优选实施方式的详述
描述了通过用游离氨基酸β-丙氨酸或其盐或酯进行膳食补充来改善涉及创伤后应激障碍(PTSD)的生理反应的方法和实施方式,并且这些方法和实施方式仅是出于说明的目的。本文中所述的方法可用于许多不同的工业,包括例如卫生保健、军队、准军事性质的组织、现场急救员例如消防员和门诊人员、急诊和外科人员、运动队和许多其他人员。以下提供本发明的某些实施方式的进一步描述。如本文中描述和要求保护的,某些术语被定义并可交换地使用。

本发明提供在一段时间内提供的营养或膳食补充品如何能改善涉及创伤后应激障碍的生理反应的重要理解。
形式和配方
β-丙氨酸可作为游离氨基酸β-丙氨酸给予,其中,所述游离氨基酸不是二肽、寡肽或多肽的一部分。该游离氨基酸可以是β-丙氨酸的酯或盐。游离氨基酸可以采取丸、片、胶囊、颗粒或粉末的形式。游离氨基酸可以作为固体、液体或半液体的一部分给予。游离氨基酸可作为饮料(例如运动饮料)或食物(例如健康棒)的一部分给予。

一旦通过使用有效量的人膳食补充品增加了β-丙氨酰组氨酸的水平,或者称为装载阶段(loading phase),出于本发明的目的可调整定量给予以维持改善涉及创伤后应激障碍的生理反应所需的β-丙氨酰组氨酸的水平。
本文中提供的形式和配方可以是美国专利第5,965,596、6,426,361、7,825,084、8,067,381和8,329,207号中描述和提供的那些,每篇专利以其整体通过引用合并入本文。
在一个方面,为一份或更多份食物配制膳食补充品,所述一份或更多份食物每天被摄取一次或更多次,以达到本发明要求的有效量。因此,满足有效量的游离β-丙氨酸或其酯或盐所需的总的每日摄取量可通过单份食物或通过一天中的多份较少的食物来获得,所述多份食物总体上满足要求的游离β-丙氨酸或其酯或盐的量,其是膳食补充品的总的每日摄入中的有效量。因此,可以用较少量的游离β-丙氨酸或其酯或盐来配制膳食补充品,目的是一天多份食物,其中多份食物的总量满足所需的每日总摄入以实现本发明定义的有效量。
β-丙氨酸可以大约100g/kg组合物的浓度给予。在某些实施方式中,浓度为约70g/kg-约130g/kg、约80g/kg-约120g/kg和/或约90g/kg-约110g/kg。
可作为游离氨基酸β-丙氨酸或其酯或盐和一种或更多种其它化合物的混合物(组合物)提供β-丙氨酸。所述一种或更多种其它化合物可以是但不限于葡甘露聚糖。在某些实施方式中,所述混合物可以是约80%β-丙氨酸或其酯或盐和约20%葡甘露聚糖。在某些实施方式中,游离β-丙氨酸或其酯或盐的百分率可以是约50%-约95%、约60%-约90%和/或约70%-约85%。
可作为溶液提供所述混合物。所述溶液可包括每约100ml水中约1.0-1.2g组合物。

如本领域的技术人员通过本发明的公开将理解的,其它成分,例如肌酸、其它氨基酸和碳水化合物可以类似于针对游离氨基酸β-丙氨酸或其酯或盐所述的类似的量存在于人膳食补充品中。
实施例
在一个实施方式中,对创伤后应激障碍(PTSD)的动物模型中的行为和神经内分泌反应研究了30天的β-丙氨酸(BA)摄取(100g·kg-1)。对动物饲喂具有或不具有(PL)BA补充(100mg·kg-1)的正常饮食达30天。在补充期后,使动物暴露于捕食者-气味压力(PSS)或对照组(UNEX),PSS已被验证用于动物模型以刺激PTSD样的行为。在暴露后7天,用高架十字迷宫和声音惊吓反应评估行为。1天后分析皮质酮浓度(CS)、脑源性神经营养因子(BDNF)的表达和脑肌肽浓度。PSS+PL中的动物比PSS+BA、UNEX+BA或UNEX+PL中的动物在EPM中的开放臂中花费显著更少的时间并且进入的次数显著更少。PSS+BA中的动物具有与UNEX+BA中的动物相当的得分。PSS+PL中的动物相比于PSS+BA中的动物或未暴露的动物,焦虑指数更高(p<0.05)。与未暴露的动物相比,在暴露于PSS的动物中ASR和冻结更剧烈(p<0.05)。与未暴露的动物相比,在暴露于PSS的动物中CS表达更高(p<0.05)。与PL动物相比,在用BA补充的动物中在海马和其它脑切片中的脑肌肽浓度显著更高。与暴露并且补充BA的动物或未暴露的动物相比,在暴露并且饲喂正常饮食的动物中海马的CA1和DG亚区中的BDNF表达更低(p<0.05)。就这点而论,BA补充可提供脑肌肽浓度的增加、和PTSD样行为的减少,其部分由维持海马中BDNF表达来介导。
总之,此研究的结果显示,30天的β-丙氨酸的摄取能改善涉及创伤后应激障碍的生理反应。
方法
动物:
使重200-250gm的成年雄性Sprague-Dawley大鼠(n=122)习惯于舍饲条件达至少七天。使所有的动物每笼四只居住在具有稳定温度和颠倒的12-h光线/黑暗循环的动物房内,可无限制地获得食物和水。在80:20共混物中以粉末形式提供β-丙氨酸与葡甘露聚糖。向大鼠每kg体重提供100mg该粉末(总共30mg的粉末溶解在25ml水中)。以相同的相对剂量向PL处理的大鼠提供载体(葡甘露聚糖)。每天处理动物一次。所有测试都在黑暗期在暗红色光条件下进行。该研究根据国立卫生研究所对于实验动物的护理和使用指南的原则和指导方针进行。所有的处理和测试程序得到以色列内盖夫的本-古里安大学的动物护理委员会(Animal Care Committee)的批准。
实验设计
将大鼠随机分配至四个处理组中的一个(每组n=30或31只):
1)未暴露的并且载体处理的组(UNEXP+PL):给大鼠饲喂普通的食物和水达30天并使它们暴露于新鲜、未使用的褥草(litter)达15min。
2)未暴露的并用β-丙氨酸处理的组(UNEXP+BA):在大鼠的水里给它们提供β-丙氨酸并且使它们暴露于新鲜的、未使用的褥草达15min。
3)暴露的并且载体处理的组(EXP+PL):给大鼠饲喂普通的食物和水达30天并使它们暴露于PSS达15min。
4)暴露的并用β-丙氨酸处理的组(EXP+BA):在大鼠的水里给它们提供β-丙氨酸并且使它们暴露于PSS。
在7天的顺应期(其中所有大鼠接受正常的粉末饮食)之后,将它们随机分成四组。在30天正常饮食或β-丙氨酸补充的饮食之后,使大鼠暴露于PSS或对照组方案。在PSS或对照组方案后7天进行所有行为测试,并随后在24小时后处死大鼠并取出脑。维持饮食直到研究结束。该模型的有效性已经在数个研究(Cohen等人,2004;2012b;Kozlovsky等人,2007)中得到证实。
捕食者-气味压力(PSS)
在30天的补充方案后,使动物暴露于PSS方案。PSS方案由在封闭的环境中将实验动物置于充分污染的猫砂(猫使用2天,筛掉粪便)达10分钟组成。使对照动物暴露于新鲜的未使用的褥草达相同量的时间。情景提示由将动物置于新鲜的、未使用的猫砂达15分钟组成。
评估计划
在高架十字迷宫、声音惊吓反应和情景冻结中评估行为反应。使用EthoVision自动跟踪系统(Noldus Information Technology,The Netherlands)记录并分析所有结果。在高架十字迷宫和声音惊吓反应中的表现发生在初始暴露于PSS后7天。情景冻结测量在初始暴露后第8天进行。从PSS延迟进行这些测量是基于以下发现:极端行为变化在暴露7天后保持恒定并代表在延长的一段时间内持续的“慢性症状”(Cohen等人2004)(Cohen和Zohar,2004;Cohen等人,2004)。在行为评估后,处死所有动物并取出脑用于分析。

高架十字迷宫(EPM)
以前已经有描述在EPM中进行的行为评估(Cohen H等人,2003;2012)。EPM是具有两条相对的开放臂和两条相对的闭合臂(仅朝中心平台开放并且在三个侧面被14-cm高的不透明壁围绕)的十字形平台。将大鼠放置在面朝开放臂的中心平台,并允许其探索迷宫5min。将每次实验过程录到录像带上并且随后由独立的观察者记分。臂进入定义为所有四个爪子都进入臂。被评估的行为是:在开放臂和闭合臂和中心平台上花费的时间(持续时间);进入开放臂和闭合臂的次数;和总探索(进入所有臂)。总探索计算为进入迷宫的任何臂的次数以便区分受损的探索行为、限制于闭合臂的探索(逃避)和自由探索。“焦虑指数”,整合EPM行为测量的指数,按如下计算:
焦虑指数值在0-1范围内,其中指数的增加表示增加的焦虑样行为。
声音惊吓反应
使用两个通风的惊吓室(SR-LAB系统,San Diego Instruments,San Diego,CA)测量惊吓反应。常规地使用SR-LAB校准单元以保证在测试室之间和随时间一致的稳定计灵敏度。每个Plexiglas圆筒安置在隔音的通风室内的平台上。由框架下的压电加速度计检测在管内的运动。使用声级计常规地测量每个测试室内的声级以确保一致的呈现。每个测试期以对68dB的背景白噪音的5-min适应期开始,随后是在六个木块中的30次声音惊吓试验刺激(40ms持续时间的110dB白噪音,30或45秒试验间间隔)。行为评估由平均惊吓幅度(全部30次试验的平均)和对声脉冲的重复呈现的惊吓适应的百分率。适应百分率-对第一个木块的声刺激和最后一个木块的声刺激的反应之间的变化百分率-按如下计算:
情景冻结测量
在情景提示/线索暴露的过程中为冻结行为打分,并定义为所有运动(除了呼吸)的缺失(Kim等人1992)。测量总累积冻结时间(在各评估期过程中冻结花费的总秒数)并计算为总时间的百分率。使用头顶摄影机记录冻结行为并使用记录的图像为静止打分。由不知道处理条件的受训练的观察者对录影带和记录的图像两者打分。
取血样
用铡刀将来自每一组的二十只大鼠斩首。注意使情景压力最小化:每次处死之间彻底清洁该区域并移除身体。收集躯干血并在室温下保留2hr并且随后用Hermle离心机离心(1000g,4℃下达10min)。收集血清(每只大鼠大约1ml)并储藏在-80℃下直到进行分析。
血清皮质酮的测量
使用ELISA测试试剂盒(Endocrine Technologies Inc.Newark,CA)根据生产商的用法说明测量血清皮质酮。在450nm读板。所有的反应都一式两份测定。批间测定变异是6.1%并且批内测定变异是5.9%。所有样品都一式两份测量。此测定中可检测的最低水平是0.1ng/mL血清。
脑BDNF水平
组织制备
在行为测试后24小时,深度麻醉(氯胺酮和甲苯噻嗪混合物)来自各组的10只动物并用冷的0.9%生理盐水、随后用在0.1M磷酸盐缓冲液(pH 7.4)中的4%多聚甲醛(Sigma-Aldrich)进行心脏灌注。快速除去脑,在4℃下后固定于相同的固定剂中达12h,并且在4℃下在0.1M磷酸盐缓冲液中的30%蔗糖中低温保存过夜。将脑在干冰上冷冻并保存在-80℃。使用低温恒温器(Leica CM 1850)对各动物收集背侧海马水平上的连续冠状切片(10μm),并装载在包被的载波片上。
免疫荧光:
将切成薄片的切片风干并在冷冻甲醇(2min)和在4%多聚甲醛(4min)中孵育。在含有Tween 20的磷酸盐缓冲盐水(PBS)(PBS/T)(Sigma-Aldrich)中洗涤三次后,在(PBS中的正常山羊或马血清)中的封闭液中孵育切片达60min,并且随后在4℃过夜,一级抗体抗BDNF(各1:250;Abcam)。在PBS/T中洗涤三次后,在含有2%正常血清的PBS中的DyLight-488中孵育切片达2h。洗涤切片,用封固剂(Vectrastain Vector laboratories,USA)装载。在缺乏一级抗体下进行对照染色。此外,交换二次荧光标记以检查交叉反应性,并且在没有任何一级抗体的情况下孵育切片以检查二级抗体的任何非特异性结合。
定量:
使用计算机辅助图像分析系统(Leica Application Suite V3.6,Leica,德国)来定量分析免疫染色并且采用50×物镜来评估海马中的BDNF-IR阳性细胞的数量,所述海马分成三个(分别计数)区域:CA1亚区、CA3亚区和齿状回(DG)。概述感兴趣的区域并且使用计算机辅助评估来计算CA1和CA3的锥体层和DG的颗粒层中的BDNF-IR细胞的数量。从来自各组的每只动物选择(在前囟-2.30和前囟-3.60之间)七个代表性的海马切片。由两位不知道处理方案的观察者来分析这些切片。对于各被调查的海马结构,使用标准技术来估计每单位面积的BDNF细胞数量概况。
脑肌肽浓度测量
通过LC-MS-MS分析来确定脑匀浆中的肌肽浓度。
将脑在冰上部分解冻并对六个脑区域取样:大脑皮层、下丘脑、海马、杏仁核、嗅球和丘脑。各样品称重并转移至个别小瓶中以进一步均质化。
使用Polytron均化器、在补充1mM EDTA的盐水(0.9%NaCl;100mg/1ml)中均质化脑样品。在LC/MS/MS分析之前将匀浆储存在-20℃±5℃。购自Sigma的L-肌肽用于制备用于校准曲线的标准品(以ppb计)。分析前样品稀释5倍。来自标准曲线的浓度读数乘以稀释系数并转化成ppm单位。
统计分析:
使用单向方差分析(ANOVA)进行统计分析。如果是显著的F比率,LSD事后分析用于成对比较。所有数据都报道为平均值±SD。皮尔森的积矩相关用于确定所选的二变量相关。p<0.05的α水平用于确定统计显著性。使用SPSS v22软件(SPSS Inc.,Chicago,IL)分析数据。
结果:
高架十字迷宫:
关于在开放臂中花费的时间,各组间的比较显示显著效果(F(3,118)=7.06,p<0.001)。事后分析显示,消耗正常饮食并暴露于PSS的动物比所有其它处理组在开放臂中花费显著更少的时间(p在<0.001–0.012范围内)。在开放臂中花费的时间上,没有注意到其它组间差异(参见图1A)。进入开放臂次数的比较(参见图1B)还显示组之间的显著的相互作用(F(3,118)=9.55,p<0.001)。暴露于PSS的正常饮食饲喂的动物比所有其它处理组显示显著(p<0.001)更少的进入次数(p<0.001)。在进入开放臂上次数没有注意到其它组差异。
所有组在EPM上的总活动性的分析(参见图1C)揭示在组之间没有显著差异(F(3,118)=0.925,p=0.431)。基于整合行为测量,注意到组之间的焦虑指数的显著差异(参见图1D)(F(3,118)=8.05,p<0.001)。饲喂正常饮食并暴露于PSS的动物显示比其它处理组显著更大程度的焦虑(p在<0.001–0.002范围内)(参见图1D)。没有注意到其它组间效果。
急性惊吓反应和惊吓适应
声音惊吓幅度中各组间的比较揭示显著的组差异(F(3,118)=14.02,p<0.001)(参见图2A)。暴露于PSS并被饲喂正常饮食的动物比未暴露的且不管饮食如何的动物(p<0.001),以及暴露于PSS并被饲喂BA的动物比被饲喂正常饮食(p<0.001)或用BA补充的饮食(p=0.0001)的未暴露的动物,惊吓幅度显著更大。此外,暴露于PSS并且其饮食中补充了BA的动物比没有被饲喂BA的暴露的动物倾向于具有更小的惊吓幅度(p=0.085)。在惊吓适应中也注意到显著的差异(F(3,118)=9.11,p<0.001)(参见图2B)。暴露于PSS且不管饮食如何的动物比所有其它组具有显著更低的适应得分(p<0.001)。没有注意到组之间的其它显著差异。
在第8天线索暴露对于冻结行为的影响:
图3描绘了线索暴露时的冻结行为。看到冻结行为在组之间存在显著差异(F(3,118)=9.342,p<0.001)。暴露于PSS并被饲喂正常饮食的动物比未暴露并且被饲喂正常饮食(p<0.001)或用BA补充的饮食(p<0.001)的动物在提供线索时具有显著更多的静止。也观察到暴露于PSS并且补充BA的动物比未暴露并且被饲喂正常饮食(p=0.01)或被饲喂用BA补充的饮食(p=0.003)的动物在提供线索时具有显著更多的静止。暴露于PSS并被饲喂BA的动物比暴露于PSS并被饲喂正常饮食的动物冻结少约6%(40.9%对34.8%)。然而,该差异不是统计学上的差异(p=0.148)。
暴露后第8天皮质酮浓度
在皮质酮浓度方面观察到组之间的显著差异(图4)(F(3,76)=13.883,p<0.001)。在暴露于PSS并被饲喂正常饮食的动物中的皮质酮浓度比未暴露并被饲喂类似饮食(p<0.001)或补充BA(p<0.001)的动物显著更高。此外,也观察到暴露于PSS并用BA补充的动物比未暴露的并饲喂正常饮食(p<0.001)或未暴露的并用BA补充(p<0.001)的动物具有显著更高的皮质酮浓度。在被饲喂正常饮食的暴露的大鼠与补充BA的暴露的大鼠之间未注意到显著的差异(p=0.837)。
PSS暴露后第8天BDNF表达
可分别在图5A–5C中观察在CA1、CA3和DG亚区中在BDNF表达方面的组之间的比较。在CA1亚区(F(3,36)=23.2,p<0.001)和DG亚区(F(1,36)=4.126,p=0.013)中注意到显著差异。在CA3亚区(F(1,36)=0.918,p=0.442)中未观察到组之间有差异。在暴露于PSS并被饲喂正常饮食的动物的CA1和DG亚区中的BDNF表达显著低于所有其它组(p在<0.001–0.011范围内)。此外,在CA1中,在暴露于PSS并补充BA的动物中的BDNF表达显著低于未暴露的动物(r在0.009–0.024范围内),但是仅显著高于被饲喂正常饮食的暴露的大鼠(p<0.001)。在DG亚区,在暴露于PSS但补充BA的动物中的BDNF表达与未暴露并被饲喂正常饮食(p=0.765)或补充BA(p=0.818)的动物在统计学上无差异,然而,在暴露的并被饲喂BA的这些动物中的BDNF的表达高于(p=0.011)暴露的并被饲喂正常饮食的动物。没有注意到其它差异。
PSS暴露后第8天脑肌肽浓度
在表1中可观察各个脑区域中肌肽的浓度。在海马(F(3,36)=5.818,p=0.002)、大脑皮层(F(1,36)=6.659,p=0.001)、下丘脑(F(3,36)=5.348,p=0.004)、杏仁核(F(3,36)=3.825,p=0.018)和丘脑(F(3,36)=5.162p=0.005)中注意到显著的差异。在嗅球(F(3,36)=2.051,p=0.124)中没有注意到显著的差异。在海马、皮层、下丘脑、杏仁核和丘脑中,摄入BA并暴露于PSS的动物比被饲喂正常饮食且无论是否暴露的动物具有显著更高的肌肽浓度(p在<0.001–0.011范围内)。未暴露于PSS并补充BA的动物相比于未暴露的并被饲喂正常饮食的动物在海马(p=0.030)和丘脑(p=0.042)中具有显著更高的肌肽浓度。在大脑皮层、下丘脑和杏仁核中,当与被饲喂正常饮食且无论是否暴露的动物相比时,未曾暴露于PSS并补充BA的动物之间的其它比较似乎仅导致强的趋势(在0.051–0.089范围内)。仅在下丘脑(p=0.113)和杏仁核(p=0.115)中,在未暴露的并被饲喂BA的动物与暴露的并被饲喂正常饮食的动物之间的比较中未注意到差异。

讨论
此研究的结果显示,大鼠摄入β-丙氨酸30天有效地减少一些所测试的并与暴露于PSS关联的行为。与未暴露的动物或暴露并补充BA的动物相比,观察到被饲喂正常饮食并暴露于PSS的大鼠当被置于高架迷宫中时显著更不活跃,并且具有更高的焦虑水平。观察到暴露但补充β-丙氨酸的动物比暴露的并且未补充的动物,惊吓反应低19%(p=0.085)并且冻结反应低15%(p=0.148)。
此研究中使用的PTSD的动物模型以前已经被确立为检查应激的特定反应模式的生物分子和生理参数的确凿和有效的方法。此模型也已经被确立为确定各种干预对应激的行为反应的影响的有效方法。在PSS+PL分别与UNEX+PL或UNEX+BA之间看到的惊吓幅度反应的差异(高2.5–2.2倍)与以前报道中的2.5倍增加一致。此研究中,与未暴露的动物相比,暴露于PSS还导致焦虑指数升高30%,和冻结反应增加几乎2倍。
暴露于PSS还伴随着压力后7天血浆皮质酮浓度的显著升高。这与使用相同压力模型的其它研究一致。糖皮质激素的升高与暴露于压力相关,但是最近的证据暗示,糖皮质激素的升高可在增强与损害恢复中起作用。以前的研究已经显示,对急性应激的减弱的下丘脑-垂体肾上腺轴反应可增加PTSD样症状的易感性,这可用给予大剂量的可的松的处理来逆转。然而,糖皮质激素浓度似乎证明倒U效果。虽然低浓度可能是PTSD的预报,已经显示高浓度也与神经元的形态改变相关,神经元的形态改变也有助于在应激过程中见到的消极的行为反应。海马对急性和慢性应激都敏感。慢性应激反应可提升大脑中糖皮质激素表达,造成海马中神经元经历可逆的重塑,特别在DG-CA3区域。这些变化通常涉及树突萎缩,这可导致在依赖于海马的记忆任务中功能下降。相比之下,BDNF被报道通过增强神经形成和树突重塑而对神经可塑性具有相反的作用。此研究中在暴露于PSS的两组大鼠中都观察到的皮质酮浓度的提高可能是那些动物经历的应激的指示。然而,关于其焦虑水平和在EPM中的表现,补充BA的动物似乎恢复或显示对应激的更强的复原。这可能是脑肌肽水平的升高和BDNF表达增加两者的作用。这得到在此研究中观察到的CA1和DG亚区中BDNF表达的增加与所检查的动物的惊吓反应(分别为r=-0.341,p=0.031和r=-0.523,p=0.001)和冻结反应(分别为r=-0.572,p<0.001和r=-0.335,p=0.035)的减少的所见到的关联的支持。
此研究中补充β-丙氨酸的大鼠经历在脑的几乎所有部分中的肌肽浓度的显著升高。补充β-丙氨酸对嗅球中肌肽浓度似乎没有任何影响。这可能与大多数哺乳动物物种的嗅球中通常见到的高肌肽浓度有关。然而,在其它脑区域中肌肽浓度的差异支持以前的研究,它们显示补充β-丙氨酸的饮食可增加小鼠和鸡中的脑肌肽浓度。脑肌肽通过减少去甲肾上腺素的脑代谢物、或通过经其组分之一-组氨酸-刺激组胺能神经元可减弱抑郁样行为。其他人曾提出,肌肽可能影响脑抗氧化活性,而其他人曾报道,在下丘脑和大脑皮层中脑肌肽浓度的升高与小鼠的海马中BDNF浓度的增加有关。在本研究中,与PSS+PL大鼠相比,在PSS+BA组中脑肌肽浓度升高与维持海马的CA1和DG亚区中的BDNF表达关联。在海马、皮层、下丘脑、杏仁核和丘脑中肌肽浓度的升高与焦虑指数(r在-0.471--0.550范围内,p<0.002)反向关联,与在开放臂中花费的时间(r在0.453–0.521范围内,p<0.003)的增多正向相关。
在暴露于PSS和被饲喂正常饮食的啮齿动物中观察到的BDNF表达的显著下降与以前的研究一致,这些研究报道BDNF表达减少和PTSD样行为应激反应的减弱。尽管β-丙氨酸摄取和随后在脑中的肌肽合成似乎并不增加未暴露的大鼠中的BDNF表达,但是,其似乎的确维持暴露于PSS的那些大鼠中的BDNF表达。在UNEX+BA和UNEX+PL两者中类似的BDNF表达暗示,脑肌肽水平的升高不直接负责改变BDNF表达。这些结果表面上似乎与显示β-丙氨酸摄入后在海马匀浆中BDNF浓度显著升高的研究截然不同。然而,那些研究者没有使用非应激组作为对照。海马中升高的脑肌肽和BDNF表达的维持的机制尚未得到很好地理解,但是,这可能与通过其充当抗氧化剂而作为神经保护剂的作用有关。脑中的氧化应激和炎症已被证明造成PTSD的发展和进一步的加剧。因此,肌肽作为抗氧化剂的作用可防止与糖皮质激素升高有关的神经变性并间接保存BDNF表达。
总之,此研究的结果显示,补充β-丙氨酸30天似乎促进由PSS复原和/或恢复。与脑肌肽升高相关的保护效果可能与海马中BDNF表达的保护相关。β-丙氨酸可用作治疗或预防PTSD的膳食补充品。

Claims (19)
1.改善涉及创伤后应激障碍的生理反应的方法,所述方法包括:
在一段时间内以有效量给予包含游离氨基酸β-丙氨酸或其盐的人膳食补充品以改善涉及创伤后应激障碍的一种或更多种生理反应。
2.权利要求1的方法,其中,所述生理反应选自下组:不想要的令人痛苦的创伤事件的记忆、再体验创伤事件如同其再次发生(重现)、关于创伤事件的令人苦恼的梦、严重的精神折磨或对使人想起创伤事件的事物的身体反应、试图避免想起或谈论创伤事件、回避使人想起创伤事件的地点、活动或人、关于自身或他人的消极的感觉、不能体验积极的情感、感觉情感麻木、对曾经喜爱的活动缺乏兴趣、对未来不抱希望、记忆力问题、维持紧密关系有困难、易怒、爆发愤怒或攻击行为、总是在提防危险、压倒性的内疚或害羞、自我毁灭行为、集中注意力有困难、睡眠有困难、容易受惊吓或害怕、以及它们的组合。
3.权利要求1的方法,其中,每天给予所述人膳食补充品一次或更多次,持续多天。
