Abstract
本发明属于生物技术领域,公开了用于促进鱼的生长的手段和方法。具体地,本发明提供了一种工程质粒、重组蛋白、重组枯草芽孢杆菌、动物食物/饲料添加剂,制备重组枯草芽孢杆菌的方法,制备培养物溶解产物的方法,和提高鱼的生长速率的方法。使用本发明提供的动物食物/饲料添加剂喂养鱼,可以促进鱼的生长。

技术领域
本发明涉及用于促进鱼的生长的生物工程手段和方法。
背景技术
生长速率是渔业(fish farm)认为最重要的焦点之一,因为在生长性能和产量(由此获得的利润)之间存在直接关系。因此,在该行业中已经进行过不同的尝试以提高鱼的生长速率。
鱼生长激素(FSH)被提出用于促进鱼的生长。使用FSH促进鱼生长的一种方式是在溶解有FSH的水浴中浸泡鱼。然而,该方式存在较严格的限制。例如,在大规模中,以该方式使用FSH是低效的、昂贵的且不切实际的。第二,许多鱼不能以这种方式很好地吸收FSH。另一个限制在于,尚不清楚食用以这种FSH处理的鱼是否安全。
本发明寻求解决上述问题,或至少为公众提供有用的备选。
发明内容
根据本发明的第一方面,提供了一种在枯草芽孢杆菌(B.subtilis)宿主中表达编码荷那龙罗非鱼(Oreochromis hornorum)的鱼生长激素(tiGH)的基因的方法,该方法包括以下步骤:
—制备重组DNA表达盒(DNA cassette),所述DNA表达盒从5’端到3’端包括:三对
—以串联的形式存在的vegA启动子(vegA)和lac操纵基因(lacO)、
枯草芽孢杆菌共有核糖体结合位点(RBS)、ATG起始密码子、
编码所述tiGH的DNA序列和编码6个组氨酸残基(6x His标签)的DNA低聚核苷酸序列;
—在枯草芽孢杆菌或大肠杆菌(E.coli)的穿梭载体中插入所述DNA表达盒,从而形成重组构建体3vegAtiGHH6;
—将所述重组构建体转化至所述枯草芽孢杆菌宿主中;
—使所述枯草芽孢杆菌宿主在细胞培养物中生长并且在细胞内表达所述tiGH;和
—使用French压力匀浆器(French press homogenizer),通过机械溶解所述枯草芽孢杆菌,释放所述tiGH。
根据本发明的第二方面,提供了一种在枯草芽孢杆菌宿主中表达编码荷那龙罗非鱼的鱼生长激素(tiGH)的基因的方法,该方法包括以下步骤:
—制备重组DNA表达盒,所述DNA表达盒包括:至少一对vegA启动子(vegA)和lac操纵基因(lacO)、枯草芽孢杆菌共有核糖体结合位点、ATG起始密码子、编码所述tiGH的DNA序列和编码6个组氨酸残基(6x His标签)的DNA低聚核苷酸序列;
—在枯草芽孢杆菌或大肠杆菌的穿梭载体中插入所述DNA表达盒,从而形成重组构建体3vegAtiGHH6;
—将所述重组构建体转化至所述枯草芽孢杆菌宿主中;
—使所述枯草芽孢杆菌宿主在细胞培养物中生长并且在细胞内表达所述tiGH;和
—释放所述tiGH。
优选地,tiGH可以为22kDa的单一多肽。重组DNA表达盒从5’端到3’端可以包括:三对以串联的形式存在的vegA启动子(vegA)和lac操纵基因(lacO)、枯草芽孢杆菌共有核糖体结合位点、ATG起始密码子、编码所述tiGH的DNA序列和编码6个组氨酸残基(6x His标签)的DNA低聚核苷酸序列。穿梭载体可以具有6.62-kb大小的pMTLBs72。在IPTG诱导剂和/或氯霉素抗生素的存在下,可以使转化后的枯草芽孢杆菌宿主生长,直至对数晚期或稳定期。
在某些实施方式中,tiGH可以通过机械的方式从枯草芽孢杆菌宿主释放,例如,通过使用French压力细胞匀浆器(French pressure cell homogenizer)。所述方法可以包括将枯草芽孢杆菌宿主的细胞离心下来以获取tiGH的步骤。
根据本发明的第三方面,提供了一种制备鱼饲料的方法,该方法包括上述在枯草芽孢杆菌宿主中表达编码荷那龙罗非鱼的鱼生长激素(tiGH)的基因的方法。
优选地,由细胞溶解产生的细胞溶解产物可以涂覆在市售可得的鱼饲料颗粒上或另外混合在市售可得的鱼饲料颗粒中。
在某些实施方式中,所述方法可以包括使用每ml基本上含有55.56μl所述细胞溶解产物的琼脂溶液制备溶液的步骤,其中,所述细胞溶解产物可以由基本上1.112ml细胞培养物制备得到,由该细胞培养物制备细胞溶解产物。琼脂溶液可以基本上含有0.5%蔗糖和1%琼脂,并且可以基本上在50℃的温度下制备。混合的颗粒可以随后在室温下干燥。鱼饲料可以基本上包括1μg所述tiGH/1g饲料。
根据本发明的第四方面,提供了一种工程质粒,该工程质粒含有编码序列,其中,所述编码序列包括以下:-
a)至少一对vegA启动子(vegA)和lac操纵基因(lacO);
b)共有核糖体结合位点(RBS);
c)编码罗非鱼(tilapia)生长激素(tiGH)的基因;和

优选地,所述质粒从5’端到3’端可以含有至少三对可操作地连接在一起的vegA启动子(vegA)和lac操纵基因(lacO)。tiGH可以为荷那龙罗非鱼的生长激素。tiGH在编码序列的3’末端与6-组氨酸密码子可操作地连接。
根据本发明的第五方面,提供了由整合了所述质粒的宿主表达的重组蛋白。优选地,所述蛋白可以在枯草芽孢杆菌中表达。
根据本发明的第六方面,提供了含有上述工程质粒的重组枯草芽孢杆菌(Bacillus subtilis)。重组枯草芽孢杆菌可以被配置以产生基因工程动物食物或饲料添加剂。重组枯草芽孢杆菌可以被配置以助于由其表达的tiGH的检测和纯化。
根据本发明的第七方面,提供了一种动物食物或饲料添加剂,其含有所述重组枯草芽孢杆菌的细胞、其溶解产物和/或由其表达的蛋白。所述动物食物或饲料添加剂可以分别为鱼食物或鱼食物添加剂。通过涂覆所述细胞、溶解产物和/或蛋白的方式,动物食物或饲料添加剂可以含有所述细胞、溶解产物和/或蛋白。动物食物或饲料添加剂可以含有培养物的溶解产物,其中悬浮着重组枯草芽孢杆菌和基本上以1:18的比率熔化的琼脂溶液,其中,所述熔化的琼脂溶液包括基本上0.5%(w/v)蔗糖。动物食物或饲料添加剂可以被配置用于喂养日本锦鲤(Japanese koi),用于提高其生长性能。
根据本发明的第八方面,提供了一种制备重组枯草芽孢杆菌的方法,该方法包括在野生型枯草芽孢杆菌中整合上述质粒的步骤。
根据本发明的第九方面,提供了一种制备培养物溶解产物的方法,该方法包括由培养物制备溶解产物的步骤,上述重组枯草芽孢杆菌在所述培养物中生长。
根据本发明的第十方面,提供了一种提高鱼的生长速率的方法,该方法包括使用所述动物饲料或饲料添加剂喂养鱼的步骤。生长速率可以包括总体重或总身长或二者。鱼可以为日本锦鲤。

具体实施方式
本发明涉及施用鱼生长激素(FGH)以提高鱼的生长速率。出于不同的原因,荷那龙罗非鱼生长激素(tiGH)为本发明的焦点之一。引出本发明的研究表明tiGH的重组形式不仅可以促进罗非鱼的硬骨鱼种属生长,而且还促进其它鱼的生长,例如,红鲑鱼(sockeyesalmon),罗非鱼、金鱼、鳟鱼的幼体,以及鲤鱼(carp)和天使鱼(angelfish)的仔鱼。更具体地,在本发明的过程期间的实验证明工程枯草芽孢杆菌(即,GRAS)系统可以在枯草芽孢杆菌宿主的细胞质中有效且高效率地表达具有活性和功能性的重组tiGH(rtiGH),在其C-末端含有6-组氨酸标签(rtiGH)。进行实验以证实产生的rtiGH的同一性,特别是通过蛋白印迹(Western blot)分析和质谱法。本发明还提出通过使用含有rtiGH的枯草芽孢杆菌的全细胞溶解产物产生的rtiGH制备鱼饲料的简易方法。在随机对照研究中进行测试以确定补充饲料与正常饲料促进鱼生长中的效力,采用日本锦鲤作为鱼模型的一个实例。在四个月的研究期间,比较处理组(分别为40.38g和2.17cm)和对照组(分别为31.58g和1.61cm)之间鱼的体重和长度。结论是处理组的鱼的生长显著快于对照组的鱼。喂食补充饲料的鱼显示出在所有时候都是健康的和身体发育良好的,活跃的并且渴望进食的。请参见图10的示例。
以下说明和实验更详细地证明本发明。
材料和方法
细菌菌株和培养基
用作rtiGH表达的宿主的枯草芽孢杆菌菌株1A751(eglSΔ102,bglT/bglSΔEV,npr,apr,his)从芽孢杆菌遗传库存中心,俄亥俄(Bacillus Genetics Stock Centre,Ohio)获得。用作重组DNA工作的中间体宿主的大肠杆菌菌株JM101(supE,thi,Δlac-proAB(F’,tra36,proAB+,lacIqZΔM15))如前所述。
枯草芽孢杆菌转化体于37℃下在补充10.5mg l-1氯霉素(Fluka)的2x LB培养基(20g l-1 Difco胰蛋白胨,10g l-1 Difco酵母提取物,20g l-1 NaCl)中生长。对于固体培养基,以1.5%(w/v)的浓度加入Bacto琼脂。分别使用氯化钙方法和Spizizen描述的方法使用重组质粒转化大肠杆菌和枯草芽孢杆菌。
鱼的种属和来源
(从香港Mermaid Koi中心)购买18条6-8个月大的日本锦鲤鱼仔鱼(18-25cm长;大多数:红白色)用于设置阴性实验。另一方面,44条10个月大的日本锦鲤鱼仔鱼(主要是红白色)由东河渔业有限公司(East River Fishing Limited)(中国广东省)友好捐赠,用于评价rtiGH的生物活性的随机对照研究。此外,8条锦鲤鱼(其中7条10个月大;1条22个月大)在玻璃缸(150cm×43cm×63cm)中单独喂养,并且喂食补充rtiGH的饲料。在5个月内监测它们的行为和外部特征。
质粒和重组tiGH的表达
6.62-kb枯草芽孢杆菌/大肠杆菌穿梭载体pMTLBs72从芽孢杆菌遗传库存中心获得。质粒pMTLBs72由一部分枯草芽孢杆菌pBS72质粒和一部分大肠杆菌pBR322序列组成。它是在枯草芽孢杆菌中低拷贝数和稳定的质粒,具有约6个拷贝/染色体。使用pMTLBs72作为载体(未公布的数据),如前构建改性质粒3vegAcenA。使用3vegAcenA,如下构建质粒3vegAtiGHH6。使用14个低聚引物:P1-P14(请参见显示为SEQ ID NOs.1-14的序列表),通过PCR重叠延伸先合成中间体;所得到的产物使用SpeI和SmaI切割,随后与经相同的两种酶限制性切割的载体3vegAcenA连接,以形成3vegAtiGHH6。请参见图1。
为了表达rtiGH,新鲜的枯草芽孢杆菌1A751(3vegAtiGHH6)转化体在补充氯霉素的50ml 2x LB培养基中于37℃下生长8小时。所有培养物随后被接种至具有氯霉素的500ml2x LB培养基中,于37℃下生长,直至A600值等于1.0,接着通过加入IPTG,至0.1mM的终浓度。继续细胞培养20小时时间段。然后,将培养物离心并溶解细胞团。
细胞溶解
将保留的细胞团溶解在25ml磷酸盐缓冲液(PB;1.44g l-1 Na2HPO4和0.24g l-1KH2PO4;pH 7.4),用压力细胞匀浆器(Stansted Fluid Power Ltd.,UK)将重悬液加工3次。收集溶解产物,用于分析和饲料制备。
rtiGH的分析
在12%(w/v)曲辛-SDS-聚丙烯酰胺凝胶(tricine-SDS-polyacrylamide gel)上分离的蛋白样品使用考马斯亮蓝R250(Coomassie Brilliant Blue R250)染色。使用His-探针抗体(H-3)(Santa Cruz,sc-8036)进行蛋白的蛋白印迹分析。通过如前所述的具有Mascot搜索的液相色谱法串联质谱法证实rtiGH的同一性。
补充细菌培养物溶解产物的鱼饲料的制备
在仅20秒的短时间内,0.9ml琼脂溶液(0.5%蔗糖,1%琼脂;50℃)以及50μl含有rtiGH(浓度100mg l-1)的细胞溶解产物的混合物显示被均匀地良好地吸附在5g鱼饲料(Aqua master,Koi Food,Wheat Germ,尺寸S)上。
为了制备补充rtiGH的鱼饲料,确定1g饲料含有1μg rtiGH;因此50μl rtiGH溶解产物被用于与0.9ml琼脂溶液混合。测定诱导的1A751(3vegAtiGHH6)培养物的溶解产物含有100mg l-1 rtiGH。确定1A751(3vegAtiGHH6)溶解产物的蛋白浓度为9.93g l-1,而宿主溶解产物(阴性对照)的蛋白浓度为9.78g l-1。由于相同浓度的溶解产物蛋白被用于制备各种饲料,1g对照饲料被校准为含有10μl宿主溶解产物。制备补充培养物溶解产物的饲料的示意性细节在图2中描述。最后,将涂覆的饲料在室温下空气干燥3天。
rtiGH的生物活性的评价
共享相同尺寸(168cm×64cm×16cm)的两个灰色塑料水槽用于喂养两组(处理组和对照组)鱼。两个槽中的所有变量保持相同。使用Qmax设定为2,200l h-1的再循环系统促进在其中换水。两个槽中的水条件如下保持恒定:温度23℃,pH为6,溶解的氧为6.3mg l-1。
在阴性实验中,给鱼喂食正常食物颗粒或补充缺乏rtiGH活性的枯草芽孢杆菌细胞溶解产物的食物颗粒。在随机对照研究中,给鱼喂食正常食物颗粒(对照组)或补充含有rtiGH的细胞溶解产物的食物颗粒(处理组)。以预定的时间间隔,对每一个鱼样品进行拍照(用于鉴定)和称重,并测量其身长。
结果
罗非鱼鱼生长激素在枯草芽孢杆菌中的表达
将通过基因组装得到的编码罗非鱼鱼生长激素(tiGH)的DNA序列克隆至3VegAcenA载体(未公布的数据)中,以形成表达rtiGH的构建体3VegAtiGHH6(参见图1)。在IPTG诱导下,枯草芽孢杆菌1A751(3VegAtiGHH6)培养物表现出表达高水平的细胞内rtiGH。当通过SDS-PAGE分析1A751的细胞溶解产物(3VegAtiGHH6)时,显示出明显的具有预期大小的rtiGH(22.6kDa)的条带(参见图3)。随后,通过质谱法证实该条带为tiGH(数据未显示)。rtiGH的产量估计为约100mg l-1。
涂覆有枯草芽孢杆菌细胞溶解产物的鱼食物颗粒的制备
加入鱼生长激素(FGH)的食物颗粒的常规用途不是新的。本发明的一个新颖性涉及将枯草芽孢杆菌溶解产物特定应用于鱼饲料的制备。虽然被认为是GRAS,但是枯草芽孢杆菌培养物存在令人反感的气味。为了解决枯草芽孢杆菌溶解产物具有不受鱼欢迎的气味的问题,本发明的另一方面涉及在食物颗粒上紧固地涂覆悬浮于细胞溶解产物中的rtiGH的有效方法。
在凝胶上显示的rtiGH良好的回收和完整的条带(参见图3)支持通过高压的细胞溶解方法为实际可行方法的结论。为了克服枯草芽孢杆菌溶解产物具有的令人讨厌的气味和在颗粒上涂覆溶解产物的效率低,解决软琼脂以及低水平的蔗糖的可加工性。使用该过程,细菌溶解产物均匀并且良好地涂覆在颗粒上(参见图2)。
选择日本锦鲤作为鱼模型
日本锦鲤(锦鲤)为在中国和日本最流行的观赏鱼之一。基于以下考虑,在研究中采用锦鲤作为鱼模型。锦鲤容易获得,不挑剔食物,快速生长,并且基本上被饲养用于观赏。
当给锦鲤喂食涂覆有枯草芽孢杆菌溶解产物的食物颗粒和涂覆有含有rtiGH的枯草芽孢杆菌溶解产物的食物颗粒时,鱼似乎很享受这两种类型的饲料,行为与喂食正常食物颗粒差不多相同。
枯草芽孢杆菌溶解产物对日本锦鲤的生长的作用
为了确定缺乏rtiGH的枯草芽孢杆菌细胞溶解产物对锦鲤的生长是否可能造成任何刺激作用,首先获得仅含有3VegA表达载体(数据未显示)的转化体枯草芽孢杆菌1A751(3VegA)。随后制备枯草芽孢杆菌1A751(3VegA)的培养物溶解产物,并且用于在食物颗粒上涂覆,从而产生用于比较的阴性对照(未经处理的颗粒)。将18条日本锦鲤分成两组,一组喂食未经处理的颗粒,另一组喂食正常食物颗粒。两组鱼的喂养方案(两次/天和0.5g/鱼)相同,研究持续50天。在研究结束时,关于在50天中鱼的总重量和长度变化进行评价(数据未显示),生长性能不存在显著差异,随后得出结论认为缺乏rtiGH活性的枯草芽孢杆菌溶解产物对日本锦鲤的生长不具有刺激作用。
rtiGH的生长刺激活性
通过对以下两组锦鲤的饮食供给,测定rtiGH的生长刺激作用。处理组喂食涂覆有枯草芽孢杆菌1A751(3VegAtiGHH6)培养物溶解产物的食物颗粒,而对照组喂食正常颗粒。以每月的间隔对每一个个体锦鲤进行生长参数(重量和长度)的测量(参见图4,图7)。在研究的前两个月,两组中大多数个体的重量减轻(参见图4),可能是鱼对各种环境影响的负反应的结果。后来,记录的两组中几乎所有的鱼的重量和长度测量结果均增加。到4个月实验结束时,处理组中的个体的重量和长度的增量显示出比对照组中的相应个体明显更好的性能(参见图5,图8)。处理组重量的总体平均增加量为27.9%,高于对照组(参见图6),而前者的长度的总体平均增加量为34.8%,高于后者(参见图9)。
喂食补充rtiGH的食物颗粒的日本锦鲤的外部观察(external view)
为了得到关于喂食补充含有rtiGH的枯草芽孢杆菌溶解产物的食物颗粒的日本锦鲤(处理组)的行为和外部特征的一些观点,在第二批鱼(材料和方法)到达后立即从其中分离出八个个体。这些鱼随后在玻璃缸中单独喂养(材料和方法),并且基本上采用与处理组相同的方式,仅喂食补充rtiGH的饲料。
喂养5个月后,所有鱼显示是健康的,多肉的(并且少数可能甚至被认为是短粗的),灵敏的且颜色清晰的。鱼不害怕站在附近的观察者,并且主动围绕水槽游动。更重要地是,它们似乎在所有时间都是饿的,并且无论何时有人经过水槽时,它们都保持高度警惕食物供应。
具有高蛋白含量的鱼饲料的可用性是在养鱼中考虑的主要成本因素。然而,因为在养鱼中饲料转化效率(feed conversion efficiency,FCE)高;例如,在喂养鲑鱼的过程中,估计FCE在1.1kg饲料/kg鲑鱼的范围。因此,为了提高鱼的生长性能,在头脑中闪过的好主意是通过增加鱼饲料中的蛋白含量以提高FCE。用于在鱼饲料中增加蛋白含量的常用成分包括鱼蛋白水解产物、盐水虾、乌贼内脏、大豆和蓝绿海藻。
在沿着相同研究路线的科学努力中,近来生物技术的发展已经能够开发转基因鱼或基因改性的(GM)鱼,其中关注的鱼种属被基因突变,从而导致内源性生长激素的过度产生,以提高鱼的生长速率。尽管FDA批准使用GM鱼用于人消费,但是这个问题一直备受争议。关于鱼生长激素(FGH)的应用的一个良好的折中办法似乎将它作为鱼膳食中的饲料补充使用。
使用多种方式已成功表达和引入了不同鱼种属的重组FGH(rFGH),包括注射、浸浴、口服孵育和供食至鱼模型,用于评价在既定的条件下它们促进生长的性能。然而,在真实情况下,由于养鱼涉及大规模养殖,前三种方法似乎不实用。关于第四种方法,供食(指的是给鱼喂食补充rFGH的食物颗粒)在技术上是可行的,只要生产关注的饲料的具有成本效益的方式是可用的即可。之前有报道表明补充表达橄榄比目鱼(olive flounder)的rFGH的冻干集胞藻(Synechocystis)(一种蓝绿海藻)的鱼饲料促进比目鱼类鱼的生长。然而,尽管饲料对生长性能具有正面影响,但是该报道未能解决存在于集胞藻中的毒素微囊藻素(Microcystin)对人健康造成的潜在危害作用。该重要的问题结合制备最终添加剂所需的相对长的时间,对所述喂养方法在养鱼中的广泛应用提出了困难挑战。
与此相反,关于本发明,rFGH表达宿主枯草芽孢杆菌被公认为是“GRAS”有机体应用。此外,其具有良好表征的遗传学,相对短的加倍时间(120分钟),简易和简单的处理过程,以及最重要的是规模扩大方便的且具有成本效益的生产均支持采用枯草芽孢杆菌作为宿主用于重组应用的可行性。实际上,制备补充表达rtiGH的枯草芽孢杆菌的全细胞溶解产物的鱼饲料(参见图2;材料和方法)的方法不仅方便而且容易扩大规模使用。无论供应是小规模还是大规模进行,补充饲料,就像其正常的相应物一样,方便配送及接近被正在被喂食的鱼。
在实验前,计划四个月的相对长的时间(除去在阴性实验上消耗的开始的两个月)用于实验。随后确定观察下的鱼由于新的环境和测量操作可能显示出不舒服的反应,从而影响它们的食欲并且导致在研究中在饮食组成和生长性能之间得出的结论的错误解释。如预测的,在处理组(喂食补充rtiGH的饲料;参见图4A)和对照组(喂食正常饲料;参见图4B)中大多数个体似乎表现出对它们的新到达和/或随后的测量确定产生非预期反应。令人感兴趣的是,这些副作用仅影响两组鱼的体重(参见图4),但是不影响身长(参见图7)。此外,在研究的第一个月和第二个月,鱼的重量减轻表现最明显。后来,鱼似乎适应了它们的环境并且恢复对食物的兴趣,因此使我们能够在饲料消耗和鱼的重量/长度增加之间得出相关性。
以上讨论的结果支持以下结论。首先,两组锦鲤鱼消耗一定的时间段以适应环境和/或将它们新引入的操作过程。第二,每一条鱼对新环境和/或新操作过程的反应不同,因此,需要监测个体鱼的反应。第三,由于适应需要时间,在研究的前两个月难以得出在饲料消耗和鱼的重量/长度增加之间可靠的相关性。第四,两组鱼的身长似乎对适应环境的反应不太敏感。然而,在研究的第三个月和第四个月,身长和体重的变化彼此密切一致,因此这使我们在饲料消耗和鱼的重量/长度增加之间得出相关性。
在研究的前两个月,不管可能引发处理组和对照组二者的鱼重量减轻的环境因素的可能存在(参见图3),两组鱼的体重和长度的总体平均增加量的差异提供了区分正常的鱼饲料和补充rtiGH的饲料之间的性能的决定性支持证据。虽然两组鱼中所用成员经四个月的研究表现出较重的重量和较长的长度(参见图5和图8),但是处理组的体重和长度的总体平均增加量分别为40.38g(参见图6)和2.17cm(参见图9),明显高于未处理组的值,分别为31.58g(参见图6)和1.61cm(参见图9)。事实上,两组数据之间的不一致均显著不同,重量总体平均增加27.9%(参见图6)和长度总体平均增加34.8%(参见图9)。
因此,在本申请中提供的我们的结果支持在枯草芽孢杆菌中表达的rtiGH在促进日本锦鲤鱼的生长中具有生物活性的结论。此外,获得枯草芽孢杆菌的全细胞溶解产物的便利性提供了用于制备补充rtiGH的鱼饲料的简易手段。继续努力改进tiGH表达,蛋白含量和枯草芽孢杆菌的生长密度以及扩大规模生产的条件,所述方法可以证明是在养鱼中大规模应用的具有成本效益的方法。
生长速率是渔业认为最重要的点之一,因为在生长性能和利润产量之间存在直接关系。由垂体产生的鱼生长激素(FGH),22kDa单一链多肽,不仅对生长具有积极影响,而且还具有多效性功能,这对于鱼在不利的条件(例如环境压力和感染)下更好地应对是关键的。
已经很好地表征了多种种属的FGH,并且它们的相应的重组体已经在不同的微生物宿主系统中表达,以用于研究。在多种FGH中,能在海水以及淡水中繁殖的成年罗非鱼之一荷那龙罗非鱼为本发明的焦点之一。罗非鱼的天然FGH(tiGH)为具有187个氨基酸的多肽,其与金枪鱼和黄尾鱼的FGH具有高水平(超过84%)的同源性,并且与鳟鱼和鲑鱼66%相同。TiGH还以重组体的形式在大肠杆菌和巴斯德毕赤氏酵母中表达。由于具有改进的可用性,已经对重组tiGH在影响鱼的生长速率的效力方面进行了充分的研究。在一个实施例中,尽管鳟鱼和鲑鱼的tiGH和FGH之间具有相对低水平(66%)的同一性,但是有报道表明来自大肠杆菌的纯化的重组tiGH不仅仅促进罗非鱼的硬骨鱼种属(Oreochromis mossambicus)的生长,而且还促进红鲑鱼(Oncorhynchus nerka)的生长。在另一个实施例中,由巴斯德毕赤氏酵母制备的重组tiGH表现出增强幼体的生长、存活和质量以及若干幼体和仔鱼的免疫,例如罗非鱼、金鱼和鳟鱼的幼体以及鲤鱼和天使鱼的仔鱼。
令人感兴趣的是,不像相对应的哺乳动物,显示可以口服给予FGH,以提高鱼的生长性能。该策略在硬骨鱼中表现良好,因为它们的肠上皮能吸收高分子量蛋白。如预期的,生物活性tiGH已显示被鱼肠良好吸收,以促进躯体生长。此外,口服给予FGH与其它种属的鱼的生长促进具有相关性,包括银鳟鲑鱼(coho salmon)、比目鱼(Paralichtysolivaceus)、鲈鱼(Perca fiuviatilis)和巨型鲶鱼(giant catfish)。更重要地是,在给予后90分钟,在鱼的血液、肠、肌肉或肝脏中不能检测到口服给予的FGH的踪迹。因此,口服给予FGH似乎提供了一种提高鱼的生长性能的安全方法。
使用FGH处理鱼样品的一种方式是通过在溶解有FGH的缓冲溶液中浸泡鱼。虽然经处理的鱼明显比用普通缓冲溶液浸泡的对照鱼更快速地生长,但是这种给予方式是低效的,劳动密集型的并且在大规模上不可行。
我们的实验室已经参与构建有效的微生物系统,用于生产商业用途的重组蛋白。除了良好表征的革兰氏阴性细菌大肠杆菌以外,我们认为未像大肠杆菌那样充分研究的革兰氏阳性细菌枯草芽孢杆菌具有很大的作为通用的重组体宿主系统的开发潜力。由于缺乏外部膜,枯草芽孢杆菌良好适合工程化用于分泌生产异种蛋白。此外,由于缺少外膜,枯草芽孢杆菌不含内毒素,因此它的使用通常被认为是安全的(GRAS)。
本发明开拓性的开发了枯草芽孢杆菌分泌系统,用于有益蛋白的重组生产,包括人表皮生长因子和分解纤维的细菌(纤维单胞菌,Cellulomonas fimi)的内切葡聚糖酶。随后,构建了枯草芽孢杆菌的veg I启动子的衍生物,并且证明它们在大肠杆菌中在功能上具有活性。
应理解的是,为了清楚,在单独的实施方式或实验的内容中描述的本发明的某些特征可以在单一实施方式中组合提供。相反地,为了简洁,在单一实施方式的内容中描述的本发明的各种特征可以单独地或以任何合适的亚组合提供。应注意的是,实施方式的某些特征通过非限制性实施例来说明。并且,本领域技术人员了解为了简洁的目的在以上未解释的现有技术。一些现有技术如下所示,它们通过引用将全文结合到本文中。

Claims (12)
1.一种在枯草芽孢杆菌宿主中表达编码荷那龙罗非鱼(Oreochromis hornorum)的鱼生长激素(tiGH)的基因的方法,该方法包括以下步骤:
—制备重组DNA表达盒,所述DNA表达盒从5’端到3’端包括:三对以串联的形式存在的vegA启动子(vegA)和lac操纵基因(lacO)、枯草芽孢杆菌共有核糖体结合位点(RBS)、ATG起始密码子、编码所述tiGH的DNA序列和编码6个组氨酸残基(6x His标签)的DNA低聚核苷酸序列;
—在枯草芽孢杆菌或大肠杆菌的穿梭载体中插入所述DNA表达盒,从而形成重组构建体3vegAtiGHH6;
—将所述重组构建体转化至所述枯草芽孢杆菌宿主中;
—使所述枯草芽孢杆菌宿主在细胞培养物中生长并且在细胞内表达所述tiGH;和
—通过溶解所述枯草芽孢杆菌释放所述tiGH。
2.一种在枯草芽孢杆菌宿主中表达编码荷那龙罗非鱼的鱼生长激素(tiGH)的基因的方法,该方法包括以下步骤:
—制备重组DNA表达盒,所述DNA表达盒包括:至少一对vegA启动子(vegA)和lac操纵基因(lacO)、枯草芽孢杆菌共有核糖体结合位点、ATG起始密码子、编码所述tiGH的DNA序列和编码6个组氨酸残基(6x His标签)的DNA低聚核苷酸序列;
—在枯草芽孢杆菌或大肠杆菌的穿梭载体中插入所述DNA表达盒,从而形成重组构建体3vegAtiGHH6;
—将所述重组构建体转化至所述枯草芽孢杆菌宿主中;
—使所述枯草芽孢杆菌宿主在细胞培养物中生长并且在细胞内表达所述tiGH;和
—释放所述tiGH。
3.根据权利要求2所述的方法,其中,所述tiGH为22kDa的单一多肽。
4.根据权利要求2或3所述的方法,其中,所述重组DNA表达盒从5’端到3’端包括:三对以串联的形式存在的vegA启动子(vegA)和lac操纵基因(lacO)、枯草芽孢杆菌共有核糖体结合位点、ATG起始密码子、编码所述tiGH的DNA序列和编码6个组氨酸残基(6x His标签)的DNA低聚核苷酸序列。
5.根据权利要求2或3所述的方法,其中,所述穿梭载体为6.62-kb大小的pMTLBs72。
6.根据权利要求2或3所述的方法,其中,在IPTG诱导剂和/或氯霉素抗生素的存在下,使转化后的枯草芽孢杆菌宿主生长,直至对数晚期或稳定期。
7.根据权利要求2或3所述的方法,所述方法包括将所述枯草芽孢杆菌宿主的细胞离心下来以获取所述tiGH的步骤。
8.根据权利要求1、2或3所述的方法,其中,通过使用French压力细胞匀浆器,以机械的方式实现所述tiGH的释放。
9.一种制备鱼饲料的方法,该方法包括权利要求1或3所述的方法。
10.一种工程质粒,该工程质粒含有:具有至少一对vegA启动子(vegA)和lac操纵基因(lacO)的编码序列、共有核糖体结合位点(RBS)、编码罗非鱼生长激素(tiGH)的基因和编码6-组氨酸密码子(6x His标签)的序列,其中,所述质粒从5’端到3’端还含有至少三对可操作地连接在一起的vegA启动子(vegA)和lac操纵基因(lacO)。
11.含有权利要求10所述的工程质粒的重组枯草芽孢杆菌。
12.一种动物食物或饲料添加剂,该动物食物或饲料添加剂含有权利要求11所述的重组枯草芽孢杆菌的细胞、其溶解产物或由其表达的蛋白。
