The invention adopts a sealing cover and a sealing plug which are respectively arranged at the top end and the rear end of a carotid artery implantation catheter, namely the sealing cover and the sealing plug can be penetrated by a needle, an injection opening covered by a unidirectional flow gate is arranged at the front end of the carotid artery implantation catheter, the flow speed and the time are controlled by a controller, a constant-speed injection micropump is driven to push an injector, and cancer cells or injection medicines flow into a carotid artery vessel through the injection opening of the carotid artery implantation catheter; the invention can simplify the operation method, simplify the operation steps and shorten the operation time; meanwhile, the operation mode of the animal model injected into the internal carotid artery of the mouse is completely changed, the external carotid artery of the mouse is not required to be sutured, the vagus nerve and the blood vessel are not damaged, and the success rate of the operation is greatly improved.

Technical Field
The invention relates to a mouse animal model technology, in particular to an implantable internal carotid artery constant-speed injection device and an operation method thereof.
Background
When preparing a mouse animal model for brain metastasis of various cancer cells, the proximal end of the common carotid artery and the external carotid artery of the mouse are usually required to be ligated, then the cancer cells cultured in vitro are injected into the internal carotid artery from the distal end of the common carotid artery by using an injector to reach the blood brain barrier, and spontaneously metastasize into the brain, so as to prepare the mouse animal model most similar to the brain metastasis of human cancer. From the local anatomy it is known that: the common carotid artery is divided forward into the external carotid artery and the internal carotid artery, with only the blood in the internal carotid artery flowing intracranially. Thus, only ligating the external carotid artery will allow blood flow in the common carotid artery to flow all the way forward into the internal carotid artery. Since the operation of the mouse needs to be carried out under a microscope, the operation must be very fine, the most critical step in the operation process is to ligate the external carotid artery of the mouse, then the cancer cells can be completely injected into the internal carotid artery to transfer to the brain by using a syringe, and the attendant vagus nerve and vein blood vessels must be protected from damage when the external carotid artery is sutured, otherwise, cardiac arrest and blood loss of the mouse are caused, which is an important cause of death of the mouse after the operation failure. In addition, after ligating the external carotid artery, the needle of the injector filled with cancer cells needs to be penetrated into the distal end of the common carotid artery (above the proximal ligature), and then the external cultured cancer cells are injected into the internal carotid artery, and the injection process needs to strictly control the injection speed, namely under the condition that the concentration of the cancer cells is fixed, the faster the injection speed is, the more the number of the cancer cells reaching the blood brain barrier in unit time is, the cancer plugs can be artificially caused to block blood vessels, which is equivalent to causing the death of mice due to the large-area thrombotic diseases, and is another important cause of death of mice with failed operations.
Disclosure of Invention
Aiming at the problems in the prior art, the invention provides an implantable carotid artery constant-speed injection device and an operation method thereof, so as to improve the success rate of manufacturing a mouse animal model and reduce the death rate of the mouse.

The implantable internal carotid artery constant-speed injection device of the invention comprises: the common carotid artery implantation catheter, the sealing cover, the sealing plug, the injection opening, the unidirectional flow gate, the marker, the needle, the injector, the propeller, the constant-speed injection micro pump and the controller; wherein, the common carotid artery implantation catheter is a cylindrical hollow catheter with hollow inside; the top end of the carotid artery implanted catheter is provided with a sealing cover, and the rear end is provided with a sealing plug; an injection opening is arranged on the side wall of the front end of the carotid artery implantation catheter; a unidirectional flow gate is arranged at the injection opening and positioned outside the carotid artery implantation catheter and covers the injection opening, the front end of the unidirectional flow gate is connected to the side wall of the front end of the carotid artery implantation catheter, and the unidirectional flow gate can be opened and closed outside the carotid artery implantation catheter around the connection position; a marker is arranged at the front end of the common carotid artery implantation catheter; the common carotid artery implantation catheter, the sealing cover, the sealing plug, the injection opening, the unidirectional flow gate and the marker form a common carotid artery implantation device; the front end of the injector is connected with a needle head, and the shape of the needle head is a folded line type with an obtuse angle; the needle penetrates the sealing plug and goes deep into the carotid artery implantation catheter; the piston handle at the bottom end of the injector is connected to the propeller; the propeller is connected to a constant-speed injection micro pump; the constant-speed injection micropump is connected to the controller; the empty cylinder of the injector is filled with the injection, the injection rate is set by the controller, the constant-speed injection micro pump is controlled to drive the propeller, the piston handle of the injector is pushed, the injection is injected into the carotid artery implantation catheter through the needle, and the injection flows out through the injection opening and the one-way flow gate; after the injection is finished, the unidirectional flow gate is closed because the pressure of the blood reflux is stronger than the pressure in the carotid artery implantation catheter, namely the carotid artery implantation catheter is negative pressure.
Before the operation, the carotid artery implantation catheter is inserted into the carotid artery blood vessel, and after the operation is finished, the carotid artery implantation catheter is taken out.
The common carotid artery implantation catheter is made of polyvinyl chloride PVC, polyethylene PE or polystyrene PS. The internal diameter of the common carotid artery implantation catheter is 0.5-0.75 mm; the outer diameter is 1-1.25 mm.
The sealing cover and the sealing plug are made of one of rubber, latex and silica gel.
The diameter of the injection opening is 1-2 mm, and the distance between the injection opening and the top end of the carotid artery implantation catheter is 3-4 mm. The invention adopts the sealing cover to seal the top end of the common carotid artery implantation catheter, and the side wall of the common carotid artery implantation catheter is provided with the injection opening, so that the injection is completely injected into the internal carotid artery, the external carotid artery of a mouse is not required to be sutured, and the vagus nerve and the blood vessel are not damaged.
The one-way flow gate is arranged outside the injection opening, in the injection process, the injection can flow out by flushing the one-way flow gate, and after the injection is finished, the one-way flow gate is closed to cover the injection opening due to negative pressure in the carotid implanted catheter, so that blood in the carotid artery is prevented from flowing back into the carotid implanted catheter.
The marker adopts an indication strip, and the surface of the indication strip is coated with a fluorescent agent; the indicator strip is tightly adhered to the inner surface of the sealing cover.
The obtuse angle of the needle head is 120-145 degrees, so that the puncture operation of the common carotid artery blood vessel is facilitated, the common carotid artery implantation catheter is more convenient to implant, and the common carotid artery blood vessel of a mouse cannot be damaged when the injection operation is performed.
The injector comprises a hollow cylinder, a piston and a piston handle; the top of the hollow cylinder is provided with a communicated needle head, the bottom of the piston is provided with a piston handle, and the front end of the piston penetrates into the hollow cylinder from the bottom of the hollow cylinder.
The propeller comprises a push head and a screw rod; the front end of the push head is connected with a piston handle of the injector, the rear end of the push head is connected with a screw rod, and the screw rod is connected to the constant-speed injection micro pump. The constant-speed injection micro pump is controlled by the controller, a control signal of the controller drives the constant-speed injection micro pump to drive the screw rod to rotate, the screw rod drives the front end push head to advance, and the push head can convert the rotary motion of the screw rod pair into linear motion, so that the piston of the injector at the front end of the push head is pushed to inject cancer cells or medicines.
The controller comprises a power supply, an input device, an output device and a control circuit board; wherein, the power supply is respectively connected to the input device, the output device and the control circuit board; the control circuit board is respectively connected to the input device and the output device; the infusion speed, the injection time and the total injection amount are set through an input device, a display screen is adopted as an output device, and the display screen is a Liquid Crystal Display (LCD). The advancing speed of the piston of the injector is controlled by setting the infusion speed and the injection time of the constant-speed injection micro pump so as to control the rotating speed of the screw.
The dosage of the injection is 500-200 mu l; the constant injection rate is 0.50-1.0 mu l/s, and the injection time is 100-200 s.
Further, the invention also comprises a protective sleeve which is sleeved outside the common carotid artery implantation device.
The injectate is various cancer cells transferred to brain or various injection medicines for treatment.
It is another object of the present invention to provide a method of operating an implantable internal carotid artery constant velocity injection device.
The operation method of the implantable internal carotid artery constant-speed injection device comprises the following steps:
1) Filling the syringe with the injectate;
2) Connecting a piston handle at the bottom end of the injector to the propeller;
3) Connecting the propeller to a constant speed injection micro pump;
4) Connecting a constant-speed injection micropump to a controller;

8) After the injection is finished, the unidirectional flow gate is closed because the internal of the carotid artery implanted catheter is negative pressure.
The invention has the advantages that:

The invention will be further elucidated by means of specific embodiments in conjunction with the accompanying drawings.

The syringe 6 comprises a hollow barrel 61, a piston 62 and a piston stem 63.
In the embodiment, the common carotid artery implantation catheter 1 adopts polyvinyl chloride PVC; rubber is adopted as the material of the sealing cover 2 and the sealing plug 3; the diameter of the injection opening was 1mm and the distance from the top of the carotid artery implanted catheter 1 was 3mm. The marker adopts an indication strip, and the surface of the indication strip is coated with fluorescent agent and is tightly adhered to the inner surface of the sealing cover 2; the obtuse angle of the needle 5 is 135 degrees; the dose of the implant was 100 μl; the constant injection rate was 0.50. 7) The injection speed is set through a controller, a constant-speed injection micro pump is controlled to drive a propeller, a piston handle of the injector 6 is pushed, an injection is injected into the common carotid artery implantation catheter 1 through a needle 5, and a unidirectional flow gate is flushed through an injection opening to flow out;
8) After the injection is finished, the common carotid artery implanted catheter 1 is under negative pressure, and the unidirectional flow gate is closed;
9) After the operation is finished, the common carotid artery implantation device is taken out.
