Abstract

本发明公开了一种双层双向控释抗氧化抗菌膜及其制备方法与应用。该膜由亲水抗氧化内层、疏水抗菌外层组成,其中亲水抗氧化内层由含有水溶性天然抗氧化剂的淀粉层组成,疏水抗菌外层由含有脂溶性抗菌精油的醇溶蛋白层组成。将这两种原料液经增塑、均质、脱气、流延法依次铺膜处理即得该膜。本发明首次采用亲水性淀粉结合水溶性天然抗氧化剂与疏水性醇溶蛋白结合脂溶性抗菌精油复合作为双层可食性膜的基材制备得到。该薄膜既能阻挡外来微生物的入侵,也能防止内部包装物的氧化,还能兼具优良的阻湿性能和机械性能,且克服了精油的易挥发性和水溶性天然抗氧化剂的有限应用性,有效弥补各单一组分的缺点,可作为饼干、鲜切果蔬、肉类等保鲜包装材料。

技术领域
本发明属于可食性包装材料技术领域,更具体地,涉及一种双层双向控释抗氧化抗菌膜及其制备方法与应用。
背景技术
可食性膜是指以天然可食性生物大分子为主要基质,辅以可食性增塑剂,通过一定的处理工序使各成膜剂分子之间相互作用、在干燥后形成具有一定力学性能和选择透过性的多孔网络结构的致密薄膜,可作为食品色、香、味、营养强化剂、抗氧化剂和抗菌剂等的载体,且对食品和环境无害。
新鲜食品保鲜期较短,尤其对于鲜切加工的果蔬、肉类、鱼类、奶制品等来说,在受到机械加工损伤后,会诱发一系列相关酶活性的变化,导致产品软化、褐变以及因微生物侵染而引起腐烂等不良症状,严重影响其商品价值和货架期以及消费者的购买欲望。因此,研究开发新的抗氧化抗菌包装材料是很有必要的。
近年来,随着活性包装概念的提出,对于食品科技工作者来说最主要的挑战就是设计活性食品包装材料。活性食品包装即在食品包装材料中添加一定的抗菌或抗氧化物质,通过一定的手段使抗氧化或抗菌物质达到缓慢释放的目的,相比于直接应用抗菌或抗氧化物质于食品表面有更理想的效果。同时,出于人们健康的考虑和环境安全问题,当前食品活性包装中添加剂的研究越来越多的集中于天然的抗氧化或抗菌物质。然而,现有的单层可食性膜通常很难将水溶性天然抗氧化剂和脂溶性抗菌剂结合起来发挥作用,在抗氧化和抗菌双功能上未能达到很好的效果,影响了食品的保藏质量,在应用上有很大局限性。因此,如何将不同性质的基质有效复配,充分发挥各组分优势的同时做到既抗氧化又抗菌,是当前解决实际问题的关键,这对促进我国食品储藏和保鲜技术的发展、提供新型包装材料等方面具有重要意义。
发明内容
本发明要解决的技术问题是克服上述现有可食性活性包装膜的缺陷和不足,制备一种能定向释放抗氧化抗菌物质的双层活性包装膜,采用疏水性醇溶蛋白结合脂溶性抗菌精油与亲水性淀粉结合水溶性天然抗氧化剂复合作为双层可食性膜的基材,并采用复配工艺制备得到一种具有抗氧化抗菌双功能的双层可食性膜。该双层可食性膜对外可阻挡外界微生物入侵,对内可防止包装材料氧化,兼具优良的阻湿性能和良好的机械性能,透明度好,且其原料来源广泛、加工工艺简单易行,水溶性天然抗氧化剂的抗氧化能力强,无毒无味;脂溶性抗菌精油天然无毒,且具有良好的抑菌活性,制备的抗菌膜性能良好且具有抑制食品表面微生物生长的作用,可广泛用于饼干、果蔬等食品包装领域。
本发明的目的是提供一种具有双层双向控释抗氧化抗菌膜及其应用。
本发明另一个目的是提供所述双层双向控释抗氧化抗菌膜的制备方法。
本发明的目的通过下述技术方案来实现。
一种双层双向控释抗氧化抗菌膜,由亲水抗氧化内层、疏水抗菌外层组成,其中所述亲水抗氧化内层由含有水溶性天然抗氧化剂的淀粉层组成,所述疏水抗菌外层由含有脂溶性抗菌精油的醇溶蛋白层组成。
优选的,所述亲水抗氧化内层中淀粉的质量百分比为5~30%,水溶性天然抗氧化剂的添加量为淀粉质量的0.5%~15%。
优选的,所述疏水抗菌外层中醇溶蛋白的质量百分比为5~30%,脂溶性抗菌精油与醇溶蛋白的添加比例为(0.005-0.15)mL:1g。
进一步优选的,所述亲水抗氧化内层中淀粉的质量百分比为6~8%(最优选6%),水溶性天然抗氧化剂的添加量为淀粉质量的2.5%~7.5%;所述疏水抗菌外层中醇溶蛋白的质量百分比为15~18%(最优选15%),脂溶性抗菌精油与醇溶蛋白的添加比例为(0.025-0.075)mL:1g。
优选的,所述亲水抗氧化内层、疏水抗菌外层的厚度均为45~65μm。
优选的,所述淀粉为原淀粉和变性淀粉中的一种或几种,所述原淀粉为玉米淀粉、木薯淀粉、马铃薯淀粉、大米淀粉和小麦淀粉中的一种或几种混合物,所述变性淀粉为羟丙基玉米变性淀粉、羧甲基淀粉、酯化淀粉、氧化淀粉、酸变性淀粉、醚化淀粉、交联淀粉、阳离子淀粉、接枝淀粉和双醛淀粉中的一种或几种混合物;所述水溶性天然抗氧化剂为迷迭香酸、鼠尾草酸、茶多酚、葡萄多酚、百里香酚、维C和维E中的一种或两种以上混合物;所述醇溶蛋白为玉米醇溶蛋白、小麦醇溶蛋白、大麦醇溶蛋白和高粱醇溶蛋白的一种或两种以上混合物,所述脂溶性抗菌精油为肉桂精油、丁香精油、牛至精油、薄荷精油、百里香精油、茶树精油、迷迭香精油、香茅精油、蔷薇精油、甜橙精油、桉叶精油和玫瑰精油中的一种或两种以上混合物。
更优选地,所述淀粉为玉米变性淀粉(优选为羟丙基玉米变性淀粉),所述水溶性天然抗氧化剂为茶多酚,所述醇溶蛋白为玉米醇溶蛋白,所述脂溶性抗菌精油为牛至精油。
更优选地,所述玉米醇溶蛋白为α-玉米醇溶蛋白、β-玉米醇溶蛋白和γ-玉米醇溶蛋白中的一种或两种以上混合物。
以上所述的一种双层双向控释抗氧化抗菌膜应用于制备包装袋或包装薄膜。
以上所述的一种双层双向控释抗氧化抗菌膜的制备方法,包括以下步骤:
S1.亲水抗氧化内层储备液的制备:将淀粉溶解到水中,并在60~80℃下加入水溶性天然抗氧化剂,得到亲水抗氧化内层储备液,备用;
S2.疏水抗菌外层储备液的制备:将醇溶蛋白溶解到有机溶剂中,并加入脂溶性抗菌精油,均质得到疏水抗菌外层储备液,备用;
S3.流延法铺膜:将亲水抗氧化内层储备液、疏水抗菌外层储备液脱气后,进行流延成膜,依次将亲水抗氧化内层储备液、疏水抗菌外层储备液流延成膜,第二层膜需待前一层膜部分脱水后再继续成膜;干燥,揭膜,即得双层双向控释抗氧化抗菌膜。
优选的,所述的有机溶剂为乙酸、乙醇、丙三醇、丁二醇或它们的混合物,进一步优选为乙酸。
优选的,所述亲水抗氧化内层储备液、疏水抗菌外层储备液中还有增塑剂。
优选的,所述增塑剂的添加量为淀粉或醇溶蛋白质量的10%~40%。
优选的,所述增塑剂为甘油、油酸、柠檬酸、聚乙二醇、乙二醇和丙二醇其中的一种或几种。
更优选地,所述增塑剂为甘油。
优选地,上述步骤S1中,加入增塑剂后,水浴磁力搅拌1.5~2h,再在60~80℃下加入茶多酚继续搅拌15-30min,使储备液中的各种成分充分混合。
优选地,上述步骤S2中,加入增塑剂后磁力搅拌1.5~2h,再加入牛至精油,并利用均质机在8000~10000rpm下均质4~5min,使储备液中的各种成分充分混合。
优选地,步骤S3中,所述脱气的操作为采用超声波脱气处理10~20min,以除去两种储备液中的气体。
优选地,步骤S3中,所述干燥的条件为:干燥温度为室温,干燥环境为通风橱,干燥时间为20~24h。
优选地,将步骤S4制得的双层双向控释抗氧化抗菌膜置于温度为25~30℃、湿度为52%~53%的密闭容器中储存。
本发明以亲水淀粉抗氧化膜为内层、以疏水醇溶蛋白抗菌膜为外层来制备双层复合膜。所用淀粉(如玉米淀粉、木薯淀粉、马铃薯淀粉、大米淀粉、小麦淀粉等原淀粉,或羟丙基玉米变性淀粉、羧甲基淀粉、酯化淀粉、氧化淀粉、酸变性淀粉、醚化淀粉、交联淀粉、阳离子淀粉、接枝淀粉、双醛淀粉等变性淀粉)基与其他物质有较好的相容性,有结合天然抗氧化物质的潜力,但其可食性膜的阻湿性差;而醇溶蛋白(如玉米醇溶蛋白、小麦醇溶蛋白、高粱醇溶蛋白)基具有亲油性,且可食性膜的阻湿性能优异,但机械强度低;在本发明的方案中,亲水淀粉抗氧化膜作为内层在包装内侧起抗氧化及保湿作用,疏水醇溶蛋白抗菌膜作为外层发挥抗菌及阻水优势,以此制备成的双层可食性膜对外可阻挡外界微生物入侵,对内可防止包装材料氧化,兼具优良的阻湿性能和良好的机械性能,对促进我国食品储藏和保鲜技术的发展、提供新型包装材料等方面具有重要意义。
与现有技术相比,本发明具有以下有益效果:
1、本发明的双层双向控释抗氧化抗菌膜首次采用亲水性淀粉结合水溶性天然抗氧化剂与疏水性醇溶蛋白结合脂溶性抗菌精油复合作为双层可食性膜的基材以合适的复配工艺制备得到,具有定向抗氧化抗菌性,兼具优良的阻湿性能和良好的机械性能。
2、本发明的双层双向控释抗氧化抗菌膜可广泛应用于饼干、鲜切果蔬、肉类等食品包装中,由于该复合膜具有定向释放抗氧化抗菌物质及阻湿的,即能在包装内侧起抗氧化及吸湿作用,又能在外侧发挥抗菌及阻水优势,在包装材料等领域具有潜在的应用前景,尤其是对促进我国食品储藏和保鲜技术的发展、提供新型包装材料等方面具有重要意义。
3、本发明的双层双向控释抗氧化抗菌膜采用造价低廉、操作简单、能耗低、制备效率高的流延成膜技术制备,且原料来源广泛,加工工艺简单易行、成本低。
4、本发明的双层双向控释抗氧化抗菌膜为可食性包装材料的研究和应用提供了一种新颖的实用方法。

具体实施方式
以下结合说明书附图和具体实施例来进一步说明本发明。以下实施例为本发明较佳的实施方式,但并不对本发明的保护范围做任何形式的限定。除非特别说明,本发明采用的试剂、方法和设备为本技术领域常规试剂、方法和设备。
除非特别说明,以下实施例所用试剂和材料均为市购。
实施例1
一种双层双向控释抗氧化抗菌膜,由以下步骤制备而成:
(1)亲水抗氧化内层储备液的制备:60℃条件下,将3g淀粉溶解于50mL水中,加入0.3g甘油,在水浴磁力搅拌条件下充分搅拌30min,升温至100℃后继续搅拌1h,降温至70℃,加入0.225g茶多酚,继续搅拌30min,静置10min;
(2)疏水抗菌外层储备液的制备:将3g玉米醇溶蛋白溶解于17mL的100%乙酸溶液中,加入0.8g甘油,在磁力搅拌条件下充分搅拌2h,加入75μL的牛至精油,利用均质机在10000rpm下均质5min;
(3)脱气:将混合均匀的储备液放入超声脱气机中进行脱气10min,除去膜液中的气体;
(4)流延铺膜:利用流延法铺膜于规格为90mm×100mm的培养皿中,具体为:将6mL脱气完的亲水抗氧化内层储备液浇铸在培养皿上,流延成膜,在室温下通风橱内放置6h使该膜部分脱水;再将3mL疏水抗菌外层储备液铺在疏水外层上,流延成膜;
(6)干燥、揭膜:将流延成膜后的培养皿放入通风橱中干燥24h后,揭膜,得到可双层双向控释抗氧化抗菌膜;
(7)保存:将上述膜片放在温度为25~30℃和湿度为52%~53%的密闭容器中储存。
实施例2
一种双层双向控释抗氧化抗菌膜,由以下步骤制备而成:
(1)亲水抗氧化内层储备液的制备:60℃条件下,将3g淀粉溶解于50mL水溶液中,加入0.4g甘油,在水浴磁力搅拌条件下充分搅拌30min,升温至100℃后继续搅拌1h,降温至70℃,加入0.15g茶多酚,继续搅拌30min,静置10min;
(2)疏水抗菌外层储备液的制备:将3g玉米醇溶蛋白溶解于17mL的100%乙酸溶液中,加入0.7g甘油,在磁力搅拌条件下充分搅拌2h,加入150μL的牛至精油,利用均质机在10000rpm下均质5min;
(3)脱气:将混合均匀的储备液放入超声脱气机中进行脱气10min,除去膜液中的气体;
(4)流延铺膜:利用流延法铺膜于规格为90mm×100mm的培养皿中,具体为:将6mL脱气完的亲水抗氧化内层储备液浇铸在培养皿上,流延成膜,在室温下通风橱内放置6h使该膜部分脱水;再将3mL疏水抗菌外层储备液铺在疏水外层上,流延成膜;
(6)干燥、揭膜:将流延成膜后的培养皿放入通风橱中干燥24h后,揭膜,得到可双层双向控释抗氧化抗菌膜;
(7)保存:将上述膜片放在温度为25~30℃和湿度为52%~53%的密闭容器中储存。
实施例3
一种双层双向控释抗氧化抗菌膜,由以下步骤制备而成:
(1)亲水抗氧化内层储备液的制备:60℃条件下,将3g淀粉溶解于50mL水溶液中,加入0.5g甘油,在水浴磁力搅拌条件下充分搅拌30min,升温至100℃后继续搅拌1h,降温至70℃,加入0.075g茶多酚,继续搅拌20min,静置30min;
(2)疏水抗菌外层储备液的制备:将3g玉米醇溶蛋白溶解于17mL的100%乙酸溶液中,加入0.6g甘油,在磁力搅拌条件下充分搅拌2h,加入225μL的牛至精油,利用均质机在10000rpm下均质5min;
(3)脱气:将混合均匀的储备液放入超声脱气机中进行脱气10min,除去膜液中的气体;
(4)流延铺膜:利用流延法铺膜于规格为90mm×100mm的培养皿中,具体为:将6mL脱气完的亲水抗氧化内层储备液浇铸在培养皿上,流延成膜,在室温下通风橱内放置6h使该膜部分脱水;再将3mL疏水抗菌外层储备液铺在疏水外层上,流延成膜;
(6)干燥、揭膜:将流延成膜后的培养皿放入通风橱中干燥24h后,揭膜,得到可双层双向控释抗氧化抗菌膜;
(7)保存:将上述膜片放在温度为25~30℃和湿度为52%~53%的密闭容器中储存。
实施例4
将实施例1、2、3制得的产品放置15天后进行性能测定,
抗氧化性能测试:使用DPPH(2,2-二苯基-1-苦基肼)测定法测量从膜基质释放的抗氧化活性。取0.1g样品放入20mL水中,超声10min,将100μL膜提取物溶液的等分试样与3.9mL DPPH/甲醇溶液(40mg/L)混合。在黑暗中反应30分钟后,在517nm处测量吸光度。以去离子水为空白对照。
其抗氧化效果如下:

由表1可知,实施例1-3所得产品具有抗氧化性。
抗菌性能测试:采用体外抑菌实验方法—纸片法。在制备好的无菌固体培养基上均匀涂布0.1mL菌悬液来制成带菌平板,用灭菌好的镊子夹取由打孔器制好的膜片放到含菌平板上,每组实施例3个平行。经恒温培养箱37℃培养12h后取出,量取抑菌圈直径。
其抗菌效果如下:

由表2可知,实施例1-3所得产品具有抗菌性。
水蒸气透过率(WVP)测试:采用拟杯子法进行水蒸气透过率的测定,分别测定膜内、外侧两侧的阻湿性。在瓶口直径2.5cm的称量瓶中加入约11g的完全干燥硅胶,干燥剂用量距离瓶口6mm,分别以膜的内侧、外侧朝外盖住瓶口并封紧使膜与瓶口之间不留下任何缝隙。然后将称量瓶置于底部为去离子水,温度为25℃的玻璃干燥器中,每隔12小时测定称量瓶的重量。
其阻湿性能如下:

由表3可知,实施例1-3所得产品具有优良的阻湿性能。
机械性能测试:机械性能试验是用配备有0.1×N负载单元的拉伸试验机(Instron5566,USA)进行的。在测量前,在25℃下,在溴化钠饱和溶液中以57%RH储存膜2天。然后将薄膜切割成哑铃形(35mm×2mm)。初始距离为25mm,样品在十字头速度5mm/min拉伸。根据应力-应变实验曲线测定了薄膜的杨氏模量(YM)、拉伸强度(TS)和断裂伸长率(EAB)。每个膜均进行六个重复处理。

由表4可知,实施例1-3所得产品具有良好的机械性能。

Claims (9)

1.一种双层双向控释抗氧化抗菌膜,其特征在于,由亲水抗氧化内层、疏水抗菌外层组成,其中所述亲水抗氧化内层由含有水溶性天然抗氧化剂的淀粉层组成,所述疏水抗菌外层由含有脂溶性抗菌精油的醇溶蛋白层组成;所述亲水抗氧化内层中淀粉的质量百分比为5~30%,水溶性天然抗氧化剂的添加量为淀粉质量的0.5%~15%;所述疏水抗菌外层中醇溶蛋白的质量百分比为5~30%,脂溶性抗菌精油与醇溶蛋白的添加比例为(0.005-0.15)mL:1g。
2.根据权利要求1所述的一种双层双向控释抗氧化抗菌膜,其特征在于,所述亲水抗氧化内层中淀粉的质量百分比为6-8%,水溶性天然抗氧化剂的添加量为淀粉质量的2.5%~7.5%;所述疏水抗菌外层中醇溶蛋白的质量百分比为15-18%,脂溶性抗菌精油与醇溶蛋白的添加比例为(0.025-0.075)mL:1 g。
3.根据权利要求1所述的一种双层双向控释抗氧化抗菌膜,其特征在于,所述亲水抗氧化内层、疏水抗菌外层的厚度均为45~65 μm。
4.根据权利要求1所述的一种双层双向控释抗氧化抗菌膜,其特征在于,所述淀粉为原淀粉和变性淀粉中的一种或几种,所述原淀粉为玉米淀粉、木薯淀粉、马铃薯淀粉、大米淀粉和小麦淀粉中的一种或几种混合物,所述变性淀粉为羟丙基玉米变性淀粉、羧甲基淀粉、酯化淀粉、氧化淀粉、酸变性淀粉、醚化淀粉、交联淀粉、阳离子淀粉、接枝淀粉和双醛淀粉中的一种或几种混合物;所述水溶性天然抗氧化剂为迷迭香酸、鼠尾草酸、茶多酚、葡萄多酚、百里香酚、维C和维E中的一种或两种以上混合物;所述醇溶蛋白为玉米醇溶蛋白、小麦醇溶蛋白、大麦醇溶蛋白和高粱醇溶蛋白的一种或两种以上混合物,所述脂溶性抗菌精油为肉桂精油、丁香精油、牛至精油、薄荷精油、百里香精油、茶树精油、迷迭香精油、香茅精油、蔷薇精油、甜橙精油、桉叶精油和玫瑰精油中的一种或两种以上混合物。
5.制备权利要求1~4任一项所述的一种双层双向控释抗氧化抗菌膜的方法,其特征在于,包括以下步骤:
S1. 亲水抗氧化内层储备液的制备:将淀粉溶解到水中,并在60~80℃下加入水溶性天然抗氧化剂,得到亲水抗氧化内层储备液,备用;
S2. 疏水抗菌外层储备液的制备:将醇溶蛋白溶解到有机溶剂中,并加入脂溶性抗菌精油,均质得到疏水抗菌外层储备液,备用;
S3. 流延法铺膜:将亲水抗氧化内层储备液、疏水抗菌外层储备液脱气后,进行流延成膜,依次将亲水抗氧化内层储备液、疏水抗菌外层储备液流延成膜,第二层膜需待前一层膜部分脱水后再继续成膜;干燥,揭膜,即得双层双向控释抗氧化抗菌膜。
6.根据权利要求5所述的制备方法,其特征在于,所述的有机溶剂为乙酸、乙醇、丙三醇和丁二醇中的一种或多种。
7.根据权利要求5所述的制备方法,其特征在于,所述亲水抗氧化内层储备液、疏水抗菌外层储备液中还有增塑剂。
8.根据权利要求7所述的制备方法,其特征在于,所述增塑剂为甘油、油酸、柠檬酸、聚乙二醇、乙二醇和丙二醇其中的一种或几种;所述增塑剂的添加量为淀粉或醇溶蛋白质量的10%~40%。
9.权利要求1~4任一项所述的一种双层双向控释抗氧化抗菌膜应用于制备包装袋或包装薄膜。
