Abstract
本发明提供了一种胰腺癌动物模型的构建方法,包括:(1)构建KC鼠并对KC鼠进行扩繁得到KC鼠的子代;(2)准备P53鼠并对P53鼠进行扩繁得到P53鼠的子代;(3)采用所述KC鼠的子代与所述P53鼠的子代进行繁殖得到KPC胰腺癌鼠模型。本发明的构建方法具有时间短、成本低的优点。
Description
技术领域
本发明属于医学生物学技术领域,具体地,本发明涉及一种动物模型的构建方法,更具体地,本发明涉及一种胰腺癌动物模型的快速构建方法。
背景技术
胰腺癌具有临床表现缺乏特异性、早期诊断困难、手术切除率低等特点,所以通常难以获得不同发病阶段的胰腺癌组织标本,也难以在临床上观察胰腺癌的发生、发展过程。因此,通过胰腺癌动物模型,可以在动物体内模拟人胰腺癌的发生、发展过程,为深入研究胰腺癌的发生、发展机制以及指导治疗奠定基础。
胰腺癌动物模型例如胰腺癌鼠模型包括Kras KO、P53KO、PC、KC和KPC鼠,这些模型均易发生肿瘤,但是周期均较长,有的甚至长达数月。在这些模型中,KPC鼠模型由于包含3种基因缺陷,因此与其它胰腺癌模型(细胞系、异种移植瘤等)相比,存在扩繁费用较高、周期更长等缺陷。
但是,KPC鼠模型保留了胰腺癌天然的肿瘤微环境和免疫系统,为胰腺癌的肿瘤免疫机制研究和免疫治疗以及化疗增敏治疗等提供了有利便捷的工具。
因此,目前急需一种能够快速且低成本的获得KPC胰腺癌动物模型的方法。
发明内容
本发明的目的是针对现有技术的缺陷,提供了一种胰腺癌动物模型的构建方法。
本发明的胰腺癌动物模型的构建方法包括:(1)构建KC鼠并对KC鼠进行扩繁得到KC鼠的子代;(2)准备P53鼠并对P53鼠进行扩繁得到P53鼠的子代;(3)采用所述KC鼠的子代与所述P53鼠的子代进行繁殖得到KPC胰腺癌鼠模型。
进一步,步骤(1)中,所述构建KC鼠采用如下方法进行:将LSL-KrasG12D鼠与Pdx1-Cre鼠按照雌:雄=1∶1至3∶1进行合笼交配,获得的LSL-KrasG12D阳性且Pdx1-Cre阳性的子代即为所述KC鼠。
进一步,步骤(1)中,所述对KC鼠进行扩繁得到KC鼠的子代采用如下方法进行:采用所述KC鼠与野生型鼠自然交配进行扩繁得到KC鼠的子代;或者,采用所述KC鼠的精子与野生型鼠的卵子通过快速扩繁进行扩繁得到KC鼠的子代。
进一步,所述KC鼠是杂合KC鼠。
进一步,步骤(2)中,所述对P53鼠进行扩繁得到P53鼠的子代采用如下方法进行:采用所述P53鼠与野生型鼠自然交配进行扩繁得到P53鼠的子代;或者,采用所述P53鼠的精子与野生型鼠的卵子通过快速扩繁进行扩繁得到P53鼠的子代。
进一步,所述P53鼠是纯合P53鼠或杂合P53鼠。

本发明的技术方案至少具有如下有益效果:
(1)本发明的构建方法基于IVF快速扩繁技术,能够在短时间内获得KPC胰腺癌鼠模型,具有时间短且成本低的优点。
(2)本发明的构建方法主要以P53纯合雄鼠来进行扩繁,使KPC胰腺癌鼠模型的获得比例提高了一倍。

本文所述的“体外受精”(in-vitro fertilization,IVF)是指分别将卵子与精子从哺乳动物内取出并在体外受精,发育成胚胎后,再移植回母体子宫内,以达到受孕目的的一种技术。
本文所述的“快速扩繁”或“快速扩繁技术”是指对适龄雌鼠采用超数排卵的方式获得大量卵母细胞,通过新鲜或冷冻精子进行体外受精的方式获得需要的受精卵,受精卵发育到合适阶段时将其移植到代孕母鼠体内,短时间(一代)获得大量子代的技术。
本文所述的“鼠”可以是小鼠、大鼠等,在本发明的具体实施例中主要以小鼠为例对本发明的技术方案进行说明。
本文所述的“KPC胰腺癌鼠模型”是指LSL-KrasG12D/+;LSL-Trp53R172H/+;Pdx-1-Cre鼠,其是经典的胰腺导管细胞癌动物模型。
本文所述的“KC鼠”是指LSL-KrasG12D/+;Pdx-1-Cre(又称Lpf1-cre)鼠。
本文所述的“P53鼠”是指LSL-Trp53R172H/+鼠。
KPC胰腺癌动物模型的构建方法
针对目前KPC胰腺癌动物模型构建周期长、费用高等问题,本发明的发明人创造性的提出了一种KPC胰腺癌动物模型的快速且高效的构建方法,该构建方法具体包括如下步骤:
S101:构建KC鼠并对KC鼠进行扩繁得到KC鼠的子代;
S102:准备P53鼠并对P53鼠进行扩繁得到P53鼠的子代
S103:采用KC鼠的子代与P53鼠的子代进行繁殖得到KPC胰腺癌鼠模型。
上述步骤S101和步骤S102顺序可互相,无先后顺序的设定。
进一步,步骤S101包括:
S1011:将LSL-KrasG12D鼠与Pdx1-Cre鼠按照雌:雄=1∶1至3∶1进行合笼交配,获得的LSL-KrasG12D阳性且Pdx1-Cre阳性的子代即为所述KC鼠。其中,LSL-KrasG12D鼠与Pdx1-Cre鼠交配得到的子代通过PCR技术来鉴定其中的LSL-KrasG12D阳性且Pdx1-Cre阳性的子代。在本发明中,LSL-KrasG12D鼠与Pdx1-Cre鼠可通过购买获得。
S1012:采用KC鼠与野生型鼠自然交配进行扩繁得到KC鼠的子代;或者,采用KC鼠的精子与野生型鼠的卵子通过快速扩繁进行扩繁得到KC鼠的子代。得到KC鼠的子代采用PCR技术进行鉴定。
优选地,步骤S1012采用KC鼠的精子与野生型鼠的卵子通过快速扩繁进行扩繁得到KC鼠的子代。
优选地,步骤S1012采用的KC鼠是杂合体。发明人通过研究发现,Kras纯合突变鼠致死,杂合KC鼠成年后会发生胰腺癌(pancreatic ductal adenocarcinoma,PDA),6月龄时死亡率大概20%,因此,步骤S1012中的扩繁采用杂合Kras Cre(KC)阳性鼠与野生型鼠交配扩繁或IVF快速扩繁。

S1021:准备P53鼠。在本发明中P53鼠可通过购买获得。

KPC胰腺癌鼠模型涉及3个基因,其中,①Kras纯合致死;②P53小鼠需要杂合保种;③Pdx-1-Cre鉴定仅区分阳性和阴性。将3种基因扩繁到一起获得KPC鼠,可以有较多的途径。但是,采用本发明的构建方法,通过IVF快速扩繁途径,起始仅需要少量雄鼠或具有受精能力的精子,根据实验需要7至8个月即可以获得大量的KPC鼠,同时提高了KPC鼠获得的比例(从12.5%提高到25%)。
下面通过具体的实施例对本发明优选实施方案作进一步详细说明,本领域技术人员应当理解的是,这不应被理解为对本发明权利要求范围的限制。
下述实施例中采用的LSL-KrasG12D(01XJ6-B6.129-Krastm4Tyj)小鼠与Pdx1-Cre(01XL5-B6.FVB-Tg(pf1-cre)1Tuv)小鼠购买自美国国立癌症研究院;P53小鼠,即LSL-Trp53R172H/+小鼠,购自美国Jackson实验室。下述实施例中采用的试剂均常规市购获得。
实施例1:KC小鼠模型的构建和鉴定
LSL-KrasG12D小鼠及Pdx1-Cre小鼠按雌∶雄=3∶1进行合笼交配,待子代小鼠出生10天后剪鼠尾提取DNA进行PCR鉴定其基因型,以获得LSL-KrasG12D阳性且Pdx1-Cre阳性的小鼠。其中,鼠尾DNA的提取使用北京维通达生物技术有限公司生产的“组织/细胞裂解试剂盒”(100份次),按照说明书步骤提取。
其中,LSL-KrasG12D(01XJ6-B6.129-Krastm4Tyj)小鼠涉及的引物序列包括:
K004:5′-gtc gac aag ctc atg cgg gtg-3′(SEQ ID NO:1)
K006:5′-cct tta caa gcg cac gca gac tgt aga-3′(SEQ ID NO:2)
K005:5′-agc tag cca cca tgg ctt gag taa gtc tgc a-3′(SEQ ID NO:3)
上游引物为K004,下游引物为K006时,扩增片段为500bp的野生型Kras基因。上游引物为K005,下游引物为K006时,扩增片段为550bp的野生型Kras基因前所引入的lox基因。
其中,Pdx1-Cre(01XL5-B6.FVB-Tg(1pf1-cre)1Tuv)小鼠涉及的引物序列包括:
Pdx-F:5′-ctg gac tac atc ttg agt tgc-3′(SEQ ID NO:4)
Pdx-R:5′-ggt gta cgg tca gta aat ttg-3′(SEQ ID NO:5)
上游引物为Pdx-F,下游引物为Pdx-R,扩增片段为650bp的转基因Cre重组酶。
分别针对Kras、Pdx1-Cre和Trp53进行PCR鉴定,具体方案如下。

(3)检测方法:琼脂糖凝胶电泳检测
采用1.5%(质量)琼脂糖凝胶对Kras和Pdx1-Cre进行检测;采用2%(质量)琼脂糖凝胶对Trp53进行检测;电压120V电泳约10分钟至20分钟。
LSL-KrasG12D/+小鼠PCR鉴定结果如图1所示。其中,图1所示的电泳图片为使用K005和K006引物扩增的条带。
Pdx-1-Cre小鼠PCR鉴定结果如图2所示。
KC小鼠PCR鉴定结果如图3所示。
实施例2:KC精子与野生型小鼠的卵进行IVF快速扩繁
(1)KC小鼠新鲜精子的准备:
①选择8周龄或以上的KC雄鼠(20周龄内最好),收集精子前1~2周安排合笼1次,采精前至少3天不要安排合笼交配。
②安乐死取出附睾尾和输精管。
③在解剖镜下,放到滤纸上去除所有脂肪和血管组织。
④在60mm的培养皿中(Falcon 353004)中央加上90μl精子预处理液(TYH+0.75mMMBCD(配方见下表);先作一个45μl的滴,盖上石蜡油,然后再补加45μl)。覆盖上石蜡油后,在37℃CO2培养箱中平衡10~20分钟。
精子预处理液:
*使用0.22μm滤器过滤,1ml等分后,在4℃有效期3个月
⑤将附睾放到精子预处理液附近(90μl TYH+MBCD),使用显微外科剪刀在附睾上剪切口。使用钟表镊从附睾中轻轻撕出一个“球形”精子团;并拖入精子分散的滴中。
⑥在37℃CO2培养箱中培养约30~60分钟,以便精子分散到整个培养液中。
(2)受精培养盘的准备
①在1ml HTF溶液的tube管中加入0.0307g GSH,盖盖,混匀。
②对于新鲜精子样品,加入10μl GSH溶液到4ml的高钙HTF溶液中,轻轻混匀备用(终浓度为0.25mM GSH)。
③使用前,用0.22μm过滤一下,备用。
④用60mm培养皿(Falcon 353004)作一个200μl的滴(用于新鲜精子样本);或者90μl的滴(用于冻融精子样本);在37℃5%CO2培养箱中孵育10~20分钟。

(3)体外受精卵母细胞的准备和IVF
①在注射hCG后约13~14小时处死经过超数排卵处理的供体C57BL/6母鼠。
注意:防止透明带变硬,排卵后时间越长,透明带会逐渐变硬,一般在注射hCG后13个小时取卵比较好。取卵应快一些,最好5分钟内完成。收集后尽快加入到IVF滴中。
②打开腹腔,取出输卵管,尽可能的减少血液、组织液和脂肪等。
③将输卵管置于灭菌过滤纸上,擦去血迹和组织液等。小鼠腹部用酒精棉擦拭后打开小鼠腹腔,用镊子等移动腹腔内脏器,露出子宫、输卵管和卵巢等。取出输卵管。
④将输卵管置于含有受精液(HTF+GSH)的培养皿的矿物油中。
⑤用一把精细镊子固定住输卵管,用一把解剖针刺破输卵管的膨大部,带出卵母细胞团至受精液(HTF+GSH)培养液中。然后将输卵管弃去。
⑥卵母细胞在培养箱中孵育30分钟后,加入精子。冻/融精子在加入受精滴前需要在培养箱中至少孵育30分钟。从孵育滴的边缘使用宽口径吸头吸取10μl精液,这个区域包含运动能力最好的精子,尽量避免吸取任何精子碎片。新鲜精子,在TYH+MBCD溶液中在CO2培养箱孵育至少30~60分钟,同上加入3~5μl运动能力最好的精子,避免加入精子碎片。
注意:这一步应当在体视镜下非常小心的操作,使用宽口的10μl枪头(AxygenInc;T-400),这样可以容易的收集运动能力好的精子,避免吸取死精子和细胞碎片。精子量约为200~600个/μl。1个IVF培养盘可以放5~8只小鼠的卵母细胞团。
⑧重复1~6步,完成下一批体外受精实验。目前是从获得输卵管开始,5分钟内完成卵母细胞团的收集及孵育。
⑨当所有的卵母细胞加入精子受精后,拿出一个培养皿评估精子的运动能力和浓度,同时观察卵丘细胞是否被离散。如果精子运动力不佳,浓度低,很少卵丘细胞离散,可以在受精滴中补加一些精子到每一个受精皿中。
⑩将受精用培养皿置于37℃,5%CO2的培养箱中培养。受精3~4小时后,用新鲜的HTF培养液清洗2次,并观察胚胎。注意:清洗中识别和分离出单性生殖卵母细胞这个步骤是很重要的,在这个过程中,如果没有分离出,这些卵母细胞可能发育到囊胚。因此,尽可能地在2细胞期将单性生殖的卵母细胞分离出去。
(4)洗涤和培养受精的卵母细胞
①使用60mm培养皿(Falcon 353004)准备4滴150μl的HTF(不添加GSH),并覆盖上石蜡油。把培养皿放到37℃CO2培养箱3个小时或过夜平衡。
②把受精卵依次过1,2,3滴,去除细胞碎片、退化的卵母细胞和死精子。
③将假定的合子(受精卵)放到第4个滴,在CO2培养箱培养过夜。
④第二天早上,将1-cell胚胎和退化的卵母细胞转移到第3个滴中。
⑤准备1商透明质酸酶溶液(即300μg/ml透明质酸酶溶液,下面简称为M2或M2溶液)。在37℃孵育几分钟,轻吹除去黏附的细胞和精子。说明:这一步仅对于存在2-cell胚胎透明带黏附有卵丘细胞的情况。
(5)2-cell期胚胎的输卵管移植和仔鼠编号
①将受体麻醉,手术取出卵巢连接输卵管,用脂肪镊固定,在显微镜下找到输卵管开口。
②吸取2-cell胚胎,吸取方法是先吸一段较长的M2溶液,吸一个气泡,然后吸取受精卵,尽量紧密排列,再吸一段液体,吸一个气泡,再吸一段液体,共四段液体三个气泡。除较长的那段液体,其余的液体大致0.5cm~1cm左右,气泡0.2cm左右。将移植管口插入输卵管口,轻轻将移植管内的液体吹入,看到输卵管壶腹部膨大并清晰地看到三个气泡,即移植成功。
③将卵巢连同输卵管放回腹腔,缝合肌肉和皮肤。
④手术后19天左右仔鼠分娩,得到的仔鼠。仔鼠7天左右,准备编号和PCR鉴定。
(6)KC仔鼠PCR鉴定
与实施例1中KC小鼠的PCR鉴定方案相同。
实施例3:P53纯合小鼠模型的构建和鉴定
P53小鼠,LSL-Trp53R172H/+小鼠,购自美国Jackson实验室(https://www.jax.org/strain/08652)。小鼠在JAX网站上建议为杂合扩繁,纯合鼠容易自发肿瘤。
P53纯合小鼠雌雄均可以扩繁用,但只可用1~2胎左右;且需要严格执行实验动物伦理要求,即(1)动物肿瘤大于1500mm3时;(2)动物体重下降20%时;(3)动物外观异常,需要安乐死时。保种采用常规方法,Trp53敲除鼠应杂合保种。
PCR鉴定涉及的引物序列如下:

实施例4:P53纯合小鼠的精子与野生型小鼠的卵进行IVF快速扩繁
(1)LSL-Trp53R172H/R172H小鼠新鲜精子的准备
①选择8周龄或以上的P53雄鼠(20周龄内最好),收集精子前1~2周安排合笼1次,采精前至少3天不要安排合笼交配。
②安乐死取出附睾尾和输精管。
③在解剖镜下,放到滤纸上去除所有脂肪和血管组织。

⑤准备1滴透明质酸酶溶液(即300μg/ml透明质酸酶的M2溶液)。在37℃孵育几分钟,轻吹除去黏附的细胞和精子。说明:这一步仅对于存在2-cell胚胎透明带黏附有卵丘细胞的情况。
⑥用2滴M2清洗胚胎;然后将胚胎进行输卵管移植;或者冻存;或者在KSOM+AA中继续培养。
(5)2-cell期胚胎的输卵管移植和仔鼠编号
①将受体麻醉,手术取出卵巢连接输卵管,用脂肪镊固定,在显微镜下找到输卵管开口。
②吸取2-cell胚胎,吸取方法是先吸一段较长的M2,吸一个气泡,然后吸取受精卵,尽量紧密排列,再吸一段液体,吸一个气泡,再吸一段液体,共四段液体三个气泡。除较长的那段液体,其余的液体大致0.5cm~1cm左右,气泡0.2cm左右。将移植管口插入输卵管口,轻轻将移植管内的液体吹入,看到输卵管壶腹部膨大并清晰地看到三个气泡,即移植成功。
③将卵巢连同输卵管放回腹腔,缝合肌肉和皮肤。
④手术后19天左右仔鼠分娩,得到的仔鼠。仔鼠7天左右,准备编号和PCR鉴定。
(5)P53仔鼠PCR鉴定
与实施例3中P53鼠的PCR鉴定方案相同。
实施例5:P53纯合和杂合小鼠与KC小鼠扩繁获得KPC小鼠
在快速扩繁的同时,采用P53纯合和杂合小鼠与KC小鼠互配扩繁,按照雌∶雄=2~3∶1进行合笼交配,待子代小鼠出生7~10天后剪鼠尾提取DNA进行PCR鉴定其基因型,以获得KPC小鼠(LSL-Trp53R172H/+;LSL-KrasG12D/+;Pdx1-Cre阳性的小鼠)。
PCR鉴定方案与实施例1相同。
KPC小鼠PCR鉴定结果如图5所示。
最后,需要注意的是:以上列举的仅是本发明的具体实施例子,当然本领域的技术人员可以对本发明进行改动和变型,倘若这些修改和变型属于本发明权利要求及其等同技术的范围之内,均应认为是本发明的保护范围。

Claims (10)
1.一种胰腺癌动物模型的构建方法,其特征在于,包括如下步骤:
(1)构建KC鼠并对KC鼠进行扩繁得到KC鼠的子代;
(2)准备P53鼠并对P53鼠进行扩繁得到P53鼠的子代;
(3)采用所述KC鼠的子代与所述P53鼠的子代进行繁殖得到KPC胰腺癌鼠模型;
所述KC鼠是LSL-KrasG12D/+;Pdx-1-Cre鼠;
所述P53鼠是LSL-Trp53R172H/+鼠;
所述KPC胰腺癌鼠是LSL-KrasG12D/+;LSL-Trp53R172H/+;Pdx-1-Cre鼠。
2.根据权利要求1所述的胰腺癌动物模型的构建方法,其特征在于,步骤(1)中,所述构建KC鼠采用如下方法进行:将LSL-KrasG12D鼠与Pdx1-Cre鼠按照雌:雄=1:1至3:1进行合笼交配,获得的LSL-KrasG12D阳性且Pdx1-Cre阳性的子代即为所述KC鼠。
3.根据权利要求1所述的胰腺癌动物模型的构建方法,其特征在于,步骤(1)中,所述对KC鼠进行扩繁得到KC鼠的子代采用如下方法进行:采用所述KC鼠与野生型鼠自然交配进行扩繁得到KC鼠的子代;或者,采用所述KC鼠的精子与野生型鼠的卵子通过快速扩繁进行扩繁得到KC鼠的子代。
4.根据权利要求3所述的胰腺癌动物模型的构建方法,其特征在于,所述KC鼠是杂合KC鼠。
5.根据权利要求1所述的胰腺癌动物模型的构建方法,其特征在于,步骤(2)中,所述对P53鼠进行扩繁得到P53鼠的子代采用如下方法进行:采用所述P53鼠与野生型鼠自然交配进行扩繁得到P53鼠的子代;或者,采用所述P53鼠的精子与野生型鼠的卵子通过快速扩繁进行扩繁得到P53鼠的子代。
6.根据权利要求5所述的胰腺癌动物模型的构建方法,其特征在于,所述P53鼠是纯合P53鼠或杂合P53鼠。
7.根据权利要求1所述的胰腺癌动物模型的构建方法,其特征在于,步骤(3)中,采用所述KC鼠的子代与所述P53鼠的子代自然交配进行繁殖得到KPC胰腺癌鼠模型;或者,采用所述KC鼠的子代的卵子与所述P53鼠的子代的精子通过快速扩繁进行繁殖得到KPC胰腺癌鼠模型。
8.根据权利要求7所述的胰腺癌动物模型的构建方法,其特征在于,所述P53鼠的子代是P53纯合雄鼠或P53杂合雄鼠。
9.根据权利要求8所述的胰腺癌动物模型的构建方法,其特征在于,所述P53鼠的子代是P53纯合雄鼠。
10.根据权利要求7所述的胰腺癌动物模型的构建方法,其特征在于,所述KC鼠的子代是杂合KC鼠。
