Abstract

发明领域
本发明涉及用于评估候选化合物的代谢作用的方法和/或测定法。该方法和/或测定包括将一种或多种三维细胞培养物或组织暴露于候选化合物。
线粒体在产生大部分细胞能量(如ATP)的方面发挥着关键作用。它们还参与其他代谢过程,例如尿素生成、血红素合成和脂肪酸β-氧化。药物对线粒体功能的破坏可通过坏死导致细胞死亡或可通过细胞凋亡(例如,细胞色素c释放后)预示细胞死亡。
损伤线粒体的药物通常通过抑制电子链的呼吸复合物;抑制或解耦氧化磷酸化;诱导线粒体氧化应激;或抑制DNA复制、转录或翻译来实现。
在药物开发早期测试线粒体毒性是重要的,因为线粒体功能的损害可诱发危及生命的各种病理状况或可增加现有线粒体疾病的进展。
据报道,线粒体损伤涉及、造成或至少与不同类型的疾病有关,包括帕金森病、阿尔茨海默病、心脏病、癌症以及胆固醇和脂质紊乱。
出于这个原因,已经开发了线粒体毒性测定。其中的一些,如Promega提供的线粒体ToxGloTM测定,包含使用单个细胞悬液的基于细胞的测定方法。该测定基于在短期暴露期间与载体处理的对照细胞相比与细胞膜完整性和细胞ATP水平的变化相关的生物标志物的差异测量。首先通过测量与坏死相关的独特蛋白酶活性的存在或不存在来评估细胞膜完整性。接下来,在细胞裂解后测量ATP。可组合这两组数据以产生代表线粒体功能障碍或非线粒体相关细胞毒性机制的谱。
其他线粒体毒性测定使用2D细胞培养物,其中细胞例如被接种在胶原蛋白I包被的微量培养板中并在加入化合物之前粘附18-24小时。对于初步筛选和二次剂量反应性实验,将特定浓度的化合物添加到接种的细胞中,并在37℃、5%CO2下孵育24小时。然后,添加细胞标记物以进行成像细胞计数分析,这导致线粒体毒性的测量(Tsiper等,2012)。
本发明的一个目的是改进现有技术的方法。本发明的另一个目的是提高现有技术方法的可预测性。本发明的另一个目的是进一步开发现有技术的方法,使得它们被用于研究细胞长期暴露于候选化合物的作用。本发明的另一个目的是增加现有技术方法的灵敏度,使得它们提供对候选化合物的线粒体毒性更灵敏和精确的预测。本发明的另一个目的是在尚未影响细胞活力且不能在根据现有技术的细胞活力/细胞毒性测定或线粒体毒性测定中检测到的毒素浓度下检测线粒体损伤。

发明内容

b)在至少一种三维细胞培养物或组织中测量这种暴露对三维细胞培养物或组织特异性呼吸速率(MTSRR)的作用。
术语“三维细胞培养物或组织”和“3D微组织”在本文中可互换使用。令人惊讶的是,本发明人已经表明,通过该测定可在尚未影响细胞活力且不能在根据现有技术的细胞活力/细胞毒性测定或线粒体毒性测定中检测到的毒素浓度下检测线粒体损伤。
发明人将该发现归因于这样的事实:三维细胞培养物或组织是天然组织的更忠实的复制。换句话说:它比例如2D细胞培养物或细胞悬液的表现更具生理性。三维细胞培养物或组织概括了组织或器官的最小功能性单元,提供了天然组织的形态和功能性类似物。因此,它们弥合了体外和体内毒性测试模型之间的差距。不受理论束缚,本发明人用不同的作用解释了这种现象。首先,在三维细胞培养物或组织中,可建立间质和细胞外结构。其次,在三维细胞培养物或组织中,细胞可发展三维细胞-细胞相互作用。第三,在三维细胞培养物或组织中,待测试的药物必须通过从外部扩散进入组织,因此更多地影响外细胞团中的细胞而不是内细胞团中的细胞。所有这些现象在2D细胞培养物和/或细胞悬液中都不存在。
在本发明的一个实施方案中,所述方法和/或测定还包括
c)在相同的或至少一种其他三维细胞培养物或组织中测量这种暴露在至少一种三维细胞培养物或组织的一个或多个细胞中对细胞活性和/或细胞毒性效应(CV-CYT)的作用,和
d)比较步骤b)和c)的作用以提供对一种或多种候选化合物的线粒体毒性的估计。
步骤a)和b)、步骤a)和c)以及步骤b)和c)可同步进行。在另一个实施方案中,步骤a)可在步骤b)和c)之前进行,其可同步或随后进行。
如已经讨论的,现有技术的线粒体毒性测定使用细胞悬液或2D细胞培养物。本发明人意外地表明,3D微组织表现出备用呼吸容量,其更能代表体内情况,即真实组织。因此,3D微组织的使用提供了对候选化合物的线粒体毒性更灵敏和精确的预测。
通常,在药物研发、临床前试验和临床试验中估计花费在5亿美元到12亿美元之间。很有可能药物在任何研发阶段会失败。与安全性相关的问题仍然是药物失败的主要原因,主要是因为预测哪些药物会被证实对患者产生毒性非常困难。上面讨论的标准测定具有有限的通量、细胞活力的丧失和可测量的组织特异性功能性降低的缺陷。体内动物试验是昂贵的,引起道德和伦理问题,并且经常不能揭示人类的重要毒性迹象。在本发明的另一个实施方案中,所述方法和/或测定因此用于确定候选化合物的线粒体毒性。在该实施方案中,候选化合物是例如候选药物,其中在毒性筛选过程中确定潜在的毒性作用或其他副作用。该实施方案还涵盖其他化合物,如除草剂、杀真菌剂或杀虫剂。

如上所述,三维细胞培养物或组织在本发明的上下文中具有显著的优点,因为它们是天然组织的更忠实的复制,并且它比例如2D细胞培养物或细胞悬液表现得更具生理性。此外,它们具有比2D细胞培养物或细胞悬液更长的寿命,这有助于处理并且允许测试候选化合物的长期作用和脱靶作用。这在筛选可能随后用于长期治疗的化合物或人类长时间暴露的化合物中特别有用。
在本发明的另一个实施方案中,所述方法和/或测定三维细胞培养物或组织包含原代细胞。与肿瘤细胞或永生化细胞相比,原代细胞没有累积的突变,且因此对于体内组织的行为更具代表性或预测性。将原代细胞转移到三维细胞培养物或组织中具有两个显著优点:a)它进一步增加细胞与体内组织的生理相似性,从而使它们更具代表性或预测性,并且b)与2D细胞培养物或细胞悬液相比它增加了原代细胞的寿命。
优选地,通过悬滴微量滴定板产生三维细胞培养物或组织,所述悬滴微量滴定板由瑞士Schlieren的InspheroAG以“GravityPLUS TM悬滴系统”品牌发售,并在国际专利申请WO2010031194A1中公开。该方法允许在更复杂的3D细胞培养情况下产生三维细胞培养物或组织,例如当使用原代细胞时、当使用抗自身聚集的细胞系时或当产生共培养微组织时。
在另一个实施方案中,三维细胞培养物或组织在超低附着(ULA)微量滴定板中产生,由瑞士Schlieren的InspheroAG以“GravityTRAP TM Ultra-Low Attachment(ULA)Plate”的品牌发售。
在本发明的另一个实施方案中,所述方法和/或测定的三维细胞培养物或组织包含肝细胞。原代培养物中的肝细胞提供最接近人体肝脏的体外模型,人体肝脏是受药物毒性影响最大的器官,并且是唯一能够对给定药物产生与体内发现类似的代谢谱的模型。原代培养中的肝细胞表达典型的肝功能并表达药物代谢酶。这些优点尤其存在于三维细胞培养物或组织中,如发明人惊奇地示出其表现得更具生理性,且因此更忠实地反映了“真正的”肝脏组织对药物暴露的反应。
在其他实施方案中,细胞可以是心肌细胞、神经元细胞、胰岛、成纤维细胞和角质形成细胞。由这些细胞类型制成的三维细胞培养物或组织特别应用在药物毒性或有效性筛选中。
在本发明的另一个实施方案中,三维细胞培养物或组织包含永生化细胞、恶性细胞、癌性细胞和/或赘生性细胞。
在该实施方案中,先前暴露对所述细胞的微组织特异性呼吸速率(MTSRR)的作用可用于研究测试分子对癌细胞的潜在化疗有效性。
此外,癌细胞固有地改变它们的细胞机器以改变它们消耗和利用葡萄糖的方式。抑制细胞呼吸中的癌症特异性关键步骤可破坏癌细胞的增殖和存活而不影响正常细胞。
在本发明的另一个实施方案中,该方法和/或测定还包括在相同或至少一种其他三维细胞培养物或组织中测定暴露对其尺寸的作用的步骤。该步骤可与步骤a)、b)和/或c)同时进行或随后进行。
优选地,尺寸确定是指选自以下的至少一个参数:

因此,尺寸可以是直接测量的参数,或者是基于这种测量计算的参数。
优选地,通过成像装置进行三维细胞培养物或组织的尺寸确定。
这种成像装置的一个例子是由InSphero AG,Schlieren,CH发售并由日本SCREEN制造的Cell3iMager。它允许通过在明场中扫描多孔板来分析球状体。它基于球状体尺寸和密度以及每个孔中的球状体数量和面积计算估计值。凭借简单高效的可操作性,其无振动的设计保护细胞免受损坏。一种优秀的应用也可用于确定球状体随时间的增殖并测量3D培养物中的颗粒分布。
在另一个实施方案中,所述三维细胞培养物或组织还包含至少一种选自以下的细胞类型:
·免疫细胞,
·间质细胞,
·成纤维细胞,
·癌症干细胞,
·已知对给定的抗癌剂具有抗性或敏感性的永生化细胞、恶性细胞、癌性细胞和/或赘生性细胞,
在这种三维细胞培养物或组织中使用免疫细胞具有特别的益处,因为还可研究先天免疫系统的作用。在研究与免疫系统共同作用的抗癌剂,即免疫治疗剂(如免疫检查点抑制剂(见下文))时,这是特别有用的。这些药物激活免疫系统以靶向癌细胞。因此,为了测试它们在体外的有效性,并研究潜在的复发,这些试剂需要免疫细胞的存在。
可用于这种共培养的合适的免疫细胞的实例尤其包括组织驻留白细胞(tissueresident leukocyte)如巨噬细胞(例如,星状巨噬细胞/枯否细胞)、CD8+T细胞、天然杀伤细胞、天然杀伤T细胞和/或调节性T细胞。
使用结缔组织成分,如间质细胞或成纤维细胞,可帮助在体外重建重要的肿瘤微环境,从而提高三维细胞培养物或组织的质量,并提高了这种培养物或组织对于测定的预测价值或筛选目的。
癌症干细胞(CSC)是具有与正常干细胞相关的特征的癌细胞。癌症干细胞有时与正常的体细胞干细胞具有共同特征,体现在它们对化疗剂具有天然抗性,因为它们表达不同的主动运出药物的转运蛋白。因此,CSC通常能够在一线癌症治疗中存活并因此导致肿瘤复发。
因此,将永生化细胞、恶性细胞、癌性细胞和/或赘生性细胞与癌症干细胞共培养可帮助开发影响原发性肿瘤并同时抑制癌症干细胞的疗法。
同样的原理适用于给定的永生化细胞、恶性细胞、癌性细胞和/或赘生性细胞类型与另一种已知对给定的抗癌剂具有抗性或敏感性的永生化细胞、恶性细胞、癌性细胞和/或赘生性细胞类型的共培养。
在一个实施方案中,微组织特异性呼吸速率通过同时测量一个或多个分离的3D微组织的氧消耗速率(OCR)和细胞外酸化速率(ECAR)来确定。
OCR是线粒体呼吸的指标,而ECAR主要是糖酵解的结果。这种测量可例如在Seahorse XF分析仪中进行,市场上存在其他合适的装置并且同样可使用。Seahorse XF分析仪能够在通常是微量培养板孔中的单层细胞之上的极小体积(约2μL)的培养基中进行OCR和ECAR的实时测量。细胞氧消耗(呼吸)和质子排泄(糖酵解)导致在这种“瞬态微室”中溶解氧和游离质子浓度的快速的、易于测量的变化,每隔几秒由位于单层细胞上方200微米的固态传感器探针测量。仪器测量浓度2-5分钟,并然后分别计算OCR和/或ECAR,例如以pmol/分钟(OCR)和mpH/分钟(ECAR)为单位。总测定时间通常为60至90分钟。
本发明人首次表明,在该方法中,出乎意料地,可使用3D微组织代替二次文献以及产品手册中公开的单层细胞。由于孔提供的微升体积,这种孔中的球状微组织提供了与孔底部上的单层细胞完全不同的微环境。因此,不可预见将相应方法转移到3D微组织是可行的且提供有意义且可再现的结果。
在一个实施方案中,在确定微组织特异性呼吸速率(MTSRR)时,使用备用呼吸容量(“SPARE”spare respiratory capacity)。
备用呼吸容量表示细胞响应能量需求的容量以及细胞呼吸至接近理论最大值的程度。备用呼吸容量定义为组织和/或细胞的最大呼吸和基础呼吸之间的差异。
为了确定备用呼吸容量,一种方法是首先测量基础呼吸速率,并然后测量最大呼吸速率。两者之间的算术差异(“delta”)是备用呼吸容量。
基础呼吸速率显示基线条件下组织或细胞的能量需求,而最大氧消耗反映组织或细胞可达到的最大呼吸速率。
基础呼吸速率可通过测量的第一步中的氧消耗速率(OCR)的测量来确定。最大呼吸速率可通过测量用解联剂如羰基氰-4(三氟甲氧基)苯腙(FCCP)处理的组织或细胞中的氧消耗速率(OCR)来确定。这种解联剂通过摧毁质子梯度和破坏线粒体膜电位刺激呼吸链以使其以最大容量运行来模拟生理能量需求。因此,通过电子传输链的电子流不受抑制,并且复合物IV最大程度地消耗氧。
根据另外的实施方案,在确定微组织特异性呼吸速率(MTSRR)时,还使用基础呼吸速率(“BASAL”)和/或最大呼吸速率(“MAX”)。
主要是,备用呼吸容量(SPARE)用于确定微组织特异性呼吸速率(MTSRR),基础呼吸速率(BASAL)和最大呼吸速率(MAX)在与SPARE相比不准确或更高灵敏度的情况下使用。
在一个实施方案中,细胞活力/细胞毒性效应的测量确定一个或多个细胞的残余活力。
细胞活力/细胞毒性效应可例如通过定量细胞中存在的ATP来确定。一种这样的测定是CellTiter-Glo发光细胞活力测定(Promega,Madison,US),而其他细胞活力/细胞毒性测定存在于市场上并且同样可以使用。
在另一个实施方案中,对微组织特异性呼吸速率的作用和/或对细胞活力/细胞毒性效应的作用被确定为抑制浓度,优选为半数最大抑制浓度(IC50)。
所述抑制浓度是指与未处理的三维细胞培养物或组织相比,在步骤a)中三维细胞培养物或组织暴露于其中的候选化合物的浓度。
本文中使用的关于微组织特异性呼吸速率的IC50的变量是IC50-MTSRR。如上所述,MTSRR优选地通过确定备用呼吸容量(SPARE)来确定。在这种情况下,IC50_MTSRR将等于IC50_SPARE。
本文中使用的关于细胞活力/细胞毒性的IC50的变量是IC50-CV/CYT。
在另一个实施方案中,暴露步骤a)在随后的测量步骤b)和/或c)之前进行。
该实施方案允许不同长度的暴露时间。通过这种方式,可研究短期作用和长期作用。该实施方案与要求保护的三维细胞培养物或组织的使用具有令人惊讶的协同作用,如上所述,与2D细胞培养物或细胞悬液相比,其具有更长的寿命,且因此适于长期暴露于候选化合物(如候选药物)。
与此相反,Seahorse XF分析仪的使用说明书规定在测定期间将测试药物分配到细胞中。为此目的,该装置提供了一种测定卡匣(assay cartridge),其可容纳多达四种药物用于在测定期间自动注射。首先将测定卡匣放入分析仪中以允许光学传感器的自动校准。然后,将细胞培养板插入仪器中。然而,在这种设置中,不能进行长期暴露测试。
在又一个实施方案中,暴露步骤a)在一个或多个第一容器中进行,而随后的测量步骤b)和/或c)在一个或多个第二和/或第三容器中进行。
优选地,在第一阵列的容器(例如,第一微量滴定板的孔)中进行暴露,而随后的测量在第二阵列的容器(例如,第二微量滴定板的孔)中进行。
在另一个实施方案中,在暴露步骤a)之后,通过合适的分配装置将微组织从一个或多个第一容器转移到一个或多个第二和/或第三容器,以进行测量步骤b)和/或c)。
所述分配装置是例如具有移液枪尖的多通道移液管,所述移液枪尖是非蛋白质结合的或预包被有蛋白质的,以避免微组织粘附或附着到移液枪尖的内表面或外表面。
在另一个实施方案中,线粒体毒性基于以下之间的定量关系确定
a)关于微组织特异性呼吸速率的IC50(IC50_MTSRR),
b)关于细胞活力/细胞毒性的IC50(IC50_CV/CYT)。
优选地,
a)IC50_MTSRR≥IC50_CV/CYT表示候选化合物没有特定的线粒体毒性,
b)IC50_MTSRR<IC50_CV/CYT且IC50-MTSRR≥0.75 x IC50_CV/CYT表示候选化合物具有低线粒体毒性风险,

实施例1:3D微组织的产生
1.1.解冻冷冻保存的肝细胞
·从冷冻罐中取出选定的细胞小瓶,并转移到水浴(37℃)中;设定计时器2分钟
·将40ml洗涤/解冻培养基移液入50ml管中
·将肝细胞转移到管中,用1ml培养基洗涤
·将50ml管放入离心机中;在室温下以50rcf(相当于600rpm)离心5分钟。
·去除上清液
·用20ml洗涤缓冲液洗涤沉淀
·将3ml洗涤/解冻培养基小心地放入50ml管中
·用2D细胞培养基重悬细胞沉淀
·用例如台盼蓝计数肝细胞
1.2.肝细胞预接种
·使用胶原蛋白包被的细胞培养皿进行预接种
·将肝细胞接种在6厘米培养皿中:100000-250000个肝细胞/cm2;0.05-0.5ml/cm2
·置于37℃的含CO2的培养箱中
·可选地:与无血清2D培养基连接后交换培养基
·12-96小时后收获肝细胞(如超过12小时每日更换培养基)
·用预热的磷酸盐缓冲盐水洗涤3次,去除非粘附的肝细胞
·将细胞分离溶液(如胶原酶/细胞消化液(accutase)混合物)添加到培养皿中,将肝细胞从孔中移出50-500μl/cm2
·在37℃孵育5-30分钟(目视观察,直到大部分细胞从表面脱离)
·小心地将分散的肝细胞转移到预装有洗涤缓冲液的50ml管中
·在室温下以50rcf(相当于600rpm)离心5分钟
·吸出上清液,在沉淀中加入1-50ml洗涤培养基,并轻轻转动管重悬沉淀
·在室温下用50rcf(相当于600rpm)重复离心步骤5分钟
·向沉淀中加入1ml再聚集培养基,并通过轻轻转动管溶解肝细胞
·计数肝细胞
1.3.产生微组织
A.GravityPLUSTM板的制备(Insphero AG,CH)
·在Omnitray中加入15ml 0.75x PBS/两性霉素并加入加湿垫
·将框架放在Omnitray上
·将板盖上盖子并标记板
B.细胞悬液的制备
·在falcon管中制备确定细胞数(即25′000肝细胞/25′000NPC/ml)的细胞悬液
·通过转动管使细胞悬液均质化
C.接种
·使用Viaflo电子多通道移液管(Integra Bioscience)
·设置Viaflo参数:重复分配:40μl,速度3,重复:1x(适用于96孔Viaflo)
·清空储存器中的细胞悬液(不要在储液器中倒入超过30ml,以确保细胞正确分布)
·从GravityPLUS板上取下盖子
·在吸入细胞悬液前,轻轻转动储存器,使细胞均匀分布
·启动Viaflo程序:重复分配
·将Viaflo水平放置在插入物上,通过轻柔地将Viaflo压入插入物避免轻敲位于顶部的液滴。
·将盖子放回板上
D.微组织的转移
·预先润湿GravityTRAP板:在每个孔的最底部加入30-50μl培养基或PBS,且通过抽吸取出或者通过轻弹取出(使用浸泡垫)
·将GravityPLUS板放在接收器底板旁边。将盖子和框架二者从储存器中取出,并小心地将框架的插头(仍然盖着盖子)锁定到接收器板的所有3个槽口中
·使用70μl培养基,其会释放内部具有微组织的悬滴
·“坠落”的液滴具有约为60–70μl的体积
·用盖子关闭接收器板,并以250x RCF离心板30秒-1分钟。这样可以去除捕获的气泡,且微组织位于孔底
·在显微镜下检查转移效率
·培养基补充:通过将枪尖放在GravityTRAP板的壁架上吸取培养基并移除培养基
·用50-80μl培养基重新填充GravityTRAP板的孔
1.4.使用的培养基
实施例2:使用Seahorse XF分析仪测量线粒体毒性
2.1 3D微组织的处理
在产生直径大小为370μm的3D原代人肝细胞微组织(IPHH_02)后,将微组织与不同浓度的化合物一起孵育48小时。从而将化合物溶解在3D InSightTM人肝维持培养基-TOX中。
2.2 Seahorse XF分析仪测量
A.制备Seahorse测定培养基
调至pH为7.4
B.Seahorse球状体板的包被
-将20μL聚-D-赖氨酸(在细胞培养级水中稀释成100μg/mL)加入Seahorse球状体板的每个孔中(室温下孵育20分钟)
-包被后,用200μL无菌水洗涤Seahorse球状体板并除去洗涤液(洗涤步骤重复一次)
-风干Seahorse球状体板几分钟
C.将微组织加入Seahorse球状体板
-将准备好的Seahorse测定培养基(见上文)加热至37℃,并用70μL填充板
-使用Seahorse捕捉筛选插入工具(Capture Screen Insert Tool)将微组织准确定位在板中
-在使用前,用FCS冲洗移液器头几次
-FCS包被后,以20μL体积将微组织浸泡在GravityTRAP板
-将移液器枪头转移到Seahorse捕捉筛选插入工具的孔中
-将枪头留在工具中约1分钟,以确保球状体完全转移
-球状体通过重力转移到孔中,因此它们会自动从枪尖中脱落,且您不必使用触发器
-完成微组织转移后,用预热的Seahorse测定培养基填充孔至最终体积为180μL
-扫描Insphero Cell3iMager中的板以在Seahorse运行之前定位板上的微组织
-在37℃无CO2的条件下孵育微组织1小时
D.Seahorse XF分析仪运行
-根据Seahorse制造商方案,在细胞孵育期间填充卡匣的端口
-设置测量的组和参数(基础:7个循环、寡霉素2μM:6个循环、FCCP 0.5μM:3个循环、FCCP 1μM:3个循环、Rot AA 0.5μM:9个循环)
-开始测量
E.细胞活力评估
-在Seahorse XF分析仪运行的同时,测量了以与Seahorse实验相同的方式处理的不同微组织的细胞活力
E.以胺碘酮安全性的分析和预测为例
-通过生成每个胺碘酮浓度的所有呼吸备用容量值的Sigmoidal剂量-反应曲线(可变斜率),我们能够获得IC50_MTSRR为25.3μM
-通过了解胺碘酮的CMax为5.3μM并将IC50_MTSRR除以CMax,我们能够产生4.8的安全边际(MOS)
-由于胺碘酮的MOS(4.8)低于30,我们预测胺碘酮是一种有毒化合物,其与DILI分类相关(Gustaffson等人2013)
-此外,我们可通过考虑以下方案来确定线粒体毒性风险范围:
IC50_MTSRR≥IC50_CV/CYT表示候选化合物没有特定的线粒体毒性,
IC50_MTSRR<IC50_CV/CYT且IC50-MTSRR≥0.75x IC50_CV/CYT表示候选化合物具有低线粒体毒性风险,

Claims (21)
1.用于评估候选化合物的代谢作用的方法和/或测定,所述方法和/或测定包括
a)将一种或多种三维细胞培养物或组织暴露于一种或多种候选化合物,和
b)在至少一种三维细胞培养物或组织中测量这种暴露对三维细胞培养物或组织特异性呼吸速率(MTSRR)的作用。
2.根据权利要求1所述的方法和/或测定,所述方法和/或测定还包括
c)在相同的或至少一种其他三维细胞培养物或组织中测量这种暴露在至少一种三维细胞培养物或组织的一个或多个细胞中对细胞活性和/或细胞毒性效应(CV-CYT)的作用,和
d)比较步骤b)和c)的作用以提供对一种或多种候选化合物的线粒体毒性的估计。
3.根据权利要求1-2中任一项所述的方法和/或测定,其中所述方法和/或测定用于确定候选化合物的线粒体毒性。
4.根据权利要求1-2中任一项所述的方法和/或测定,其中所述方法和/或测定用于确定候选化合物的化疗的有效性。

10.根据权利要求9所述的方法和/或测定,在所述方法和/或测定中的所述尺寸确定是指选自以下的至少一个参数:
·直径,
·周长,
·体积,
·光学横截面的区域。
11.根据权利要求8-10中任一项所述的方法和/或测定,其中所述三维细胞培养物或组织还包含至少一种选自以下的细胞类型:
·免疫细胞,
·间质细胞,
·成纤维细胞,
·癌症干细胞,
·已知对给定的抗癌剂具有抗性或敏感性的永生化细胞、恶性细胞、癌性细胞和/或赘生性细胞。
12.根据前述权利要求中任一项所述的方法和/或测定,其中所述微组织特异性呼吸速率通过同时测量一个或多个分离的3D微组织的氧气消耗速率(OCR)和细胞外酸化速率(ECAR)来确定。
13.根据权利要求12所述的方法和/或测定,其中在确定微组织特异性呼吸速率(MTSRR)时,使用备用呼吸容量(“SPARE”)。
14.根据权利要求12或13所述的方法和/或测定,其中在确定所述微组织特异性呼吸速率(MTSRR)时,还使用基础呼吸速率(“BASAL”)和/或最大呼吸速率(“MAX”)。
15.根据前述权利要求中任一项所述的方法和/或测定,其中测量细胞活力/细胞毒性效应确定一个或多个细胞的残余活力。
16.根据前述权利要求中任一项所述的方法和/或测定,其中对微组织特异性呼吸速率的作用和/或对细胞活力/细胞毒性效应的作用被确定为抑制浓度,优选为半数最大抑制浓度(IC50)。
