Abstract

背景技术
全世界约有六千万患者患有青光眼,这是一种毁灭性的疾病,无法治愈,导致全球800万人双侧失明。目前的疗法只能减缓疾病的进展。导致失明的最主要的风险因素是眼内压升高。

用于本发明小鼠的构建体、引物和组分与用于先前在美国专利申请号14/790,884(公开号US 2016/0000871 A1)中描述的小鼠的相同。根据该参考文献,已知A1A2FloxWB ΔE16.5(cKO或条件性敲除)小鼠发生双侧眼积水且在促血管生成素1和促血管生成素2条件性敲除小鼠(“Ang1/2条件性敲除小鼠”)中缺少施勒姆管和眼内压(“IOP“)升高。
Angpt/Tie2信号传导在管形成中具有剂量依赖性作用。虽然Angpt 1/2双敲除完全缺少施勒姆管,但Angpt 1敲除的小鼠仅具有保留一些管组织的亚型表型。单独的Angpt2敲除没有效果,这表明Angpt 1是主要配体,而Angpt 2可提供一些补偿。眼压(“IOP”)测量值验证了这些组织学结果,因为Angpt 1敲除小鼠的压力升高(尽管不像双敲除那样升高),而Angpt 2敲除是正常的。Tie2活化(即Angpt/Tie2信号传导的水平)对管形成具有剂量依赖性影响。

Claims (11)
1.一种产生具有降低的VE-PTP的小鼠的方法,其包括用VE-PTP无效等位基因替换至少一个野生型VE-PTP等位基因。
