Abstract

本发明公开了一种防治仔猪腹泻的复合益生菌制剂及其制备方法和应用。该复合益生菌制剂包括5种益生菌,所述5种益生菌分别为Lactobacillus frumenti、Lactobacillus gasseri LA39、Butyricicoccus pullicaecorum、Eubacterium hallii和Blautia hansenii。将制得的五种细菌悬液分别离心获取细菌沉淀,用磷酸盐缓冲液PBS重悬细菌沉淀,将五种活菌悬液混合均匀,得到复合益生菌制剂;每毫升复合益生菌制剂中总活菌数量为101~1020个。本发明的益生菌制剂的制作过程简单,生产成本较低,具有显著的防治仔猪腹泻效果。

Description

防治仔猪腹泻的复合益生菌制剂及其制备方法和应用
技术领域
本发明涉及防治仔猪腹泻的生物制剂,具体地指一种防治仔猪腹泻的复合益生菌制剂及其制备方法和应用。
背景技术
仔猪由于免疫系统发育不完善,在受到外界刺激时易发生腹泻。尤其是仔猪的早期断奶措施会从营养、心理和环境等方面刺激仔猪,导致仔猪发生早期断奶应激,严重时引起仔猪发生腹泻甚至死亡。生产实践中广泛使用抗生素来缓解仔猪腹泻,但随着抗生素的耐药性和食品安全等问题的日益突出,禁用抗生素已经成为一种必然趋势。
目前常用益生菌制剂来部分替代猪用抗生素的使用。但单一益生菌制剂由单一微生物构成,存在作用方式单一、抗仔猪腹泻效果不显著等不利因素,从而在一定程度上限制了单一益生菌制剂的推广与应用。复合益生菌制剂是由两种或两种以上的微生物构成的制剂,微生物之间可优势互补,具有协同作用,其抗腹泻效果比单一益生菌制剂更佳。因此如何制备具有高效防治仔猪腹泻效果的复合微生态制剂成为当前的研究热点。
发明内容
本发明的目的是提供一种防治仔猪腹泻的复合益生菌制剂及其制备方法和应用。该复合益生菌制剂由5种益生菌组合而成,用于防治仔猪腹泻,具有显著的效果,从而可以在防治仔猪腹泻过程中减少猪用抗生素的使用量。
为了实现上述目的,本发明提供了一种防治仔猪腹泻的复合益生菌制剂,所述复合益生菌制剂包括5种益生菌,所述5种益生菌分别为Lactobacillus frumenti、Lactobacillus gasseri LA39、Butyricicoccus pullicaecorum、Eubacterium hallii和Blautia hansenii。
进一步地,所述复合益生菌制剂中,总活菌数量为101~1020个/ml。
再进一步地,所述复合益生菌制剂中,总活菌数量为103~1015个/ml。
再进一步地,所述复合益生菌制剂中,Lactobacillus frumenti、Lactobacillusgasseri LA39、Butyricicoccus pullicaecorum、Eubacterium hallii和Blautiahansenii的活菌数量比为1:0.1~10:0.1~10:0.1~10:0.1~10。
再进一步地,所述复合益生菌制剂中,Lactobacillus frumenti、Lactobacillusgasseri LA39、Butyricicoccus pullicaecorum、Eubacterium hallii和Blautiahansenii的活菌数量比为1:0.2~5:0.2~5:0.2~5:0.2~5。
本发明还提供了一种上述防治仔猪腹泻的复合益生菌制剂的制备方法,包括以下步骤:
1)将Lactobacillus frumenti、Lactobacillus gasseri LA39、Butyricicoccuspullicaecorum、Eubacterium hallii和Blautia hansenii五种菌株分别接种到新鲜的液态培养基中进行培养,得到五种细菌悬液;
2)将制得的五种细菌悬液分别离心获取细菌沉淀,用磷酸盐缓冲液PBS重悬细菌沉淀,将五种活菌悬液混合均匀,得到复合益生菌制剂;其中,每毫升复合益生菌制剂中总活菌数量为101~1020个,优选为103~1015个。
再进一步地,所述复合益生菌制剂中,Lactobacillus frumenti、Lactobacillusgasseri LA39、Butyricicoccus pullicaecorum、Eubacterium hallii和Blautiahansenii的活菌数量比为1:0.1~10:0.1~10:0.1~10:0.1~10,优选为1:0.2~5:0.2~5:0.2~5:0.2~5。
本发明还提供了一种防治仔猪腹泻的复合益生菌制剂在防治仔猪腹泻中的应用,将制备的复合微生态制剂灌服至仔猪胃中,灌服剂量为1~10毫升/次,日服一次。
上述Lactobacillus frumenti、Lactobacillus gasseri LA39和Blautiahansenii购于日本微生物菌种保藏中心(JCM)。上述Butyricicoccus pullicaecorum、Eubacterium hallii购于德国生物资源保藏中心(DSMZ)。
如需长途运输或者长期存放,为了保持复合益生菌制剂中菌的活性,可以使用以下两种方法:
1)向不同菌株的活菌悬液中分别加入甘油得到细菌冻存液(甘油的体积分数为10%),置于液氮中低温保存。使用前通过37℃水浴快速解冻菌液,然后将制得的细菌悬液分别离心获取细菌沉淀,用磷酸盐缓冲液(PBS)重悬细菌沉淀,按Lactobacillusfrumenti、Lactobacillus gasseri LA39、Butyricicoccus pullicaecorum、Eubacteriumhallii和Blautia hansenii的活菌数量比1:0.2~5:0.2~5:0.2~5:0.2~5混合均匀(优选为1:0.2~5:0.2~5:0.2~5:0.2~5),得到复合益生菌制剂。
2)向制得的复合益生菌制剂中加入甘油得到细菌冻存液(甘油的体积分数为10%),置于液氮中低温保存。使用前通过37℃水浴快速解冻菌液,离心获取细菌沉淀,用磷酸盐缓冲液(PBS)重悬细菌沉淀,得到复合益生菌制剂。
本发明的有益效果在于:
本发明的益生菌制剂的制作过程简单,生产成本较低,具有显著的防治仔猪腹泻效果。
具体实施方式
为了更好地解释本发明,以下结合具体实施例进一步阐明本发明的主要内容,但本发明的内容不仅仅局限于以下实施例。
复合益生菌制剂的制备方法为:
1)通过37℃水浴快速复苏液氮冻存的菌液,接种到新鲜的液态培养基中富集培养。待液态培养基变浑浊,用接种环沾取适量菌液划线接种于固态培养基中。固态培养基中长出单菌落后,挑取单菌落接种到新鲜的液态培养基中富集培养。待液态培养基变浑浊,提取菌液中细菌基因组DNA,PCR扩增16S rDNA全长并纯化回收。将回收的16S rDNA片段送至测序公司进行测序,经测序结果与NCBI数据库进行比对,验证挑取的单菌落是否为原菌株。将经测序比对验证成功的菌液接种到新鲜的液态培养基扩大培养。采用活菌计数法对菌液中的活菌计数,并根据计数结果将菌液中的活菌数量调整为101~1020个/毫升。向不同菌株的菌液中分别加入甘油得到细菌冻存液(甘油的体积分数为10%),置于液氮中低温保存。
2)使用前通过37℃水浴快速解冻菌液,然后将制得的细菌悬液分别离心获取细菌沉淀,用磷酸盐缓冲液(PBS)重悬细菌沉淀,按Lactobacillus frumenti、Lactobacillusgasseri LA39、Butyricicoccus pullicaecorum、Eubacterium hallii和Blautiahansenii的活菌数量比1:0.2~5:0.2~5:0.2~5:0.2~5混合均匀,得到复合益生菌制剂。其中,每毫升复合益生菌制剂中总活菌数量为103~1015个。
在仔猪日龄第6-20天,将制备的复合益生菌制剂灌服至长×大二元杂交仔猪胃中,灌服剂量为1~10毫升/次,日服一次。与实施磷酸盐缓冲液(PBS)灌服的对照组相比,该益生菌制剂灌服组在断奶(21日龄)后第1~11天阶段的腹泻率显著降低了80%。该复合益生菌制剂有效预防了仔猪腹泻。
同时,将制备的复合益生菌制剂分别灌服至15头断奶前发生腹泻的仔猪胃中,灌服剂量为1~10毫升/次,日服一次。次日,仔猪的腹泻率降低了50%。第三日,所有腹泻仔猪的腹泻症状基本消失。该复合益生菌制剂有效治疗了仔猪腹泻。
其它未详细说明的部分均为现有技术。尽管上述实施例对本发明做出了详尽的描述,但它仅仅是本发明一部分实施例,而不是全部实施例,人们还可以根据本实施例在不经创造性前提下获得其他实施例,这些实施例都属于本发明保护范围。

1.一种防治仔猪腹泻的复合益生菌制剂,其特征在于:所述复合益生菌制剂由5种益生菌组成,所述5种益生菌分别为Lactobacillus frumentiLactobacillus gasseri LA39Butyricicoccus pullicaecorumEubacterium halliiBlautia hansenii;所述复合益生菌制剂中,总活菌数量为103~1015个/ml;所述复合益生菌制剂中,Lactobacillus frumentiLactobacillus gasseri LA39Butyricicoccus pullicaecorumEubacterium halliiBlautia hansenii的活菌数量比为1: 0.2~5:0.2~5:0.2~5:0.2~5。
2.一种权利要求1所述防治仔猪腹泻的复合益生菌制剂的制备方法,其特征在于:包括以下步骤:
1)将Lactobacillus frumentiLactobacillus gasseri LA39Butyricicoccus pullicaecorumEubacterium halliiBlautia hansenii五种菌株分别接种到新鲜的液态培养基中进行培养,得到五种细菌悬液;
2)将制得的五种细菌悬液分别离心获取细菌沉淀,用磷酸盐缓冲液PBS重悬细菌沉淀,将五种活菌悬液混合均匀,得到复合益生菌制剂。
