萄糖、0.5%-1.5%的蔗糖和1%-3%的甘油的水溶液。
8.根据权利要求2所述淀粉基大肠靶向双层益生菌片的制备方法,其特征在于:步骤(6)中的离心条件为4-6℃,4000-8000rpm/min,10-30min;真空冷冻干燥的条件为0.01mBar,-80℃。
9.根据权利要求2所述淀粉基大肠靶向双层益生菌片的制备方法,其特征在于:所述步骤(7)中的压片所得益生菌片中的菌体密度为108-1011cfu/g。
