Abstract
本发明涉及腐食酪螨消化系统的一种染色方法,其特征是在温度15℃‑35℃、湿度70%‑95%条件下,将0.05%‑5%的荧光素钠或品红溶液倒入培养皿中,再将拉伸好的parafilm膜剪成直径小于培养皿的圆形,然后将剪好的parafilm膜平放在培养皿内的荧光素钠或品红溶液表层使parafilm膜浮在培养皿溶液上,最后将饥饿48‑72小时的腐食酪螨放在parafilm膜上,3小时后即可在显微镜下观察到腐食酪螨消化系统。本发明操作简单,能够为在荧光或普通体式显微镜下观察到腐食酪螨的消化道、整个消化系统提供技术参考。

技术领域
本发明涉及腐食酪螨消化系统的一种染色的方法,属于现代农业害虫生物防治领域。
背景技术
目前,国内对腐食酪螨的研究还主要局限于分类、饲养及应用方面,对于腐食酪螨体内的微观结构,尤其是消化系统的机构研究甚少。
腐食酪螨消化系统是研究其取食消化食物的重要结构,能够观察到腐食酪螨取食食物后食物在螨类体内的走向,对于研究腐食酪螨消化系统,以及简便有效标记食物的走向具有重要意义。
然而,对于如何便于观察腐食酪螨消化系统,并没有相关简易方法的报道。
本发明的是为了提供一种简易高效的腐食酪螨消化系统染色方法,并且不会导致螨类致死。
发明内容
本发明的目的是提供一种便于观察腐食酪螨消化系统的简易、具体染色方法,能够实现在显微镜下即可观察到腐食酪螨食物走向、储存位置的目的。
本发明是在温度15℃-35℃、湿度70%-95%条件下,将0.05%-5%的荧光素钠或品红溶液倒入培养皿中,再拉伸parafilm膜,并将拉伸好的parafilm膜剪成直径小于培养皿的圆形,然后将剪好的parafilm膜平放在培养皿内的荧光素钠或品红溶液表层使parafilm膜浮在培养皿溶液上,最后将饥饿48-72小时的腐食酪螨放在parafilm膜上,3小时后即可在显微镜下观察到腐食酪螨消化系统。
本发明操作简单,在显微镜下就能够观察到腐食酪螨的消化道、整个消化系统的轮廓。
具体实施方式
实施例 腐食酪螨的染色方法
在温度25℃湿度85%条件下,将0.1%荧光素钠30ml倒入直径为5cm的培养皿中,再拉伸parafilm膜,并剪出直径为3cm的圆形,然后将剪好的parafilm膜平放在培养皿内的荧光素钠溶液表层使parafilm膜浮在培养皿溶液上,最后将挑好放在2毫升离心管中饥饿72小时的腐食酪螨放在膜上,5小时后即可在荧光体式显微镜下观察到腐食酪螨消化系统。

1.腐食酪螨消化系统的一种染色方法,其特征是在温度15℃-35℃、湿度70%-95%条件下,将0.05%-5%的荧光素钠或品红溶液倒入培养皿中,再拉伸parafilm膜,并将拉伸好的parafilm膜剪成直径小于培养皿的圆形,然后将剪好的parafilm膜平放在培养皿内的荧光素钠或品红溶液表层使parafilm膜浮在培养皿溶液上,最后将饥饿48-72小时的腐食酪螨放在parafilm膜上,3小时后即可在显微镜下观察到腐食酪螨消化系统。
