该方法组合使用头皮冷却和调节人角质形成细胞中ROS产生的分子细胞内信号传导通路的抑制剂。
典型地,使用冷却装置(例如Paxman Orbis冷却帽)来冷却区域或头皮的区域。
在一个实施方案中,ROS抑制剂为细胞渗透性抗氧化剂和/或ROS清除剂。典型地,ROS抑制剂为直接药理学抑制剂。
在一个实施方案中,ROS抑制剂为N-乙酰基-半胱氨酸(NAC)。
在一个实施方案中,ROS抑制剂为合成的。典型地,抑制剂为合成的NAC。
在一个实施方案中,ROS抑制剂包括一种或多种天然化合物。在一个实施方案中,ROS抑制剂包括维生素C。
在一个实施方案中,ROS抑制剂包括一种或多种类黄酮。典型地,ROS抑制剂包括一种或多种黄烷醇。在一个实施方案中,ROS抑制剂包括槲皮素。
在一个实施方案中,一种或多种ROS抑制剂为细胞渗透性的。典型地,ROS抑制剂为干扰ROS生物合成(产生细胞信号传导通路的ROS)的细胞渗透性生物学或药理学抑制剂。
在本发明的又一方面,提供一种治疗化学治疗诱发的脱发的药品,所述药品包括ROS抑制剂和适于局部施用的载体。
典型地,ROS抑制剂为NAC或其衍生物。
典型地,ROS递送方法包括以下任一种或任意的组合:
-通过由皮肤的直接吸收(例如经由乳霜)局部施用
-基于纳米颗粒或脂质体的靶向途径
-其它载体/生物学工具(例如病毒)
在本发明的又一方面,提供一种组合物,其用于和/或增强化学诱发的脱发(CIA)的冷却疗法和治疗,所述组合物含有活性氧类(ROS)抑制剂。
在本发明的又一方面,提供使用含有活性氧类(ROS)抑制剂的组合物来增强治疗化学诱发的脱发(CIA)的冷却疗法。
具体实施方式
实验和实施例。
在6孔板中将HaCaTa细胞培养于KSFM培养基中,并在37℃下用3μg/mL多柔比星处理2小时,然后用5μM的H2DCF-DA在处理后的规定的时间点标记(30分钟),用于测量细胞内ROS的水平。然后收集细胞并通过流式细胞术分析。在FL-1(或FL-2通道)中检测荧光强度作为细胞内ROS产生水平的指标并在图1中表示为相对于荧光测量的时间点(x轴)的中值荧光强度或MFI(y轴)。
结果表明,用化疗药物处理细胞后检测ROS的最佳时间点为30分钟。在该特定的最佳时间点,流式细胞术方法允许细胞内ROS精确且灵敏的检测。
参考图2,HaCaTa细胞在37℃下或在22℃、18℃和14℃下用3μg/mL多柔比星处理2小时来模拟不同的冷却条件。如上所述(图1),在通过流式细胞术分析前用5μM的H2DCF-DA标记细胞。代表性结果绘制成重叠直方图,且所代表的条件在图例中示出(图右侧,其中Con37℃表示为2,37℃为4,Con 22℃为6,22℃、18℃和14℃为8)。
结果表明,冷却可以防止人角质形成细胞中ROS的产生,并且所用温度越低,ROS产生的程度越小。有趣的是,尽管22℃显著减少ROS产生,但将温度降低至18℃和14℃几乎完全减弱ROS产生–这与这两个温度值几乎完全防止这些细胞中的细胞毒性的能力直接相关(我们发表的评论,在Al-Tameemi等人2014中被报道)。
参考图3,在96孔板中以5×103/孔的密度将HaCaTa细胞培养于KSFM中,并将细胞在37℃/5%CO2下温育过夜。将一些细胞用0.625mM NAC预处理1小时(NAC的最佳浓度通过预滴定实验来确定)。之后,用一系列浓度的多西紫杉醇、多柔比星(Sigma)或4-羟基环磷酰胺(4-OH-CP)(环磷酰胺的活性代谢物(Niomech))在37℃下(对照条件)或在冷却条件下(22℃)、在有和没有NAC的适当培养基(pH7.4)中处理细胞2小时。在所有实验中包括溶剂(DMSO)对照(代表对应于最高药物浓度的DMSO的最大量)。处理后,除去药物,用PBS洗涤细胞两次并加入新鲜培养基。接着将培养物在37℃下温育72小时,然后使用CellTiter96AQueous One细胞增殖检测(Promega)评估细胞生长。简言之,向每个孔中加入20μl的CellTiter试剂并将板在5%CO2的条件中在37℃下孵育4小时,然后采用分光光度法地在492nm处获得吸光度读数。结果表示为相对于对照(未处理细胞)的%。“dox”、“dce”和“CP”分别表示多柔比星、多西紫杉醇和4-OH-CP。37℃下的药物处理表示为10,22℃下的药物为12,37℃下的药物+NAC为14,22℃下的药物+NAC为16。
结果表明,尽管单独用NAC冷却或处理可以提供一些细胞保护,但冷却加上抗氧化剂/ROS清除剂NAC的组合提供比单独的每种药剂更好的保护。对于较高的药物浓度,结果尤为显著(由框表示)。

通过扩展4-OH-CP的药物浓度范围,该图中的结果增强了我们在图3中呈现的发现。结果表明,即使在药物剂量导致完全细胞毒性(完全死亡)的该特定药物的极高剂量下(红色框),冷却加ROS抑制剂的组合对药物诱发的毒性提供极高水平的保护。

参考图7,在96孔板中以5×103/孔的密度将HaCaTa细胞培养于KSFM中,并将细胞在37℃/5%CO2下温育过夜。将一些细胞用0.625mM NAC预处理1小时(NAC的最佳浓度通过预滴定实验来确定)。之后,用组合AC在37℃下(对照条件)或在冷却条件下(26℃或22℃)、在有和没有NAC的适当培养基(pH7.4)中处理细胞。特别地,使用3μg/ml(1小时)的多柔比星和5μg/ml(1小时)的4-OH-CP进行多柔比星(A-表示阿霉素)和4-OH-CP(C,表示环磷酰胺)的依次处理。接着洗涤细胞并替换培养基,然后,使用CellTiter 96AQueous One细胞增殖检测(Promega)评估72小时后的细胞生长。简言之,向每个孔中加入20μl的CellTiter试剂并将板在5%CO2的条件中在37℃下孵育4小时,然后采用分光光度法地在492nm处获得吸光度读数。结果表示为相对于对照(未处理细胞)的%。37℃下的处理表示为22,26℃下的为24,22℃下的表示为26。
结果表明,尽管在26℃或22℃下的冷却显示出显著的细胞保护效果,但冷却与ROS抑制剂/清除剂NAC的组合导致更好的细胞保护。这些发现表明冷却加ROS抑制比单独冷却更好地保护人角质形成细胞免受不仅是由单一的而且是由组合的药物治疗所引起的细胞毒性。
使用抗氧化剂槲皮素的结果示于图8。HaCaTa细胞(5×103细胞/孔)以每孔100μlKSFM中培养于96孔板中,并在37℃/5%CO2下温育过夜。24小时后将细胞用3μM槲皮素或0.625mM的NAC预处理。还包括不含有任何抗氧化剂的对照。预处理1小时后,用多柔比星在0.25μg/ml、0.5μg/ml和1μg/ml下处理细胞。然后在37℃下(对照条件)或在冷却条件下(26℃)温育细胞。接着在37℃下温育培养物72小时,然后使用CellTiter 96AQueous One细胞增殖检测(Promega)评估细胞生长。结果表示为相对于含有抗氧化剂但无多柔比星的对照的%。
结果表明,尽管冷却或仅用槲皮素处理可以提供一些细胞保护,但是冷却加上抗氧化剂/ROS清除剂槲皮素的组合提供比单独的每种药剂更好的保护(如红色框所示)。
37℃下仅DOX表示为30,26℃下仅DOX表示为32,37℃下的NAC+DOX表示为34,26℃下的NAC+DOX表示为36,37℃下的槲皮素+DOX表示为38,26℃下的槲皮素+DOX表示为40。
+NAC为14,26℃下的药物为18,26℃下的药物+NAC为20。
Claims (32)
1.一种包含活性氧类(ROS)抑制剂的组合物,其用于治疗化学诱发的脱发(CIA)的冷却疗法。
2.根据权利要求1所述的组合物,其中所述组合物包括一种或多种ROS抑制剂和所述冷却疗法包括头皮冷却。
3.根据权利要求1所述的组合物,其中在控温环境中递送所述组合物。
4.根据权利要求1所述的组合物,其中局部给药所述组合物。
5.根据权利要求4所述的组合物,其中含有ROS抑制剂的所述组合物被施用于头皮。
6.根据权利要求1所述的组合物,其中所述ROS抑制剂为细胞渗透性抗氧化剂和/或ROS清除剂。
7.根据权利要求1所述的组合物,其中所述ROS抑制剂为直接药理学抑制剂。
8.根据权利要求1所述的组合物,其中所述ROS抑制剂为N-乙酰基-半胱氨酸(NAC)。
9.根据权利要求1所述的组合物,其中所述ROS抑制剂为合成的。
10.根据权利要求8所述的组合物,其中所述抑制剂为合成的NAC。
11.根据权利要求1所述的组合物,其中所述ROS抑制剂包括一种或多种天然化合物。
12.根据权利要求11所述的组合物,其中所述ROS抑制剂包括维生素C。

17.根据权利要求1所述的组合物,其中所述ROS抑制剂为干扰ROS生物合成(产生细胞信号传导通路的ROS)的细胞渗透性生物学或药理学抑制剂。
18.一种用于治疗化学治疗法诱发的脱发的药品,所述药品含有ROS抑制剂和适于局部施用的载体。
19.一种通过冷却并通过使用含有ROS抑制剂的组合物来保护毛发或毛囊的方法。
20.根据权利要求19所述的方法,其中将至少所述头皮的局部区域冷却至低于室温和/或至低于体温。
21.根据权利要求19所述的方法,其中在控温环境中递送ROS抑制性化合物或组合物。
22.根据权利要求19所述的方法,其中使用冷却装置在身体部位上局部实施所述冷却。
23.根据权利要求22所述的方法,其中所述身体部位为头皮。
