背景技术
实体肿瘤(solid tumor)周围的微环境通常缺氧。肿瘤周围的组织中脱氧血红蛋白与氧合血红蛋白浓度的比率高于健康组织中的比率。通过测量组织氧合,可以评估肿瘤的功能行为。组织氧合状态也可用于评估常规或新辅助化疗的有效性以及确定疾病进展和帮助预后。

发明内容

本公开描述了一种使用光子混合器设备(Photonic Mixer Device,PMD)通过频域NIR光谱学和断层成像测量实体肿瘤周围的组织的氧合状态的技术。PMD是基于CCD技术或CMOS技术的半导体结构,其中每个像素包括两个电荷存储位置(即,子像素)。由射频(Radiofrequency,RF)控制电压确定,PMD中的光电子交替地分配给两个子像素。RF电压信号被锁相到以相同频率调制的场景照射光源。从场景反射的光生成光电子,这导致电荷在两个子像素处积累。在两个子像素处收集的电荷给出反射光信号的同相分量和180°异相分量。使用该信息,使用下面所示的方程(1)计算反射光的相移。

在示例性实施例中,NIR光源20可以以10-1000MHz范围内的频率进行振幅调制。例如,在一些实施例中,NIR光源20可以以200MHz进行振幅调制。在另一实施例中,NIR光源可以以30MHz进行振幅调制。

图5示出了根据示例性实施例的NIR光谱学设备10'的示意图。由于PMD ToF相机的小外形因素,设备10'可以被实施为手持式设备、可穿戴设备或可植入设备。除了NIR光源20和检测器40之外,设备10'可以包括处理器50、通信系统60和电力系统80。处理器50可以控制设备10'的整体操作。处理器50也可以处理和/或分析记录的相位和振幅数据,并确定目标组织的氧合状态。通信系统60可以包括有线或无线通信能力,包括天线、收发器、编码器、解码器等。在一些实施例中,通信系统60可以将处理后的数据传输到远程存储设备或其中可以显示氧合状态的远程显示设备。在一些实施例中,通信系统60可以将原始数据传输到其中可以执行计算并且结果可以传输到患者或健康护理提供商的远程处理设备或云服务器。在一些实施例中,设备10'可以包括片上电子器件,以在由处理器50处理之前或者在传输到远程处理设备或云服务器之前预处理记录的数据。在这样的实施例中,设备10'可以包括放大器、模数转换器、多路复用器和其他电子电路,以预处理获取的数据。电力系统80可以为处理系统50、通信系统60、NIR光源20和检测器40供电。在一些实施例中,设备10'可以无线供电。在这样的实施例中,电力系统80可以包括超级电容器、电池或可以由远程设备无线地充电的一些其他类型的充电系统。在一些实施例中,使用光伏电池阵列的光供电可用于为设备10'的嵌入式电子器件供电或为其电池再充电。
示例:PMD-ToF相机针对相移测量的应用
对于半无限介质(具有类似于组织的光学特性)中的许多不同的调制频率,计算入射光和反射光之间的相移,以验证代表性相移可使用PMD ToF相机测量。图4示出了在不同的源-检测器间隔ρ下不同调制频率(f)的相移。结果表明,随着源-检测器间隔的增加,相移增加。作为示例,当ρ=5cm和f=30MHZ时,相移为35°,这相当于~3ns的时间延迟。当用于距离感测时,~3ns的时间延迟对应于45cm的相机到物体的距离,这可以被PMD ToF相机检测到,因此意味着相应的35°的相移也可以被PMD ToF相机检测到。

Claims (20)
1.一种用于连续监测组织区域中的肿瘤的设备,该设备包括:
PMD相机芯片;和
至少一个调幅近红外光源,其与PMD相机芯片水平地分离,使得PMD相机芯片和近红外光源处于反射几何结构。
2.根据权利要求1所述的设备,其中,所述PMD相机芯片是多像素PMD相机芯片。
3.根据权利要求1所述的设备,其中,所述至少一个调制的近红外光源包括排列在所述PMD相机芯片周围的调制的近红外光源阵列。
4.根据权利要求1所述的设备,其中,所述至少一个调制的近红外光源以不同的频率调制。
5.根据权利要求1所述的设备,还包括电机,以在PMD相机芯片周围移动所述至少一个调制的近红外光源。
6.根据权利要求1所述的设备,还包括无线通信系统,以向远程处理单元或云服务器传输记录的数据。
7.根据权利要求1所述的设备,还包括无线电力系统。
8.根据权利要求1所述的设备,其中,所述调制的近红外光源和PMD相机芯片之间的距离是肿瘤距组织区域的表面的深度的大约两倍。
9.一种用于连续监测肿瘤缺氧的方法,该方法包括:
a.用紧邻肿瘤提供的调幅近红外光源照射具有肿瘤的组织区域;
b.使用紧邻肿瘤提供的多像素PMD相机芯片记录从组织区域反射的光,其中所述近红外光源与所述PMD相机芯片水平地分离;
c.测量反射光的振幅和相移;
d.使用反射光的振幅和相移计算吸收系数和约化散射系数;
e.对至少两种不同波长的光重复步骤a-d;以及
f.使用针对至少两种不同波长的光计算的吸收系数和约化散射系数来计算组织区域中氧合血红蛋白和脱氧血红蛋白的浓度。
10.根据权利要求9所述的方法,其中,所述组织区域用排列在所述PMD相机芯片周围的多个近红外光源照射。
11.根据权利要求10所述的方法,其中所述多个红外光源以多路复用的方式操作,使得每个光源的振幅和相移被单独记录。
12.根据权利要求9所述的方法,其中所述至少两种波长的光的第一波长高于血红蛋白的等吸收点,并且所述至少两种波长的光的第二波长低于血红蛋白的等吸收点。
13.根据权利要求12所述的方法,其中所述第一波长为约780nm,并且所述第二波长为约830nm。
14.根据权利要求7所述的方法,其中所述近红外光源以10-1000MHz范围内的频率进行调制。
15.根据权利要求14所述的方法,其中所述近红外光源以30MHz的频率进行调制。
16.根据权利要求14所述的方法,其中所述近红外光源以200MHz的频率进行调制。
17.根据权利要求9所述的方法,其中所述振幅和相移值被无线传输到云服务器,用于计算吸收系数和约化散射系数以及氧合血红蛋白和脱氧血红蛋白的浓度。
18.根据权利要求9所述的方法,其中所述调制的近红外光源和所述PMD相机芯片位于肿瘤距组织区域的表面的深度的大约两倍的距离处。
19.根据权利要求9所述的方法,其中所述近红外光源和所述PMD相机芯片被无线供电。
20.根据权利要求9所述的方法,其中,所述近红外光源围绕所述PMD相机芯片旋转。
